对毛茛科铁线莲属Clematis L.的单性铁线莲组sect. Aspidanthera Spach s.l.进行了全面修订,确定此组共含72种和15变种,这些种被归类于6个亚组中,其中包括首次描述的1系、5种和2变种,以及做出的2新等级.对单性铁线莲组的分类学简史和地理分布做了介绍.写出了组、亚组、系的形态特征和地理分布,分亚组检索表及各亚组的分种检索表;以及各种植物的形态描述、地理分布、生长环境等,并附有多幅插图.在研究了此组全部种类的标本之后,观察到此组几个重要形态特征的演化趋势(1)萼片在数目上由4枚演变到8枚,在开展方向上由平展到直上展,在形状上由长圆形到狭条形,在长度上由比雄蕊稍长到2-4倍长于雄蕊,其卷叠式由镊合状到次生的覆瓦状;(2)花药药隔顶端由不具突起到具短或长的突起;(3)雌花退化雄蕊的数目由多数到定数、少数,以至完全消失;(4)花序自当年生枝的叶腋发生演变到与数叶同自老枝的一腋芽中发出.根据上述演化趋势推断本组各亚组间的亲缘关系如下subsect. Dioicae (34种,广布于北美洲和南美洲)的花构造(萼片4,镊合状排列,平展,通常呈长圆形,稍长于雄蕊或近等长,外面边缘被短绒毛;雄蕊花药呈长圆形,药隔顶端无突起;雌花具多数退化雄蕊;花序自当年生枝的叶腋发出)与具两性花的威灵仙组欧洲铁线莲亚组sect. Clematis subsect. Clematis 的花极为相似,区别主要在于本亚组的花为单性,由此判断subsect. Dioicae可能是单性铁线莲组sect. Aspidanthera的原始群,源出于欧洲铁线莲亚组.本组的第二亚组subsect. Lasianthae(2种,分布于北美西南部)与subsect. Dioicae 在亲缘关系上极为相近,区别只在于其花序或具花当年生枝与数叶一同由老枝的一个腋芽中发出,此亚组当是从subsect. Dioicae衍生出的一个小群.第三亚组subsect. Microphyllae(7种,特产澳大利亚)也与subsect. Dioicae相近缘,但此亚组的萼片变狭长,多呈条形或狭条形,常2-4倍长于多少变短的雄蕊,雌花的退化雄蕊数目变少,16-2枚,根据这些进化特征,推测此亚组也源出于subsect. Dioicae.第四亚组subsect. Aristatae (16种,分布于澳大利亚、新几内亚及邻近岛屿)与subsect. Microphyllae 在亲缘关系上相近,但本亚组的花药药隔顶端具短或长的突起而不同,根据此进化特征,推测本亚组系由后者演化而出.第五亚组subsect. Hexapetalae (11种,特产新西兰)的花构造与分布于澳大利亚的subsect. Microphyllae相似,但其萼片为覆瓦状排列,外面边缘不被短绒毛,在多数种多于4枚,为5-8枚而不同.据Godley 的研究,本亚组中的C. afoliata的花有4枚萼片,排成2轮,每轮的2枚萼片均为近镊合状排列;另外,在C. paniculata花的6枚萼片中,4枚为覆瓦状排列,其他2枚有时内向镊合状排列.从上述情况可见此亚组的萼片覆瓦状卷叠式可能是由镊合状卷叠式演变而来的一种次生现象,并由此推测,此亚组可能与subsect. Microphyllae相同,也源自美洲的subsect.Dioicae.最后一个亚组subsect. Insidiosae(2种,特产马达加斯加)的雌花萼片直上展,退化雄蕊完全消失,具有这些进化特征,当是单性铁线莲组的进化群,可能源出自具定数或少数退化雄蕊的subsect. Microphyllae.  相似文献

