用添加5%异麦芽寡糖的四种培养基观察对双歧杆菌等5种细菌的生长促进作用。结果显示,在营养较丰富的培养基中对双歧杆菌生长促进作用不显著;在营养较差的培养基中显著(P<0.05),对嗜酸乳杆菌也显著,而对大肠杆菌、脆弱类杆菌和艰难梭菌不显著。  相似文献

目的比较不同培养基成分对福赛类杆菌ATCC43037生长的影响,寻找一种有效促进福赛类杆菌生长的培养基.方法将福赛类杆菌ATCC430 37 接种到5种不同培养基(包括琼脂板和液体培养基)中,在厌氧培养箱内37℃厌氧培养7 d,观察平板上菌落生长情况,在λ=550 nm处每24 h测定液体培养菌液A值.细菌通过革兰染色和PCR法鉴定.结果1.生长在血平板上的福赛类杆菌ATCC43037菌落形态呈粉红色或白色小斑点状,在以BHI 为基础培养基中添加氯化血红素、维生素K3、N-乙酰胞壁酸的No2血琼脂培养基生长最好,而在无氯化血红素、维生素K3及血的No4琼脂板上不生长.2.福赛类杆菌ATCC43037在以TSB 为基础培养基中添加酵母提取物、植物蛋白胨、氯化血红素、维生素K3、N-乙酰胞壁酸和D TT的No1液体培养基生长最好,培养7 d后A值>0.8,在缺乏N-乙酰胞壁酸的No5液体培养基中几乎不生长.3.菌落细菌革兰染色为G 梭状杆菌,PCR法鉴定为福赛类杆菌.结论福赛类杆菌在血平板和液体培养基中存在不同的生长特性,N-乙酰胞壁酸是福赛类杆菌在液体培养基中生长最重要的成分,DTT有益于细菌生长.  相似文献

从土壤里分离到两类在碱性培养基中生长并产生环状糊精葡糖基转移酶的细菌,一类与国外已在工业生产中使用的芽孢杆菌相似,另一类为无芽孢的杆状细菌,尚未见报道。两类细菌的代表菌株,芽孢杆菌32—2要求的产酶因子以酵母膏最好;无芽孢杆菌52—2号则用玉米浆最好。碳源和氮凉也有所不同。52—2产酶过程比32—2稍晚,酶反应条件基本相近。  相似文献

从饲料用麦麸中分离出的12种细菌均能支持无菌家蝇Musca domestica L.幼虫在胰化酪蛋白大豆卵黄琼脂(TrypticaseSoy EggYolkAgar,TSEYA)培养基中完成整个生长发育过程。1)幼虫在接种香味类香味菌Myroidesodoratimimus的TSEYA培养基中生长时间最短,仅需97.61±1.14h;2)幼虫在接种醋酸钙不动杆菌Acinetobacter calcoaceticus的TSEYA培养基中的化蛹率可达到86.81%;3)从接种嗜水汽单胞菌Aeromonas hydrophila的TSEYA培养基中得到的蝇蛹重量最高,达到20.15±0.23mg/个;4)除铜绿假单胞菌Pseudomonasaeruginosa饲养的家蝇羽化率较低(60.87%)外,其余各种细菌饲养的羽化率在84.33%~97.47%之间。此外,枯草芽孢杆菌Bacillus subtilis、香味类香味菌、聚团肠杆菌Enterobacter agglomerans以及成团肠杆菌Pantoeaagglomerans可作为单一营养来源支持幼虫完成整个生长发育过程。对枯草芽孢杆菌、金黄色葡萄球菌Staphylococcusaurous、成团肠杆菌、大肠杆菌Escherichiacoli、奇异变形杆菌Proteusmirabilis以及香味类香味菌中各种营养成分进行分析,结果发现,6种细菌均能提供大量的维生素如(0.105~1.08g/kg)。在氨基酸方面,香味类香味菌和枯草芽孢杆菌中的10种昆虫必需氨基酸含量与总氨基酸含量之比(Essential amino acid/Totalaminoacid,EAA/TAA)最高,而金黄色葡萄球菌最低。这个比例与蛹重呈正相关(p=0.031)。在脂肪酸相对含量方面,金黄色葡萄球菌具有最高的饱和脂肪酸含量(76.38%),香味类香味菌含有56.79%的不饱和脂肪酸,而枯草芽孢杆菌则具有最高的支链脂肪酸含量(42.16%)。  相似文献

