Fluorescent lectins were used to study the chemical nature of carbohydrate moieties present on the surface of female and male germ cells isolated from mouse gonads during fetal and early posnatal development. Concanavalin A (ConA), lens culinaris agglutinin (LCA), ricinus communis agglutinin (RCAI) and wheat germ agglutinin (WGA) bound intensely to the germ cell plasma membrane at all stages studied. Other lectins such as ulex europaeus agglutinin (UEAI) and agglutinin (SBA) did not bind or bound moderately (SBA to female germ cells only). Distinct developmental-related changes were observed when female germ cells were labeled with fluorescein-conjugated peanut agglutinin (PNA) or dolichos biflorus agglutinin (DBA). DBA and PNA binding was absent or weak in fetal female and male germ cells, but became intensely positive in oocytes in the immediate postnatal period. The percentage of oocytes stained with DBA increased during the first three days after birth, and from day 3–4 onwards all oocytes were strongly labeled. I suggest that these changes in lectin binding reflect changes in biochemical structure of the oocyte surface related to differentiative events occurring in the mouse ovary immediately after birth.  相似文献

Germ cells produce sperm and eggs for reproduction and fertility. The Asian seabass (Lates calcarifer), a protandrous marine fish, undergoes male-female sex reversal and thus offers an excellent model to study the role of germ cells in sex differentiation and sex reversal. Here we report the cloning and expression of vasa as a first germ cell marker in this organism. A 2241-bp cDNA was cloned by PCR using degenerate primers of conserved sequences and gene-specific primers. This cDNA contains a polyadenylation signal and a full open reading frame for 645 amino acid residues, which was designated as Lcvasa for the seabass vasa, as its predicted protein is homologous to Vasa proteins. The Lcvasa RNA is maternally supplied and specific to gonads in adulthood. By chromogenic and fluorescent in situ hybridization we revealed germ cell-specific Lcvasa expression in both the testis and ovary. Importantly, Lcvasa shows dynamic patterns of temporospatial expression and subcellular distribution during gametogenesis. At different stages of oogenesis, for example, Lcvasa undergoes nuclear-cytoplasmic redistribution and becomes concentrated preferentially in the Balbiani body of stage-II~III oocytes. Thus, the vasa RNA identifies both female and male germ cells in the Asian seabass, and its expression and distribution delineate critical stages of gametogenesis.  相似文献

Degeneration of germ line cells in amphibian ovary. —Acta Zoologica (Stockholm) 91 : 319–327 We studied the morphology of degenerating ovarian follicles in juvenile and adult frogs Rana temporaria, Rana lessonae and Rana ridibunda. Degeneration of primordial germ cells was never observed and was extremely rare in oogonia and early oocytes in a cyst phase in juveniles. Previtellogenic oocytes were rarely affected. Three main types of atresia were identified. In type I (subdivided into stages A–D), vitellogenic oocytes are digested by proliferating follicle cells that hypertrophy and become phagocytic. A – germinal vesicle shrinks, nucleoli fuse, oocyte envelope interrupts, and follicular cells hypertrophy; B – follicular cells multiply and invade the oocyte; C – entire vesicle is filled by phagocytic cells; D – degenerating phagocytes accumulate black pigment. Type II is rare and resembles breakdown of follicles and release of ooplasm. In type III, observed in previtellogenic and early vitellogenic oocytes, ooplasm and germinal vesicle shrink, follicle cells do not invade the vesicle, and condensed ooplasm becomes fragmented. The residual oogonia in adult ovaries (germ patches) multiply, but soon degenerate.  相似文献

Halichoeres poecilopterus (Labridae) is a protogynous, hermaphroditic fish common in temperate waters around Japan. Although oocyte degeneration is an established event at the onset of sex change, there is little information on the process. We examined cell numbers and proportional ratios of ovarian germ cells in sex-changing gonads of H. poecilopterus. This confirmed a lack of YV (yolk vesicle stage) and Y (yolk stage) oocytes, a rapid decrease of P (perinucleolus stage) oocytes, and a slight increase of CN (before/at chromatin nucleolus stage) germ cells. We discuss gonadal conditions necessary for the protogynous sex change of H. poecilopterus.  相似文献

