Light Control of Anthocyanin Biosynthesis in Zea Seedlings   总被引:2,自引:0,他引:2  
Evidence for involvement of two non-photosynthetic pigments in photoinduction of anthocyanin biosynthesis in the roots and mesocotyls of Zea mays L. seedlings is presented. Short (5 min), low energy (4.5 × 103 J m?2) fluences of red light neither induced anthocyanin synthesis nor enhanced phenylalanine ammonia-lyase activity in dark-grown maize seedlings. Little anthocyanin synthesis and no enhancement of phenylalanine ammonia-lyase activity was induced by continuous far-red light. Continuous white or blue light induced both anthocyanin synthesis and enhanced phenylalanine ammonia-lyase activity. These results show that phytochrome alone cannot induce anthocyanin synthesis in maize seedlings. However, a strong phytochrome mediation of white light induced pigment synthesis was demonstrated. This effect was not demonstrable with white light enhanced phenylalanine ammonia-lyase activity, indicating that phytochrome controls another step in anthocyanin biosynthesis.  相似文献

The promotion of anthocyanin synthesis in red-cabbage seedlings by 5 min exposure to R light is inhibited by subsequent application of CaCl2. The stimulation of dark synthesis of anthocyanin by n-PrOH and by kinetin is also reduced by Ca2+ and by cholesterol, both of which are well known to stabilize cell membranes. By contrast, EDTA, which chelates Ca2+, promotes dark synthesis of anthocyanin. Assay of native Ca2+ extractable from seedlings immersed in EDTA demonstrates that R light exposure promotes a highly significant increase in extractable Ca2+. It is suggested that the molecular configuration of the phytochrome molecule affects the ability of a membrane to bind Ca2+ and that this in turn affects the permeability to substrates which are required for anthocyanin biosynthesis.  相似文献

-The synthesis of anthocyanin in red-cabbage is very sensitive to control by light, R/FR reversibility being effected by exposures of 5 min duration, The demonstration of this control does not depend upon a preceding irradiation of high-intensity light but depends upon the duration of incubation in darkness subsequent to irradiation. R/FR reversibility is well shown in seedlings kept in darkness for 48 hr after exposure but after 120 hr this reversibility is no longer evident. This is due to the fact that a further synthesis of anthocyanin occurs in unexposed seedlings and in FR and R/FR treated material in the period from 48 to 120 hr but does not occur in the R treatment after 48 hr. Reagents such as n-propanol which are believed to increase membrane permeability, greatly increase anthocyanin synthesis in dark grown material. n-PrOH also reverses the effect of 5 min FR irradiation but, by contrast with R light, does not promote PAL activity. It is concluded that the limitation to synthesis in material unexposed to light is substrate availability at the site of flavonoid biosynthesis, rather than the level of PAL activity. The evidence presented supports the hypothesis that R/FR reversible phytochrome action involves the control of the passage of substrate through a membrane to the site of anthocyanin biosynthesis.  相似文献

Differences in the extent of anthocyanin production between intermittent light treatments with short and long dark intervals between successive irradiations are more pronounced in dark-grown than in light-pretreated cabbage seedlings. This observation is consistent with the hypothesis, based on destruction kinetics data, that there might be two pools of phytochrome, a labile one and a stable one, present in different proportions in dark-grown and light-pretreated seedlings, and suggests that light-dependent changes of the stable to labile phytochrome ratio might be physiologically significant in the photoregulation of photomorphogenic responses.  相似文献

