Recent studies in two species of Drosophila have demonstrated a negative effect of parental age on offspring fitness, including a reduced hatch rate of eggs and larval-to-adult viability. This has led to a call to consider the decline of offspring quality as a function of parental age in theoretical considerations of the evolution of ageing. We have tested whether a decline in egg and larval quality of older mothers is a general feature of senescence by examining it in the cockroach Nauphoeta cinerea. We also tested whether maternal age affected the reproductive potential of daughters. Although maternal age at first reproduction profoundly affected maternal fitness, there was no difference in hatch rate or larval viability between the offspring of young and old mothers. Likewise, the reproductive potential of the daughters of young and old mothers was the same. Thus, while maternal age effects may be important aspects of ageing in some systems, the generality and overall importance for theories of ageing remain unclear.  相似文献

Egg quality in fish: what makes a good egg?   总被引:15,自引:0,他引:15
Factors affecting egg quality are determined by the intrinsic properties of the egg itself and the environment in which the egg is fertilized and subsequently incubated. Egg quality in fish is very variable. Some of the factors affecting egg quality in fish are known, but many (probably most) are unknown. Components that do affect egg quality include the endocrine status of the female during the growth of the oocyte in the ovary, the diet of the broodfish, the complement of nutrients deposited into the oocyte, and the physiochemical conditions of the water in which the eggs are subsequently incubated. In captive broodfish, the husbandry practices to which fish are subjected are probably a major contributory factor affecting egg quality. Our knowledge of the genetic influences on egg quality is very limited indeed. We know that parental genes strongly influence both fecundity and egg quality, but almost nothing is known about gene expression and/or mRNA translation in fish oocytes/embryos. This is surprising because the products synthesized in ovoand the mechanisms controlling their expression are likely to play a central role in determining egg quality. The genetic mechanisms underpinning oocyte and embryo growth and development are a priority for research  相似文献

Conventional culture for the detection, enumeration and identification of micro-organisms has been the traditional tool of the microbiologist. It is, however, time-consuming and labour-intensive and confirmed results often require several days of analysis. Culture may not grow the organisms being sought and for enumeration may only detect a small proportion of the total population. However, it does have the advantage of being simple to use and relatively inexpensive. It is also a direct means of assessing cell viability. Novel fluorogenic dyes and fluorgenic and chromogenic substrates have overcome some of these problems by providing a means of rapid and specific detection and enumeration whilst removing the need for subculture and confirmation tests. Immunological tests such as ELISA have significantly reduced analysis time by providing specific target organism detection. Molecular techniques have removed the need for culture. Improvements in sensitivity, and removal of the inhibitory nature of sample matrices, have allowed analysts to detect low levels of micro-organisms but the questions of viability and comparability with cultural techniques still remain. Are we about to see a change of culture in water quality assessment, or can cultural techniques be developed that reduce analysis time to a few hours and can rapid methods be used for detecting the presence and viability of organisms?  相似文献

Computer system validation of a LIMS is often confused with the ongoing task of entering and quality-assuring the material, instrument, specification, method, calculation, and study data required for laboratory sample and results management. A practical approach to LIMS template data quality assurance includes preparation of one or more standard operating procedures for entry and quality assurance of material specifications, test limits, secondary calculation formulae, and other analytical laboratory testing information.  相似文献

A number of evolutionary theories of human life history assume a quantity-quality tradeoff for offspring production: parents with fewer offspring can have higher biological fitness than those with more. Direct evidence for such a tradeoff, however, is mixed. We tested this assumption in a community of Ecuadorian Shuar hunter-horticulturalists, using child anthropometry as a proxy for fitness. We measured the impact of household consumer/producer (CP) ratio on height, weight, skinfold thicknesses, and arm and calf circumferences of 85 children and young adults. To control for possible "phenotypic" correlates that might mask the effect of CP ratio on anthropometry, we also measured household garden productivity, wealth, and social status. Regression models of the age-standardized variables indicated a significant negative impact of CP ratio on child growth and nutrition. The age-standardized height and weight of children in households with the largest CP ratio (10) were 1.38 and 1.44 standard deviations, respectively, below those of children in households with the smallest CP ratio (2). Surprisingly, garden productivity, wealth, and status had little to no effect on the fitness proxies. There was, however, an interesting and unexpected interaction between status and sex: for females, but not males, higher father status correlated significantly with higher values on the proxies.  相似文献