The aim of this study was to clarify the developmental and ultrastructual characteristics of oocytes grown in vitro from primordial germ cells. The female genital ridges at 12.5 days post coitus were cultured for 18 days on an insert membrane in Waymouth’s MB752/1 medium, supplemented with 15% fetal bovine serum and 1 mM sodium pyruvate; subsequently, the follicles isolated from the tissue were cultured for eight days in Waymouth’s medium supplemented with 5 ng/ml insulin, 5 ng/ml transferrin, 5 ng/ml selenium, 10 mlU/ml follicle stimulating hormone, and 100 ng/ml stem cell factor. The primordial germ cells developed in vitro into oocytes of more than 60 nm in diameter. The transmission electron microscopic analysis indicated that the oocytes, which developed in vitro, showed no obvious abnormality in their ultrastructure and had organelles appropriate for the oocyte size. However, a delay in the progressive changes of morphology in some of the organelles during oocyte growth was often found when comparing them to oocytes grown in vivo.  相似文献

Early in the spawning season female D. excentricus can be induced to spawn oocytes in the late stages of oogenesis. Observations with light microscopy indicate that pigment cells migrate in the jelly coat away from the surface of oocytes coincident with the final growth of the oocyte and its maturation. The pigment cells undergo a series of changes in shape and the oocyte elaborates arrays of long microvilli as the cells elevate from the surface of the oocyte. Transmission electron microscopy (TEM) observations of oocytes fixed within the ovary indicate that features of the sequence observed in the spawned oocytes normally take place within the ovary, prior to spawning. Extracts of the pigment cells induce a low level of germinal vesicle breakdown in asteroid oocytes. It is proposed that these cells are homologous to the follicle cells of other echinoderms and are involved in stimulating the maturation of oocytes.  相似文献

In Hirudo medicinalis and Haemopis sanguisuga, two convoluted ovary cords are found within each ovary. Each ovary cord is a polarized structure composed of germ cells (oogonia, developing oocytes, nurse cells) and somatic cells (apical cell, follicular cells). One end of the ovary cord is club-shaped and comprises one huge apical cell, numerous oogonia, and small cysts (clusters) of interconnected germ cells. The main part of the cord contains fully developed cysts composed of numerous nurse cells connected via intercellular bridges with the cytophore, which in turn is connected by a cytoplasmic bridge with the growing oocyte. The opposite end of the cord degenerates. Cord integrity is ensured by flattened follicular cells enveloping the cord; moreover, inside the cord, some follicular cells (internal follicular cells) are distributed among germ cells. As oogenesis progresses, the growing oocytes gradually protrude into the ovary lumen; as a result, fully developed oocytes arrested in meiotic metaphase I float freely in the ovary lumen. This paper describes the successive stages of oogenesis of H. medicinalis in detail. Ovary organization in Hirudinea was classified within four different types: non-polarized ovary cords were found in glossiphoniids, egg follicles were described in piscicolids, ovarian bodies were found characteristic for erpobdellids, and polarized ovary cords in hirudiniforms. Ovaries with polarized structures equipped with apical cell (i.e. polarized ovary cords and ovarian bodies) (as found in arhynchobdellids) are considered as primary for Hirudinea while non-polarized ovary cords and the occurrence of egg follicles (rhynchobdellids) represent derived condition.  相似文献

