Advantages and disadvantages in using functional male sterility (positional sterile — ps, positional sterile 2 — ps 2, and excerted stigma — ex) in tomato hybrid seed production and attempts to elaborate systems for their more efficacious use in breeding were discussed in this review. It was concluded that the application of one of these types of sterility, (ps 2) in practice, although in a limited number of countries, showed the functional male sterility in tomato was a potential not to be underestimated in developing approaches that aimed at reducting the time and cost associated with hybrid seed production.  相似文献

Fruit set and fruit development in tomato is largely affected by changes in environmental conditions, therefore autonomous fruit set independent of fertilization is a highly desirable trait in tomato. Here, we report the production and characterization of male‐sterile transgenic plants that produce parthenocarpic fruits in two tomato cultivars (Micro‐Tom and Moneymaker). We generated male‐sterility using the cytotoxic gene barnase targeted to the anthers with the PsEND1 anther‐specific promoter. The ovaries of these plants grew in the absence of fertilization producing seedless, parthenocarpic fruits. Early anther ablation is essential to trigger the developing of the transgenic ovaries into fruits, in the absence of the signals usually generated during pollination and fertilization. Ovaries are fully functional and can be manually pollinated to obtain seeds. The transgenic plants obtained in the commercial cultivar Moneymaker show that the parthenocarpic development of the fruit does not have negative consequences in fruit quality. Throughout metabolomic analyses of the tomato fruits, we have identified two elite lines which showed increased levels of several health promoting metabolites and volatile compounds. Thus, early anther ablation can be considered a useful tool to promote fruit set and to obtain seedless and good quality fruits in tomato plants. Vertical block SDS-PAAG electrophoresis of individual tomato seeds was used to examine the potential of seed proteins as gene markers for distinguishing male sterile from fertile lines and cultivars. Seeds of three genic male sterile lines and four fertile cultivars and lines were investigated. The line 6944 is carrier of the gene ms 10 35for pollen male sterility and the lines P-1a and PPS-3 carry the gene ps-2 for positional sterility. A modified extract solution which includes 1M tris — buffer pH 7.0, 7 % SDS, dimethylformamide and distilled water was applied. Qualitative variation in the A protein locus which could be used as a marker for distinguishing male sterile from fertile tomato lines was established.  相似文献

We report a novel cytoplasmic male sterility (CMS) system in Brassica juncea (oilseed mustard) which could be used for production of hybrid seed in the crop. A male sterile plant identified in a microspore derived doubled haploid population of re-synthesized B. napus line ISN 706 was found to be a CMS as the trait was inherited from the female parent. This CMS, designated ‘126-1’, was subsequently transferred to ten different B. juncea varieties and lines through inter-specific crosses followed by recurrent backcrossing. The F1s of inter-specific crosses were invariably partially fertile, but irrespective of the variety/line used, the recipient lines became progressively male sterile over five to seven generations and could be maintained by crossing the male sterile lines with their normal counterparts. The male sterile lines were found to be stable for the trait under both long and short day conditions. CMS lines when crossed with lines other than the respective maintainer line were restored for fertility, implying that any variety could act as a restorer for ‘126-1’ cytoplasm in B. juncea. These unique features in maintenance and restoration of CMS lines coupled with near normal floral morphology of the CMS lines have allowed the use of ‘126-1’ cytoplasm for hybrid seed production. The uniqueness of ‘126-1’ has been further established by Southern hybridization with mitochondrial DNA probes and by a histological study of the development of male sterile anthers.  相似文献

Incorporating male sterility into hybrid seed production reduces its cost and ensures high varietal purity. Despite these advantages, male‐sterile lines have not been widely used to produce tomato (Solanum lycopersicum) hybrid seeds. We describe the development of a biotechnology‐based breeding platform that utilized genic male sterility to produce hybrid seeds. In this platform, we generated a novel male‐sterile tomato line by clustered regularly interspaced short palindromic repeat (CRISPR)/CRISPR‐associated protein 9 (Cas9)‐mediated mutagenesis of a stamen‐specific gene SlSTR1 and devised a transgenic maintainer by transforming male‐sterile plants with a fertility‐restoration gene linked to a seedling‐colour gene. Offspring of crosses between a hemizygous maintainer and the homozygous male‐sterile plant segregated into 50% non‐transgenic male‐sterile plants and 50% male‐fertile maintainer plants, which could be easily distinguished by seedling colour. This system has great practical potential for hybrid seed breeding and production as it overcomes the problems intrinsic to other male‐sterility systems and can be easily adapted for a range of tomato cultivars and diverse vegetable crops.  相似文献

