有毒蓝藻在形成水华破坏水体环境的同时,也对人畜产生危害。运用分子生物学技术对水环境中产毒蓝藻进行监测,由于其简单、快速和经济等优点,逐渐成为各国学者关注的热点。本文从以下3个方面对其进行了综述:1)早期分子生物学技术在产毒蓝藻诊断中的运用;2)以产毒基因为靶标进行的产毒蓝藻诊断;3)运用基因芯片技术对产毒蓝藻进行检测。与传统的分子生物学技术相比,生物芯片技术具有体积小、重量轻、便于携带、分析自动快速、高通量等许多传统方法所不能比拟的优势。因此,该技术在蓝藻毒素检测中的运用必将给目前的产毒蓝藻的检测带来一场新的革命。  相似文献

本文报道了采集广西产虎纹捕鸟蛛(Selenocosmia huwena)毒液的方法,并对粗毒进行了部分生物学活性的测定,该粗毒对小鼠和蜚蠊的LD50分钟为1.16mg/kg和300ug/g,该粗毒具有透明质酸酶,碱性磷酸酶,蛋白水解酶和脱氧核糖核酸酶活性,未测到磷脂酶A和胆碱脂酶性,通过电生理实验发现该粗毒含有阻断蟾蜍神经肌肉接头传递的毒素,并观察到当小鼠接受腹腔注射致死剂量(5mg/kg)粗毒后便迅速出现呼吸麻痹。  相似文献

对某卫生防疫站委托本研究室分离病原菌的一份引起肉毒中毒的食品一"黄豆冬瓜酱"进行检测和病原菌的分离,从中再次检出了E型肉毒毒素并分离到一产毒菌种,对该菌种的生物学及生化学特性进行检查,并检测其毒素基因(PCR试验)。结果:该分离菌能产生E型肉毒毒素,PCR检测结果也证明其具有E型肉毒神经毒素基因,但其多项生化特性与E型肉毒梭菌有明显差异,而与酪酸梭菌完全一致。结果说明该分离菌系产生E型肉毒毒素的酪酸梭菌,而非E型肉毒梭菌。由酪酸梭菌引起的食物中毒型肉毒中毒并从中毒食品中分离到该病原菌,这在国际上尚属首次报告。  相似文献

全球范围内,高频次、大范围暴发的蓝藻水华对淡水水体环境造成严重影响.微囊藻因其在生长特别是衰亡过程中向水体释放微囊藻毒素而威胁人类健康.因此,分析其产毒株及非产毒株在环境样品中的组成,建立产毒蓝藻的预报及评价体系显得极为重要.本文采用荧光原位杂交技术结合流式细胞技术实现对环境样品中产毒藻株的鉴别与定量.针对目标基因mcyA设计的、以地高辛标记的双链DNA探针可有效应用于产毒微囊藻FACHB905和PCC7806的鉴别.分别对来自滇池、太湖和关桥的11个样品进行分析显示,该方法与传统的形态学鉴定及PCR方法有较好的匹配.荧光原位杂交技术与流式细胞相结合可有效鉴别产毒与非产毒微囊藻,尤其可以对野外样品中产毒与非产毒藻株进行简便、可视化地鉴别,从而达到对产毒微囊藻水华早期预警的目的.  相似文献

不同pH和盐度下海洋细菌对赤潮藻生长和产毒的影响   总被引:17,自引:4,他引:13  
研究了在可控生态条件下。一株分离自厦门西海域沉积物的海洋细菌S10在不同pH和盐度条件下对赤潮原因种塔玛亚历山大藻(Alexandrium tamarense)生长和产毒的影响.结果表明。实验用藻株适宜生长pH为6~8,适宜盐度为20~34;该藻株在不同pH及不同盐度条件下,藻细胞毒力差异显著,且随着pH升高而下降。随着盐度增加而加大,到盐度为30时达到最高值。然后逐渐下降;菌株Sl0(1.02×10^l0cells·ml^-1)在pH7~9和盐度15~34下均能有效抑藻生长和产毒。且在pH7、盐度34时其抑藻生长作用最强;在pH7时抑藻产毒效果较好,且其抑藻产毒作用强度不随盐度变化而异.  相似文献

研究以珠海市20座水库两次(2018年和2022年)调查数据为基础,分析华南热带地区产毒拟柱孢藻的发生规律和拟柱孢藻毒素(CYN)的健康风险。基于rpoC1和cyrJ基因的荧光定量PCR分析发现,在两次采样的40个样品中均检到拟柱孢藻,在其中的33个样品中检到产毒拟柱孢藻。种群内产毒基因型的比例在0.046%—38.66%,表明产毒株广泛存在但不占优势。与2018年相比, 20座水库的总拟柱孢藻丰度均值在2022年增加近10倍,但两年的产毒拟柱孢藻丰度无显著差异;产毒基因型的比例明显下降,比例均值和最大值分别从2018年的14.86%和38.66%降至2022年的4.17%和17.24%; CYN浓度与产毒细胞比例具有类似的变化趋势,平均浓度也从0.56降至0.19μg/L。线性混合效应模型分析表明,非产毒基因型丰度随无机磷的升高及硝氮的下降而显著增加,而产毒拟柱孢藻比例随水温升高而增加。  相似文献