Transgenic tomato [Lycopersicon esculentum (=Solanum lycopersicum)] lines overexpressing tomato PHYA, PHYB1, or PHYB2, under control of the constitutive double-35S promoter from cauliflower mosaic virus (CaMV) have been generated to test the level of saturation in individual phytochrome-signalling pathways in tomato. Western blot analysis confirmed the elevated phytochrome protein levels in dark-grown seedlings of the respective PHY overexpressing (PHYOE) lines. Exposure to 4 h of red light resulted in a decrease in phytochrome A protein level in the PHYAOE lines, indicating that the chromophore availability is not limiting for assembly into holoprotein and that the excess of phytochrome A protein is also targeted for light-regulated destruction. The elongation and anthocyanin accumulation responses of plants grown under white light, red light, far-red light, and end-of-day far-red light were used for characterization of selected PHYOE lines. In addition, the anthocyanin accumulation response to different fluence rates of red light of 4-d-old dark-grown seedlings was studied. The elevated levels of phyA in the PHYAOE lines had little effect on seedling and adult plant phenotype. Both PHYAOE in the phyA mutant background and PHYB2OE in the double-mutant background rescued the mutant phenotype, proving that expression of the transgene results in biologically active phytochrome. The PHYB1OE lines showed mild effects on the inhibition of stem elongation and anthocyanin accumulation and little or no effect on the red light high irradiance response. By contrast, the PHYB2OE lines showed a strong inhibition of elongation, enhancement of anthocyanin accumulation, and a strong amplification of the red light high irradiance response.  相似文献

It has been reported that pigmentation In plants Is stimulated by light and cytoklnln (CTK); however, the signaling pathways and the relationship between light and CTK Involved In the regulation of anthocyanln accumulation remain to be elucidated. We Investigated (i) the role of blue light (BL) and CTK In anthocyanln accumulation ; and (ii) the relationship between BL and CTK In wild type (WT) and by4 mutants of Arabidopsis thaiiana. Two-d-old seedlings grown on medium with or without klnetln (KT) or zeatln (ZT) In darkness were Irradiated using BL at different fluence rates for 3 d before the anthocyanln content was determined using a spectrophotometrlc method. Anthocyanln accumulation was strongly Induced by BL In WT seedlings but not In hy4 seedlings, which demonstrated that CRY1 Is the main photoreceptor for BL. Both KT and ZT enhanced the response of the WT seedlings to BL In a dose-dependent manner, whereas they were not sufficient to promote anthocyanln eccumulatlon In darkness. In addition, data from experiments using the hy4 mutant showed that the CTK effect of BL was also CRYl-dependent. The results from experiments with three different treatment programs showed that the relationship between BL and KT In anthocyanln accumulation of Arabidopsis seedlings seems neither muItlpllcatlve nor additive coactlon, but rather Interaction. BL Is necessary for anthocyanln accumulation, and KT might be Involved In the BL signaling pathway.  相似文献

Gibberellic acid (GA3) inhibits amaranthin synthesis whereas the growth retardant, phosphon D, enhances pigment levels in A. caudatus seedlings exposed to light. No effect was observed on chlorophyll and carotenoid synthesis. Radioactive tyrosine and DOPA were incorporated into amaranthin. The specific activity of amaranthin synthesised in the presence of 14C-tyrosine or 14C-DOPA in seedlings treated with GA3 is higher than water controls. The specific activity of pigment from phosphon D treated tissue is relatively low. GA3 treated tissue has lower active tyrosine and DOPA pools compared to phosphon treated seedlings. Tyrosine and DOPA-oxidase activity increases in GA3 treated and H2O control seedlings exposed to light. Kinetin stimulates the synthesis of amaranthin in dark-grown seedlings and this is not overcome by simultaneous GA3 application. Dark-grown seedlings treated with different kinetin concentrations and incubated in 14C-tyrosine synthesise radioactive amaranthin of similar specific activity. Kinetin treatment of dark-grown seedlings brings about an increased tyrosine and DOPA-oxidase activity. The results indicate that GA3 controls the production and/or availability of tyrosine whereas kinetin can mimic light treatment and controls the utilisation of tyrosine probably by bringing about the synthesis or activation of tyrosine and DOPA-oxidase protein.  相似文献

