A morphological basis for transcellular potassium transport in the midgut of the mature fifth instar larvae of Hyalophora cecropia has been established through studies with the light and electron microscopes. The single-layered epithelium consists of two distinct cell types, the columnar cell and the goblet cell. No regenerative cells are present. Both columnar and goblet cells rest on a well developed basement lamina. The basal portion of the columnar cell is incompletely divided into compartments by deep infoldings of the plasma membrane, whereas the apical end consists of numerous cytoplasmic projections, each of which is covered with a fine fuzzy or filamentous material. The cytoplasm of this cell contains large amounts of rough endoplasmic reticulum, microtubules, and mitochondria. In the basal region of the cell the mitochondria are oriented parallel to the long axes of the folded plasma-lemma, but in the intermediate and apical portions they are randomly scattered within the cytoplasmic matrix. Compared to the columnar cell, the goblet cell has relatively little endoplasmic reticulum. On the other hand, the plications of the plasma membrane of the goblet cell greatly exceed those of the columnar cell. One can distinguish at least four characteristic types of folding: (a) basal podocytelike extensions, (b) lateral evaginations, (c) apical microvilli, and (d) specialized cytoplasmic projections which line the goblet chamber. Apically, the projections are large and branch to form villus-like units, whereas in the major portion of the cavity each projection appears to contain an elongate mitochondrion. Junctional complexes of similar kind and position appear between neighboring columnar cells and between adjacent columnar and goblet cells as follows: a zonula adherens is found near the luminal surface and is followed by one or more zonulae occludentes. The morphological data obtained in this study and the physiological information on ion transport through the midgut epithelium have encouraged us to suggest that the goblet cell may be the principal unit of active potassium transport from the hemolymph to the lumen of the midgut. We have postulated that ion accumulation by mitochondria in close association with plicated plasma membranes may play a role in the active movement of potassium across the midgut.  相似文献

利用透射电镜观察了稻纵卷叶螟 Cnaphalocrocis medinalis(Guenée)幼虫取食转Bt基因水稻后中肠的组织病理变化。结果表明:稻纵卷叶螟幼虫取食转cry1Ab基因水稻后,中肠上皮细胞的线粒体先发生形态变化,随连续取食时间的延长线粒体出现凝聚、内嵴稀疏、空泡化等,在后期还呈凝聚态随突起脱落或沿杯腔边沿单一排列。内质网的变化也很明显,病变过程中伴随着粗糙内质网的肿胀、核糖体脱落,粗糙内质网增多等现象。细胞核的变化较小,在处理后期出现细胞核拉长、核仁聚集等变化。组织病变程度不一,有的细胞在病变早期就出现了空泡化。  相似文献

The epithelium of anterior midgut of adult Cenocorixa bifida was examined with light and electron microscopy. The folded epithelium is composed of tall columnar cells extending to the lumen, differentiating dark and light cells with interdigitating apices and regenerative basal cells in the nidi surrounded by villiform ridges that penetrate deeply into the epithelium. The columnar cells display microvilli at their luminal surface. Microvilli lined intercellular spaces and basal plasma membrane infoldings are associated with mitochondria. These ultrastructural features suggest their role in absorption of electrolytes and nutrients from the midgut lumen. The columnar cells contain large oval nuclei with prominent nucleoli. Their cytoplasm is rich in rough endoplasmic reticulum, Golgi complexes and electron-dense secretory granules indicating that they are also engaged in synthesis of digestive enzymes. The presence of secretory granules in close proximity of the apical plasma membrane suggests the release of secretion is by exocytosis. The presence of degenerating cells containing secretory granules at the luminal surface and the occurance of empty vesicles and cell fragments in the lumen are consistent with the holocrine secretion of digestive enzymes. Apical extrusions of columnar cells filled with fine granular material are most likely formed in response to the lack of food in the midgut. The presence of laminated concretions in the cytoplasm is indicative of storageexcretion of surplus minerals. The peritrophic membrane is absent from the midgut of C. bifida.  相似文献

