为了合理利用羌活和宽叶羌活的药用植物资源,同时保护其物种多样性,该研究利用SSR分子标记技术对羌活与宽叶羌活邻域及异域分布的23个自然种群,共计227个个体进行多样性和种间分化研究。结果显示:(1)两个物种具有中等水平的遗传多样性;羌活的平均等位基因数(N_a)、有效等位基因数(N_e)和期望杂合度(H_e)分别为2.603、1.777和0.313,均高于宽叶羌活(分别为2.200、1.641和0.308)。(2)分子方差分析表明,两个物种的遗传变异主要存在于群体内,羌活和宽叶羌活群体间分化系数分别为0.181和0.191。(3)Structure聚类分析和主坐标分析(PCoA)将所有取样个体分为两大遗传组分,分别对应于羌活和宽叶羌活两个物种,二者间存在着有限的基因交流。研究表明,羌活与宽叶羌活物种间存在较高程度的遗传分化,并且遗传变异主要源自群体内,应各自划分为不同的地理单元进行多样性保护。  相似文献

近缘物种筛选法是获取微卫星引物的主要方法之一。利用松属植物EST-SSRs的种间通用性,从马尾松微卫星引物中筛选出红松EST-SSR标记,并与已有的红松基因组SSR标记进行对比分析。采集长白山露水河红松母树林4个种群的248个样本,分析种群遗传多样性。结果表明:马尾松与红松的种间通用率为27.27%,松属内不同亚属物种间的种间通用性偏低;与红松基因组SSRs相比,EST-SSRs的单位点等位基因数(N a)、有效等位基因数(Ne)、观测杂合度(Ho)、期望杂合度(He)等遗传多样性参数均较低;用2组引物分析4个种群的遗传多样性,发现有效等位基因均能反映出不同种群遗传变异的趋势,基因多样性均能反映出遗传同一性的差异,说明两组引物都能准确地反映红松种群的遗传多态性水平,新筛选的EST-SSRs可以用于红松种群遗传多样性研究。  相似文献

薄叶金花茶、小花金花茶和小瓣金花茶是三种濒危金花茶植物,为了解珍稀濒危植物遗传多样性和遗传结构,该研究利用微卫星标记对他们的7个种群共184个个体进行了遗传多样性和遗传结构分析。结果表明:11个位点共检测到等位基因92个。在物种水平上,小瓣金花茶平均等位基因数(N_A)为3.9、有效等位基因数(N_E)为2.328、观测杂合度(H_o)为0.520、期望杂合度(H_e)为0.501,高于薄叶金花茶和小花金花茶。在种群水平上,有效等位基因数(N_E)在1.788~2.466之间,期望杂合度(H_e)在0.379~0.543之间;种群间遗传分化系数(FST)在0.143 7~0.453 3之间,种群间基因流(N_m)在0.301 5~1.488 9之间。AMOVA分子变异分析显示65.72%的变异存在于种群内。三种金花茶具有较低水平的遗传多样性和高水平的种群间遗传分化。STRUCTURE和PCoA种群遗传结构分析结果将取样种群分为2组,即薄叶金花茶和小花金花茶大部分个体分为一组,小瓣金花茶大部分个体分为一组。现存所有种群应根据实际情况尽快采取就地保护或迁地保护措施。  相似文献

岛屿具有独特的生态系统,常被生态学家和进化生物学家视为研究生物进化的天然实验室,岛屿生物地理学也受到了越来越多科学家的关注。对舟山群岛8个面积不等岛屿的黄毛鼠(Rattus losea)种群进行了调查,分析了8个种群的遗传变异特征,对探讨岛屿理论中的种群动态和种群分化具有重要意义。采用线粒体分子标记技术,利用PCR扩增得到D-loop区基因序列815 bp,在330个黄毛鼠样本中共识别出15个单倍型,平均核苷酸多样性(P_i)为0.001,平均单倍型多样性(H_d)为0.364,表明舟山群岛黄毛鼠种群的遗传多样性较低。Tajima′s D中性检验显示除了小盘峙种群,均为显著负值(P0.01),表明种群受到了自然选择的作用,历史上发生过种群扩张。AMOVE显示,群体间的遗传分化指数平均值为0.745,处于较高的分化水平,表明遗传变异主要来自种群间,占74.5%。基于线粒体D-loop区序列构建的系统发育树和中值网络都表明8个岛屿的黄毛鼠种群起源于两个母系。此外,Mental检验显示不同岛屿种群间的遗传距离与岛屿间地理距离之间存在显著正相关关系(r=0.6077,P=0.004),种群遗传多样性与岛屿面积并未发现显著相关性(r=0.6255,P=0.1840)。研究结果可为岛屿黄毛鼠种群的微观演化以及一些岛屿物种的进化理论提供参考。  相似文献

