植物的水分基本是通过器官表皮气孔和角质层两种通道散失到大气中去的,尤以气孔蒸腾为主,而且与光合作用有着十分密切的关系。气孔蒸腾(Tr)可用多种方法测定。最常用的为气孔计法,但往往限于叶片,较难应用于其它器官或部位。角质层蒸腾则常用离体叶片称重测定其失水速率(**U。二者同步联合测定的报道较少(棉花有Quisenberry等[‘j,小麦有Wang和Clarke[‘j及马瑞昆等[‘j),叶片以外的其它器官测定则更未见报道。本文以小麦为材料,提出用离体植株器官连续自动称重方法同步联合测定气孔和角质层蒸腾的技术,同时对取样、数…  相似文献

打孔称重法是基于相近叶位叶片的比叶重(单位面积叶片重量)相对恒定的原理,通过测定采样区植物叶片干重和少量叶片比叶重计算叶面积和叶面积指数的方法。比较试验结果表明:打孔称重法与长宽校正法和复印称重法测定的水稻叶面积呈极显著相关;打孔称重法和复印称重法所测定的叶面积指数平均值经统计检验差异不显著。2种方法均能很好地袁征不同处理间叶面积指数的差异。  相似文献

通过对毛乌素沙地几种主要植物的枝条在剪断前后的蒸腾速率的研究 ,发现这些植物枝条的蒸腾速率在剪离母株后一般会发生较大的变化 ,说明用快速称重法测定得到的剪断枝条的蒸腾速率与剪断前植物的实际蒸腾速率也存在较大的偏差。由于这类变化与植物的生物学特性、生长环境的水分条件、测定时叶片的水分状况、剪断的部位和测定的时间等多种因素都有密切关系 ,定量地预测某个植物剪断枝条的蒸腾速率的变化一般也比较困难。用快速称重法测定植物的蒸腾速率需要有同步的校正措施 ,否则结果不可靠。气孔计在测定植物枝条剪断前后蒸腾速率的相对变化率时比较准确。因而 ,用快速称重法测定植物的蒸腾速率 ,用气孔计来校正其偏差 ,这种二合一的方法是测定植物蒸腾速率比较准确而理想的方法  相似文献

通过离体子叶冷冻法、离体叶片冷冻法和10%二甲基亚砜+5%脱脂乳冷冻法等3种方法来保藏大白菜霜霉病菌寄生霜霉,并采用离体子叶接种法测定孢子囊的致病力,以筛选出一种较好的保藏方法。结果表明,保藏6个月后,离体子叶冷冻法和离体叶片冷冻法保藏的孢子囊的萌发率、发病率和病情指数均较高;而10%二甲基亚砜+5%脱脂乳冷冻法保藏6个月后仅有2.16%的孢子囊能够萌发,菌株致病力丧失,发病率和病情指数均为0。保藏12个月后,离体子叶冷冻法保藏效果最好,孢子囊萌发率达到62.22%,发病率为90%,病情指数为48.89;离体叶片冷冻法保藏的孢子囊的致病力较弱;10%二甲基亚砜+5%脱脂乳冷冻法保藏12个月的孢子囊全部失活。此外,采用离体子叶冷冻法保藏12个月的32株寄生霜霉复壮成功率达到了93.75%。离体子叶冷冻法适用于寄生霜霉的保藏,其孢子囊可长时间保持较高的萌发率。  相似文献

为了减轻UV-B辐射对植物叶片的伤害, 本研究以离体红芸豆叶片为实验材料, 通过外源施加NaHSO3的方法探讨了UV-B辐射下NaHSO3 对离体红芸豆叶片的保护作用。结果表明:与未处理对照相比较, 用0.5 mmol.L-1 NaHSO3处理的离体红芸豆叶片表面褐色斑减少、边缘蜷曲及萎蔫程度降低;且能延缓叶片中叶绿素和类胡萝卜素含量的降低;使类黄酮含量升高; 叶片中过氧化物酶(POD)和抗坏血酸过氧化物酶(APX)活性升高, 过氧化氢(H2O2)含量降低。进一步研究发现NaHSO3处理能明显延缓PSⅡ原初光能转换效率的降低;增强PSⅡ的电子传递能力, 减少叶绿体内有害自由基的产生, 减缓叶绿体内光合机构遭受破坏的程度。以上结果表明NaHSO3可能通过提高POD和APX的活性、降低自由基产生及保护光合色素等来实现UV-B胁迫下对红芸豆叶片的保护作用。  相似文献

