新疆干旱区植物藜的种子异型性及其萌发机理   总被引:2,自引:0,他引:2  
新疆干旱区分布的植物藜(Chenopodium album)的种子有黑色和褐色两种类型。对藜的异型性种子从形态结构、不同环境因素及激素或化学物质对萌发的影响以及同工酶谱等方面进行了研究,并对其萌发及适应异质环境的机理进行了讨论。结果表明:(1)藜的异型性种子在形态结构、萌发休眠特性等方面都存在明显差异:黑色种子种皮厚且硬,休眠,萌发慢,萌发率低;褐色种子种皮薄而软,不休眠,萌发快且萌发率高;(2)黑色种子的休眠可通过切除胚根外缘种皮得以完全解除;(3)赤霉素、乙烯利对黑色种子的萌发无明显促进作用;KNO3可较显著促进黑色种子的萌发;协同使用乙烯利和KNO3时,可显著提高黑种子萌发率,完全打破休眠;(4)黑色种子和褐色种子的酯酶、过氧化物酶及过氧化氢酶同工酶谱带存在差异;(5)黑色种子的萌发需要光照,而褐色种子则对光不敏感;低温贮藏对二者的萌发均无显著影响,尽管黑色种子的萌发率有波动。研究结果初步显示黑色种子的休眠是内源(胚)和外源(种皮)因素共同所致。藜的种子异型性及其萌发机理的形成是其对新疆干旱区异质化环境的高度适应。  相似文献

角果碱蓬(Suaeda corniculata)是藜科一年生盐生植物, 在我国分布于北方盐碱滩涂和盐碱荒漠地区。角果碱蓬具有棕色和黑色两种异型体种子(简称棕色和黑色种子)。对采自内蒙古鄂托克前旗盐渍化生境的角果碱蓬二型种子的形态、休眠和萌发特性开展对比研究, 测定了二型种子休眠和萌发行为对温度、光照和盐分(NaCl)的响应, 以揭示盐生植物异型种子对温带盐漠生境的适应对策。结果表明: (1)二型性种子在大小、种皮特性和结实比例方面有显著差异。与黑色种子相比, 棕色种子个体较大, 种皮透水性强。黑色种子与棕色种子的结实比例约为5.6 : 1。(2)新成熟的棕色种子的萌发对各温度梯度和光照条件不敏感, 萌发率较高(84%-100%); 而新成熟的黑色种子萌发率较低(8%-78%), 萌发对光照敏感。(3)黑色种子具有浅度生理休眠, 种皮划破、赤霉素处理和低温层积均可有效地提高种子的萌发率。(4)二型种子萌发对土壤盐分的胁迫具有不同的响应。与黑色种子相比, 棕色种子对盐分胁迫不敏感, 在较高的盐分浓度下仍有较高的萌发率, 低温层积处理能够降低黑色种子对盐胁迫的敏感性, 有效地提高种子的初始萌发率、萌发恢复率和最终萌发率。角果碱蓬二型种子不同的形态、休眠和萌发特性, 提高了该物种在高度异质性生境中的适合度, 对种群成功地适应温带盐漠环境具有重要的意义。  相似文献

以两种菊属野生植物'菊花脑'(Dendranthema nankingense)和'乙立寒菊'(D.indicum var.maruyamanum)幼苗为材料,通过Hogland营养液水培试验研究了120和180 mmol·L NaCl的渐进和非渐进胁迫处理对幼苗叶片受害程度及植株生物量、根系活力、叶片叶绿素含量、相对电导率和丙二醛含量的影响,以探讨它们对盐冲击的生理响应规律.结果显示:(1)在两种浓度盐胁迫下,两种菊属植物的整株鲜重、根系活力、叶片叶绿素含量大多显著下降,而同期的叶片丙二醛(MDA)含量、相对电导率显著增加;渐进盐胁迫下材料受害程度均轻于非渐进胁迫;盐胁迫浓度越大,各指标变化幅度越大,且两种胁迫方式下各指标的差异亦越大;(2)'菊花脑'对盐胁迫敏感,而'乙立寒菊'对盐胁迫的耐性较强;相同盐胁迫浓度下,非渐进胁迫与渐进胁迫间的差异在盐敏感材料上表现较明显.研究表明,两种菊属植物均存在盐冲击现象,并以耐盐性弱的材料表现更加突出,且高盐胁迫下植物所受的冲击伤害更明显.  相似文献