以千岛湖地区13个岛屿上的社鼠(Niviventer confucianus)种群为研究对象,利用13个微卫星位点分析了13个社鼠种群的遗传现状,并探讨了种群遗传现状与岛屿面积大小之间的关系。研究结果显示,13个社鼠种群的平均观测等位基因数(Na)与平均有效等位基因数(Ne)分别为7.385,5.952,平均期望杂合度(He)与平均观测杂合度(Ho)分别为0.819,0.930,平均多态信息含量(PIC)为0.760,说明13个社鼠种群均具有较高的遗传多样性。13个种群间的分化系数(Fst)为0.053,表明种群间的遗传分化较小。3个面积较大且相近岛屿上社鼠种群,即高仙阁种群 (Gaoxiange Island,G);乌石T岛种群 (Wushi T Island,WT);东门岛种群 (Dongmen Island,DM)间的Fst值均小于0.05,表明种群间无分化,而小岛种群与大岛种群间的分化却比较明显。此外,一元回归结果显示,多态信息含量(PIC)和Shannon多样性指数(I)与岛屿面积大小之间均存在显著地正相关关系。以上结果说明,在社鼠种群进化过程中,生境面积的缩小可能使种群的遗传结构发生改变,并有可能导致种群的快速进化。  相似文献

【目的】中华蜜蜂Apis cerana cerana是一种兼有生态价值和经济价值的授粉昆虫。本研究拟揭示秦巴山区中华蜜蜂种群遗传多态性现状,探讨中华蜜蜂的种群分化机制及其影响因素。【方法】使用8个微卫星DNA标记评估秦巴山区17个样点共979个蜂群的中华蜜蜂遗传多态性,以长白山中华蜜蜂和阿坝中华蜜蜂作为外群进行种群遗传分化分析。【结果】秦巴山区中华蜜蜂95%的差异来源于样点内,样点间遗传分化系数(Fst)为0.002~0.037,基因流参数Nm为6.51~124.75,与外群的遗传分化分析中均显示秦巴山区中华蜜蜂不存在遗传分化。平均期望杂合度(He)为0.6877±0.1098,平均观察杂合度(Ho)为0.6364±0.1367,平均有效等位基因数(Ne)为3.7488±1.6201个,平均等位基因数(Na)为7.71±2.52个,多态信息含量(PIC)为0.6418±0.1152,香农指数(I)为1.5026±0.3754,这些参数均表明微卫星遗传多态性丰富。中蜂囊状幼虫病流行过的地区,其杂合度、多态信息含量以及等位基因数均显著低于未发病地区。【结论】秦巴山区中华蜜蜂种群数量大,分布均匀,基因流水平高,在650 km距离范围内没有发生种群分化;秦巴山区中华蜜蜂微卫星遗传多态性丰富,仅部分地区中华蜜蜂遗传多态性受中蜂囊状幼虫病的选择压而降低。  相似文献

为分析甘肃金鳟遗传资源状况,研究利用18个微卫星DNA标记,对甘肃金鳟、日本金鳟和道氏虹鳟3群体各50尾个体进行了遗传分析,以评价甘肃金鳟遗传资源状况并研究其与日本金鳟和道氏虹鳟遗传分化情况。结果表明甘肃金鳟具有较丰富的遗传变异,15个微卫星DNA座位共获得了51个等位基因,有效等位基因数(Ne)为1.13,多态信息含量(PIC)为0.440,平均观察杂合度(Ho)和期望杂合度(He)分别为0.430和0.498。在Hardy-Weinberg平衡条件下,进行了P检验,发现3个群体均有位点发生了显著偏离;对3个群体进行了Fst值计算,表明群体间存在着一定程度的遗传分化,但遗传变异绝大部分存在于品种内。综合分析,认为甘肃金鳟遗传性状基本稳定,遗传变异度较高,遗传多样性相对丰富,具有较大的遗传潜力,与道氏虹鳟和日本金鳟群体间分化明显,人工选育对群体的遗传变异产生了一定的影响。研究结果对甘肃金鳟的保种、选育及其合理的推广应用具有一定的指导作用。  相似文献

