2017年7-8月和11-12月在贺兰山岩画风景区采集到369份岩羊新鲜粪便样本,用多态性较高的10个微卫星位点进行基因分型,共识别出282只岩羊个体。通过复合扩増和牙釉蛋白基因PCR法共鉴定出152只雄性,130只雌性。微卫星分析得到平均等位基因(Na)为10.1,平均观测杂合度(Ho)为0.7737,平均期望杂合度(He)为0.6512,平均多态信息(PIC)为0.6129。运用Cervus3.0软件进行亲权鉴定,并用Kingroup V2计算个体间的亲缘系数(r),作为亲权鉴定的补充,共鉴定出父-母-子7对,父-子21对,母-子19对。根据亲缘关系对岩羊的婚配制进行推测分析,表明岩羊婚配制应属一雄多雌。  相似文献

近年来,由于人类活动的干扰使黑麂的栖息地明显减少并呈斑块状分布.因此,深入了解该物种各地理分布种群的遗传现状是制定保护及管理策略的重要科学依据.本研究运用非损伤取样技术在浙江省九龙山国家级自然保护区的黑麂分布区共采集61份粪便样品、2份肌肉样品和2份皮张样品,采用8个特异微卫星位点对61份粪便样品中提取的DNA进行检测,肌肉和皮张样品DNA的检测结果作为对照,分析了九龙山国家级自然保护区黑麂种群的遗传多样性.经多次重复提取和PCR扩增后,发现61份粪便样本中有38份可被稳定扩增,且不同样本间的条带大小有较大差异,因此我们认为38份粪便样本来源于38只不同的黑麂个体.8个微卫星位点共检测到50个等位基因,平均等位基因数(A)为6(5~8);平均有效等位基因数(Ne)为5.297 (4.031 ~6.353);平均期望杂合度(He)为0.817 (0.763 ~0.855);平均多态信息含量(PIC)为0.775 (0.709 ~0.822).8个位点的平均个体识别率(DP)为0.931,累积个体识别率(CDP)为0.999 9.有1个微卫星位点(BM1706)极显著地偏离了Hardy-Weinberg平衡(P<0.01).以上结果显示该分布区黑麂种群具有较高的遗传多样性.  相似文献

东平湖麦穗鱼群体遗传结构的微卫星标记分析   总被引:5,自引:0,他引:5  
利用微卫星标记技术,采用26对鲤微卫星引物对山东东平湖麦穗鱼进行全基因组扫描.结果表明,有13对引物能获得稳定的特异性条带(占总数的50%),其中有6个微卫星位点具有多态性(占总数的23.1%).6个多态位点共检测到22个等位基因,每个位点的等位基因数从2个到7个不等,大小在80~406bp之间;平均多态信息含量(PIC)为0.5115,平均观测杂合度(H0)为0.6812,平均期望杂合度(HE)为0.5775.研究结果表明,东平湖麦穗鱼群体遗传多样性较丰富,种群结构合理,种质资源处于安全状态.  相似文献

菲律宾蛤仔EST_SSR标记与生长性状的相关分析   总被引:1,自引:0,他引:1  
研究利用20个微卫星标记对菲律宾蛤仔斑马蛤F2代家系107个个体进行遗传多样性分析,并对标记位点与生长相关性状进行分析。在20个微卫星位点共检测到41个等位基因,各位点等位基因数为2—3个,等位基因片段大小为109—430 bp,平均等位基因数为2.05个。平均有效等位基因数为1.71个,观测杂合度平均值为0.504,期望杂合度的平均值为0.431,平均多态信息含量为0.324。经卡方检验,3个位点SSR11,SSR164和SSR213的基因型分布显著偏离了孟德尔定律(P0.01)。运用SPSS 20.0对20个微卫星位点与菲律宾蛤仔斑马蛤家系生长性状的相关性(壳长、壳宽、壳高和体重)进行连锁显著性检验。结果表明,SSR9位点与壳高存在显著的相关关系(P0.05),SSR135和SSR164位点与壳宽呈显著相关(P0.05),SSR142位点与体重呈显著性相关(P0.05)。研究结果可为菲律宾蛤仔的分子标记辅助选育提供参考。  相似文献

选择12对微卫星标记检测了于2011年采集自元江(红河上游中国江段)5个样点192尾鲤的群体遗传多样性.共检测到201个等位基因,每个位点等位基因2-27个.各群体各位点平均等位基因(NA)12.25-14.67个,平均有效等位基因(NE)8.28-9.73个,平均观察杂合度(Ho)o.7765-0.8037,平均期望杂合度(HE)0.7761-0.8080,平均多态信息含量(PIC)0.7534-0.7843.元江鲤种群192个个体各位点NA、NE、Ho、HE、PIC分别为16.50、11.26、0.7927、0.8049、0.7966,种群遗传多样性水平高.元江鲤群体之间遗传分化小,可作为一个种群管理单元进行管理.增殖放流要防止遗传多样性丧失.  相似文献