Solenopsis saevissima has a midgut composed of columnar, regenerative, and goblet cells. The midgut epithelium was covered by a basal lamina. Outside the basal lamina, layers of inner oblique, circular, and outer longitudinal muscles were present. Columnar cells showed a basal plasma membrane containing numerous folds, mitochondria, and the nucleus. Rough endoplasmic reticulum, Golgi bodies, membrane bounded vacuoles, and spherocrystals were found in this region. The apical plasma membrane was constituted by microvilli, which were above a region rich in mitochondria. Regenerative cells were found in groups lying by the basal lamina. Goblet cells were associated with an ion-transporting mechanism between the haemolymph and the midgut epithelium. These cells were lying by the midgut lumen and large microvilli were evident, but the cytoplasmic features were similar to the columnar cells.  相似文献

The epithelium of the Malpighian tubules in the housefly is comprised of four distinct cellular types. Type I cells are characterized by the presence of intimate associations between infoldings of basal plasma membrane and mitochondria. On the luminal surface, cytoplasm is extended into microvilli which contain mitochondria. Membrane-bound vacuoles in the cytoplasm seem to progressively accumulate granular material. Type II cells have dilated canaliculi. Microvilli lack mitochondria. The Type III cell has not been reported previously in Malpighian tubules. It has very well-developed granular endoplasmic reticulum which contains intracisternal bundles of tubules. Cytoplasm contains numerous electron dense bodies. Type IV cells occur in the common duct region of the Malpighian tubules. Mitochondria do not extend into the microvilli.  相似文献

The midgut epithelium of larval and early postlarval brown shrimp has been studied with light and electron microscopy. Ultrastructurally the features of the midgut do not change during these stages of development. On the basis of electron density, two epithelial cell types can be distinguished, and these are referred to as light and dark cells. The dark cells contain more rough endoplasmic reticulum and more free ribosomes than the light cells. Mitochondria in the dark cells have a matrix which is less electron dense than the mitochondrial matrix of the light cells. Both cell types have a microvillous border with a surface coat. The microvilli lack microfilaments within their core, and a terminal web is not differentiated in the stages examined. Tubular smooth endoplasmic reticulum is abundant in the basal portions of the cells. Electron dense, membrane bound vesicles are consistently seen in association with the Golgi apparatus, apical cell surface, and gut lumen and therefore are believed to be secretory granules. Cells in the anterior portion of the midgut often contain very large lipid droplets in the cytoplasm.  相似文献

Bacillus thuringiensis crystal toxin induced a cytoplasmic response in columnar cells within 1 min after ingestion although external symptoms were not exhibited by larvae until 15 min after ingestion. Microvilli became less consistently uniform in diameter; their organized internal microfilaments were disrupted and disappeared. The cisternae of rough endoplasmic reticulum were enlarged and denuded of ribosomes. By 5 min after ingestion, microvilli of some columnar cells disappeared entirely and gross ultrastructural changes were observed in other regions of the cells. Up to 5 min after ingestion there were few, if any, ultrastructural changes observed within goblet cells. Mitochondria in columnar cells were swollen but did not exhibit the condensed configuration reported by other workers. Both the buffer system used in the fixation medium and its osmolarity influenced the changes in the ultrastructure of midgut cells exposed to B. thuringiensis crystal toxin.  相似文献

为了明确Cry2Ab杀虫蛋白的作用机制, 利用透射电镜观察了棉铃虫Helicoverpa armigera (Hübner)3龄末幼虫取食含Cry2Ab蛋白(8 μg/g)饲料后中肠的组织病理变化, 并与分别取食含Cry1Ac蛋白(0.97 μg/g)饲料和正常饲料的棉铃虫进行了比较。结果表明: 棉铃 虫取食Cry2Ab蛋白后中肠细胞及其细胞器均发生了明显的病变, 主要表现为: 中肠杯状细胞的杯腔肿胀或拉长, 部分柱状细胞被杯状细 胞挤压出来, 微绒毛脱落, 细胞核皱缩, 质膜和核膜不清晰, 染色质凝聚, 线粒体拉伸变形, 内质网肿胀断裂; 并且随着取食时间 的延长病变越来越明显。与取食Cry1Ac蛋白的棉铃虫相比, Cry2Ab引起棉铃虫中肠组织发生病变的速度较慢。本研究可为Cry2Ab作为转基 因棉花的重要杀虫蛋白在将来更好地发挥作用提供理论依据。  相似文献

