It is shown that dynamics of percentage of conditioned food refusals by snails Helix pomatia and Helix lucorum is satisfactorily approximated by regression equation of exponential type with included coefficient reflecting the animals state before the beginning of learning. The ability is established of the snails to practically completely differentiate various alimentary conditioned stimuli. The introduction of differentiation always improved the reflex consolidation. Conditioned reactions to differentiation stimulus appeared at the elaboration stage and were absent at the stage of conditioned reflex consolidation.  相似文献

The effects of direct application of acetylcholine (ACh) and m- and n-cholinoreceptor blockers on test cells were investigated in waking cats having developed instrumental lever-pressing conditioned reflex. Changes were recorded in both spontaneous and invoked firing activity in a functionally homogeneous group of motor cortex cells, in which increased discharge rate usually preceded the start of conditioned reflex movements. It was found, however, that ACh increased spontaneous activity considerably in some of the neurons tested and reduced it moderately in others. Atropine sharply reduced background activity in cortical neurons while preserving spike response to presentation of a conditioned stimulus and n-cholino-blockers such as hexonium and (occasionally) tubocurarine inhibited spike response produced by conditioned stimuli; background activity was slightly inhibited by hexonium and reinforced by tubocurarine. It was concluded that ACh put out by cholinergic fibers helps to maintain background firing activity level in cortical neurons under naturally occurring conditions, acting via m-cholinoreceptors, whereas factors influencing generation of spike discharges associated with performance of conditioned reflex movements are mediated by n-cholinoreceptors.A. A conditioned alimentary instrumental reflex was elaborated in three dogs with chronically implanted electrodes to a consecutive complex conditioned stimulus consisting of two equal acoustic stimuli separated by a pause. At first, the animals reacted with motor reactions during the whole action of the complex signal. Then inhibition of reaction was elaborated at first in pause and then in response to the preparatory stimulus. In the process of stabilization of inhibition, there was an increase of similarity of spectral composition of hippocampal theta-rhythm with the values obtained in the inter-signal period. Conclusion is made that the hippocampus is not a structure necessary for the elaboration of internal inhibition. The frequency of hippocampal theta-rhythm may serve as a characteristic of the property called in Pavlov's school mobility of the nervous system.  相似文献

The basic design used in our human fear-conditioning studies on disrupting reconsolidation includes testing over different phases across three consecutive days. On day 1 - the fear acquisition phase, healthy participants are exposed to a series of picture presentations. One picture stimulus (CS1+) is repeatedly paired with an aversive electric stimulus (US), resulting in the acquisition of a fear association, whereas another picture stimulus (CS2-) is never followed by an US. On day 2 - the memory reactivation phase, the participants are re-exposed to the conditioned stimulus without the US (CS1-), which typically triggers a conditioned fear response. After the memory reactivation we administer an oral dose of 40 mg of propranolol HCl, a β-adrenergic receptor antagonist that indirectly targets the protein synthesis required for reconsolidation by inhibiting the noradrenaline-stimulated CREB phosphorylation. On day 3 - the test phase, the participants are again exposed to the unreinforced conditioned stimuli (CS1- and CS2-) in order to measure the fear-reducing effect of the manipulation. This retention test is followed by an extinction procedure and the presentation of situational triggers to test for the return of fear. Potentiation of the eye blink startle reflex is measured as an index for conditioned fear responding. Declarative knowledge of the fear association is measured through online US expectancy ratings during each CS presentation. In contrast to extinction learning, disrupting reconsolidation targets the original fear memory thereby preventing the return of fear. Although the clinical applications are still in their infancy, disrupting reconsolidation of fear memory seems to be a promising new technique with the prospect to persistently dampen the expression of fear memory in patients suffering from anxiety disorders and other psychiatric disorders.  相似文献