利用培养皿种子萌发及土壤盆栽方法对新疆特有藜科1年生盐生植物异子蓬(Suaeda aralocaspica)在不同盐分(NaCl和Na2SO4)、变温(4~30℃)和激素(GA3)处理下的种子萌发及成苗行为进行了研究.结果表明:异子蓬2种种子在NaCl和Na2SO42种盐胁迫下的萌发和休眠特性表现出明显差异,其中黑色种子的萌发率比褐色种子低;黑色种子属于非深度生理休眠,可分别通过15/30℃变温处理、4℃低温层积30 d及用800mg·L-1赤霉素处理促进其萌发;盐处理及温度变化对褐色种子的出苗率和成苗率影响显著,对黑色种子则不明显,说明黑色种子萌发存在着持续性的缓萌机制,有利于其形成持久种子库,以上特性均显示出其对初春盐生环境的适应性.  相似文献

采用砂培方式,研究了外源5-氨基乙酰丙酸(ALA)对盐胁迫下菘蓝种子的萌发、幼苗叶片的可溶性糖含量、丙二醛(MDA)含量及其抗氧化酶活性的影响,探讨ALA缓解菘蓝受盐胁迫伤害的响应机制。结果显示:(1)菘蓝种子萌发及幼苗生长在100 mmol·L-1 NaCl胁迫下受到明显的抑制,种子发芽率、发芽势、发芽指数、活力指数与自然含水量均显著降低,丙二醛含量、可溶性糖含量以及超氧化物歧化酶(SOD)、过氧化物酶(POD)活性显著升高。(2)盐胁迫下适宜浓度的ALA处理显著提高了种子萌发率、自然含水量及SOD、POD和CAT活性,降低了可溶性糖和丙二醛的含量,并以16.7 mg·L-1 ALA处理盐胁迫下菘蓝种子的发芽率、发芽势最大,其幼苗的SOD、POD、CAT活性最强。研究表明,盐胁迫显著抑制菘蓝种子的萌发及幼苗生长,适宜浓度的ALA能够有效缓解盐胁迫对菘蓝种子萌发及幼苗生长的伤害,提高植株的抗盐性,并以16.7 mg·L-1 ALA处理效果最佳。  相似文献

为筛选能够应用于干旱地区及盐性土壤的花海植物,以10种具有代表性的菊科植物为材料,利用PEG-6000溶液和盆栽控水法分别在种子萌发期和幼苗期进行干旱胁迫,利用NaCl溶液分别在这两个时期进行盐胁迫,测定供试植物种子的发芽指标、幼苗的质膜相对透性以及抗氧化酶活性等生理指标,通过隶属函数法全面评价10种菊科植物的抗旱性和抗盐性。结果表明:干旱或盐胁迫下,10种测试植物的生理指标变化显著;其中,相对电导率、丙二醛(MDA)含量均增加,但抗性强的品种增幅较小;过氧化物酶(POD)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)活性因物种不同存在差异;抗旱性强弱次序为波斯菊万寿菊小丽花硫华菊五色菊向日葵麦秆菊矢车菊勋章菊异果菊;抗盐性强弱次序为波斯菊硫华菊小丽花万寿菊五色菊矢车菊向日葵麦秆菊勋章菊异果菊。  相似文献

