The role of defence gene expression triggered by Cd toxicity in the plant’s response to Botrytis cinerea was investigated in Arabidopsis thaliana Columbia 0. Silicon (0 or 1.5 mM) and Cd (0, 1 or 10 μM) were supplied to 3-month-old solution-cultured plants. After 3 days, half of the plants of each treatment were inoculated with Botrytis. Supplied Cd concentrations were below the toxicity threshold and did not cause shoot growth inhibition or evidence of oxidative stress, while Botrytis infection severely decreased plant growth in all treatments. The expression of marker genes PR1 and BGL2 for the salicylic acid (SA) and the PDF1.2 for the jasmonic acid–ethylene (JA–ET) signalling pathways was enhanced in 10 μM Cd-treated non-infected plants. Twenty hours after inoculation, PDF1.2 expression showed a strong increase in all treatments, while enhanced PR1, BGL2, and CHIB expression was only found 7 days after infection. A great synergistic effect of Cd and Botrytis on PDF1.2 expression was found in 10 μM Cd-treated plants. Silicon decreased PR1, BGL2, and CHIB, while increasing PDF1.2 expression, which indicates its role as a modulator of the signalling pathways involved in the plant’s response to fungal infection. Botrytis growth decreased in 10 μM Cd-treated plants, which could be due to the combined effects of Cd and Botrytis activating the SA and JA–ET-mediated signalling pathways. Taken together, our results provide support for the view that Cd concentrations close to the toxicity threshold induce defence signalling pathways which potentiate the plant’s response against fungal infection.  相似文献

Increasing evidence indicates that selenium (Se) could antagonize metal toxicity, including cadmium (Cd) toxicity. However, the effects of Se on Cd-induced changes in the ion profile in the pancreas of chickens have not been reported. In the present study, 128 Hy-Line brown laying chickens were divided into the control group, Se-treated group, Se/Cd-treated group, and Cd-treated group, and we detected the concentrations of 28 ions in the four groups by inductively coupled plasma mass spectrometry. In the Cd-treated group, the accumulation of Cd in the pancreas was 836.8 times higher that than in the control group (27,353.71 ppb/32.69 ppb). Meanwhile, the Ca, Ti, Fe, Mo, Li, Al, and Pb levels increased and the Cr, Mn, Ni, Cu, Zn, Se, Sr, and Sb levels decreased due to sub-chronic Cd poisoning. The Fe, Mo, Ba, and Pb levels decreased in the Se/Cd-treated group. Our findings suggest that Cd can accumulate in the chicken pancreas and affect the ion profiles, whereas Se can ameliorate the accumulation of Cd and change the ion profiles in the chicken pancreas.  相似文献

Formation of lipid hydroperoxides, malondialdehyde (MDA) and hydroxyalkenals (HAEs), membrane damages and antioxidative response of plants expressed as changes in glutathione S-transferase activity (GST) and anthocyanin accumulation were studied in Arabidopsis thaliana (L.) Heynh cv. Columbia plants treated for 7 days with various concentrations: 5, 25, 50, 100 μM Cd and Cu. Increased lipid hydroperoxide content was metal concentration-dependent. The level of MDA + HAE was elevated in Cd- and Cu- treated plants, but it was metal concentration-dependent under Cu stress. Electrolyte leakage measurements showed a larger membrane damage under Cu- than Cd-treatment. In Cu-stressed plants, GST activity was always enhanced in comparison with control, while in plants exposed to Cd it dropped slightly at lower metal concentrations; but at 100 μM Cd it was even higher than in plants treated with the same Cu concentration. Anthocyanin accumulation was considerably higher under Cu than Cd stress. Both lipid peroxidation and antioxidative response was stronger in Cu- than Cd-treated Arabidopsis thaliana plants. Various mechanisms of defense against the lipid peroxidation products, depending on the metal type, are discussed.  相似文献

