贫营养湖泊花神湖和紫霞湖浮游细菌群落结构分析   总被引:1,自引:0,他引:1  
以南京市花神湖和紫霞湖两个贫营养型湖泊为研究对象,通过构建花神湖和紫霞湖16S rRNA基因克隆文库探讨了浮游细菌群落结构组成的变化。结果表明,花神湖和紫霞湖两湖泊水体中浮游细菌群落结构相似,主要隶属于放线菌门(Actinobacteria)、蓝藻门(Cyanobacteria)、α-变形菌门(Alphaproteobacteria)、β-变形菌门(Betaproteobacteria)、杆菌门(Bacteroidetes)、浮霉菌门(Planctomycetes)、疣微菌门(Verrucomicrobia)和芽单胞菌门(Gemmatimonadetes),其中放线菌门(Actinobacteria)、蓝藻门(Cyanobacteria)、β-变形菌门(Betaproteobacteria)是优势细菌类群。两个湖泊水体中75%的细菌与GenBank中已有的未培养细菌同源性高于97%,同时在两个克隆文库中还发现了6个淡水细菌新类群。通过对低纬度区域贫营养型湖泊浮游细菌群落结构的分析,加深了我们对浮游细菌多样性的了解,表明湖泊浮游细菌多样性有待进一步认识。  相似文献

Current trophic status and trend of Chinese freshwater lakes were investigated in this study. The results showed that all lakes studied were commonly undergoing the eutrophica-tion process, water quality decreased and lake's ecosystem is being declined. Most of the urban lakes are facing serious eutrophication. Many medium-sized lakes are in metrophic or eutrophic status, some local water are even approaching the hypertrophic level. The famous five freshwater lakes in China have entered into eutrophication in the condition of higher nutrient load. Lake Taihu, Hongze and Caohu are already in eutrophic state. Eutrophic lakes are mainly distributed in the middle and lower reaches of Yangtze River and Yungui plateau. Lake eutrophication developed rapidly. Among the 34 lakes studied in 1970's, most of lakes were in the mesotrophic status, mesotrophic water area accounted for 91.8%. With the nine year of 1978-1987 the area percentage of oligotrophic lakes decreased from 3.2% to 0.53%, and that of eutrophic lakes increased from 5.0% to 55.01%. Recent data showed 57.5% lakes were in eutrophic and hyper trophic status of the 40 surveyed lakes. Eutrophic trend of Lake Taihu, Chaohu and Xuanwu in the region of the middle and lower reaches of Yangtze River was predicated using the ecological stress model. The results showed that in 2008 Lake Taihu, Chaohu and Xuanwu might be of eutrophication, eutrophication and hypertrophication, respectively if no control measurement is taken. Provided the pollution water treatment rate is 60% in 2030, approximately 30 billion ton pollution water would still be discharged directly in the lakes. Therefore, in 2030 the urban lakes in China might be eutrophication or hypertrophication, and most of the medium-sized lakes at the urban-rural fringe might be in eutrophication or hypertrophication. The famous five biggest freshwater lakes in China might be eutrophication if control countermeasures are taken as now. Lake eutrophication has become a serious environmental problem in China. Based on the domestic and foreign experiences of the eutrophic control technologies, both nutrient pollution control and lake ecological restoration should be carried out and this may be the guidance for the eutrophic control of lakes in China.  相似文献

Lake Neuchâtel (Switzerland), oligotrophic until 1950, was meso-eutrophic in 1980. The relative abundance in worm communities of Peloscolex velutinus and Stylodrilus heringianus was used to monitor the trophic state of the lake. In 1980, the median relative abundance of these oligotrophic species was 9% in the whole of Lake Neuchâtel compared with 70% in oligotrophic lakes, 35% in mesotrophic lakes, and 0% in eutrophic lakes. The scarcity of oligotrophic species in the deepest area (153 m) characterized better the meso-eutrophic state of Lake Neuchâtel than oxygen concentrations which never descended below 6 mg·1-1. Location of the area within the lake from where worms were sampled was of critical importance to assess the trophic state: some areas reflected the past rather than the present state of the lake.  相似文献