The effects of external stimulus presentation on neuronal spike response induced in area 7 by conditioned and unconditioned stimuli were investigated in waking cats. Pointing reaction arising in response to the action of external stimuli led to the phasing out of spike response to unconditioned cutaneous stimulation, as well as depression or complete suppression of response in area 7 neurons to conditioned stimulus presentation, despite occurrence of conditioned reflex motion. External stimuli no longer exerted an inhibitory effect on the neuronal spike response induced by protracted use of conditioned and unconditioned stimuli. The fact that the conditioned lever-pressing reflex could occur either following or without neuronal spike response in area 7 would raise the question of whether neurons belonging to this area always participate directly in the triggering and occurrence of conditioned reflex motion.A. Neuronal response in the cat association cortex (area 5) to conditioned and non-conditioned acoustic stimulation was investigated. Numbers of neurons responding to a conditioned acoustic stimulus according to the traditional reflex pattern were twice as high. Numbers of inhibitory neuronal responses to the stimulus increased when instrumental reflex occurred. Neurons were found which only reacted to a conditioned acoustic stimulus in the absence of conditioned reflex movement occurring with instrumental food reflex. Although findings do not exclude the possibility of this cortical area contributing to the analysis of sensory signals and evaluation of their biological significance, it might be supposed that its main functional property lies in its involvement in the process of initiating behavioral response to a conditioned response.A. Unit activity of the somatosensory cortex was studied in chronic experiments on cats during alimentary instrumental conditioned reflex performance and elaboration of conditioned inhibition (CI). First presentations of the CI signal at the beginning of CI formation caused, due to orienting reaction to a new stimulus, a levelling of the response of the neurones to the positive conditioned signal included into the CI complex. At the stage of consolidated CI, this depression proceeded gradually during the development of conditioned inhibition caused by consecutive presentations of the nonreinforced CI combination. Two groups of neurones were involved in the process of the CI performance: one of them being the same which was activated also in response to positive stimulation during performance of the conditioned response, and the second one being areactive to the positive conditioned stimulus but reducing the frequency of the background activity.  相似文献

Fear‐potentiated acoustic startle paradigms have been used to investigate phasic and sustained components of conditioned fear in rats and humans. This study describes a novel training protocol to assess phasic and sustained fear in freely behaving C57BL/6J mice, using freezing and/or fear‐potentiated startle as measures of fear, thereby, if needed, allowing in vivo application of various techniques, such as optogenetics, electrophysiology and pharmacological intervention, in freely behaving animals. An auditory Pavlovian fear conditioning paradigm, with pseudo‐randomized conditioned–unconditioned stimulus presentations at various durations, is combined with repetitive brief auditory white noise burst presentations during fear memory retrieval 24 h after fear conditioning. Major findings are that (1) a motion sensitive platform built on mechano‐electrical transducers enables measurement of startle responses in freely behaving mice, (2) absence or presence of startle stimuli during retrieval as well as unpredictability of a given threat determine phasic and sustained fear response profiles and (3) both freezing and startle responses indicate phasic and sustained components of behavioral fear, with sustained freezing reflecting unpredictability of conditioned stimulus (CS)/unconditioned stimulus (US) pairings. This paradigm and available genetically modified mouse lines will pave the way for investigation of the molecular and neural mechanisms relating to the transition from phasic to sustained fear.  相似文献

The responses of motor cortex neurons in the cat to the presentation of a single auditory click and a series of 10 clicks presented with 1,000/sec frequency were studied under conditions of chronic experiments before and after the development of an instrumental food reflex. After reflex development a single presentation of a positive conditioned stimulus (single click) markedly influenced for 7 sec the appearance of instrumental movements. At the same time, the immediate responses of motor cortex neurons to presentation of the conditioned auditory stimulus had no impact on the appearance in the motor cortex of discharges leading to the realization of instrumental movements. Consequently, motor cortex neurons do not require activation from afferent sensory inputs for the generation of such discharges. The immediate neuronal responses to conditioned stimulation did not inhibit the realization of the instrumental reflex. It is proposed that they are associated with the realization of motor function in the unconditioned defensive response evoked by the presentation of an auditory stimulus. The presence or absence of responses to auditory conditioned stimulation was dependent upon the signal meaning of the stimulus, its physical parameters, and the degree of excitability of the animal.A. A reversal of signal meaning of stimuli signalling experimental situation was performed on dogs with conditioned defensive motor reflexes to clicks: clicks and electro-cutaneous stimulation were stopped, and the animals were started to be given food in the same experimental situation. Reversal of conditioned defensive tonic stimuli to alimentary ones led to a complex of behavioral and electrographic reactions. Isolated click tests showed that signals of experimental situation considerably affect a fully-elaborated defensive motor reflex. At the same time the preparatory role of certain situational elements may be specialized with regard to the components of the conditioned reflex, in particular to the local flexor reaction.  相似文献

Intravenous naloxone injection (0.1 mg/kg) facilitated blood pressure increase in response to conditioned sound stimulus followed by electrocutaneous shock in conscious chair-restrained baboons (Papio hamadryas). Naloxone at a dose of 1.0 mg/kg had an opposite effect and led to the decrease in blood pressure and heart rate in conditioned fear reflex. Naloxone microinjections (50 microM) into the periventricular hypothalamus led to a significant diminution of blood pressure and heart rate increment in response to electrocutaneous shock; naloxone microinjections into tractus solitarius nuclei suppressed blood pressure and heart rate reactions both to conditioned (sound) and unconditioned (electrocutaneous shock) stimuli. Microinjections of equimolar morphine quantities in these brain regions facilitated such reactions. It is concluded that endogenous opioid system participates in the formation of cardiovascular reactions to emotional stimuli in monkeys, with multiple opioid receptors of periventricular hypothalamus and tractus solitarius nuclei involved in the generation of such reactions.  相似文献