The molecular genetic mechanisms of sex determination are not known for most vertebrates, including zebrafish. We identified a mutation in the zebrafish fancl gene that causes homozygous mutants to develop as fertile males due to female-to-male sex reversal. Fancl is a member of the Fanconi Anemia/BRCA DNA repair pathway. Experiments showed that zebrafish fancl was expressed in developing germ cells in bipotential gonads at the critical time of sexual fate determination. Caspase-3 immunoassays revealed increased germ cell apoptosis in fancl mutants that compromised oocyte survival. In the absence of oocytes surviving through meiosis, somatic cells of mutant gonads did not maintain expression of the ovary gene cyp19a1a and did not down-regulate expression of the early testis gene amh; consequently, gonads masculinized and became testes. Remarkably, results showed that the introduction of a tp53 (p53) mutation into fancl mutants rescued the sex-reversal phenotype by reducing germ cell apoptosis and, thus, allowed fancl mutants to become fertile females. Our results show that Fancl function is not essential for spermatogonia and oogonia to become sperm or mature oocytes, but instead suggest that Fancl function is involved in the survival of developing oocytes through meiosis. This work reveals that Tp53-mediated germ cell apoptosis induces sex reversal after the mutation of a DNA–repair pathway gene by compromising the survival of oocytes and suggests the existence of an oocyte-derived signal that biases gonad fate towards the female developmental pathway and thereby controls zebrafish sex determination.  相似文献

The ultrastructure of the ovary of the amphilinidean cestode Amphilina japonica Goto & Ishii, 1936 from the body-cavity of the American sturgeon Acipenser transmontanus Richardson is described using transmission electron microscopy. The characters of the ovary of Amphilina japonica are different from those of all other cestodes. The most important difference is in the nature of the relationship between the germ and accessory cells within the ovary. In A. japonica the oocytes and accessory cells form numerous different intercellular contacts (desmosome-like junctions and zonulae adherentes). Gap junctions are present between the narrow cytoplasmic processes of the accessory cells. Numerous micropinocytotic vesicles and vacuoles from the accessory cells discharge their content into spaces between the oocytes and the accessory cells. The accessory cells are closely associated with the oocytes during the early and middle stages of oogenesis. As the volume of oocytes increases, the accessory cells gradually lose their association with the oocyte surfaces. Peripherally located individual accessory cells of A. japonica give rise to a cellular epithelial layer of irregular shape and thickness which breaks down via numerous invaginations of the basal membrane and underlying basal matrix. The different arrangements of the interconnection of cell components in the Amphilinidea compared with the Gyrocotylidea and Eucestoda (the absence of specialised cell contacts and the syncytial nature of the accessory ‘interstitial’ cells) are evidence suggesting the presence of unrelated groups within the Cestoda. The nature of the association of the accessory and germ cells in ovary of A. japonica more closely resembles the ovary of non-platyhelminth invertebrates rather than that of other neodermatans.  相似文献

Cytological status of hepatocytes in respect to seasons influencing ovarian activities has been studied in Mastacembelusarmatus (Hamilton). Histologically, the hepatocyte of female liver was provided with distinct nucleus and basophilic cytoplasmic granules. In M. armatus different germ line cells were recognized on the basis of size and histoarchitecture of the cells. By examining seasonal changes in relation to hepatocytes and ovary, it was found that during growth and maturation phases the density of cytoplasmic granules of hepatocytes was increased in number as well as the nucleus become hypertrophied. These features were well correlated with the occurrence of cortical alveolus and yolk granule stage in the ovary. During spawning phase the cytoplasmic granules were sparse in the hypertrophied hepatic cells. This was due to the dynamic cytological activities like vitellogenesis and occurrence of mature oocytes in the ovary. However, no significant alterations were noticed in the hepatocytes during post spawning phase due to the appearance of new germ line cells in the ovary. Cytological changes in the hepatocytes were correlated with the reproductive phases in female M. armatus.  相似文献

