绿僵菌对小麦纹枯病菌的抑制作用研究   总被引:1,自引:0,他引:1  
在实验室条件下,研究了金龟子绿僵菌(Metarhizium anisopliae)对小麦纹枯病菌(Rhizoctonia cerealis)的拮抗作用及其机理。结果表明,金龟子绿僵菌与小麦纹枯病菌对峙培养以及在培养基中加入金龟子绿僵菌孢子悬浮液,对小麦纹枯病菌菌丝生长均有较好的抑制作用。测定了培养不同天数的金龟子绿僵菌Ma55发酵液对小麦纹枯病菌菌丝生长、菌核产生量及菌核萌发率的影响。结果表明,液体振荡培养25 d的金龟子绿僵菌Ma55发酵液对小麦纹枯病菌的菌丝生长、菌核产生量及菌核萌发具有显著的抑制作用,且Ma55发酵液中的抑菌活性物质具有较好的热稳定性。在光学显微镜下,未观察到Ma55对小麦纹枯病菌的重寄生现象,但发现金龟子绿僵菌与小麦纹枯病菌对峙培养处小麦纹枯病菌营养菌丝的细胞质变稀薄、菌丝部分消解或断裂。上述结果显示,金龟子绿僵菌对小麦纹枯病菌的拮抗机制主要是营养竞争、空间竞争及抗生作用等。  相似文献

分析比较了 33种不同来源的氨基酸产品红外图谱的差异 ,其中丝氨酸、门冬氨酸、醋酸赖氨酸、谷氨酸 (白色结晶性粉末 )、苏氨酸、缬氨酸、丙氨酸、亮氨酸、脯氨酸、盐酸组氨酸、盐酸精氨酸、酪氨酸、胱氨酸等 13种与标准图谱完全一致 ;甲硫氨酸、盐酸赖氨酸、甘氨酸、谷氨酸 (白色结晶 )等 4种与标准图谱不一致 ,其原因是 :甘氨酸和谷氨酸由晶型不同造成 ,甲硫氨酸因旋光性不同而造成 ,盐酸赖氨酸与相应的生化试剂图谱一致。  相似文献

运用生态位原理与方法,研究了水稻纹枯病菌的矿质元素营养生态位.结果表明,在分蘖盛期、孕穗期、抽穗期和蜡熟期,水稻纹枯病菌矿质营养生态位宽度指数分别为0.2710、0.3865、0.4252和0.4817,即随着水稻的生长而逐渐增大,但各生育期的生态位宽度总体上仍偏小.表明在水稻生长的各时期水稻纹枯病菌只占有了矿质营养类型中的较少部分.水稻纹枯病菌总是优先占有低Mg、低Zn、低Si的营养位,说明Mg、Zn、Si的含量与水稻对纹枯病的抗性紧密相关.  相似文献

八种氨基酸对水稻硝酸盐吸收的影响   总被引:2,自引:0,他引:2  
目的:在较低NO3-浓度下,研究不同氨基酸对水稻硝酸盐吸收的调控作用.方法:使用非侵害性硝酸盐检测系统测定了谷氨酸、天门冬氨酸、精氨酸、赖氨酸、脯氨酸、苯丙氨酸、缬氨酸和丝氨酸8种氨基酸对水稻硝酸盐吸收的动力学影响.结果:加入谷氨酸、天门冬氨酸后对水稻硝酸盐的吸收有短暂的促进作用,硝酸盐净吸收率分别上升258±123%和217±34%;另外,加入赖氨酸和精氨酸以后发现水稻硝酸盐的吸收率降低-63±34%和-64±27%,反应延迟时间为30min以上,而其他4种氨基酸并没有产生明显的影响.结论:氨基酸不是直接调控水稻高亲和力NO3-转运系统,而可能是对NO3-同化过程进行调控.  相似文献

[目的]研究大黄鱼共生菌Bacillus coagulans LL1103的次生代谢产物及其抑菌活性。[方法]利用色谱层析技术对B.coagulans LL1103发酵液乙酸乙酯和正丁醇提取物进行分离纯化;运用波谱技术鉴定化合物的结构;采用微量肉汤稀释法对化合物进行抗菌活性测定。[结果]从大黄鱼内生菌B.coagulans LL1103发酵液中分离并鉴定9个环二肽类化合物,分别为:(1)环(4-羟基-脯氨酸-亮氨酸)、(2)环(异亮氨酸-丙氨酸)、(3)环(脯氨酸-缬氨酸)、(4)环(脯氨酸-苯丙氨酸)、(5)环(脯氨酸-亮氨酸)、(6)环(甘氨酸-丙氨酸)、(7)环(脯氨酸-甘氨酸)、(8)环(脯氨酸-丙氨酸)和(9)环(络氨酸-甘氨酸)。活性评价显示化合物3、7和9对大肠埃希菌(Escherichia coli)具有较强抑制作用,最低抑菌浓度(minimal inhibitory concentration,MIC)分别为8.0、4.0和16.0μg/mL。[结论]从大黄鱼共生菌B.coagulans LL1103的发酵液中分离得到3个对大肠埃希菌(E.coli)具有较好抑制活性的化合物。  相似文献