In three dogs the dynamics was studied of changes in the number of instrumental motor reactions, the heart rate and the hippocampus theta rhythm was studied at the change of constant reinforcement to various regimes of probable reinforcement of alimentary conditioned stimulus. At 70 and 50% levels of the reinforcement the instrumental reflex appeared in response to all presentations of the signal stimulus. The greatest decrease in the number of reactions took place at 5% probability. The heart rate both in the intersignal period and at getting of empty feeder depended on the used probability of reinforcement and individual characteristics of animals. The frequency of the theta rhythm in all dogs was the highest at the 30% level and the lowest values were found at the 5% level of reinforcement. The obtained facts testify once more to complex genesis of emotional excitation, the value of which depends both on informational (uncertainty) and motivational (significance of reinforcement) factors.  相似文献

In analysis of neuronal activity of g. proreus of dogs brain in response to positive and differentiation conditioned cutaneous stimuli, in 44% of 78 studied neurons "late responses" appeared in the form of impulse activity intensification elicited by a positive conditioned stimulus after the end of eating and by a differentiation one--during conditioned signal action; it persisted in different neurones for 30--60 s. By means of various functional tests it has been shown that these reactions are a neurophysiological correlate of animals emotional state gradient.  相似文献

Acute animal preparations have been used in research prospectively investigating electrode designs and stimulation techniques for integration into neural auditory prostheses, such as auditory brainstem implants and auditory midbrain implants. While acute experiments can give initial insight to the effectiveness of the implant, testing the chronically implanted and awake animals provides the advantage of examining the psychophysical properties of the sensations induced using implanted devices. Several techniques such as reward-based operant conditioning, conditioned avoidance, or classical fear conditioning have been used to provide behavioral confirmation of detection of a relevant stimulus attribute. Selection of a technique involves balancing aspects including time efficiency (often poor in reward-based approaches), the ability to test a plurality of stimulus attributes simultaneously (limited in conditioned avoidance), and measure reliability of repeated stimuli (a potential constraint when physiological measures are employed). Here, a classical fear conditioning behavioral method is presented which may be used to simultaneously test both detection of a stimulus, and discrimination between two stimuli. Heart-rate is used as a measure of fear response, which reduces or eliminates the requirement for time-consuming video coding for freeze behaviour or other such measures (although such measures could be included to provide convergent evidence). Animals were conditioned using these techniques in three 2-hour conditioning sessions, each providing 48 stimulus trials. Subsequent 48-trial testing sessions were then used to test for detection of each stimulus in presented pairs, and test discrimination between the member stimuli of each pair. This behavioral method is presented in the context of its utilisation in auditory prosthetic research. The implantation of electrocardiogram telemetry devices is shown. Subsequent implantation of brain electrodes into the Cochlear Nucleus, guided by the monitoring of neural responses to acoustic stimuli, and the fixation of the electrode into place for chronic use is likewise shown.  相似文献

In conditioned taste aversion (CTA) training performed on the pond snail Lymnaea stagnalis, a stimulus (the conditional stimulus, CS; e.g., sucrose) that elicits a feeding response is paired with an aversive stimulus (the unconditional stimulus, US) that elicits the whole-body withdrawal response and inhibits feeding. After CTA training and memory formation, the CS no longer elicits feeding. We hypothesize that one reason for this result is that after CTA training the CS now elicits a fear response. Consistent with this hypothesis, we predict the CS will cause (1) the heart to skip a beat and (2) a significant change in the heart rate. Such changes are seen in mammalian preparations exposed to fearful stimuli. We found that in snails exhibiting long-term memory for one-trial CTA (i.e., good learners) the CS significantly increased the probability of a skipped heartbeat, but did not significantly change the heart rate. The probability of a skipped heartbeat was unaltered in control snails given backward conditioning (US followed by CS) or in snails that did not acquire associative learning (i.e., poor learners) after the one-trial CTA training. These results suggest that as a consequence of acquiring CTA, the CS evokes conditioned fear in the conditioned snails, as evidenced by a change in the nervous system control of cardiac activity.  相似文献

