高原鼠兔是新开发的实验动物,能很好地适应高原低氧环境。本研究报道了驯化高原鼠兔的部分血液指标,并与野生高原鼠兔及部分其它实验动物和人的相应指标作了比较。同时对高原鼠兔能适应高原低氧环境的机理作了探讨。高原鼠兔红细胞数量高、体积小、红细胞膜表面积大,从而提高了Hb的利用率,使尽可能多的Hb参与摄氧和释氧。  相似文献

高原鼠兔(Ochotona curzoniae)是生活在青藏高原海拔3000-5000m地区的特有物种,具有极强的低温、低氧耐受能力。近十几年来,许多国内外学者从整体水平及分子水平对高原鼠兔的低氧适应机制进行了大量研究,认为该动物是研究低氧适应的理想动物模型。本文对高原鼠兔的低氧适应机制从血液学特征、肺血管的结构和功能及分子生物学研究等方面作一系统阐述,旨在阐明高原土生动物在高寒缺氧环境中生存的适应机制,这对人类适应高原及高原疾病的防治有着重要的指导意义。  相似文献

高原鼢鼠和高原鼠兔是青藏高原特有的物种,对高原低氧环境有很强的适应性。本研究测定了不同海拔的高原鼢鼠和高原鼠兔的红细胞数目、血红蛋白浓度、平均红细胞压积、平均红细胞体积等;通过质谱测序明确两种高原动物血红蛋白亚型;采用PAML4.8程序分析两种动物血红蛋白亚基中的正向选择位点;采用同源建模的方法分析正向选择位点对血红蛋白氧亲和力的影响,基于对两种高原动物血液特征的分析,探讨了它们适应低氧的策略。结果表明,随着海拔升高,高原鼢鼠通过增加红细胞数目、减小红细胞体积应答低氧,相反,高原鼠兔通过减少红细胞数目、增大红细胞体积应答低氧;高原鼠兔红细胞中具有α2β2成年型血红蛋白和α2ε2胎儿型血红蛋白,高原鼢鼠红细胞中只有α2β2成年型血红蛋白,但高原鼢鼠血红蛋白氧亲和力和别构效应显著高于高原鼠兔;高原鼢鼠与高原鼠兔的血红蛋白α和β亚基中,正向选择的氨基酸位点数目、位置以及它们的侧链基团极性和取向具有明显不同,这可能是造成两种动物血红蛋白氧亲和力不同的重要原因。总之...  相似文献

目的:克隆青藏高原高原鼠兔Na ,K -ATP酶β2亚基(ATP1B2)的基因编码区,并分析其序列特征,以揭示高原鼠兔低氧适应的分子基础。方法:采用RT-PCR技术从高原鼠兔脑组织中扩增出ATP1B2基因编码区cDNA序列并进行序列测定,采用生物信息学技术对其进行分析。结果:ATP1B2基因编码区由873bp组成,编码290个氨基酸残基。序列分析结果显示,高原鼠兔ATP1B2编码区的核酸序列与兔、人、牛、大鼠、小鼠及狗分别有99%、93%、91%、91%、90%和90%的同源性。结论:克隆出青藏高原高原鼠兔ATP1B2基因编码区,为进一步了解高原鼠兔低氧适应的分子机制提供了基础。  相似文献

低氧诱导因子—1(HIF—1)是一种由低氧诱导因子—1α(HIF—1α)和芳香烃核受体转录因子(ARNT)组成的二聚体转录因子,在生物体氧平衡调节中起关键作用。高原鼠兔(Dchotorua curzoniae)是生活在青藏高原海拔3000m以上区域,有着极强的低氧、低温耐受能力的特有自然种群。它通过高的基础代谢率和高的氧利用率来适应高原低温、缺氧环境。将高原鼠兔带到北京,常氧室温条件下适应7d。RT—PCR扩增高原鼠兔HIF—1α cDNA片断,^32P标记作为特异性探针,Northern杂交检测了常氧条件下高原鼠兔HIF—1α的组织特异性表达。结果显示,HIF—1α mRNA在高原鼠兔多种组织中均有表达,并且表现出明显的组织差异性。其中脑的表达量最大,肾其次,心、脾和肝的表达相对较少。  相似文献

