A domain of epiglucan was synthesized by beta-glucosidases. Two beta-glucosidases, an extracellular beta-glucosidase derived from Sclerotinia sclerotiorum grown on xylose, and a commercial lyophilized preparation of beta-glucosidase from Aspergillus niger, were used to synthesize gluco-oligosaccharides from cellobiose and, specially, beta-(1-6) branched beta-(1-3) gluco-oligosaccharides, corresponding to the structure of epiglucan. Gentiobiose, cellotriose, cellotetraose, beta-Glc-(1-3)-beta-Glc-(1-4)-Glc, beta-Glc-(1-6)-beta-Glc-(1-4)-Glc and beta-Glc-(1-6)-beta-Glc-(1-3)-Glc were synthesized from cellobiose by both enzymes. The latter compound was preferentially synthesized by the beta-glycosidase from Sclerotinia sclerotiorum. Under the best conditions, only 7 g l(-1) of beta-Glc-(1-6)-beta-Glc-(1-3)-Glc was synthesized by the beta-glycosidase from Aspergillus niger compared to 20 g l(-1) synthesized with beta-glycosidase from Sclerotinia sclerotiorum.  相似文献

The presynaptic protein RIM1α mediates multiple forms of presynaptic plasticity at both excitatory and inhibitory synapses. Previous studies of mice lacking RIM1α (RIM1α(-/-) throughout the brain showed that deletion of RIM1α results in multiple behavioral abnormalities. In an effort to begin to delineate the brain regions in which RIM1 deletion mediates these abnormal behaviors, we used conditional (floxed) RIM1 knockout mice (fRIM1). By crossing these fRIM1 mice to previously characterized transgenic cre lines, we aimed to delete RIM1 selectively in the dentate gyrus (DG), using a specific preproopiomelanocortin promoter driving cre recombinase (POMC-cre) line , and in pyramidal neurons of the CA3 region of hippocampus, using the kainate receptor subunit 1 promoter driving cre recombinase (KA-cre). Neither of these cre driver lines was uniquely selective to the targeted regions. In spite of this, we were able to reproduce a subset of the global RIM1α(-/-) behavioral abnormalities, thereby narrowing the brain regions in which loss of RIM1 is sufficient to produce these behavioral differences. Most interestingly, hypersensitivity to the pyschotomimetic MK-801 was shown in mice lacking RIM1 selectively in the DG, arcuate nucleus of the hypothalamus and select cerebellar neurons, implicating novel brain regions and neuronal subtypes in this behavior.  相似文献

1-(1′-2′ S-Nornicotino)-1-deoxy-β-d-fructofuranose was first isolated from flue-cured leaves of Cherry Red tobacco, Nicotiana tabacum, cv. Bright Yellow. Its structure was established spectrometrically and synthetically. This substance was shown to be formed from nornicotine during flue-curing. Its smoking effect was mild.  相似文献

Acyclic diastereoisomers, namely, 1-(N-acetyl-α-methylbenzylamino)-1-deoxyalditol acetates are readily obtained by reductive amination of aldoses with chiral α-methylbenzylamine (MBA) in the presence of sodium cyanoborohydride, and may be separated by reversed-phase 1.c. and, more effectively, by adsorption 1.c. According to this procedure, enantiomers of the common, neutral aldoses are resolved. In adsorption 1.c., l-l* [defined as an adduct of an l-aldose and l-(-)-MBA] is eluted before d-l* for erythrose, xylose, ribose, glucose, 4-O-methylglucose, galactose, and fucose, and the elution order is the reverse for arabinose, lyxose, mannose, rhamnose, and glyceraldehyde. This behavior is probably related to the configuration of C-2 of the aldoses.  相似文献

The p-nitrophenyl and p-aminophenyl 1-thio-α- and -β-d-idopyranosides were synthesized for use in structure-activity studies of glycosidases. Zinc chloride-catalyzed fusion of α-d-idopyranose pentaacetate with p-nitrobenzenethiol gave p-nitrophenyl 2,3,4,6-tetra-O-acetyl-1-thio-α-d-idopyranoside as an amorphous glass in 67% yield, and the crystalline β anomer in 13% yield. Deacetylation with catalytic amounts of sodium methoxide in methanol, followed by hydrogenation under pressure over palladium-on-barium sulfate catalyst, afforded p-aminophenyl 1-thio-α- and -β-d-idopyranosides.  相似文献