 The Zfcth1 gene is, as the previously cloned carp cth1 gene, related to the mammalian TIS 11 family of primary response genes and encodes a protein with two putative CCCH zinc fingers. This report describes the RNA expression of this gene during oogenesis and early embryogenesis up to gastrulation in the zebrafish (Danio rerio). Maternal cth1 message is present in the ovary of 1-month-old fish and of adult fish in oocytes at all stages of maturation. In the youngest oocytes the message is localized in the cytoplasm all around the nucleus, in larger oocytes the message becomes restricted to the future animal pole of the embryo, and in mature oocytes the expression is sharply localized in the cortical layer under the micropyle. After ovulation the cth1 messenger spreads over the cytoplasmic cap and is distributed over the blastomeres during subsequent cleavages. In subsequent stages maternal expression of cth1 gradually disappears. From early epiboly stages onward embryonic cth1 expression is localized to the germ ring and the hypoblast cells in the central part of the embryonic shield. In the shield, cth1 expression largely overlaps with the area of gooscoid expression in the first involuting cells. In stages after 70% of epiboly cth1 expression diminishes and soon can no longer be detected in the embryo. The aim of the present study is to describe the organization of the ovary and mode of oogenesis at the ultrastructural level in two representatives of Lumbriculida – Lumbriculus variegatus and Stylodrilus heringianus. In both species studied, the ovaries are small and conically shaped structures that are attached to the intersegmental septum via a thin ligament. The ovaries are composed of germline cysts formed by germ cells interconnected by stable cytoplasmic bridges. As a rule, the cyst center is occupied by a poorly developed anuclear cytoplasmic mass, termed a cytophore, whereas the germ cells are located at the periphery of the cyst. Germline cysts are enveloped by somatic cells. The ovaries of the species studied are polarized, i.e., along the long axis of the ovary there is an evident gradient of germ cell development. The data obtained suggest ovary meroism, i.e., two categories of germ cells were found: oocytes, which continue meiosis, gather nutrients, grow and protrude into the body cavity, and nurse cells, which do not grow and are supposed to supply oocytes with cell organelles and macromolecules via the cytophore. The ovary structure and mode of oogenesis in the species studied were compared with those of other clitellate annelids. As a rule, in all clitellates studied to date, the ovaries are composed of germline cysts equipped with a cytophore and associated with somatic cells; however, the ovary morphology differs between taxa regarding several quantitative and qualitative features. The ovary organization and mode of oogenesis in L. variegatus and S. heringianus strongly resemble those found in Tubificinae and Branchiobdellida studied to date. Our results also support a sister-group relationship between Lumbriculida and a clade comprising ectoparasitic clitellates (i.e., Branchiobdellida, Acanthobdellida and Hirudinida) with Branchiobdellida as a plesiomorphic sister group to Acanthobdellida and Hirudinida.  相似文献

The aim of the present study was to know the cyclic changes in gonadal maturation and to investigate the various developmental stages of oocytes and testicular germ cells of snow trout, Schizopyge niger. A total of 316 fish specimens were procured from three sites of river Jhelum during November 2016 to October 2017 and the histological analysis of their gonad was carried out. The results of the present study indicates that the male population was found significantly (p < 0.05) higher compared to female population and were reported as (1.6:1; M:F). The highest mean GSI value was recorded in male (12.24 ± 0.89) during the month of March and for female (13.74 ± 0.92) also in the month of March, which indicated that the gonadal development reached its peak during this month. The highest mean oocyte diameter was 1.04 ± 0.20 mm observed in March, showing that the oocyte reached its maturity during this month. Histological study of ovary revealed the evidence of vitellogenic yolk granule stage and ripe mature stage from December to April. While in case of male, four stages of spermatogenesis were distinguished and spermatozoa were highly abundant in the months of March and April. Mean ± SE of absolute fecundity was 3,224 ± 421 eggs and also, the relative fecundity was 69 egg/g body weight. Based on the results of GSI and the analysis of gonad histology, it was observed that the peak breeding season of S. niger occurred in the month of March. It was also noted that spawning period of this fish begins in the month of February and lasts till April with a peak breeding season in the month of March. The present study will provide a baseline information about the appropriate peak breeding of the fish, which will be useful for artificial breeding as well for prevention of fishing from natural habitat.  相似文献

The morphological characteristics of Synbranchus marmoratus female germ cells in various development stages were described in details; then measurements of ovarian follicle diameters were taken from primary and secondary growth as during these development stages the oocyte size varied considerably along the fish growth. The results were correlated to total fish length, using the individuals division in six size classes. It was possible to group oocytes by stages according to histological characteristics but not according to morphometric diameter, as there was a wide variation in diameter in each stage and overlap between different maturation stages. These data make available new information on the reproductive biology of Synbranchidae.  相似文献
