Induction of resistance to crown rust, caused by Puccinia coronata Cda. var, avenae Frazer et Led., occurred in seedlings of three genetic lines of oats (Avena sativa L.) inoculated sequentially with one of three inducer-challenger combinations of the pathogen. There was significant reduction in both the number of pustules per leaf and the weight of urediospores harvested in all three host genotypes tested. There were highly significant differences between individual spore harvests correlated with age of the pustules. Development of telia by the challenger was found to occur considerably later, and was less intense, when the challenger was inoculated alone in contrast to treatments in which the challenger followed pre-inoculation with the inducer. There was no correlation between the numbers of resistant and susceptible lesions within the range of inoculum density used in these experiments.  相似文献

Under restrictive vegetative conditions, cells of cell-division cycle (cdc) temperature-sensitive mutants arrest at specific points in the cycle. Meiotic and mitotic behaviour of such arrested cells was examined under permissive sporulation conditions. Those mutants which were committed to mitosis at their specific point of arrest finished the cell cycle and could only then go into meiosis. It was found that commitment to mitosis occurred early in the cell cycle, prior to DNA replication, and that this commitment was dependent upon the gene function of cdc4.  相似文献

Inactivation of liver cytosol proteins has been measured in vitro in the presence of various membranes and disulphides. Inactivation rates correlate with the known degradation rate constants of the enzymes in the intact liver. More extensive studies were carried out with glucose-6-phosphate dehydrogenase (G6PD) and phosphoenolpyruvate carboxykinase (PEPCK) using either cytosol as a source of these enzymes or alternatively highly purified preparations of each enzyme. All membranes purified from liver had a considerable capacity to inactivate the enzymes with higher activity found in the hepatocyte plasma membrane. Various lipid preparations or plasma membranes from other tissues were virtually ineffective. Inactivation was dependent on disulphides in the membranes as shown by the inhibition of activity if membranes were pretreated with thiols. Preliminary experiments of the fate of inactivated G6PD or PEPCK show binding to membranes and subsequent proteolysis. A model is proposed for the degradation of labile enzymes.  相似文献

During the course of radical oxidation, cholesterol may exert seemingly contradictory effects. In order to gain a better understanding of the relationship between cholesterol levels and membrane susceptibility to oxidative damage induced by reactive oxygen species (ROS), here we analyze the integrity and structural stability of cholesterol-modified (enriched or depleted) and unmodified (control) erythrocytes exposed to tert-butyl hydroperoxide. The oxidant significantly increased ROS production, with almost complete oxidation of hemoglobin and a reduction in GSH content in the different erythrocyte groups at 2 mM concentration. These changes were accompanied by losses of cholesterol and total phospholipids, the main decreases being in phosphatidylethanolamine and phosphatidylcholine. The highest lipid loss was found in the cholesterol-depleted group. Fatty acid analyses revealed changes only in peroxidized cholesterol-modified erythrocytes, with decreases in linoleic and arachidonic acids. Fluorescence anisotropy studies showed an increase in the fluidity of the negatively charged surface of peroxidized control erythrocytes. Increased hemolysis and a positive correlation between cellular osmotic fragility and malondialdehyde contents were found in all peroxidized groups. These findings provide evidence that the modification of cholesterol levels in the erythrocyte membrane has provoking effects on peroxidation, with corresponding increases in oxidative damage in the treated cell, possibly as a consequence of lipid bilayer destabilization.  相似文献

The heterogeneous responses of clonal cancer cells to treatment is understood to be caused by several factors, including stochasticity, cell-cycle dynamics, and different micro-environments. In a tumor, cancer cells may encounter fluctuating conditions and transit from a stationary culture to a proliferating state, for example this may occur following treatment. Here, we undertake a quantitative evaluation of the response of single cancerous lymphoblasts (L1210 cells) to various treatments administered during this transition. Additionally, we developed an experimental system, a “Mammalian Mother Machine,” that tracks the fate of thousands of mammalian cells over several generations under transient exposure to chemotherapeutic drugs. Using our developed system, we were able to follow the same cell under repeated treatments and continuously track many generations. We found that the dynamics of the transition between stationary and proliferative states are highly variable and affect the response to drug treatment. Using cell-cycle markers, we were able to isolate a subpopulation of persister cells with distinctly higher than average survival probability. The higher survival rate encountered with cell-cycle phase specific drugs was associated with a significantly longer time-till-division, and was reduced by a non cell-cycle specific drug. Our results suggest that the variability of transition times from the stationary to the proliferating state may be an obstacle hampering the effectiveness of drugs and should be taken into account when designing treatment regimens.  相似文献