根据胚乳蛋白在不同溶荆中的溶解能力,可将小 麦胚乳蛋白分为四个组份臼”,其中可溶于70%酒精 的贮藏蛋白,称为小麦醇溶蛋白(gliadin),醇溶蛋白只 存在于禾本科植物种子中,它们构成了大部分具有重 要经济价值的谷物籽粒的主要贮藏蛋白〔,.〕。在小麦 中,它的含量约占籽粒总蛋白的一半。醇溶蛋白位于 胚乳的蛋白体内,主要存在于亚糊粉层,其富含疏水性 氨基酸,如亮氨酸, 氨酸、脯氨酸和谷酞胺等,缺乏亲 水性氨基酸,如赖氨酸、色氨酸和蛋氨酸等。氨基酸谱 的不平衡是使醇溶蛋白含量较高的小麦在实际应用上 受限制的一个重要原因。  相似文献

从抗纹枯病小麦品种CI12633中克隆出一个小麦防御素基因TaPDF35,并对其表达特性及功能进行了分析。TaPDF35基因包含一个长为249 bp的开放阅读框(ORF,open reading frame),编码82个氨基酸组成的多肽TaPDF35。预测分析表明,TaPDF35能形成由1个α螺旋和3股反向平行的β-折叠片组成的αβ(CSαβ)基序,αβ(CSαβ)基序由8个半胱氨酸形成的4个二硫键固定。TaPDF35的N端有一段27个氨基酸的信号肽。表达分析结果表明,TaPDF35基因在抗纹枯病小麦品种CI12633和山红麦中的表达量显著高于在感纹枯病小麦品种扬麦158和温麦6号中的表达量;该基因在小麦的叶鞘和茎中均有表达,且受纹枯病原菌诱导而上调表达。利用大麦条形花叶病毒(BSMV,barley stripe mosaic virus)诱导的基因沉默技术(VIGS,virus-induced gene silencing)降低抗纹枯病小麦品种CI12633中TaPDF35基因的转录水平,再接种纹枯病原菌进行纹枯病抗性鉴定。结果显示,与接种BSMV:GFP的CI12633对照植株相比,TaPDF35表达水平降低的CI12633植株对纹枯病的抗性显著降低,表明TaPDF35表达是小麦防御纹枯病反应所需的。  相似文献

Aspergillus flavus and Penicillium italicum were cultivated on Dox medium amended with different concentrations of a vanadium salt; vanadium pentoxide (V2O5). The fungal isolates were found to absorb considerable quantities of vanadium. Fungal contents of carbohydrates and proteins were decreased by the presence of vanadium in the growth environment. Vanadium was incorporated into several types of low and high molecular weight protein(s) and peptides. In case of P. italicum, lysine, tyrosine, glycine, methionine, valine, alanine, proline, glutamic acid and histidine were detected in the fungal cell free extract. While in case of A. flavus, the following amino acids were detected: cystine, methionine, leucine, phenylalanine, glycine, alanine, tyrosine, lysine, histidine, arginine and valine.  相似文献

Amino acid synthesis from glucose-U-14C was investigated in 2 day post-emergent and pregnant females of Glossina morsitans. This insect can synthesize alanine, aspartic acid, cystine, glutamic acid, glycine, proline, and serine from glucose. Arginine, histidine, hydroxyproline, isoleucine, leucine, lysine, methionine, phenylalanine, taurine, threonine, and valine showed no radioactivity and hence may be classified as nutritionally indispensable amino acids. Although tyrosine and hydroxyproline were not synthesized from glucose, they are at least partially dispensable nutrients for this insect because their synthesis from phenylalanine has been demonstrated. After the labelled glucose injection the highest radioactivity was recovered in the proline fraction. This is probably related to its rôle as an important energy reserve for flight. The radioactive amino acids recovered from females and from their offspring following glucose-U-14C injection were similar to those recovered from younger females. Radioactivity was also detected in the expired CO2 and the excreta. The amino acids alanine, arginine, cystine, glycine, histidine, leucine/isoleucine, lysine, methionine, proline, and valine were identified in the excreta, of which arginine and histidine were in the largest amounts. Only excreted alanine, glycine, and proline showed radioactivity.  相似文献