During the past several years, the use of a smokable form of methamphetamine hydrochloride called "ice" has increased rapidly. The heaviest use has occurred on the West Coast and in Hawaii. Many regional emergency departments treat more methamphetamine users than cocaine-intoxicated patients. The ease of synthesis from inexpensive and readily available chemicals makes possible the rampant abuse of a dangerous drug that can produce a euphoria similar to that induced by cocaine. The absence of the larval Anisakis simplex s. l. in samples from central Chile but heavy infections in fish from southern localities suggest the existence of two stocks of Strangomera bentincki, one closely associated with the central coast of Chile (landing port San Antonio and Talcahuano, c. 33° 30′ S–36° 40′ S) and the other associated with southern Chile (landing port Puerto Montt, c. 41° 30′ S). Results confirm the usefulness of metazoan parasites, like A. simplex s. l. as biological markers.  相似文献

The sequences of chloroplast trn K intron 5' region were determined for seven genera of Ulmaceae s.l. ( 12 species), two genera of Cannabaceae, two of Moraceae and one genus from each of the three families, i.e. , Urticaceae, Eucommiaceae and Malvaceae. The aligned sequences used in PAUP analyses were 805 bp, and the maximum parsimony analysis resulted in a single most parsimonious tree with tree length = 665, CI = 0.7714 and RI = 0.7965. The topology of the tree is relatively strongly supported by the bootstrap test and congruent with those generated from rbc L sequence analysis and cpDNA restriction site analysis. The phylogenetic tree indicated that Ulmaceae s. l. is polyphyletic, and that the monophyletic Ulmaceae s. str. is sister to the other Urticales taxa. Cannabaceae, represented by Humulus and Cannabis, is nested within Celtidaceae, revealing that Celtidaceae is a paraphyletic group. Gironniera and Aphananthe are both clades of Celtidaceae. The study also suggested that the trn K intron should be suitable for phylogenetic analyses at intergenericor interspecific levels. Scutcheon noninitial, exuvial touchiness alitizing. An RFLP analysis of the chloroplast genetrnK of 32 species of the generaPapaver, Roemeria, Stylomecon, andMeconopsis leads to the following conclusions: (1) AsianMeconopsis consists of two distinct clades and is paraphyletic in relation toPapaver, Roemeria, Stylomecon, and the W EuropeanMeconopsis cambrica. (2) Sister group relationships ofRoemeria toPapaver sect.Argemonidium and ofStylomecon toPapaver californicum are well-supported. (3)Meconopsis cambrica is nested withinPapaver (incl.Roemeria andStylomecon). The consideration of morphology, geographical distribution and ecology leads to the conclusion thatM. cambrica is best regarded as a member ofMeconopsis, and thatPapaver arose polyphyletically from within a paraphyleticMeconopsis in response to Tertiary climatic aridification. — The removal ofM. cambrica from the taxon matrix is discussed. It is concluded that this experiment illuminates the importance of critical taxon sampling, and shows that at least potentially the assessment of taxa as mono-, para-, or polyphyletic may characterize their present status only and need not reflect their phylogenetic history.  相似文献

Protosilvius gurupi sp. n. (Tabanidae, Pangoniinae) is described and illustrated based on seven female and 53 male specimens collected in the Amazonian region at Reserva Biológica Gurupi, Centro Novo do Maranhão municipality, northwest Maranhão, Brazil. This is the first record of Protosilvius in northern Brazil and in the Amazon Basin. An illustrated key to all Protosilvius species is also presented.  相似文献

In this paper, we describe the new genus Pauropygus gen. n. which includes three minute species, blind and unpigmented, living in interstitial littoral habitats in tropical or subtropical countries. Two of these species are new to science (type species Pauropygus projectus sp. n. from New Caledonia and Pauropygus pacificus sp. n. from China); the third one, originally described in the genus Cryptopygus (Cryptopygus caussaneli Thibaud, 1996), has a larger pantropical distribution. We synonymize here Cryptopygus riebi Barra, 1997 from South Africa with Pauropygus caussaneli. Two paratypes of the Mexican species Cryptopygus axayacatl Palacios & Thibaud, 2001 turned also to be Pauropygus caussaneli, while the holotype and remaining paratypes of this species support its placement in Proisotomodes. Among the Cryptopygus complex, Pauropygus gen. n. is easily recognized by characters of mouthparts (presence of two large projections on pleural fold, basolateral field with 6 chaetae, modified mouthparts) and reduced sensillar chaetotaxy (tergal sensilla 2-3,0-1/0-1,0-1,1-2,1-2,1-3, microsensilla reduced in number: 00/0-100, with sensilla situated in p-row on the abdomen). Small size, absence of eyes and pigment are also shared by all its species. The three species belonging to the genus differ by sensillar chaetotaxy.  相似文献

