The life histories of the pond smelt Hypomesus nipponensis collected from Japanese fresh waters and brackish (sea) waters were studied by examining the strontium (Sr) and calcium (Ca) concentrations in their otoliths. The Sr:Ca ratios in the otoliths changed with the salinity of the habitat. The pond smelt living in a freshwater environment showed consistently low Sr:Ca ratios throughout the otolith, averaging 1·2–1·3 × 10−3. These samples were identified as a standard freshwater type. In contrast, fish collected from the intertidal zone showed higher otolith Sr:Ca ratios than those in the standard freshwater type, and the ratios fluctuated along the growth phase. In addition to the two representative life‐history types of H. nipponensis , i. e . freshwater and anadromous life‐history types, other pond smelts were found to have an estuarine resident life history‐type with no freshwater phase, indicating that the pond smelt has a flexible migration strategy with a high degree of behavioural plasticity and an ability to utilize the full range of salinity in its life history.  相似文献

A significantly higher concentration of testicular spermatozoa was obtained from freshwater Oreochromis mossambicus (9·9×109 spermatozoa ml−1) than seawater O. mossambicus (4·6×109 spermatozoa ml−1). The mean osmolality of the urine of freshwater fish (78·5 mOsmol kg−1) was significantly different from that of seawater fish (304·8 mOsmol kg−1). The mean length of the mid-piece of the spermatozoa together with the tail was more variable in freshwater O. mossambicus (8·80±0·23μm) than in seawater specimens (8·27±0·18 μm). Stripped sperm of freshwater O. mossambicus was highly contaminated by urine which was a good activator of sperm motility in O. mossambicus held in both fresh and sea water. The osmolality for initiation of motility in freshwater O. mossambicus spermatozoa was from 0 to 333 mOsmol kg−1 while for seawater O. mossambicus spermatozoa it was from 0 to 1022 mOsmol kg−1. The optimum osmolality for motility was from 70 to 333 mOsmol kg−1 for freshwater O. mossambicus spermatozoa and from 333 to 645 mOsmol kg−1 for seawater fish. In freshwater O. mossambicus spermatozoa, the presence of 20 mM CaCl2 increased the permissive osmolality of NaCl from 184 to 645 mOsmol kg−1. For seawater O. mossambicus spermatozoa, solutions of NaCl devoid of CaCl2 were unable initiate motility, but the addition of 1·5 to 30 mM CaCl2 to the NaCl solution (0–934 mOsmol kg1) had a full motility initiating effect.  相似文献

A toxicity test with cadmium concentrations ranging from 0·1 to 100·0 mg Cd 1−1 was used to assess the effect of cadmium pretreatment on rainbow trout ( Salmo gairdneri Richardson) alevins. The median period of survival for fish pretreated at 0·01 mg Cd 1−1 was found to be increased at test concentrations up to 10mg Cd 1−1 compared with alevins pretreated with dilution water. However, at concentrations above 10mg Cd 1−1 pretreatment at 0·01 mg Cd 1−1 reduced the median period of survival.  相似文献

Abstract— The content of ATP was measured in neurons from dorsal root ganglia and in cortical and spinal astrocytes with the luciferase method. No ATP could be demonstrated in the cortical cells but in the dorsal root neurons and spinal astrocytes an average amount of 7·9 and 11·9 × 10−15 mol per cell respectively was found. An increase in the external potassium concentration had no effect on the content of ATP in neurons (7·1 × 10−15 mol/cell) but led to a significant ( P < 0·005) decline to 6·4 × 10−15 mol per cell in the astrocytes.  相似文献