Brain-derived neurotrophic factor (BDNF) has been implicated in hippocampal-dependent learning processes, and carriers of the Met allele of the Val66Met BDNF genotype are characterized by reduced hippocampal structure and function. Recent nonhuman animal work suggests that BDNF is also crucial for amygdala-dependent associative learning. The present study sought to examine fear conditioning as a function of the BDNF polymorphism. Fifty-seven participants were genotyped for the BDNF polymorphism and took part in a differential-conditioning paradigm. Participants were shocked following a particular conditioned stimulus (CS+) and were also presented with stimuli that ranged in perceptual similarity to the CS+ (20, 40 or 60% smaller or larger than the CS+). The eye blink component of the startle response was measured to quantify fear conditioning; post-task shock likelihood ratings for each stimulus were also obtained. All participants reported that shock likelihood varied with perceptual similarity to the CS+ and showed potentiated startle in response to CS ± 20% stimuli. However, only the Val/Val group had potentiated startle responses to the CS+. Met allele carrying individuals were characterized by deficient fear conditioning – evidenced by an attenuated startle response to CS+ stimuli. Variation in the BDNF genotype appears related to abnormal fear conditioning, consistent with nonhuman animal work on the importance of BDNF in amygdala-dependent associative learning. The relation between genetic variation in BDNF and amygdala-dependent associative learning deficits is discussed in terms of potential mechanisms of risk for psychopathology.  相似文献

The activity of 109 neurons in the compact and diffuse parts of the pedunculopontine tegmental nucleus (PPTg) was recorded in freely moving rabbits during the acquisition and actualization of the defensive instrumental conditioned reflex. It was found that 47% of the recorded neurons responded to a conditioned stimulus (CS). This finding suggests the involvement of the PPTg in the instrumental conditioning. A significant prevalence of the excitatory conditioned responses to the CS suggests the predominantly activating influence of the PPTg on its projection structures during conditioning. The neuronal responses to the CS were classified into several basic patterns reflecting stimulus effects, the structure of the behavioral act, and the nature of the reinforcement. They indicated the involvement of the PPTg in attention, motor learning, and reinforcement. A significant decrease in the reactivity of the CS of the PPTg neurons as a result of learning specialization was shown. The revealed differences in the associative and reactive properties (with respect to CS) between the neurons of the compact and diffuse PPTg parts testify to the functional heterogeneity of this structure and suggest the leading role of the cholinergic compact part of the PPTg in the instrumental defensive conditioning. Thus, the obtained evidence suggests the involvement of the PPTg in the mechanisms of attention and acquisition of the active defensive motor conditioning.  相似文献

Activity of 91 neurons in the compact and diffuse parts of the pedunculopontine tegmental nucleus (PPTg) was recorded in freely moving rabbits during execution of instrumental appetitive conditioning. Of the recorded neurons, 37.4% changed their activity in response to a conditioned stimulus, which is the evidence for the PPTg involvement in instrumental conditioning. Excitatory conditioned neuronal responses to the conditioned stimulus and food reinforcement significantly prevailed over inhibitory reactions. Neuronal responses to the conditioned stimulus were classified in several basic patterns reflecting stimulus effects, structure of the behavioral act, and reinforcement properties. These reactions indicate the involvement of the PPTg in attention, motor learning, and reinforcement. The revealed differences in associative reactive properties of the compact and diffuse parts of the PPTg to the conditioned stimulus and reinforcement point to the functional heterogeneity of this structure and suggest the leading role of the cholinergic part of the PPTg in instrumental appetitive conditioning and reinforcement as well as the essential contribution of the diffuse part into classical appetitive conditioning.  相似文献

Fear is one of the most potent emotional experiences and is an adaptive component of response to potentially threatening stimuli. On the other hand, too much or inappropriate fear accounts for many common psychiatric problems. Cumulative evidence suggests that the amygdala plays a central role in the acquisition, storage and expression of fear memory. Here, we developed an inducible striatal neuron ablation system in transgenic mice. The ablation of striatal neurons in the adult brain hardly affected the auditory fear learning under the standard condition in agreement with previous studies. When conditioned with a low-intensity unconditioned stimulus, however, the formation of long-term fear memory but not short-tem memory was impaired in striatal neuron-ablated mice. Consistently, the ablation of striatal neurons 24 h after conditioning with the low-intensity unconditioned stimulus, when the long-term fear memory was formed, diminished the retention of the long-term memory. Our results reveal a novel form of the auditory fear memory depending on striatal neurons at the low-intensity unconditioned stimulus.  相似文献

Paired heterogeneous stimulation of the lateral geniculate body (LGB) and pulvinar (Pulv) as a conditioned stimulus of alimentary instrumental conditioned reflex (CR), resulted in a change of relations between afferent inputs from LGB and Pulv to the visual and associative cortex of cats. At stimulation of LGB preceding by 40 ms, facilitation of the response to testing Pulv stimulation observed in untrained cats, appeared only at the beginning of the learning and was suppressed by the end of elaboration, when the amplitude of the response to the conditioning LGB stimulation greatly increased. In the process of CR elaboration (in the middle of learning), Pulv stimulation preceding by 40 ms facilitated the response to the testing LGB stimulation and simultaneously increased the amplitude of the response to the conditioning Pulv stimulation.  相似文献
