Growth analysis of plants raised under controlled environments(10–5, 12, 15, 18 and 20 °C, and 21 h photoperiod)was used to examine whether varietal differences in the minimumgermination temperature of four bean cultivars persist duringgrowth at suboptimal temperatures. A method to estimate theminimum vegetative growth temperature, based on axis relativegrowth rate, was developed. In order to compensate for ontogeneticdrift, the harvests were conducted at the same stage of developmentof the plants. Axis relative growth rates, reduction rates ofthe cotyledons and other growth parameters were calculated inorder to compare the cultivars. Cultivar ‘Marschall’showed better growth potential at 12 °C than the others,‘Pergousa‘ at 15 °C, and ‘Marschall’,‘Olsok’ and ‘Pergousa’ at 18 and 20°C. The effect of temperature on axis RGR was similar for‘Marschall’, ‘Olsok’ and ‘Pergousa’(Q10 = 2·1) and more pronounced than for ‘Processor’(Q10 = 1·3). Although there were significant differencesin the growth parameters among the cultivars within each temperatureused, the differences did not correspond with the differencesduring germination at low temperatures. The minimum vegetativegrowth temperature was close to 10 °C for all the cultivarstested. Requirements for seed germination (emergence of radicle) andseedling formation (emergence of both radicle and cotyledons)of a hemi-root parasite Sopubia delphinifolia were studied inthe absence of any host stimulus, in Petri dish and asepticcultures. Both water washing in trickling tap water as wellas cold treatment were effective in inducing/stimulating germinationand seedling formation in the light. Although ethrel, an ethylenereleasing compound, stimulated radicle emergence it inhibitedthe emergence of the cotyledons. Light was found to be essentialfor germination; none of the growth substances could replacethe light requirement. Light responses seem to be mediated throughthe phytochrome system. The results indicate that the emergenceof the radicle, its further growth into the root and the emergenceof cotyledons are controlled by different factors. Oscillation in the growth rate as well the lag time for an IAAresponse in bean (Phaseolus vulgaris) hypocotyls were studiedwith a displacement transducer. The growing zone of the hypocotylshowed continuous oscillation in the growth rates for intactseedlings, hypocotyl explants with or without cotyledons, andexplants submerged in water. The lag time for the IAA response was shortened by the presenceof cotyledon tissues as well as by preliminary treatment withgibberellic acid and sucrose. When IAA first was applied tothe basal cut surface then to the upper cut surface of the hypocotylexplant, two growth responses with distinct lag periods werefound. Two Phaseolus vulgaris L. cultivars were grown at 20/15, 25/20,and 30/25 °C day/night temperatures in growth chambers witha 16 h thermoperiod corresponding to the photoperiod. When thefirst trifoliolate leaf was fully expanded rates of CO2 exchange(CER) were measured at 27 °C and saturating light usinginfrared gas analysis. Stomatal (rs) and mesophyll resistances,CO2 compensation points, activities of the enzymes ribulosebisphosphate carboxylase (RuBPCase), glycolate oxidase (GAO),malate dehydrogenase (MDH), and fructose-1, 6 diphosphate (FDP),chlorophyll content, Hill activities, and leaf anatomy at boththe light and electron microscope level were also investigatedin these leaves. Rates of CO2 exchange in the light, transpiration rate, andchlorophyll content increased with increasing growth temperaturewhile leaf thickness, specific leaf weight, RuBPCase activity,compensation point, and stomatal resistance decreased. Mesophyllresistance also decreased when calculated assuming zero chloroplastCO2 concentration (rm, o), but not when calculated assuminga chloroplast CO2 concentration equal to the CO2 compensationconcentration (rm, g). Average leaf size was maximal in 25/20°C plants while dark respiration, MDH activity, stomataldensity, and starch were minimal. Nutrient absorption in wheat and sugarbeet was studied duringpre-emergence growth by adding 0, 7, 10.5 or 14 mol m-3nitrogen(N) to the growth medium. Seedling growth and carbon, N and15Ncontents of the seedling parts were measured. Differences betweenthe natural abundance of15N in seeds and in nutrient solutionwere used to determine the proportion of N in the organs originatingfrom seed reserves and from absorption. Absorption began laterfor wheat than for sugarbeet and had less effect on seedlinggrowth. The absorbed N was found mainly in roots. Compared towheat, sugarbeet seedling N