Plant virus diseases cause major losses in agricultural and horticultural products, especially in tropical and subtropical regions. The first step to manage these diseases is detecting, identifying and determining the pathogen characteristics. Cucumber mosaic disease is one of the most prevalent and damaging plant diseases in the world which is caused by Cucumber mosaic virus (CMV). Each year, this virus causes yield decreasing and substantial economic damages in its host plants worldwide including the north of Iran. In order to study and identify CMV, 935 leaf samples were collected based on typical symptoms from 10 crops (tomato, pea, tobacco, soybean, watermelon, broad bean, squash, cucumber, eggplant and lettuce) in different regions of Golestan and Mazandaran provinces (north of Iran) during 2009–2010. Suspicious samples were analysed by DAS-ELISA and polyclonal antibodies. The results showed that 275 samples (29.4%) were infected by CMV. Between these hosts, the highest and the lowest CMV infection was associated to watermelon (62.44%) and lettuce (0%), respectively. Among sampling locations, Behshahr (100%) and Minoodasht (3.47%) showed the maximum and minimum infection, respectively.  相似文献

Micropropagated bananas derived from Banana Bunchy-Top Virus (BBTV) infected plants, but displaying no symptoms of the disease, were established in the field. They were grown for three years and produced a plant crop and ratoon crops. No disease symptoms were observed. There was uncertainty as to whether
–  micropropagation eliminated the virus from the material,
–  the plants were symptomlessly infected, or
–  the plants were somaclonal variants that suppress symptom expression.
To resolve this question, suckers from these plants and non-infected controls were established in pots and infected with aphids, Pentalonia nigronervosa Coq, collected from BBTV-infected plants. All plants produced characteristic Banana Bunchy-Top Disease (BBTD) symptoms in five months. In addition, samples collected from the original symptomless field plants were tested by DAS-ELISA using a polyclonal antibody specific for BBTV. No detectable levels of virus were found in any of the samples. These studies support the view that the symptomless plants were virus free and not symptomlessly infected or BBTV-resistant somaclones generated from micropropagation. Nevertheless there continues to be a need for caution in dissemination of micropropagated bananas, especially when obtained from regions where BBTV is known to occur.  相似文献

Eight sweet pepper plant samples showing viral and viral like symptoms were collected from open field and used for detecting viral infections through biological, serological and biochemical methods. DAS-ELISA, DBIA and TPIA have relative effectiveness for detecting parenchymal viruses (CMV, TMV and PVY) and vascular virus (TYLCV), and the DAS-ELISA and TPIA are found more efficient (87.5%) than DBIA (78.1%). The examined leaf samples were found co-infected with different mixed types of viruses including (CMV, TMV, PVY and TYLCV), (CMV, PVY and TYLCV), (TMV, PVY and TYLCV) and (TMV and TYLCV) that enhanced different degrees of severe external symptoms. There are 2 out of 8 samples infected with Phytoplasma sp. by Diene’s stain and PCR using generated 16S rDNA gene primer with expected amplicon size of 680?bp. The co-infections with various viruses and phytoplasma has 12.5% frequency that reduced the levels of protein content, peroxidase and polyphenol oxidase activity quantitatively and qualitatively in 2 samples in comparison with other mixed categories. The sweet pepper plant can be considered as a reservoir for parenchymal and vascular viruses and Phytoplasma sp. due to the synergistic and antagonistic effects causing unusual and unpredictable biological and epidemiological, viral and viral-like via host biochemical effects.  相似文献