Bacteria are crucial components in lake sediments and play important role in various environmental processes. Urban lakes in the densely populated cities are often small, shallow, highly artificial and hypereutrophic compared to rural and natural lakes and have been overlooked for a long time. In the present study, bacterial community compositions in surface sediments of three urban lakes (Lake Mochou, Lake Qianhu and Lake Zixia) in Nanjing City, China, were investigated using the terminal restriction fragment length polymorphism (T-RFLP) of PCR-amplified 16S rRNA gene and clone libraries. Remarkable differences in the T-RFLP patterns were observed in different lakes or different sampling stations of the same lake. Canonical correspondence analysis indicated that total nitrogen (TN) had significant effects on bacterial community structure in the lake sediments. Chloroflexi were the most dominant bacterial group in the clone library from Lake Mochou (21.7?% of the total clones) which was partly associated with its higher TN and organic matters concentrations. However, Bacteroidetes appeared to be dominated colonizers in the sediments of Lake Zixia (20.4?% of the total clones). Our study gives a comprehensive insight into the structure of bacterial community of urban lake sediments, indicating that the environmental factors played a key role in influencing the bacterial community composition in the freshwater ecosystems.  相似文献

In this study,PCR-denaturing gradient gel electrophoresis (DGGE) was applied to analyze the microbial communities in lake sediments from Lake Xuanwu,Lake Mochou in Nanjing and Lake Taihu in Wuxi.Sediment samples from seven locations in three lakes were collected and their genomic DNAs were extracted.The DNA yields of the sediments of Lake Xuanwu and Lake Mochou were high (10 μg/g),while that of sediments in Lake Taihu was relatively low.After DNA purification,the 16S rDNA genes (V3 to V5 region) were amplified and the amplified DNA fragments were separated by parallel DGGE.The DGGE profiles showed that there were five common bands in all the lake sediment samples indicating that there were similarities among the populations of microorganisms in all the lake sediments.The DGGE profiles of Lake Xuanwu and Lake Mochou were similar and about 20 types of micro-organisms were identified in the sediment samples of both lakes.These results suggest that the sediment samples of these two city lakes (Xuanwu,Mochou) have similar microbial communities.However,the DGGE profiles of sediment samples in Lake Taihu were significantly differ-ent from these two lakes.Furthermore,the DGGE pro-files of sediment samples in different locations in Lake Taihu were also different,suggesting that the microbial communities in Lake Taihu are more diversified than those in Lake Xuanwu and Lake Mochou.The differences in microbial diversity may be caused by the different environmental conditions,such as redox potential,pH,and the concentrations of organic matters.Seven major bands of 16S rDNA genes fragments from the DGGE profiles of sediment samples were further re-amplified and sequenced.The results of sequencing analysis indicate that five sequences shared 99%-100% homology with known sequences (Bacillus and Brevibacillus,uncultured bacteria),while the other two sequences shared 93%-96% homology with known sequences (Acinetobacter,and Bacillus).The study shows that the PCR-DGGE tech-nique combined with sequence analysis is a feasible and efficient method for the determination of microbial com-munities in sediment samples.  相似文献

In 1978–80, oligochaete communities of meso-eutrophic Lake Léman (Lake of Geneva) were compared to those of mesotrophic Lake Neuchâtel. Worm species were classified into three groups corresponding to their increasing tolerance to eutrophication: (1) oligotrophic species, mostly Peloscolex velutinus, Stylodrilus heringianus; (2) mesotrophic species, mostly Potamothrix vejdovskyi, P. bedoti; (3) eutrophic species, mostly Potamothrix hammoniensis, P. heuscheri, Tubifex tubifex. In both lakes, eutrophic species constituted the bulk of the communities in terms of absolute abundance. However, relative abundance of mesotrophic and eutrophic species was higher in Lake Léman; oligotrophic species were more important in Lake Neuchâtel. These data confirmed the trophic classification of lakes based on chemical parameters. The number of zero values, which perturbated statistical analysis, was reduced by using species groupings instead of isolated species. Thus, making the lakes more comparable even if different species were present in each one. Relative density values based on all samples were distributed among 4 density classes for the 3 species groupings. The 12 resulting frequencies described the community structure expressed in terms of eutrophication. Furthermore, these frequencies may be used for comparison of eutrophication levels in several lakes.  相似文献