选用35个多态性微卫星分子标记对天津换新良种场镜鲤一个繁殖群体的有效等位基因数(Ae)、观测杂合度(Ho)、期望杂合度(He)、多态信息含量(PIC) 等进行了检测, 以卡方检验估计群体Hardy-Weinberg平衡。结果表明:在35个基因座共检测到118个等位基因, 平均等位基因数为3.37个, 每个座位检测到的等位基因数2~7个不等, 平均有效等位基因数为2.16, 观测杂合度平均值0.431, 无偏期望杂合度的平均值为0.4736, 平均多态信息含量0.42, 说明这个群体属于中度多态, 遗传多样性水平不高。卡方检验的P值显示多于半数的位点都发生了偏离。并将35个基因座的不同基因型与个体的体重、体长值进行了连锁分析, 得到了4个与体重、体长连锁的基因型, 并将所得结果与鲤鱼体长性状QTL定位结果进行对比, 其中HLJ319标记与QTL定位结果基本一致。分析了几个严重偏离平衡的基因型, 并讨论出现这种现象的可能原因。  相似文献

目的开发诸氏鲻虾虎鱼(Mugilogobius chulae)多态性微卫星标记,为诸氏鲻虾虎鱼的遗传背景评价提供基础数据。方法采用磁珠富集法,以生物素标记的(AC)15为探针,构建了诸氏鲻虾虎鱼微卫星富集文库。经克隆、筛选、测序后,设计合成微卫星引物,利用野生群体对微卫星引物进行多态性筛选和评价。结果共获得74个含有微卫星重复单元的序列,根据微卫星序列共设计出33对微卫星引物。利用30个野生诸氏鲻虾虎鱼样本对33对引物的微卫星位点进行了评价,其中有12个位点表现出多态性,平均有效等位基因数(Ne)、观测杂合度(Ho)、期望杂合度(He)及多态信息含量(PIC)分别为2.9167、2.2311、0.4583、0.5633和0.4474。结论所筛选的微卫星标记将为诸氏鲻虾虎鱼遗传质量控制及监测提供实验依据。  相似文献

梅花鹿3个种群遗传多样性的微卫星标记分析   总被引:1,自引:0,他引:1  
利用16个微卫星标记对黑龙江省部分地区(兴凯湖农场、大庆市银浪牧场、五大连池大庆农场鹿苑)的3个梅花鹿(Cervus nippon)群体进行了遗传多样性检测.统计了3个鹿群的等位基因组成、平均有效等位基因数(Ne)、平均遗传杂合度(h)和多态信息含量(PIC).结果表明,除5个位点外,其余11个微卫星位点均表现出不同的多态信息含量,其中高度多态位点5个,中度多态位点4个.这说明本研究所选用的微卫星位点可较准确地评估3个梅花鹿群体的遗传多样性,并为今后相关研究筛选出了有价值的引物.3个梅花鹿群体的平均h在0.454~0.636之间变动,其中兴凯湖梅花鹿群体最高,为0.636,具有较大的遗传潜力.  相似文献

达氏鲟(Acipenser dabryanus)是我国特有的珍稀鱼类,主要分布在长江上游干支流。由于人类活动的影响,其种群数量急剧下降,被列为国家Ⅰ级重点保护动物、IUCN红色名录极危(CR)物种。本研究针对当前达氏鲟群体的濒危现状,应用微卫星标记开展达氏鲟保护遗传学研究。通过高通量测序成功筛选出25个微卫星位点,采用18尾野生个体和30尾人工繁殖个体对其多样性进行了检验,结果表明,25个位点共检测出181个等位基因,每个位点的等位基因数(A)为4~11(平均值7.2),观测杂合度(HO)为0.160~1.000(平均值0.744),期望杂合度(HE)为0.346~0.875(平均值0.727)。所有位点均不偏离"Hardy-Weinberg"平衡(P0.05),各位点间也无连锁不平衡现象。除了其中一个位点以外,所有位点的多态信息含量(PIC)均大于0.5,其多态性较好。本次筛选的微卫星位点将有助于达氏鲟资源保护和群体遗传研究。  相似文献