Since April 2010, Tembusu virus (TMUV) which is a contagious pathogen of waterfowls, causing symptoms of high fever, loss of appetite and fall in egg production, has been reported in east of China. A double antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) which detects for TMUV was developed, using two monoclonal antibodies (mAbs) against the TMUV envelope (E) protein. BALB/c mice were immunized with purified recombinant E protein expressed in E. coli. Three hybridoma cell lines designated as 12B1, 10C6 and 2D2, were screened by cell fusion and indirect ELISA for their ability to recognize different linear epitopes on the E protein, and were characterized subsequently. High-affinity mAbs 12B1 and 2D2 were used as capture and detection antibodies, respectively. The reaction conditions for the DAS-ELISA were optimized for TMUV detection. The cross-reactivity of the DAS-ELISA was determined using TMUV, duck plague virus, avian influenza virus subtype H9, Newcastle disease virus, duck hepatitis A virus type 1 and duck reovirus samples. A total of 191 homogenized tissues of field samples were simultaneously detected by DAS-ELISA and by RT-PCR. The former was found to have a high specificity of 99.1% and a sensitivity of 93.1%. These results reveal a positive coincidence between DAS-ELISA and RT-PCR at a coincidence rate of 95.8%. The method developed in this study can be used for the diagnosis of TMUV infection of duck origin.  相似文献

During surveys carried out in 2008 in the nurseries of some ornamental and medical plants, about 90 plant samples belonging to six plant species were collected. Cucumber mosaic virus (CMV) was detected by routinely double antibody sandwich-enzyme linked immunosorbent assay (DAS-ELISA) in most tested plants. In Vinca rosea, Ocimum basilicum and Pelargonium sp., which reacted positively for CMV, 100% of the samples were infected. High absorbance values were obtained when extracts were prepared from these species and then examined by DAS-ELISA. The results indicated that CMV concentration on O. basilicum is greater than 50 μg ml?1 when compared with purified CMV standards. High absorbance values occur even under conditions fully unsuitable for the detection of antigens. This result suggested that the strong ELISA absorbance values were nonspecific reactions with materials in plant extracts. So, other detection methods including dot blot, Western blot and bioassay was used for comparing with ELISA test. The nonspecific reactions were substantiated when CMV was not detected by dot blot and Western blot. Also, infectious CMV was not detected in bioassay involving inoculation of extracts onto Chenopodium amaranticolor and Nicotiana tabacum var. White Burley plants as indicator hosts. Addition of bovine serum albumin (BSA) or polyvinylpyrolidene (PVP) to extraction buffer or to wells after IgG coating for DAS-ELISA reduced nonspecific reactions. Finally, CMV was isolated from V. rosea symptomatic plants, which give a positive reaction by DAS-ELISA, dot blot, Western blot and bioassay. Lettuce mosaic virus (LMV) Cucumber mosaic virus (CMV) and Tomato spotted wilt virus (TSWV) were identified in fields of Tehran province. In this study 452 leaf samples were collected from the fields throughout the Tehran province during 2002 and 2003. Distribution of Lettuce mosaic virus (LMV), Cucumber mosaic virus (CMV), Tomato spotted wilt virus (TSWV)and Arabis mosaic virus (ArMV) was determined with DAS-ELISA. Percentage of single Infection to LMV. CMV or TSWV was 20.58, 15.93 and 9.96% respectively. Also 15.28% of samples were co- infected with LMV+CMV, 8.19% with LMV+TSMV and 7.74% with CMV+TSWV. 4.65% of samples were Infected to all of these three viruses. LMV was found in 48.69%, CMV in 43.59% and TSWV in 30.54% of samples totally. Therefore LMV is major dominant agent of lettuce mosaic disease in Tehran province. This is the first report of occurrence of TSWV on lettuce in Iran and first report of CMV and LMV in Tehran province.  相似文献

Squash (Cucurbita pepo) belongs to Cucurbitaceae family. Every year Cucurbitaceae are planted world wide. They are one of the most important economic crops. Cucurbitaceae are threatened by viruses. Many viruses damage the plants of this family. Since nine viruses have been reported on squash from Iran. In this survey, during 2002--2003, to determine the distribution of Cucumber mosaic virus (CMV), Zucchini yellow mosaic virus (ZYMV) and Watermelon mosaic virus (WMV), 466 samples were collected from squash field in Tehran province. Infected plants showing symptoms such as: mosaic, yellowing, deformation, shoestring of leaves and fruit deformation and yield reduction. Distribution of CMV, ZYMV and WMV were determined by DAS-ELISA. Thepercentage of ZYMV, WMV and CMV were 35.6, 26.1 and 25.1% respectively. Triple infection (CMV+ZYMV+WMV) were found in 6.4% of samples. ZYMV were found the most frequently the viruses. This is the first report of WMV on squash in Tehran province.  相似文献

