Detailed histochemical studies have been conducted on the distribution of various enzymes such as thiamine pyrophosphatase, alpha-glucan phosphorylase, hexokinase, glucose-6-phosphate dehydrogenase, aldolase, lactate dehydrogenase and succinate dehydrogenase in various components of the nucleus Edinger-Westphali, nucleus n. oculomotorii, nucleus ruber and nucleus niger of healthy adult male Wistar strain rats. The thiamine pyrophosphatase reaction showed the morphological patterns of the Golgi apparatus characteristic for each nucleus. The Golgi apparatus was well developed in the nucleus Edinger-Westphali, composing a network of highly fenestrated plates in the nucleus n. oculomotorii and nucleus ruber, and a simple network in the nucleus niger. These results indicate that the former three nuclei need a rich energy supply and argue against the possibility that the four nuclei have a secretory role. The neurons of the nucleus Edinger-Westphali may derive their energy mainly from glucose of the circulating blood, but glial cells may serve as energy donators to the neurons in the pars compacta of the nucleus niger, and the neurons of the other nuclei may derive energy from both sources. These conclusions are consistent with the morphological patterns of the Golgi apparatus.  相似文献

Summary Detailed histochemical studies have been made on the distribution of various enzymes such as thiamine pyrophosphatase (TPPase), hexokinase and glucokinase (HK), phosphoglucomutase (PG), glucose-6-phosphate dehydrogenase (G6PD) and succinate dehydrogenase (SDH) in every component of the dorsal vagal nucleus (X) and the hypoglossal nucleus (XII) of adult healthy male rabbits.The neurons of both nuclei were classified into the same six categories on the basis of the morphology of the Golgi apparatus (GA). Many intermediate forms were observed among these different categories. In general, the neurons of the X showed much more developed GA compared with those of the XII. The present results concerning TPPase may indicate that two different types of neurons are present in both nuclei and that they go through cyclic activity. The GA extensions in the dendrites may also change their forms depending on the functional state.The cytoplasm of the neuron and the neuropil reacted more strongly in the X than in the XII for the HK test. All the glial cells were very strongly positive for HK and they were closely arranged in the X. Both nuclei showed a low level of PG activity. These results indicate that glucose in the blood is the main energy source, that not only oligodendrocytes but also astrocytes work as energy donators to the neurons, and that the neurons of the X get more energy supply than those of the XII. Inverse relationship was found on the distribution of G6PD and SDH in the neurons of the X and XII: The cytoplasm of the neuron in the X was rich in G6PD and relatively poor in SDH whereas this, in the XII, was rich in SDH and poor in G6PD. These findings may provide the support to the conception of Exceptional Nuclei. The predominance of hexose monophosphate shunt in the neurons of the X may provide TPNH which is important for synthesis.The present author assumes that the neurons of the X have vigorous secretory activity on the basis of well-developed GA, rich energy supply and the predominance of hexose monophosphate shunt.  相似文献

Histochemical studies have been conducted by applying hexokinase (HK), aldolase (AD), glyceraldehyde-3-phosphate dehydrogenase (G3), succinate dehydrogenase (SDH), glucose-6-phosphate dehydrogenase (G6PD), and thiamine pyrophosphatase (TPPase) methods, as well as Nissl staining and Gomori's chrome-alum-hematoxylin-phloxine (CHP) methods to intercalated neurons of the supraoptic nucleus (SO) on Wistar strain rats. Intercalated neurons reacted weakly to the AD, G3, G6PD, and SDH tests, indicating that they belong to the category of ordinary neurons with low carbohydrate metabolism. Many fibrous astrocytes showing strong HK reactions surround neurosecretory neurons. However, they do not surround intercalated neurons with mild HK activity. These results indicate that the latter receive a poor supply of energy from glucose in the circulating blood in contrast to the former. Intercalated neurons are very rich in Nissl substance but lack CHP-positive material. They may have a high potential for synthesizing protein. The principal morphological features of the TPPase-positive Golgi material are peculiar and heterogeneous shape and poor development. These findings together with mild G6PD activity suggest that intercalated neurons are very likely to have poor synthesizing activity.  相似文献