The retinal pigment epithelium (RPE), the choriocapillaris and Bruch's membrane (complexus basalis) have been studied by light and electron microscopy in the bobtail goanna (Tiliqua rugosa) an Australian diurnal lizard. The RPE consists of a single layer of cuboidal cells which display very deep and tortuous basal (choroidal) infoldings as well as numerous apical (vitreal) processes which interdigitate with the photoreceptor cells. The lateral cell borders are relatively smooth and joined by basally located tight junctions. Internally smooth endoplasmic reticulum is abundant while rough endoplasmic reticulum is not. The RPE cell nucleus is large and vesicular and basally located in the light-adapted state. Polysomes, mitochondria and myeloid bodies are present and widely distributed. Melanosomes are plentiful in the apical region of the epithelial cells in light-adaptation. Bruch's membrane is pentalaminate with the basal lamina of the choriocapillaris being exceptionally thick. The choriocapillaris is a single layer of large-caliber capillaries with thin but only moderately fenestrated endothelium. Numerous dense granules are always present within these endothelial cells.  相似文献

The midgut of Rhynchosciara americana larvae consists of a cylindrical ventriculus from which protrudes two gastric caeca formed by polyhedral cells with microvilli covering their apical faces. The basal plasma membrane of these cells is infolded and displays associated mitochondria which are, nevertheless, more conspicuous in the apical cytoplasm. The anterior ventricular cells possess elaborate mitochondria-associated basal plasma membrane infoldings extending almost to the tips of the cells, and small microvilli disposed in the cell apexes. Distal posterior ventricular cells with long apical microvilli are grouped into major epithelial foldings forming multicellular crypts. In these cells the majority of the mitochondria are dispersed in the apical cytoplasm, minor amounts being associated with moderately-developed basal plasma membrane infoldings. The proximal posterior ventriculus represents a transition region between the anterior ventriculus and the distal posterior ventriculus. The resemblance between the gastric caeca and distal posterior ventricular cells is stressed by the finding that their microvilli preparations display similar alkaline phosphatase-specific activities. The results lend support to the proposal, based mainly on previous data on enzyme excretion rates, that the endo-ectoperitrophic circulation of digestive enzymes is a consequence of fluid fluxes caused by the transport of water into the first two thirds of midgut lumen, and its transference back to the haemolymph in the gastric caeca and in the distal posterior ventriculus.  相似文献

为了进一步明确Vip3Aa的作用机制, 利用透射电镜观察了棉铃虫4龄幼虫取食含Vip3Aa蛋白饲料后中肠杯状细胞的病理变化, 并比较了其病变与取食含Cry1Ac饲料后棉铃虫组织病变的差异。取食含Vip3Aa饲料后, 棉铃虫幼虫的中肠杯状细胞逐渐发生病变, 主要表现为: 微绒毛肿胀、 脱落; 细胞核核膜界限不清晰, 染色质分布不均匀; 线粒体变形、 数量减少, 内脊不清晰; 内质网杂乱不规则、 数量减少。与取食Cry1Ac的棉铃虫相比, 取食Vip3Aa的棉铃虫中肠杯状细胞发生病变较为缓慢, 在取食12 h后才发现明显病变, 随着取食时间的增加病变越来越明显; 而取食Cry1Ac的棉铃虫2 h后中肠杯状细胞就出现明显病变。本研究可为Vip3Aa作为新毒素策略的重要蛋白在棉铃虫Helicoverpa armigera综合防治中更好地发挥作用提供理论依据。  相似文献

To improve the current knowledge about the digestive system in opisthobranchs, light and electron microscopy methods were used to characterize the epithelial cells in the mid‐intestine of Aplysia depilans. This epithelium is mainly formed by columnar cells intermingled with two types of secretory cells, named mucous cells and granular cells. Columnar cells bear microvilli on their apical surface and most of them are ciliated. Mitochondria, multivesicular bodies, lysosomes and lipid droplets are the main components of the cytoplasm in the region above the nucleus of these cells. Peroxisomes are mainly found in middle and basal regions, usually close to mitochondria. Mucous cells are filled with large secretory vesicles containing thin electron‐dense filaments surrounded by electron‐lucent material in which acidic mucopolysaccharides were detected. The basal region includes the nucleus, several Golgi stacks and many dilated rough endoplasmic reticulum cisternae containing tubular structures. The granular cells are characterized by very high amounts of flat rough endoplasmic reticulum cisternae and electron‐dense spherical secretory granules containing glycoproteins. Enteroendocrine cells containing small electron‐dense granules are occasionally present in the basal region of the epithelium. Intraepithelial nerve fibres are abundant and seem to establish contacts with secretory and enteroendocrine cells.  相似文献