Somatic fusion hybrids between the diploid potato and tomato were backcrossed to several genotypes of potato. Two ploidy levels of fusion hybrids, 4x and 6x, were used as female parents in backcrosses with five clones of 4x-potato. An estimate of the berry set and seed set in immature berries harvested 14–21 days after pollination indicated that crosses between certain combinations of 6x-fusion hybrids and male parents were more successful than others. The culture of over 4000 young seeds from berries harvested 2–2.5 weeks after pollination gave rise to a single seedling, 93.6701, from the cross between the 6x-fusion hybrid C 31-17-1 and the 4x-potato AM 66.42. This seedling was found to possess a pentaploid chromosome number, which was expected of a 6x × 4x cross. Isozyme analysis and DNA hybridisation studies confirmed that the seedling 93.6701 was indeed a backcross (BC1) progeny. Morphologically, this BC1 plant resembled potato with respect to plant habit, leaf shape, stolons and tuber characteristics, while some of the characters, such as floral morphology and the fragrance of the crushed leaves (typical of tomato), were intermediate. It was male sterile but could be successfully hybridized with 4x-potato through in vitro culture of yound seeds; thus, BC2 plants were obtained. The possibilities of backcrossing and the potential use of BC1 and BC2 plants in genetics and breeding are discussed.  相似文献

Production of hybrid seeds and pursuing heterosis breeding of many crops have been accomplished using male sterile lines. However, not all crops have valuable male sterile lines due to instability of male sterility and absence of a restorer system. In this study, male sterile lines have been induced using a two-component system. The extracellular ribonuclease Barnase was cleaves into two inactive yet complementary fragments, designated as ??Bn-5?? and ??Bn-3??. Both components were controlled by a TA29 promoter. They were transferred into the tomato inbred line ??Yellow tomato?? by Agrobacterium method. Southern blotting identified that 11 transgenic Bn-5 plants (T0) and 10 transgenic Bn-3 plants (T0) were obtained. The vegetative phenotypes of all T0 plants were similar to wild-type, and they were capable of producing viable pollen grains and normal fruit with seeds, indicating that Barnase had lost its function after it being split two partial fragments. After self-pollination, homozygous progenies (T1) of transgenic Bn-5 and Bn-3 plants were chosen to cross each other, Barnase could be reconstituted and co-expressed in the same cell, which caused the hybrid plants to produce collapsed pollen grains with no viability and thus100?% male sterile plants were obtained. Stamens of male sterile plants were shorter than those of the wild type plants. PCR detection demonstrated that all male sterile plants contained Barnase, but male fertile plants did not. The male sterile plants were crossed with the male fertile inbred lines, and the result showed that hybrid (F1) plants were capable of producing normal fruit with seeds, and their pollen grain fertility was restored. The co-segregation ratio of Bn-5 and Bn-3 fragments showed 1:1 among hybrid plants. In conclusion, the results verified that the male sterility could be generated by two component system and be used in hybrid seed production. The F1 between the male sterile plant and the inbred line showed heterotic comparing to both parents. This system needs not breed restoration line.  相似文献

Brassica plants, with a combination of chloroplasts encodingtriazine resistance and mitochondria encoding cytoplasmic malesterility, offer new opportunities for hybrid seed production.Such plants can be grown as a random mixture with a male parent,thereby allowing more effective pollen transfer for greaterefficiency of hybrid seed production. Harvested seed consistsof hybrid seed from the ‘female’ parent, which istriazine-resistant, and non-hybrid, triazine-sensitive seedresulting from self pollination of the ‘male’ parent. This study demonstrates that triazine-sensitive broccoli (BrassicaoleraceaL. var.italica) progeny can easily be eliminated byallowing the seed to imbibe a solution of 10 g l-1atrazine overnightthen drying-back the seed for subsequent germination. Such atreatment prevents triazine-sensitive broccoli from developingbeyond the cotyledon stage following germination, and remainseffective for at least 3.5 years after seed treatment. Both the pollination control system and genetic distance are major factors in the utilization of crop heterosis. The recessive genic male sterile line (RGMS) 7-7365A (Bnms3ms3ms4ms4) has been widely applied to hybrid seed production because it can generate a completely male sterile population by crossing with the 7-7365C temporary line (Bnms3ms3rfrf). In this study, the sterile genes of 7-7365A were transferred to the new Brassica napus lines 7-749 and 7-750 with a high content of subgenomes by backcross breeding. We used the amplified fragment length polymorphism (AFLP) technique combined with bulk segregant analysis (BSA) to identify markers linked to the BnMs4 gene. Twelve AFLP markers linked to the BnMs4 gene were identified. Of them, SA06MG09 and P08MG16 were the closest makers, which were on either side of the gene at a distance of 0.9 and 0.8?cM, respectively. Twenty AFLP primer combinations were used to screen the F2, BC1F3, and BC2F4 populations from the breeding program, and the markers linked to the BnMs3 and BnMs4 genes were used to screen the BC2F4 populations. As a result, we obtained two types of improved sterile lines, 7-749A and 7-750A, and their indexes of subgenomic components (ISG) were 44.2?C49.8 and 20.2?C26.6%, respectively. The combining ability analyses of seed yield character were conducted in the crosses from the three sterile lines and ten restorers within a random block design in three environments for two successive years. The general combining ability (GCA) of the two improved sterile lines were significantly higher than the GCA of 7-7365A in every environment tested. The two improved sterile lines had stability in seed yield, and they will be used in the future for hybrid seed production.  相似文献