水稻过氧化氢酶B (catalase B, CatB)主要在非光合组织、愈伤组织、根和种子中表达,参与调控活性氧动态水平的平衡和根系的生长。目前, CatB参与非生物胁迫响应的功能在很大程度上是未知的。本研究利用基因编辑技术CRISPR/Cas9获得OsCatB基因敲除的纯合突变体catb,并分析了catb在盐、高温和氧化等胁迫处理下的生理和生化表型。研究结果发现,在盐处理下,突变体catb的生理和生化表型与野生型植株无显著差异;在热胁迫下, catb的过氧化氢酶活性和存活率与野生型植株相比显著降低,而H2O2含量显著升高;在氧化胁迫下, catb的幼苗高度低于野生型。这些结果说明,水稻CatB参与盐胁迫响应的调控不明显,而主要参与了高温和氧化胁迫响应,并正调控水稻的高温和氧化胁迫耐受性。该研究为进一步探究Os CatB参与逆境响应的分子机制和培育抗逆水稻品种提供了一定的理论指导和遗传材料。  相似文献

为探讨盐胁迫下两类不同地区藜麦品种幼苗的耐盐机制,该文利用不同浓度的NaCl溶液,对来自青海省海东市乐都区的‘LD-13’(低盐地区)和来自青海省海西州乌兰县的‘WL-192’(高盐地区)的2个藜麦品种的种子和幼苗进行盐胁迫处理,研究了种子萌发指标(发芽率、发芽势、发芽指数),生长指标(鲜重、根长、茎长)及生理指标(MDA含量及SOD、POD、CAT活性)等的变化。结果表明:低浓度盐(NaCl浓度小于250 mmol·L~(-1))胁迫下,2个藜麦品种种子的萌发、幼苗生长及生理活性表现均较适宜;但中、高浓度盐(NaCl浓度大于250 mmol·L-1)胁迫下,其种子萌发、生长及幼苗生理活性均受到不同程度的抑制。从耐盐性综合评价值D值来看,虽然同为山谷型的藜麦品种,但适应盐碱地栽培的‘WL-192’品种比低盐土壤中生长的‘LD-13’品种更耐盐。推测‘WL-192’比‘LD-13’耐盐性除了受选育地区土壤盐度的影响外,还可能与品种自身及光周期、温度、海拔、纬度等外部生长环境等因素有关。结合青海西部地区气候环境和盐碱土地资源开发利用,‘WL-192’品种更适合在青海地区推广种植。  相似文献

NaCl胁迫对藜麦幼苗生长和抗氧化酶活性的影响   总被引:4,自引:0,他引:4  
以国内首个藜麦自育品种"陇藜1号"为材料,采用温室盆栽法,以蒸馏水处理作为共同对照(CK),分别用100、200、300、400和500mmol/L NaCl水溶液处理藜麦种子和盆栽幼苗,通过测定种子萌发指标及处理后第5、10、15天藜麦幼苗叶片叶绿素、可溶性糖、脯氨酸、MDA含量及抗氧化酶活性,分析NaCl胁迫对藜麦生长发育及其生理特性的影响,探讨藜麦的耐盐生理机制。结果表明:(1)随NaCl浓度的升高,藜麦种子发芽率、发芽势、发芽指数和活力指数先升高后下降,且在200 mmol/L NaCl处理下种子各发芽指标均达到最高,比CK分别升高了6.40%、28.18%、20.77%和30.91%。(2)随NaCl浓度的升高,藜麦幼苗根部和茎部生长均受到抑制,且茎部生长受到抑制程度大于根部。(3)随NaCl浓度的升高和处理时间的延长,藜麦幼苗叶片叶绿素含量先升高后下降,可溶性糖、脯氨酸和MDA含量逐渐升高,SOD、POD、CAT和APX活性增强。研究发现,低浓度盐胁迫处理可增加藜麦幼苗叶片内渗透调节物质含量,增强抗氧化酶活性,清除多余活性氧,从而促进幼苗根系生长,提高幼苗耐旱性;初步推断藜麦耐盐阈值为200~300mmol/L NaCl。  相似文献