The development of a nuclear polyhedrosis virus (NPV) in larval midgut cells of the armyworm, Pseudaletia unipuncta, is similar to that of other NPV. In the nucleus, the envelopes around the nucleocapsids seem to be derived de novo or from the inner layer of the nuclear envelope wich forms cisternae, blebs, or infoldings. The nucleocapsids are also enveloped by synhymenosis during passage through the nuclear membrane, the cell membrane, or the endoplasmic reticulum membrane. Both enveloped and unenveloped nucleocapsids may enter the cytoplasm through the nuclear pore or budding through the nuclear membrane. From the cytoplasm the virions may enter the hemocoel through the basal cell and basement membranes or through the endoplasmic reticulum, intercellular space, and the basement membrane.  相似文献

The ultrastructure of the anal organ of the full-grown larva of Drosophila melanogaster is described. The thin cuticle is characterized by epicuticular depressions which contain particulate material. In AgNO(3)-treated larvae, silver grains tend to penetrate the cuticle at the epicuticular depressions. At the basal surface, the epithelial cells exhibit narrow, parallel membrane infoldings which bear a particulate coat on the cytoplasmic surface. The infoldings are also attached around the cytoplasmic surface of endocuticular tubercles, thereby greatly increasing the absorptive surface area. At the apical surface, the membrane invaginations, which are closely associated with mitochondria, anastomose freely and extend deeply into the cytoplasm. The lateral membranes are linked by desmosomes and septate junctions. They are highly folded, are closely associated with mitochondria, and enclose intercellular channels and spaces. The epithelial cells are rich in mitochondria, glycogen particles and tracheoles. Numerous vesicles, multivesicular bodies, lysosome-like dense bodies and sparse endoplasmic reticulum are found in the cytoplasm. In concentrated medium, the epithelial cells show complete absence of the membrane infoldings and invaginations and reduction in the number of mitochondria. The ultrastructural features of the anal organ are consistent with its function in ion transport.  相似文献

Summary The structure of the salivary gland of the dipteran insect Rhynchosciara angelae in a defined stage of the larval development, characterized by the synthesis and storage of secretion product, is described. Observations were made with both Nomarski optics and electron microscopy. Filiform projections extending into the lumen of the gland were observed in the apical portion of the cells. At the basal region junctions, characterized as hemidesmosomes, were observed between the membrane of the cell and the basal lamina. The plasma membrane presents numerous infoldings into the cell increasing considerably the surface area at this region. Throughout the cytoplasm of the gland cells numerous mitochondria, Golgi complexes, microtubules, profiles of endoplasmic reticulum, secretion granules and glycogen granules were observed. Carbohydrates were detected on ultrathin sections by using the periodic acid-silver methenamine and the periodic acid-thiosemicarbazide-silver proteinate techniques.  相似文献

The peritrophic membrane of Drosophila melanogaster consists of four layers, each associated with a specific region of the folded epithelial lining of the cardia. The epithelium is adapted to produce this multilaminar peritrophic membrane by bringing together several regions of foregut and midgut, each characterized by a distinctively differentiated cell type. The very thin, electron-dense inner layer of the peritrophic membrane originates adjacent to the cuticular surface of the stomadeal valve and so appears to require some contribution by the underlying foregut cells. These foregut cells are characterized by dense concentrations of glycogen, extensive arrays of smooth endoplasmic reticulum, and pleated apical plasma membranes. The second and thickest layer of the peritrophic membrane coalesces from amorphous, periodic acid-Schiff-positive material between the microvilli of midgut cells in the neck of the valve. The third layer of the peritrophic membrane is composed of fine electron-dense granules associated with the tall midgut cells of the outer cardia wall. These columnar cells are characterized by cytoplasm filled with extensive rough endoplasmic reticulum and numerous Golgi bodies and by an apical projection filled with secretory vesicles and covered by microvilli. The fourth, outer layer of the peritrophic membrane originates over the brush border of the cuboidal midgut cells, which connect the cardia with the ventriculus.  相似文献