Small eukaryotes, cells with a diameter of less than 5 mum, are fundamental components of lacustrine planktonic systems. In this study, small-eukaryote diversity was determined by sequencing cloned 18S rRNA genes in three libraries from lakes of differing trophic status in the Massif Central, France: the oligotrophic Lake Godivelle, the oligomesotrophic Lake Pavin, and the eutrophic Lake Aydat. This analysis shows that the least diversified library was in the eutrophic lake (12 operational taxonomic units [OTUs]) and the most diversified was in the oligomesotrophic lake (26 OTUs). Certain groups were present in at least two ecosystems, while the others were specific to one lake on the sampling date. Cryptophyta, Chrysophyceae, and the strictly heterotrophic eukaryotes, Ciliophora and fungi, were identified in the three libraries. Among the small eukaryotes found only in two lakes, Choanoflagellida and environmental sequences (LKM11) were not detected in the eutrophic system whereas Cercozoa were confined to the oligomesotrophic and eutrophic lakes. Three OTUs, linked to the Perkinsozoa, were detected only in the Aydat library, where they represented 60% of the clones of the library. Chlorophyta and Haptophyta lineages were represented by a single clone and were present only in Godivelle and Pavin, respectively. Of the 127 clones studied, classical pigmented organisms (autotrophs and mixotrophs) represented only a low proportion regardless of the library's origin. This study shows that the small-eukaryote community composition may differ as a function of trophic status; certain lineages could be detected only in a single ecosystem.  相似文献

Phyloplankton structure and its relation to physical and chemical properties of the water was studied in 58 central Finnish lakes. The biomass ranged from 0.2 to 14.2 g m−3 and the number of taxa per sample ranged from 33 to 152. The lakes were grouped into 5 types according to their trophic state: eutrophic, dyseutrophic, mesotrophic, oligotrophic, and acid oligotrophic lakes. The average biomass in eutrophic lakes was 5.57 g m−3, in dyseutrophic 3.54 g m−3, 1.23 g m−3 in mesotrophic, 0.52 g m−3 in oligotrophic and 0.39 g −3 in acid oligotrophic lakes. The average number of taxa per sample in the corresponding lake types were 109. 1, 79.3, 97.9, 90.9 and 43.8, respectively. The phytoplankton communities in eutrophic lakes were characterized by blue-green algae (21.2% of total biomass) and green algae (18.7% of total biomass). In dyseutrophic lakes the proportion of green algae was much smaller (7.2% of total biomass) than in eutrophic lakes, whereas the proportion of diatoms and cryptophytes was higher (28.2 and 20.4% of total biomass, respectively). Chrysophytes dominated in the oligotrophic and mesotrophic lakes (27.3–39.9% of total biomass). The contribution of dinoflagellates to the total biomass was highest in the most oligotrophic acidified lakes and in those lakes the relative proportions of blue-green and green algae were much higher than in the typical oligotrophic lakes. The lakes were also grouped into 8 community types according to the dominating algal group. Cyanophyceae- and Chlorophyceae-types characterized the eutrophic lakes, whereas Chrysophyceae-Dinopheceae-type was typical for most oligotrophic lakes. The other 5 types occurred in mesotrophic and oligotrophic lakes but the physical and chemical properties of these lakes did not differ much.  相似文献

