鼠李糖脂是最常见,研究最深入,应用最广泛的一类生物表面活性剂。从油田附近、沼气池旁的土壤中分离得到了25株菌,通过硫酸-苯酚反应,乳化实验,排油性实验,薄层色谱实验筛选产鼠李糖脂的菌株并表征产生的鼠李糖脂,通过16S r DNA序列确定细菌的种属。硫酸-苯酚反应显示有2株菌可能产鼠李糖脂;5株菌的发酵液具有明显的乳化效果,命名为"其红"这株菌的乳化指数可达58.97%;1株菌的发酵液上清稀释10倍后排油圈直径仍可达3.53 cm;薄层色谱实验也显示"其红"菌产鼠李糖脂。在四个实验中"其红"菌都检测到了鼠李糖脂,故确认"其红"菌可以产鼠李糖脂。16S r DNA序列分析表明"其红"菌属于希瓦氏菌属(Shewanella)并与腐败希瓦菌(S.putrefaciens LMG 26268(T))相似度最高。  相似文献

为了适应精酿啤酒对个性化风味的需求,能产生特定风味化合物的产香酵母成为研究者的研究重点。从精酿啤酒原液中分离到1株产香酵母LX15菌,该菌细胞呈圆形或卵圆形、多极芽殖生长;LX15菌在玉米粉培养基上培养7~10 d不形成假菌丝,在酵母膏蛋白胨培养基上培养3 d能够形成子囊孢子。经生理生化特征和系统发育分析,确认该生香酵母为Pichia myanmarensis菌中的一个菌株,所产主要风味化合物包括乙酸乙酯、乙酸异戊酯、己酸乙酯和辛酸乙酯。当LX15菌与啤酒酵母C1菌共发酵时,能够产生协同效应,提高酯类化合物和高级醇类的含量,并与LX15菌的接种比例正相关,但并不影响啤酒酿造的整体发酵速率和发酵能力。因此,LX15菌是一株适于提高精酿啤酒风味的产香酵母菌。  相似文献

【目的】槐糖脂是一类生物表面活性剂,不仅具有常规表面活性剂所具有的增溶、乳化、润湿、发泡、分散、降低表面张力等通用性能,且对环境的耐受性极强。熊蜂生假丝酵母(Starmerella bombicola)能够发酵生产槐糖脂,但槐糖脂具有酸型、内酯型和乙酰化型等不同类型,结构多样,难以分离。本文拟通过代谢工程改造,构建高产酸型槐糖脂的熊蜂生假丝酵母工程菌株。【方法】利用潮霉素抗性基因构建了标记基因重复利用系统Rec-six基因编辑系统,在此基础上将合成内酯型槐糖脂的关键基因——内酯酶基因SBLE敲除获得一株只产酸型槐糖脂的工程菌株Δsble,进一步同源过量表达葡萄糖基转移酶基因UGTB并敲除过氧化物酶体膜转运蛋白编码基因PXA1,构建了高产酸型槐糖脂的酵母工程菌。【结果】与出发菌株相比,重组熊蜂生假丝酵母发酵油酸能够合成单一的酸型槐糖脂,而不再合成内酯型槐糖脂,同时酸型槐糖脂的产量由20 g/L提高到44 g/L,提高了2.1倍。【结论】通过敲除PXA1、SBLE和过表达UGTB来改造熊蜂生假丝酵母,能够有效提高重组菌的酸型槐糖脂产量,为发酵法生产酸型槐糖脂奠定了基础。  相似文献

目的 探讨FUS-100全自动尿沉渣分析仪(简称FUS-100)中的细菌和酵母定量计数在筛查住院患者尿路感染时的价值.方法 用定量细菌培养法和FUS-100检测505例疑似泌尿系统感染患者清洁中段尿标本.结果 505份尿标本培养阳性192份,阳性率为38.0%,其中36份标本有两种细菌生长,共分离出228株菌,革兰阳性球菌30株(13.2%),革兰阴性杆菌108株(47.4%),真菌90株(39.4%).以尿细菌计数≥23.72/μL、酵母计数≥0.65/μL为感染阳性标准,与中段尿培养相比较,FUS-100细菌定量计数的敏感性为85.5%,特异性为89.5%,阳性似然比为8.14,阴性似然比为0.16;酵母定量计数的敏感性为80.4%,特异性为97.2%,阳性似然比为28.71,阴性似然比为0.20.结论 FUS-100定量计数尿细菌和酵母可用于住院患者尿路感染的快速筛查.  相似文献