Summary Detailed histochemical studies have been conducted on the morphology of the Golgi apparatus by applying the thiamine pyrophosphatase technique (Novikoff and Goldfisher, 1961) to the neurons of supraoptic and paraventricular nuclei of normal and dehydrated rabbits. The neurons in both nuclei were classified into five categories on the basis of the morphology of the Golgi apparatus. The number of cells in individual categories were counted to evaluate the percentage of each category in the whole nucleus.Neurons have many vesicles which show the tendency to form clusters. Such clusters are present also in the basal bodies. The Golgi apparatus is localized near one side of the nucleus in many neurons. The neurons indicate phasic activity of resting, anabolic and catabolic stages under normal conditions.During dehydration, the Golgi apparatus went through the three stages of network formation, the increase of the budding-off process and later on disintegration. The supraoptic nucleus reacted to the TPPase test more severely than the paraventricular nucleus, whereas the former went through the stages more slowly than the latter. The paraventricular nucleus also revealed sensitivity to osmotic stress.  相似文献

Abstract: The dopamine (DA)-containing nerve terminals in the caudate nucleus arise from cell bodies located in the substantia nigra (pars compacta), and it is possible that p-tyramine- and m-tyramine-containing neurons may also exist in this nucleus. We have studied the effects of unilateral electrolytic lesions of the pars compacta in rat on levels of DA, p-tyramine, m-tyramine, and homovanillic acid in the caudate nucleus after various survival times. At 12 and 24 h following lesioning the ipsilateral level of p-tyramine was significantly reduced compared with the contralateral side, whereas the concentrations of m-tyramine, DA, and homovanillic acid were significantly increased. Thus, in the short term, the lesion results in an increase in DA turnover, which is accompanied by an increase in m-tyramine levels and a decrease in p-tyramine levels. Similar changes occur following pharmacological treatments (chlorpromazine, d-amphetamine, l-DOPA) that increase DA turnover. At survival times of 2, 11, and 25 days, the ipsilateral concentrations of m-tyramine, DA, and homovanillic acid were reduced along with p-tyramine. These longer-term alterations in amine levels are most likely a consequence of degeneration of nigro-striatal axons. Placement of a lesion 1 mm dorsal to the usual position centering on the pars compacta produced different biochemical changes from those seen after the pars compacta lesion. One day following this lesion the concentration of p-tyramine was slightly reduced; DA was unaffected, but the concentration of m-tyramine was profoundly increased, even more so than after the pars compacta lesion. This could indicate the existence of specific m-tyramine-containing cell bodies located dorsal to the substantia nigra. The results suggest that p- and m-tyramine in the caudate nucleus originate from neurons in or close to the substantia nigra. The results in the short term following the lesion support the observation that there is an inverse relationship between p-tyramine concentration and DA turnover in the caudate nucleus.  相似文献

Cell-free preparations of Chlorella pyrenoidosa Chick, van Niel's strain, were assayed for oxidative enzymes, utilizing isotopic and spectrophotometric techniques. The enzyme activity of heterotrophic and autotrophic cells was compared. The study was divided into categories, one concerned with the spectrophotometric detection of enzymes involved in the initial reactions of glycolysis and the hexose monophosphate shunt, and the other with the direct oxidation of glucose as compared with that oxidized via glycolysis. The reduction of pyridine nucleotides in crude extracts was studied with glucose, glucose-6-phosphate, 6-phosphogluconate, and fructose-1-6-diphosphate as substrates. Enzymes detected in both heterotrophic and autotrophic cells were hexokinase, fructose-diphosphate-aldolase, NAD-linked 3-phosphoglyceraldchyde dehydrogenase, glucose-6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase, and a NADP-linked 3-phosphoglyceraldchyde dehydrogenase. In addition to isotopic studies designed to make an appraisal of the hexose monophosphate shunt, a comparison of the rate of reduction of NADP by glucose-6-phosphate and 6-phosphogluconate in relation to the reduction of NAD by 3-phosphoglyceraldehyde was made in light- and dark-grown cells. The rate of reduction of NADP appeared to be lowered in the light-grown cells, suggesting, as did also the isotopic studies, that the hexose monophosphate shunt is less active in autotrophic metabolism than in heterotrophic metabolism.  相似文献

