为探讨白蚁工蚁品级性腺不育的原因,采用组织学观察与测量对尖唇散白蚁Reticulitermes aculabialis Tsai et Hwang的工蚁和繁殖蚁的卵子发生各阶段进行比较和分析。结果表明,成熟工蚁的卵子发生与繁殖蚁相比,仅有卵母细胞的分化期和生长期,没有卵黄形成期,其分化期和生长期的卵母细胞大小与繁殖蚁这2个时期的卵母细胞大小呈显著性差异;生长期时,工蚁的卵母细胞和滤泡细胞之间出现较大间隙并且呈现萎缩退化状态;工蚁卵子发生相当于雌性繁殖蚁末龄若虫水平。该结果将为工蚁不能生殖提供组织学上的证据,同时又表明了工蚁具有潜在生殖能力的结构基础。  相似文献

为探讨白蚁非生殖品级和生殖品级生殖细胞发育差异,采用组织学染色技术对尖唇散白蚁Reticulitermes aculabialis Tsaiet Hwang繁殖蚁、工蚁和兵蚁的精巢发育以及精子发生进行了显微观察和比较研究。结果发现3个品级间精巢发育的程度差异很大,三者精巢切面面积相对大小之比为:繁殖蚁∶工蚁=1.7∶1;繁殖蚁∶兵蚁=29.3∶1;工蚁∶兵蚁=17.1∶1。繁殖蚁和工蚁精巢管内有精子的形成,工蚁和繁殖蚁精子的发生都经历了精原细胞、初级精母细胞、次级精母细胞、精细胞和精子时期,但工蚁有大量次级精母细胞呈细胞凋亡状态。兵蚁生殖细胞发育仅有精原细胞、初级精母细胞、次级精母细胞,没有精细胞和精子产生。工蚁的生殖细胞显著小于同一时期繁殖蚁的生殖细胞,兵蚁的各时期生殖细胞均极显著小于繁殖蚁同一时期的生殖细胞。研究表明各品级之间生殖功能分化与生殖细胞发育有直接关系,工蚁有转化为补充繁殖蚁和兵蚁的能力;而兵蚁由于精巢极度退化不能产生精细胞和精子,因此是非生殖品级分化的终极形式,不具有转化成补充繁殖蚁或其它品级的能力。  相似文献

为了探讨c-fos原癌基因在白蚁生殖品级和非生殖品级性腺发育中的作用,揭示白蚁不同品级性腺发育的调节机理,本研究运用免疫细胞化学定位方法对尖唇散白蚁Reticulitermes aculabialis繁殖蚁和工蚁精、卵发生过程中的c-fos蛋白表达进行了研究。结果显示:雌性繁殖蚁在末龄若虫期的卵子发生过程中有c-fos-like表达, c-fos-like免疫阳性物质定位于生长期的卵母细胞核和滤泡细胞核中;而繁殖蚁成虫的卵子发生过程中没有c-fos-like免疫阳性反应。雄性繁殖蚁在末龄若虫期时精子发生过程中没有c-fos-like表达, 而发育到成虫期有c-fos-like免疫阳性反应,阳性物质定位于精原细胞的细胞核中。工蚁精、 卵发生过程中均没有c-fos-like的表达。结果提示:c-fos在调节繁殖蚁精子和卵子发生方面有重要作用,c-fos可能通过调节精原细胞增殖参与精子的发生;在卵巢中可以直接作用于生长期的卵母细胞和滤泡细胞来调节卵子的发生。在工蚁性腺中c-fos表达缺失可能导致卵母细胞和滤泡细胞无法正常发育,精原细胞停止增殖而使精子发生处于相对抑制状态。工蚁性腺退化不育可能与c-fos没有正常表达有关。  相似文献

在黑胸散白蚁Reticulitermes chinensis发育和分化过程中,发现有假工蚁、假若蚁两种虫态。假工蚁由6龄和7龄工蚁转化发育而来,假若蚁由4龄和5龄若蚁转化发育而来。显微测量结果表明,黑胸散白蚁的胚后发育主要包括2个龄期的幼蚁期、6个以上龄期的工蚁期、4个龄期的若蚁期和有翅成虫。在此基础上分析了其他虫态的分化来源,发现兵蚁由3~7龄工蚁分化发育而来,翅鳞型和长翅芽型补充生殖蚁由6龄若蚁转化发育而来,短翅芽型补充生殖蚁由4龄和5龄若蚁转化发育而来,微翅芽型补充生殖蚁既可由4~6龄工蚁转化发育而来,又可由假工蚁和假若蚁转化发育而来,无翅型补充生殖蚁由3~7龄工蚁转化发育而来。提出了黑胸散白蚁群体中不同品级个体的可能分化途径。  相似文献