The amylase-producing ability of the intestinal microflora in cultured specimens of ayu, carp, channel catfish, Japanese eel and tilapia was determined. Mean viable counts of aerobes and anaerobes ranged from 1·1×106 to 3·7×108 cfu g−1 and from 1·3×103 to 1·6×108 cfu g−1, respectively. Aeromonas spp. and Bacteroidaceae were predominant in four to five fish species. Of 206 strains examined, 65 (31·6%) produced ≥0·01 U amylase ml−1. The percentage of producers differed among families and genera of bacteria and fish species. While 56% of the anaerobes produced amylase, only 20% of the aerobes did. More than 50% of Aeromonas , Bacteroidaceae and Clostridium strains produced amylase efficiently while Acinetobacter , coryneforms, Enterobacteriaceae, Moraxella , Plesiomonas and Streptococcus strains did not. High amylase production (≥0·05 U ml−1) was found in 12 strains, 11 from Aeromonas and one Pseudomonas . The percentage of high amylase producers in Japanese eel was lower than the other four fish (2–30%). These results strongly suggest that the amylase produced by the intestinal microflora play an important role in the digestion of starch in freshwater fish to some extent.  相似文献

The acute effects of a herbicide based on 2,4-D on embryos and larvae of bleak, Alburnus alburnus L. exposed to concentrations of 25–3200 mg 1−1 for 12–48 hours were investigated. The 2,4-D delayed or stopped the development of embryos in their early stages, caused behavioural changes and morphological alterations. The L.C.50 values for embryos varied between 12–9 and 15·9–4mg 1−1 and for larvae from 51·6 to 111·2mg 1−1 according to the time of exposure. The level of 2,4-D in Lake Balaton, or similar shallow lakes, without substantial injury of the littoral fish fauna should not surpass 0·5–1·0 mg 1−1.  相似文献

This study describes a broad host transformation protocol that enables the uptake of plasmid DNA into 10 different species of Bifidobacterium , some of which have never been transformed before. The vector pNC7 (4·9 kb) was used to optimize the electroporation protocol. Transformation efficiencies ranged from 3·6×10−1 to 1·2×105 transformations per μg DNA. The impact of growth medium composition and electric field strength on transformation efficiency were independently optimized. Electrocompetent cells were grown in Iwata medium broth enriched with ActilightRP 16%, harvested during the early exponential growth phase, and pulsed at 12·5 kV cm−1, 100 Ω and 25 μF.  相似文献

Standard lengths of cyprinids at several sites in the River Great Ouse, England, were compared between catches taken using a conventional 15 × 3 m micromesh seine (pore diameter 2 mm) and using point sampling by electrofishing (PSE). No statistically significant differences in sites were found in six out of 10 trials. In nine out of 10 trials, the difference between the mean lengths differed by <1 mm. No serial bias was found between PSE and seine netting for cyprinid fishes between 14 and 100 mm SL, although variation between size classes was high, owing to small sample sizes. The coefficient of variation in fish length with size tended to increase with the age of 0+ Rutilus rutilus . The relationship between sample size (n) and variance ( s2 ) was explained by s 2=13·9 n −1·24. Sampling more than 30 fish resulted in little increase in precision. The relationship between the mean catch per site (̄) and the variance of the mean estimate was log s 2=1·6039 log ̄ +0·973. The number of samples required to estimate density within a given variance range was: n =9·33 ̄ 1·6–2 CV̄ −2. Given the high variance-mean ratio, great care should be taken when interpreting density data collected using PSE, and 50 point samples is the minimum required for density estimation.  相似文献

During the auxin-sensitive phase of root initiation, rates of 3-indolyl- [2-14C] acetic acid (IAA) uptake into the 1 cm bases of shoots of the apple rootstock M.9 ( Malus pumila Mill.) 'in vitro' were not significantly affected by the presence of 10−3 M phloroglucinol (PG) using either liquid or agar-solidified media. The use of a liquid medium did however reduce rates of uptake over a 10-day period of auxin application. The distribution of labelled IAA between the 1-cm base and the shoot remainder was not affected by PG.
Exposure of shoots of the difficult-to-root M.9 and the easy-to-root M.26, to 2.8 × 10−5 M IAA containing [2-14C] IAA revealed no positive correlation between the amount of label taken up by the 1-cm base and rooting performance. M.9 bases absorbed almost twice as much label as M.26 after 9 days but had produced only one-third as many roots. Measurements of label distribution between the 1-cm base and the shoot remainder showed that less than 10% of the label moved to the shoot remainder over a 6-day period of auxin application. Dose-response curves of IAA and rooting over the range 1 × 10−5 M and 3 × 10−3 M showed that root number in M.9 was at an optimum at 1 × 10−3 M IAA after 6 days whilst M.26 required only 1 × 10−4 M for a similar response. These data support the hypothesis that differences in rooting of the two rootstocks reflect differences in the endogenous metabolism of exogenous IAA and not differences in its rates of uptake or distribution in the shoots.  相似文献