The role of endogenous gibberellin (GA) in seed and fruit development was studied with the use of the GA-deficient ga-1 mutant of tomato ( Lycopersicon esculentum Mill. cv. Moneymaker). Flowers of the ga-1 mutant were abnormal and sterile, but parthenocarpic fruit development was observed occasionally on the dwarf plants. A single application of GA4+7 restored the fertility of the mutant flowers and resulted in seed set. Development of GA-producing and GA-deficient seeds in GA-deficient fruits was compared by pollination of ga-1/ga-1 flowers with wild-type or ga-1 pollen, respectively. In ga-1/ga-1 seed dehydration started about 1 week earlier than in Ga-1/ga-1 seeds. Ultimate fresh and dry weights of mature Ga-1/ga-1 seeds were higher than those of ga-1/ga-1 seeds and showed negative correlations with the total number of seeds per fruit. Total content and composition of seed proteins were not influenced by the GA-deficiency. Germination of the mature seeds depended on embryonal GA synthesis and was not influenced by maternal GA production. Final fresh weight of the ga-1/ga-1 fruits was positively correlated with the number of seeds per fruit. In these fruits, the minimum number of seeds for growth above the parthenocarpic level was about 10 or 35 in the presence of Ga-1/ga-1 or ga-1/ga-1 seeds, respectively. Fruits containing GA-producing seeds reached a higher fresh weight than those containing GA-deficient seeds, and their ripening was delayed by one week. It is concluded that gibberellin is indispensable for the development of fertile flowers and for seed germination, but only promoting in later stages of fruit and seed development.  相似文献

To characterize the phenomenon of natural parthenocarpy in tomato ( Lycopersicon esculentum Mill.) two different approaches have been followed. At a developmental level, the ovary weights of three non-parthenocarpic lines and three near-isogenic parthenocarpic ( pat-2 ) lines were compared. Four developmental stages were considered: flower bud, preanthesis, anthesis and 4 days after anthesis. The parthenocarpic lines displayed ovary weights higher than their respective non-parthenocarpic lines from preanthesis to 4 days after anthesis. A molecular approach involved comparison of in vitro translation products from flower RNAs taken from the same developmental stages of non-parthenocarpic and near-isogenic parthenocarpic ( pat-2 and pat-3/pat-4 ) lines. Analysis by two-dimensional polyacrylamide gel electrophoresis showed the differential expression of a 30-kDa product in parthenocarpic materials from preanthesis to anthesis. These results suggest that the physiological and molecular events responsible for parthenocarpy begin at the preanthesis stage, before the flower is completely mature and receptive to pollination. The differential expression of this in vitro translation product in pat-2 and pat-3/pat-4 genotypes also suggests a common or confluent molecular basis in genetically controlled parthenocarpy.  相似文献

The functional male sterility controlled by ps gene proved to be a useful tool in hybrid tomato varieties breeding in Poland. The climat conditions such as excessive temperature and high humidity have a bad effect on the expression and stability of functional male sterility. Using the RAPD methods we have identified two RAPD markers linked to the ps gene. The markers OPW 131230 and OPAX 10780 were generated by 5′CACAGCGACA 3′ and 5′CCAGGCTGAC 3′ decamers respectively in F2 population of combination 24/29 × G-1.  相似文献