为了比较和探讨类雄激素受体 (androgen receptor-like,AR-like)在白蚁生殖品级和非生殖品级精子发生过程中的作用,运用免疫细胞化学方法对尖唇散白蚁Reticulitermes aculabialis繁殖蚁和工蚁精子发生中的AR-like的定位进行了研究。结果显示:在繁殖蚁和工蚁精子发生过程中都有AR-like免疫阳性细胞的分布,均分布于初级精母细胞的细胞质和细胞核中;与繁殖蚁相比,AR-like在工蚁中的表达较弱。结果提示,AR-like的表达与精子发生过程中初级精母细胞的减数分裂有关,雄激素及其受体通过调控精母细胞的第一次减数分裂来影响白蚁精子的发生。虽然工蚁的精巢发育受抑制,但其精子发生和维持具有与繁殖蚁相同的激素调节机制,能形成精子。本研究为工蚁性腺退化不育而根据群体的变化又可以发育为补充繁殖蚁这一特殊的生理功能提供了组织学依据。  相似文献

【目的】为了探讨圆唇散白蚁Reticulitermes labralis雌性工蚁向补充生殖蚁转化过程中的卵巢发育特征以及卵母细胞从滞育到恢复生长发育的起点。【方法】观察圆唇散白蚁雌性工蚁从3龄-6龄-转化成补充生殖蚁发育过程中的卵巢和卵母细胞动态变化;在若蚁向原始生殖蚁和补充生殖蚁转化的转录组中筛选出与卵母细胞生长期相关的基因表达,利用qRT-PCR方法检测这些基因在工蚁向生殖蚁转化发育过程中的表达水平。【结果】圆唇散白蚁工蚁从低龄向老龄发育过程中卵巢逐渐增大;前补充生殖蚁卵巢长度和宽度分别是工蚁的约2和3倍,而前补充生殖蚁转化为补充生殖蚁之后,卵巢没有显著增大。工蚁转化为前补充生殖蚁之后,卵母细胞大小(长径)和滤泡细胞层厚度仍然没有显著改变;前补充生殖蚁转化成补充生殖蚁之后,卵母细胞大小和滤泡细胞层厚度显著增加。调控卵母细胞生长期发育的6个基因cyclin-dependent kinase 1, cell division cycle protein 20, G2/mitotic-specific cyclin-B3, G2/mitotic-specific cyclin-A, aurora kinase A和serine/threonine-protein kinasepolo在补充生殖蚁中的表达水平极显著增加,分别是前补充生殖蚁的约34, 62, 91, 36, 57和106倍。工蚁转化为前补充生殖蚁之后,仅G2/mitotic-specific cyclin-B3, G2/mitotic-specific cyclin-A和aurora kinase A表达水平显著增高,分别是工蚁的约3, 3和2倍。【结论】圆唇散白蚁工蚁卵巢发育的停滞发生在老龄期,卵母细胞发育的恢复和减数分裂启动从工蚁转化为补充生殖蚁之后开始。  相似文献

为探讨类雄激素受体 (androgen receptor-like,AR-like)在白蚁卵子发生过程中的作用,采用免疫细胞化学方法对尖唇散白蚁Reticulitermes aculabialis雌性繁殖蚁和工蚁卵子发生中的类雄激素受体进行了定位检测。结果发现:繁殖蚁和工蚁卵巢中均有类雄激素受体免疫阳性反应。类雄激素受体在末龄若虫和成虫卵母细胞的分化期和生长期定位于卵母细胞质,而在成虫的卵黄形成期定位于滤泡细胞;在工蚁卵巢中,类雄激素受体定位于分化期和生长期的卵母细胞质,无卵黄形成期。结果提示类雄激素受体在白蚁的卵子发生过程中有重要作用: 工蚁的卵巢发育受抑制,其卵子发生相当于末龄若虫水平,但根据群体的变化又可以发育为补充繁殖蚁,有发育的潜能。  相似文献