To determine the negative effects of cadmium (Cd) exposure and the protective role of selenium (Se) on Cd-spiked neutrophils of chicken, forty-eight 28-day-old Isa Brown male chickens were divided randomly into four groups. Group I (control group) was fed with the basic diet containing 0.2 mg/kg Se. Group II (Se-treated group) was fed with the basic diet supplemented with Na2SeO3, and the total Se content was 2 mg/kg. Group III (Se/Cd-treated group) was fed with the basic diet supplemented with Na2SeO3; the total Se content was 2 mg/kg and supplemented with 150 mg/kg CdCl2. Group IV (Cd-treated group) was fed with the basic diet supplemented with 150 mg/kg CdCl2. Analyses of inflammatory factors, cytokines, and heat shock protein (Hsp) messenger RNA (mRNA) expression were detected by real-time PCR (RT-PCR). Additionally, we evaluated the phagocytic rate of neutrophils in peripheral blood. First, we observed that Cd significantly induced the mRNA expression levels of inflammatory factors NF-κB, iNOS, COX-2, and TNF-α, while Se/Cd treatment reduced their mRNA expression, although these expression levels remained higher than that of the control group. In addition, the mRNA expression levels of cytokines (IL-2, IL-4, and IL-10) for the Se-treated group exhibited significant differences between the Se/Cd-treated group and the Cd-treated group. Furthermore, the mRNA expression levels of Hsps demonstrated that the Se/Cd-treated group and the Cd-treated group were significantly higher (P < 0.05) than the control group and the Se-treated group. These results demonstrated that Se presented partial protection on Cd-spiked neutrophils of chicken with Hsps being involved in the process of the Cd-spiked toxic effects in chicken peripheral blood neutrophils.  相似文献

Oxidative stress in obesity leads to insulin resistance in type 2 diabetes. Some selenoproteins possess antioxidant properties, suggesting that selenium (Se) may protect against type 2 diabetes; however, evidence from epidemiological studies is contradictory. We hypothesized that Se status before supplementation (baseline) contributes to the supplementation outcome. This study aimed to clarify the influence of baseline Se status on the effect of Se supplementation on the diabetic condition. Six-week-old KKAy mice were fed a diet without supplemental Se or with 0.1 ppm Se in the form of l-selenomethionine (SeM) for 2 weeks to create low-Se and sufficient-Se baseline statuses, respectively. For the next 4 weeks, low-Se mice were given a SeM (0.5 ppm Se)-supplemented diet, and sufficient-Se mice were given either a SeM (0.5 ppm Se)- or sodium selenite (0.5 ppm Se)-supplemented diet; control groups continued on baseline diets. Serum Se concentrations, glutathione peroxidase (GPx) activities, adiponectin levels, glucose tolerance, and insulin sensitivity were analyzed. All mice became diabetic during the 2-week baseline induction period. At the end of the supplementation period, Se-receiving groups demonstrated significantly higher Se concentrations and GPx activities than their respective controls. Sufficient-Se mice receiving SeM had lower blood glucose levels and better insulin sensitivity than control and sodium selenite-receiving mice, whereas low-Se mice receiving SeM showed no such improvements compared with their controls. Our results suggest that Se supplementation in the form of SeM may help prevent type 2 diabetes aggravation in people taking the 55 μg/day Se recommended dietary allowance.  相似文献