The activity of the enzymes of glycolysis (phosphofructokinase, aldolase, pyruvate kinase, lactate dehydrogenase) and hexose monophosphate shunt (glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase) was determined in the eye tissues of the rabbit at different stages of ontogenesis. The activity of these enzymes in the retina was shown to be higher than in other eye tissues. In the uveal tract (iris, ciliary bodies, uvea) the activity of glycolytic enzymes changes with the age. The greatest changes in the activity of enzymes were found during the period of the opening of eyelids. The activity of the enzymes of hexose monophosphate shunt in the eye tissues increases with the age. The relative activity of dehydrogenases of the hexose monophosphate shunt after the establishment of visual function is, however, not high and does not exceed that of phosphofructokinase and pyruvate kinase in the eye tissues of the rabbit.  相似文献

The activity of some enzymes of intermediary metabolism, including enzymes of glycolysis, the hexose monophosphate shunt, and polyol cryoprotectant synthesis, were measured in freeze-tolerant Eurosta solidaginis larvae over a winter season and upon entry into pupation. Flexible metabolic rearrangement was observed concurrently with acclimatization and development. Profiles of enzyme activities related to the metabolism of the cryoprotectant glycerol indicated that fall biosynthesis may occur from two possible pathways: 1. glyceraldehyde-phosphate glyceraldehyde glycerol, using glyceraldehyde phosphatase and NADPH-linked polyol dehydrogenase, or 2. dihydroxyacetonephosphate glycerol-3-phosphate glycerol, using glycerol-3-phosphate dehydrogenase and glycerol-3-phosphatase. Clearance of glycerol in the spring appeared to occur by a novel route through the action of polyol dehydrogenase and glyceraldehyde kinase. Profiles of enzyme activities associated with sorbitol metabolism suggested that this polyol cryoprotectant was synthesized from glucose-6-phosphate through the action of glucose-6-phosphatase and NADPH-linked polyol dehydrogenase. Removal of sorbitol in the spring appeared to occur through the action of sorbitol dehydrogenase and hexokinase. Glycogen phosphorylase activation ensured the required flow of carbon into the synthesis of both glycerol and sorbitol. Little change was seen in the activity of glycolytic or hexose monophosphate shunt enzymes over the winter. Three distinct neurons were identified in the substantia nigra of the rat using Golgi, light, and electron microscopic techniques. A large neuron, found in the pars reticulata, is characterized by well-developed RER, a tubular cytoplasmic inclusion, and somatic and dendritec thorns. A medium-sized neuron, found in the pars compacta, has an eccentric nucleus, distinct Nissl bodies, and an inclusion composed of whorls of concentric cisternae. A small neuron, found in both nigral regions, contains a highly invaginated nucleus, fibrous nuclear inclusion, and paucity of cytoplasmic organelles. Its axon synapses around other nigral dendrites. The presence of these neurons was correlated with the efferent projections and function of the substantia nigra.  相似文献

The spontaneous degeneration of neurons of the Nucl. tractus mesenceph. n. trigemini of 33 male NMRI-mice between 19 and 60 days of ontogenesis has been analyzed fitting the exponential function (see article). The number of neurons decreases from 1500 at birth to 1200 at the adult level. The damping of the curve is 4.8. This nucleus is compared with other nuclei of the albino mouse (Nucl. n. oculomotorii, Nucl. cochleares dorsalis et ventralis). The spontaneous reduction of the number of neurons seems to be a common phenomen during the perinatal time of the mammals. There is a heterochronous reduction for the different analyzed nuclei.  相似文献

Synopsis The activity and distribution of the following eighteen oxidative and hydrolytic enzyme systems have been investigated in the lung of the adult rat: reduced NAD dehydrogenase, reduced NADP dehydrogenase, succinate dehydrogenase, malate dehydrogenase, isocitrate dehydrogenase, lactate dehydrogenase, glucose-6-phosphate dehydrogenase, glucose dehydrogenase, glutamate dehydrogenase, -hydroxybutyrate dehydrogenase, acid phosphatase, alkaline phosphatase, glucose-6-phosphatase, adenosine triphosphatase, 5-nucleotidase, non-specific esterase, cytochrome oxidase and -glucuronidase.The low concentration of cells in sections of inflated lung may have made histochemical demonstration of some enzymes impossible because the enzyme concentration was below that detectable by the method employed.The carboxylic acid cycle and the hexose monophosphate shunt were potentially active but fatty acid metabolism was not indicated.The granular reaction sometimes encountered in alveolar cell cytoplasm may be useful for differentiating alveolar cell types, but further cytochemical studies are required to resolve the possible metabolic differences of alveolar cells.  相似文献