以新疆荒漠盐生植物异子蓬(Suaeda aralocaspica)异型种子为材料,通过外源ABA直播(长时间)与浸种(短时间)处理分别研究不同浓度(0、0.5、1.0、5.0、10.0μmol/L)ABA对异型种子萌发、幼苗生长的影响,并通过定量PCR技术分析与萌发相关的ABC转运蛋白基因、丝裂原活化蛋白激酶激酶基因和DNA修复和转录因子基因在5.0μmol/L ABA处理下的表达规律,探讨外源ABA影响异子蓬异型种子萌发差异的机制。结果显示:(1)外源ABA直播处理下,褐色种子萌发及幼苗生长均受到明显抑制,黑色种子的萌发和幼苗生长在低浓度被促进,较高浓度被显著抑制;短时间浸种处理,能够同时促进异型种子的萌发及幼苗生长,且随ABA浓度增加促进效应增强。(2)直播萌发后,褐色种子中3个基因的表达量与对照相比均不变或降低表达(除了DNA修复和转录因子基因在8 h显著升高),黑色种子与对照相比均上调表达(除了ABC转运蛋白基因在8h显著下调);短时间浸种萌发后,褐色种子中3个基因的表达量均比对照显著升高,黑色种子中升高不明显。基因表达规律与种子萌发结果趋势一致,暗示ABA浸种可能触发了异子蓬种子萌发内在机制并对随后的萌发过程产生促进作用。而黑色种子对ABA处理表现出较好的萌发响应,可能是其幼苗能抵御荒漠地区逆境环境的重要原因。  相似文献

Background and Aims

Chenopodium album is well-known as a serious weed and is a salt-tolerant species inhabiting semi-arid and light-saline environments in Xinjiang, China. It produces large amounts of heteromorphic (black and brown) seeds. The primary aims of the present study were to compare the germination characteristics of heteromorphic seeds, the diversity of plant growth and seed proliferation pattern of the resulting plants, and the correlation between NaCl stress and variation of seed heteromorphism.

Methods

The phenotypic characters of heteromorphic seeds, e.g. seed morphology, seed mass and total seed protein were determined. The effects of dry storage at room temperature on dormancy behaviour, the germination response of seeds to salinity stress, and the effect of salinity on growth and seed proliferation with plants derived from different seed types were investigated.

Key Results

Black and brown seeds differed in seed morphology, mass, total seed protein, dormancy behaviour and salinity tolerance. Brown seeds were large, non-dormant and more salt tolerant, and could germinate rapidly to a high percentage in a wider range of environments; black seeds were salt-sensitive, and a large proportion of seeds were dormant. These characteristics varied between two populations. There was little difference in growth characteristics and seed output of plants produced from the two seed morphs except when plants were subjected to high salinity stress. Plants that suffered higher salinity stress produced more brown (salt-tolerant) seeds.

Conclusions

The two seed morphs of C. album exhibited distinct diversity in germination characteristics. There was a significant difference in plant development and seed proliferation pattern from the two types of seeds only when the parent plants were treated with high salinity. In addition, seed heteromorphism of C. album varied between the two populations, and such variation may be attributed, at least in part, to the salinity.  相似文献

Suaeda salsa, a leaf succulent shrub in the family Chenopodiaceae, is one of the most important halophytes in China. Suaeda salsa produces dimorphic seeds (soft brown seeds and hard black seeds). Seeds of S. salsa were collected from the coastal salt flats near Huanghua City, China. Experiments were conducted to determine the salinity-alleviating effect of plant growth regulators, nitric oxide, nitrate, nitrite and light on the germination of dimorphic seeds of S. salsa. Brown seeds had a higher germination rate than black seeds in all experiments. Black seeds were more sensitive to salt in the absence of light in comparison to brown seeds. Brown seeds absorbed water more quickly in comparison to black seeds and were found to be more tolerant of salt stress. Our results showed that 1-aminocyclopropane-1-carboxylate (ACC, the immediate precursor of ethylene), nitrite, GA4 and BA improved seed germination in the presence of salt. However, nitrate, GA1, GA3 failed to alleviate salt stress. ABA inhibited seed germination and seedling growth. Possible mechanisms involved in the alleviation of salt stress in S. salsa seeds and the ecological adaptation of the seeds to the environment are discussed.  相似文献

