Summary Using fresh frozen (with and without semipermeable membranes), freeze-dried or sections from aldehyde fixed material and hexazotized p-rosaniline for simultaneous coupling more than 20 different unsubstituted or substituted l-amino acid naphthylamides are split especially in the microvilli and/or stereocilia of the small intestine, kidney and epididymis from rats. Further sites of positive reactions can be revealed by l-alanyl, l-leucyl, l-lysyl, ,l-glutamyl, ,l-glutamyl, l-asparaginyl, N-benzoyl-l-arginyl, N-carbobenzoxy-l-arginyl and N-benzoyl-l-phenylalanyl 2-naphthylamide. Among the substituted and unsubstituted peptide 2-naphthylamides l-prolyl-l-arginyl 2-naphthylamide is not hydrolysed in visible amounts; l-arginyl-l-arginyl, l-alanyl-l-arginyl-l-arginyl, l-alanyl-l-leucyl-l-tyrosyl, l-histidyl-l-seryl, l-seryl-l-tyrosyl and l-glycyl-l-phenylalanyl 2-naphthylamide are metabolized in the renal and intestinal brush border; the reaction pattern obtained with N-carbobenzoxy-l-glycyl-l-glycyl-l-arginyl 2-naphthylamide differs from that of N-carbobenzoxy-l-arginyl 2-naphthylamide. In addition l-glycyl-l-prolyl, l-leucyl-l-alanyl, l-lysyl-l-alanyl and l-alanyl-l-phenylalanyl-l-prolyl 2-naphthylamide are also split in the lysosomes of many organs and the secretion granules of gland cells.

Zusammenfassung Mit hexazotiertem p-Rosanilin als Simultankuppler werden in frischen (mit und ohne semipermeable Membranen), gefriergetrockneten und in Schnitten von aldehyd-fixiertem Material mehr als 20 unsubstituierte oder substituierte Aminosäure-2-naphthylamide nahezu ausschließlich in den Mikrovilli und/oder Stereocilien von Dünndarm, Nebenhoden und Niere von Ratten umgesetzt. Weitere positive Reaktionsorte liefern l-Alanyl-, l-Leucyl-, l-Lysyl-, ,l-Glutamyl-, ,l-Glutamyl-, l-Asparaginyl-, N-Benzoyl-l-arginyl-, N-Carbobenzoxy-l-arginyl- und N-Benzoyl-l-phenylalanyl-2-naphthylamid. Unter den Peptid-2-naphthylamiden wird l-Prolyl-l-arginyl-2-naphthylamid nicht in sichtbarer Menge hydrolysiert; l-Alanyl-l-arginyl-l-arginyl-, l-Arginyl-l-arginyl-, l-Asparaginyl-l-arginyl-, l-Alanyl-l-leucyl-l-tyrosyl-, l-Histidyl-l-seryl-, l-Seryl-l-tyrosyl- und l-Glycyl-l-phenylalanyl-2-naphthylamid setzen der renale und intestinale Bürstensaum um. N-Carbobenzoxy-l-glycyl-l-glycyl-l-arginyl-2-naphthylamid liefert verglichen mit Benzoyl- und N-Carbobenzoxy-l-arginyl-2-naphthylamid zusätzliche Resultate. Mit l-Glycyl-l-prolyl-, l-Leucyl-l-alanyl-, l-Lysyl-l-alanyl-und l-Alanyl-l-phenylalanyl-l-prolyl-2-naphthylamid reagieren die Lysosomen zahlreicher Organe und Sekretgranula positiv.

Summary Cell extracts ofCandida guilliermondii grown ind-xylose,l-arabinose,d-galactose,d-glucose,d-mannose and glycerol as sole carbon sources possessed NADPH-dependent aldose reductase activity, but no NADH-dependent activity was detected.d-xylose andl-arabinose were the best inducers of aldose reductase activity. The highest enzyme activity ind-xylose orl-arabinose-grown cells was observed first withl-arabinose followed byd-xylose as substrates of the enzymatic reaction. However, only low activity was found ind-glucose,d-mannose andd-galactose-grown cells, indicating that these carbon sources cause catabolite repression. Enzyme activities induced ind-xylose-grown cells were twice as high as those obtained from the cells under resting conditions. Furthermore, the level of induction of aldose reductase activity depended on the initial concentration ofd-xylose. The present study shows that aldose reductase activity may be efficiently induced by pentose sugars of hemicellulosic hydrolysates and weakly by hemicellulosic hexoses.  相似文献

The ability of Aspergillus fumigatus l-amino acid oxidase (l-aao) to cause the resolution of racemic mixtures of dl-amino acids was investigated with dl-alanine, dl-phenylalanine, dl-tyrosine, and dl-aspartic acid. A chiral column, Crownpak CR+ was used for the analysis of the amino acids. The enzyme was able to cause the resolution of the three dl-amino acids resulting in the production of optically pure d-alanine (100% resolution), d-phenylalanine (80.2%), and d-tyrosine (84.1%), respectively. The optically pure d-amino acids have many uses and thus can be exploited industrially. This is the first report of the use of A. fumigatus l-amino acid oxidase for racemic resolution of dl-amino acids.  相似文献