为探讨凋亡调节因子Bcl-2和Bax蛋白对白蚁生殖蚁和工蚁性腺发育的影响, 揭示白蚁生殖品级与非生殖品级性腺发育的调节机理, 以尖唇散白蚁Reticulitermes aculabialis为研究对象, 运用免疫细胞化学定位方法对生殖蚁和工蚁精子发生过程中的Bcl-2和Bax蛋白表达进行了研究。结果显示: 生殖蚁和工蚁精子发生过程中从精原细胞至精子时期均有Bcl-2-like和Bax-like的阳性表达。生殖蚁的次级精母细胞、 精子细胞和精子中Bcl-2-like阳性表达率较高, 而在精原细胞和初级精母细胞中阳性率较低; 工蚁在次级精母细胞中最高, 在精原细胞和初级精母细胞中较低。除初级精母细胞期外, 工蚁生殖细胞其他发育阶段Bax-like阳性表达率均显著高于生殖蚁同一阶段生殖细胞。生殖蚁的生殖细胞在精原细胞、 初级精母细胞和次级精母细胞时期Bax-like阳性表达率较高, 发育至精子时期阳性率最低; 工蚁在次级精母细胞、 精子细胞和精子时期Bax-like表达率较高, 在初级精母细胞中表达率最低。在精子发生过程中, 生殖蚁生殖细胞Bax/Bcl表达量比值逐步下降; 而工蚁生殖细胞发育过程中Bax/Bcl表达量比值仅在次级精母细胞期下降, 其他发育时期均升高; 根据Bax/Bcl判断, 精原细胞和初级精母细胞是生殖蚁精子发生过程中主要的凋亡点, 而工蚁除了精原细胞和初级精母细胞外, 精子细胞和精子也是主要的凋亡目标。研究结果表明, 白蚁生殖细胞凋亡与其他动物一样受Bcl-2家族的调节, 在精子发生过程中Bcl-2-like和Bax-like表达具有动态变化规律, 正是这种变化调控生殖细胞在不同发育阶段的生或死; Bcl-2-like和Bax-like对生殖细胞凋亡调节不仅在精子发生中有非常重要的作用, 而且可能与工蚁品级的形成有关。  相似文献

【目的】研究圆唇散白蚁Reticulitermes labralis(Hsia)补充生殖蚁分化及发育过程中卵黄蛋白原(Vitellogenin,Vg)基因的表达、卵母细胞的发育以及子代品级分化。【方法】通过使用实时定量PCR(Real-time quantitative polymerase chain reaction)技术测定补充生殖蚁从分化到产卵的过程中Vg基因相对表达量的动态变化;采用组织染色的方法进行卵母细胞发育阶段的观察;对子代的数量及分化进行统计分析。【结果】补充生殖蚁发育过程中,Vg基因表达呈现先升高后降低的动态变化模式。在补充生殖蚁分化后的第10天开始形成具卵黄的卵母细胞,在20 d时具卵黄卵母细胞的数量达到最高值。第1个月至第3个月,补充生殖蚁的产卵数逐渐增加,分别为(2.45±1.43)、(7.68±2.53)和(12.10±7.09)粒。第3个月,巢内开始出现幼蚁,数量为(5.15±2.41)头;第10个月,巢群内有工蚁、前兵蚁和兵蚁的分化,分别为(17.03±2.28)、(1.45±0.31)和(0.79±0.18)头。【结论】在新建巢群中,若蚁的分化在工蚁和兵蚁分化之后。圆唇散白蚁若蚁在巢群需要的时候,可以在很短时间内转化为补充生殖蚁;Vg基因的表达水平与卵母细胞的卵黄形成相关,Vg基因表达量的增加启动了卵母细胞的卵黄摄取过程。虽然单个补充生殖蚁产卵量不如单个原始生殖蚁的多,但是在一个巢内补充生殖蚁总的产卵数目要远远超过原始生殖蚁,因为补充生殖蚁的数量比原始生殖蚁的数量多。因此补充生殖蚁对巢群稳定和发展具有重要作用。  相似文献

