1. The nature and distribution of the electron transport system of Acetobacter suboxydans (ATCC 621) has been investigated, with particular reference to cytochrome o.

2. A highly active membrane-bound electron transport system has been demonstrated, and functional roles suggested for ubiquinone, two c-type cytochromes ( peaks at 549 and 553 nm at — 196°), and two b-type cytochromes ( peaks at 558 and 564 nm at — 196°).

3. Evidence is presented suggesting that both the b-type cytochromes may be terminal oxidases of the cytochrome o type, and that cytochrome o (558) has an O2 affinity approx. 10 times greater than cytochrome o (565), and a CO affinity only half as great.  相似文献

Low-temperature photodissociation spectra of membranes from the thermophile PS3 reveal cytochromes o and a3. The latter reacts with O2 at −103°C to give a light-insensitive compound(s), but the initial stages of O2 binding to cytochrome o could not be studied under these conditions. Photochemical action spectra identify cytochromes a3 and o, but not a CO-binding c-type cytochrome, as functional terminal oxidases in this bacterium.  相似文献

Two functional terminal oxidases, cytochrome aa3 and cytochrome o, are present in the cytochrome system in cyanide-sensitive trypanosomatids. The organisms examined included Crithidia fasciculata, Leptomonas sp., Blastocrithidia culicis, Herpetomonas muscarum, Leishmania tarentolae, Trypanosoma lewisi, Trypanosoma conorhini and Trypanosoma cruzi. A CO-binding protein with the spectral properties of cytochrome o has been solubilized and partially purified from C. fasciculata. In L. tarentolae, T. conorhini and C. fasciculata, 10–20% of the respiration is cyanide-resistant but is inhibited by salicylhydroxamic acid. The various possibilities of a branched electron transport system are examined and discussed.  相似文献

Extracellular oxidase of the white rot fungus Panus tigrinus earlier reported as laccase)contains copper but has no absorption spectrum typical of ‘blue’ oxidases. Thioglycolate and sodium azide inhibit the activity of this enzyme at concentrations 2.5–3 orders lower than those needed for fungal laccases. The oxidase of P. tigrinus oxidizes syringaldazine, coniferyl alcohol, ABTS, syringic acid, diaminobenzidine, guaiacol, catechol and vanillylacetone with different efficiencies. Oxygen consumption and no hydrogen peroxide formation were detected during substrate oxidation by P. tigrinus oxidase. It is proposed that P. tigrinus oxidase is a new ligninolytic enzyme.  相似文献

Polymerase chain reaction using degenerate primers was used to identify genes encoding proteins of the ATP-binding cassette superfamily in Aspergillus fumigatus and Aspergillus flavus. In A. fumigatus, two genes (AfuMDR1 and AfuMDR2) encoding proteins of the ATP-binding cassette superfamily were identified. One gene (AflMDR1) was isolated from A. flavus and is the apparent homologue to AfuMDR1. AfuMDR1and AflMDR1 encode proteins of molecular weights 148 000 and 143 000, respectively, each containing 12 putative transmembrane regions and two ATP-binding sites. These proteins are arranged in two homologous halves, each half consisting of a hydrophobic region (encoding six putative transmembrane domains) and an ATP-binding site. The AfuMDR1 and AflMDR1-encoded proteins bear a high degree of similarity to the Schizosaccharomyces pombe leptomycin B resistance protein and to human MDR1. The second gene identified in A. fumigatus, AfuMDR2, encodes a protein of molecular weight 85 000, containing four putative transmembrane domains and an ATP binding domain. The encoded protein is similar to those encoded by MDL1 and MDL2, two MDR-like genes of Saccharomyces cerevisiae. Expression of AFUMDR1 in S. cerevisiae conferred increased resistance to the antifungal agent cilofungin (LY121019), an echinocandin B analog.  相似文献

The essential oils of two endemic Centaurea species from Turkey, C. mucronifera and C. chrysantha, collected in the same habitat, have been studied. The main compounds of the former were germacrene D (29.3%), β-eudesmol (17.4%) and β-caryophyllene (7.3%), while in the latter germacrene D (27.4%), caryophyllene oxide (9.5%) and bicyclogermacrene (5.4%) were detected among its major constituents. The two species produced many similar compounds in their essential oils that could be justified by the similar ecological conditions of their habitat, but also many differences were found that could confirm their taxonomic separation.  相似文献