Aphis fabae was reared on synthetic diets omitting individual amino acids and amino acid groups. Methionine and histidine deletions induced apterae formation and prevented the aphids from reaching the adult stage, whereas arginine, leucine, lysine, and proline deletions induced alatae formation. The omission of alanine, cysteine, phenylalanine, proline, serine, and tyrosine reduced the size attained by adult alatae and together with methionine and histidine, these amino acids are considered to be essential for normal growth by A. fabae. A delay in parturition occurred in aphids reared on certain deficient diets and there is an association between the size attained by alatae and the delay in parturition.  相似文献

The skeletal muscle of Ophicephalus punctatus contains nine essential free amino acids, arginine, histidine, isoleucine, leucine, methionine, phenylalanine, threonine, valine and lysine, and eight non-essential amino acids, alanine, aspartic acid, cystine, glutamic acid, glycine, tyrosine, proline and serine. Histidine and lysine dominated the free amino acids pool. Seasonal variation was detected in the levels of histidine, arginine, leucine, phenylalanine, glycine, cystine and serine with highest values occurring in April and again in November. Changes were also detected in the concentrations of certain amino acids as the fish grew in size. Levels of free amino acids did not significantly differ between sexes. This medium (medium B4) contains sodium lactate as a partial carbon source, proline as both a partial carbon source and a partial nitrogen source, aspartate as a partial nitrogen source, and the growth factors arginine, glycine, and methionine. Either aspartate, glutamate, or proline could serve as sole nitrogen source, but growth occurred at a significantly better rate if proline was present together with either aspartate or glutamate, or with both aspartate and glutamate. With the exception of strain ATCC 23246, all the strains had an absolute requirement for arginine and either a partial or absolute requirement for glycine. The concentration of glycine required for optimal growth was found to be relatively high for an amino acid growth factor. Heart infusion broth was found to be growth inhibitory for spontaneous mutants of one strain able to grow in the absence of arginine, and such mutants reverted readily to arginine dependence accompanied by the ability to grow faster on the complex medium. Growth rates in the defined medium B4 were enhanced by the simultaneous addition of asparagine, glutamate, glutamine, leucine, lysine, histidine, and phenylalanine.  相似文献

Rats fasted overnight were allowed to consume single meals containing 0, 18, or 40% protein or continued to fast; after 2 h, brains and sera were taken and assayed for various amino acids. In general, serum levels of most amino acids were reduced by the 0% protein meal and elevated by the high-protein meal when compared with those associated with fasting conditions. Exceptions were those not diminished by the 0% protein meal (tryptophan, methionine, proline) and those increased (alanine) or decreased (glycine) by all of the test meals. Amino acids exhibiting the broadest normal ranges (estimated by comparing their serum levels after 40% protein with those after 0% protein) were tyrosine, leucine, valine, isoleucine, and proline; serum lysine and histidine, two basic amino acids, also varied more than threefold. Brain levels of lysine, histidine, and some of the large neutral amino acids (LNAAs) also exhibited clear relationships to the protein content of the test meal: those of valine, leucine, and isoleucine were depressed by the 0% protein but increased (compared with 0% protein) when protein was added to the meal: brain tyrosine was increased by all of the test meals in proportion to their protein contents; tryptophan, phenylalanine, and glutamate were increased after the 0% protein meal but not by protein-containing meals; brain lysine, histidine, and methionine were increased after the high-protein meal, and brain alanine was increased slightly by all of the meals.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The intracellular pool of free amino of Streptococcus lactis--lysine producer contains a good number of amino acids when cultivated on the corn medium. Glutamic acid, proline, alanine, lysine, leucine, histidine and arginine are in predominance. An almost complete amino acid pool develops at an early exponential phase of Str. lactis growth under stationary cultivation conditions. The content of free amino acids increases 4-fold during the transition from the early exponential phase to the stationary phases under submerged cultivation conditions. This can be attributed to a more intensive amino acid exchange during the medium stirring than during stationary cultivation.  相似文献

The effect of the amino acid concentration on sporulation of five mutants ofBacillus subtilis requiring histidine, isoleucine, leucine, lysine and methionine for growth was studied. Low amino acid concentrations, permitting about 10–20% of maximum growth, inhibit the ability of the bacteria to sporulate on completing vegetative growth. In medium concentrations (from 25% of maximum growth), leucine, isoleucine, histidine and methionine induce sporulation, while the concentration of the other nutrients in the medium is still adequate for further vegetative growth. The percentage of sporulation depends on the amino acid concentration and is characteristic for each amino acid. The lysine mutant strain 1399 was not capable of sporulation unless cultivated in the presence of the lysine concentration optimal for growth. The effect of amino acid deficiency depends on the cultivation method.  相似文献