低氧性肺血管收缩反应钝化是高原鼠兔适应低氧环境的重要策略,但参与该生理代偿反应的功能基因尚不明确。间隙连接蛋白40 (Connexin40, Cx40) 在哺乳动物肺血管内皮表达。本研究对生活在海拔3 200 m的高原鼠兔进行28 d模拟海拔5 000 m低氧处理,以Sprague Dawley (SD) 大鼠为对照,采用免疫组化法分析模拟低氧处理后高原鼠兔和SD大鼠肺组织形态结构,qPCR和蛋白印记法检测Cx40基因和蛋白表达量变化,探究Cx40在高原鼠兔低氧性肺血管收缩反应钝化中的潜在作用。结果显示,低氧处理后,高原鼠兔肺泡呈空泡囊状,Cx40蛋白在支气管和肺血管中均表达,Cx40 基因mRNA水平随着低氧暴露而升高,但其蛋白质水平呈下降趋势,肺支气管Cx40蛋白无明显变化。SD大鼠肺血管和肺支气管表达的Cx40蛋白均无明显变化。暗示生活在高海拔低氧环境中的高原鼠兔,Cx40蛋白下调可抑制血管收缩信号,减弱低氧性肺血管收缩反应,使低氧性肺血管收缩反应钝化,以适应高原缺氧环境。研究结果可为高原土著动物适应高寒缺氧环境提供基础理论数据。  相似文献

本实验用氧电极法测定在急性及慢性低氧条件下,S.D大白鼠及高原鼠兔(Ochotona curzoniae)肝线粒体的氧化磷酸化效率及呼吸控制率之变化。结果表明:大白鼠和高原鼠兔经24小时急性低氧暴露后,其氧化磷酸化效率无明显变化;呼吸控制率在高原鼠兔中无明显变化,而在大白鼠中却有明显增加,这种增加是由于呼吸状态Ⅳ呼吸速度降低而引起的。经25天的慢性低氧暴露后,大白鼠和高原鼠兔的氧化磷酸化效率仍无明显变化,但大白鼠在海拔7000米时的呼吸控制率则有明显下降,而高原鼠兔却明显增加。实验结果提示:在细胞呼吸水平方面,高原鼠兔对低氧的耐受力明显高于大鼠。  相似文献

In normal eyes, the amplitude of the b-wave of the photopic ERGs increases during light adaptation, but the mechanism causing this increase has not been fully determined. The purpose of this study was to evaluate the contribution of receptoral and post-receptoral components of the retina to this phenomenon. To accomplish this, we examined the ERGs during light adaptation in Pikachurin null-mutant (Pika -/-) mice, which have a misalignment of the bipolar cell dendritic tips to the photoreceptor ribbon synapses. After dark-adaptation, photopic ERGs were recorded from Pika -/- and wild type (WT) mice during the first 9 minutes of light adaptation. In some of the mice, post-receptoral components were blocked pharmacologically. The photopic b-waves of WT mice increased by 50% during the 9 min of light adaptation as previously reported. On the other hand, the b-waves of the Pika -/- mice decreased by 20% during the same time period. After blocking post-receptoral components, the b-waves were abolished from the WT mice, and the ERGs resembled those of the Pika -/- mice. The extracted post-receptoral component increased during light adaptation in the WT mice, but decreased for the first 3 minutes to a plateau in Pika -/- mice. We conclude that the normal synaptic connection between photoreceptor and retinal ON bipolar cells, which is controlled by pikachurin, is required for the ERGs to increase during light-adaptation. The contributions of post-receptoral components are essential for the photopic b-wave increase during the light adaptation.  相似文献

A collaborative study was conducted to investigate the teratological susceptibility of the Pika (Ochotona rufescens rufescens) to selected teratogenic chemicals: cyclophosphamide, 6-mercaptopurine, 5-fluorouracil, 6-aminonicotinamide, actinomycin D, ethylurethan, ampicillin, tetracycline, thalidomide, diphenylhydantoin, hypervitaminosis A, aspirin, dexamethasone, betamethasone and bredinin. Some of the chemicals were shown to be teratogenic in the Pika, but this animal was generally more resistant to their teratogenicity than the rabbit and rodents. In the Pika, thalidomide did not induce any typical limb defects, which have been produced in the rabbit. Pikas reproduce well and appear to have no substantial disadvantages as an animal species for teratological studies. Thus, the Pika may be useful as a new non-rodent species for teratological testings.  相似文献