为了解猴耳环(Archidendron clypearia)种质资源的遗传多样性,以广东省12个野生猴耳环群体的146份种质资源为材料,采用SSR分子标记技术对其遗传多样性和亲缘关系进行分析。结果表明,21对SSR引物共检测到249个等位基因,平均每对SSR引物检测的等位基因数(Na)为11.857,有效等位基因数(Ne)为3.500,期望杂合度(He)为0.718,多态信息含量(PIC)为0.676;12个群体中博罗群体的Shannon多样性指数(I=0.528)和有效等位基因数(Ne=0.716)均最大,是遗传多样性最丰富的群体;群体间的遗传分化系数为0.071,AMOVA分析表明,猴耳环的遗传变异主要在群体内(97%),群体内的遗传分化大于群体间。聚类分析表明,遗传系数在0.16时,可将12个群体分为6大类,与主坐标分析的结果大致相同。这为发掘、利用与保护猴耳环群体种质资源,开展猴耳环优良品种的遗传育种提供重要的理论依据。  相似文献

按胸径将福建武夷山大安源样地的甜槠(Castanopsis eyrei)群体划分为成体、小树、幼苗3个世代,利用SSR分子标记对不同世代的甜槠遗传多样性及遗传分化进行分析,旨在揭示其不同世代间的遗传变异规律,为甜槠资源的保护与利用提供科学依据。14对SSR引物共检测到92个等位基因,平均每位点的等位基因数A=6.571 4,居群的平均有效等位基因数Ae=3.905 4,平均期望杂合度He=0.722 9,表明甜槠群体具有丰富的遗传变异。SSR分析显示3个世代的Ae、He、Nei指数(h)、Shannon信息指数(I)均以幼苗最高,小树次之,成体最低,幼苗的遗传多样性指数高于成体及小树,且幼苗中出现最多的稀有等位基因数,表明甜槠种群世代间的遗传多样性呈稳定上升趋势。分子方差分析(AMOVA)表明甜槠群体不同世代内、世代间均存在遗传变异,但遗传变异主要存在于世代内。SSR分析显示,甜槠不同世代间的遗传分化系数Fst=0.074 3,基因流Nm=3.115 4。甜槠不同世代间的遗传相似度以成体与幼苗最小,遗传距离以成体与幼苗间最大。基于甜槠群体SSR的研究结果,认为自然保护区的建立对物种遗传多样性的保护具有重要作用,并提出在遗传多样性保护中应注重保护成体和幼苗中稀有的等位变异。  相似文献

用ISSR分子标记对内蒙古地区刺叶柄棘豆(Oxytorpis aciphylla Ledeb.) 5个地理种群进行了种群遗传多样性分析。结果表明:刺叶柄棘豆种群具有较高的遗传多样性,11个ISSR引物扩增出215条带,总的多态位点百分率为98.14%,Shannon多样性指数I=0.2108,Nei基因多样性指数 H=0.341 6,种内总基因多样性(Ht) 为0.2108,种群内基因多样性(Hs)为0.160 4, 大部分遗传变异(76.11%)的遗传变异存在于种群内, 23.89%的遗传变异存在于种群间。遗传分化系数(Gst)为0.238 9,基因流(Nm)为1.592 9。5个居群间已有遗传分化趋势,遗传漂变不会引起遗传分化。UPGMA遗传距离聚类结果表明, 5个地理种群中,植被类型为荒漠草原的4个种群之间遗传距离较近,与1个荒漠种群距离较远。  相似文献

Multiple paternity within litters has been recorded among a variety of small mammal species, including some species of rodents. Although multiple mating has been observed in wild Rattus populations, whether such mating results in litters with multiple paternities has not been established previously. For studies involving invasive species, awareness is useful of the level of genetic diversity a single pregnant invader can bring to a population. Multiple paternity is a means of providing additional genetic diversity to founding populations of rats on islands, which might improve population fitness. We used a genetic approach to confirm that multiple paternity occurs in wild populations of two rat species (Rattus norvegicus and R. rattus) in New?Zealand. This was accomplished by genotyping litters of embryos in pregnant females, and subtracting the known maternal alleles to find the number of paternal alleles necessary to form the litter. The number of paternal contributors cannot be overestimated by this method, befitting a conservative approach to the detection of multiple paternity, but can be underestimated. We used simulations to investigate the level of underestimation likely under two possible scenarios involving multiple paternity.  相似文献

