Synchronous cultures of Chlamydomonas reinhardii have been examined for the total amounts of carotenoid and chlorophyll present throughout a 12 hrs light–4 hrs dark life cycle. Variations in the carotenoid distribution at different points within the cell cycle have been found. During the greater part of the light period all major carotenoids increased at a proportionally similar rate. However, the increases in lutein and violaxanthin preceded those in β-carotene and neoxanthin by some 2 hrs and that in loroxanthin, an algal xanthophyll, by about 3 hrs. A marked drop in total carotenoid accumulation, corresponding to similar temporary falling away in the accumulation of β-carotene, lutein and violaxanthin occurred at 9 hrs. The correspondence of this with the established drop in RNA accumulation and the break-up of the nucleolus was pointed out. Considerable redistribution among the carotenoids occurred during the dark period, notably the amount of β-carotene increased relative to the total xanthophylls. The full significance of these results can not be estimated in the absence of comparative data on related organisms.  相似文献

The wild type (WT) of Scenedesmus obliquus and a mutant lacking chlorophyll b and the light-harvesting complexes (WT-LHC1) were synchronized by a light-dark regime. Both cultures contained the same type of carotenoids. However, concentrations and patterns of carotenoids were different during their synchronous life cycles. The concentration of total carotenoids followed more or less that of chlorophyll. The WT contained more carotenoids per cell mass, but slightly less per chlorophyll. It is discussed that part of the carotenoids of the mutant, lacking the peripheral antenna of PSII, might be located in the chlorophyll b-less apoprotein or in an enlarged core antenna of PSII. During the life cycle of Scenedesmus the carotenes are initially synthesized and most of the α-carotene is immediately oxidized to lutein which is inserted in the antennae systems of PSII and PSI. The further oxidation of lutein to loroxanthin seems to depend on both the change from dark to light, and on stages of the life cycle itself. Although the major part of β-carotene appears to be inserted in the reaction centers, a fraction of the total pool is rapidly converted to violaxanthin, following the onset of illumination. The conversion may serve to protect against photooxidation. Further conversion of violaxanthin to neoxanthin occurs to a greater extent in the mutant, WT-LHC1. The results demonstrate (1) the close connection between the carotenoid pattern and the modulation of the photosynthetic apparatus during the life cycle of Scenedesmus and (2) the flexibility of the organism in compensating for the absence of the light-harvesting complexes of photosystems II by adjusting the carotenoid distribution.  相似文献

In this work we characterize the changes induced by iron deficiency in the pigment composition of sugar beet (Beta vulgaris L.) leaves. When sugar beet plants were grown hydroponically under limited iron supply, neoxanthin and β-carotene decreased concomitantly with chlorophyll a, whereas lutein and the carotenoids within the xanthophyll cycle were less affected. Iron deficiency caused major increases in the lutein/chlorophyll a and xanthophyll cycle pigments/chlorophyll a molar ratios. Xanthophyll cycle carotenoids in Fe-deficient plants underwent epoxidations and de-epoxidations in response to ambient light conditions. In dark adapted Fe-deficient plants most of the xanthophyll cycle pigment pool was in the epoxidated form violaxanthin. We show, both by HPLC and by in vivo 505 nanometers absorbance changes, that in Fe deficient plants and in response to light, the de-epoxidated forms antheraxanthin and zeaxanthin were rapidly formed at the expense of violaxanthin. Several hours after returning to dark, the xanthophyll cycle was shifted again toward violaxanthin. The ratio of variable to maximum chlorophyll fluorescence from intact leaves was decreased by iron deficiency. However, in iron deficient leaves this ratio was little affected by light conditions which displace the xanthophyll cycle toward epoxidation or de-epoxidation. This suggests that the functioning of the xanthophyll cycle is not necessarily linked to protection against excess light input.  相似文献