Male three-spined stickleback Gasterosteus aculeatus have a fixed amount of sperm during the breeding season because spermatogenesis is inhibited at this time. A method was developed to estimate ejaculate size in situ by removing the sperm from the male's nest. The reliability of the method was tested using known numbers of sperm. In their first mating, males ejaculated 11·64 × 106 sperm (median), representing c. 5% of the male's sperm store (median 27·88 × 107 sperm). The amount of sperm in the testes was significantly reduced in males that had mated several times (median 8·09 × 107). Additionally, ejaculate size was smaller in these experienced males (median 8·79 × 105). Heavier and larger fish invested absolutely and relatively more sperm in a mating than did lighter and smaller fish. Ejaculate size did not correlate with the mass of the egg clutch.  相似文献

The gonad weight and gonadosomatic index of the male Baltic herring Clupea harengus membras L., were highest at the beginning of the reproductive season (April/May), the values decreasing towards the end of it (July/August) during 1988–1991. The decline could not be explained by fish size but may have been due to fish condition. A high individual variation was typical for both gonad weights and gonadosomatic indices in fish of the same size and maturity stage. The mean density of sperm cells was significantly higher in June (34·9 × 109 ml−1) than in July (19·2 × 109 ml−1, Mann-Whitney U= 17; P<0·05), the variation among the males being high in both groups. Electron microscope analysis showed a severe disruption of the mitochondrial elements in males spawning at 22°C.  相似文献

Protoplast fusion induced by polyethylene glycol and Ca2+, was performed between auxotrophic mutants of pectinolytic fungi Aspergillus sp. CH-Y-1043 (A13) ade and Aspergillus flavipes ATCC-16795 (F7) lys . Prototrophic colonies were developed on minimal medium with a fusion frequency of 1·0×10−2. The reversion frequency of the mutation in spores and protoplasts was low and ranged from 2·0 to 4·0×10−7. Four prototrophic hybrids (HH, HE, HF and HJ) exhibited enhanced production of endo-pectinase and pectin-lyase. The highest production was observed in HJ ; maximum activities were 150 and 160% respectively, whereas the exo-pectinase production was similar to the wild-type strain Aspergillus sp. CH-Y-1043. Hybrid HJ showed the greatest growth ; nevertheless, specific endo-pectinase and pectin-lyase activities were higher in all hybrids than those produced by the wild-type strains.  相似文献

An in vitro method of growing bacteria as a defined nutrient-depleted biofilm is proposed. The medium was defined nutritionally in terms of the quantitative composition and by the total amount of nutrient required to achieve a defined population size. Escherichia coli and Burkholderia cepacia were incubated on a filter support placed on a defined volume of solid medium. The change of biomass of the biofilm population was compared with the change in a planktonic culture. The size of the population in stationary phase was proportional to the concentration of limiting substrate up to 40 μmol cm−2 glucose for E. coli and up to 2·7 × 10−9 mol cm−2 iron for B. cepacia . Escherichia coli growing exponentially had a growth rate of μ = 0·30 h−1 in a biofilm and μ = 0·96 h−1 in planktonic culture. The growth rate, μ, for exponentially growing B. cepacia in a biofilm was 1·12 h−1 and in planktonic culture 0·78 h−1. This method allows the limitation of the size of a biofilm population to a chosen value.  相似文献

