Functional architecture of the AChE active center appears to be characterized by both structural “rigidity”, necessary to stabilize the catalytic triad as well as by flexibility in accommodating the different, high affinity AChE ligands. These seemingly conflicting structural properties of the active center are demonstrated through combination of structural methods with kinetic studies of the enzyme and its mutant derivatives with plethora of structurally diverse ligands and in particular with series of stereoselective covalent and noncovalent AChE ligands. Thus, steric perturbation of the acyl pocket precipitates in a pronounced stereoselectivity toward methylphosphonates by disrupting the stabilizing environment of the catalytic histidine rather than through steric exclusion demonstrating the functional importance of the “rigid” environment of the catalytic machinery. The acyl pocket, the cation-binding subsite (Trp86) and the peripheral anionic subsite were also found to be directly involved in HuAChE stereoselectivity toward charged chiral phosphonates, operating through differential positioning of the ligand cationic moiety within the active center. Residue Trp86 is also a part of the “hydrophobic patch” which seems flexible enough to accommodate the structurally diverse ligands like tacrine, galanthamine and the two diastereomers of huperzine A. Also, we have recently discovered further aspects of the role of both the unique structure and the flexibility of the “hydrophobic patch” in determining the reactivity and stereoselectivity of HuAChE toward certain carbamates including analogs of physostigmine. In these cases the ligands are accommodated mostly through hydrophobic interactions and their stereoselectivity delineates precisely the steric limits of the pocket. Hence, the HuAChE stereoselectivity provides a sensitive tool in the in depth exploration of the functional architecture of the active center. These studies suggest that the combination of “rigidity” and flexibility within the HuAChE gorge are an essential element of its molecular design.  相似文献

A new method for monitoring reactions catalyzed by an immobilized enzyme, cross-linked penicillin acylase aggregates (PA CLEA), is suggested. Appropriate chromogenic substrates for spectrophotometric assay of catalytic activity of immobilized enzyme were chosen and their kinetic parameters determined. Active sites in PA CLEA preparations were titrated by the suggested method; it is shown that almost all active sites are retained during immobilization. This method is characterized as highly expressive, simple, and precise and may be used for control of PA immobilization efficiency as well as for study of operational, thermal, and pH stability of immobilized enzyme preparations.  相似文献

Contrary to previous reports, the functional and spectral properties of “monomeric” shark cytochrome c oxidases are not entirely similar to those of the “dimeric” beef enzyme. Most significantly, unlike the behavior of beef oxidase, the fully oxidized shark enzyme is not reducible by carbon monoxide. Also, preparations of the shark enzyme, isolated at pH 7.8-8.0, lead to more than 60% of the sample always being obtained in a resting form, whereas similarly prepared beef oxidase is very often obtained, both by ourselves and others, exclusively in the pulsed form. Although the electronic absorption, magnetic circular dichroism and electron paramagnetic resonance (EPR) spectra of cytochrome c oxidase obtained from several shark species are similar to those of the beef enzyme, there are some significant differences. In particular, the Soret maximum is at 422 nm in the case of the fully oxidized resting shark oxidases at physiological pH and not 418 nm as commonly found for the beef enzyme. Moreover, the resting shark oxidases do not necessarily exhibit a “g = 12” signal in their EPR spectra. The turnover numbers of recent preparations of the shark enzyme are higher than previously reported and, interestingly, do not differ within experimental uncertainty from those documented for several beef isoenzymes assayed under comparable conditions.  相似文献