The action of light on ribosome formation was examined in the cabbage seedlings, a system extensively used in the studies of anthocyanin synthesis. Ribosomes were extracted 18 h after the beginning of the irradiation and separated by sucrose gradient centrifugation. In the cotyledons of dark-grown cabbage seedlings, a brief red light induces an increase both in total ribosomes and in the fraction present as polysomes; the effect of red light is reversed by far red light, indicating the involvement of phytochrome in polysome formation in cabbage seedlings. Continuous red and continuous far red light are about equally effective in bringing about an increase of total ribosomes and of the polysome fraction. Streptomycin, which inhibits chlorophyll synthesis and chloroplast development, and enhances anthocyanin synthesis in cabbage seedlings, causes a decrease of total ribosomes and of the fraction present as polysomes. In hypocotyls, the red-far red reversibility is evident only for the polysome content and streptomycin does not decrease the polysome/monosomo ratio as it does in cotyledons.  相似文献

The vacuoles in the epidermal cells of light-grown seedlingsof Polygonum cuspidatum were found to contain intensely pigmentedspherical bodies (anthocyanoplasts). Dark-grown seedlings containedsimilar spherical bodies that were unpigmented. The unpigmentedspherical bodies accumulated anthocyanin and turned into anthocyanoplastswhen dark-grown seedlings were irradiated. The unpigmented sphericalbodies of dark-grown seedlings were heavily stained upon treatmentof seedlings with neutral red. The absorption spectra of sphericalbodies and vacuoles showed that the contents of the sphericalbody were different from those of the vacuole. The sphericalbody was stable in darkness or at low temperature but was unstableat 25°C in the light. Abstract. A brief high temperature treatment (45 C) promoted anthocyanin synthesis in 2-d-old dark-grown red cabbage seedlings. Polygonum cuspidatum seedling. Anthocyanin accumulated first in the lower part of hypocotyls and then the site of accumulation gradually extended toward the upper part of hypocotyls when seedlings were irradiated with white light (WL) at 25 C. Etiolated seedlings accumulated anthocyanin only in the upper parts (hook and cotyledons) when the seedlings were irradiated with WL at 5 C. De-etiolated seedlings that had been pre-irradiated with WL for 1 day at 25 C accumulated anthocyanin both in upper and lower parts of the seedlings when the seedlings were irradiated with WL at 5 C. Spectral sensitivity was dependent on the temperature during irradiation. Red light (R), blue light (B), and near ultra-violet light (NUV) induced the accumulation of anthocyanin at 5 C but only NUV was effective in inducing the accumulation of anthocyanin at 25 C. Dichlorophenyl dimethylurea (DCMU) inhibited WL-induced anthocyanin accumulation but did not NUV-induced anthocyanin accumulation at 25 C. However, sucrose promoted NUV action at 25 C, indicating that photosynthesis can promote NUV-induced anthocyanin accumulation. Distribution of phytochrome in etiolated seedlings, that was examined by spectrophotometry, was similar to the distribution of anthocyanin at 5 C. Seedlings of 17 rice (Oryza sativa L.) cultivars were classified on the basis of anthocyanin pigmentation into three groups: an acyanic group with 9 cultivars, a moderately cyanic group with 5 cultivars, and a cyanic group with 3 cultivars. Seedlings of the cyanic group were deep purple in color, possessing copious amounts of anthocyanin in shoots. Sunlight (SL)-mediated anthocyanin and phenylalanine ammonia lyase (PAL) induction in a cyanic cultivar, purple puttu, was compared with an acyanic cultivar, black puttu. A brief exposure of dark-grown purple puttu seedlings to SL induced anthocyanin formation during a subsequent dark period with a peak at 24 h. The magnitude of SL-mediated anthocyanin induction is age dependent, the 4-d-old seedlings being the most responsive to SL. The anthocyanin induction in purple puttu seedlings is mediated exclusively by the ultraviolet-B (UV-B) component of SL. The SL-triggered anthocyanin induction was reduced by about 30% by a terminal far-red light pulse and was restored by a red light pulse, indicating the role of phytochrome in modulation of anthocyanin level. The SL-mediated induction of PAL showed two peaks, one at 4 h and the other at 12 h. Whereas the first PAL peak (4 h) was induced by phytochrome and was seen in both cultivars, the second PAL peak (12 h) was inducible by UV-B only in the cyanic purple puttu cultivar.  相似文献