从能利用正十二烷产生1,12-十二碳二元酸的热带假丝酵母突变株D28出发,经两次紫外线照射诱变,选育到一株从正十三烷产生1,13-十三碳二元酸较高的突变株2—23号菌。该突变株较出发菌株提高产酸率20%,达40.4g/L。突变株2—23也能将一定链长的长链烷烃以较高的产率转变成相应的单一二元酸。此外,在产酸摇瓶条件试验中观察到烷烃的诱导作用,使突变株产酸能力得以提高。用烷烃预培养的种子发酵正十三烷,其产生1,13一十三碳二元酸的量较糖质碳源培养的种子发酵时要提高30%。  相似文献

为了探究新疆本土蟠桃园可培养酵母菌多样性,并挖掘功能酵母资源,本研究以新疆石河子蟠桃园3年、8年、15年树龄的根际和非根际土壤以及桃树叶片为材料,经过传统的分离培养方法获得可培养的酵母菌菌株,并进行形态学、生理生化以及26S r DNA的D1/D2区序列分析,共获得可培养酵母菌129株,从属于12个属17个种,其中子囊菌酵母为优势菌群,占分离属的88%,分布于威克汉姆酵母属(Wickerhamomyces),Vanrija属,Barnettozyma属和有孢圆酵母属(Torulaspora)等11个属的15个种。担子菌占分离属的12%,分布于隐球酵母属(Cryptococcus)的2个种。其中优势属威克汉姆酵母属,包括异常威克汉姆酵母(W. anomalus)和W. pijperi两个种,占总比例的33%,优势种异常威克汉姆酵母所占总株数比例为17%。从可培养酵母中共筛选出23株功能酵母,其中富硒酵母21株,优势种为白地霉(Galactomyces candidum),产蛋白酶酵母2株均属于隐球酵母属的Cryptococcus albidus。结果表明,新疆桃园中蕴含丰富的酵母菌资源,非根际土壤中的酵母多样性大于根际及叶片酵母多样性,且分离得到富硒酵母及产蛋白酶酵母。本研究挖掘了新疆本土可培养酵母菌资源,同时也为功能酵母的开发和利用提供理论指导。  相似文献

【目的】从油页岩环境中筛选可降解油页岩干酪根的产生物表面活性剂菌株。【方法】从抚顺油页岩矿废水样品中用血平板法初筛,排油圈法、乳化法和表面张力法复筛,获得产生物表面活性剂菌株。对目标菌株进行生理生化鉴定、16S r RNA基因序列和系统发育分析,用薄层色谱鉴定其发酵液表面活性成分,优化产表面活性剂的培养条件,初步考察其对油页岩干酪根的降解能力。【结果】筛选到一株产糖脂表面活性剂菌株B-1,初步鉴定为Pseudomonas sp.,该菌株有良好的排油和乳化能力以及较低的表面张力,可利用烷烃、不饱和脂肪酸和糖类作为碳源。在30-34°C范围内添加0.3%Na Cl的葡萄糖培养基(p H 7.0)中该菌生长旺盛,发酵液表面张力最低为27 m N/m。菌株B-1在添加一定量葡萄糖的无机盐培养基中作用30 d后对干酪根的降解率为2.85%,高于不添加葡萄糖无机盐培养基对照组的降解率(1.04%)。【结论】菌株B-1是一株性能良好的产糖脂表面活性剂细菌,有降解干酪根的潜力。  相似文献

【背景】土壤中铁主要以难溶态铁氧化物的形式存在,有效性较低,产铁载体细菌对铁氧化物的活化是提高铁利用效率的有效途径。【目的】从林木土壤筛选产铁载体细菌,并观察菌株对难溶性铁氧化物的利用效应,可为土壤微生物资源开发及其在养分调控中的作用提供理论依据。【方法】通过CAS检测法从林木根系附近表层土壤中分离产铁载体细菌,借助生物培养实验分析温度和pH对微生物生长和铁载体产生的影响,通过振荡平衡实验,探究细菌产铁载体对铁氧化物的活化效应。【结果】通过CAS检测法从林木根系附近表层土壤中分离得到12株产铁载体细菌,16S rRNA基因扩增子测序初步鉴定结果显示筛选细菌均为假单胞菌属。选取铁载体产生能力和生长活性较高的两株细菌ARSB02和CNRSB01作为重点研究对象,结果显示,不同条件下CNRSB01的生物量和铁载体产量均高于菌株ARSB02,22 h时菌株ARSB02和CNRSB01的铁载体活性单位分别达到67.07%和84.60%。pH 5.0-8.0的范围内两株细菌可以保持较好的铁载体产生能力,菌株ARSB02和CNRSB01在pH7.0时铁载体产生能力最强,铁载体活性单位分别达到38.9...  相似文献