Elucidating the demographic and landscape features that determine the genetic effects of habitat fragmentation has become fundamental to research in conservation and evolutionary biology. Land-bridge islands provide ideal study areas for investigating the genetic effects of habitat fragmentation at different temporal and spatial scales. In this context, we compared patterns of nuclear microsatellite variation between insular populations of a shrub of evergreen broad-leaved forest, Loropetalum chinense, from the artificially created Thousand-Island Lake (TIL) and the Holocene-dated Zhoushan Archipelago of Southeast China. Populations from the TIL region harboured higher levels of genetic diversity than those from the Zhoushan Archipelago, but these differences were not significant. There was no correlation between genetic diversity and most island features, excepting a negative effect of mainland–island distance on allelic richness and expected heterozygosity in the Zhoushan Archipelago. In general, levels of gene flow among island populations were moderate to high, and tests of alternative models of population history strongly favoured a gene flow-drift model over a pure drift model in each region. In sum, our results showed no obvious genetic effects of habitat fragmentation due to recent (artificial) or past (natural) island formation. Rather, they highlight the importance of gene flow (most likely via seed) in maintaining genetic variation and preventing inter-population differentiation in the face of habitat ‘insularization'' at different temporal and spatial scales.  相似文献

Three polymorphic microsatellite loci were isolated and employed to examine paternity patterns in two natural populations of live-bearing mosquitofish, Gambusia holbrooki. Each locus displayed four to five alleles per population in samples of presumably unrelated adults. Nearly 900 embryos from a total of 50 pregnant females were assayed individually, and paternal alleles in each embryo were identified. Counts of paternal alleles, Mendelian segregation patterns, multilocus allelic associations and genetic relatedness coefficients were employed to estimate the minimum and effective numbers of fathers per brood. At least 90% of the assayed broods were shown to have been fathered by multiple males, a figure substantially higher than previous estimates based on less polymorphic genetic loci. However, the genetic data yield a face-value estimate of only about 2.2 fathers per brood, a number that seems perhaps surprisingly low based on frequencies of attempted copulations by males. Both biological and sampling factors that might bias mean sire counts downward are considered. Although higher sire counts per brood might be obtained from loci with even greater numbers of alleles, little statistical room remains for higher frequency estimates of multiple paternity in Gambusia.  相似文献

【目的】揭示岛屿隔离效应对舟山群岛及邻近大陆碧蛾蜡蝉Geisha distinctissima种群遗传多样性的影响。【方法】依据本研究获取的舟山群岛(11个岛屿)及邻近大陆15个种群共247头碧蛾蜡蝉样本的16S rRNA基因序列,对该地区种群进行遗传多样性评估;采用贝叶斯推断法构建系统发育树并绘制单倍型网络图,以解析种群间的遗传结构;通过中性检验、错配分布分析和贝叶斯天空线(Bayesian skyline plot, BSP)分析,探讨该地区碧蛾蜡蝉的种群历史动态。【结果】贝叶斯系统树及单倍型网络图均没有解析出舟山群岛碧蛾蜡蝉种群明显的遗传结构。且分子方差分析发现,遗传变异主要集中在种群内部(种群内变异占68.26%),种群间没有明显的遗传分化,显示了该地区碧蛾蜡蝉种群遗传多样性的均质化。不同种群间的单倍型多态性和核苷酸多态性与大陆-岛屿距离及岛屿隔离时间不相关。中性检验分析检测到Tajima’s D和Fu’s Fs均呈显著的负值,且错配分布分析呈现出典型的单峰,表明该地区碧蛾蜡蝉种群发生了近期的种群扩张事件。BSP分析表明,这种扩张事件发生在大约0.5~3.5千年前。【结论】岛屿隔离效应可能不足以驱使舟山群岛及邻近大陆的碧蛾蜡蝉种群遗传多样性产生明显的差异。  相似文献

The identification of the factors responsible for genetic variation and differentiation at adaptive loci can provide important insights into the evolutionary process and is crucial for the effective management of threatened species. We studied the impact of environmental viral richness and abundance on functional diversity and differentiation of the MHC class Ia locus in populations of the black‐spotted pond frog (Pelophylax nigromaculatus), an IUCN‐listed species, on 24 land‐bridge islands of the Zhoushan Archipelago and three nearby mainland sites. We found a high proportion of private MHC alleles in mainland and insular populations, corresponding to 32 distinct functional supertypes, and strong positive selection on MHC antigen‐binding sites in all populations. Viral pathogen diversity and abundance were reduced at island sites relative to the mainland, and islands housed distinctive viral communities. Standardized MHC diversity at island sites exceeded that found at neutral microsatellites, and the representation of key functional supertypes was positively correlated with the abundance of specific viruses in the environment (Frog virus 3 and Ambystoma tigrinum virus). These results indicate that pathogen‐driven diversifying selection can play an important role in maintaining functionally important MHC variation following island isolation, highlighting the importance of considering functionally important genetic variation and host–pathogen associations in conservation planning and management.  相似文献