By using two polyclonal antisera against WH 7803 strain (Synechococcus sp.) and WH 5701 strain (Synechococcus bacillaris) it is possible to detect and to enumerate cells of the two cyanobacterial serogroups. The immunofluorescence technique was used to study the distribution of the two serogroups in the estuarine, coastal and upwelling waters of the Mediterranean Sea surrounding Messina. In the estuarine waters of the Alcantara River (Ionian Sea), the WH 7803 serogroup was present at a concentration in the order of 102 cells ml−1 and the WH 5701 serogroup at a concentration of 5·5 × 102 cellsml−1. In the coastal waters of Messina, where urban and industrial wastes are usuallydumped, the concentration of total phycoerythrin- Synechococcus ranged from 1·3 × 102 to 4·1 × 103 cells ml−1; the WH 7803 serogroup accounted for 50–94% of the totalpopulation in Ionian stations, whereas the WH 5701 serogroup ranged from1·4 × 101 to6·7 × 102cells ml−1. In the upwelling area (Straits of Messina) bothserogroups were found. Vertical distribution of two Synechococcus strains had anopposite trend and their concentrations were of the order of 101–102cells ml−1. Theuse of the Scan laser system allows both autofluorescent and labelled organismsto be distinguished in a preparation for optical microscopy. The body composition of protein and fat in Atlantic salmon Salmo salar and brown trout Salmo trutta before and after winter was investigated in a temperate, small river, normally ice covered from the middle of November until the end of March. Fat, protein and specific energy declined greatly in winter but were replenished rapidly in spring. Rates of decline were slower for the smallest fish, which also had the lowest specific content of fat, protein, and energy, while the differences in absolute amounts were greatest for the largest fish. The mean specific fat content was reduced by 45–70% during winter, relative to the pre-winter period (September). Mean daily reductions in specific enegy of the larger size groups of brown trout (3·7 × 10−3 kJ g−1 day−1) were almost half of the corresponding values for the largest Atlantic salmon (6·3 × 10−3 kJ g−1 day−1) during winter. A minor reduction in protein content was found during winter, with mean reductions of 6–10% in comparison to those in September. During spring the fat content was replenished rapidly, particularly for the smallest salmon fry (a threefold increase from April to June). Fat content in the larger salmon and trout increased by about 1·8 times. Based on estimated metabolic rates, digested energy during wintertime may contribute about two-thirds of the brown trout fry's energy demand. For Atlantic salmon, the corresponding value is about 50%. The winter period put considerable stress on the young salmonids living in lotic environments, in particular for the smallest fry with the lowest energy content before winter and the largest losses during winter. This should make the fry more vulnerable to adverse abiotic and biotic factors.  相似文献

The schooling behaviour of Atlantic mackerel was studied in a large tank at different light intensities in the range 12.6–1.8 × 10−10μEs−1 m−2. Variable light intensity was produced by accurately controlling the current to a green light-emitting diode (LED) 3 m above the experimental tank. Under high light levels (1.8 × 10−6μEs−1 m−2) mackerel always formed a single school, whereas at lower levels (1.8 × 10−8μEs−1 m−2) they swam as individuals. At light levels down to 1.0 × 10−6μEs−1 m−2 the mean nearest neighbour distance in a school remained relatively constant (0.3–0.9 body lengths), and individual mackerel swam along a path which deviated from the position of their nearest neighbours by less than 14°. As light dropped below 1.8 × 10−7μEs−1 m−2, both nearest neighbour distance and heading angle between nearest neighbours increased, with mean values of 1–1.8 body lengths and 23–92°, respectively, at 1.8 × 10−9μEs−1 m−2. The results are discussed in terms of ambient light conditions in the sea.  相似文献