Previously, it has been demonstrated that the red light-inducedanthocyanin accumulation in mung bean seedlings is mediatedby phytochrome [Dumortier and Vendrig Z. Pflanzenphysiol. 87:313 (1978)]. In this paper the importance of phytochrome forthe accumulation of anthocyanins in seedlings of mung beanswas studied in non-irradiated seedlings and in seedlings irradiatedwith 5 min R. A short FR-irradiation given early after sowing reduced theamount of anthocyanins which were normally found in non-irradiatedseedlings. This indicates that PFR may be important for at leastpart of the anthocyanin synthesis in the dark. As for the redlight-mediated anthocyanin accumulation, irradiation appearedto be most effective when given to seedlings at the age of 36–48hr. Although the seedlings were sensitive to red light irradiationbefore that time, they were not able to synthesize anthocyaninsuntil they had reached the age of 36 hr. Complete escape ofred/far-red reversibility occurred only when far-red was given12 hr after red, although partial escape could be observed witha shorter time-interval. Furthermore, the time-course of anthocyaninaccumulation after a two-fold R-irradiation was compared withthe effect of a single R-exposure. From the results could beconcluded that the pattern of anthocyanin accumulation is dependenton the time during which PFR is present in the seedlings. Theseexperiments also indicate that PFR not only plays a role inthe synthesis of anthocyanins but probably also in their degradation. The results of our study show that phytochrome is importantfor anthocyanin accumulation in non-irradiated mung bean seedlingsas well as in R-irradiated, and that it probably is also involvedin the degradation of the pigment. Although the growth of intact plants is inhibited by irradiation with blue light, the growth rate of isolated stem segments is largely unaffected by blue light. We hypothesized that this loss of responsiveness was a result of ethylene production as part of the wounding response. However, we found no interaction between ethylene- and blue light-induced growth inhibition in dark- or red light-grown seedlings of pea (Pisum sativum L.). Inhibition of growth begins in dark-grown seedlings exposed to blue light within 3 min of the onset of blue light, as was known for red light-grown seedlings. By contrast, ethylene-induced inhibition of growth occurs only after a lag of 20 to 30 min or more (dark-grown seedlings) or 60 min (red light-grown seedlings). Also, the inhibition response of red light-grown seedlings is the same whether ethylene is present from the onset of continuous blue-light treatment or not. Finally the spatial distribution of inhibition following blue light was different from that following ethylene treatment.  相似文献

Results are reported which support the view that the photosynthetic photosystems are not involved in the high irradiance response (HIR) phenomenon of light-dependent anthocyanin biosynthesis in dark-grown Zea mays L. seedlings. A negative correlation between change in greening rates and change in light-dependent anthocyanin accumulation rates with age was demonstrated. Lack of chlorophyll synthesis in a strain of maize possessing a temperature-sensitive lesion for chlorophyll synthesis could not be correlated with light-induced anthocyanin accumulation. Furthermore, seedlings totally lacking photosynthetic capabilities, either due to a genetic lesion or to excision of all photosynthetic tissue, had an enhanced rate of photoinduced anthocyanin formation. This evidence indicates that the HIR results in the initiation of processes that are in competition with chloroplast development for substrate in normal, intact seedlings.  相似文献