ProjectGolestan province, located in northeast of Iran, has been known as a high risk area for esophageal cancer (EC). This study was conducted to assess the relationship between soils selenium (Se) level and development of EC in this region.ProceduresIn this ecological study, 135 blocks were identified in Golestan province based on geographical altitude and longitude on the map. One soil sample was collected from the center of each block. Then we investigated Se concentration in soil samples by flame atomic absorption spectrometry. Statistical analysis was performed by the Pearson correlation test and Student t-tests. P-values of less than 0.05 were considered as significant.ResultsThe mean±SD of soils Se level in Golestan province was 3.7±1.61 mg/kg. There was a positive correlation between soils Se level and EC rates in this area (P=0.03) (Pearson correlation coefficient=0.19). Soils Se concentration was significantly higher in high (4.13 mg/kg) than in the low (3.39 mg/kg) EC rate areas (P=0.01).ConclusionsWe found high soils Se concentration and a significant positive relationship between soils Se level and EC rate in Golestan province of Iran. So, high soils Se level may play a possible role in developing EC in this area, specifically in Turkmensahra (very high EC rates).  相似文献

A purification procedure, which yielded up to 15–30 mg of beet yellows virus (BYV) per 100 g of infected Tetragonia expansa leaves, has been developed. The procedure included sap clarification with Triton X-100, and two cycles of ultracentrifugation through sucrose cushion, which contained PEG-6000 and NaCl. A specific antiserum was prepared, and BYV infection was successfully detected by the double-antibody sandwich (DAS) ELISA in infected sugar beet leaves and roots diluted up to 1 × 105 and 1 × 104, respectively. The virus concentration was demonstrated to decrease in infected sugar beet roots slowly during 7 months, thus allowing successful diagnosis of planting material in winter storage. BYV presence in Myzus persicae aphids was also reliably detectable using the DAS-ELISA. In a competitive DAS-ELISA test, the Ukraine and the British BYV isolates were found serologically indistinguishable.  相似文献

A virus was isolated from squirting cucumber (Ecballium elaterium L.) collected in France (Alpes de Haute Provence). After mechanical inoculation, eight species belonging to the Cucurbitaceae were found susceptible to this virus with systemic symptoms of mosaic, chlorotic spots, and fruit deformations. The French ZYFV (ZYFV-Fr) isolate differed from the type strain in its ability to infect some differential host plants. Elongated flexuous particles with a modal length of 752 nm were observed by electron microscopy in leaf extracts. Cytoplasmic inclusions similar to those associated with infections by members of the potyvirus group were observed by light microscopy. The virus was found by SDS-immunodiffusion and DAS-ELISA to be serologically related but distinct from the type strain of zucchini yellow fleck virus from Italy (ZYFV-lt). ZYFV has not yet been found in cultivated cucurbit plants in France; this virus appears to be restricted to squirting cucumbers in a few localities. A survey was conducted in Al-Kharj governorate, Riyadh region to identify viruses causing variety of virus-like symptoms on tomato plants. A total of 135 samples were collected from symptomatic tomato plants. Symptoms included mottling, deformation, necrosis of leaves and fruits. Eighteen viruses were tested by DAS-ELISA. Tomato black ring virus (TBRV) was the virus of concern as it was not detected in Saudi Arabia before and was detected in 52.6% of the collected samples in this study. RT-PCR was used to confirm detection of TBRV and to sequence the amplified products to determine molecular characteristics of this virus. In the host range test study that was performed using a purified isolate of TBRV, sixteen out of the twenty two tested plants showed symptoms. Brassica oleracea was not infected by this virus. Gel electrophoreses (2% agarose) yielded fragments of 978 bp of coat protein gene of TBRV. Nucleotide sequences of purified RT-PCR products for three TBRV Saudi isolates were deposited in the GenBank with the following accession numbers MT274656, MT274657, and MT274658. These isolates of TBRV indicated a close Phylogenetic relationship of (99–100%) among themselves and with five isolates from Poland (95–98%) but a distant relationship of 85% with isolates from England and Lithuania deposited in the GenBank. This is the first report for detection and molecular characterization of TBRV infecting tomato plants in Saudi Arabia.  相似文献