Significant insight into the mechanisms that contribute to dopaminergic neurodegeneration in Parkinson disease has been gained from the analysis of genes linked to rare heritable forms of parkinsonism such as PINK1 and parkin, loss-of-function mutations of which cause autosomal recessive parkinsonism. PINK1 encodes a mitochondrially targeted Ser/Thr kinase and parkin encodes a ubiquitin-protein ligase. Functional studies of PINK1 and Parkin in animal and cellular model systems have shown that both proteins play important roles in maintaining mitochondrial integrity. Genetic studies of PINK1 and Parkin orthologs in flies have shown that PINK1 acts upstream from Parkin in a common pathway that appears to regulate mitochondrial morphology. Mitochondrial morphology is regulated by mitochondrial fission and fusion-promoting proteins, and is important in a variety of contexts, including mitochondrial trafficking and mitochondrial quality control. In particular, mitochondrial fission appears to promote the segregation of terminally dysfunctional mitochondria for degradation in the lysosome through a process termed mitophagy. Recent work has shown that Parkin promotes the degradation of dysfunctional mitochondria in vertebrate cell culture. Here we postulate a model whereby the PINK1/Parkin pathway regulates mitochondrial dynamics in an effort to promote the turnover of damaged mitochondria.  相似文献

While there has been considerable interest in female choice for male sexual signals, there have been few studies of the underlying information that different aspects of the signal calls convey. Such studies, however, are essential to understand the significance of signals as honest handicaps, arbitrary Fisherian traits and/or in species recognition. We studied the somewhat exceptional system of audible drumming in the wolf spider Hygrolycosa rubrofasciata. We estimated the repeatabilities of signal components, the levels of between-male variance, the symmetry of the signal, the correlations between different aspects of drumming and their correlations with body weight. While in other taxa the frequency of audible signals may convey honest information of male size, in this species signal frequency was not related to male size and had a low repeatability. The pulse rate within each drum was highly repeatable but had a relatively small between-male coefficient of variation. In previous studies on this species, these traits were not important for male mating success. Among the traits directionally preferred by females, signal volume had considerable repeatability. Signal length was repeatable with high variability between males. In one population, signal length and volume were positively correlated with the rate at which males produced the drumming signals, a trait essential for male mating success. Thus, while signal length may reliably indicate male quality, other signal characteristics such as peak frequency and symmetry were not repeatable or were static and not related to any other male traits. Mutations in human mitochondrial tRNA genes are associated with a number of multisystemic disorders. These single nucleotide substitutions in various domains of tRNA molecules may affect different steps of tRNA biogenesis. Often, the prominent decrease of aminoacylation and/or steady-state levels of affected mitochondrial tRNA have been demonstrated in patients' tissues and in cultured cells. Similar effect has been observed for pathogenic mutations in nuclear genes encoding mitochondrial aminoacyl-tRNA-synthetases, while over-expression of mitochondrial aminoacyl-tRNA synthetases or elongation factor EF-Tu rescued mutated tRNAs from degradation. In this review we summarize experimental data concerning the possible regulatory mechanisms governing mitochondrial tRNA steady-state levels, and propose a hypothesis based on the tRNA channelling principle. According to this hypothesis, interaction of mitochondrial tRNA with proteins ensures not only tRNA synthesis, maturation and function, but also protection from degradation. Mutations perturbing this interaction lead to decreased tRNA stability.  相似文献