The pigment composition of leaves from a number of different plant species collected from field sites in the region of Sheffield, UK, have been compared using high-performance liquid chromatography. Expression of pigment content per unit leaf area was dominated by variation in the total leaf chlorophyll. Neither chlorophyll per unit area nor the chlorophyll a/b ratio were found to be correlated with the habitat from which the plants originated. When the amounts of different carotenoids were expressed relative to the total carotenoid pool, it was found that whilst neither total carotene (α- +β-carotene) nor neoxanthin correlated with ability to grow in shade, the leaf content of both lutein and the total xanthophyll cycle carotenoids (zeaxanthin, anther-axanthin and violaxanthin) did, with lutein content being high in shade species and xanthophyll cycle intermediates low. There was a strong negative correlation between the relative amounts of each of these groups of carotenoids. The ratio of lutein to xanthophyll cycle carotenoids was strongly correlated to an index of shade tolerance.  相似文献

A new reversed phase-high performance liquid chromatography method has been developed to analyze the full complement of higher plant photosynthetic pigments (cis-neoxanthin, neoxanthin, violaxanthin, taraxanthin, anteraxanthin, lutein, zeaxanthin, cis-lutein, chlorophyll b, chlorophyll a, α- and β-carotene). The separation is carried out on a C18 column in about 10 minutes, using a single high-pressure pump and three different mobile phases in three isocratic steps. This method introduces a major improvement in higher plant photosynthetic pigment analysis, resolving in only 10 minutes all photosynthetic pigments while achieving good separation of lutein from its isomer zeaxanthin. Zeaxanthin is involved in the xanthophyll cycle, which recently has been proposed to play a significant role in the protection of the photosynthetic apparatus from photoinhibitory conditions (Demmig et al. [1987] Plant Physiol 84: 218-224).  相似文献

A short impulse of red light has a varying effect on carotenoid synthesis in dark-grown wheat seedlings. Except for β-carotene, which remains unchanged in the dark (it increases in continuous light), the carotenoid synthesis shows the same tendency as during constant irradiation. Thus, lutein and neoxanthin slowly increases, while violaxanthin decreases. During a period of constant light following various periods of darkness after the short impulse of light, the pigment changes correspond to those occurring in the dark, but are much more pronounced. The changes are discussed on the basis of phytochrome action.  相似文献

The pigment and quinone content of wild-type barley ( Hordeum vulgare L., cv. Svalöfs Bonus) and of two photosynthetic mutants was assayed. Wild type plants and the photosystem Hacking mutant viridis zb63 contained chlorophyll a and b. whereas chlorina-f2 contained only chlorophyll a The inability of the mutant chlorina-f2 to convert chlorophyll a into chlorophyll a appears to he the primary effect of the mutation. In both mutants, the carotenoid composition was virtually identical to that of the wild type. As compared to the wild type. chlorina-f2 contained less lutein and neoxanthin. The mutant viridis-zb63 contained less β-carotene but more antheraxanthin and xeaxanthin than the wild type. The quinone content and composition of the wild type and the photosynthetic mutants was similar, and both mutants biosynthesized plastid quinones and chromanols starting from [14C]-labeled tyrosine. The data indicate that carotenoid and quinone biosynthesis are not altered in the two mutants as compared to the wild type.  相似文献

The photosynthetic pigments of twigs in five tree and shrub species possessing chlorenchyma under a well developed, stomata-less, and highly photon absorptive periderm were analysed and compared to those of the corresponding canopy leaves. We asked whether the unavoidable shade acclimation of corticular chlorenchyma results in photosynthetic pigment complements typically found in shade leaves. As expected, chlorophyll (Chl) a/b ratios in twigs were consistently low. However, carotenoid (Car) analysis did not confirm the initial hypothesis, since twigs generally contained increased Chl-based pool sizes of the xanthophyll cycle components. The contents of photo-selective neoxanthin and lutein were high as well. Yet, β-carotene content was extraordinarily low. In addition, twigs retained high pre-dawn ratios of the deepoxidized antheraxanthin and zeaxanthin, although environmental conditions were not pre-disposing for such a state. The unexpected Car composition allows the conclusion that other micro-environmental conditions within twigs (hypoxia, increased red to blue photon ratios, and extremely high CO2 concentrations) are more important than shade in shaping the Car profiles.  相似文献