The flavonoid pathway leading to anthocyanin biosynthesis in maize is controlled by multiple regulatory genes and induced by various developmental and environmental factors. We have investigated the effect of the regulatory loci R, B, and Pl on anthocyanin accumulation and on the expression of four genes (C2, A1, Bz1, and Bz2) in the biosynthetic pathway during an inductive light treatment. The results show that light-mediated anthocyanin biosynthesis is regulated solely by R; the contributions of B and Pl are negligible in young seedlings. Induction of the A1 and Bz2 genes by high fluence-rate white light requires the expression of a dominant R allele, whereas accumulation of C2 and Bz1 mRNA occurs with either a dominant or recessive allele at R. A1 and Bz2 mRNA accumulate only in response to high fluence-rate white light, but Bz1 is fully expressed in dim red light. Some C2 mRNA is induced by dim red light, but accumulation is far greater in high fluence-rate white light. Furthermore, expression from both dominant and recessive alleles of the regulatory gene R is enhanced by high fluence-rate white light. Seedlings with a recessive allele at R produce functional chalcone synthase protein (the C2 gene product) but accumulate no anthocyanins, suggesting that, in contrast to the R-mediated coordinate regulation of C2 and Bz1 observed in the aleurone, C2 expression in seedlings is independent of R and appears to be regulated by a different light-sensitive pathway.  相似文献

A comparative study of the spectral sensitivity of anthocyanin production in dark-grown and light-pretreated systems was carried out in Brassica oleracea L., Lycopersicon esculentum Mill., Secale cereale L. and Spirodela polyrrhiza L. Light pretreatments bring about an enhancement of the inductive, red-far red reversible response in all systems, a decrease of the continuous irradiation response in cabbage, rye, and tomato seedlings, and an enhancement of the continuous irradiation response in cabbage leaf disks. Light pretreatments also bring about a marked change in the spectral sensitivity of the continuous irradiation response. The different effect of light pretreatments on the photosensitivity of the response to short and long wavelength irradiations suggests that two photoreceptors, phytochrome and cryptochrome, may be involved in the photoregulation of anthocyanin production.  相似文献

Relative growth rate determinations on 5-millimeter regions of cucumber (Cucumis sativus L.) hypocotyls show that dim-red light-grown seedlings have an even distribution of growth along the stem axis. This contrasts with the apical to basal graded decline in growth rate seen in dark-grown seedlings, including dark-grown cucumber seedlings used as controls in this study. Dark-grown seedlings convert to the nongradient pattern when transferred to dim-red light. The small amount of light required suggests that the change in developmental pattern may happen in the natural light environment.  相似文献

The effect of different concentrations of growth regulators and sucrose on anthocyanin production in cell suspension cultures of Camptotheca acuminata Decaisne (Nyssaceae) was described for the first time and qualitatively and quantitatively evaluated. Anthocyanin production was significantly greater in the presence of kinetin, compared to benzyladenine, with the greatest concentration observed in the presence of 2 microM kinetin. No significant differences in anthocyanin production were observed when comparing 2,4-dichlorophenoxyacetic acid to alpha-naphthaleneacetic acid, except when using 2 microM, 2,4-dichlorophenoxyacetic acid, which resulted in greater anthocyanin production. High sucrose concentration enhanced the production of anthocyanins. Based on the absence of anthocyanin production in the dark, we concluded that light was essential for stimulating anthocyanin production. The optimised medium consisted of: 2 microM kinetin, 2 microM 2,4-dichlorophenoxyacetic acid and 292 mM sucrose. HPLC/DAD and HPLC/MS analyses revealed that the main anthocyanin was Cy 3-O-galactoside and that the minor derivative was Cy 3-O-glucoside.  相似文献

The exposure of dark-grown Pharbitis nil seedlings to continuous R induces a rapid decrease in PHYA mRNA abundance with a half-life of about 2 h. A 5 min R pulse also induces this decline, and the effect is partially reversible by subsequent FR irradiation, confirming that the regulation of expression is mediated via the Pfr form of a phytochrome. When de-etiolated seedlings are returned to darkness after a W photoperiod, PHYA mRNA slowly reaccumulates from 20% to 50% of the dark level within 24 h. The rate of reaccumulation is greatly accelerated by the removal of Pfr with a FR pulse, resulting in reaccumulation to 100% within approximately 11 h. Without FR irradiation PHYA mRNA expression remains fully repressed for at least 11 h after the end of the photoperiod, suggesting that the controlling Pfr is highly stable.  相似文献