We present here three expression plasmids for Trypanosoma cruzi adapted to the Gateway® recombination cloning system. Two of these plasmids were designed to express trypanosomal proteins fused to a double tag for tandem affinity purification (TAPtag). The TAPtag and Gateway® cassette were introduced into an episomal (pTEX) and an integrative (pTREX) plasmid. Both plasmids were assayed by introducing green fluorescent protein (GFP) by recombination and the integrity of the double-tagged protein was determined by western blotting and immunofluorescence microscopy. The third Gateway adapted vector assayed was the inducible pTcINDEX. When tested with GFP, pTcINDEX-GW showed a good response to tetracycline, being less leaky than its precursor (pTcINDEX).  相似文献

The hypothesis that the elements of the modern species-rich flora of the Cape Floristic Region (CFR), South Africa, originated more or less simultaneously at the Miocene/Pliocene boundary, in response to the development of a mediterranean climate, has been challenged by numerous molecular dating estimates of Cape floral clades. These studies reveal a more gradual emergence, with the oldest clades originating in the Eocene, but others appearing later, some as recently as the Pliocene. That there are factors which might affect the dates recovered, such as choice of calibration point, analysis method, sampling density and the delimitation of Cape floral clades, suggests a need for further critical evaluation of the age estimates presented to date. In this study, the dates of origin of two Cape floral clades (the legume Crotalarieae p.p. and Podalyrieae) are estimated, constrained by a shared calibration point in a single analysis using an rDNA ITS phylogeny in which 633 taxa are sampled. The results indicate that these two clades arose contemporaneously 44-46 mya, not at the Miocene/Pliocene boundary as had been previously supposed. The contemporaneous origin of these Cape floral clades suggests that additional more inclusive analyses are needed before rejecting the hypothesis that a single environmental trigger explains the establishment of Cape floral clades.  相似文献

Pollen grains of species of the subgenera of Cybianthus s.l. were analyzed in order to determine if pollen morphology is consistent with the currently accepted infrageneric classification, and if it can provide characters useful for distinguishing species or groups of species. Acetolyzed pollen grains were analyzed, measured, described and photomicrographed, while non-acetolyzed pollen grains were analyzed using scanning electron microscopy. The pollen grains of all species investigated were isopolar monads, small- to medium-sized, subprolate or prolate, 3-colporate, and rugulate or rugulate-perforate. The different analytical methods (statistical analyses, exploratory analysis of the data and cluster analysis) used demonstrated that pollen morphology is informative in delimiting this genus. However, the pollen characters are generally quite uniform among the subgenres of Cybianthus s.l. Thus, the use of the pollen attributes in the separation of the subordinate species to different subgenera is not effective. Pollen morphology did not give support to current subgeneric systematics, but it did support the genus Cybianthus s.l.  相似文献

Type material of several polychaete species described by Enrique Rioja from Mexican coasts are lost, and the current status of some species is doubtful. Nereis oligohalina (Rioja, 1946) was described from the Gulf of Mexico, but it has been considered a junior synonym of Nereis occidentalis Hartman, 1945, or regarded as a distinct species with an amphiamerican distribution. On the other hand, Nereis garwoodi González-Escalante & Salazar-Vallejo, 2003, described from Chetumal Bay, Caribbean coasts, could be confused with Nereis oligohalina. In order to clarify these uncertainties, Nereis oligohalina is redescribed based on specimens from the Mexican Gulf of Mexico, including a proposed neotype; further, Nereis garwoodi is redescribed including the selection of lectotype and paralectotypes, and Nereis confusa sp. n. is described with material from the Gulf of California. A key for the identification of similar species and some comments about speciation in nereidid polychaetes are also included.  相似文献