The two malonylated pigments, malonylmalvin and malvidin 3-malonylglucoside, were identified in petals of Lavatera maritima, which belongs to the Malvaceae, a family known to synthesise such pigments. Zwitterionic anthocyanins could not be detected in four other newly examined sources and common unacylated pigments were recorded. Thus, the fruits of the palms Euterpe edulis and Pinanga polymorpha have a mixture of cyanidin 3-glucoside and cyanidin 3-rutinoside, while the fruit of Cephaelis subcoriacea is coloured by cyanidin 3-glucoside. The latter pigment was also obtained from the reddish brown inflorescence of the parasitic plant Cynomorium coccineum.  相似文献

The deleterious effects of temperature-induced coral bleaching, a process by which corals lose their endosymbiotic algae (zooxanthellae; genus Symbiodinium) primarily at temperatures above mean yearly maximums, has not been well described for alcyonacean soft corals (Coelenterata, Octocorallia). The study of Symbiodinium cells lost from Sarcophyton ehrenbergi, Sinularia sp., and Xenia sp., which have not been compared in bleaching studies, indicate that the soft coral S. ehrenbergi released the greatest number of symbiont cells, however, it was less susceptible to heat stress surviving temperatures of 34 °C for >39 h. Sinularia sp. showed intermediate levels of bleaching tolerance to elevated temperatures, surviving prolonged exposures at 32 °C, but dying within 24 h at 34 °C. Xenia sp., however, was the most vulnerable to high heat stress maximally releasing Symbiodinium at temperatures ≤30 °C. This evidence indicates that Xenia sp. is even more susceptible to elevated temperatures than Acropora spp., previously reported to be the most vulnerable coral species to elevated temperature-induced bleaching.

Molecular analysis showed that the more resistant soft coral species (S. ehrenbergi) had the same type of Symbiodinium (clade C) as less resistant soft corals (Xenia sp.). In comparison to scleractinian corals collected from the same region that show similar bleaching resistance to high temperatures (e.g. Porities solida—more robust; Favites complanata—moderate resistance; Acropora hyacinthus—less robust), all scleractinian corals were symbiotic with Symbiodinium from clade C. A. hyacinthus, however, was found to possess multiple symbionts (clades B and C), and this represents a first report of Clade B in any Acropora species.  相似文献

The Teloschistaceae is a widespread family with considerable morphological and ecological heterogeneity across genera and species groups. In order to provide a comprehensive molecular phylogeny for this family, phylogenetic analyses were carried out on sequences from the nuclear ribosomal ITS region obtained from 114 individuals that represent virtually all main lineages of Teloschistaceae. Our study confirmed the polyphyly of Caloplaca, Fulgensia and Xanthoria, and revealed that Teloschistes is probably non-monophyletic. We also confirm here that species traditionally included in Caloplaca subgenus Gasparrinia do not form a monophyletic entity. Caloplaca aurantia, C. carphinea and C. saxicola s. str. groups were recovered as monophyletic. The subgenera Caloplaca and Pyrenodesmia were also polyphyletic. In the subgenus Caloplaca, the traditionally recognized C. cerina group was recovered as monophyletic. Because this study is based solely on ITS, to maximize taxon sampling, the inclusion of phylogenetic signal from ambiguously aligned regions in MP (recoded INAASE and arc characters) resulted in the most highly supported phylogenetic reconstruction, compared with Bayesian inference restricted to alignable sites.  相似文献

Leaf resins in the leguminous genera Hymenaea and Copaifera may play a role in restricting infection by the associated leaf fungus, Pestalotia. We tested this hypothesis by assessing growth of the geographically widespread Pestalotia subcuticularis presented with different compositions of resins. Leaf resins of Hymenaea and Copaifera are composed of the same suite of sesquiterpene hydrocarbons, but these vary quantitatively to form discrete compositional patterns among individuals and populations. Leaf resins may also contain sesquiterpene oxides; caryophyllene oxide, which may have been formed from the caryophyllene precursor common in these resins, inhibited fungal growth in vitro.  相似文献

Previous reports on the alkaloids of Z. leprieurii, Z. lemairei and Z. rubescens are reviewed. The alkaloids from nine specimens collected in Ghana have been examined chromatographically and the implications of the findings discussed, with special reference to the distinction between Z. leprieurii and Z. rubescens. In addition three further compounds, dihydrochelerythrine, lupeol and sesamin are reported for the first time from Z. leprieurii. The overall value of alkaloids as systematic markers in Zanthoxylum in Africa is briefly discussed.  相似文献