Leptin, an adipocyte-derived hormone, plays an important role in body energy homeostasis. Plateau pika (Ochotona curzoniae), an endemic and keystone species living only at 3000-5000 m above sea level on Qinghai-Tibet Plateau, is a typically high hypoxia and low temperature tolerant mammal with high resting metabolic rate (RMR), non-shivering thermogenesis (NST), and high ratio of oxygen utilization to cope with harsh plateau environment. To explore the molecular mechanism of ecological acclimation in plateau pika, we first cloned pika leptin cDNA and compared its mRNA expression in different altitudes (3200 and 3900 m) using real-time RT-PCR (Taqman probe) technology. The full-length pika leptin cDNA was 3015 with 504 bp open-reading frame encoding the precursor peptide of 167 amino acids including 21 residues of signal peptide. Pika leptin was 70-72% homologous to that of other species and was of similarly structural characteristics with other species. The pika-specific genetic diversity in leptin sequence occurred at twenty sites. With the increase in altitude, there were larger fat store and high level of ob gene expression in plateau pika. Our results indicated that leptin is sensitive to cold and hypoxia plateau environment and may play one of important roles in pika's ecological adaptation to harsh plateau environment.  相似文献

Pikas originated in Asia and are small lagomorphs native to cold climates. The plateau pika, Ochotona curzoniae is a keystone species on the Qinghai-Tibet Plateau and an ideal animal model for hypoxic adaptation studies. Altered mitochondrial function, especially cytochrome c oxidase activity, is an important factor in modulation of energy generation and expenditure during cold and hypoxia adaptation. In this study, we determined the complete nucleotide sequence of the O. curzoniae mitochondrial genome. The plateau pika mitochondrial DNA is 17,131bp long and encodes the complete set of 37 proteins typical for vertebrates. Phylogenetic analysis based on concatenated heavy-strand encoded protein-coding genes revealed that pikas are closer to rabbit and hare than to rat. This suggests that rabbit or hare would be a good control animal for pikas in cold and hypoxia adaptation studies. Fifteen novel mitochondrial DNA-encoded amino acid changes were identified in the pikas, including three in the subunits of cytochrome c oxidase. These amino acid substitutions potentially function in modulation of mitochondrial complexes and electron transport efficiency during cold and hypoxia adaptation.  相似文献

高原鼠兔(Ochotona curzoniae)是青藏高原的特有动物和关键种。本实验应用ISSR分子标记和细胞色素b(Cytochrome b,Cytb)基因序列分析了雅鲁藏布江两岸高原鼠兔4个种群的遗传多样性和遗传分化。结果显示,两种标记所得到的4个种群的遗传多样性都较高,并以Cytb基因为指标的遗传多样性水平在种群间体现出较大差异。在ISSR标记中,分子方差分析(AMOVA)表明种群间的遗传变异为13.50%,种群分化较低且UPGMA聚类时江北岸与南岸的种群有交叉;而在Cytb基因中遗传变异主要发生在种群间,占79.24%,种群间存在着显著的遗传分化,且江北岸和南岸的两个种群分别聚为一类。研究结果表明,mtDNA基因在反映种群遗传多样性和遗传分化上较ISSR分子标记相对具有优势。  相似文献

2009年3~7月,在西藏自治区墨竹工卡县日多乡念村(29o46′N,92o19′~92o20′E,海拔4 423~5 015 m),采用目标动物抽样法对高原鼠兔(Ochotona curzoniae)的警戒行为和取食行为进行了观察,利用广义线性模型(GLM)的泊松回归模型分析了不同土地利用、植被覆盖度、植物类别和植被优势种对高原鼠兔行为影响的差异。研究结果表明,雌性高原鼠兔取食行为频率高于雄性(β=﹣0.203,SE=0.096,P0.05),警戒行为频率低于雄性(β=0.199,SE=0.088,P0.05)。高原鼠兔的取食行为(β=﹣0.009,SE=0.004,P0.05)随着栖息地内双子叶植物覆盖比例增高而呈递减趋势,相应地,随着栖息地内单子叶植物覆盖比例的增高呈现递增趋势(β=0.009,SE=0.004,P0.05)。高原鼠兔的警戒行为在放牧地(β=0.273,SE=0.131,P0.05)以及植被覆盖度高的栖息地内(β=0.007,SE=0.003,P0.05)均呈递增趋势。随着薹草属植物覆盖度比例增高,高原鼠兔的取食行为(β=0.023,SE=0.006,P0.001)呈现递增趋势,而警戒行为呈递减趋势(β=﹣0.018,SE=0.007,P0.05)。  相似文献