Relative to their scarcity, large, deep lakes support a large proportion of the world’s freshwater species. This biodiversity is threatened by human development and is in need of conservation. Direct comparison of biodiversity is the basis of biological monitoring for conservation but is difficult to conduct between large, insular ecosystems. The objective of our study was to conduct such a comparison of benthic biodiversity between three of the world’s largest lakes: Lake Tahoe, USA; Lake Hövsgöl, Mongolia; and Crater Lake, USA. We examined biodiversity of common benthic organism, the non-biting midges (Chironomidae) and determined lake trophic status using chironomid-based lake typology, tested whether community structure was similar between the three lakes despite geographic distance; and tested whether chironomid diversity would show significant variation within and between lakes. Typology analysis indicated that Lake Hövsgöl was ultra-oligotrophic, Crater Lake was oligotrophic, and Lake Tahoe was borderline oligotrophic/mesotrophic. These results were similar to traditional pelagic measures of lake trophic status for Lake Hövsgöl and Crater Lake but differed for Lake Tahoe, which has been designated as ultra-oligotrophic by traditional pelagic measures such as transparency found in the literature. Analysis of similarity showed that Lake Tahoe and Lake Hövsgöl chironomid communities were more similar to each other than either was to Crater Lake communities. Diversity varied between the three lakes and spatially within each lake. This research shows that chironomid communities from these large lakes were sensitive to trophic conditions. Chironomid communities were similar between the deep environments of Lake Hövsgöl and Lake Tahoe, indicating that chironomid communities from these lakes may be useful in comparing trophic state changes in large lakes. Spatial variation in Lake Tahoe’s diversity is indicative of differential response of chironomid communities to nutrient enrichment which may be an indication of changes in trophic state within and across habitats.  相似文献

It is shown that meiobenthos plays an important role in the secondary production by zoobenthos in lakes, as well as in the degradation of organic matter. In large lakes (Lake Ladoga, Lake Onega, Lake Päijänne, Lake Constance), the ratio of meiobenthic production to the production of macrobenthos is on average 50–61%. In the small Latgalian lakes (Latvia), this proportion is different: in the profundal of these lakes it varies from 92.5% in a naturally clean mesotrophic lake to 0.0004% in the most eutrophic lake, and in the littoral of lakes – from 578–1476% in mesotrophic lakes to 148–306% in eutrophic ones. The level of production of littoral meiobenthos does not depend on the trophic status of the lake, and can be equally high both in undisturbed mesotrophic lakes and in strongly eutrophicated lakes. The intensity of production of the littoral meiobenthos in oligotrophic and mesotrophic lakes, on the one hand, and in eutrophic lakes on the other, are not reliably distinguished. There is a clear tendency for a decrease of the role of profundal meiobenthos with regard to the transformation of energy flows in lake ecosystems, both with an increase in eutrophication and with an increase in the amount of organic matter in the benthal available from phytoplankton.  相似文献

Bacterioplankton plays a central role in the microbial functioning of lacustrine ecosystems; however, factors that constrain its structural variation are still poorly understood. Here we evaluated the driving forces exerted by a large set of environmental and biological parameters on the temporal and spatial dynamics of free-living bacterial community structures (BCS) in two neighboring perialpine lakes, Lake Bourget and Lake Annecy, which differ in trophic status. We analyzed monthly data from a 1-year sampling period at two depths situated in the epi- and hypolimnia for each lake. Overall, denaturing gradient gel electrophoresis (DGGE) revealed significant differences in the BCS in the two lakes, characterized by a higher number of bands in the oligotrophic ecosystem (i.e., Lake Annecy). The temporal dynamics of BCS differed greatly between depths and lakes, with temporal scale patterns being much longer in the mesotrophic Lake Bourget. Direct-gradient multivariate ordination analyses showed that a complex array of biogeochemical parameters was the driving force behind BCS shifts in both lakes. Our results indicated that 60 to 80% of the variance was explained only by the bottom-up factors in both lakes, indicating the importance of nutrients and organic matter from autotrophic origin in controlling the BCS. Top-down regulation by flagellates together with ciliates or viruses was found only in the hypolimnion and not in the epilimnion for both lakes and explained less than 18% of the bacterial community changes during the year. Our study suggests that the temporal dynamics of the free-living bacterial community structure in deep perialpine lakes are dependent mainly on bottom-up factors and to a lesser extent on top-down factors, whatever the specific environmental conditions of these lakes.  相似文献