Trypsin inhibitors have been shown to be present in Vigna unguiculata (a variety of bean found in Nigeria). Four inhibitors were isolated by gel filtration and ion exchange chromatography. The most active of the inhibitors was shown to be electrophoretically homogeneous and to have a molecular weight of 20 000 estimated by molecular sieve chromatography. It was found to have a maximum activity at pH 8.5 which was almost completely lost after boiling for 25 min at 100°C.  相似文献

The objective of this work was to study the partial consumption of malic acid in a semi-synthetic medium by Schizosaccharomyces yeast under wine-making conditions. The aim was to control the extent of deacidification by stopping the activity of Schizosaccharomyces by the addition of Saccharomyces yeast at different delayed intervals of time. This was thought possible because the Schizosaccharomyces yeasts are much more slower than the Saccharomyces yeasts and may be inhibited by the latter. In fact, the opposite was observed. Schizosaccharomyces exhibited a amensal effect against Saccharomyces: the Saccharomyces growth was inhibited by Schizosaccharomyces, this inhibition being proportional to the Schizosaccharomyces concentration, reversible, and sensitive to temperature. Such interactions similar to a killer effect have not been described before. At the same time a competition towards sugar was noted, being negative for Schizosaccharomyces since these yeasts have slower metabolic rates. Nevertheless, total demalication was nearly always possible because sufficient concentrations of Schizosaccharomyces were always reached. The delayed inoculation of Saccharomyces after Schizosaccharomyces was not a good solution to obtain partial deacidification.  相似文献

The granular structure and gelatinisation properties of starches from a range of pea seed mutants were studied. Genes which affect the supply of substrate during starch synthesis (rb, rug3, rug4) affected the total crystallinity and possibly increased the content of A polymorphs in the starch. Conversely, genes directly affecting the synthesis of starch polymers (r, rug5, lam) increased the content of B polymorphs, but had a minimal effect on total crystallinity. During gelatinisation, starches from the rb, rug3, rug4 and lam mutants had narrow endothermic peaks which were similar to starch from the wild-type, although all the starches had different peak temperatures and enthalpy changes. Starches from r and rug5 mutants were very different to all other starches, having a very wide transition during gelatinisation. In addition, the amylopectin in starch from these mutants had altered chain lengths for those parts of the polymer which form the ordered structures in the granule.  相似文献

Experiments were performed to investigate the effects of 3 polycyclic aromatic hydrocarbons, benz[a]anthracene, dibenz[a,c]anthracene and dibenz[a,h]anthracene and K-regio epoxides and some of their related dihydrodiols on the chromosomes of Chinese hamster ovary cells in vitro. Of the 3 hydrocarbons only benz[a]anthracene showed any activity in inducing sister-chromatid exchanges. The K-region epoxide and the 3,4-dihydrodiol have been found to be more active than the corresponding K-region or the other non K-region dihydrodiols derived from benz[a]anthracene. Athough dibenz[a,c]anthracene was almost inactive, the K-region 5,6-epoxide and all 3 possible dihydrodiols, the 1,2-, 3,4- and 10,11-diols were active in inducing increased numbers of sister-chromatid exchanges in the chromosomes of these cells. The 3,4-dihydrodiol of dibenz[a,h]anthrecene was also active in inducing sister-chromatid exchanges whereas the 1,2- and 5,6-dihydrodiols were only weakly active. This study provides some support for the suggestiion that the activation of these 3 hydrocarbons proceeds by the metabolic conversion of non K-region dihydrodiols into vicinal diol-epoxides.  相似文献