GlcNAc1-2Man and GlcNAc1-6Man were synthesized using the reverse hydrolysis activity of -N-acetylglucosaminidase from both jack beans and Bacillus circulans. In turn, Gal1-4GlcNAc1-2Man and Gal1-4GlcNAc1-6Man were synthesized regioselectively using the transglycosylation activity of -galactosidase from Diplococcus pneumoniae and B. circulans, respectively. These di- and trisaccharides are important components of complex type sugar chains and will be used as intermediates in our synthetic studies. Six new products of oxidation of indolyl-3-acetic add catalyzed by horseradish peroxidase were isolated, along with four known ones, 3-hydroxymethyloxindole (1), 3-methyleneoxindole (2), indolyl-3-aldehyde (4), and 3,3-diindolylmethane (10). Based on spectroscopic and chemical evidence, the new products were identified as 3-acetoxyindole (3), 3-(indol-3-ylmethyl)oxindole (6), 3-[(2-mdol-3-ylmethyl)indol-3-ylmethyl]oxindole (9), the 3-hydroxymethyl compounds of 6 and 9 (5 and 7), and 2-(indol-3-ylmethyl)indolyl-3-acetic acid (8), respectively.  相似文献

The SAR of a series of 1-amino-3-(1H-indol-1-yl)-3-phenylpropan-2-ols as monoamine reuptake inhibitors, with a goal to improve both potency toward inhibiting the norepinephrine transporter and selectivity over the serotonin transporter, is reported. The effect of specific substitution on both the 3-phenyl group and the indole moiety were explored. This study led to the discovery of compound 20 which inhibited the norepinephrine transporter with an IC50 value of 4 nM while exhibiting 86-fold selectivity over the serotonin transporter.  相似文献

Abstract

ROESY and NOESY NMR spectroscopic analyses of the ribofuranosyl (1a), 2′-deoxyribofuranosyl (1b), and arabinofuranosyl (1c) derivatives of 6-formyluracil in (CD3)2SO and D2O solutions have established that each exclusive 7,05′-cyclic hemiacetal diastereomer of 1a,b and the major 7,O2′-cyclic hemiacetal diastereomer of 1c possess the 7R configuration. In addition, (7R)-1c has been shown to be thermodynamically more stable than (7S)-1c, contrary to our previous indication. A new, higher yielding synthetic route to 1a has been developed, 1b has been obtained for the first time in crystalline form, the route to 1c has been modified to better accommodate large scale preparations, and a new, fourth member of this class, 6-formylthymidine (1d), has been synthesized and its solution structures in (CD3)2SO, D2O, and CD3OD have been determined. Antitumor and antiviral evaluations of 1a-c have revealed no significant levels of activity.  相似文献

Cytoplasmic membranes from mycelium or protoplasts of Saprolegnia monoica (a cellulosic cell-wall fungus) were separated by continuous sucrose-density-gradient centrifugation. Glucan synthases assayed at low (micromolar uridine 5-diphosphate (UDP) glucose for -1-4-glucan synthase) and high (millimolar UDP glucose for -1-3-glucan synthase) substrate concentrations were associated with membranes exhibiting vanadate-sensitive, oligomycin-insensitive ATPase and equilibrating at density 1.16 g cm-3. Synthase activities were also bound to membranes of lower density (1.10 and 1.145 g cm-3). Plasma membranes were stabilized by coating protoplasts with concanavalin A. After lysis of the protoplasts, plasma membranes recovered by low centrifugal forces were isolated in continuous isopycinic gradients. Both synthase activities peaked with [3H]concanavalin A and Na-vanadate ATPase indicating that the synthetases are located at the plasma membrane. Alzheimer's disease (AD) is the most prevalent form of neurodegenerative disorders even so the exact pathomechanism is still unclear. Recently, it is widely accepted that amyloid-beta peptide (Aβ) toxicity is positively linked to Aβ oligomers, which may be responsible for the initiation of AD. For this reason, AD research requires well defined aggregation state and structure of Aβ. Precursor peptide 'iso-Aβ1-42' makes it possible to use Aβ1-42 with well- defined aggregation state for in vitro and in vivo experiments. The aim of this study was to identify protein expression changes from differentiated SH-SY5Y neuroblastoma cells after treatment with oligomeric Aβ1-42 prepared in situ from 'iso-Aβ1-42'. In our experiment, a cell viability assay revealed a strong and time-dependent toxic effect of oligomeric Aβ1-42 which was supported by dramatic morphological changes. Our proteomics study also revealed numerous significant protein expression changes (22 proteins down- and 25 proteins up-regulated) after comparison of the untreated and Aβ1-42-treated cell lysates by two-dimensional electrophoresis. From the functional classification of the identified proteins, we found deregulations of proteins involved in metabolic processes, cytoskeleton organisation and protein biosynthesis and a huge number of up-regulated stress proteins displayed oligomeric Aβ1-42-induced cell stress.  相似文献