A chloroform crude extract (aerial part) and two compounds, apigenin (1) and cynaropicrin (2), isolated from Moquinia kingii were evaluated against Trypanosoma cruzi trypomastigotes in vitro. Antimicrobial activity was also screened using twenty-two strains including gram-positive and gram-negative bacteria and the yeasts Candida albicans and C. tropicalis. The chloroform crude extract, fractions and isolated compounds from M. kingii were active for both activities. The IC50 values for trypanocidal activity obtained for cynaropicrin and apigenin were 93.5 microg/ml and 181 microg/ml, respectively, while the minimum inhibitory concentrations (MICs) varied from 100 microg/ml to 2500 microg/ml, against the strains of bacteria and yeasts evaluated.  相似文献

Many bacteria are known to inhibit food pathogens, such as Listeria monocytogenes, by secreting a variety of bactericidal and bacteriostatic substances. In sharp contrast, it is unknown whether yeast has an inhibitory potential for the growth of pathogenic bacteria in food. A total of 404 yeasts were screened for inhibitory activity against five Listeria monocytogenes strains. Three hundred and four of these yeasts were isolated from smear-ripened cheeses. Most of the yeasts were identified by Fourier transform infrared spectroscopy. Using an agar-membrane screening assay, a fraction of approximately 4% of the 304 red smear cheese isolates clearly inhibited growth of L. monocytogenes. Furthermore, 14 out of these 304 cheese yeasts were cocultivated with L. monocytogenes WSLC 1364 on solid medium to test the antilisterial activity of yeast in direct cell contact with Listeria. All yeasts inhibited L. monocytogenes to a low degree, which is most probably due to competition for nutrients. However, one Candida intermedia strain was able to reduce the listerial cell count by 4 log units. Another four yeasts, assigned to C. intermedia (three strains) and Kluyveromyces marxianus (one strain), repressed growth of L. monocytogenes by 3 log units. Inhibition of L. monocytogenes was clearly pronounced in the cocultivation assay, which simulates the conditions and contamination rates present on smear cheese surfaces. Extracellular enzyme‐producing yeasts might be involved in the supplementation of enzymes within the gastrointestinal tract of fish. The present study was intended to detect yeasts in the intestine of three Indian major carps (Labeo rohita, Catla catla, Cirrhinus mrigala), three exotic carps (Hypophthalmichthys molitrix, Ctenopharyngodon idella, Cyprinus carpio), as well as Nile tilapia (Oreochromis niloticus), and to identify the most promising extracellular enzyme‐producing (e.g. amylase, protease, lipase, cellulase, xylanase and phytase) yeast strains by 18S rDNA sequence analysis. Selected for qualitative enzyme assay were 121 yeast strains, from which 28 were further studied for quantitative enzyme assay. The strain CMH6A isolated from C. mrigala exhibited the best extracellular enzyme activities except for amylase and cellulase. The strain ONF19B isolated from O. niloticus was noted as the best extracellular enzyme producer among the strains that produced all of the extracellular enzymes studied. Sequencing of the 18S rDNA fragment followed by nucleotide blast in the National Centre for Biotechnology Information (NCBI) GenBank revealed that strains CMH6A and ONF19B were similar to Pichia kudriavzevii (Accession no. KF479403 ) and Candida rugosa (Accession no. KF479404 ), respectively. The test of antagonism (in vitro) revealed that the isolated yeasts could not affect the growth of the autochthonous gut bacteria. This might indicate likely co‐existence of autochthonous yeasts and bacteria in the fish gut. Further research is necessary to explore the possibilities of utilizing the extracellular enzyme‐producing yeasts detected in the present study for commercial aquaculture.  相似文献