目的分析口臭患者龈下菌斑和舌苔上主要相关厌氧菌的分布情况。方法选择口腔门诊中口臭患者29例,鼻闻法来确定产臭部位和非产臭部位;分别采集龈下菌斑和舌苔标本接种在非选择性培养基和核梭杆菌选择性培养基,厌氧培养5d后记录非选择性培养基上生长的细菌总数、产黑色素细菌总数及核梭杆菌选择性培养基上生长的目的菌总数。结果29例患者中,15例患者的口臭主要来源于龈缘菌斑,10例主要来源于舌苔,4例患者的口臭由龈缘菌斑和舌苔共同产生;产臭部位和非产臭部位相比,细菌总数、产黑色素菌和具核梭杆菌数都明显上升(P〈0.01)。结论口源性口臭患者口气变化与产黑色素细菌、核梭杆菌相关。  相似文献

不同培养基组合提高土壤细菌可培养性的研究   总被引:8,自引:1,他引:7  
为选择性采用多培养基组合以提高土壤细菌可培养性,利用变性梯度凝胶电泳(DGGE)技术研究了贫营养、富营养和自然营养培养基在3种培养方式下获得细菌种群的差异。结果表明:平板培养条件下,细菌在贫营养培养基上生长较慢,菌落连续稳定形成。培养5d后,富营养的LB培养基和贫营养的R2A培养基获得菌落数最多,分别是贫营养的0.1×LB培养基获得菌落数的5.1倍和5.3倍。7种培养基中,LB培养基获得细菌种群数目最多,营养成分适当稀释后,培养物中有新的种群出现。贫营养培养基和富营养培养基培养物DGGE图谱相似性低,条带互补性强。三角瓶静置培养时,R2A和LB培养基获得细菌种群数目较多,其它几种培养基获得的细菌类群都能在这2种培养基中找到。试管静置培养条件下,LB培养基获得细菌种群数目最多,某些种群也只出现在R2A培养基和TSB培养基上,R2A及LB培养基与TSB培养基获得的细菌种群差异较为明显。研究结果为特殊培养基设计及选用合适培养基分离土壤细菌提供参考。  相似文献

广东南岭森林土壤中蕴藏着丰富的生物资源,但对其中的可培养细菌种类仍缺乏系统了解。本研究采用贫营养型的R2A培养基和富营养型的TSA培养基对南岭森林土壤中细菌进行了分离,获得细菌408株,分别从属于厚壁菌门、变形菌门、放线菌门和拟杆菌门的35属。其中的优势类群为厚壁菌门,占分离总数量的71%。在属水平,芽胞杆菌及其近缘属为优势类群。除芽胞杆菌外,假单胞菌、伯克霍尔德氏菌草酸杆菌科Collimonas属和罗丹诺杆菌科Dyella属是分离获得的主要类群。R2A培养基在分离革兰氏阴性的变形菌门菌株方面表现出一定的偏好性,而TSA培养基分离得到的更多为快速生长的芽胞杆菌及其近缘的革兰氏阳性细菌。发现了15属的菌株具有一定的水解酶活性,大多表现出对淀粉和牛奶的水解活性,对有机磷的水解性能优于对无机磷的水解。降解纤维素的菌株则主要集中于芽胞杆菌及其近缘属中。发现了潜在新物种26株,分布于芽胞杆菌、Dyella、类芽孢杆菌等9属中。本研究仅使用了两种营养类型的培养基,进一步借助培养组学技术有望能更加全面反映南岭森林土壤中的可培养微生物多样性。  相似文献

【目的】研究添加泥浸汁与否对太湖沉积物中可培养细菌的影响。【方法】采用R2A培养基和添加泥浸汁R2A培养基对沉积物中细菌进行分离培养,16S r RNA基因系统发育分析比较种群结构。【结果】培养基中添加泥浸汁,可使可培养细菌的种类数量增加到1.6倍。16S r RNA基因序列分析表明,培养的优势细菌类群存在明显差别。R2A培养基上生长的细菌主要为厚壁菌门(52%)、放线菌门(24%)、变形菌门(20%)和拟杆菌门(4%),其中大部分细菌与芽孢杆菌属、假单胞菌属、节杆菌属等关系密切;而添加泥浸汁的R2A培养基上生长的细菌则主要为变形菌门(40%)、放线菌门(35%)、厚壁菌门(22.5%)和拟杆菌门(2.5%),与鞘脂单胞菌属、芽孢杆菌属、副球菌属、节杆菌属等关系密切。【结论】添加泥浸汁原始营养因子可提高沉积物中可培养细菌的多样性,提高菌种可培养效率。  相似文献