The result of the Leptosphaeria maculans/Brassica napus interaction is usually assessed by symptom scoring following a cotyledon-inoculation test. However, an early evaluation of the interaction, and reliable quantitaive data of fungal growth inside plant tissues are needed to supplement the visual assessment of the symptoms. For this purpose, we developed a quantitatve double antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) using rabbit polyclonal antisera directed against soluble mycelial proteins. The specificity of the serum was first assessed by immunoblotting following isoelectric focusing of soluble proteins (Western blot) and by DAS-ELISA. Except for Alternaria brassicae, no cross-reactions were observed with my celial extracts of saprophytes or pathogens of B. napus following DAS-ELISA. Although Tox+ and Tox0 isolates of L. maculans were unequivocally discriminated by Western blot, they were quantitatively indistinguishable following ELISA, thus enabling us to analyse a wide range of L. maculans isolates in planta. The detection limit of the assay was less than 10 ng of fungal proteins per ml of plant extract. For a given isolate, time-course studies showed that fungal growth in cotyledons was correlated with symptom scoring. In the case of hypersensitive response, only 34% of the plants were ELISA-positive, and these plants never contained more than 10 ng of fungal protein per cotyledon. In contrast, in the cases, of susceptibility, 100% of the plants were ELISA-positive and fungal protein content was higher than 10 μg per cotyledon. Moreover, significant differences in ability to colonize the tissues were observed among Tox+ isolates. Finally, using the ELISA quantification, intermediate symptoms could be differentiated as lateresistance responses or susceptibility.  相似文献

Occurrence and distribution of cassava begomoviruses in Kenya   总被引:1,自引:0,他引:1  
A survey for cassava mosaic disease (CMD) was conducted in Kenya, to investigate the factors contributing to the generally increased incidence and severity of CMD in the cassava growing regions and to study the distribution of the disease's causal begomoviruses, African cassava mosaic virus (ACMV) and East African cassava mosaic virus (EACMV) and their strains. Special emphasis was given to the occurrence of the destructive recombinant Uganda variant strain of EACMV (EACMV-UG2). Samples from 91 farmers' fields in the main cassava-growing areas of coastal and western Kenya were collected and subjected to ELISA and PCR for detection and typing of the begomoviruses present. CMD incidence was highest in western Kenya (80–100%) and lowest in the Coast province (25–50%). In Western and Nyanza provinces, 52% of the samples tested contained EACMV-UG2, 22% ACMV and 17% contained both ACMV and EACMV-UG2. EACMV was found in four cases at different sites. In cassava samples from the coast province, only EACMV with DNA-A sequences similar to EACMV strains present in Kenya and Tanzania was found. East African cassava mosaic Zanzibar virus (EACMZV) was present in several farms in the Kilifi district. In 15% of all cassava samples with CMD symptoms, flexuous, filamentous virus-like particles were also found, providing evidence for a more complex virus situation in cassava grown at the Kenyan coast. In western Kenya, where intense cassava cultivation takes place, CMD is rampant and EACMV-UG2 was found in mixed virus infections with ACMV driving the epidemics. In coastal areas, where farms are scattered and in isolation, EACMV is endemic, however, with a lower disease incidence and with a limited impact to cassava production.  相似文献