Rat peritoneal macrophages derive energy differently from other tissues. Resting rat peritoneal macrophages have been taken for the present investigation. Lactate produced by extracellular glycolysis in the peritoneal lavage fluid, is readily converted into pyruvate by resting peritoneal macrophages and is oxidised in mitochondria. Glycolytic enzymes other than phosphoglucoisomerase and lactate dehydrogenase could not be substantially demonstrated. Glucose-6-phosphate dehydrogenase was detected. The presence of glucose-6-phosphate dehydrogenase along with phosphoglucoisomerase indicates the operation of the hexose monophosphate shunt as a pathway supplementary to glycolysis. Resting rat peritoneal macrophages thus appear to utilize extracellular lactate as their main energy source instead of glucose, bypass glycolysis and have active hexose monophosphate shunt.  相似文献

Neurons of the substantia nigra show severe morphological changes in Parkinson's disease. Pathological alterations of cell bodies have been described, whereas those of neuronal processes have hardly been investigated. Golgi impregnation has been the chosen method for demonstrating neuronal processes and dendritic and somatic spines. We therefore used the Golgi-Braitenberg method to qualitatively and semi-quantitatively study the substantia nigra of eight patients with Parkinson's disease compared with eight control cases. Golgi impregnation of substantia nigra neurons was good in all control cases. In full agreement with the analysis of Braak and Braak (1986) three neuronal types within the substantia nigra were found. In cases of Parkinson's disease, severe pathological changes such as decrease of dendritic length, loss of dendritic spines and several types of dendritic varicosities were found only in the melanin-containing pars compacta neurons. Pars reticulata nerve cells were intact. These findings support the predominant role played by the dopaminergic efferent pathway in the degenerative process. The afferent pathway was not affected. This suggests that the substantia nigra lesion is primary in Parkinson's disease. Loss of neurons found in H & E sections corresponded to a lesser amount of impregnated pars compacta neurons in cases with Parkinson's disease when compared to controls. Evidences exist that the duration of the disease may be related to the extent of pathologically altered Golgi-impregnated pars compacta cells. The amount of Lewy bodies in H & E sections corresponded to the quantity of round varicosities in impregnated pars compacta neurons. The integrity of the erythrocyte (RBC) hexose monophosphate shunt was investigated in a group of 33 healthy elderly individuals by determining their RBC glutathione content, glucose-6-phosphate dehydrogenase activity and glutathione regeneration. When these parameters were compared with those of the controls, 44 young healthy adults, no significant differences were found. This study indicates that the RBC hexose monophosphate shunt in healthy elderly individuals is intact. Factors other than senescence per se should be sought in elderly individuals who exhibit dysfunction of this shunt.  相似文献

Summary The ovaries of 70 mature Mongolian gerbils (Meriones unguiculatus) were investigated morphologically and enzyme histochemically for the appearance of acid phosphatase, non-specific esterases, succinate dehydrogenase and thiamine pyrophosphatase. In the oocyte two particular enzyme active zones exist depending on the state of development. In young oocytes acid phosphatase, succinate dehydrogenase and thiamine pyrophosphatase can be found only in the perinuclear zone. From the late secondary follicle on, activity of acid phosphatase, succinate dehydrogenase and non-specific esterases can be detected only in a peripheral area of cytoplasm, whereas thiamine pyrophosphatase is present in the entire ooplasm.In the follicular epithelium a different pattern of enzyme distribution suggests a functional differentiation of the epithelial cells during folliculogenesis.This investigation was supported by the Deutsche Forschungsgemeinschaft  相似文献