The objective of this study was to investigate the effect of selenium (Se) supply (0, control; 2.5, 5, 10, or 20 μM) on cucumber (Cucumis sativus L.) cv. Polan F1 plants grown under short-term low temperature stress. About 14–16 day-old seedlings, grown at an optimal temperature (25/20°C; day/night), were exposed to short-term chilling stress with a day/night temperature of 10°C/5°C for 24 h, for a further 24 h at 20°C/15°C, and then transferred to 25/20°C (re-warming) for 7 days. Se did not affect the fresh weight (FW) of plants at a concentration of 2.5–10 μM, but in the presence of 20 μM Se, the biomass of shoots significantly decreased. The contents of chlorophylls and carotenoids witnessed no significant change after Se supplementation. Compared with the control, the Se-treated plants showed an increase of proline content in leaves, once after chilling and again after 7 days of re-warming. However, proline levels were much higher immediately after chilling than after re-warming. The malondialdehyde (MDA) content in the root of plants treated with 2.5–10 μM Se decreased directly after stress. This was in comparison with the plants grown without Se, whereas it increased in roots and leaves of plants exposed to 20 μM Se. Seven days later, the MDA level in the root of plants grown in the presence of Se was still lower than those of plants not treated with Se and generally witnessed no significant change in leaves. Although Se at concentrations of 2.5–10 μM modified the physiological response of cucumber to short-term chilling stress, causing an increase in proline content in leaves and diminishing lipid peroxidation in roots, the resistance of plants to low temperature was not clearly enhanced, as concluded on the basis of FW and photosynthetic pigments accumulation.  相似文献

The effects of varying concentrations of cadmium (Cd) on the development of Lycopersicon esculentum cv. Micro‐Tom (MT) plants were investigated after 40 days (vegetative growth) and 95 days (fruit production), corresponding to 20 days and 75 days of exposure to CdCl2, respectively. Inhibition of growth was clearly observed in the leaves after 20 days and was greater after 75 days of growth in 1 mM CdCl2, whereas the fruits exhibited reduced growth following the exposure to a concentration as low as 0.1 mM CdCl2. Cd was shown to accumulate in the roots after 75 days of growth but was mainly translocated to the upper parts of the plants accumulating to high concentrations in the fruits. Lipid peroxidation was more pronounced in the roots even at 0.05 mM CdCl2 after 75 days, whereas in leaves, there was a major increase after 20 days of exposure to 1 mM CdCl2, but the fruit only exhibited a slight significant increase in lipid peroxidation in plants subjected to 1 mM CdCl2 when compared with the control. Oxidative stress was also investigated by the analysis of four key antioxidant enzymes, which exhibited changes in response to the increasing concentrations of Cd tested. Catalase (EC 1.11.1.6) activity was shown to increase after 75 days of Cd treatment, but the major increases were observed at 0.1 and 0.2 mM CdCl2, whereas guaiacol peroxidase (EC 1.11.1.7) did not vary significantly from the control in leaves and roots apart from specific changes at 0.5 and 1 mM CdCl2. The other two enzymes tested, glutathione reductase (EC 1.6.4.2) and superoxide dismutase (SOD, EC 1.15.1.1), did not exhibit any significant changes in activity, apart from a slight decrease in SOD activity at concentrations above 0.2 mM CdCl2. However, the most striking results were obtained when an extra treatment was used in which a set of plants was subjected to a stepwise increase in CdCl2 from 0.05 to 1 mM, leading to tolerance of the Cd applied even at the final highest concentration of 1 mM. This apparent adaptation to the toxic effect of Cd was confirmed by biomass values being similar to the control, indicating a tolerance to Cd acquired by the MT plants.  相似文献

Despite the well-established toxicity of cadmium (Cd) to animals and the ameliorative effects of selenium (Se), some specific mechanisms in the chicken ovary are not yet clarified. To explore the mechanism by which the toxicity effect of Cd is induced and explore the effect of supranutritional Se on Cd toxicity in female bird reproduction, forty-eight 50-day-old Isa Brown female chickens were divided randomly into four groups. Group I (control group) was fed the basic diet containing 0.2 mg/kg Se. Group II (Se-treated group) was fed the basic diet supplemented with sodium selenite (Na2SeO3), and the total Se content was 2 mg/kg. Group III (Se + Cd-treated group) was fed the basic diet supplemented with Na2SeO3; the total Se content was 2 mg/kg, and it was supplemented with 150 mg/kg cadmium chloride (CdCl2). Group IV (Cd-treated group) was with the basic diet supplemented with 150 mg/kg CdCl2. The Cd, estradiol (E2), and progestogen (P4) contents changed after subchronic Cd exposure in chicken ovarian tissue; subsequently, oxidative stress occurred and activated the endoplasmic reticulum (ER) pathway to induce apoptosis. Further, Se decreased the accumulation of Cd in ovarian tissue, increased the E2 and P4 contents, alleviated oxidative stress, and reduced apoptosis via the ER stress pathway. The present results demonstrated that Cd could induce apoptosis via the ER stress pathway in chicken ovarian tissue and that Se had a significant antagonistic effect. These results are potentially valuable for finding a strategy to prevent Cd poisoning.  相似文献