Abstract.  Termites (Isoptera) are eusocial insects and express polyphenism. Soldiers have specialized morphology for colony defense, but their feeding activity is dependent on other colony members. To determine differences in cellulose degradation between soldier and worker termites, enzymatic activity and cellulase gene expression, as well digestive tract histology, are examined in two phylogenetically distant species. In Hodotermopsis sjostesti (family Termopsidae) , endo-β-1,4-glucanase activity is identified in the salivary glands, whereas β-glucosidase activity is identified in salivary glands and hindgut. The relative expression levels of endo-β-1,4-glucanase genes in soldiers are significantly lower than in workers. Thin sections of salivary gland of workers and soldiers are different in H. sjostedti . In Nasutitermes takasagoensis (family Termitidae), the endo-β-1,4-glucanase activity is restricted to the midgut in four tested castes (i.e. three types of workers and soldier). Examination of activity per termite reveals the highest activity in minor workers and the lowest activity in major workers and soldiers. The β-glucosidase activity is also concentrated on the midgut in all four castes. The relative expression level of the endo-β-1,4-glucanase gene does not correspond with its activity in the midgut. In thin sections prepared from N. takasagoensis , the folds and pulvillus in the gizzards, and cuticle structure of soldiers are less developed compared with the other three worker castes. The differences in digestive system among termite castes in terms of caste development in each species are discussed.  相似文献

Abstract.  The relationship between scent composition and antennal sensitivity in different castes of Atta mexicana is investigated under laboratory conditions. Extracts of dead ants are analysed by gas chromatography-mass spectrometry to identify the compounds presumably responsible for the specific undertaking behaviour. Oleic acid is identified as one compound that triggers undertaking behaviour. To determine differences in odour reception between workers of different castes (i.e. foragers, undertakers and soldiers), further antennal sensitivity to task-related odours is tested using electroantennographic techniques. Soldiers are the most specialized caste because of their low response to all odours, except odourants related to alarm pheromones. The behavioural specialization of soldiers and their reduced behavioural repertoire are discussed.  相似文献

The bacterial microbiota from the whole gut of soldier and worker castes of the termite Reticulitermes grassei was isolated and studied. In addition, the 16S rDNA bacterial genes from gut DNA were PCR-amplified using Bacteria-selective primers, and the 16S rDNA amplicons subsequently cloned into Escherichia coli. Sequences of the cloned inserts were then used to determine closest relatives by comparison with published sequences and with sequences from our previous work. The clones were found to be affiliated with the phyla Spirochaetes, Proteobacteria, Firmicutes, Bacteroidetes, Actinobacteria, Synergistetes, Verrucomicrobia, and candidate phyla Termite Group 1 (TG1) and Termite Group 2 (TG2). No significant differences were observed with respect to the relative bacterial abundances between soldier and worker phylotypes. The phylotypes obtained in this study were compared with reported sequences from other termites, especially those of phylotypes related to Spirochaetes, Wolbachia (an Alphaproteobacteria), Actinobacteria, and TG1. Many of the clone phylotypes detected in soldiers grouped with those of workers. Moreover, clones CRgS91 (soldiers) and CRgW68 (workers), both affiliated with 'Endomicrobia', were the same phylotype. Soldiers and workers also seemed to have similar relative protist abundances. Heterotrophic, poly-β-hydroxyalkanoate-accumulating bacteria were isolated from the gut of soldiers and shown to be affiliated with Actinobacteria and Gammaproteobacteria. We noted that Wolbachia was detected in soldiers but not in workers. Overall, the maintenance by soldiers and workers of comparable axial and radial redox gradients in the gut is consistent with the similarities in the prokaryotes and protists comprising their microbiota.  相似文献

Trophallaxis and feeding relationships in the eastern subterranean termite, Reticulitermes flavipes (Kollar), were examined using a novel marking technique, rabbit IgG protein coupled with an enzyme linked immunosorbent assay (ELISA) to detect the marker. Transfer experiments in small dishes evaluated the trophallactic transfer of the marker from donor workers fed IgG-treated paper to recipient workers or larvae. Worker donors rapidly acquired the marker, and 100% of donors tested positive within 24 h. Trophallactic transfer from donors to recipients was relatively inefficient, and 51 +/- 2% of recipient workers and 31 +/- 2% of recipient larvae tested positive at 72 h. Based on the mean optical density counts, approximately 27% of marker ingested by the donors was passed on to the recipient workers in the first 24 h, 14% to recipient larvae, and 26% to recipient soldiers. The ability of soldiers to feed independently of workers was examined in dish assays. Soldiers showed no significant uptake of the marker when isolated from the workers, and uptake increased significantly when workers were present. The distribution of the marker was further studied in larger colony fragments composed of workers, soldiers, nymphs, and larvae. Marker acquisition by the different castes/developmental stages was highly variable, with workers and nymphs acquiring the marker at a faster rate than soldiers and larvae. The results of this study contribute to our understanding of the foraging ecology and social behavior in R. flavipes. In addition, they may help design improved control programs for subterranean termites based on baits.  相似文献