Plants protect themselves from excess absorbed light energy through thermal dissipation, which is measured as nonphotochemical quenching of chlorophyll fluorescence (NPQ). The major component of NPQ, qE, is induced by high transthylakoid ΔpH in excess light and depends on the xanthophyll cycle, in which violaxanthin and antheraxanthin are deepoxidized to form zeaxanthin. To investigate the xanthophyll dependence of qE, we identified suppressor of zeaxanthin-less1 (szl1) as a suppressor of the Arabidopsis thaliana npq1 mutant, which lacks zeaxanthin. szl1 npq1 plants have a partially restored qE but lack zeaxanthin and have low levels of violaxanthin, antheraxanthin, and neoxanthin. However, they accumulate more lutein and α-carotene than the wild type. szl1 contains a point mutation in the lycopene β-cyclase (LCYB) gene. Based on the pigment analysis, LCYB appears to be the major lycopene β-cyclase and is not involved in neoxanthin synthesis. The Lhcb4 (CP29) and Lhcb5 (CP26) protein levels are reduced by 50% in szl1 npq1 relative to the wild type, whereas other Lhcb proteins are present at wild-type levels. Analysis of carotenoid radical cation formation and leaf absorbance changes strongly suggest that the higher amount of lutein substitutes for zeaxanthin in qE, implying a direct role in qE, as well as a mechanism that is weakly sensitive to carotenoid structural properties.  相似文献

In addition to chlorophylls a and b, β-carotene, lutein, violaxanthin and neoxanthin, leaves of tobacco (Nicotiana tabacum L. cv. Virginia Gold) contain antheraxanthin in some harvests. In lower leaves, chlorophylls decreased more rapidly than carotenoids during senescence, but both types of pigment decreased at equal rates in upper leaves. The chlorophyll a:b ratio decreased only in post-mature leaves. Total carotenoid decreased with age, with the relative proportion of β-carotene increasing in lower leaves. Seasonal influences rather than age of leaf determines whether antheraxanthin is present. No esterified xanthophylls were found in senescent leaves.  相似文献

Among three species, Gossypium hirsutum, Rhizophora mangle, and Monstera deliciosa, which were transferred from low to high growth PFD, only small decreases in the efficiency of photochemical energy conversion were observed in those plants which exhibited an increase in photosynthetic capacity. Leaves of plants which showed no increase in photosynthetic capacity experienced a continuous decrease in photochemical efficiency, accompanied by a more pronounced loss of chlorophyll than that observed in the former group. In all species marked increases in the xanthophyll/β-carotene ratio resulted from small increases in lutein, and several-fold increases in the sum of the three components of the xanthophyll cycle, zeaxanthin, antheraxanthin, and violaxanthin. A strong increase in the level of zeaxanthin was only partially matched by a decrease of violaxanthin to zero, and was further paralleled by a decrease in β-carotene. Antiparallel changes in the sum of zeaxanthin + antheraxanthin + violaxanthin and β-carotene between morning and evening were observed in all species. These diel changes were overlaid on a net increase in β-carotene as well as total carotenoid content in those plants in which photosynthetic capacity increased. In those, however, which exhibited no photosynthetic acclimation upon transfer to high light, a decrease in both β-carotene and total carotenoid content was observed. Rhizophora mangle grown at 100 % seawater exhibited a particularly high capacity for increasing the level of zeaxanthin in response to high light.  相似文献

Abstract: We studied the biochemical composition and photosynthetic characteristics of the aerial parasite eastern dwarf mistletoe (Arceuthobium pusillum) and the effect of infection on the needles of host white spruce (Picea glauca) in a coastal forest stand in Maine, USA. Eastern dwarf mistletoe was capable of photosynthetic oxygen evolution; however, rates were low and were exceeded by respiratory oxygen consumption at all light intensities through full sunlight. Therefore, eastern dwarf mistletoe acts as a net sink for host photosynthate. Relative to those of uninfected trees, needles from infected branches of white spruce were significantly smaller in terms of length, fresh weight, maximum cross section and the diameter of the vascular cylinder. Needles of uninfected and infected trees did not differ in terms of fresh weight to dry weight ratio, nor in nitrogen, soluble sugar or starch content. Needles of infected trees possessed significantly less α-carotene and neoxanthin, but did not otherwise differ from uninfected needles in terms of chlorophyll and carotenoid composition. Since specific physiological roles for α-carotene and neoxanthin have not been described, the functional significance of the decreases in their content is not known. Photosynthetic capacities of needles from infected and uninfected white spruce did not differ significantly, as measured by oxygen evolution. These findings suggest that dwarf mistletoe infection does not substantially perturb host white spruce source-sink balance at the end of the growing season and that carbon exchange dynamics between the host and parasite are unlikely to fully explain the detrimental effects of infection on white spruce.  相似文献