Courtship behaviour in spiders in the form of premating vibrations by males may function (1) as a male identity signal used for species recognition, (2) in suppression of female aggressiveness, (3) to stimulate female mating behaviour, or (4) as a quality signal used in female choice. We investigated the function of web vibration by male Stegodyphus lineatus in a series of experiments. Regardless of vibratory performance, all males mated successfully with virgin females but only 56.4% of males mated with nonvirgin females. Vibratory performance did not influence male mating success, but heavier males had a higher probability of mating with mated females. Males vibrated less often and produced fewer vibrations when introduced on the web of a mated female. Males that vibrated webs of virgin females mated faster than nonvibrating males, but there was no effect of vibration rate or body mass. There was no effect of male vibratory effort or vibration rate on female reproductive success measured as time to egg laying, clutch size, number of hatched young, number of dispersed young and offspring body mass after a single mating. Males vibrated on abandoned virgin female webs but the response decreased with increasing duration of female absence, suggesting that females produce a web-borne pheromone, which elicits male vibrating behaviour. Mated females were less receptive and not stimulated by male vibrating behaviour. We conclude that male premating vibrations in S. lineatus do not function as a male quality signal selected via female choice. Rather, the primary function of this behaviour may be to stimulate a receptive female to mate. Several quantitative trait loci (QTL) for different meat quality traits have been localized on the q arm of porcine chromosome 2 at position 55-78 cM. Association analyses were performed in a commercial Landrace × Chinese-European (LCE) crossbred population (n = 446) slaughtered at approximately 127 kg and an average age of 198 days with records for performance (growth, fat and meat accretion) and meat quality [intramuscular fat (IMF), Minolta L*, Minolta a*, Minolta b* and pH at 45 m]. Polymorphisms within positional candidate genes cloned from homologous regions on human chromosome 19, ubiquitin-like 5 (UBL5- AM950288:g.566G>A), resistin (RETN- AM157180:g.1473A>G causing substitution p.Ala36Thr), insulin receptor (INSR- AM950289:g.589T>C) and complement factor D (adipsin) (CFD- AM950287:g. 306C>T) were located at positions 62.1, 64.0, 68.0 and 70.7 cM respectively on the current USDA USMARC map of porcine chromosome 2 and had the following allele frequencies in the LCE: UBL5 566G - 0.57; RETN 1473G - 0.84; INSR 589C - 0.70; and CFD 306C - 0.73. The effects of alleles within the candidate genes on the recorded traits were estimated using an animal model. Significant effects (P < 0.05) were found for pH(45) in m. semimembranosus (m. sm.) (UBL5), IMF (RETN) and Minolta L* (RETN, CFD). Differences between phenotypic means of homozygotes at UBL5, RETN and either RETN or CFD explained 0.34 SD for pH(45) in m. sm., 0.47 SD for IMF and 0.68 SD for Minolta L* respectively. Suggestive effects (P < 0.10) on IMF (UBL5, CFD), Minolta a* (INSR, CFD) and Minolta b* (INSR) were also observed. Our results support the localization of further QTL for meat quality traits in this region and suggest that there are several genes affecting different meat quality traits.  相似文献

Glucocorticoids (GCs) are often interpreted as indicators of disturbance, habitat quality, and fitness in wild populations. However, since most investigations have been unable to examine habitat variability, GC levels, and fitness simultaneously, such interpretations remain largely unvalidated. We combined a quantification of two habitat types, a manipulation of foraging ability (feather‐clipping just prior to nestling rearing), multiple baseline plasma GC measures, and multi‐year reproductive monitoring to experimentally examine the linkages between habitat quality, GCs, and fitness in female tree swallows Tachycineta bicolor. Control females experiencing the higher early‐season food resources of inland–pasture habitat laid larger clutches, but fledged an equal number but lower mass offspring compared to those in riparian–cropland habitat. Despite these differences in reproductive success, females nesting in the two habitat types did not differ in baseline GC levels at the early‐ or late‐breeding stage. Feather‐clipping reduced provisioning rate in both habitat types. However, baseline GC levels were affected in a habitat‐specific way; only individuals in inland–pasture habitats showed an increase in GCs. Despite this difference in GC levels, the manipulation did not influence offspring mass, reproductive output, adult return rate (a proxy for survival) to the following year, or reproductive success in the subsequent year. Nonetheless, regardless of treatment, individuals with higher GC levels during the late breeding stage returned in the following year with higher GC levels at incubation, indicating a long‐term effect on future GC levels. Our results indicate that environmental changes (e.g. foraging conditions) can have consequences for body condition, behaviour, and current and future baseline GC levels without concomitant influences on fitness, and that differences in fitness components between habitats may not be reflected in baseline GC levels. These results illustrate that baseline GCs may not simultaneously reflect environmental quality and fitness, potentially limiting their application in ecological and conservation settings.  相似文献