The preparation of crosslinked aggregates of pancreatic porcine lipase (PPL‐CLEA) was systematically studied, evaluating the influence of three precipitants and two crosslinking agents, as well as the use of soy protein as an alternative feeder protein on the catalytic properties and stability of the immobilized PPL. Standard CLEAs showed a global yield (CLEA’ observed activity/offered total activity) of less than 4%, whereas with the addition of soy protein (PPL:soy protein mass ratio of 1:3) the global yield was approximately fivefold higher. The CLEA of PPL prepared with soy protein as feeder (PPL:soy protein mass ratio of 1:3) and glutaraldehyde as crosslinking reagent (10 μmol of aldehyde groups/mg of total protein) was more active mainly because of the reduced enzyme leaching in the washing step. This CLEA, named PPL‐SOY‐CLEA, had an immobilization yield around 60% and an expressed activity around 40%. In the ethanolysis of soybean oil, the PPL‐SOY‐CLEA yielded maximum fatty acid ethyl ester (FAEE) concentration around 12‐fold higher than that achieved using soluble PPL (34 h reaction at 30°C, 300 rpm stirring, soybean oil/ethanol molar ratio of 1:5) with an enzyme load around 2‐fold lower (very likely due to free enzyme inactivation). The operational stability of the PPL‐SOY‐CLEA in the ethanolysis of soybean oil in a vortex flow type reactor showed that FAEE yield was higher than 50% during ten reaction cycles of 24 h. This reactor configuration may be an attractive alternative to the conventional stirred reactors for biotransformations in industrial plants using carrier‐free biocatalysts. © 2018 American Institute of Chemical Engineers Biotechnol. Butyrylcholinesterase (BChE, EC 3.1.1.8) is important in human cocaine metabolism despite its limited ability to hydrolyze this drug. Efforts to improve the catalytic efficiency of this enzyme have led to a quadruple mutant cocaine hydrolase, “CocH”, that in animal models of addiction appears promising for treatment of overdose and relapse. We incorporated the CocH mutations into a BChE–albumin fusion protein, “Albu-CocH”, and evaluated the pharmacokinetics of the enzyme after i.v. injection in rats. As assessed from the time course of cocaine hydrolyzing activity in plasma, Albu-CocH redistributed into extracellular fluid (16% of estimated total body water) with a t1/2 of 0.66 h and it underwent elimination with a t1/2 of 8 h. These results indicate that the enzyme has ample stability for short-term applications and may be suitable for longer-term treatment as well. Present data also confirm the markedly enhanced power of Albu-CocH for cocaine hydrolysis and they support the view that Albu-CocH might prove valuable in treating phenomena associated with cocaine abuse.  相似文献

The concept of the effective population size is discussed. It is shown that the “eigenvalue” and the “inbreeding” effective population sizes are in principle different, even though they have been sometimes identified in the literature. On the other hand the “eigenvalue” and “variance” effective sizes are usually both close when the latter exists. Since, however, there are many models for which a variance effective size cannot in principle exist, it seems useful to introduce the eigenvalue effective size and to examine some of its properties.  相似文献

Cage enrichment devices (ED), frequently termed cage “toys,” are often provided to captive parrots as a means of promoting a behaviorally stimulating environment, but it is not clear whether particular properties of EDs are more effective than others in eliciting engagement with them. We tested preference for color, size and hardness of cube-shaped EDs constructed from wood and of color preference for EDs constructed from flat rawhide rectangles. Orange-winged Amazon parrots (Amazona amazonica; N = 8–10, mixed-sex, 4–5 years of age) were individually housed in cages each equipped with two computer-monitored omni-directional lever-type switches attached to cage ceilings. EDs were attached to the switches; any interactions generating lateral movement and causing switch closure (operationally constituting “use” of EDs) were continuously recorded. Preference for 3.8 cm3 softwood (Douglas fir) cubes dyed in eight different colors was tested by presenting each bird with all combinations of colors, two colors at a time. Daily switch activity averages were computed for each bird and subjected to repeated-measures ANOVA: yellow cubes elicited greater use than red, green, blue, violet or natural cubes (P < 0.05; but see below) and orange cubes were preferred over green and blue cubes (P < 0.05). Color exerted no effect in a comparable trial of rawhide rectangles. Preference for size of yellow hardwood cubes was tested by presenting combinations of cubes of three sizes: 2.5, 3.8, and 5.1 cm3; the smallest blocks were preferred over the largest size (P < 0.001). Preference for hardness of wood was tested by presenting birds with 3.8 cm3 yellow cubes and blue cubes made of either Douglas fir (“soft”) or birch/maple (“hard”) wood; birds preferred softer cubes (P < 0.0002), but there was no significant preference of yellow over blue. The results show that color, hardness, size and material all influence ED use by captive Amazon parrots. This constellation of preferences may reflect properties of foods native to Orange-winged Amazon natural habitat.  相似文献