Abstract Termite workers and soldiers differ markedly in their morphology and behaviour. We sought evidence for genetic influences on caste determination in the giant northern termite, Mastotermes darwiniensis , by investigating if workers and soldiers from the same colony differed genetically. The genotypes of 795 termites from 11 distinct colonies were assayed at six polymorphic microsatellite loci. We found that the multilocus genotypes of workers and soldiers from 8 of the 11 colonies did not differ significantly. Thus, the majority of the data provided no evidence for a genetic association with caste in workers and soldiers of M. darwiniensis . However, the genotype frequencies of workers and soldiers from three colonies differed, suggesting that genotype is occasionally associated with caste in this species. The genetic differentiation of castes within these colonies could reflect differences in the propensities of termites with distinct genotypes to develop into particular castes and provide a selective advantage to colonies headed by multiple reproductives.  相似文献

Workers of Reticulitermes flavipes were isolated in groups of increasing numbers to determine the in vitro rates of juvenile hormone (JH) synthesis by individual pairs of corpora allata (CA) as other castes differentiated. Only neotenic reproductives developed in groups of 12. Mean JH synthesis rates increased after 5 weeks but only a few individuals had significantly higher rates, about 0.4 pmol/pair/h, which occurred at about 3 weeks before neotenics developed. Soldiers and neotenics developed in groups of 50. Mean rates increased to a peak at week 6 after isolation, but only a few individuals had rates approaching 1 pmol/pair/h, which occurred at the same time after isolation as the development of pre-soldiers. JH synthesis by CA of pharate pre-soldiers and soldiers was low compared to that of pharate workers and neotenics. CA of pre-soldiers attained a peak mean rate of JH synthesis of 0.9 pmol/pair/h at 6 days of age, whereas CA of soldiers attained only a peak mean rate of 0.3 pmol/pair/h. These measurements of JH synthesis by individual pairs of CA suggest that the few workers destined to become pre-soldiers have 2.5-fold higher JH synthesis than the few that would develop into neotenic reproductives, and show that a cycle of synthesis accompanies the development of pre-soldiers into soldiers.  相似文献

The social organization of termites, unlike that of other social insects, is characterized by a highly plastic caste system. With the exception of the alates, all other individuals in a colony remain at an immature stage of development. Workers in particular remain developmentally flexible; they can switch castes to become soldiers or neotenics. Juvenile hormone (JH) is known to play a key role in turning workers into soldiers. In this study, we analyzed differences in cuticular hydrocarbon (CHC) profiles among castes, paying particular attention to the transition of workers to soldiers, in the subterranean termite species Reticulitermes flavipes. CHCs have a fundamental function in social insects as they serve as cues in inter- and intraspecific recognition. We showed that (1) the CHC profiles of the different castes (workers, soldiers, nymphs and neotenics) are different and (2) when workers were experimentally exposed to a JH analog and thus induced to become soldiers, their CHC profiles were modified before and after the worker-presoldier molt and before and after the presoldier-soldier molt.  相似文献

While intercolonial and interspecific agonism has been broadly documented in termites, detailed studies of individual behavior and its differentiation among castes are scarce. We studied the responses of non-reproductive castes of Prorhinotermes canalifrons (pseudergates, presoldiers, soldiers of different age and artificial intercastes) to conspecific and heterospecific aliens. Each caste displayed a specific behavioral repertoire. Soldiers, but to a lesser extent pseudergates as well, are responsible for colony defense. The behavioral repertoire of the soldier appears on the second day after exuviation and becomes complete on the third day, well before its defensive secretion is present. The behavior of artificial intercastes induced by a JH analog is intermediate between that of pseudergates and soldiers. Reactions to heterospecific aliens were much quicker and more violent than reactions to conspecifics, which suggests that individuals experience difficulties in identifying conspecific aliens.  相似文献