Medium chain β-1-alkylglycosides show interesting mild detergent properties. Therefore, their synthesis and purification have been investigated and improved so as to permit preparation of 50–100 g amounts. Preparatory methods are presented for the already known compounds β-1-octyl-, β-1-nonyl and β-1-decyl-glucose and for the new compounds β-1-undecylglucose and β-1-dodecylmaltose. Some relevant properties such as melting point, optical rotation, critical micelle concentration and NMR-spectra have been determined. They illustrate the suitability of this class of detergents for membrane research.  相似文献

It has been found that aqueous–ethanol extracts of brown seaweeds contain substances inhibiting endo-13--D-glucanases, the digestive enzymes of marine mollusks. The inhibitors were detected in 14 of 21 brown seaweeds investigated. An irreversible protein inhibitor possessing high specificity toward endo-13--D-glucanases of marine mollusks was isolated from the brown seaweed Laminaria cichorioides. As determined by gel filtration, the molecular mass of the inhibitor is 46 kD. The value of [I]50 (10–8 M) for the inhibitor is comparable with the corresponding value for natural inhibitors of amylases from terrestrial plants. The results of chemical modification indicated that tryptophan, glutamic acid, aspartic acid, histidine, and probably tyrosine residues are important for the interaction of the inhibitor with the enzyme.(subject: to Zvyagintseva)  相似文献

A -1-3-glucanase from O. xanthineolytica was purified from a fermentation broth, in two chromatographic steps using information obtained from electrophoretic titration curves. Signal transduction and cell responses after stimulation with heregulin β-1 (HRG) are examined in HCC2998 and MKN45-1 cells, which have been used for a model system to study the formation of signet ring carcinomas, one of poorly differentiated adenocarcinomas. HRG stimulation causes rounding of the cells, responding to HRG. The adherens junction, which is present in the control cells, is disrupted and cell-cell interaction is lost after stimulation. Inhibition of phosphatidylinositol (PI)-3 kinase or p38 MAP kinase blocked this reaction, which indicates that the PI-3 kinase-p38 MAP kinase pathway is required for this reaction. Inhibition of the p38 MAP kinase pathway resulted in immediate restoration of cell-cell interaction. This result indicates that signaling for adherent molecules is strictly regulated by growth factor signaling. Expression of MUC1 at the cell surface is also observed and found to be expressed only after HRG stimulation. The total amount of MUC1 remains unchanged, suggesting that this amount is not due to induction of gene expression but to translocation of MUC1 from the inner membrane to the plasma membrane. This reaction is independent of the cytohesin pathway but dependent on PI-3 kinase activity. In addition to these reactions, HRG stimulates cell growth of both HCC2998 and MKN45-1 cells, depending on the ERK pathway given that the MEK inhibitor abolishes this effect. Therefore, HRG induces various reactions in HCC2998 and MKN45-1 cells by different pathways. These reactions are all related to characteristics of tumors, which implicates that HRG signaling can contribute to the formation of tumors.  相似文献

Novel 1-(1-aryl-4,5dihydro-1H-imidazoline)-3-chlorosulfonylourea derivatives 3a3f were synthesized in the reaction of 1-aryl-4,5-dihydro-1H-imidazol-2-amines with chlorosulfonyl isocyanate. The second series of compounds 4a4f was prepared from the respective 1-(1-aryl-4,5-dihydro-1H-imidazoline)-3-chlorsulfonylureas 3a3f and 1,1′-carbonyldiimidazole (CDI). The selected compounds were tested for their activity against Herpes simplex virus and coxsackievirus B3 (CVB3). It was determined that three derivatives, i.e 3d, 4a and 4d are active against Herpes simplex virus (HSV-1). Compounds 3d and 4c are active against CVB3. Their favorable activity can be primarily attributed to their low lipophilicity values. Moreover, the lack of substituent in the phenyl moiety or 4-methoxy substitution can be considered as the most beneficial for the antiviral activity.  相似文献
