Zinc (Zn), lead (Pb) and cadmium (Cd) tolerance in populationsof seedlings ofPhragmites australisraised from seeds collectedfrom a mine site (Plombières, Belgium) contaminated withZn, Pb and Cd and three ‘clean’ sites (Felixstowe,UK; Wisbech, UK; and Mai Po, Hong Kong) were studied under glasshouseconditions. Small differences were found between the metal-contaminatedpopulation and the three ‘clean’ populations whenseedlings were grown in 1.0 µg  ml-1Zn and 10.0 µgml-1Pb treatment solutions. In general, however, different populationsof seedlings showed similar growth responses, metal uptake andindices of Zn, Pb and Cd tolerance when cultured in the samemetal-contaminated media for 89 d or in the same metal treatmentsolutions (ZnSO4:1.0 and 4.0 µg ml-1Zn; Pb(NO3)2: 10.0and 25.0 µg ml-1Pb; CdSO4: 0.5 and 1.0 µg ml-1Cd)for 3 weeks. There was insufficient evidence to support thehypothesis that the metal-contaminated population has evolvedto a Zn-, Pb- or Cd-tolerant ecotype but the results indicatedsome differentiation between the populations with that fromHong Kong being the least productive under the experimentalconditions used. The implications of the findings on selectionof provenances for use in constructed wetlands for wastewatertreatment are discussed. The increase of -amylase activity in embryoless rice endosperminduced by the addition of gibberellin A3 was examined undervarious conditions with an aim to establish a bioassay methodfor gibberellins. Sterilized embryoless rice endosperms were incubated in a testtube containing 0.2–1.0 ml of test solution for 4 daysat 30. -Amylase activity in the endosperm was determined bymeasuring digestion of added starch. The increase of amylaseactivity during the incubation was not affected by the additionof various vitamins, amino acids, organic acids, protease, sucrose,indoleacetic acid or kinetin. Helminthosporol, helminthosporicacid and sclerin (1–10 µg/ml) had weak promotingeffects. Under appropriate conditions, 10–5 µg/mlof gibberellin A3 could be detected. In double logarithmic plot,the increase in the enzyme activity was proportional to thegibberellin A3 concentration in the range from 10–5 to10–2 µg/ml. (Received April 13, 1966; )  相似文献

The amount of gibberellin-like substances in the seed of Pharbitisnil increased in parallel with the growth of the seed, and attained20 days after anthesis to a maximum of 0.115 µg gibberellinA3 eqivalent per seed, when the seed reached its maximum freshweight or four-fifths of its final dry weight. At this maximumlevel, 0.03 µg and 0.1 µg gibberellin A3 equivalentswere localized in the embryo and in the "endosperm", respectively.Three gibberellin-like substances (Factors I, II and III) wereseparated upon paper chromatography. In view of the changesin amount of the factors with respect to the seed maturation,these factors, especially Factor II, in the "endosperm" andembryo were assumed to participate in the initial or the mainpart of growth of the embryo. Dwarf rice seedling and maizedwarfs 1, 3 and 5 responded to the three factors nearly in thesame way. Pharbitis dwarf, however, responded only to FactorI, but not to Factors II and III. Effects of three phytohormones (IAA1, GA3 and kinetin) on grain-fillingand the pattern of 32P translocation from individual leavesto grains were studied at intervals of 7 days during the progressof reproductive development of rice (Oryza saliva L. cv. Jaya).The plants were sprayed with 100 µg ml–1 aqueoussolutions of the hormones at 100 days, when the plants wereentering the reproductive stage. Kinetin produced a pronouncedeffect on grain-filling as well as on 32P mobilization fromindividual leaf to grains and increased yield, possibly by increasingleaf longevity. GA3 and IAA also increased the grain-fillingand 32P mobilization significantly over control but the effectswere less marked than those of kinetin. Maize (Zea mays L., hybrid Cargill 147) seedlings, grown inaseptic conditions, were inoculated with three strains of Azospirillumlipoferum (Al op 33, Al iaa 320, and ATCC 29708) or culturedin different concentrations of indol-3-acetic acid (IAA) orgibberellin A3 (GA3). After 48 h, root length, root surfacearea, root dry weight, and shoot dry weight were measured inall treatments. Gibberellin content was evaluated in selectedroots of control and Azospirillum inoculated seedlings by gaschromatography-mass spectrometry-selected ion monitoring withthe use of deuterated gibberellins as internal standards. Thethree