It has been suggested that previous infection by a vascular fungus causes induced resistance against two-spotted spider mites. To test the generality of this phenomenon, a series of experiments was carried out using two lines of tomato, differing only in resistance againstFusarium. In addition, tests were done in order to see whether the defense response against the fungus also affects the phytophagous mite directly. Inoculation of tomato plants with a vascular fungus (Fusarium oxysporum f.sp.lycopersici race 1) prior to infestation with spider mites caused a decrease in the rate of oviposition of two-spotted spider mites (Tetranychus urticae) on aFusarium-susceptible line, but only when plants were moderately to severely wilted. Spider mite oviposition did not change significantly of a previously inoculatedFusarium-resistant line.AsFusarium causes vascular occlusion and wilting of the plants, drought stress was experimentally induced to determine its influence on the reduction of oviposition. Drought caused a significant reduction in spider mite oviposition. We conclude that the effect of previousFusarium-inoculation on spider mite oviposition is primarily due to the fungus affecting the quality of the host plant (including the effect it may have on the composition of defensive compounds), rather than due to the stimulation of the defense system of the plant. SinceFusarium seals off the xylem vessels, thereby causing wilting of susceptible plants, the reduction in mite oviposition may well be due to drought stress in the leaves, rather than due to the production of phytoalexins.  相似文献

Benthic component of an ecosystem is considered in ecological status assessment of the key European Directives. Most of the metrics proposed for the benthic quality assessment are biodiversity based. Their robustness and applicability are widely discussed in many recent studies. However an impact of invasive alien species on biotic indices and environmental quality assessments has been largely overlooked by researchers so far. In the current study we assessed Benthic Quality Index (BQI) in a coastal ecosystem, highly affected by the invasive zebra mussel Dreissena polymorpha. Zebra mussel is able of modifying benthic habitats and enhancing local biodiversity. In the analyzed ecosystem it affected benthic species richness, abundance and community structure. As a result the calculated BQI values were significantly higher in the presence of zebra mussel with evident outliers in samples with particularly high zebra mussel abundances. Therefore we found that BQI determined in our study was artificially elevated providing false signal of the ecological status improvement. Based on the results presented, we suggested data correction framework that has been tested on the current dataset and proved to be effective minimizing zebra mussel impact on BQI assessment. Our experience could be applied for other coastal ecosystems invaded by the zebra mussel or any other aquatic invasive species with resembling biological traits and bioinvasion impacts.  相似文献

Mate choice is favored by indirect selection if choosy females mate with males of high genetic quality. We believe, however, that testing hypotheses about indirect selection has been constrained by how we conceptualize and therefore empirically measure male genetic quality. Here, we argue that genetic quality is the breeding value of an individual for total fitness. We can therefore learn little about genetic quality from measures of only a few fitness components. We explain breeding value for total fitness, drawing on concepts from life-history theory and quantitative genetics, and suggest how approaches incorporating these insights might result in empirical progress.  相似文献