Background: Golestan province in northeastern Iran has been known as a high-risk area for esophageal cancer (EC). This study was conducted to assess aflatoxin (AF) contamination of wheat flour (WF) samples in high and low EC-risk areas of Golestan province. Methods: Four WF samples were collected randomly from each of 25 active silos throughout the province in 2009. The levels of AFs were measured using the High-performance liquid chromatography method. Using the data of EC rates obtained from Golestan population-based cancer registry, the province was divided into high and low risk areas for EC. Student t-test and multivariate regression analysis were used to compare the levels of aflatoxins as well as the condition of silos between the two areas. Results: One hundred WF samples were collected. The mean levels of total aflatoxin and aflatoxin B1 was 1.99 and 0.53 ng g?1, respectively. The levels of total AF (p = 0.03), AFG2 (p = 0.02) and AFB1 (p = 0.003) were significantly higher in samples obtained from high risk area. Multivariate regression analysis showed that humidity of silo was the most important source of difference between silos of the two areas (p = 0.04). Conclusion: We found a positive relationship between AF level of WF samples and the risk of EC. So, AF contamination may be a possible risk factor for EC in our region. We also found that humidity of silos was the most important determinant of AF contamination of WF. Intensive control of silos conditions including humidity and temperature are needed especially in high EC-risk areas.  相似文献

An intensive survey was conducted to identify virus diseases affecting pea crops in Tehran province of Iran. A total of 270 pea samples were collected randomly from pea fields. samples were tested by Double Antibody Sandwich Enzyme Linked Immunosorbent Assay (DAS-ELISA) using polyclonal antisera prepared against PSBMV (AS-0129, DSMZ, Braunschweig, Germany) and TSWV (AS-0580, DSMZ, Braunschweig, Germany). Virus disease incidence in pea samples was followed by PSBMV (33%) TSWV (24.4%) and PSBMV+TSWV (17.77). The positive samples with PSBMV were extracted in 0.05M phosphate buffer pH 6.5-7 containing 2% pvp and inoculated on Pisum sativum, Vicia faba, Chenopodium quinoa, Chenopodium amaranticolor. That produced in Pisum sativum; leaflets roll downwards, shoots curl, internodes shorten and plants are rosetted. Early infections reduce flower and fruit formation or eliminate their development. Broad bean has symptoms accompanied by a certain margin rolling and leaflet distortion. In Chenopodium amaranticolor necrotic local lesions and Chenopodium quinoa chlorotic local lesions had produced. The positive samples with TSWV were extracted in 0.01 M phosphate buffer containing 1% Na2 SO3 and inoculated on Petunia hybrida, Pisum sativum. TSWV causes several symptoms in infected peas, including brown leaf petiole and stem coloration, leaflet spotting, vein necrosis. In petunia hybrida after approximately 5 days showed local necrotic lesion. Biological purification in TSWV with chlorotic local lesions in Petunia hybrida and in PSBMV; chlorotic local lesions in Chenopodium quinoa were done. In PSBMV, back inoculated on Pisum sativum and Vicia faba also tested with DAS-ELISA. RT-PCR confirmed the results. This is the first report of PSBMV and TSWV naturally infecting pea in Iran.  相似文献

IntroductionBreast cancer is the most frequent cancer among women worldwide. Breast cancer incidence in young women is a health issue of concern, especially in middle-income countries such as Iran. The aim of this study is to report the breast cancer incidence variations in Golestan province, Iran, over a 10-year period (2004–2013).MethodsWe analyzed data from the Golestan Population-based Cancer Registry (GPCR), which is a high-quality cancer registry collecting data on primary cancers based on standard protocols throughout the Golestan province. Age-standardized incidence rates (ASRs) and age-specific incidence rates per 100,000 person-years were calculated. Time trends in ASRs and age-specific rates were evaluated using Joinpoint regressions. The average annual percentage change (AAPC) with correspondence 95% confidence intervals (95%CIs) were calculated.ResultsA total of 2106 new breast cancer cases were diagnosed during the study period. Most cases occurred in women living in urban areas: 1449 cases (68%) versus 657 cases (31%) in rural areas. Statistically significant increasing trends were observed over the 10-year study period amongst women of all ages (AAPC = 4.4; 95%CI: 1.2–7.8) as well as amongst women in the age groups 20–29 years (AAPC = 10.0; 95%CI: 1.7–19.0) and 30–39 years (AAPC = 5.1; 95%CI: 1.4–9.0).ConclusionThe incidence of breast cancer increased between 2004 and 2013 in Golestan province amongst all age groups, and in particular amongst women aged 20–39 years. Breast cancer should be considered a high priority for health policy making in our community.  相似文献