strains of A. lipoferum, IAA (2 ng ml–1), and GA3(40 to 400 pg ml–1) significantly enhanced root growth.Improvement of root hair growth and density was obtained mainlywith A. lipoferum strain Al op 33 and GA3 40 pg ml–1.GA3 was identified by gas chromatography-mass spectrometry (byboth, full scan and selected ion monitoring) in a free acidfraction from roots of the seedlings inoculated with A. lipoferum.In the roots of the non inoculated seedlings GA3 was found afterhydrolysis of a fraction expected to contain glucosyl conjugates. A synthetic brassinosteroid, 22,23(S,S)-homobrassinolide (hBR),was examined for its interaction with IAA and GA3 in the elongationof hypocotyl sections of light-grown cucumber (Cucumis salivusL. cv. Aonagajibai) seedlings. hBR alone was less active thanIAA. Its optimal concentration was around 10 µM and thelowest effective concentration between 10 and 100 µM,which is more than 100 times higher than that of brassinolide.hBR was more active in sections from younger seedlings. Itsgrowth-promoting effect was negated or greatly reduced by inhibitorsof auxin-induced elongation such as p-chlorophenoxyisobutyricacid and kinetin. hBR acted synergistically with IAA and 2,4-Dbut not with GA3 showing only an additive effect. Sequentialtreatment of sections with hBR and then with IAA also resultedin synergistic enhancement of auxininduced elongation, but whenthe order of treatment was reversed, hBR was inactive. The synergisticeffect was obtained with 1 h pretreatment with hBR and couldbe reduced by subsequent washing with water. There was no sequentialinteraction between hBR and GA3. The synergistic pretreatmenteffects of hBR and GA3 were simply additive to each other. Amembrane-bound ATPase inhibitor, dicyclohexylcarbodiimide, inhibitedthe hBR-induced elongation, but did not affect GA3-induced elongation.The findings led to the conclusion that brassinosteroids enhanceauxin action and possess growth-promoting activity which isindependent of that of gibberellin. The spawning rate of laboratory-reared Centropages typicus fedHymenomonas elongala increases with food concentration, up toa value of {small tilde}2800 µg C (16 500 cells) ml–1.An alternation of a low food (1000 cells ml–1) and highfood concentration (16 500 cells ml–1) is not favourableto egg release when its periodicity is 1 or 2 days, whereasit may be of advantage if it is longer (3–6 days). Inthe latter case, Centropages typicus will benefit best fromthe rich food diet if this coincides with (or just follows)the last moult.  相似文献

Two gibberellin-like substances were separated chromatographicallyfrom the ethanol-extract of germinating cotyledons of Pharbitisnil. In the dark-grown seedling, these substances in the cotyledonincreased in amount gradually with the slow increase in freshweight and area of the tissue, and attained, 4 days after sowing,a maximum level of 0.03 µg gibberellin A3 equivalent pera couple of cotyledon; in the light-grown seedling, they increasedrapidly with the rapid increase in the fresh weight and area,and attained a maximum content of 0.07 µg gibberellinA3 equivalent per cotyledon on the 6th day. When the etiolatedseedling was exposed to light, the amount of the gibberellin-likesubstances in the cotyledon increased in parallel with openingof the hypccotyl hook and expansion of the cotyledon. The openingof the hook and increase of the substances occurred in red lightas well as in far-red. Relatively small amounts of the gibberellin-like substanceswere also contained in the germinating hypocotyl and root. The growth of cotyledonary disks was promoted by gibberellinA3 and the gibberellin-like substances from the expanding cotyledon. Plants of Lamium amplexicaule, grown under short-day field conditionsin Northern California produce predominantly closed flowers.Under long-day field conditions, plants may produce up to 50per cent open flowers, the same total number of flowers beingproduced under each daylength regime. A 20 ml drop of GA3 or GA7 (100µM) was applied to themain shoot apex of plants growing under short-day conditions,and all subsequently produced flowers opened. CCC, an inhibitorof gibberellin synthesis, was applied (0.06 per cent) to thesoil of seedlings grown under long-day conditions. The CCC-treatedplants were dwarfed and bore only closed flowers. With GA7 appliedexogenously to the CCC-treated shoots, inhibition was released,resulting in elongated internodes and open flower production.The timing of flower production