恶劣环境下,人工海防林因面临养分胁迫而经营困难。为探讨盐、磷胁迫对主要海防林树种木麻黄和台湾相思种子萌发及生长的影响,该研究分别用不同浓度的NaCl(盐)和KH2PO4(磷)溶液处理种子和浇灌幼苗,测定种子萌发和幼苗生长指标。结果表明:(1)高盐胁迫显著抑制种子萌发,对幼苗生长有一定影响,但两种植物影响程度不同;台湾相思种子萌发耐盐性高于木麻黄,前者相对盐害率最大值为23.03%,后者为89.15%;随着盐浓度增加,木麻黄和台湾相思种子的发芽率、发芽势、发芽指数和活力指数均降低,对应最大值分别为38.70%、34.67%、18.70、0.055和76.67%、62.22%、48.46、6.11。(2)两种植物的株高和根长随盐浓度增加而降低,木麻黄和台湾相思株高分别为12.29~6.01 mm和48.27~17.33 mm,根长分别为8.57~1.45 mm和33.41~5.88 mm;台湾相思根、茎、叶生物量及根冠比均随盐浓度的增加逐渐减小,木麻黄各处理差异较小。(3)台湾相思的种子和幼苗较木麻黄更耐低磷环境,二者最适磷浓度存在差异;木麻黄种...  相似文献

Salsola ikonnikovii (Chenopodiaceae), a drought-tolerant plant species that is distributed in sand or light-saline soil in Xinjiang, China, produces seeds (fruits) with attached winged perianths. To study the role of the wing in seed germination under salt stress and to further investigate the growth and physiological responses of the plants to salt stress, the germination behaviour of S. ikonnikovii was determined after winged and non-winged seeds were treated with 0–1000 mmol · L?1 NaCl. Several parameters of two-month old plants that had been treated with NaCl for three weeks were measured. The results revealed that the winged perianths limited germination but protected the seeds from salt damage. The growth of the plants was stimulated by lower concentrations of salt (≤100 mmol · L?1 NaCl), while increasing salt concentrations inhibited growth. The level of reactive oxygen species and malondialdehyde increased significantly at high concentrations of salt. Correspondingly, concentrations of the osmolytes proline, betaine, and soluble sugars, and the activities of antioxidative enzymes (catalase, peroxidase and superoxide dismutase) increased, but the levels of non-enzymatic antioxidants (carotenoids, glutathione) were significantly reduced at high salt concentrations. These results imply that osmotic adjustment and the antioxidative system may work synergistically to ensure that a plant grows normally under high salt concentrations.  相似文献

Salinity and nitrogen are two important environmental factors that affect the distribution of halophytes in their natural saline habitats. Seeds of the euhalophyte Suaeda salsa L. were harvested from plants that had been treated with 1 or 500 mm NaCl combined with 0.5 or 5 mm NO3?‐N (nitrate) for 115 days in a glasshouse. Germination was evaluated under different concentrations of NaCl and nitrate. Plants exposed to high salinity (500 mm ) and low nitrate (0.5 mm ) tended to produce heavy seeds. Either high salinity (500 mm ) or high nitrate (5 mm ) increased the brown/black seed ratio. The concentrations of Na+, K+, and Cl? were higher in brown than in black seeds, and NO3? concentrations were higher in black than in brown seeds, regardless of NaCl and nitrate treatments during plant culture. Regardless of NaCl and nitrate concentrations during germination, seeds from plants grown with 0.5 mm nitrate generally germinated more rapidly than seeds from plants grown with 5 mm nitrate, and the difference was greater for black than for brown seeds. Exogenous nitrate during germination enhanced the germination of brown seeds less than that of black seeds. Producing more brown seeds and heavy black or brown seeds appears to be an adaptation of S. suaeda to saline environments. Producing more black seeds, which tend to remain dormant, should reduce competition for nitrogen and appears to be an adaptation to nitrogen‐limited environments. In conclusion, nitrate provided exogenously or by mother plants to black seeds may act as a signal molecule that enhances the germination of black S. suaeda seeds.  相似文献