Several analytical methods were compared to evaluate characteristic wood decaying fungi for their potential to depolymerise lignin on spruce wood particles. Wood samples were treated with the white rot fungi Phlebia brevispora, Ceriporiopsis subvermispora, Merulius tremellosus, Pycnoporus sanguineus, Trametes pubescens and with the brown rot fungus Gloeophyllum trabeum. The UV absorbancies of crude ethanol extracts, total extractives content from sequential extraction, ligninolytic enzyme activities, lignin solubilisation and decrease of lignin content were compared. It was shown, that, in early decay stages, UV absorbancies of crude ethanol extracts and total extractives content correlate well with lignin degradation, increase of acid soluble lignin and increased production of ligninolytic enzymes (total peroxidase). Lignin content was determined using FT-NIR spectroscopy as well as by wet-chemical analysis, indicating a very good correlation between the two methods. According to the different analytical methods, the tested fungi can be classified into three categories based on their characteristic behaviour: brown rot, “slow” and “fast” white rot.  相似文献

Pseudomonas aeruginosa produces a fucose-binding lectin (PA-IIL) which strongly binds to human cells. This lectin was shown to be highly sensitive to inhibition by fucose-bearing human milk glycoproteins. Since the glycans of these glycoproteins mimic human cell receptors, they may function as decoys in blocking lectin-dependent pathogen adhesion to the host cells. Human saliva and seminal fluid also contain such compounds, and body fluids of individuals who are “secretors” express additional fucosylated (alpha 1,2) residues. The latter are selectively detected by Ulex europaeus lectin UEA-I. The aim of the present research was to compare the PA-IIL and UEA-I interactions with human salivas and seminal fluids of “secretors” and “nonsecretors” with those obtained with the respective milks. Using hemagglutination inhibition and Western blot analyses, we showed that PA-IIL interactions with the saliva and seminal fluid glycoproteins were somewhat weaker than those obtained with the milk and that “nonsecretor” body fluids were not less efficient than those of “secretors” in PA-IIL blocking. UEA-I, which interacted only with the “secretors” glycoproteins, was most sensitive to those of the seminal fluids.  相似文献

Peptide:N-glycanase (PNGase) releases N-glycans from glycoproteins/glycopeptides. Cytoplasmic PNGase is widely recognized as a component of machinery for ER-associated degradation (ERAD), i.e. proteasomal degradation of misfolded, newly synthesized (glyco)proteins that have been exported from the ER. The enzyme belongs to the “transglutaminase superfamily” that contains a putative catalytic triad of cysteine, histidine, and aspartic acid. The mammalian orthologues of PNGase contain the N-terminal PUB domain that serves as the protein–protein interaction domain. The C-terminus of PNGase was recently found to be a novel carbohydrate-binding domain. Taken together, these observations indicate that C-terminus of mammalian PNGase is important for recognition of the substrates while N-terminus of this enzyme is involved in assembly of a degradation complex.  相似文献

Photosynthetic responses to irradiance and temperature of “leaves” and receptacles were compared in February (vegetative stage) and May (reproductive stage) in the seaweed, Hizikia fusiforme (Harvey) Okamura (Sargassaceae, Phaeophyta) from Nanao Island, Shantou, China. Irradiance-saturated photosynthesis (Pmax) was significantly higher in receptacles than in “leaves” on a fresh weight basis, and that of “leaves” was greater in May than in February at ambient seawater temperatures. The optimum temperature for Pmax was 30C for both “leaves” and receptacles, being 5–10C higher than the ambient seawater temperature. The apparent photosynthetic efficiencies were greater in receptacles than in “leaves” within the tested temperature range of 10–40C. The irradiance for saturating photosynthesis for both “leaves” and receptacles was temperature-dependent, with the highest values (about 200μmolphotonsm−2s−1) at 30C.  相似文献

The role played by the surface charge density of the phospholipid coat of nanometer-sized Fe3O4 colloids (so-called “magnetoliposomes”) in the catalytic activity of beef heart cytochrome c oxidase was investigated. Screening of various binary mixtures of the anionic dimyristoylphosphatidylglycerol and the zwitterionic dimyristoylphosphatidylcholine demonstrated that the highest degree of reactivation was found in the lower negative charge range. Pre-incubation of the charged colloidal biocatalytic particles with cytochrome c induced aggregation and reduced overall enzymatic activity. The results are interpreted in terms of a different affinity of the substrate for the various membrane types and of a reorganisation of the enzyme within the membrane matrices.  相似文献

