本文论述了植物防御系统中抗病相关基因(resistance gene,R基因)的研究进展。列表总结了迄今已克隆的R基因,并将其归为四种不同的类型。综述了不同基因表达产物-R蛋白在细胞中的定位及其相应的功能。此外,还对R基因编码区的多态性、R基因在染色体上排列方式以及R基因的进化与起源等问题进行了讨论。 Abstract:This review comments on recent advances in research of disease resistance genes(R Genes) in defence system of plants.The R genes cloned up to date are summarized and classified roughly into four classes listed in the Table 1.The location and the founction of the R proteins,i.e.,the expressed products of different R genes in the cells are reviewed.In addition,the polymophism of coding region of R genes,the different fashions of R gene arrangement on the chromosomes,and the evolution and origin of R genes are discussed.  相似文献

将含有硫霉素环化酶基因的重组质粒p6BCl2转化变铅青链霉菌(Streptomyceslividans)TK24,含有p6BCl2的转化子细胞抽提液分别与琉霉素生物合成阻断变株Y,发酵液以及纯化的Y。中间产物经过体外共培养可产生活性物质.化学分析表明与Y,发酵液混合后产生的是硫霉素,与纯化的Y。中间产物混合产生的是一种不稳定的活性物质。说明硫霉素环化酶基因在S.lividans TK24中得到了表达,其产物以Y。中间产物为底物并弥补了Y,中的缺陷。对p6Bcl2中4.5kb外源片段进行了限制酶酶切分析,建立了酶切图谱.利用含硫霉素环化酶基因的S.Lividans TK24转化子体外转化Y,的应用体系,将硫霉素环化酶基因定位在0.9kb Hinc I—Pst I片段上,并证明了硫霉紊环化酶的活性与IPNS同源片段无关。以上实验为进一步研究琉霉素环化酶基因的结构打下了基础。  相似文献

ASR(abscisic acid,stress,ripening-induced)基因是近年来从植物中发现的一类受ABA、胁迫和成熟诱导表达的基因,具有保守的ABA/WDS结构域。ASR基因不仅参与植物对干旱、高盐、低温以及脱落酸的胁迫应答,而且参与植物生命活动的许多过程,如果实发育、成熟和糖代谢等。本文综述了近年来国内外ASR基因的研究进展,主要包括ASR基因和蛋白结构特点、ASR基因家族的进化、ASR基因的表达及可能具有的功能,为植物ASR基因研究提供参考。  相似文献

水平基因转移(horizontal gene transfer, HGT)是生物体获得遗传信息的方式之一,对生物体进化起重要作用。近年来,越来越多昆虫中的水平基因转移现象被报道,如在鳞翅目(如家蚕、甜菜夜蛾、小菜蛾、斜纹夜蛾)、半翅目(如柑橘粉蚧、烟粉虱)、鞘翅目(如咖啡果小蠹、米象、光肩星天牛)、膜翅目(如金小蜂)、双翅目(如果蝇、白纹伊蚊)等昆虫中广泛存在水平转移基因,且不同的水平转移基因对昆虫的营养合成与共生、吸收与消化、毒素产生与解毒、生长和发育、体色改变等方面有着重要作用。本文结合国内外专家学者的相关报道,就HGT的研究步骤与技术方法、评判HGT发生的方法、昆虫HGT的供体与功能几个方面进行了总结和讨论,以期更加深入地了解水平基因转移现象,为探究水平基因转移的作用机制、理解昆虫的进化、遗传和行为、并将水平基因转移应用到农业生产中为农业害虫的绿色防治提供更多思路。  相似文献

Use of germ cells as vectors for transgenesis in mammals has been well developed and offers exciting prospects for experimental and applied biology, agricultural and medical sciences.Such approach is referred to as either male germ cell mediated gene transfer (MGCMGT)or female germ cell mediated gene transfer(FGCMGT)technique.Sperm-mediated gene transfer (SMGT),including its alternative method,testis-mediated gene transfer(TMGT),becomes an established and reliable method for transgenesis.They have been extensively used for producing transgenic animals.The newly developed approach of FGCMGT,ovary-mediated gene transfer(OMGT) is also a novel and useful tool for efficient transgenesis.This review highlights an overview of the recent progress in germ cell mediated gene transfer techniques,methods developed and mechanisms of nucleic acid uptake by germ cells.  相似文献