种子异型性是指同一植株产生不同形状或行为种子的现象。根据异型种子在植株上的生长位置, 种子异型性可划分为地上下结实性和地上种子异型性两类。此现象已在26科129属292种被子植物中报道。异型性种子植物主要分布于干旱半干旱区、荒漠和盐渍土地区等干扰强烈的环境, 在菊科和藜科中最为常见, 主要出现在一年生植物中。种子异型性在避免密集负效应、减弱同胞子代间的竞争、采取两头下注策略以适应时空异质性环境等方面具有重要的进化生态意义。该文系统总结了国内外种子异型性的研究工作, 主要内容包括: 1)种子异型性的概念、类型和种类, 2)具有异型种子植物的生境和生活型, 3)异型种子的生态学特性, 4)种子异型性的理论模型, 5)种子异型性的生态意义。在综述文献的基础上, 对今后的研究进行了展望。针对国内外的研究现状, 提出两点建议: 1)系统调查具有种子异型性现象的植物种类, 摸清其生物学特性; 2)确定研究种子异型性现象的模式植物, 从生态学、生理学和分子生物学等学科角度来研究种子异型性的个体发育机制及分子调控机理。  相似文献

Abstract

Salt stress is one of the major environmental factors limiting crop growth and yield. To understand the effect of salt stress on plant growth, we investigated the response of three perilla varieties (Suyin 1, Ziye 7, and Ziye 10) to NaC1 exposure at concentrations of 0, 50, 100, 150, 200, and 250 mM in terms of seed germination, seedling growth, root activity, contents of soluble sugar, proline, and malondialdehyde (MDA), and peroxidase (POD) enzyme activity. Germination characteristics, such as the percentage of seed germination, tended to decrease with increasing NaC1 concentrations. After three weeks of salt stress, the three varieties exhibited different salt tolerance in terms of seed germination, seedling growth, and physiological changes: seedling growth was inhibited to various degrees, seedling vigor and root activities decreased, and MDA, proline, and soluble sugar contents increased with increasing NaCl concentrations. POD enzyme activity, a protective mechanism against salt stress, increased at low NaC1 concentrations in Suyin1 (0–150 mM) and Ziye 7 (0–100 mM), and then decreased at higher NaCl concentrations. In Ziye 10, on the other hand, POD activity dropped significantly with increasing NaCl concentrations. These results suggest that among the three varieties Suyin 1 is more salt tolerant than Ziye 7 and Ziye 10, and that Ziye 10 is the most sensitive to salt stress.  相似文献

Summary The R gene of soybean is involved in anthocyanin synthesis in the seed coat, and its r-m allele conditions a variegated distribution of black spots and/or concentric rings of pigment superimposed on an otherwise brown seed coat. We describe an unusual feature of r-m that causes expression at the R locus to switch between active and inactive phases both somatically and germinally. Non-heritable somatic changes of the allele produce single plants containing mixtures of seed with different coat colors (black+striped or brown+striped). Heritable changes of the r-m allele are manifested in progeny plants which produce all black seed or all brown seed. Surprisingly, subsequent generations from revertant sublines show continued instability of the allele such that brown revertants (r*/r*) or homozygous black seed revertants (R*/R*) can give rise to striped or striped+black-seeded plants. Thus, the revertants produced by the r-m allele are not stable but interconvert between all three forms (R*, r*, and r-m) at detectable frequencies. Mutability of the r-m allele in a different genetic background has also been found after inter-crossing various soybean genotypes.  相似文献