content was greatly altered andthe hypocotyl showed increased elongation when nutrients wereadded. Most of the absorbed N was found in the radicle and hypocotylwith less in the cotyledons. Heterogeneity within the root environment results in differentialgrowth within root systems. The response of five Phaseolus vulgarisL. cultivars to non-uniform root aeration was evaluated. Threetreatments were applied to a split root system for a periodof 72 h. Treatments consisted of an aerated control, a non-aeratedcontrol (both halves non-aerated, using N2). and localized anoxia(one-half the root system aerated and the remaining half subjectedto N2). Shoot and root growth were reduced in the anoxic controlbut not in the aerated control or localized anoxia treatment. Root growth was greatest in the aerated portion of the localizedanoxia treatment for all genotypes. Contributions of the rootcomponents to the compensatory responses differed dependingon the plant cultivar examined. The growth of branched and lateralroots present before the treatment period increased by 65% inline 31908. A 50% increase in the growth of lateral roots whichemerged during the treatment period occurred in another line(Swan Valley). Other genotypes responded in an intermediatemanner. Growth during pre-emergence of three sugarbeet genotypes, alltolerant to the root disease rhizomania but with poor emergencefeatures, was studied. Dry mass, carbon and nitrogen content,and N isotope composition were measured for the seeds and eachseedling part. These results were compared with those obtainedfor Vega, a genotype with no emergence problems. The depletionof seed reserves and carbon use were similar for all genotypes.Tolerant genotypes had altered nitrogen absorption: the netnitrogen balance (total N in the seedling/N initial mass inthe seed) was 20% lower in these genotypes at the end of theexperiments. Differences in the natural abundance of15N in seedsand nutrient solutions were used to determine the proportionof N originating from seed reserves or from absorption by theorgans. N absorbed from the nutrient solution was found mainlyin the hypocotyl and radicle, with less in the cotyledons ofall genotypes. Tolerant genotypes had less absorbed N in allparts than non-tolerant genotypes. Hypocotyl and radicle elongationwas the same in all genotypes, but the emergence forces in rhizomania-tolerantgenotypes were smaller. These results suggest that breedingto introduce tolerance to rhizomania has also led to a loweredmineral absorption capacity. A method is described for estimating gibberellin levels in plantmaterial, using a separation method avoiding organic solvents.High molecular weight inhibitors were separated from growth-promotingfractions by column chromatography on Sephadex. Of the severalgrades of Sephadex tried, G15 was found to give the sharpestseparation. Ammonium chloride was included in the sample appliedto the column to increase the retention of anionic substances.The inhibitory effect of other substances, probably abscisicacid, which are eluted from the gel together with the gibberellins,was reversed by addition of kinetin to test solutions in thelettuce hypocotyl bioassay. Using this method an increase ingibberellin level in cotyledons of Phaseolus multiflorus duringgermination was measured in terms of GA3 activity, with referenceto the lettuce hypocotyl bioassay.  相似文献

Seedlings with leaf-like photosynthetic cotyledons such as Cucumissativus, Sinapis alba and Trifolium pratense have higher growthrates and a greater requirement for an external supply of potassiumthan hypogeal species (Pisum sativum, Vicia faba) or epigealspecies with very fleshy cotyledons (Phaseolus vulgaris, Lupinusalbus, Lupinus angustifolius). The initial stages of seedlingdevelopment in the latter two groups involve mainly transferof reserves from cotyledons to axis and a relatively small increasein total seedling dry weight takes place during the first 2to 3 weeks. Potassium may be drawn either from the supply already presentin the cotyledons or from an external source. Species with expandingphotosynthetic cotyledons utilize the reserve potassium duringcotyledon development and it is not transported to the epicotyl.Thus, in these species an external supply of potassium is essentialfor full development of the photosynthetic system and the roots.In species whose cotyledons are not major photosynthetic organsthe reserve potassium is transported from the cotyledons tothe developing axis. In these species culture in a nutrientsolution deficient in potassium or in distilled water increasesroot dry weight but reduces plumule weight (although to a lesserextent than in species with leaf-like cotyledons). In general,the response of seedlings to culture in a nutrient solutiondeficient in potassium was very similar to culture in distilledwater only. Thus, although all the species studied had similar levels ofpotassium in the seed (µg potassium/mg dry weight) itssite of utilization differs. It appears that seedlings withleaf-like cotyledons are more susceptible during early seedlinggrowth to deficiencies in external factors, such as potassium,than are species with fleshy cotyledons.  相似文献