厌氧条件下以羧卡青霉素诱导金黄色葡萄球菌、大肠杆菌和蜡样芽胞杆菌形成L型,观察细菌L型在厌氧条件下的形成、形态、生长及时渗透压的敏感性等特性。结果表明:蜡样芽胞杆菌在厌氧条件下不能形成L型或其L型在厌氧条件下亦不能返祖。金黄色葡萄球菌和大肠杆菌在厌氧条件下虽能诱生L型,但形成丝状体的构成L型菌落难以传代培养,厌氧培养未见L型圆球体和典型L型油煎蛋样菌落。金黄色葡萄球菌L型在含1%~10%NaCl的L型培养基上可生长形成L型菌落或非菌落形式存在的L型巨形体;大肠杆菌和蜡样芽胞杆菌的L型在含2%~6%NaCl的L型培养基上可生长形成L型菌落或非菌落形式存在的L型巨形体。涂片染色或返祖试验证实细菌L型在含0.5%NaCl的L型培养基或常规细菌学培养基上亦可生存。非菌落性L型巨形体和丝形体是细菌L型在琼脂培养基上广泛的存在形式。  相似文献

棉铃虫的人工饲养是对其进行昆虫病原菌生物测定的重要步骤,而饲料污染是影响棉铃虫培养成功率的主要因素。因此,选择合适的抗生素对棉铃虫的饲养具有重要意义。为明确棉铃虫饲料中污染细菌的组成,筛选可抑制污染菌的抗生素,本研究以受污染棉铃虫饲料中分离出的9个菌株为实验材料,观察菌株形态。采用16S rDNA系统发育分析法构建进化树,同时根据药敏与生理生化实验,确定分离菌株的生理生化特性。结果表明,9个菌株共呈现5种生理生化特性,其中,2株与肠球菌属(Enterococcus sp.)接近,2株与埃希氏菌属(Escherichia sp.)接近,2株与志贺氏菌属(Shigella sp.)接近,2株与肠杆菌属(Enterobacter sp.)接近,1株与沙雷氏菌属(Sarratia sp.)接近。抗生素筛选实验结果显示,卡那霉素对于不同菌株的抑菌效果最好。这为提高棉铃虫人工饲养成活率具有重要意义。  相似文献

We studied the effects of predation on the cytometric and phylogenetic features of two enriched bacterial communities obtained from two cultures of marine heterotrophic nanoflagellates: Jakoba libera and a mixed culture of Cafeteria sp. and Monosiga sp. Protists were harvested by flow cytometric cell sorting and eight different treatments were prepared. Each bacterial community was incubated with and without protists, and we added two treatments with protists and the bacteria present after the sorting procedure (cosorted bacteria). The bacterial community derived from the culture of Jakoba libera had higher green fluorescence per cell (FL1) than that derived from the mixed culture of Cafeteria sp. and Monosiga sp. When the experiment began all treatments presented bacterial communities that increase in fluorescence per bacterium (FL1); after that the FL1 decreased when bacteria attained maximal concentrations; and, finally, there was a new increase in FL1 toward the end of the experiment. Cosorted bacteria of Jakoba libera had the same fluorescence as the bacterial community derived from this protist, while the bacteria derived from the mixed culture of Cafeteria sp. and Monosiga sp. was nearly twice as fluorescent than that of the parental community. All treatments presented a general decline of SSC along the incubation. Therefore, there was a small influence of protists on the cytometric signature of each bacterial community. However, each bacterial community preyed by Jakoba libera or the mixed culture of Cafeteria sp. and Monosiga sp. led to four different phylogenetic fingerprint. Besides, the final Communities were different from the fingerprint of controls without protists, and most of them diverge from the fingerprint of cosorted bacteria. Our results confirm that changes in the phylogenetic composition of marine bacterial communities may depend on the initial communities of both bacteria and protists.  相似文献