Developmental morphology and anatomy of Botrychium s.l. were studied to clarify rhizome ontogeny and patterns of tissue maturation that can be used to test the hypothesis that ferns of the Ophioglossales may represent living progymnosperms. Serial anatomical sections of the rhizomes of B. virginianum and B. dissectum reveal that apical meristematic activity and vascular tissue maturation occur over an extended period of several years and then stop. Most of the xylem consists of radial rows of tracheids and interspersed ray-like xylem parenchyma cells that are similar in these respects to secondary xylem, but pits occur on all tracheid walls as is characteristic of primary xylem. No vascular cambium is initiated in mature primary tissues, nor is there secondary phloem. Radial rows of xylem cells are produced by the direct continuation of divisions that begin at the shoot apical meristem, forming a cylinder of radially aligned procambial cells before the differentiation of protoxylem. Continuing divisions over a period of several years increase the number of thin-walled cells and tracheids in each radial row back to about one internode behind where the current year's frond trace diverges from the rhizome stele. At more proximal levels of the rhizome, procambial cell divisions cease and there is no additional tracheid differentiation. These data reveal that the rhizome matures over an exceedingly long period of several years, but that growth is ultimately determinate, thus supporting hypotheses that the Ophioglossales is more closely related to other groups of living ferns than to progymnosperms and seed plants.  相似文献

Taxaceae s.l. comprise six genera (including Cephalotaxus) and about 35 species; The present study aims to give new insights into the evolution of this family, especially into the phylogenetic position of Cephalotaxus. Moreover, only little is known about comparative leaf anatomy of this family and this study aims to expose and interpret the diversity and evolution of leaf anatomical characters and to assess their applicability to identify taxa at the generic and species level. A detailed phylogeny was reconstructed, using both maximum likelihood and Bayesian inference, with a combined dataset of four molecular markers from the plastid and nuclear genomes. Leaf sections from 132 specimens, representing 32 species and four varieties (fresh and herbarium material) were inspected, using fluorescence microscopy. Ancestral characters were reconstructed using Mesquite. The phylogenetic analyses provided full support for Cephalotaxus as sister group to Taxaceae s.str. Within the latter, two monophyletic tribes Taxeae (comprising Austrotaxus, Pseudotaxus, and Taxus) and Torreyeae (comprising Amentotaxus and Torreya) were fully supported. Fluorescence microscopy was shown to be very useful for identifying leaf tissues and their constitution. We were able to show that particularly sclerified tissues have highest potential for the discrimination of both freshly collected samples and rehydrated herbarium specimens at the generic and species level. A correlation between the presence of different sclereid types could be shown and sclereids were hypothesized to pose a primitive trait in the evolution of Taxaceae s.l. New identification keys were generated on the basis of leaf anatomical characters. The microscopic method presented here is applicable for further studies within gymnosperms and probably in angiosperms, as well.  相似文献