Triatoma vitticeps (Stal, 1859) is a hematophagous Hemiptera that, although being considered wild, can be found in households, being a potential Chagas’ disease vector. This work describes the histology and ultrastructure of the midgut of T. vitticeps under different starvation periods. Fifteen adults of both sexes starved for 3, 7, 20 and 25 days were studied. In general, digestive cells had apical microvilli, basal plasma membrane infoldings and central nucleus. The perimicrovillar membrane was found in all insects examined. Digestive cells of anterior midgut had lipid droplets, glycogen granules, developed basal labyrinth associated with mitochondria suggesting their role in nutrient storage and in fluid and ion transport. The cells of median and posterior regions of the midgut were rich in rough endoplasmic reticulum, lysosomes, vesicles and granules with different electron-densities. Moreover, cells of the posterior portion of the midgut had hemozoyn granules and mitochondria in the apical cytoplasm close to microvilli, suggesting their role in blood digestion and active nutrient absorption. The midgut of T. vitticeps showed differences in digestive cells associated with the time after feeding, and the increase of vesicles amount in long starvation periods, which suggests enzyme storage, which is readily used after a blood meal.  相似文献

The effects of salinity adaptation and of composition and tonicity of fixatives upon the ultrastructure of the posterior midgut caecum (PMC) of Pachygrapsus crassipes have been studied. The PMC epithelium consists of a single layer of columnar cells with a microvillous border. The apical cytoplasm contains numerous mitochondria, lysosomes, and much smooth endoplasmic reticulum. Rough endoplasmic reticulum and Golgi apparatus are situated in the perinuclear cytoplasm. This epithelium resembles other transporting epithelia in that the basal cytoplasm has an extensive system of branched tubules formed from invaginations of the lateral and basal plasma membrane. Numerous mitochondria are associated with the basal tubular system. To determine the possible contribution of the PMC to the osmoregulatory ability of Pachygrapsus, the ultrastructure of the PMC from animals adapted to 40, 50, 100 and 150% sea water was investigated. Enlargement of basal tubules and intercellular spaces at low salinity, suggestive of fluid-transport activity, was found to be an artifact of fixation. The most consistent response when animals were acclimated to dilute salinities was that some basal mitochondria assume a more complex shape, usually appearing as rings in cross sections of the caecum. A hypothesis concerning the functional significance of these mitochondria is proposed.  相似文献

Small intestinal crypt epithelium obtained from normal fasting humans by peroral biopsy of the mucosa was studied with the electron microscope. Paneth cells were identified at the base of the crypts by their elaborate highly organized endoplasmic reticulum, large secretory granules, and small lysosome-like dense bodies within the cytoplasm. Undifferentiated cells were characterized by smaller cytoplasmic membrane-bounded granules which were presumed to be secretory in nature, a less elaborate endoplasmic reticulum, many unattached ribosomes and, in some cells, the presence of glycogen. Some undifferentiated cells at the base of the crypts contained lobulated nuclei and striking paranuclear accumulations of mitochondria. Membrane-bounded cytoplasmic fragments, probably originating from undifferentiated and Paneth cells, were frequently apparent within crypt lumina. Of the goblet cells, some were seen actively secreting mucus. In these, apical mucus appeared to exude into the crypt lumen between gaps in the microvilli. The membrane formerly surrounding the apical mucus appeared to fuse with and become part of the plasma membrane of the cell, suggesting a merocrine secretory mechanism. Enterochromaffin cells were identified by their location between the basal regions of other crypt cells and by their unique intracytoplasmic granules.  相似文献

The urinary bladder of a euryhaline marine teleost, Gillichthys mirabilis , was studied by light and electron microscopy. An enlargement of the mesonephric ducts forms a sac-like structure lined by an epithelium composed of two major cell types. Tall columnar cells continuous with the duct epithelium are characterized by a large number of mitochondria and well-developed rough and smooth endoplasmic reticulum. Tubular smooth endoplasmic reticulum is more developed in the basal cytoplasm and often opens into the extracellular space. A second cell type, the low cuboidal cells, forming most of the bladder epithelium, has fewer mitochondria. Basal cells are rarely observed and mucous cells are absent.
In seawater Gillichthys , cells of both types are separated by narrow intercellular spaces. In 5% seawater fish, the columnar cells show functional activation, as evidenced by an increased number of mitochondria and more extensive tubular smooth endoplasmic reticulum. No such changes were noted in the cuboidal cells; however, the lateral intercellular spaces are dilated probably owing to hypotonicity of the urine in the hypotonic environment. A functional difference between the two cell types is strongly suggested. The columnar cells may be responsible for active sodium uptake in hypotonic seawater environments.  相似文献