Sunflower is one of the most important oilseed crops, which is grown in many countries. Increasing demand to use sunflower oil has expanded the under cultivation area throughout the world. Sunflower viruses have been reported as one of the pathogens that reduce the quantity and the quality of this product in a number of countries. In our research to study the occurrence and distribution of viruses in sunflower fields of Iran, 562 samples were collected from different fields in Kerman and Isfahan provinces during growing seasons from 2009 to 2011. Potato virus Y (PVY), Cucumber mosaic virus (CMV) and Tomato spotted wilt virus (TSWV) were detected by double antibody sandwich enzyme-linked immunosorbent assay. Also, numbers of samples were positive for infection to potyviruses (except PVY) in antigen-coated-plate enzyme-linked immunosorbent assay test using potyviruses general antiserum. Infected plants had symptoms like mosaic, yellowing, deformation, necrotic and chlorotic lesions and mottling on leaves and stunting. The infection rates of potyviruses, PVY, CMV and TSWV in Isfahan province were 33, 22, 4.18 and 3.25% of collected samples, respectively. The corresponding rates for samples from Kerman province were 15, 5, 0.8 and 0.4% of collected samples. According to these results, viral infection in Isfahan province was more than surveyed in Kerman province during the mentioned period of three years. Furthermore, generally there was a decrease in the percentage of viral infection during this three growing seasons in both provinces. This is the first report of the detection of CMV and TSWV in sunflower fields of Isfahan and Kerman and first report of PVY in Isfahan.  相似文献

With the aim of understanding virus movement and fluctuations in the virus concentration in bulb and leaves of onion (Allium cepa L.) plants after infection, Onion yellow dwarf virus (OYDV) was analysed by double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA). OYDV concentrations were higher in onion leaves of plants grown from tested bulbs compared with bulbs, although the virus was successfully detected in bulb of onion.  相似文献

Musa landraces (469) from 73 villages in 10 states in 1999, 2000 and 2004 in southern Nigeria were visually assessed to determine virus symptoms associated with CMV disease. CMV indexing was done using dot-blot immunoassay and ELISA. Monoclonal antibodies were used to group CMV isolates in 2004. Symptoms of interveinal chlorosis and chlorotic streaks were observed in 22.4 and 17.9% of the Musa samples with CMV disease incidences of 17.1% and 14.7%, respectively. Other virus symptoms accounted for between 1.5 and 12.2% of the total leaf samples while 13.9% were asymptomatic. CMV incidence in asymptomatic leaf samples was as high as 10.4% while the virus incidence in the other symptom types were between 0.9 and 7.9%. The incidences of CMV in both the symptomatic and asymptomatic plants in 1999, 2000 and 2004 were between 1.3 and 18.7%, 1.9 and 26.9% and 4.9 and 26.4%, respectively.  相似文献

1818 collected samples of potato plant showing virus infection symptoms from 85 fields were tested for PVS infection using DAS-ELISA. Average of infection to this virus varied from 0 to 100%. Least infection was belonging to fields with new introduced varieties. On the other hand native and old introduced cultivars showed heavy infection. In field condition, PVS infected plants didn't show very obvious symptoms, so some infected plants may be missed in field sample collecting. The physical properties of 3 isolates, Avaj, Stanboly and Agria No 15 were determined. TIP 55-60 degrees C, DEP 10(-3) and Liv measured 3-4 days. Ouchterlony agar double diffusion test using SDS was useful for virus detection and precipitation lines didn't show any spur between isolates, although isolates differs slightly in symptomatology. SDS-Page and Western blotting methods used successfully for virus detection and determining and measuring viral protein components.  相似文献