Duvalius (sg. Neoduvalius) gejzadunayi sp. n. from Pećina u Dubokom potoku cave ( Donje Biševo village near Rožaje, Montenegro), the first known representative of this subgenus from the territory of Montenegro is described, illustrated and compared with the related species of the subgenus Neoduvalius Müller, 1913. This new species is characterised by depigmented, medium sized body, totally reduced eyes, deep and complete frontal furrows, 3–4 pairs of discal setae in third elytral stria, as well as by the shape of aedeagus. Data on the distribution and the ecology of this remarkable species, as well as a check-list of the subgenus Neoduvalius are also provided. Recently described genera Serboduvalius Ćurčić, S. B. Pavićević & Ćurčić, B.P.M., 2001, Rascioduvalius Ćurčić, S. B. Brajković, Mitić & Ćurčić, B.P.M., 2003, Javorella Ćurčić, S. B. Brajković, Ćurčić, B.P.M. & Mitić, 2003 and Curcicia Ćurčić, S. Tissue transglutaminase (tTG) has been implicated in the pathogenesis of Parkinson disease (PD). However, exactly how tTG modulates the structural and functional properties of α-synuclein (α-syn) and contributes to the pathogenesis of PD remains unknown. Using site-directed mutagenesis combined with detailed biophysical and mass spectrometry analyses, we sought to identify the exact residues involved in tTG-catalyzed cross-linking of wild-type α-syn and α-syn mutants associated with PD. To better understand the structural consequences of each cross-linking reaction, we determined the effect of tTG-catalyzed cross-linking on the oligomerization, fibrillization, and membrane binding of α-syn in vitro. Our findings show that tTG-catalyzed cross-linking of monomeric α-syn involves multiple cross-links (specifically 2-3). We subjected tTG-catalyzed cross-linked monomeric α-syn composed of either wild-type or Gln → Asn mutants to sequential proteolysis by multiple enzymes and peptide mapping by mass spectrometry. Using this approach, we identified the glutamine and lysine residues involved in tTG-catalyzed intramolecular cross-linking of α-syn. These studies demonstrate for the first time that Gln79 and Gln109 serve as the primary tTG reactive sites. Mutating both residues to asparagine abolishes tTG-catalyzed cross-linking of α-syn and tTG-induced inhibition of α-syn fibrillization in vitro. To further elucidate the sequence and structural basis underlying these effects, we identified the lysine residues that form isopeptide bonds with Gln79 and Gln109. This study provides mechanistic insight into the sequence and structural basis of the inhibitory effects of tTG on α-syn fibrillogenesis in vivo, and it sheds light on the potential role of tTG cross-linking on modulating the physiological and pathogenic properties of α-syn.Parkinson disease (PD)2 is a progressive movement disorder that is caused by the loss of dopaminergic neurons in the substantia nigra, the part of the brain responsible for controlling movement. Clinically, PD is manifested in symptoms that include tremors, rigidity, and difficulty in initiating movement (bradykinesia). Pathologically, PD is characterized by the presence of intraneuronal, cytoplasmic inclusions known as Lewy bodies (LB), which are composed primarily of the protein “α-synuclein” (α-syn) (1) and are seen in the post-mortem brains of PD patients with the sporadic or familial forms of the disease (2). α-Syn is a presynaptic protein of 140 residues with a “natively” unfolded structure (3). Three missense point mutations in α-syn (A30P, E46K, and A53T) are associated with the early-onset, dominant, inherited form of PD (4, 5). Moreover, duplication or triplication of the α-syn gene has been linked to the familial form of PD, suggesting that an increase in α-syn expression is sufficient to cause PD. Together, these findings suggest that α-syn plays a central role in the pathogenesis of PD.The molecular and cellular determinants that govern α-syn oligomerization and fibrillogenesis in vivo remain poorly understood. In vitro aggregation studies have shown that the mutations associated with PD (A30P, E46K, and A53T) accelerate α-syn oligomerization, but only E46K and A53T α-syn show higher propensity to fibrillize than wild-type (WT) α-syn (6-8). This suggests that oligomerization, rather than fibrillization, is linked to early-onset familial PD (9). Our understanding of the molecular composition and biochemical state of α-syn in LBs has provided important clues about protein-protein interactions and post-translational modifications that may play a role in modulating oligomerization, fibrillogenesis, and LB formation of the protein. In addition to ubiquitination (10), phosphorylation (11, 12), nitration (13, 14), and C-terminal truncation (15, 16), analysis of post-mortem brain tissues from PD and Lewy bodies in dementia patients has confirmed the colocalization of tissue transglutaminase (tTG)-catalyzed cross-linked α-syn monomers and higher molecular aggregates in LBs within dopaminergic neurons (17, 18). Tissue transglutaminase catalyzes a calcium-dependent transamidating reaction involving glutamine and lysine residues, which results in the formation of a covalent cross-link via ε-(γ-glutamyl) lysine bonds (Fig. 2F). To date, seven different isoforms of tTGs have been reported, of which only tTG2 seems to be expressed in the human brain (19), whereas tTG1 and tTG3 are more abundantly found in stratified squamous epithelia (20). Subsequent immuno-histochemical, colocalization, and immunoprecipitation studies have shown that the levels of tTG and cross-linked α-syn species are increased in the substantia nigra of PD brains (17). These findings, combined with the known role of tTG in cross-linking and stabilizing bimolecular assemblies, led to the hypothesis that tTG plays an important role in the initiation and propagation of α-syn fibril formation and that it contributes to fibril stability in LBs. This hypothesis was initially supported by in vitro studies demonstrating that tTG catalyzes the polymerization of the α-syn-derived non-amyloid component (NAC) peptide via intermolecular covalent cross-linking of residues Gln79 and Lys80 (21) and by other studies suggesting that tTG promotes the fibrillization of amyloidogenic proteins implicated in the pathogenesis of other neurodegenerative diseases such as Alzheimer disease, supranuclear palsy, Huntington disease, and other polyglutamine diseases (22-24). However, recent in vitro studies with full-length α-syn have shown that tTG catalyzes intramolecular cross-linking of monomeric α-syn and inhibits, rather than promotes, its fibrillization in vitro (25, 26). F, schematic depiction of the tTG-catalyzed chemical reaction (isodipeptide formation) between glutamine and lysine residues.In this study, we have identified the primary glutamine and lysine residues involved in tTG-catalyzed, intramolecularly cross-linked monomeric α-syn and investigated how cross-linking these residues affects the oligomerization, fibrillization, and membrane binding of α-syn in vitro. Using single-site mutagenesis and mass spectrometry applied to exhaustive proteolytic digests of native and cross-linked monomeric α-syn, we identified Gln109 and Gln79 as the major tTG substrates. We demonstrate that the altered electrophoretic mobility of the intramolecularly cross-linked α-syn in SDS-PAGE occurs as a result of tTG-catalyzed cross-linking of Gln109 to lysine residues in the N terminus of α-syn, which leads to the formation of more compact monomers. Consistent with previous studies, we show that intramolecularly cross-linked α-syn forms off-pathway oligomers that are distinct from those formed by the wild-type protein and that do not convert to fibrils within the time scale of our experiments (3-5 days). We also show that membrane-bound α-syn is a substrate of tTG and that intramolecular cross-linking does not interfere with the ability of monomeric α-syn to adopt an α-helical conformation upon binding to synthetic membranes. These studies provide novel mechanistic insight into the sequence and structural basis of events that allow tTG to inhibit α-syn fibrillogenesis, and they shed light on the potential role of tTG-catalyzed cross-linking in modulating the physiological and pathogenic properties of α-syn.  相似文献