The application of 8[14C]t-zeatin to the cotyledons of germinatingbean seeds demonstrated that cytokinins are not readily exportedfrom the cotyledons to the embryonic axis during the early stagesof this process. In the cotyledons the applied zeatin is metabolizedextensively to metabolites which are polar and which occur atRF 0·2–0·5 on paper chromatograms. Thesemetabolites are stable and are not readily exported from thecotyledons. In contrast the metabolites found at RF 0–0·2are more readily exported. When exported to the radicles andplumules a large proportion of the translocated metaboliteswere converted to compounds which on paper co-chromatographedwith zeatin. This seems to suggest that the embryonic axis hasthe capacity to synthesize cytokinins and that some of the metabolitesformed during its catabolism can also be used for its synthesis. Plants of the C3 species Phaseolus vulgaris and Xanthium strumariumand of the C4 salt-sensitive Zea mays and the C4 halophyte Atriplexhalimus were grown with and without NaCl salt-stress at normal(340 µl I–1) and at high (2500 µl I–1)ambient CO2. In all four species growth (dry weight increment)was enhanced by CO2 supplementation. The relative response wasgreater in the salinized than in the control plants. Plant topsresponded more to CO, than the roots. CO2 supplementation appearsto increase plant tolerance of low levels of salinity. Key words: Salinity, CO2, Growth  相似文献

Changes in the dry weight (dry wt.), total and neutral lipids,and fatty acid composition were determined in the cotyledonsand axis of Citrullus lanatus cv. Sugar baby seedlings duringtheir first 12 d growth in the dark, at 25 ? 1 ?C. The major stored reserves were mobilized between days 3 and8. The lipid concentration after day 4 decreased rapidly inthe cotyledons. On the first day the lipid concentration inthe axis was higher than in the cotyledons but decreased rapidlyand from day 5 up to the end of the experiments it remainedat very low values. In the cotyledons, the change in the fattyacid composition was relatively small compared to the significantchange observed in the axis. The neutral lipids of the cotyledonsdecreased sharply between days 2 and 6 while their fatty acidcomposition suffered no significant change. The axis, comparedto the cotyledons, contained a small but concentrated amountof neutral lipids, which decreased up to day 6. Thereafter,an increase was observed. The fatty acid composition of theaxis neutral lipids changed significantly, linoleic and oleicacid decreased while palmitic and linolenic acid increased. Effects of Acetylcholine on the Growth of the Vigna Seedling   总被引:2,自引:0,他引:2  
Acetylcholine applied to cotyledons influenced the growth patternof etiolated seedlings of Vigna sesquipedalis in a specificmanner. The reagent as well as red light suppressed hypocotylgrowth and simultanously promoted epicotyl growth. Such effectswere not found with choline. Neostigmine, a potent inhibitorof cholinesterases from plants, caused fundamentally the sameeffects as acetylcholine. Atropine, an acetylcholine antagonistin animals showed effects opposite to those of acetylcholineor neostigmine. Atropine also partially negated the red light-inducedeffects. Indole-3-acetic acid, kinetin and gibberellic acid could notproduce effects like those of cholinergic reagents. From these results, it was surmised that acetylcholine as agrowth substance in the Vigna seedling regulates the transitionfrom hypocotyl growth to subsequent epicotyl growth. Ethanolic extracts from the cotyledons of mature dry Phaseolusvulgaris L. seed yielded cytokinin-like activity which co-chromatographedwith zeatin and ribosylzeatin. Under conditions which stimulatedgermination and cotyledon expansion, the level of these cytokininsdecreased rapidly in both intact embryos and excised cotyledons.In the excised cotyledons the decrease was continuous, resultingin very low levels of cytokinin being detected after 4 daysof incubation. With the embryonic axis present, however, theinitial decrease was arrested and