The biomass and production of the most important zooplankton species were followed for two years in three lakes of varying trophic status in the Lake Washington watershed. Cladocerans and copepods were of equal importance in the biomass of lakes Findley and Chester Morse (both oligotrophic), whereas, copepods were the main biomass component in Lake Sammamish (mesotrophic). Cladocerans dominated production in lakes Sammamish and Chester Morse, while in Findley Lake their productive role, like that of biomass, was equal to that of the copepods. Rotifers contributed a relatively small biomass and production.Data from this study supported Hillbricht-Ilkowska's postulate that the energy transfer efficiency between the primary and secondary trophic levels decreases with increasing trophic state. Energy transfer efficiencies for the lakes of this study expressed as a two year mean of the ratio-secondary: primary production, were as follows: Findley Lake-0.13; Chester Morse Lake-0.08; and Lake Sammanish-0.04.On the other hand, the hypothesis of Patalas that the secondary productivity: biomass ratio (P/B) tended to increase in proportion to the productivity of a lake, could not be supported. Lake Sammamish, the most productive of the lakes studied, had a P/B ratio of 0.03 while lakes Findley and Chester Morse had P/B ratios of 0.04.  相似文献

The aim of this study was to examine whether littoral nematode community patterns are shaped by lake trophic state. It was hypothesized that trophic level is associated negatively with the proportion of omnivores and positively with the percentages of bacterial feeders, but not at all with the diversity, abundance, and biomass of freshwater nematodes. Sediment samples were taken at littoral sites of eight southern Swedish lakes of different trophy in spring and autumn 2007. Trophic level was found to strongly influence species richness, as oligotrophic and mesotrophic lakes supported the greatest species numbers, whereas nematode abundance, biomass, and Shannon index were unaffected. Furthermore, our results indicated effects on the nematode community’s trophic structure, with a larger proportion of predatory nematodes in oligotrophic and mesotrophic lakes but no differences in the other feeding types (bacteria, algae and suction feeders, omnivorous species). Multivariate analysis indicated a shift in species compositions along the threshold from mesotrophic to eutrophic conditions, with the presence of Tobrilus gracilis, Monhystera paludicola, Brevitobrilus stefanskii, and Ethmolaimus pratensis related to the latter. Nematode communities in oligotrophic and mesotrophic lakes were characterized by a similar species composition, with pronounced occurrences of Eumonhystera longicaudatula, Semitobrilus cf. pellucidus, Prodesmodora circulata, and Rhabdolaimus terrestris. Vertical distribution of bacterial community structure was investigated in the sediments of two eutrophic lakes of China, Lake Taihu and Lake Xuanwu. Profiles of bacterial communities were generated using a molecular fingerprinting technique, denaturing gradient gel electrophoresis (DGGE) followed by DNA sequence analysis, and the results were interpreted with multivariate statistical analysis. To assess changes in the genetic diversity of bacterial communities with changing depth, DGGE banding patterns were analysed by cluster analysis. Distinct clusters were recognized in different sampling stations of Lake Taihu. Canonical correspondence analysis (CCA) was carried out to infer the relationship between environmental variables and bacterial community structure. DGGE samples collected at the same sampling site clustered together in both lakes. Total phosphorus, organic matter and pH were considered to be the key factors driving the changes in bacterial community composition.  相似文献