以感染内生真菌的天然禾草羊草为实验材料,通过体外纯培养条件下的内生真菌、感染内生真菌的离体叶片和在体叶片对3种病原菌的抑菌实验,以探讨内生真菌对宿主植物羊草在抗病性方面的贡献。结果表明:体外纯培养条件下,分离自羊草的内生真菌Epichlobromicola对新月弯孢(Curvularia lunata)、根腐离蠕孢(Bipolaris sorokiniana)和枝孢霉(Cladosporium sp.)这3种病原菌都具有抑制作用,抑菌率分别达56.22%,46.93%和45.15%,且内生真菌培养滤液可以有效抑制这3种病原菌的孢子萌发,平均萌发率分别为30.4%,15.7%和16.4%;宿主植物叶片在离体条件下,内生真菌感染可以有效降低羊草叶片受C.lunata和C.sp.侵染后的病斑数或病斑长度,但对B.sorokiniana不起作用,甚至提高了叶片的病斑数及病斑长度,而离体叶片提取液对不同病原菌均有不同程度的抑制作用;在体条件下,内生真菌均可以通过降低叶片病斑数来增强羊草植株对这3种病原菌的抗性。由此看来,内生真菌E.bromicola对宿主植物羊草在抗病原菌侵染方面有一定的增益作用。  相似文献

采用数码相机获取叶片的数字图像,用Photoshop图像处理软件计算叶面积,并与目前常用的剪纸法和叶面积仪测定法进行比较分析。结果表明,本方法和上述传统测定方法测定结果存在极显著线性相关;不同拍摄分辨率、单位叶面积存贮像素个数和拍摄角度对测定结果无显著影响。和其它方法相比,本方法具有准确、快速、成本低廉、适合非破坏性动态连续观测等优点,适用于植物生理生态学研究中叶面积的测定。  相似文献

We report a protocol using a common desk-top scanner and public domain software for measuring existing leaf area and leaf area removed as a result of herbivory. We compared the accuracy and precision of this method to that of a standard leaf area meter. Both methods were used to measure metal disks of a known area, the area of soybean (Glycine max L.) leaves, and the area removed by simulating leaf feeding with a hole-punch. We varied the amount of injury across a low, medium, and high degree of simulated feeding. The mean area of 10 cm2 and 50 cm2 metal disks was more accurately estimated with the leaf area meter than the desk-top scanner. Leaf area estimates from both methods were highly correlated. The desk-top scanner accurately estimated the leaf area removed from the low, medium, or high degree of simulated leaf feeding. However, the leaf area meter overestimated low levels of simulated feeding injury. Though measuring a leaf's surface area with a desk-top scanner requires two steps (creating a digital image file and calculating the area represented by that image), the overall time required to measure leaf injury is shorter than with a leaf area meter. This relatively simple and inexpensive method of estimating leaf area and feeding damage has advantages in certain experimental situations where a prefeeding measurement of the leaf is impossible or undesirable, or when small amounts of feeding occur.  相似文献

毛竹扩张对常绿阔叶林生态系统的影响问题已成为近年国内外关注的热点问题之一。通过对常绿阔叶林和竹阔混交林内甜槠、青冈、枳椇以及青榨槭4树种主要叶结构型性状的调查与测定,分析毛竹扩张对常绿阔叶林主要树种叶结构型性状的影响。结果表明:(1)毛竹扩张不会显著改变乔木层4树种总体叶结构型性状特征。(2)不同生活型阔叶树种对毛竹扩张的响应策略不同。受毛竹扩张影响,常绿阔叶树种叶组织密度(LTD,leaf tissue density)显著减小;落叶阔叶树种叶宽(LW,leaf width)显著减小,叶形态指数(LMI,leaf morphology index)则显著增大。(3)毛竹扩张会使乔木层4树种间部分叶结构型性状差异特征发生改变,竹阔混交林中乔木层4树种间LMI、LTD、比叶面积(SLA,specific leaf area)及叶干物质含量(LDMC,leaf dry matter content)的差异特征不同于常绿阔叶林;毛竹向常绿阔叶林扩张后,甜槠叶长(LL,leaf length)、LW及叶面积(LA,leaf area)显著增大;青冈LMI显著增大,LDMC和LTD显著减小;枳椇的LMI显著增大,LW、LTD显著减小;青榨槭的LW显著减小。(4)毛竹扩张会造成乔木层树种部分叶结构型性状间关系发生改变,使林木的LL与LW、LA与LW间的截距减小,造成林木LL与LW、LA与LW之间的数量关系发生改变。  相似文献

Human perception of plant leaf and flower colour can influence species management. Colour and colour contrast may influence the detectability of invasive or rare species during surveys. Quantitative, repeatable measures of plant colour are required for comparison across studies and generalisation across species. We present a standard method for measuring plant leaf and flower colour traits using images taken with digital cameras. We demonstrate the method by quantifying the colour of and colour difference between the flowers of eleven grassland species near Falls Creek, Australia, as part of an invasive species detection experiment. The reliability of the method was tested by measuring the leaf colour of five residential garden shrub species in Ballarat, Australia using five different types of digital camera. Flowers and leaves had overlapping but distinct colour distributions. Calculated colour differences corresponded well with qualitative comparisons. Estimates of proportional cover of yellow flowers identified using colour measurements correlated well with estimates obtained by measuring and counting individual flowers. Digital SLR and mirrorless cameras were superior to phone cameras and point-and-shoot cameras for producing reliable measurements, particularly under variable lighting conditions. The analysis of digital images taken with digital cameras is a practicable method for quantifying plant flower and leaf colour in the field or lab. Quantitative, repeatable measurements allow for comparisons between species and generalisations across species and studies. This allows plant colour to be related to human perception and preferences and, ultimately, species management.  相似文献