and internodal extension inLamium amplexicaule are positively correlated. When floral primordiawere removed from main shoots, the average internode lengthsdecreased. The number of nodes produced in treated plants wasincreased as a result of flower removal. Exogenous GA7 (10 µM)applied to the nodes from which flowers were removed resultedin internodal extension but had no effect on node number. Twoprocesses that may contribute to the control of the productionof open and closed flowers in Lamium amplexicaule are: (1) anincreasing anther sac size from lower to upper node flowersthat may exert control locally, via GN production, on corollaexpansion, and (2) a photoperiodic control. Gibberellins present in cotyledonary embryos of Phaseolus coccineusL. have been identified by combined gas chromatography-massspectrometry as GA1, GA4, GA5, GA6; also an unidentified gibberellinwas present. The total amount of gibberellins was estimated by gas chromatographyto be 26.5 µg per g fresh weight; the individual gibberellinscontributed to the total amount as follows: GA1 18.4µg,GA5 2.6 µg, GA4 2.3 µg, GA6 1.9 µg, unidentifiedGA 1.3 µg. The amount of gibberellin-like substances in seed, pod, embryoand "endosperm" of Lupinus luteus in relation to their developmentwas studied by means of the rice seedling test. The amount of gibberellin-like substances per seed increasedremarkably at the first stage of development, when the growthof seed and pod was very slow, and attained its maximum at suchan early stage as the dry weight of seed reached a value ofonly 10 per cent and that of pod 28 per cent of their respectivemaximum levels. Changes in amount of gibberellin-like substancespresent in the pod were very closely related to the changesin growth of pod. Gibberellin-like substances equivalent to0.3 µg gibberellin A3 per seed and 0.028 µg perpod were found at the 20th day after anthesis, and no activityin mature seed and 0.09 µg per pod at the 50th day. Noactivity was found in the embryo itself at all stages, indicatingthat the gibberellin-like substances in seed occur only in the"endosperm," i.e. in the tissues that will eventually form testain fully ripened stage. Growth in lengths of tomato stems and leaves was acceleratedby 5.0 µg gibberellic acid (GA2) applied to the seed,or by 5.0, 0.5, and 0.05 µg given to the roots. Treatmentwith 5.0 µg also decreased bud number and lengthened thetime between bud appearance and fruit formation on the firsttruss by 1–8 days. Smaller amounts applied to roots shortenedthis time by 1–4 days. Indol-3yl-acetic acid at 0.5 µghad no effect, nor was simultaneous application of GA3 and IAAto the roots more effective than GA3, alone. Single applicationsof very small amounts of GA3 to seeds or seedling roots thusproved capable of changing growth-rates of stems, leaves, andtrusses.The effects of treating tomatoes with GA2 and with culturesof Azotobacter chroococcum, which contain small amounts of GA3,and IAA, are compared.  相似文献

Interactions between the growth retardants daminozide (a substitutedsuccinamic acid) or piproctanyl bromide (a quaternary ammonium,piperidinium compound), and a subsequent application of a singledose (40 µg) of either gibberellin A1, A3, A4+7 or A12,showed that, in Chrysanthemum morifolium Ramat. cv. Bright GoldenAnne, a strong interdependence exists between elongation ofthe lateral shoot and the rate of development of its terminalinflorescence. A1, A3, and A4+7 were highly active in overcoming the restrictionson both internode extension and the rate of flower-bud developmentimposed by either retardant, suggesting that these two retardanteffects are caused by a deficiency of active gibberellins (GN).In the absence of retardant, A1, A3, and A4+7 markedly increasedstem elongation, and flowering occurred earlier than in plantsreceiving neither retardant nor GN. A13 the only 20-carbon GNtested, was much less active, while A5 had a relatively greatereffect on the time of flowering than on shoot elongation. Thus,it is not necessarily the rate of stem extension which determinesthe rate of inflorescence development. The response to different amounts of A1, A3 or A13 (1, 5, 10,20, or 50 µg per shoot) neither suggest that different‘threshold’ levels of a particular GN are requiredto induce increases in either stem elongation or in the rateat which inflorescences develop, nor did a change in the dosegiven lead to any consistent differential effect on these twoprocesses. By using light-grown cucumber (cv. Aonagajibae) seedlings withfully expanded cotyledons, the osmotic potential of the epidermalcells of the hypocotyl, as determined by the plasmolysis methodwas examined in relation to the effects of gibberellin (GA7)and cotyledons. GA7 at a dosage level of 0.05 µg/plantapplied to the apical bud lowered the osmotic potential; thedecrease was significant 5 hr after the GA7 treatment. Removalof the cotyledons, covering them with aluminum foil, or applyinglinuron, a DCMU analogue, to them caused an increase in thepotential. The causes of interspecific differences in the µ-l relationshipare examined in the context of a mechanistic model which relatesµ to irradiance in terms of six factors:, kc photosyntheticquotient (PQ), Chl a:C, respiration and excretion. The effectof cell size on the light saturated growth rate is also considered.It is shown that photosynthetic efficiency and PQ exhibit remarkablylittle interspecific variability, and average 0.024 ±0.005 µg C(µg Chl a)–1 h–1 (µEm–2 s–1)–1 and 1.5 ± 0.2 mol 02 molC–1 (when NO3 is the nitrogen source) respectively.Two useful relationships were derived: (i) between growth efficiency,g and Chl a:C at µ. = 0; (ii) between the compensationintensity, Ic and the Chl a-specific maintenance respirationrate. Both relationships were independent of temperature anddaylength. Species best adapted to growth at low light werefound to exhibit high Chl a:C ratios and low maintenance respirationrates. As a group, diatoms were consistently the best adaptedfor growth at low irradiance. Chiorophytes, haptophytes, chrysophytesand cryptophytes were intermediate in their performance at lowirradiance. Dinoflagellates exhibited extreme diversity, withspecies spanning the spectrum from very good performance atlow irradiance to very poor. A new µmax-cell carbon relationshipis given based on growth rates normalized to 15°C. Evidenceis presented to show that noise in this relationship can besignificantly reduced by using only carbon-specific growth ratesand using only data for species grown at the same daylength.  相似文献

The stomatal response of seedlings grown in 360 or 720 µmolmol–1 to irradiance and leaf-to-air vapour pressure deficit(VPD) at both 360 and 720 µmol mol–1 to CO2 wasmeasured to determine how environmental factors interact withCO2 enrichment to affect stomatal conductance. Seedlings offour species with different conductances and life histories,Cercis canadensis (L.), Quercus rubra (L.), Populus deltoides(Bartr. ex Marsh.) P. nigra (L.), and Pinus taeda (L.), weremeasured in hopes of identifying general responses. Conductanceof seedlings grown at 360 and 720 µmol mol–1 CO2were similar and responded in the same manner to measurementCO2 concentration, irradiance and VPD. Conductance was lowerfor all species when measured at 720 than when measured at 360µmol mol–1 CO2 at both VPDs ({small tilde}1.5 and{small tilde}2.5 kPa) and all measured irradiances greater thanzero (100, 300, 600,>1600 µmol m–2 S–2)The average decrease in conductance due to measurement in elevatedCO2 concentration was 32% for Cercis, 29% for Quercus, 26% forPopulus, and 11% for Pinus. For alt species, the absolute decreasein conductance due to measurement in CO2 enrichment decreasedas irradiance decreased or VPD increased. The proportional decreasedue to measurement in CO2 enrichment decreased in three of eightcases: from 0.46 to 0.10 in Populus and from 0.18 to 0.07 inPinus as irradiance decreased from>1600 to 100 µmolm–2 s–1 and from 0.35 to 0.24 in Cercis as VPD increasedfrom 1.3 to 2.6 kPa. Seeds of Sorghum bicolor (L.) Moench. were germinated on moistfilter paper for 6 d, before the seedlings were transferredto pots containing 500 µmol l-1 Ca(NO3)2 for 2 d. Theseedlings were then treated with 0 or 100 µmol l-1 Alin factorial combination with 0, 1400 or 2800 µmol l-1Si for 8 d. The background solution used throughout was 500µmol l-1 Ca(NO3)2. Aluminium treatment reduced root growthand caused a significant increase in shoot/root ratio. The presenceof silica in the solution significantly ameliorated the effectsof aluminium on root growth. Three treatment were selected for a microanalytical investigationof the basal region of the root: 2800 µmol l-1 Si only;100 µmol l-1 Al only; and a combination of the two. Inthe 2800 µmol l-1 treatment silica was deposited in theendodermis, with the greatest accumulation being in the innertangential wall (ITW). When plants were treated with 100 µmoll-1 Al only, aluminium concentration was highest