The composition of coloured carotenoids in the milo shoot was investigated quantitatively (high performance liquid chromatography) during light-mediated plastidogenesis, including the time span of photodelay as caused by medium and high light fluxes. It was found that as long as only the far-red-absorbing form of phytochrome operates, the carotenoid pattern remains virtually the same as in complete darkness (violaxanthin and lutein as major constituents, traces of -carotene). On the other hand, the pattern changes dramatically in white or red light with increasing amounts of chlorophyll (lutein and -carotene dominate, -carotene showing the strongest relative increase). Photodelay during the early phase of plastidogenesis affects the carotenoid composition strongly. Increase of neoxanthin, violaxanthin and -carotene contents are diminished while lutein accumulation proves resistant towards chlorophyll-mediated photoinhibition. The photodelay can be diminished by an appropriate light pretreatment. Iron deficiency induced decreases in neoxanthin and β-carotene concomitantly with decreases in chlorophyll a, whereas lutein and carotenoids within the xanthophyll cycle were less affected. Iron deficiency caused major increases in the lutein/chlorophyll a and xanthophyll cycle pigments/chlorophyll a molar ratios. The chlorophyll a/chlorophyll b ratio increased in response to iron deficiency. The carotenoids within the xanthophyll cycle in iron-deficient and in iron-sufficient (control) leaves underwent epoxidations and de-epoxidations in response to ambient light conditions. In control leaves dark-adapted for several hours, most of the xanthophyll cycle pigment pool was in the epoxidated form vio-laxanthin, whereas iron-deficient leaves had significant amounts of zeaxanthin. Iron-deficient leaves also exhibited an increased non-photochemical quenching, supporting the possibility of a role for pigments within the xanthophyll cycle in photoprotection.  相似文献

There is a particularly high interest to derive carotenoids such as β-carotene and lutein from higher plants and algae for the global market. It is well known that β-carotene can be overproduced in the green microalga Dunaliella salina in response to stressful light conditions. However, little is known about the effects of light quality on carotenoid metabolism, e.g., narrow spectrum red light. In this study, we present UPLC-UV-MS data from D. salina consistent with the pathway proposed for carotenoid metabolism in the green microalga Chlamydomonas reinhardtii. We have studied the effect of red light-emitting diode (LED) lighting on growth rate and biomass yield and identified the optimal photon flux for D. salina growth. We found that the major carotenoids changed in parallel to the chlorophyll b content and that red light photon stress alone at high level was not capable of upregulating carotenoid accumulation presumably due to serious photodamage. We have found that combining red LED (75 %) with blue LED (25 %) allowed growth at a higher total photon flux. Additional blue light instead of red light led to increased β-carotene and lutein accumulation, and the application of long-term iterative stress (adaptive laboratory evolution) yielded strains of D. salina with increased accumulation of carotenoids under combined blue and red light.  相似文献