Pure reaction center preparations from the thermophilic species Chromatium tepidum have been obtained by lauryldimethylamine N-oxide treatment of chromatophores. The light-induced difference spectrum in presence of 10 mM sodium ascorbate revealed the presence of two high-potential cytochrome c hemes (-band, 555 nm; γ-band, 422 nm). The dithionite-minus-oxidized difference spectrum in the -band suggests the presence of additional hemes of low potential. These hemes are associated with a single polypeptide (Mr = 36 000). The reaction center pigments, probably four bacteriochorophyll a and two bacteriopheophytin a molecules, are associated with three polypeptides of apparent molecular weights equal to 33 000, 30 000 and 22 000. A carotenoid molecule is also bound to the reaction center. The three main absorption bands of this molecule are located at 480, 510 and 530 nm at liquid helium temperature. Photochemical activity is found to be stable, even after heating for 10 min at temperatures higher than 60 °C in intact chromatophore membranes. On the other hand, isolated reaction centers or chromatophores treated with 1% lauryldimethylamine N-oxide are fully inactivated after heating at temperatures higher than 50 °C. From these results, we propose that lipid-protein interactions are of prime importance in the thermal stabilization of Chromatium tepidum reaction centers.  相似文献

The flavonoid profiles of Astilbe (four taxa studied) and Rodgersia (two taxa studied) are based on simple flavonol glycosides. Astilbe has 3-O-mono-, 3-O-di-, and 3-O-triglycosides of kaempferol, quercetin, and myricetin, while Rodgersia has only mono- and diglycosides of kaempferol and quercetin. Astilbe×arendsii was also shown to accumulate dihydrochalcone glycosides. The flavonoid profile of Rodgersia is the simplest recorded so far in the herbaceous Saxifragaceae. The flavonoids of two species of Aruncus were shown to be based upon kaempferol and quercetin 3-O-mono- and 3-O-diglycosides. One of the species also exhibited an eriodictyol glycoside. The triglycoside differences were not considered important, but the differences in myricetin occurrences were taken as evidence against derivation of Saxifragaceae from an Aruncus-like ancestor. Should such an event be proposed, however, serious consideration would have to be given to the current pattern of myricetin occurrence in the two families.  相似文献

We examined the effects of 48 compounds isolated from Ferula pallida, F. penninervis, Inula macrophylla, Prangos pabularia, P. tschimganica and Rheum maximowiczii collected in Uzbekistan on ADP/Fe2+-induced lipid peroxidation of egg yolk phosphatidylcholine liposomes. Of those compounds, 23 inhibited ADP/Fe2+-induced lipid peroxidation and nine showed especially strong inhibition of lipid peroxidation. Most compounds that inhibited peroxidation scavenged the 1,1′-diphenyl-2-picrylhydrazyl (DPPH) radical, indicating that the inhibition was due to radical scavenging. However, some compounds did not scavenge DPPH but inhibited lipid peroxidation significantly, suggesting that their inhibitory effect was not due to radical scavenging but to some other mechanism, such as prevention of Fe2+ function. Thus, we found various new antioxidants, some of which had a unique mechanism of action, in Ferula, Inula, Prangos and Rheum plants collected in Uzbekistan as seeds used in medicine.  相似文献

In order to undertake a comparative analysis of carbon catabolite repression in two Aspergillus species, the creA gene has been isolated from A. niger by cross hybridization, using the cloned A. nidulans gene. The A. niger gene has been shown to be functional in A. nidulans by heterologous complementation of the creA204 mutation of A. nidulans. Overall, the genes show 90% sequence similarity (82% identity) at the amino acid (aa) level. There were some striking similarities between the aa sequences encoded by the two fungal creA genes and two genes involved in carbon catabolite repression in Saccharomyces cerevisiae. The zinc-finger regions showed 96% similarity (84% identity) with the zinc-finger region of the MIG1 gene of S. cerevisiae. The CREA protein contains a stretch of 42 aa that is identical in A. niger and A. nidulans, and these show 81% similarity (33% identity) with a region of the S. cerevisiae RGR1 gene.  相似文献

Three liverworts, Trichocolea tomentella, Neotrichocolea bissethii and Trichocoleopsis sacculata belonging to Jungermanniales were chemically investigated. Isoprenyl benzoates are the important chemical markers of Trichocolea tomentella. Neotrichocolea bissethii elaborates sesquiterpenes as the major components. The sacculatane-type diterpenes and pinguisane-type sesquiterpenes are the significant chemosystematic markers of Trichocoleopsis sacculata. These three species are chemically quite different. Trichocoleopsis sacculata is chemically rather close to Porella species. The present chemical results support the recent morphological classification of the above three species proposed by Schuster. Some species of Jungermanniales are chemically identical to those of Metzgeriales and these results also support the phylogenetic classification in which the two orders have been united within the subclass Jungermanniae.  相似文献