辣椒疫毒(Phytophthora capsici)毒素的产生是由一个不完全显性基因所控制,通过RAPD/BSA分析证明,用OPW12扩增得到一条约1300bp的RAPD标记带,该带与辣椒疫霉产毒共分离。纯化回收OPW121300DNA,共转化感受态E.coli DH5α,筛选出3个白色阳性克隆,序列分析发现该标记DNA为1291bp。这一标记DNA序列的阐明为进一步分析产毒遗传机理提供了新的信息  相似文献

The pyruvate dehydrogenase complex of Escherichia coli was isolated in a simple three-step procedure. Its chain stoichiometry, determined by trinitrobenzoate modification was found to be 1.4 E1:1 E2:0.6 E3. It was reproducible within 10% from preparation to preparation. The E. coli complex was resolved by chromatography on activated thiol Sepharose. Reconstitution of activity yielded a stoichiometry of 1.0 E1:1 E2:0.5 E3. The optimum binding stoichiometry of E1E2 and E2E3 subcomplexes was determined by sedimentation experiments and found to be 2.0 E1:1 E2 and 2.5 E3:1 E2, respectively. Competition between E1 and E3 was observed in the binding experiments, but not in the kinetic experiments. Hybrid active complexes could be reconstituted from either an E1E2 subcomplex from Azotobacter vinelandii and the E3 component from E. coli or from E2E3 subcomplex from E. coli and the E1 component from A. vinelandii. Low activity and weak binding was observed when E1 from E. coli was recombined with an E2E3 subcomplex from A. vinelandii or when E3 from A. vinelandii was recombined with an E1E2 subcomplex from E. coli. The association behaviour and stoichiometry of the reconstituted complexes is determined by the nature of the E2 component. The formation of hybrid complexes indicates a considerable structural similarity between the complexes from both sources, despite the differences in size and stoichiometry.  相似文献

Easily solubilized carotenoid-containing proteins have been found in aqueous extracts from three genera of cyanobacteria. The three proteins have been purified, and the absorption spectra have been determined to be virtually identical with absorption maxima at 495 and 465 nm. During the purification the orange protein spontaneously changed to a red protein with a single, broad absorption maximum at 505 nm. The orange protein showed a molecular weight of 47 000 on gel filtration while that of the red protein was 26 700. Sodium dodecyl sulfate polacrylamide gel electrophoresis indicated a single polypeptide of Mr 16 000 in both the red and orange forms, but this method removed the chromophore from the proteins. The main carotenoid component of the complex was determined to be 3′-hydroxy-4-keto-ββ-carotenoid or 3′-hydroxyechinenone. The number of carotenoid molecules per molecule of orange protein of molecular weight 47 000 was between 20 and 40. The stoichiometry of carotenoid to protein seemed reasonably constant.  相似文献

Core histones have been isolated from Schizosaccharomyces pombe and compared electrophoretically to core histones from Saccharomyces cerevisiae and rat liver. The molecular masses of all cognate histones examined were found to be very similar as determined by SDS gel electrophoresis. Histones H3, H2A and H2B from Sch. pombe migrated almost identically to their respective counterparts from S. cerevisiae as determined by acid/urea gel electrophoresis. Two-dimensional gel electrophoresis with a Triton X-100 acid/urea gel in the first dimension followed by an SDS gel in the second dimension was used to separate Sch. pombe histones from contaminating ribosomal proteins.  相似文献

Prolactin coding mRNA was shown to be a prevalent part of chum salmon (Oncorhynchus keta) pituitary poly(A)-RNA during the spawning period. Clone lambda gtPrk12 was selected from the pituitary cDNA library by means of hybridization with the prolactin probe, and a nucleotide sequence of the insertion was determined and compared to the prolactin coding sequences from rainbow trout and Pacific chinook salmon, which had been published earlier. The sequences compared exhibited a significant homology. The deduced amino acid sequence of the chum salmon prolactin differed from a sequence determined directly in a single position. The prolactin-coding sequence can be used for constructing the bacterial strain producing prolactin.  相似文献

The primary structure of thioredoxin, a redox protein isolated from Chromatium vinosum, was determined by high-performance tandem mass spectrometry, which permitted sequencing of the 14 peptides (ranging in length from 2 to 18 amino acids) generated by digestion with trypsin and of several peptides produced by Staphylococcus aureus protease. The mass spectrometrically determined molecular weights of the peptides from the latter digest were used to properly align the tryptic peptides, which could also be accomplished on the basis of the considerable homology with Escherichia coli thioredoxin. Finally, the molecular weight of the Chromatium thioredoxin was determined by mass spectrometry and found to be 11,748.0, in good agreement with 11,750.2 calculated for the proposed sequence. Although it was difficult to establish by mass spectrometry, five leucines and three isoleucines could be identified, leaving only eight undifferentiated.  相似文献

A polyomavirus was isolated from the eyes of horses, and the sequence was determined. A nearly identical VP1 sequence was amplified from the kidney of another animal. We report the complete genome sequence of the first polyomavirus to be isolated from a horse. Analysis shows it to be most closely related overall to human and nonhuman primate polyomaviruses.  相似文献