Exposing female tilapia to Cd did not affect reproduction and egg quality, nor the survival of their offspring. The LT 50 of Cd-exposed larvae (within 24 h post hatching) from Cd-treated female Oreochromis mossambicus (mean ±S.E.=10·9±0·5 days, n =4) was greater than that of Cd-exposed larvae from control females (6·9± 1·2 days, n =4). A substantial amount of metallothionein mRNA (MT mRNA) was found in both oocytes and newly hatched larvae from Cd-treated females, but MT mRNA levels were low in newly fertilized eggs. The amount of MT mRNA was very low in all these stages in the control group. Cadmium could not be detected at any stage for either control or Cd-treated group, and metallothionein protein levels were also very low in both groups. The early appearance of MT mRNA in oocytes and newly hatched larvae from Cd-treated females may result in early translation after hatching and explain their tolerance to Cd.  相似文献

This study aims at revealing the effects of the combined treatment of selenium and cadmium on ovary morphology, oxidative stress, and 28 kinds of ion concentrations in laying hens. In this experiment, 128 healthy 31-week-old chickens were selected and divided into four treatment groups, three of which were separately fed the basic diets supplemented with either Se or Cd or both Se and Cd for 90 days, and the remaining group was fed the basic diet and treated as a control. The chickens were sacrificed for collecting ovarian tissues. Morphological structure and ultrastructure analysis of ovaries in the Cd-treated group revealed ovarian damage, with decreased activities of SOD and GPx, along with increased levels of MDA and H2O2. Cd treatment also resulted in disturbances in ion balance. The concentrations of Ca, Ti, Cu, Zn, and Ba were significantly reduced, while the concentrations of Fe, Mo, and Cd were significantly increased when compared with the control group. Interestingly, the damages caused by cadmium were alleviated in the Se+Cd-treated group. These results indicate that selenium can alleviate cadmium-induced ovarian damages.  相似文献

Salicylic acid (SA) may accelerate the cell death of cadmium-stressed roots to avoid cadmium (Cd) uptake by plants or may play positive roles in protecting the stressed roots from Cd-induced damage. To test these hypotheses, we performed a series of split-root hydroponic experiments with one-half of rice (Oryza sativa L. cv. Jiahua 1) roots exposed to 50 microM Cd and the other half not exposed. The objectives were to elucidate the effects of SA pretreatment on the time-dependent changes of H(2)O(2) levels in roots, antioxidant defense system in different organs, root cell death and the dynamic distribution of Cd in the plants. In the split-root system, a higher Cd uptake rate was observed in the Cd-stressed portions of roots compared with the treatment with the whole roots exposed to Cd. Furthermore, an appreciable amount of Cd was translocated from the Cd-exposed roots to the unexposed roots and trace amounts of Cd were released into the external solution. The split-root method also caused the two root portions to respond differently to Cd stress. The activities of major antioxidant enzymes (superoxide dismutase, SOD; peroxidase, POD; and catalase, CAT) were significantly suppressed in the Cd-treated roots, hence leading to H(2)O(2) burst, lipid peroxidation, cell death and growth inhibition. By contrast, in the non-Cd-treated roots, the activities of enzymes (SOD, CAT, and POD) and root growth were persistently stimulated during the experimental period. The H(2)O(2) accumulation and lipid peroxidation were also induced in the non-Cd-treated roots, but they were significantly lower than those of the Cd-treated roots. The concentrations of glutathione (GSH) and non-protein thiols (NPT) in the Cd-treated roots were significantly higher than those of the untreated roots. SA pretreatment elevated enzymatic and non-enzymatic antioxidants, and the concentrations of GSH and NPT in roots and shoots, hence leading to alleviation of the oxidative damage as indicated by the lowered H(2)O(2) and MDA levels. Furthermore, SA pretreatment mitigated the Cd-induced growth inhibition in both roots and shoots and increased transpiration compared with non-SA-pretreatment under Cd exposure. It is concluded that Cd can be partly transferred from the Cd-exposed roots to Cd-unexposed roots and that cell death can be accelerated in the Cd-stressed roots in response to Cd stress. The SA-enhanced Cd tolerance in rice can be attributed to SA-elevated enzymatic and non-enzymatic antioxidants and NPT, and to SA-regulated Cd uptake, transport and distribution in plant organs.  相似文献