叶面积指数(LAI)的田间测量是生态和农业等领域的常规工作之一,测量方法分为直接测量和间接测量,间接测量中有一类方法基于数字相机照片提取冠层孔隙率,再用有限长度平均法同时估算LAI和聚集指数。然而,有限长度平均法自提出以来缺少进一步的发展,在有限长度的样线/样方上应用比尔定律的方式具有理论缺陷,可能造成无效值或高估LAI。从模拟的训练数据中提取经验公式以取代比尔定律进行样线/样方的LAI估算,提高了有限长度平均法的精度和鲁棒性。进一步分析在一定精度需求下对样线/样方大小和数量的要求,对于非均匀样地,提出样线长度为8倍等效叶片边长、样方边长为3倍等效叶片边长的推荐设置。在基于数字相机照片提取非均匀样地LAI的应用中,使用样方采样比样线采样更为适宜。  相似文献

枝条与叶片的生长关系是植物形成不同冠层结构充分利用空间资源的一种策略, 有利于植株通过构型调整增强自身的光合效率和竞争力, 以适应不同的生境条件。在石羊河上游高寒退化草地, 利用ArcGIS建立研究区域的数字高程模型(DEM), 并提取样地坡度数据, 采用标准化主轴估计(SMA)方法, 研究了不同坡度狼毒(Stellera chamaejasme)种群枝与叶的生长。结果表明: 随着坡度增大, 狼毒叶大小、枝长度和分枝数均呈逐渐减小趋势; 狼毒分枝数与枝长度、叶片数与枝长度均呈异速生长关系, 枝长度增加的速度大于叶片数增加的速度, 分枝数增加的速度大于枝长度增加的速度; 不同坡度间的比较显示, 较大坡度上狼毒分枝数与枝长度、叶片数与枝长度的异速斜率均较大, 在枝长度一定的条件下, 较大坡度的狼毒具有更大的叶片数与枝长度的比值和分枝数与枝长度的比值。坡度差异造成环境因子和植被群落环境的变化, 进而影响狼毒的资源利用策略, 表现为枝条与叶片构型以及二者之间关系的变化, 反映了毒杂草较强表型可塑性的适应机制。  相似文献

A digital image analysis method previously used to evaluate leaf color changes due to nutritional changes was modified to measure the severity of several foliar fungal diseases. Images captured with a flatbed scanner or digital camera were analyzed with a freely available software package, Scion Image, to measure changes in leaf color caused by fungal sporulation or tissue damage. High correlations were observed between the percent diseased leaf area estimated by Scion Image analysis and the percent diseased leaf area from leaf drawings. These drawings of various foliar diseases came from a disease key previously developed to aid in visual estimation of disease severity. For leaves of Nicotiana benthamiana inoculated with different spore concentrations of the anthracnose fungus Colletotrichum destructivum, a high correlation was found between the percent diseased tissue measured by Scion Image analysis and the number of leaf spots. The method was adapted to quantify percent diseased leaf area ranging from 0 to 90% for anthracnose of lily-of-the-valley, apple scab, powdery mildew of phlox and rust of golden rod. In some cases, the brightness and contrast of the images were adjusted and other modifications were made, but these were standardized for each disease. Detached leaves were used with the flatbed scanner, but a method using attached leaves with a digital camera was also developed to make serial measurements of individual leaves to quantify symptom progression. This was successfully applied to monitor anthracnose on N. benthamiana leaves. Digital image analysis using Scion Image software is a useful tool for quantifying a wide variety of fungal interactions with plant leaves.  相似文献

 经测定分析陕西南部28—37年生巴山松天然林的生物量和净初级生产量,结果表明,林分乔木层生物量为81.28t/ha,草本下木生物量为1.45t/ha,林分凋落物量为11.16t/ha;乔木层净初级生产量为6.94t/ha·yr,约45%分配到树叶。巴山松林的消光系数平均为0.33。在地上部生产结构方面,枝重主要集中在树冠中、下部,叶龄愈大,叶重在树冠下部的比例愈高。巴山松林木的根系总长度约99.4km/ha,其中细根占58%。  相似文献

Current methods for assessing leaf injury in Zostera marina (eelgrass) utilize subjective indexes for desiccation injury and wasting disease. Because of the subjective nature of these measures, they are inherently imprecise making them difficult to use in quantifying complex leaf injuries from multiple sources. We have developed a method using color digital photography of eelgrass leaves which are then manipulated using image processing programs and analyzed using geographic digital image analysis. The resulting false color images are then assigned by the user into uninjured and injured groupings which may then be reported as a percentage of leaf area affected. If images are rectified, leaf area (cm2) of injured and uninjured leaf segments may be determined. Although this method is time consuming and still requires some subjective judgments, it does allow for precise analysis of highly complex leaf injuries and has the potential to be a substantial improvement over existing leaf injury indexes.  相似文献