We studied seasonal changes in the content and ratio between photosynthetic pigments in one-yearold needles of Scotch pine (Pinus sylvestris L.) growing in Central Yakutia. Maximum accumulation of chlorophylls in developed young needles occurred in July when light and temperature conditions were favorable. In this period, the needles were notable for a relatively high level of β-carotene and neoxanthin and a reduced content of lutein and the pigments of violaxanthin cycle (VXC). In the course of autumn hardening, the content of chlorophylls decreased two times. Total content of carotenoids remained the same, but pigment composition considerably changed when plants progressed from a vegetating to frost-resistant state. We revealed time and temperature ranges of variation for individual carotenoids. In the beginning of hardening at reduced and low abovezero temperatures, the content of β-carotene in the needles decreased, the pigment-protein complexes (PPC) became enriched in lutein, the pigment pool of VXC gradually increased, and the content of neoxanthin transiently rose. When average daily air temperature further decreased to near- zero values, the content of zeaxanthin sharply rose. In winter, high levels of lutein and zeaxanthin were maintained. Main changes in pigment complex of the needles of P. sylvestris were completed before the coming of steady below-zero temperatures. The obtained data suggested that, upon seasonal decrease in temperature in early stages of hardening, a decrease in the level of chlorophyll promotes a reduction in the quantity of absorbed radiant energy. Apparently, this is accompanied by activation of the role of lutein and neoxanthin that perform specific photoprotective functions in antenna PPC associated with a gradual decrease in plants’ ability to quench singlet energy of excited chlorophyll. Accumulation of zeaxanthin as a result of inhibition of back reaction of epoxidation at near-zero temperatures creates necessary prerequisites for turning on the mechanisms of steady dissipation of absorbed light energy, which do not depend on transmembrane proton gradient of thylakoids. At the same time, zeaxanthin can perform antioxidant functions both in PPC and in the lipid phase of thylakoid membranes. The obtained data point to an adaptive nature of the observed reactions and a specific role of individual pigments in structural and functional reorganization of photosynthetic machinery in the course of development of frost-resistance in the needles.  相似文献

Carotenoids play critical roles in both light harvesting and energy dissipation for the protection of photosynthetic structures. However, limited research is available on the impact of irradiance on the production of secondary plant compounds, such as carotenoid pigments. Kale ( Brassica oleracea L.) and spinach ( Spinacia oleracea L.) are two leafy vegetables high in lutein and β-carotene carotenoids. The objectives of this study were to determine the effects of different irradiance levels on tissue biomass, elemental nutrient concentrations, and lutein β-carotene and chlorophyll (chl) pigment accumulation in the leaves of kale and spinach. 'Winterbor' kale and 'Melody' spinach were grown in nutrient solution culture in growth chambers at average irradiance levels of 125, 200, 335, 460, and 620 μmol m−2 s−1. Highest tissue lutein β-carotene and chls occurred at 335 μmol m−2 s−1 for kale, and 200 μmol m−2 s−1 for spinach. The accumulations of lutein and β-carotene were significantly different among irradiance levels for kale, but were not significantly different for spinach. However, lutein and β-carotene accumulation was significant for spinach when computed on a dry mass basis. Identifying effects of irradiance on carotenoid accumulation in kale and spinach is important information for growers producing these crops for dry capsule supplements and fresh markets.  相似文献

The carotenoids of the loquat fruit Eriobotrya japonica Golden Nugget variety, were investigated. They were identified according to their chromatographic, spectrophotometric and chemical properties and compared with standard pigments. For some of the carotenoids, MS were determined. Pulp and peels were investigated separately. The main pattern of the pulp carotenoids was β-carotene (33%), γ-carotene (6%), cryptoxanthin (22%), lutein, violaxanthin and neoxanthin, each about 3–4%. The peel, with a carotenoid content 5 times as high, had a similar pattern, but the ratio between the main pigments differed: β-carotene (50%); γ-carotene (5%); cryptoxanthin (5%); lutein (13%); violaxanthin, neoxanthin, 3–4%. The carotenoids of the loquat (subfamily Maloideae) were very similar to those of the apricot (Prunus armeniaca-subfamily Prunoideae) both of the family of Rosaceae. The intergeneric differences are more pronounced, which is of possible taxonomic significance. The lower concentration of cryptoxanthin and the high concentration of lutein in the peels is noteworthy and of biosynthetic interest.  相似文献

Thylakoid membranes from spinach were fragmented mechanically and separated into vesicles originating from grana and stroma-exposed lamellae (Andreasson et al. (1988) Biochim Biophys Acta 936: 339–350). The grana vesicles were further fragmented and separated into smaller vesicles originating from different parts of the grana (Svensson and Albertsson (1989) Photosynth Res 20: 249–259). All vesicles so obtained were analyzed with respect to chlorophyll and carotenoid composition by reverse phase HPLC. For all fractions the following relations (mole/mole) were found: 1 carotenoid per 4 chlorophyll (a+b), 2 lutein per 5 chlorophyll b and 5 violaxanthin per 100 chlorophyll (a + b). The contents of lutein and neoxanthin were each linearly related to chlorophyll b and -carotene was linearly related to chlorophyll a.  相似文献