Summary The isthmic nuclei of the pigeon were studied with the use of three different Golgi techniques. The nucleus isthmo-opticus (IO) consists of a single cell type in which all dendrites of one neuron take the same direction and ramify at identical distances from the perikaryon to form dense dendritic arborizations. The cell bodies of the IO neurons form two parallel layers. The dendrites of these neurons always extend to the area between the two layers so that the dendritic arborizations of opposite neurons overlap. A model of the cellular organization of the IO was constructed based upon these morphological characteristics. The neurons of the n. isthmi/pars parvocellularis (Ipc) have oval perikarya and long, smooth, infrequently branching dendrites. All neurons except those at the borders of the nucleus show the same dorsoventral orientation in their dendritic arborizations and together with their afferents seem to have a columnar organization. The dendrites of the neurons located at the margin of the nucleus ramify within the Ipc along its border. The n. semilunaris (Slu) consists of neurons with round somata that have on an average three dendrites with small spines. The axons leave the nucleus from the medial side and join the lemniscus lateralis. The neurons of the n. isthmi/pars magnocellularis (Imc) comprise a generalized isodendritic type resembling the cells of the reticular formation. Axons from the tectum penetrate the nucleus, making numerous en-passant contacts with several neurons.  相似文献

A major output nucleus of the basal ganglia is the substantia nigra pars reticulata, which sends GABAergic projections to brainstem and thalamic nuclei. The GABAergic (GABA) neurons are reciprocally connected with nearby dopaminergic neurons, which project mainly to the basal ganglia, a set of subcortical nuclei critical for goal-directed behaviors. Here we examined the impact of motivational states on the activity of GABA neurons in the substantia nigra pars reticulata and the neighboring dopaminergic (DA) neurons in the pars compacta. Both types of neurons show short-latency bursts to a cue predicting a food reward. As mice became sated by repeated consumption of food pellets, one class of neurons reduced cue-elicited firing, whereas another class of neurons progressively increased firing. Extinction or pre-feeding just before the test session dramatically reduced the phasic responses and their motivational modulation. These results suggest that signals related to the current motivational state bidirectionally modulate behavior and the magnitude of phasic response of both DA and GABA neurons in the substantia nigra.  相似文献

Summary The occurrence and distribution of neurotensin-immunoreactive (NT-IR) perikarya was studied in the central nervous system of the guinea pig using a newly raised antibody (KN 1). Numerous NT-IR perikarya were found in the nuclei amygdaloidei, nuclei septi interventriculare, hypothalamus, nucleus parafascicularis thalami, substantia grisea centralis mesencephali, ventral medulla oblongata, nucleus solitarius and spinal cord. The distribution of NT-IR perikarya was similar to that previously described in the rat and monkey. In the gyrus cinguli, hippocampus and nucleus olfactorius, though, no NT-IR neurons were detected in this investigation. Mice were infected with cysts of the ALT strain Toxoplasma by intraperitoneal injection. After 2-8 weeks disseminated cysts could be demonstrated in the brain tissue. All cysts showed identical histochemical characteristics, independent of their sizes or their cell number. The encysted organisms were intensely stained after the PAS-reaction. This polysaccharide is highly diastase and acid resistant. Glycogen synthetase activity could not be demonstrated, but phosphorylase activity was very high. The energy metabolism was characterized by a high lactate dehydrogenase activity, whereas the reaction for succinate dehydrogenase activity only leads to sparse deposits of reaction products. The carbohydrate content is interpreted to be not only a store of energy substrate but also a store of biosynthetic substrate. It is assumed that a part of the liberated glucose at high activities of G-6-P-DH and 6-P-G-DH is metabolized by the hexose monophosphate shunt, the pentoses of which may contribute to nucleic acid synthesis which is necessary for the proliferation of the encysted organisms.  相似文献

Aldosterone stimulates Na+ transport in toad bladder and, simultaneously with a coincident dose-response relationship, inhibits the hexose monophosphate shunt pathway. Amiloride, an acylguanidine diuretic, inhibits sodium transport when applied to the apical surface of the bladder. In this study amiloride was found to partially reverse the inhibitory effect of aldosterone on the hexose monophosphate shunt pathway. The amiloride effect upon glucose metabolism was detected when it was applied to both surfaces of the bladder simultaneously, in flask experiments, and when it was applied to the apical surface. No effect of amiloride on the shunt pathway was detected when it was applied to the serosal surface only, even at very high concentrations. It may be, but has not been proven, that the effects of aldosterone and amiloride on the hexose monophosphate shunt pathway are mediated by a common site at the apical membrane.  相似文献