The structure of microbial communities of brown algae, red algae, and of the red alga Gracilaria verrucosa, healthy and affected with rotten thallus, were comparatively investigated; 61 strains of heterotrophic bacteria were isolated and characterized. Most of them were identified to the genus level, some Vibrio spp., to the species level according to their phenotypic properties and the fatty acid composition of cellular lipids. The composition of the microflora of two species of brown algae was different. In Chordaria flagelliformis, Pseudomonas spp. prevailed, and in Desmarestia viridis, Bacillus spp. The composition of the microflora of two red algae, G. verrucosa and Camphylaephora hyphaeoides, differed mainly in the ratio of prevailing groups of bacteria. The most abundant were bacteria of the CFB cluster and pseudoalteromonads. In addition, the following bacteria were found on the surface of the algae: Sulfitobacter spp., Halomonas spp., Acinetobacter sp., Planococcus sp., Arthrobacter sp., and Agromyces sp. From tissues of the affected G. verrucosa, only vibrios were isolated, both agarolytic and nonagarolytic. The existence of specific bacterial communities characteristic of different species of algae is suggested and the relation of Vibrio sp. to the pathological process in the tissues of G. verrucosa is supposed.  相似文献

对采自辽宁省前屯煤矿的6种不同风化程度的褐煤样品进行扫描电子显微镜观察发现:刚采掘出来的褐煤表面几乎没有微生物存在.经5个月、1年及4年堆积风化的褐煤中也只见到休眠孢子和少量菌丝.将褐煤样品在潮湿状态下培养10天后,扫描电镜观察发现:刚采出来的褐煤及经5个月风化的褐煤表面有大量放线菌生长,而且菌落周围有褐煤被降解迹象.经1年风化的褐煤中除有大量放线菌及细菌生长外,真菌也有所增加.而在经4年风化褐煤中主要是真菌明显增加.平板计数结果同样说明褐煤风化过程中微生物存在演替现象:放线菌为褐煤初期降解的主要微生物,随后是细菌,在风化程度较高的褐煤中,真菌则为优势降解菌.三株优势放线菌为诺卡氏菌(Nocardia Sp.),束丝放线菌(Actinosynnema Sp.)和链霉菌(Streptomyces sp.).两株优势细菌均为节杆菌(Arthrobacter sp.).两株曲霉为栖土曲霉(A.terricola)及褚曲霉(A.ochraceous),为褐煤风化过程的优势真菌.  相似文献

植物根际促生菌是一类可促进植物生长的有益细菌,有效的根际促生菌剂可以减少化肥施用。以束村氏菌属(Tsukamurella sp.)P9、伯克霍尔德氏菌属(Burkholderia sp.)P10、以及P9和P10混合菌液作为接种菌株,研究促生菌对花生生长、植株及土壤营养、根际土壤微生物类群及功能的影响。30 d盆栽实验结果表明,接种组的花生鲜重、株高及根长均显著提高;根际土壤细菌总数、固氮菌和溶磷菌数均明显高于未接种组;氮循环功能菌群数量有不同程度提高,土壤蔗糖酶、脲酶及过氧化氢酶均高于对照;土壤碱解氮及速效钾显著提高,植株营养指标有所提升,尤以P10接种效果更优。本研究初步结论表明2株促生菌通过活化土壤微生物、提高植株的有效营养元素含量,促进了花生的生长。  相似文献

Do detached root-cap cells influence bacteria associated with maize roots?   总被引:2,自引:0,他引:2  
Abstract. A model rhizosphere has been used which consisted of detached root-cap cells of maize in their surrounding root-cap mucilage on the surface of Noble agar. These cells were co-cultured for periods up to 32 d with eight different bacterial isolates from soil-grown roots and surrounding soil and two laboratory cultures. Cap cells were unaffected by the bacteria. There were five different type-specific responses of the bacteria in proximity to the cap cells. There were, strong growth inhibition ( Rhizobium sp. and Escherichia coli ), strong stimulation ( Pseudomonas fluorescens , laboratory strain), mixed weak inhibition or stimulation ( Pseudomonas fluorescens , field isolate), early inhibition followed by strong stimulation then spore formation ( Bacillus spp.), no effect ( Streptomyces sp. and Cytophaga sp.). It is concluded that detached root-cap cells are actively involved in the establishment of characteristic rhizosphere bacterial microflora.  相似文献