This study was carried out to investigate the effects of selenium (Se) on the uptake and translocation of cadmium (Cd) and essential elements in paddy rice (Oryza sativa L., Shuangyou 998). Selenium could alleviate/aggravate Cd toxicity in paddy rice, which depended on the dosages of Se and/or Cd. When Cd treatment level was as low as 35.6 μM, ≤12.7 μM Se could inhibit the uptake of Cd in paddy rice and increase the biomass of paddy rice; however, with Cd levels reaching 89–178 μM, the addition of Se resulted in increases in Cd uptake and exacerbated the growth of paddy rice. Cd always inhibited the uptake of Se. Cd alone suppressed the uptake of Ca, Mg, Mn, Cu, and Zn; however, Se reversed the decreases in the concentrations of the said elements, suggesting an element regulation mechanism to relieve Cd toxicity. Without Cd in the solution, low doses of Se increased the biomasses of shoots and roots at the expense of the more or less decreases in the concentrations of Ca, Mg, K, Fe, Mn, Cu, and shoot Zn, indicating an antagonistic effect of Se on these cations. The presence of Cd could also reverse these decreases especially at the highest treatment levels for both Se and Cd, also suggesting an element regulation mechanism responsible for the detoxification of high dosages of Se. Consequently, when Se is used to alleviate Cd toxicity, attention must be paid to the Cd pollution extent and doses of Se supplement.  相似文献

Aims

This study aimed to screen and identify low-cadmium (Cd) hot pepper (Capsicum annuum L.) cultivars and to clarify the mechanisms of low Cd accumulation in fruits.

Methods

A pot experiment was conducted to investigate the variations of fruit Cd concentration among 30 hot pepper cultivars and to determine the differences in uptake and translocation of Cd between low- and high-Cd cultivars in the control and two Cd treatments.

Results

There are significant differences among the cultivars in their ability to accumulate Cd in fruits. Fruit Cd concentrations are positively and significantly correlated with the translocation of Cd from roots to aboveground parts and the Cd concentrations of leaves and stems. However, no correlation was observed between the fruit’s Cd concentration and the root’s Cd uptake ability.

Conclusions

Two hot pepper cultivars, Yeshengchaotianjiao (No. 16) and Heilameixiaojianjiao (No. 23), were identified as low-Cd cultivars, and two, Jinfuzaohuangjiao (No. 13) and Shuduhong (No. 18), were treated as high-Cd cultivars. The difference in fruit Cd concentrations between low- and high-Cd cultivars is attributable to the difference in Cd translocation from roots to aboveground parts and from leaves and stems to fruits, rather than to the root’s Cd uptake ability.  相似文献