Summary L-Phenylalanine is a potent inhibitor of growth in a marine species of blue-green bacteria, Agmenellum quadruplicatum. The growth inhibition is reversed by many amino acids when added to the culture medium simultaneously with L-phenylalanine. The most effective L-phenylalanine antagonists are L-tyrosine, L-alanine, L-leucine, L-methionine, L-tryptophan, and L-isoleucine. However, L-tyrosine is the only effective L-phenylalanine antagonist when growth is inhibited by L-phenylalanine for two or more hours prior to addition of an equimolar concentration of the compound tested as an antagonist. Various explanations that could account for inhibition of growth by L-phenylalanine are discussed. Inhibition of growth by L-phenylalanine is not a feature peculiar to the general physiology of blue-green bacteria. For example, the growth of Anacystis nidulans, a fresh water species, was not inhibited by L-phenylalanine, although a different pattern of metabolite sensitivity was found.  相似文献

The metabolism of the natural amino acid l-valine, the unnatural amino acids d-valine, and d-, l-phenyglycine (d-, l-PG), and the unnatural amino acid amides d-, l-phenylglycine amide (d-, l-PG-NH2) and l-valine amide (l-Val-NH2) was studied in Pseudomonas putida ATCC 12633. The organism possessed constitutive l-amidase activities towards l-PG-NH2 and l-Val-NH2, both following the same pattern of expression, suggesting the involvement of similarly regulated enzymes, or a common enzyme. Quite surprisingly, growth in mineral media with l-PG-NH2 resulted in variable, long lag phases of growth and strongly reduced l-amidase activities. Conversion of d-PG-NH2 into d-PG and l-PG also occurred and could be attributed to the presence of an inducible d-amidase and the racemization of the amino acid amide in combination with l-amidase activity, respectively. The further degradation of l-PG and d-PG involved constitutive l-PG aminotransferase and inducible d-PG dehydrogenase activities, respectively, both with a high degree of enantioselectivity. N-Acyl-D-glutamate amidohydrolase (D-AGase) was inhibited by 94 % when 1 mol/l N-acetyl-DL- glutamate was used as a substrate. The addition of 1 mM Co2+ stabilized D-AGase. Moreover, the substrate inhibition was weakened to 88% with the addition of 0.4 mM Co2+ to the reaction mixture. Although D-AGase is a zinc-metalloenzyme, the addition of Zn2+ from 0.01 to 10 mM did not increase the D-glutamic acid production in the saturated substrate. Under optimal conditions, 0.38 M D-glutamic acid was obtained from N-acyl-DL-glutamate with 100% of the theoretical yield after 48 h.  相似文献

Results of a karyological analysis of eight species collected from northeast Anatolia, Turkey are presented. The following chromosome numbers were determined:H. medianiforme (Litv. etZahn)Juxip 2n=36,H. karagollense (Zahn)P.D. Sell etC. West 2n=36,H. argillaceoides (Litv. etZahn)Juxip 2n=36,H. amblylepis Boiss. 2n=27,H. lanceolatum Vill. 2n=27,H. asterodermum (Woronow etZahn)Juxip 2n=27,H. umbellatum L. 2n=18,H. laevigatum Willd. 2n=27. Summary It is suggested that the construction of molecular models of the chromosome may prove to be a fruitful way of attacking the fundamental problems of cytology and genetics. Certain micelles consisting of molecules of protamine-nucleate are suggested for consideration. They are designed to interpret in molecular terms the postulates of genetics and the nature of the gene, and are constructed with special reference to the findings of the cytologists as to the capacity of the chromosomes to swell, to contract, to grow and to divide.Based upon a lecture given in the physics department of the University of Manchester on May 13, 1935. Summary A new species from Egypt (Sinai) and Arabia is described asStipagrostis multinerva H.Scholz. Stipagrostis arabiaefelicis Bor 1967 is a synonym ofSt. drarii (V.Täckholm)de Winter 1963 (Aristida drarii V.Täckholm 1941). The taxonomic status ofStipagrostis foexiana (Maire etWilczek)de Winter (Aristida obtusa subsp.pubescens Andreánszky) in North and South Africa is discussed. l-2-Aminobutyric acid can be synthesized in a transamination reaction from l-threonine and l-aspartic acid as substrates by the action of threonine deaminase and aromatic aminotransferase, but the by-product l-alanine was produced simultaneously. A small amount of l-alanine increased the complexity of the l-2-aminobutyric acid recovery process because of their extreme similarity in physical and chemical properties. Acetolactate synthase has been introduced to remove the pyruvate intermediate for reducing the l-alanine concentration partially. To eliminate the remnant l-alanine, alanine racemase of Bacillus subtilis in combination with d-amino acid oxidase of Rhodotorula gracilis or Trigonopsis variabilis respectively was introduced into the reaction system for the l-2-aminobutyric acid synthesis. l-Alanine could be completely removed by the action of alanine racemase of B. subtilis and d-amino acid oxidase of R. gracilis; thereby, high-purity l-2-aminobutyric acid was achieved. The results revealed that alanine racemase could discriminate effectively between l-alanine and l-2-aminobutyric acid, and selectively catalyzed l-alanine to d-alanine reversibly. d-Amino acid oxidase then catalyzed d-alanine to pyruvate stereoselectively. Furthermore, this method was also successfully used to remove the by-product l-alanine in the production of other neutral amino acids such as l-tertiary leucine and l-valine, suggesting that multienzymatic whole-cell catalysis can be employed to provide high purity products.  相似文献