Summary Previously we used a simple, sensitive agar plate method to screen lipase activity from 1229 selected cultures including 508 bacteria, 479 yeasts, 230 actinomycetes and 12 fungi covering many genera and species. About 25% of the cultures tested were lipase-positive. These lipase-positive strains were further classified as good, moderate or weak enzyme producers. We have expanded our screening method to focus specifically on the pH dependence and thermostability of these lipase activities. The lipases exhibited various pH sensitivities and were divided into three groups: (i) lipases which are active at pH 5.5 but not at pH 7.5—produced by 36 bacteria, 23 yeasts and four actinomycetes; (ii) lipases which are active at pH 7.5 but not at pH 5.5—produced by 17 bacteria, four yeasts, two actinomycetes and one fungus; and (iii) lipases which are active at both pH 5.5 and pH 7.5—produced by 112 bacteria, 90 yeasts, 15 actinomycetes and five fungi. By screening at 60°C and pH 9.0, we further identified 50 bacteria and 26 yeasts that produce thermostable alkali-tolerant lipases. Product analyses confirmed our screening results. Lipases with specific pH dependency and thermostability have potential to be developed into industrial enzymes.  相似文献

AIMS: To synthesize and evaluate fluorogenic substrates for the detection of microbial nitroreductases. These substrates, all based on 7-nitrocoumarin, may be reduced to form fluorescent aminocoumarins. METHODS AND RESULTS: Thirty pathogenic microbial strains, including both bacteria and yeasts, were examined for nitroreductase activity in a whole-cell assay. All strains readily reduced each of the seven substrates to generate fluorescence, suggesting the widespread presence of nitroreductase activity in pathogenic bacteria. CONCLUSIONS, SIGNIFICANCE AND IMPACT OF THE STUDY: These novel substrates facilitate the direct detection of nitroreductase activity and have potential as sensitive indicators of microbial growth.  相似文献

Four strains of bakers' yeast were analysed for their hyperosmotic responses when in media that mimic conditions occurring in bread doughs. Two of the strains produced strong fermentative activity in medium with low osmotic stress, but produced considerably less ethanol in high sucrose concentration medium. Two other strains produced more similar fermentation activities across the range of media tested. The strains that were inhibited by high sucrose concentration were unable to produce significant amounts of glycerol under hyperosmotic conditions. By contrast, the yeasts that were not inhibited significantly by high sucrose produced a considerable amount of glycerol. The strains that produced significant glycerol exhibited efficient expression of the glycerol-3-phosphate dehydrogenase gene GPD1. These novel data on the molecular responses of industrially relevant strains of bakers' yeasts are prerequisite to designing strategies for improving the performance of industrial yeasts in high sugar concentration media.  相似文献

The antimicrobial activities of the three diterpene dialdehydes, miogadial, galanal A and galanal B, isolated from flower buds of the myoga (Zingiber mioga Roscoe) plant were investigated with some strains of bacteria, yeasts and molds. Among the three compounds, miogadial exhibited relatively greater antimicrobial activity than the others against Gram-positive bacteria and yeasts. Galanals A and B also behaved as antimicrobial agents against Gram-positive bacteria and yeasts. The content of miogadial in the flower buds was much higher than that in the leaves, whereas galanals A and B were contained at high levels in the leaves and rhizomes.  相似文献

从中国北部湾海域采集的海蜇样品中分离到577株海洋细菌,以条件致病真菌白色念珠菌(Candida albicans)作为敏感测试菌株,采用琼脂块法测定了577株海洋细菌的抗真菌活性,结果表明有119株具有抗白色念珠菌活性,占总数的20.6%。对其中的40株高活性菌株进行抗真菌和抗细菌活性研究,结果表明,有21株海洋细菌对3种或3种以上的真菌有抑制活性,23株对细菌有不同程度的抑制作用,其中1株海洋细菌I0s4-18具有广谱的抗菌作用,对革兰氏阴性细菌、革兰氏阳性细菌、酵母菌及丝状真菌均具有抑制作用。海蜇共附生的微生物中能产生抗真菌活性的微生物的比例是很高的,I0s4-18菌株具有良好的开发前景。  相似文献

Stroptomyces strains INA T-511 and INA 3946 were isolated from 2 different soil samples. Both the strains produced an antibiotic of the group of alpha-hydroxyketopentaenes. The antibiotic was active against gram-positive bacteria, yeasts and fungi belonging to Penicillium. In addition, the strains produced a pigment antibiotic of the prodigiosin type and a pigment identified as ferroverdin. The study of the cultural and morphological features and the natural variation of the strains has shown that they belong to a new species, designated as Streptomyces variegatus sp. nov.