The objective of this study was to assess the effect of different Cd and Si concentrations on the maize plants. The following Cd and/or Si treatments were used: 5 Cd; 10 Cd; 100 Cd; 5 Cd + 0.08 Si; 10 Cd + 0.08 Si; 100 Cd + 5 Si treatments (Cd concentration in μM, Si concentration in mM). The plant growth, photosynthetic pigments content, antioxidant enzymes activities (POX, SOD, CAT), Cd and Si accumulation, translocation and cell wall deposition of the maize plants was observed. Changes in the endodermal cell walls development and late metaxylem elements lignification due to Cd and/or Si treatment were also evaluated. The negative effect of Cd (5 and 10 μM) on the growth parameters was alleviated by Si at 0.08 mM. The positive effect of Si was not observed at higher Cd and Si concentrations. This indicates that the alleviating effect of Si on Cd toxicity depends on the Cd and Si concentrations. Plants responded to Cd toxicity by an increase of antioxidant enzyme activity. Silicon addition in Cd + Si treatment stimulated an increase in the activity of antioxidant enzymes in comparison with the Cd treatment. Chlorophyll and carotenoid content in the Cd treated plants was not significantly affected by Si. The young maize plants retained much more Cd in their roots as they translocated into the shoots. 5 Cd + 0.08 Si and 10 Cd + 0.08 Si treatments correlated with an increase in Cd concentration in the roots and shoots, and in the cell walls. Silicon caused a slight decrease of the Cd translocation into the shoots in 5 Cd + 0.08 Si and 10 Cd + 0.08 Si treatments. Negative correlation between the root Cd cell wall deposition and Cd translocation was observed. Cadmium and/or Si altered root anatomy. Cadmium enhanced suberin lamellae development and late metaxylem lignification; silicon in Cd + Si treatments accelerated suberin lamellae deposition and enhanced the tertiary endodermal cell walls formation in comparison with Cd treatments. Negative correlation between the endodermal cell walls development and Cd translocation was observed.  相似文献

The Shuang'an site is the most serious selenosis site in Ziyang County, Shaanxi Province, which was the second selenosis site in China. In order to investigate the relationships between Se and other mineral elements and selenosis, bone coals, V-Mo ores, rocks, soils and plants were sampled from each site. The higher mean concentrations of Se, Mo, V and F in bone coals and ores may be the main environmental geochemical sources for soils and plants in this local ecosystem. Inappropriate revegetation in the Shuang'an mining site have posed the greatest risk of pollution, resulting in elevated mean concentrations of Se (16.9 μg/g), Mo (99 μg/g), V (1134 μg/g), F (1041 μg/g) and As (111 μg/g) in the soil directly derived from bone coal and V-Mo ore dumps. Most plants, which grow in the revegetated soil, contain elevated Se, Mo, V and F concentrations. The revegetated soil derived from bone coal and V-Mo ore dumps with excess Se, Mo, V and F concentrations in this ecosystem might have been essentially responsible for selenosis incidence of Shuang'an site in Ziyang County, Shaanxi Province. It is proposed that this selenosis area coincides with Mo, F and V toxicity based on their higher concentrations in rocks, soils and plants.  相似文献

The potential suitability of Eucalyptus camaldulensis for Cd phytoextraction was tested in a hydroponic study. Saplings were exposed to 4.5 and 89 μM Cd for one month, with and without EDTA and s,s-EDDS at 0.1, 1, and 5 mM. The saplings’ growth was not affected at the 4.5 μM Cd concentration, yet it decreased 3-fold at 89 μM, and almost all the Cd taken up was immobilized in the roots, reaching 360 and 5300 mg Cd kg?1, respectively (approximately 75% of which was non-washable in acid). The respective Cd root-to-shoot translocation factors were 0.14 and ≈5*10?4. At 0.1 mM concentration, EDTA and EDDS had no effect or even a positive effect on the saplings growth. This was reversed at 1 mM, and the chelants became lethal at the 5 mM concentration. At 89 μM Cd in the growth medium, 0.1 mM EDTA increased Cd translocation into the shoots by almost 10-fold, however it strongly reduced Cd content inside the roots. This hydroponic study indicates the feasibility of E. camaldulensis use for cleanup Cd-contaminated soils at environmental concentrations, both for site stabilization (phytostabilization) and gradual remediation (phytoextraction). EDTA was shown to be much more efficient in enhancing Cd translocation than s,s-EDDS.  相似文献