Abstract If predators select for or against contaminant-degrading bacteria, it will affect bacterial survival and has important implications for bioremediation. Protozoa are important predators of bacteria. In order to determine whether protozoa preyed differentially on bacteria with different degradation abilities, two ciliates (Euplotes sp. and Cyclidium sp.) and three strains of PAH-degrading bacteria (Vibrio spp., degrading naphthalene, anthracene, or phenanthrene) were isolated from sediment from New York/New Jersey Harbor. By manipulating growth conditions, bacterial strains with different PAH-degradation abilities and different cell properties were produced. Stepwise regression models were used to analyze how clearance rates on suspended bacteria and grazing rates on bacteria attached to particles were affected by bacterial size, hydrophobicity, C:N ratio, protein content, and PAH-degradation ability. Clearance rates ranged from 0 to 49 nl ciliate−1 h−1 for Euplotes sp. and from 0 to 1.7 nl ciliate−1 h−1 for Cyclidium sp. Clearance rates of both ciliates were positively correlated with bacterial size, hydrophobicity, and protein content, and negatively correlated with C:N ratio. PAH degradation ability had no (for Euplotes sp.) or small (for Cyclidium sp.) effects on clearance rates. The models accounted for 63–75% of the variation in clearance rates on different bacteria. Only Euplotes sp. grazed on attached bacteria, at rates from 3 to 176 bacteria ciliate−1 h−1. A regression model with only C:N ratio and protein content explained 45% of the variation in grazing rates. These models indicate that multiple properties of bacteria affect their susceptibility to predation by ciliates, but PAH-degradation ability per se has little effect. 【目的】分析赣南脐橙黄龙病植株和健康植株叶片内生菌,对比不同培养条件下培养出的内生菌,为筛选出对柑橘黄龙病原菌有影响的伴生菌奠定理论基础。【方法】通过PCR方法对脐橙中黄龙病菌进行验证,并基于16S r RNA基因高通量测序技术对患病与健康赣南脐橙叶片内生菌以及不同培养基富集培养后的内生菌进行多样性分析。【结果】所采集样品中有5株患病株,5株健康株。5株病株中共同含有的细菌属有13个,其中7个在5株健株中也共同存在。Defluviicoccus属和Granulicella属在病健株植物中都是优势菌属,且在健株中的平均含量高于病株。病株与健株的样品相似度存在明显界线。富集培养后不同样本和不同培养基中菌属分布不同。肠杆菌属(Enterobacter)、短小杆菌属(Curtobacterium)、假单胞菌属(Pseudomonas)和泛菌属(Pantoea)得到了大量富集,不动杆菌属和沙雷氏菌属等9个菌属富集量较少。另外,培养和未培养各样本间未分类菌(Unclassified)含量差异也较大。【结论】赣南脐橙患病植株和健康植株叶片内生菌有着明显差异,黄龙病菌的存在改变了脐橙叶片原有内生细菌的菌群结构。从活体植物组织内直接检测才能得到真正的植物内生菌群落分布情况。通过分析菌群的差异,有望找到与柑橘黄龙病菌生长相关的伴生菌。  相似文献

昆虫肠道微生物对其寄主的生长发育、营养代谢、免疫以及农药抗性等方面都发挥着重要作用。为研究斜纹夜蛾Spodoptera litura幼虫肠道细菌的多样性,并为其功能验证做准备,本文利用传统微生物分离纯培养方法从斜纹夜蛾4龄幼虫肠道中共分离鉴定得到10株细菌,分别为属于变形菌门(Proteobacteria)的脱氮假单胞菌(Pseudomonas denitrificans),不动细菌(Acinetobacter sp.),肺炎克雷伯氏菌(Klebsiella pneumoniae)和肠杆菌(Enterobacter sp.);属于厚壁菌门(Firmicutes)的鸡葡萄球菌(Staphylococcus gallinarum),蒙氏肠球菌(Enterococcus mundtii),蜡样芽胞杆菌(Bacillus cereus)和枯草芽胞杆菌(Bacillus subtilis)以及放线菌门(Actinobacteria)的微杆菌(Microbacteriums sp.)和乳酪棒杆菌(Corynebacterium casei)。变形菌门和厚壁菌门是斜纹夜蛾肠道可培养细菌中的优势菌群。功能验证实验表明肠杆菌具备纤维素降解能力,微杆菌具备很强的苯酚降解能力。本研究为未来深入研究斜纹夜蛾肠道微生物的功能提供了方向和菌株材料。  相似文献