Among the features of protein kinase CK2, autophosphorylation at its β-subunit(s) upon incubation with ATP/Mg++ was early detected as a rapid and stoichiometric event occurring through an intramolecular mechanism as judged from kinetic analyses. The autophosphorylation site was mapped to Ser2 and, to a lesser extent, Ser3 both fulfilling the CK2 consensus sequence (MSSSEEV). The crystal structure of the heterotetrameric holoenzyme, however, is not compatible with an intramolecular autophosphorylation of the N-terminal stretch of either of the two β subunits. Here we show that efficient “intramolecular” autophosphorylation of the β subunit is crucially dependent on the formation of oligomers composed by several holoenzyme heterotetrameric protomers. Increasing ionic strength of the incubation medium promoting dissociation of the supramolecular oligomers abrogates β subunit autophosphorylation, although CK2 catalytic activity, as judged from the phosphorylation of exogenous substrates, is still quite evident. These findings, in conjunction with graphic modelization, support the view that CK2 autophosphorylation at its β subunits takes place through an “intraoligomeric” mechanism where the β subunits of a protomer are phosphorylated by the catalytic subunits of another adjacent protomer. It appears therefore that in vivo β autophosphorylation is symptomatic of supramolecular CK2 oligomers.  相似文献

A sample of 84 female subjects (students) was taken and split up on the basis of the IST-ratings (“Intelligence Structure Test” by Amthauer) into two extreme groups of “more intelligent” (N = 14) and “less intelligent” (N = 11) subjects with an aim to testing Kozeny's findings. Using his photographic-statistical method, Kozeny had observed that “more intelligent” people have a more open eye, a smaller and straighter mouth, a stronger chin, and—above all in its side dimensions—a better developed forehead. These physiognomic indicators of differences in intelligence were confirmed to a large extent by the measurements taken from the portraits of the subjects.  相似文献

The large molecular weight polyaldehyde (macrofixative) obtained by periodate oxidation of dextran has been shown to react with the external cell surface. Chick embryo fibroblasts (CEF), which had been treated with macrofixative (MFx), were examined for the effect on the rates of sugar transport. The low “basal” rate of sugar uptake, seen in confluent (high density) cultures of CEF was unaffected by such treatment. Low density (rapidly growing) cultures, oncogenically transformed cultures, and glucose-starved cultures have rates of sugar uptake that are significantly higher than the “basal” level. Macrofixation was found to inhibit the induction of higher rate of glucose transport under all of these conditions. The results indicate that a difference may exist between the sugar transport system in resting (confluent) cells, and that in “derepressed” cells.  相似文献

Historians and philosophers of science have furnished a wide array of theoretical-historiographical terms to emphasize the discontinuities among different systems of knowledge. Some of the most famous include Thomas Kuhn’s “paradigm”, Michel Foucault’s “episteme”, and the notion of “styles of reasoning” more recently developed by Ian Hacking and Arnold Davidson. This paper takes up this theoretical-historiographical thread by assessing the values and limitations of the notion of “style” for the historical and philosophical study of science. Specifically, reflecting on various methodological and theoretical concerns prompted by sexuality, translation, and East Asian studies, this paper argues that the heretofore ways in which historians and philosophers of science have used the notion of “style” are severely restricted in terms of its mere applicability to the intellectual history of Western science. The particular example of the translation of “homosexuality” into Chinese during the May Fourth era reveals that the notion of “style” has the potential of carrying a much more dynamic conceptual weight, as when used in “styles of argumentation”. The paper also engages briefly with the historiography of scientific “national styles” and ends with some concluding remarks on the limitations of “social histories from below” and the under appreciated importance of “epistemological histories of possibilities”.  相似文献