Twelve separate outbreaks of confirmed cowpox, 10 involving humans, were reviewed. Six of the patients, including three children, had severe infections and five were admitted to hospital. In three outbreaks both people and cows were affected but it was not known how the infections entered the herds. In seven outbreaks no direct contact with cattle was established and clinical and serological examination failed to show evidence of cowpox in the bovine population. Comparison of these data with information about infections known to be enzootic in cattle leads to the suggestion that cows are not the natural reservoir of cowpox. This should be remembered when diagnosis is considered. The role of small wild animals as hosts and vectors of "cowpox" should be investigated.  相似文献

Herves is a functional Class II transposable element in Anopheles gambiae belonging to the hAT superfamily of elements. Class II transposable elements are used as gene vectors in this species and are also being considered as genetic drive agents for spreading desirable genes through natural populations as part of an effort to control malaria transmission. In this study, Herves was investigated in populations of Anopheles gambiae s.s., Anopheles arabiensis and Anopheles merus in Mozambique over a period of 2 years. The copy number of Herves within these three species was approximately 5 copies per diploid genome and did not differ among species or between years. Based on the insertion-site occupancy-frequency distribution and existing models of transposable element dynamics, Herves appears to be transpositionally active currently or, at least recently, in all species tested. Ninety-five percent of the individuals within the populations of the three species tested contained intact elements with complete Herves transposase genes and this is consistent with the idea that these elements are currently active.  相似文献