Small mammals mediate trajectories of vegetation change where both their density and the growing season are moderated by temperature and precipitation. On the Tibetan Plateau, the cold and arid climate particularly restricts the plant growing season, but the role of mammals’ density and climate in moderating small mammal populations remains unknown. We conducted a 5-year mark-recapture study of plateau pikas (Ochotona curzoniae) to test the relative importance of density-dependent and climatic factors on survival and reproduction. Plateau pikas had seasonal fluctuations in density and survival. During the warm summer season (May–August), monthly survival was density-independent, ranging from 74.7 to 90.4%, but varied with sex and age, increased with precipitation and NDVI, and decreased with temperature. During the cold season (September–April), monthly survival was around 98%. Density and precipitation had negative effects on reproductive success for the first and second litters of the year, and temperature showed consistently positive effects on reproductive success for both litters of the year. Pika density and climate regulated together the population dynamics of plateau pikas. These data on the relationships between density and climatic factors on survival and reproduction are critical for the management and conservation of plateau pikas on the Tibetan Plateau.  相似文献

We studied the diversity of all forms of the RuBisCO large subunit-encoding gene cbbL in three RuBisCO uncharacterized hydrothermal vent communities. This diversity included the archaeal cbbL and the forms IC and ID, which have not previously been studied in the deep-sea environment, in addition to the forms IA, IB and II. Vent plume sites were Fryer and Pika in the Mariana arc and the Suiyo Seamount, Izu-Bonin, Japan. The cbbL forms were PCR amplified from plume bulk microbial DNA and then cloned and sequenced. Archaeal cbbL was detected in the Mariana samples only. Both forms IA and II were amplified from all samples, while the form IC was amplified only from the Pika and Suiyo samples. Only the Suiyo sample showed amplification of the form ID. The form IB was not recorded in any sample. Based on rarefaction analysis, nucleotide diversity and average pairwise difference, the archaeal cbbL was the most diverse form in Mariana samples, while the bacterial form IA was the most diverse form in the Suiyo sample. Also, the Pika sample harbored the highest diversity of cbbL phylogenetic lineages. Based on pairwise reciprocal library comparisons, the Fryer and Pika archaeal cbbL libraries showed the most significant difference, while Pika and Suiyo showed the highest similarity for forms IA and II libraries. This suggested that the Fryer supported the most divergent sequences. All archaeal cbbL sequences formed unique phylogenetic lineages within the branches of anaerobic thermophilic archaea of the genera Pyrococcus, Archaeoglobus, and Methanococcus. The other cbbL forms formed novel phylogenetic clusters distinct from any recorded previously in other deep-sea habitats. This is the first evidence for the diversity of archaeal cbbL in environmental samples.  相似文献

随着统计模型及空间信息数据的不断发展和完善,物种分布模型已经成为全球变化背景下研究大尺度物种分布情况的重要工具。高原鼠兔(Ochotona curzoniae)是青藏高原特有的关键物种,在青藏高原生态系统中占有重要地位。通过采集高原鼠兔的分布点数据及环境变量数据,基于R语言中BIOMOD包中的7个模型对其在青海湖流域的分布进行了模拟。结果表明,高原鼠兔主要分布于青海湖西岸和北岸、天峻县周边及布哈河流域上游,影响高原鼠兔分布的主要环境因子为距道路距离、距居民点距离、最暖月最高气温、NDVI标准差、最冷季和最干季降水量。BIOMOD组合模型中,推进式回归树模型(GBM)和最大熵模型(MAXENT)的模拟效果最好,广义线性回归模型(GLM)结果较差。而优化后的结果显示,模拟结果的集成和筛选能有效提高模型的精度和效果。  相似文献
