对鳖血进行了10项生化指标测定。结果表明:乳酸脱氢酶、肌酸激酶分别为人的248和310倍,似与其长时间适应水底栖息或活动的需要有关;其甘油三酯和胆固醇含量却分别为人的034和042倍。测定了18种氨基酸,其总量为(7627±2151)mg/100mg样品,人体所需氨基酸均有,8种必需氨基酸含量占氨基酸总量的(4583±873)%。测定了18种矿物元素,与生长发育关系密切的铁、锌、铜、锰、铬等微量元素均含有。结果提示:作为功能食品,这种鳖具有良好的开发前景。  相似文献

蛋白质类生化药物,如L-天门冬酰胺酶、激肽释放酶、尿激酶、细胞色素C、胰岛素等,在临床应用上具有针对性强、毒副作用小、疗效好、容易为人体吸收和代谢等特点;但它们往往稳定性差易变性,而失去生物活性。异体蛋白具有抗原性而引起过敏反应等不足,因此人们一直希望能得到比天然蛋白质性能更为优良的蛋白质类生化药物,以便更好地为人类健康服务。 获得更为良好的蛋白质有以下四种方法:1.随机的基因突变、诱变种的筛选来获得新的蛋白质。但是,这种方法受到许多限制,特别是当要求改变蛋白质分子中某一特定的残基或区域是难以控制的;2.用化学修饰方法改良蛋白质的某些性能,这种方法已经取得了不少可喜的成果,但这种方法还是有很大局限性的,往往不易按人们的意愿来  相似文献

氨基酸分析在近几十年得到广泛应用,一般均采用进口的氨基酸混合标液进行,仅用一种方法来测定各种各样食品,饲料,药物中的氨基酸或进行课题研究,显然是不能满足需要的。本实验采用的模拟样品的方法自配标样,对200多个批号六种配方的产品作了测定,与传统方法比较。本方法结果精确可靠,且节省时间。  相似文献

Amino acid transport is a ubiquitous phenomenon and serves a variety of functions in prokaryotes, including supply of carbon and nitrogen for catabolic and anabolic processes, pH homeostasis, osmoprotection, virulence, detoxification, signal transduction and generation of electrochemical ion gradients. Many of the participating proteins have eukaryotic relatives and are successfully used as model systems for exploration of transporter structure and function. Distribution, physiological roles, functional properties, and structure-function relationships of prokaryotic α-amino acid transporters are discussed.  相似文献

Effects of Free Fatty Acids on Synaptosomal Amino Acid Uptake Systems   总被引:3,自引:11,他引:3  
Abstract: The Na+-dependent synaptosomal uptakes of proline, aspartic acid, glutamic acid, and γ-aminobutyric acid were strongly inhibited by monounsaturated fatty acids. With oleic acid, half-maximal inhibition was observed at about 15 μM. The Na+-independent uptakes of leucine, phenylalanine, histidine, and valine were less sensitive to inhibition by the unsaturated fatty acids. In contrast, the uptakes of all of these amino acids were unaffected by saturated fatty acids. The inhibition of proline uptake (and that of the other Na+-dependent amino acids) by oleic acid was overcome by the addition of serum albumin and the data presented further indicate that the previously reported stimulation of proline uptake by albumin could be related to its fatty acid binding properties.  相似文献

目的:建立液相色谱串联质谱同位素内标法检测神经递质类氨基酸并用于癫痫患者临床评价。方法:选用AAA-C18柱色谱柱,以乙腈水(含有0.01%七氟丁酸、0.1%甲酸)为流动相,采用梯度洗脱进行分离,血浆样品用iTRAQ-115衍生化试剂处理后,加入iTRAQ-114衍生化的氨基酸内标并进样,选用3200QTRAP型质谱仪的多重反应监测(MRM)扫描方式进行检测。疾病组与健康组的统计采用t检验和主成份分析。结果:疾病组和健康组氨基酸测定结果显示:Trp、GABA两组间没有显著性差异(P〉0.05),Arg、Gly、Ser、Tau、Asp、Glu、EtN、两组间有显著性差异(P〈0.05),通过PCA分析显示,疾病组与健康组之间差异明显,Asp、Glu、Ser等是引起差异的主要氨基酸。结论:试验方法灵敏、专属性强,并初步的用于癫痫患者体内氨基酸评价。  相似文献

Abstract: The uptake of amino acids by microvessels isolated from brains of rats was studied. Previous studies have demonstrated alterations in blood-brain amino acid transport after portacaval shunt in rats. In order to elucidate whether such changes in the blood-brain barrier were located in the microvessels, brain microvessels were isolated from both rats with portacaval shunt and controls. Brain microvessels from rats 2 weeks after shunt operations took up significantly greater amounts of 14C-labeled neutral amino acids, but not of glutamic acid. lysine, or α-methylaminoisobutyric acid than microvessels from sham-operated controls. Measurement of uptake kinetics showed a higher V max for phenylalanine and leucine uptake and a lower V max for lysine uptake in microvessels from shunted rats compared with control, whereas the respective K m's of uptake were similar in both preparations. The results suggest that changes in brain microvessel transport activity account for altered brain neutral amino acid concentrations after portacaval shunt and that such changes can be studied in vitro in isolated microvessels.  相似文献

石蒜(Lycoris radiata Herb.)是一种传统的中药.分别利用HP高效液相色谱仪(LC1100)和原子吸收分光光度计(TAS-986),测定石蒜鳞茎中17种氨基酸和7种矿质元素的含量;在测定的17种氨基酸中7种未检测到;除Zn以外,石蒜鳞茎中K、Ca、Mg、Fe、Mn、Cu都相对较低.  相似文献

Using the data from Protein Data Bank the correlations of primary and secondary structures of proteins were analyzed. The correlation values of the amino acids and the eight secondary structure types were calculated, where the position of the amino acid and the position in sequence with the particular secondary structure differ at most 25. The diagrams describing these results indicate that correlations are significant at distances between −9 and 10. The results show that the substituents on Cβ or Cγ atoms of amino acid play major role in their preference for particular secondary structure at the same position in the sequence, while the polarity of amino acid has significant influence on α-helices and strands at some distance in the sequence. The diagrams corresponding to polar amino acids are noticeably asymmetric. The diagrams point out the exchangeability of residues in the proteins; the amino acids with similar diagrams have similar local folding requirements. Extracellular amino acid levels in the rat piriform cortex, an area highly susceptible to seizure-induced neuropathology, were determined by means of intracranial microdialysis. Seizures were induced by systemic administration of either soman (O-1,2,2-trimethylpropyl methylphosphonofluoridate), a potent inhibitor of acetylcholinesterase, or the excitotoxin kainic acid. Extracellular glutamate levels increased in animals with seizures shortly after administration of either convulsant, but this change was statistically significant only in the case of soman-treated animals. Extracellular taurine levels increased markedly, reaching two- and fourfold baseline levels during the second hour of soman- and kainic acid-induced seizures, respectively. Taurine levels did not increase in the subpopulation of soman-treated animals without seizures, a finding indicating that elevation of extracellular taurine level is seizure related. Thus, we propose that taurine efflux may be a physiological cellular response to neuronal changes produced by excitotoxic chemicals, either directly or as a consequence of seizures.  相似文献