Present study was undertaken to study the effect of 28-days exposure of female adult rats to cadmium (Cd) in drinking water @ 3, 10 and 30 parts per million (ppm) on myometrial responsiveness to different spasmogens and unravel the possible mechanism of alterations in myometrial activity. Cadmium and Ca2+ levels in blood and uterus were measured by atomic absorption spectroscopy while isometric tension in myometrial strips was measured using data acquisition system-based physiograph. Dose-dependent increase in levels of cadmium was observed in both blood and uterus while calcium was increased only in the uterus as compared to those in control. Significant increase in absolute tension and mean integral tension along with non-significant increase in frequency of myometrial contraction was observed in rats of Cd-treated groups. As compared to the control, cadmium decreased and increased the effects of calcium chloride, 80 mM KCl, histamine (0.1 μM) and oxytocin (10?2 IU/ml) in lower-dose (3 ppm) and higher-dose groups (10 and 30 ppm), respectively. Cadmium potentiated and inhibited the relaxant response to phenylephrine in myometrium of rats at lower-dose (3 ppm) and highest-dose (30 ppm) Cd-treated groups, respectively. Results of our study revealed that Cd accumulates in the myometrium of rats and alters its responsiveness to oxytocin, histamine, 80 mM KCl, calcium chloride and phenylephrine, and these effects are differentially mediated depending on levels of exposure possibly through voltage-dependent calcium channel (VDCC) and Ca2+-mimicking pathways.  相似文献

Using a short-term Cd treatment (5–30 min), we analysed the effect of Cd on apoplastic ascorbate redox status and their regeneration during the recovery period in barley root tips. Root growth inhibition induced by 15 μM Cd was detectable after 5 min of exposure and increased in a time-dependent manner up to 15 min of exposure. High 30 μM Cd concentration completely inhibited root growth during the first 6 h after short-term treatment. In parallel with Cd-induced root growth inhibition, a rapid decrease of apoplastic ascorbate dehydroascorbate ratio was observed immediately after short-term treatments. During the recovery from 15 μM Cd short-term treatment, apoplastic ascorbate was rapidly regenerated to the control level in the first root segment containing meristem and elongation zone. In contrast to 15 μM Cd treatment, in 30 μM Cd-treated roots apoplastic ascorbate level was sustained at a significantly lower level compared to control roots. We confirmed that a decrease of apoplastic ascorbate/dehydroascorbate ratio in the elongation zone was associated with root growth inhibition or arrest.  相似文献

The objective of this study was to investigate Cd phytoremediation ability of Indian mustard, Brassica juncea. The study was conducted with 25, 50, 100, 200 and 400 mg Kg?1 CdCl2 in laboratory for 21 days and Cd concentrations in the root, shoot and leaf tissues were estimated by atomic absorption spectroscopy. The plant showed high Cd tolerance of up to 400 mg Kg?1 but there was a general trend of decline in the root and shoot length, tissue biomass, leaf chlorophyll and carotenoid contents. The tolerance index (TI) of plants were calculated taking both root and shoot lengths as variables. The maximum tolerance (TI shoot = 87.4 % and TI root = 89.6 %) to Cd toxicity was observed at 25 mg Kg?1, which progressively decreased with increase in dose. The highest shoot (10791 μg g?1 dry wt) and root (9602 μg g?1 dry wt) Cd accumulation was achieved at 200 mg kg?1 Cd treatment and the maximum leaf Cd accumulation was 10071.6 μg g?1 dry wt achieved at 100 mg Kg?1 Cd, after 21 days of treatment. The enrichment coefficient and root to shoot translocation factor were calculated, which, pointed towards the suitability of Indian mustard for removing Cd from soil.  相似文献