Changes in phenology are among the most pervasive effects of current climate change. Modifications in the timing of life-cycle events can affect the behavior, physiology and life-history of wildlife. However, organisms can develop compensatory strategies in order to reduce the costs of phenological alterations. Here, we examine the extent and limits of compensatory developmental responses in amphibian larvae exposed to variation in hatching timing. Using a common-garden experiment, we analyze how changes in temperature and food conditions alter compensatory responses to hatching delay, paying particular attention at how adverse environmental conditions can constrain these responses. We found that under benign conditions (warm temperature, unrestricted food) larvae fully compensate for the hatching delay, without cost in mass. However, under detrimental conditions (cold temperature and restricted food) these responses were prevented, and the combination of adverse conditions with long hatching delay completely disrupt compensatory responses. This study highlights the need of examining ecological responses to climate variation across a broad spectrum of environmental conditions in order to accurately predict the putative effect that climatic alterations can have on the life-histories and survival of wildlife.  相似文献

The expected increase in drought severity and frequency as a result of anthropogenic climate change leads to concerns about the ability of native tree species to cope with these changes. To determine the susceptibility of Fagus sylvatica (European beech) and Quercus robur (pedunculate oak) – the two dominant deciduous tree species in Central Europe – to drought, we quantified the climate sensitivity and drought-response of radial growth for both species using an array of dendroecological methods. Tree-ring data were collected from a site east of Coburg, Bavaria which had shown pronounced stress-symptoms (early leaf coloration) during the record drought of 2018. Climate-growth relationships were used to establish the sensitivity of radial growth to multiple climatic variables. The impact of specific drought events on tree growth was quantified using tolerance indices. In addition, we employed a Principal Component Gradient Analysis (PCGA) and remote sensing data (MODIS Normalized Difference Vegetation Index (NDVI)) to delineate the species specific drought responses. Using these methods we were able to show a clear difference in drought susceptibility between beech and oak. Beech displayed a higher sensitivity to temperature and the standardized precipitation evapotranspiration index (SPEI) and showed lower resistance and resilience to drought events than oak. In particular, beech was unable to fully recover from the 2003 drought, after which it expressed a stark growth decline, i.e. drought legacies, which was not observed for oak. The PCGA revealed a clear differentiation in the grouping of drought responses between beech and oak, supporting the findings of the climate-growth analysis and the tolerance indices. Correlations of NDVI and ring-width indices (RWI) indicated that under normal climatic conditions NDVI variability is linked to the start of the growing season. This is in contrast to drought years, such as 2003, where summer NDVI mirrored the drought response of beech and oak. These results reveal beech to have both a higher sensitivity to summer temperature and SPEI and a higher susceptibility to drought events. Although, in the past high plasticity and adaptability to drought have been attributed to both beech and oak, our study assigns beech a higher risk than oak to suffer from anticipated increases in drought frequency and intensity as a consequence of climate change.  相似文献

In the last few years, a variety of self-assembling short peptides that consist exclusively of simple amino acids have been designed and modified. These peptides exhibit self-assembling dynamic behaviors. At the molecular structural level, they form α-helical, β-sheet and β-hairpins structures in water. These structures further undergo spontaneous assembly to form nanofibers which aggregate into supramolecular scaffolds that entrap large volumes of water. Furthermore, nanostructures and supramolecular structures that self-organized from these short peptides also have a broad spectrum of biotechnological applications. They are useful as biological materials for 2D and 3D tissue cell cultures, regenerative and reparative medicine, tissue engineering as well as injectable drug delivery matrices that gel in situ. We have endeavored to do a comprehensive review of short peptides that form nanofibrous hydrogels. In particular, we have focused on recent advances in peptide assembly motifs and applications.  相似文献

The effects of a 60 min exposure to photosynthetic photon flux densities ranging from 300 to 2200 mol m–2s–1 on the photosynthetic light response curve and on PS II heterogeneity as reflected in chlorophyll a fluorescence were investigated using the unicellular green alga Chlamydomonas reinhardtii. It was established that exposure to high light acts at three different regulatory or inhibitory levels; 1) regulation occurs from 300 to 780 mol m–2s–1 where total amount of PS II centers and the shape of the light response curve is not significantly changed, 2) a first photoinhibitory range above 780 up to 1600 mol m–2s–1 where a progressive inhibition of the quantum yield and the rate of bending (convexity) of the light response curve can be related to the loss of QB-reducing centers and 3) a second photoinhibitory range above 1600 mol m–2s–1 where the rate of light saturated photosynthesis also decreases and convexity reaches zero. 以结球甘蓝高抗TuMV自交不亲和系84075为材料,构建了cDNA文库。根据抗病基因保守序列(NBS-LRR)设计一对简并引物,以84075的基因组DNA和cDNA为模板,进行PCR扩增,分别扩增出一条513bp片段,扩增片段进行克隆测序。选取两个与抗病基因同源性较高的克隆片段作探针(命名Borl,Bor2),对构建的cDNA文库进行筛选,得到一个阳性克隆(命名TuR2),测序及序列分析表明,该基因总长为762bp,编码226个氨基酸、包含681bp的开放阅读框。与已克隆的抗病基因有不同程度的同源性。利用TuR2作探针,进行了Southern杂交、Northern杂交以及抗病性的共分离检测分析。结果表明,TuR2可能吧单拷贝形式存在,其表达是组成成型的,且无组织特异性;初步确定是一个与结球甘蓝抗TuMV相关的基因。  相似文献