The length of the auxin-sensitive phase of root initiation 'in vitro' in the apple rootstock M.9 ( Malus pumila Mill.) has been determined using the auxins indol-3yl-acettc acid (IAA) at 2.8 × 10−5 M and indol-3yl-butyric acid (IBA) at 1.5 × 10−5 M in the presence and absence of 10−3 M phloroglucinol (PG). PG synergised IBA-induced rooting after 4 days exposure, but contact times exceeding 8 days decreased root number. In contrast, PG consistently synergised IAA-induced rooting in the dark for contact periods up to 13 days with the highest rooting being recorded at 9 days. An irradiance of 20 W m−2 from fluorescent lamps halved IAA-induced rooting irrespective of the presence or absence of PG. The culture of shoots at temperatures of 22,25 and 29°C during the root initiation phase (auxin present) and the root emergence phase (auxin absent) produced no difference in rooting response. In the presence of PG the use of liquid culture in place of agar-solidified culture during the auxin-sensitive phase reduced root number but not rooting percentage.  相似文献

The survival of Atlantic salmon smolts on exposure to constant concentrations of ammonia has been measured under laboratory conditions. At concentrations of dissolved oxygen close to the air-saturation value, the 24-h LC50 of un-ionised ammonia is 0.15 mg NH31−1 in fresh water (hardness 264 mg 1−1 as CaCO3) and 0.3 mg NH31−1 in 30% sea water; at concentrations of dissolved oxygen of 3.5 mg 1−1 in fresh water and 3.1 mg 1−1 in 30% sea water, the 24-h LC50 is 0.09 mg NH3 1−1 and 0.12 mg NH3 1−1 respectively; for fish acclimated for 1 day to a concentration of ammonia close to the 24-h median for un-acclimated fish, the median is increased between 38 and 79%, depending on test conditions.  相似文献

To examine otolith resorption induced by anaerobic stress, 45Ca-prelabelled goldfish, Carassius auratus , were kept in oxygen-deficient ambient water (O2 < 0.5 .1 1−1, 26° C) for 2 days and otoliths (asterisci) were analysed for 45Ca retention. In a second experiment, fish were anaerobically stressed for 2 days, and then received a single intraperitoneal injection of 45Ca. They were maintained under stress for one more day and killed to examine 45Ca deposition in otoliths. Plasma was analysed for total and radioactive calcium. Otoliths (lapilli) were examined for stress-induced check formation by scanning electron microscopy (SEM).
Stress significantly reduced plasma calcium levels and the rate of calcium retention in otoliths, which was calculated from the 45Ca specific activity of the plasma. On the other hand, the rate of calcium deposition in otoliths was the same in the stressed as in the control fish. SEM observation revealed that the applied stress resulted in a check formation in otoliths. These results indicate that a 48-h stress of oxygen deficiency induces calcium resorption in otoliths.  相似文献

Aims:  To verify the taxonomic affiliation of bacterium Butyrivibrio fibrisolvens strain A from our collection and to characterize its enzyme(s) responsible for digestion of sucrose.
Methods and Results:  Comparison of the 16S rRNA gene of the bacterium with GenBank showed over 99% sequence identity to the species Pseudobutyrivibrio ruminis . Molecular filtration, native electrophoresis on polyacrylamide gel, zymography and thin layer chromatography were used to identify and characterize the relevant enzyme. An intracellular sucrose phosphorylase with an approximate molecular mass of 52 kDa exhibiting maximum activity at pH 6·0 and temperature 45°C was identified. The enzyme was of inducible character and catalysed the reversible conversion of sucrose to fructose and glucose-1-P. The reaction required inorganic phosphate. The K m for glucose-1-P formation and fructose release were 3·88 × 10−3 and 5·56 × 10−3 mol l−1 sucrose, respectively – while the V max of the reactions were −0·579 and 0·9  μ mol mg protein−1 min−1. The enzyme also released free glucose from glucose phosphate.
Conclusion:  Pseudobutyrivibrio ruminis strain A utilized sucrose by phosphorolytic cleavage.
Significance and Impact of the Study:  Bacterium P. ruminis strain A probably participates in the transfer of energy from dietetary sucrose to the host animal.  相似文献