本研究对厚唇裸重唇鱼(Gymnodiptychus pachycheilus)基因组进行Illumina HiSeq高通量测序,共获得85 905条序列,包含567 200个微卫星位点,从中筛选出15个微卫星位点,采用雅砻江新龙种群和黄河渭河种群对其多态性进行了验证。新龙种群中,15个位点的平均等位基因数为6.9(3 ~ 13),观测杂合度(Ho)为0.712 4,多态信息含量(PIC)为0.630 3,有8个位点显著偏离哈迪-温伯格平衡,1对位点表现出连锁。渭河种群中,15个位点的平均等位基因数为8.4(4 ~ 16),观测杂合度(Ho)为0.719 5,多态信息含量(PIC)为0.680 7,有7个位点显著偏离了哈迪-温伯格平衡,4对位点表现出连锁。筛选获得的15个微卫星位点多态性较高,适合用于厚唇裸重唇鱼种群遗传学研究。  相似文献

The black muntjac (Muntiacus crinifrons), endemic to China, was categorized as a Grade I National Key Protected Animal by the Ministry of Agriculture of China and classified as Vulnerable (VU) by IUCN. Recent years, studies had been conducted on this species mainly focusing on habitat selection, food habit, gene flow etc., only with a few reports on the population dynamics. Individual identification of wild animals is one of the most important subjects in the population dynamic research. Of various molecular markers, microsatellite DNA fingerprinting has been used most frequently and successfully on many kinds of animals. Here, we constructed identification system for the black muntjac using 8 microsatellite loci. 31 black muntjacs were identified from 141 fecal samples, whereas 43 samples could be used for PCR after repeated trials. Further, the sequencing for Cytb gene was also conducted for convincing us the identity of fecal samples. The results, highly consistent between sequencing consequence and sequence data from Genebank, implied that those experienced local people are of the convincing knowledge about wild animals, especially at the respects of identification to black muntjac’ feces pellets. Moreover, we detected the specificity of identification system to black muntjac. BM1225 was the only one locus that unsuccessful PCR for the muscle samples of Muntiacus reevesi was observed, which suggested that our identification system could be used for excluding the non-researched objects in some cases. Analyses using softwares CERVUS 3.0 and POPGENE 1.21 showed that the present identification system had strong discrimination power: 0.938 per loci (DP) or 0.999 in total (CDP). The mean observed number of alleles (Na), mean effective number of alleles (Ne), and mean excepted heterogosity (He) were 8.875, 6.375, and 0.829 respectively. Considering that the change of sex ratio in population could exert significant impact on population growth, density to some extent, we also analyzed the sex ratio of those individuals that had been identified based on fecal samples from filed using SRY (Sex-determining region Y) gene amplification, which identified 19 males and 12 females.  相似文献

The Indian muntjac (Muntiacus muntjak vaginalis) has a karyotype of 2n = 6 in the female and 2n = 7 in the male. The karyotypic evolution of Indian muntjac via extensive tandem fusions and several centric fusions are well documented by molecular cytogenetic studies mainly utilizing chromosome paints. To achieve higher resolution mapping, a set of 42 different genomic clones coding for 37 genes and the nucleolar organizer region were used to examine homologies between the cattle (2n = 60), human (2n = 46), Indian muntjac (2n = 6/7) and Chinese muntjac (2n = 46) karyotypes. These genomic clones were mapped by fluorescence in situ hybridization (FISH). Localization of genes on all three pairs of M. m. vaginalis chromosomes and on the acrocentric chromosomes of M. reevesi allowed not only the analysis of the evolution of syntenic regions within the muntjac genus but also allowed a broader comparison of synteny with more distantly related species, such as cattle and human, to shed more light onto the evolving genome organization. For convenience and to avoid confusion we added for each species a three letter abbreviation prior to the chromosomal band location discussed in this paper: BTA, Cattle chromosome; HSA, Human chromosome; MMV, M. m. vaginalis chromosome; MRE, M. reevesi chromosome.  相似文献

Similar to many other pinniped species, harbour seals ( Phoca vitulina ) mate exclusively at sea. Here we present the first attempt to measure male mating success in an aquatically mating pinniped. Male mating success was estimated by paternity analysis in two cohorts of pups born at Sable Island, Nova Scotia, Canada, using microsatellite DNA markers. The genotypes of 275 pups born in 1994 and 1995 were compared to those of 90 candidate males at six microsatellite loci using a likelihood approach to resolve paternity. Paternity could be assigned for two, 22, 40 and 85 pups at confidence levels of 95, 80, 65 and 50%, respectively. Most successful males were assigned the paternity of a single offspring, suggesting a low variance in male mating success relative to most pinniped species. The proportion of paternal half sibs within cohorts and between maternally related sibs estimated by maximum likelihood were not significantly different from zero. It is thus unlikely that most offspring were sired by a small number of highly successful unsampled males, and that female harbour seals do not usually exhibit fidelity to the same male in sequential breeding seasons. A low level of polygyny in Sable Island harbour seals is consistent with predictions based on their breeding ecology, as females are highly mobile and widely dispersed in the aquatic mating environment at Sable Island.  相似文献