Herbivore–carnivore interactions are influenced by the plants on which herbivores feed. Accordingly, dietary generalist herbivores have been shown to experience differential risk of mortality from carnivores on different host-plant species. Here, we investigate whether caterpillar density and host-plant quality play a role in driving variation in generalist forest caterpillar mortality from insect parasitoids using a large-scale, multi-year observational study. A total of 4,500 polyphagous caterpillars were collected from eight host-tree species in Connecticut deciduous forests over 5 years, and frequencies of mortality from insect parasitoids (flies and wasps) were compared across the eight host-plant species for the entire generalist caterpillar assemblage (76 species). Separate comparisons were made using seven numerically dominant generalist species, allowing us to account for variation in caterpillar species-specific parasitism risk. We find significant variation in parasitism frequencies of generalist caterpillars across the eight host-plant species when accounting for variation in caterpillar density. We find no support for an influence of caterpillar density on parasitism and no clear evidence for an effect of host-plant quality on parasitism. Therefore, the results of this study discount the hypotheses that variation in caterpillar density and host-plant quality are responsible for variation in parasitism frequencies across host-plant species. Instead, our findings point to other plant-related characteristics, such as plant-derived parasitoid attractants, which may have robust, community-wide effects.  相似文献

The hypothesis that specific components of seawater, such as particulate, dissolved and colloidal organic and inorganic material, render virions non-infective has long been postulated, but never rigorously tested. To address this hypothesis, the plaque assay method was used to derive infective decay rates, k, of two bacteriophages—P1 (marine host: PWH3a) and T4 (enteric host: E. coli B). We compared k values of bacteriophage suspended in serial filtrations of seawater, with and without autoclaving and UV oxidation. Both phages exhibited reduced decay rates in particle-free water (<0.2 μm) compared to <10 μm filtrate. The largest decrease in virion decay rates was achieved by autoclaving the 0.2 μm filtrate. UV oxidation of <0.2 μm filtrate, however, yielded higher decay rates than observed in autoclaved treatments. The lowest k values were seen in ultra-filtered seawater (<10 kDa). Exposure to a wide range of concentrations of Pronase E (a proteolytic enzyme), inorganic clay (kaolinite or montmorillonite), and organic particles (phytoplankton debris) did not promote phage inactivation. P1 infective titers were also not consistently reduced by exposures to axenic cultures of a resistant host mutant (PWH3a-R) and a non-host marine bacterium (MB-5). Finally, phage were exposed to a range of temperatures to derive activation energies required for phage inactivation. Application of the Arrhenius model to inactivation of T4 and P1 yielded activation energies (E a) of 49 and 40 kJ mol−1, respectively. This is the first comprehensive analysis in which specific seawater components were assayed for their ability to inactivate bacteriophage. Inactivation of these phage does not appear to depend on capsomere denaturation, proteolytic extracellular enzymes, sorption to non-host bacteria, clay particles or particulate organic debris, but is accelerated by naturally occurring particles, which include living organisms, and heat-labile colloids and macromolecules >10 kDa.  相似文献

Pork quality is an economically important trait and one of the main selection criteria for breeding in the swine industry. In this genome-wide association study (GWAS), 455 pigs from a porcine Large White × Minzhu intercross population were genotyped using the Illumina PorcineSNP60K Beadchip, and phenotyped for intramuscular fat content (IMF), marbling, moisture, color L*, color a*, color b* and color score in the longissimus muscle (LM). Association tests between each trait and the SNPs were performed via the Genome Wide Rapid Association using the Mixed Model and Regression-Genomic Control (GRAMMAR-GC) approach. From the Ensembl porcine database, SNP annotation was implemented using Sus scrofa Build 9. A total of 45 SNPs showed significant association with one or multiple meat quality traits. Of the 45 SNPs, 36 were located on SSC12. These significantly associated SNPs aligned to or were in close approximation to previously reported quantitative trait loci (QTL) and some were located within introns of previously reported candidate genes. Two haplotype blocks ASGA0100525-ASGA0055225-ALGA0067099-MARC0004712-DIAS0000861, and ASGA0085522-H3GA0056170 were detected in the significant region. The first block contained the genes MYH1, MYH2 and MYH4. A SNP (ASGA0094812) within an intron of the USP43 gene was significantly associated with five meat quality traits. The present results effectively narrowed down the associated regions compared to previous QTL studies and revealed haplotypes and candidate genes on SSC12 for meat quality traits in pigs.  相似文献