Gene switches have wide utility in synthetic biology, gene therapy, and developmental biology, and multiple orthogonal gene switches are needed to construct advanced circuitry or to control complex phenotypes. Endogenous vascular endothelial growth factor (VEGF‐A) is crucial to angiogenesis, and it has been shown that multiple alternately spliced VEGF‐A isoforms are necessary for proper blood vessel formation. Such a necessity limits the utility of direct transgene delivery, which can provide only one splice variant. To overcome this limitation, we constructed a gene switch that can regulate the (VEGF‐A) locus in mammalian cells by combining an engineered estrogen receptor (ER) ligand‐binding domain (LBD), a p65 activation domain, and an artificial zinc‐finger DNA binding domain (DBD). Our gene switch is specifically and reversibly controlled by 4,4′‐dyhydroxybenzil (DHB), a small molecule, non‐steroid synthetic ligand, which acts orthogonally in a mammalian system. After optimization of the gene switch architecture, an endogenous VEGF‐A induction ratio of >100‐fold can be achieved in HEK293 cells at 1 µM DHB, which is the highest endogenous induction reported to date. In addition, induction has been shown to be reversible, repeatable, and sustainable. Another advantage is that the ligand response is tunable by varying the clonal composition of a stably integrated cell line. The integration of our findings with the technology to change ligand specificity and DNA binding specificity will provide the framework for generating a wide array of orthogonal gene switches that can control multiple genes with multiple orthogonal ligands. 选用不同来源的转Bt基因抗虫棉中棉所30号和新棉33B、转tfdA基因抗除草剂棉花材料TFD,通过与不同主栽推广品种正反交,研究了外源Bt基因和tfdA基因在棉株体内的遗传和分离行为。结果表明,由于外源基因提供给转基因棉花的抗虫和抗除草剂性状均是由一对于基因控制的显性性状,且该性状不受细胞质效应的影响,符合孟德尔遗传规律。  相似文献

黑曲霉纤维素酶三类不同酶系中,纤维二糖水解酶基因表达处于很低水平,导致纤维素酶总体活力水平不高。为构建黑曲霉纤维素酶高产菌株,采用基因工程方法,全基因合成拼接黑曲霉高表达葡萄糖淀粉酶基因glaA的强启动子片段与纤维二糖水解酶基因cbhB编码区片段,然后将杂合基因克隆到二元载体pCAMBIA1301上,重组质粒通过农杆菌介导转化黑曲霉分生孢子,携带杂合基因的T-DNA片段插入到黑曲霉转化子的染色体上,共筛选到48个具有潮霉素抗性的转化子。纤维素酶活力水平测定结果显示,转化子A3-9的CMC酶活力最高,为野生型黑曲霉菌株的1.31倍;转化子B1-7与A3-6的滤纸酶活力最高,为野生型黑曲霉菌株2.51倍。另外,初步分析了杂合基因在黑曲霉中的表达所需的诱导条件。  相似文献

RNA干扰(RNA interference,RNAi)是指双链RNA(double-strand RNA,dsRNA)特异性降解同源mRNA,从而引发基因转录后水平沉默的现象,是一种高效、高特异性抑制基因表达的途径。自1998年Fire等发现RNA干扰现象以来,其特异性降解目的基因的优势吸引了众多研究者的目光。本文在简要综述RNAi技术在基因功能研究、抗病毒治疗,肿瘤基因治疗等领域的应用后,重点归纳了基因枪技术在RNAi研究即siRNA导入细胞中的应用,并简单分析其优势与意义。  相似文献

Summary The ability to transfer mammalian genes parasexually has opened new possibilities for gene mapping and fine structure mapping and offers great potential for contributing to several aspects of mammalian biology, including gene expression and genetic engineering. The DNA transferred has ranged from whole genomes to single genes and smaller segments of DNA. The transfer of whole genomes by cell fusion forms cell hybrids, which has promoted the extensive mapping of human and mouse genes. Transfer, by cell fusion, of rearranged chromosomes has contributed significantly to determining close linkage and the assignment of genes to specific chromosomal regions. Transfer of single chromosomes has been achieved utilizing microcells fused to recipient cells. Metaphase chromosomes have been isolated and used to transfer single-to-multigenic DNA segments. DNA-mediated gene transfer, simulating bacterial transformation, has achieved transfer of single-copy genes. By utilizing DNA cleaved with restriction endonucleases, gene transfer is being employed as a bioassay for the purification of genes. Gene mapping and the fate of transferred genes can be examined now at the molecular level using sequence-specific probes. Recently, single genes have been clones into eucaryotic and procaryotic vectors for transfer into mammalian cells. Moreover, recombinant libraries in which entire mammalian genomes are represented collectively are a rich new source of transferable genes. Methodology for transferring mammalian genetic information and applications for mapping mammalian genes is presented and prospects for the future discussed.