Wild abortive cytoplasmic male sterility has been extensively used in hybrid seed production in the tropics. Using protoplast fusion between cytoplasmic male sterile and fertile maintainer lines; we report here, transfer of wild abortive cytoplasmic male sterility to the nuclear background of RCPL1-2C, an advance breeding line which also served as maintainer of this cytoplasm. In total, 27 putative cybrids between V20A and RCPL1-2C and 23 lines between V20A and V20B were recovered and all of them were sterile. DNA blots prepared from the mitochondrial DNA of the cybrid lines from both the sets were probed with orf155 that is known to exhibit polymorphism between the mitochondrial DNA of the male-sterile and fertile maintainer lines. Hybridization of orf155 to 1.3 kb HindIII-digested mitochondrial DNA fragment of the cybrids showed transfer of mitochondrial DNA from wild abortive cytoplasmic male-sterile line to the maintainers, viz. RCPL 1-2C and V20B. Expression of male sterility was confirmed by the presence of sterile pollen grains and the lack of seed setting due to selfing in all the cybrid lines. These cybrids, on crossing with respective fertile maintainers set seeds that in turn, produced sterile BC1 plants. DNA blots from HindIII-digested mitochondrial DNA of these BC1 plants when probed with orf155 again exhibited localization of orf155 in wild abortive cytoplasm-specific 1.3 kb HindIII-digested mitochondrial DNA fragments. This demonstrated that the cytoplasmic male sterility transferred through protoplast fusion retained intact female fertility and was inherited and expressed in BC1 plants. Fusion-derived CMS lines, on pollination with pollen grains from restorer, showed restoration of fertility in all the lines. The results demonstrate that protoplasts fusion can be used for transferring maternally inherited traits like cytoplasmic male sterility to the desired nuclear background which can, in turn, be used in hybrid seed production programme of rice in the tropical world.  相似文献

A series of experiments is described in which various growthregulators were applied in lanolin emulsions to the flowersof two pistillate varieties of strawberry, in an attempt topromote parthenocarpic development of their receptacles. Parthenocarpic development was obtained in a large majorityof the treated flowers over a range of concentrations, varyingfrom 500 p.p.m. to up to 6,000 p.p.m., after treatment withthe following growth regulators; 4-(indole-3-) butyric acid,2-naphthoxyacetic acid, and 1-naphthaleneacetic acid. Gibberellic acid promoted growth of the ‘neck’ regionin Freya and increased the response of this variety to 4-(indole-3-)butyric acid. It also reduced the period of development of thefruit from anthesis till ripening. An attempt to promote parthenocarpic receptacle developmentwith 4-(indole-3-) butyric acid after the removal of all carpelsbefore pollination was unsuccessful. It was concluded that theactive growth substances probably stimulated growth initiallyin the tissues of the unpollinated carpels, which in turn promotedthe development of the receptacle.  相似文献

The paper brings data set of seed reproduction of about 500 wild herbaceous species of Central Europe and presents a number of produced seeds in a new way. Number of seeds — reproductive capacity of a population (RCP) was defined as a number of seeds produced by one species per 1 m2 at its one-hundred-per-cent cover per one season. About 23% of seed weight variability and about 26% of variability in RCP were explainable on a family level. The trade-off between RCP and seed weight was confirmed both within families and also among families. Both characteristics had higher variability on a family level and on lower taxonomic levels than on subclass and class levels. Species with an annual life cycle have a larger number of seeds than perennials. Geophytes and species without lateral spread have a tendency to produce a large amount of relatively weighty seeds in comparison to other life forms, and to species with vegetative lateral spread. Species with seeds dispersed by wind usually have a large number of lightweight seeds. Multivariate analyses confirmed some tendencies of reproductive traits which correspond to the definition of C-, S-, R- strategies by Grime — “verification” of the RCP as ecological trait. The R- strategy was well distinguished from the C- and S- strategies by reproductive traits, whereas C- and S- strategies are very similar to each other. Species with insect pollination agent often correspond to C- strategy, wind agent to S- strategy, and species with self-pollination to R- strategy.  相似文献

Results from creation and comparative test of the new hybrid tomato cultivars are presented: Jar and Dar — semi-determinate suitable for early production in plastic hothouses and in the field; Viki, Asja and Lorin — indeterminate suitable for middle-early field production. Jar and Dar excel the standard in earliness and fruit size. Seed production of Dar, Viki and Asja is realized on a male sterile base (ps-2) and genetic morphological marker (ah) that makes easier the flower emasculation, increases the hybrid seeds purity and precipitates their realization the whole year because of exclusion of the necessity of carrying out a ground test. Above mentioned tomato cultivars are recognized for a spread in all regions of Bulgaria.  相似文献