Understanding the factors that contribute to loss of genetic diversity in fragmented populations is crucial for conservation measurements. Land‐bridge archipelagoes offer ideal model systems for identifying the long‐term effects of these factors on genetic variations in wild populations. In this study, we used nine microsatellite markers to quantify genetic diversity and differentiation of 810 pond frogs (Pelophylax nigromaculatus) from 24 islands of the Zhoushan Archipelago and three sites on nearby mainland China and estimated the effects of the island area, population size, time since island isolation, distance to the mainland and distance to the nearest larger island on reduced genetic diversity of insular populations. The mainland populations displayed higher genetic diversity than insular populations. Genetic differentiations and no obvious gene flow were detected among the frog populations on the islands. Hierarchical partitioning analysis showed that only time since island isolation (square‐root‐transformed) and population size (log‐transformed) significantly contributed to insular genetic diversity. These results suggest that decreased genetic diversity and genetic differentiations among insular populations may have been caused by random genetic drift following isolation by rising sea levels during the Holocene. The results provide strong evidence for a relationship between retained genetic diversity and population size and time since island isolation for pond frogs on the islands, consistent with the prediction of the neutral theory for finite populations. Our study highlights the importance of the size and estimated isolation time of populations in understanding the mechanisms of genetic diversity loss and differentiation in fragmented wild populations.  相似文献

本文利用13对微卫星标记,对我国3个核心种源地(巴山、秦岭、川西高原)圈养林麝种群进行遗传多样性和遗传结构分析。在167份样品中共检测到142个等位基因(Na),每个位点等位基因数介于7~16,均值为10.92,平均有效等位基因数(Ne)为6.3730,期望杂合度(He)和观测杂合度(Ho)均值分别为0.8302和0.3897。这些圈养林麝种群遗传多样性水平较高,但较低的观测杂合度表明圈养群体存在近交现象。两两群体间的Fst 值和AMOVA分析结果均表明种群之间分化程度不明显。群体遗传结构分析显示,全部样本聚为3个遗传簇(最佳K值=3),其主体与3个地理来源相符,但种群间存在基因渗透现象。本研究中的秦岭种群遗传变异最为丰富,可以作为种质改良的基因池。  相似文献

Genetic variation within and among populations is influenced by the genetic content of the founders and the migrants following establishment. This is particularly true if populations are small, migration rate low and habitats arranged in a stepping-stone fashion. Under these circumstances the level of multiple paternity is critical since multiply mated females bring more genetic variation into founder groups than single mated females. One such example is the marine snail Littorina saxatilis that during postglacial times has invaded mainland refuge areas and thereafter small islands emerging due to isostatic uplift by occasional rafting of multiply mated females. We modelled effects of varying degrees of multiple paternity on the genetic variation of island populations colonised by the founders spreading from the mainland, by quantifying the population heterozygosity during both the transient colonisation process, and after a steady state (with migration) has been reached. During colonisation, multiple mating by males increased the heterozygosity by in comparison with single paternity, while in the steady state the increase was compared with single paternity. In the steady state the increase of heterozygosity due to multiple paternity is determined by a corresponding increase in effective population size. During colonisation, by contrast, the increase in heterozygosity is larger and it cannot be explained in terms of the effective population size alone. During the steady-state phase bursts of high genetic variation spread through the system, and far from the mainland this led to short periods of high diversity separated by long periods of low diversity. The size of these fluctuations was boosted by multiple paternity. We conclude that following glacial periods of extirpation, recolonization of isolated habitats by this species has been supported by its high level of multiple paternity.  相似文献

利用改良FIASCO法(Fast Isolation by AFLP Sequences COntaining repeats)开发出的9对多态性SSR引物评价了薇菜(Osmunda japonica Thunb.)2个野生居群(庐山和恩施)、1个栽培居群(恩施)的遗传多样性和遗传分化水平。结果显示,9个SSR标记在3个薇菜居群中共检测到47个等位基因,每个SSR位点的平均等位基因数为5.222个,观测杂合度和期望杂合度分别为0.000~0.944和0.577~0.834,香农指数为0.962~1.860,表明各SSR位点多态性较高;各居群的平均期望杂合度均大于平均观测杂合度且种内近交系数均为正值,说明3个薇菜居群中都存在非随机交配现象;对各居群的相关遗传多样性参数分析表明,恩施野生居群遗传多样性最高,而其栽培居群最低;庐山野生居群与恩施野生居群间遗传分化系数为0.092,说明两地野生薇菜居群的遗传分化程度较低,AMOVA分析也表明遗传变异主要存在于野生居群内部。  相似文献