The diversity of small eukaryotes (0.2 to 5 mum) in a mesotrophic lake (Lake Bourget) was investigated using 18S rRNA gene library construction and fluorescent in situ hybridization coupled with tyramide signal amplification (TSA-FISH). Samples collected from the epilimnion on two dates were used to extend a data set previously obtained using similar approaches for lakes with a range of trophic types. A high level of diversity was recorded for this system with intermediate trophic status, and the main sequences from Lake Bourget were affiliated with ciliates (maximum, 19% of the operational taxonomic units [OTUs]), cryptophytes (33%), stramenopiles (13.2%), and cercozoa (9%). Although the comparison of TSA-FISH results and clone libraries suggested that the level of Chlorophyceae may have been underestimated using PCR with 18S rRNA primers, heterotrophic organisms dominated the small-eukaryote assemblage. We found that a large fraction of the sequences belonged to potential parasites of freshwater phytoplankton, including sequences affiliated with fungi and Perkinsozoa. On average, these sequences represented 30% of the OTUs (40% of the clones) obtained for each of two dates for Lake Bourget. Our results provide information on lacustrine small-eukaryote diversity and structure, adding to the phylogenetic data available for lakes with various trophic types.  相似文献

Very small eukaryotic organisms (picoeukaryotes) are fundamental components of marine planktonic systems, often accounting for a significant fraction of the biomass and activity in a system. Their identity, however, has remained elusive, since the small cells lack morphological features for identification. We determined the diversity of marine picoeukaryotes by sequencing cloned 18S rRNA genes in five genetic libraries from North Atlantic, Southern Ocean, and Mediterranean Sea surface waters. Picoplankton were obtained by filter size fractionation, a step that excluded most large eukaryotes and recovered most picoeukaryotes. Genetic libraries of eukaryotic ribosomal DNA were screened by restriction fragment length polymorphism analysis, and at least one clone of each operational taxonomic unit (OTU) was partially sequenced. In general, the phylogenetic diversity in each library was rather great, and each library included many different OTUs and members of very distantly related phylogenetic groups. Of 225 eukaryotic clones, 126 were affiliated with algal classes, especially the Prasinophyceae, the Prymnesiophyceae, the Bacillariophyceae, and the Dinophyceae. A minor fraction (27 clones) was affiliated with clearly heterotrophic organisms, such as ciliates, the chrysomonad Paraphysomonas, cercomonads, and fungi. There were two relatively abundant novel lineages, novel stramenopiles (53 clones) and novel alveolates (19 clones). These lineages are very different from any organism that has been isolated, suggesting that there are previously unknown picoeukaryotes. Prasinophytes and novel stramenopile clones were very abundant in all of the libraries analyzed. These findings underscore the importance of attempts to grow the small eukaryotic plankton in pure culture.  相似文献

The influence of the size distribution of phytoplankton on changes in the planktonic food web structures with eutrophication was examined using natural planktonic communities in two world-famous lakes: Lake Baikal and Lake Biwa. The size distribution of phytoplankton and the ratio of heterotrophic to autotrophic biomass (H/A ratio), indicating the balance between primary production and its consumption, were investigated in the lakes of different trophic status. The results revealed that microphytoplankton (>20μm) in mesotrophic Lake Biwa, and picophytoplankton (<2μm) or nanophytoplankton (2–20μm) in oligotrophic Lake Baikal, comprised the highest proportion of the total phytoplankton biomass. The H/A ratio was lower in Lake Biwa (<1) than in Lake Baikal (>1). The low H/A ratio in Lake Biwa appeared to be the consequence of the lack of consumption of the more abundant microphytoplankton, which were inferior competitors in nutrient uptake under oligotrophic conditions but less vulnerable to grazing. As a result, unconsumed microphytoplankton accumulated in the water column, decreasing the H/A ratio in Lake Biwa. Our results showed that food web structure and energy flow in planktonic communities were greatly influenced by the size distribution of phytoplankton, in conjunction with bottom-up (nutrient uptake) and top-down (grazing) effects at the trophic level of primary producers.  相似文献