Harbour seals sometimes breed along inland travel corridors where females become clustered in space and time and males establish underwater acoustic display territories similar to terrestrial arenas known as resource-based leks. Under these conditions, we predicted that higher levels of polygyny would be observed than has been previously reported for this species mating in open coast environments without travel corridors. Reproductive success (RS) of 70 males was measured using 20 microsatellite DNA loci and likelihood-based paternity analysis of 136 offspring collected over 3 years. Most males were assigned either zero or one paternity with 80% confidence. The greatest number of pups assigned to one male in a season was two. Variance in RS was higher for males than females (which are biologically limited to one offspring per year) indicating low to mild polygyny. In addition, distributions of relatedness values among pups within year classes did not differ significantly from a simulated distribution with R = 0, indicating that half-siblings were uncommon. Overall, polygyny levels were low relative to terrestrial pinniped mating systems and similar to observations from a harbour seal population along an open coast. Due to large confidence intervals associated with our results, we cannot rule out the hypothesis that a travel corridor might increase the degree of polygyny skew relative to that observed in open coast environments. Habitat appeared to influence male strategies as the most successful males in open coast environments patrolled offshore, while the most successful male in this study defended a territory along the travel corridor.  相似文献

We studied patterns of genetic relatedness and paternity in moustached tamarins, small Neotropical primates living in groups of 1–4 adult males and 1–4 adult females. Generally only one female per group breeds, mating with more than one male. Twin birth are the norm. In order to examine the genetic consequences of this mating pattern, DNA was extracted from fecal samples collected from two principal and six neighboring groups. DNA was characterized at twelve microsatellite loci (average: seven alleles/locus). We addressed the following questions: Do all adult males have mating access to the reproductive female of the group? How is paternity distributed across males in a group? Can polyandrous mating lead to multiple paternity? Are nonparental animals more closely related to the breeders than to the population mean? And, are mating partners unrelated? Breeding females mated with all nonrelated males. In at least one group the father of the older offspring did not sire the youngest infant although he was still resident in the group. We also found evidence for multiple paternity in a supposed twin pair. Yet, within each group the majority (67–100%) of infants had the same father, suggesting reproductive skew. Relatedness within groups was generally high (average R = 0.31), although both nonrelated males and females occurred, i.e., immigrations of both sexes are possible. Mating partners were never found to be related, hence inbreeding seems to be uncommon. The chromosomes of the Indian muntjac (Muntiacus muntjak vaginalis) are unique among mammals due to their low diploid number (2N=6, 7) and large size. It has been proposed that the karyotype of this small Asiatic deer evolved from a related deer the Chinese muntjac (Muntiacus reevesi) with a diploid chromosome number of 2n= 46 consisting of small telocentric chromosomes. In this study we utilized a kinetochore-specific antiserum derived from human patients with the autoimmune disease scleroderma CREST as an immunofluorescent probe to examine kinetochores of the two muntjac species. Since CREST antiserum binds to kinetochores of mitotic chromosomes as well as prekinetochores in interphase nuclei, it was possible to identify and compare kinetochore morphology throughout the cell cycle. Our observations indicated that the kinetochores of the Indian muntjac are composed of a linear beadlike array of smaller subunits that become revealed during interphase. The kinetochores of the Chinese muntjac consisted of minute fluorescent dots located at the tips of the 46 telocentric chromosomes. During interphase, however, the kinetochores of the Chinese muntjac clustered into small aggregates reminiscent of the beadlike arrays seen in the Indian muntjac. Morphometric measurements of fluorescence indicated an equivalent amount of stained material in the two species. Our observations indicate that the kinetochores of the Indian muntjac are compound structures composed of linear arrays of smaller units the size of the individual kinetochores seen on metaphase chromosomes of the Chinese muntjac. Our study supports the notion that the kinetochores of the Indian muntjac evolved by linear fusion of unit kinetochores of the Chinese muntjac. Moreover, it is concluded that the evolution of compound kinetochores may have been facilitated by the nonrandom aggregation of interphase kinetochores in the nuclei of the ancestral species.  相似文献

The short-nosed fruit bat Cynopterus sphinx is known to exhibit resource defence polygyny as its primary mating strategy. Tent construction by harem males to recruit females represents a heavy investment of time and effort, which is not done by nonharem males. The previously unobserved mode of harem formation by the solitary males was studied using mark-recapture and radio-telemetry. In our observation, the solitary males roosting near to harems started recruiting females by occupying the tent abandoned by the harems. This result suggests that the transition of nonharem male to harem male status possibly by a previously unobserved mode and the female recruitment is associated with resource (roost). It implies that the solitary males are actively involving in female recruitment and also presumably mating.  相似文献