Aspergillus flavus is a common filamentous fungus that produces aflatoxins and presents a major threat to agriculture and human health. Previous phylogenetic studies of A. flavus have shown that it consists of two subgroups, called groups I and II, and morphological studies indicated that it consists of two morphological groups based on sclerotium size, called “S” and “L.” The industrially important non-aflatoxin-producing fungus A. oryzae is nested within group I. Three different gene regions, including part of a gene involved in aflatoxin biosynthesis (omt12), were sequenced in 33 S and L strains of A. flavus collected from various regions around the world, along with three isolates of A. oryzae and two isolates of A. parasiticus that were used as outgroups. The production of B and G aflatoxins and cyclopiazonic acid was analyzed in the A. flavus isolates, and each isolate was identified as “S” or “L” based on sclerotium size. Phylogenetic analysis of all three genes confirmed the inference that group I and group II represent a deep divergence within A. flavus. Most group I strains produced B aflatoxins to some degree, and none produced G aflatoxins. Four of six group II strains produced both B and G aflatoxins. All group II isolates were of the “S” sclerotium phenotype, whereas group I strains consisted of both “S” and “L” isolates. Based on the omt12 gene region, phylogenetic structure in sclerotium phenotype and aflatoxin production was evident within group I. Some non-aflatoxin-producing isolates of group I had an omt12 allele that was identical to that found in isolates of A. oryzae.  相似文献

Testing German shepherd puppies to assess their chances of certification   总被引:1,自引:0,他引:1  
Behavioral activity of 7-week-old German shepherd puppies was tested and the activities analyzed if they could be used for predicting police efficiency of the individual. In total 206 individuals sired by 42 sires and 44 dams were used. The activities were divided into 10 tasks in which reactions and behavior of pups were scored from 0 to 5 points. All pups were tested separately from other conspecifics. Probability that the puppy will pass the certification was tested by a logistic regression. Of the 206 puppies, 148 passed the certification while 58 failed. Some tested behavioral variables were moderately to highly correlated with one another. Therefore we applied a factor analysis. Three factors were retained accounting for 100% of the shared variance. After inspection of the rotated factor pattern matrix and its confidence intervals, it appeared that variables “Independent movement and interactions with the tester”, “Negotiating obstacles”, “Entering a room”, “Behavior toward a person”, and “Behavior in new environments” loaded on Factor 1 (“Factor for movement”), while variables “Response to distracting stimuli caused by a shovel”, “Response to a distracting noise while left alone in a room”, and “Response to loud distracting stimuli” on Factor 2 (“Factor for responding to noise”) and variables “Retrieval” and “Tug of war” on Factor 3 (“Factor for attitude to predation”). In the final logistic regression model, the probability that the puppy will pass the certification depended on the higher weight at the time of testing (, P = 0.0005), on the “Factor for attitude to predation” (, P = 0.0007), on the “Factor for responding to noise”, where the higher the score, the weaker was the response (, P = 0.0232), and on the “Factor for movement” showing an increasing probability with decreasing score (, P = 0.0219). The tests in our study seem to be a good base which might enable selection for suitable dogs as early as 7 weeks of age. The puppies having high probability to pass certification in adulthood were heavy individuals willing to chase, catch, and fetch a tennis ball, and follow a rag drawn away from them, while weakly responding to a distracting noise in various situations and showing low activity while negotiating obstacles and moving and interacting with the tester. To conclude, contrary to skeptical assumptions expressed by various authors, the specific puppy tests for police dogs provide a reliable tool for predicting future service ability of a puppy. Differences in methodology are likely to contribute to a lack of consensus among various studies.  相似文献

This study focused on spatial relationships between female domestic goats and their kids from birth to 15 weeks of age. Results indicate that goat mothers are primarily responsible for the initiation and maintenance of mother-infant contact only during the first week following parturition. A shift to infant responsibility for mother-infant proximity was seen during Week 2, and kids remained primarily responsible throughout all subsequent weeks of observation. Mothers and their young spent most of their time more than 10 mother-lengths from each other during the course of the study; this lack of close physical proximity was particularly evident during the infant's first 6–8 weeks following birth. Thereafter, kids exhibited significant increases in the amount of time spent in close proximity to their dams. This general trend in the development of mother-infant spatial relationships in domestic goats is similar to those reported for other “hider” ungulates, and confirms that the domestic goat has a “hider” rather than a “follower” type of mother-infant relationship.  相似文献