reversed after 3 days. Thissuggests that the cotyledons do not synthesize cytokinins butthat these hormones are imported from the embryonic axis, particularlyonce radicle growth is well under way. The growth of the first pair of leaves of Phaseolus vulgaris(French bean) has been studied during germination and followingemergence of the seedling. The leaves are well developed inthe embryo and, at 22.5° C, show an exponential increasein fresh weight, dry weight, and leaf area up until about eightdays from planting. Cell division commences about two days afterplanting and is exponential for a short period. Considerablechanges in cell volume occur during the period over which celldivision occurs. Cell division ceases soon after emergence andunfolding, when the leaf has reached only 17 per cent of itsfinal area. Cessation of cell division is followed by a phaseof growth which is due entirely to cell expansion. The significanceof these findings is discussed in relation to recent work onother genera.  相似文献

Three techniques used to investigate whole-plant growth areplant growth analysis, yield component analysis and demographicanalysis. Each subdivides growth into morphological or physiologicalcomponents. This paper derives several relationships which definethe contributions made by components to the performance of thewhole plant. For example, the additive contributions by differentplant parts to overall unit leaf rate may be determined. Also,for multiplicative components, the relative growth rate of yieldis the sum of the relative growth rates of yield components.The relationships developed here serve to link different approachesto growth analysis, and they are illustrated using data fromgrowth studies of bean and sunflower. Changes in lamina area, dimensions of epidermal and palisadecells, acid invertase activity and content of sucrose and hexosein the primary leaves of Phaseolus vulgaris L. were determinedbetween emergence of the hypocotyl hook and the completion ofleaf expansion. Growth in area and thickness of the primaryleaf after emergence was attributable to the expansion of cellsalready present in the lamina at emergence. The major invertasein the expanding leaf was a readily soluble acid invertase;little insoluble invertase activity was detected. Soluble andinsoluble fractions of leaf homogenates contained little neutralinvertase activity. The specific activity of the soluble acidinvertase increased rapidly during the early stages of leafexpansion, reaching a peak at the time of most rapid cell enlargement(5 d after emergence) and then declining as the leaf matured.Highly significant positive correlations were found betweenenzyme specific activity and the rates of cell and leaf enlargement. The early, rapid phase of lamina expansion was characterizedby high concentrations of hexose sugar and low concentrationsof sucrose. As the rates of leaf cell enlargement declined theconcentration of hexose fell and that of sucrose increased.Between 5 d and 11 d after hypocotyl emergence, the hexose/sucroseratio in the primary leaf decreased approximately 10-fold asthe specific activity of acid invertase decreased. The results are discussed with reference to sources of carbonsubstrates for cell growth and to the sink/source transitionduring leaf development. The application of 8[14C]t-zeatin to the radicle tips of germinatingPhaseolus vulgaris seeds revealed that cytokinins are transportedrapidly from the embryonic axis to the cotyledons, and are utilizedand metabolized extensively in these organs. The informationobtained on the transportation between the different parts ofthe embryo is consistent with the view that the mobilizationof food reserves from the cotyledons is controlled by cytokininswhich originate in the embryonic axis. Tentative identificationof the radioactive metabolites formed indicate that the appliedzeatin was altered by side-chain cleavage and by substitutionto the adenine ring. Bot. 36: 1900–1905.Changes in abscisic acid (ABA)content with time were measured in embryonic axes and in cotyledonsof Phaseolus vulgaris embryos using a radio–immunoassay.During embryogenesis, a similar pattern was observed in bothtissues: ABA increased to a maximum 29 d after an thesis, followedby a decrease as the seed matured. The level of ABA in the cotyledonswas always much higher than that in the axes. In in vitro cultures,the duration of the lag phase before germination of isolatedembryonic axes increased with ABA content. The presence of cotyledonsalways lengthened the lag phase; longer lag phases were associatedwith greater concentrations of ABA in the cotyledons.