in the outertangential wall (OTW) of the epidermis. The element was presentin the hypodermal walls and OTW of the endodermis and was notdetectable in the stele. With both 2800 µmol l-1 Si and100 µmol l-1 Al in the nutrient solution the two biomineralizationsites were the ITW of the endodermis, where silicon was themajor element deposited, and atypically in the OTW of the epidermiswhere both aluminium and silicon were present. The sequestrationof aluminium in the Al-Si deposit in the OTW of the epidermismay represent the mechanism that allows greater root growthin this treatment.Copyright 1993, 1999 Academic Press Sorghum bicolor (L.) In 5-d-old etiolated seedlings of Sorghum bicolor, 12 h of darknessafter 5 min in red light eliminated a lag before the accumulationof chlorophylls in subsequent continuous white light. Increasingthe dark period to 24 h and 36 h, increased the rate of chlorophyllaccumulation in the later stages of greening. Exogenous -aminolevulinicacid neither completely removed the lag, nor increased the rateof chlorophyll accumulation. Cycloheximide (25 µg ml–1)and 6-methyl purine (5.0 µg ml–1), given continuouslyor only until the 12 h dark period following the red light irradiation,restored the lag and decreased the rate of chlorophyll accumulation.D-threo-chloramphenicol (400µg ml–1) also decreasedthe rate of chlorophyll accumulation but did not restore thelag. Addition of these inhibitors even 12 h after red lightirradiation decreased the rate of chlorophyll accumulation.Rifampicin (Rifamycin SV, 400 µg ml–1) did not havesuch effects. 3rß-Fluorogibberellin A9 (3rß-fluoro-GA9),3rßfluoro-GA20, 3rß-fluorodeoxygibberellinC (3rß-fluoro-DGC) and 13-fluoro-GA9 were prepared,and their effects on plant growth and gibberellin (GA) 3rß-hydroxyIaseswere examined. 3rß-Fluoro-GA9 and 3rß-fluoro-GA20promoted the growth of dwarf rice (Oryza sativa L. cv. Tan-ginbozu)seedlings to three times higher than the control seedlings ata dosage of 3 µ plant–1, and 3rßfluoro-DGCto twice higher at the same dosage. 3rßg-Fluoro-GA9was active in cucumber (Cucumis sativus L.) hypocotyl assay,its activity being about one-thirtieth as much as that of GA4.3rß-Fluoro-GAs were active per se in promoting theshoot elongation of rice. 3rß-Fluoro-DGC inhibitedthe 3rß-hydroxylation of [3H2]GA9 to [3H]GA4 by GArß-hydroxylase from bean (Phaseolus vulgaris L.),but 3rß-fluoro-GA9 and 3rß-fluoro-GA20 didnot show any effects on the enzyme activity. These 3rß-fluoro-GAsalso showed no or only a weak inhibitory effect on the rß-hydroxylasefrom pumpkin (Cucurbita maxima L.). 13-Fluoro-GA9 promoted growthof rice and cucumber seedlings, and inhibited the 3rß-hydroxylasesfrom both bean and cucumber. 13-Fluoro-GA9was converted into13-fluoro-GA4 and 2,3-didehydro-13-fluoro-GA9, in a cell-freesystem from bean, and conversion of 13-fluoro-GA9 into 13-fluoro-GA4was also observed in a cell-free system from pumpkin. Theseresults suggest that 13-fluoro-GA9 is one of the substratesof GA 3rß-hydroxy-lases, and that 13-fluoro-GA9 isactive as a result of the conversion to 13-fluoro-GA4 in riceand cucumber seedlings. Acclimation of fully developed leaves of Bischofia javanicaBlume to shadelight was examined. Seedlings were grown undersimulated daylight (1000 µmol m–2 s–1), thentransferred to a simulated shadelight (40 µmol m–2s–1). When a high-light leaf was transferred to low light, large negativenet photosynthetic rates (Pm) were recorded. This decrease wasrapid, but within 7 d the rate increased and became equal tothe low-light control leaf. These changes in photosynthesisdid not follow the pattern of changes in stomatal conductance(gs). Transfer to the low light resulted in a dramatic decreasein leaf weight per unit area (Lw), and most of the decreasesin Lw occurred within 3 d of transfer when the Pm of the transferredleaf was well below that of the low-light control leaf. There was a significant decrease in chlorophyll a in the transferredleaf without an appreciable change in chlorophyll b resultingin a large decrease in the chlorophyll a to chlorophyll b ratio.Leaf chlorophylls per unit area were higher in the transferredleaf than the low-light control leaf. Maximum photosyntheticrate in the transferred leaf was decreased by 40% compared tothat for the high-control leaf, but was almost at the same extenthigher than the low-light control leaf The results are discussedin the context of carbon gain capacity of its seedlings underlight-limiting forest understorey habitats.
