Eurasian lynx (Lynx lynx) have a wide distribution across Eurasia. The northern edge of this distribution is in Norway, where they reach up to 72 degrees north. We conducted a study of lynx space use in this region from 2007 to 2013 using GPS telemetry. The home range sizes averaged 2,606 (± 438 SE) km2 for males (n = 9 ranges) and 1,456 (± 179 SE) km2 for females (n = 24 ranges). These are the largest home ranges reported for any large felid, and indeed are only matched by polar bears, arctic living wolves, and grizzly bears among all the Carnivora. The habitat occupied was almost entirely treeless alpine tundra, with home ranges only containing from 20% to 25% of forest. These data have clear implications for the spatial planning of lynx management in the far north as the current management zones are located in unsuitable habitats and are not large enough to encompass individual lynx movements.  相似文献

Site fidelity refers to the restriction of dispersal distance of an animal and its tendency to return to a stationary site. To our knowledge, the homing ability of freshwater turtles and their fidelity is reportedly very low in Asia. We examined mark–recapture data spanning a 4‐year period in Diaoluoshan National Nature Reserve, Hainan Province, China, to investigate the site fidelity and homing behavior of big‐headed turtles Platysternon megacephalum. A total of 11 big‐headed turtles were captured, and all individuals were used in this mark–recapture study. The site fidelity results showed that the adult big‐headed turtles (n = 4) had a 71.43% recapture rate in the original site after their release at the same site, whereas the juveniles (n = 1) showed lower recapture rates (0%). Moreover, the homing behavior results showed that the adults (n = 5) had an 83.33% homing rate after displacement. Adult big‐headed turtles were able to return to their initial capture sites (home) from 150 to 2,400 m away and precisely to their home sites from either upstream or downstream of their capture sites or even from other streams. However, none of the juveniles (n = 4) returned home, despite only being displaced 25–150 m away. These results indicated that the adult big‐headed turtles showed high fidelity to their home site and strong homing ability. In contrast, the juvenile turtles may show an opposite trend but further research is needed.  相似文献

The use of camera traps in ecology helps affordably address questions about the distribution and density of cryptic and mobile species. The random encounter model (REM) is a camera‐trap method that has been developed to estimate population densities using unmarked individuals. However, few studies have evaluated its reliability in the field, especially considering that this method relies on parameters obtained from collared animals (i.e., average speed, in km/h), which can be difficult to acquire at low cost and effort. Our objectives were to (1) assess the reliability of this camera‐trap method and (2) evaluate the influence of parameters coming from different populations on density estimates. We estimated a reference density of black bears (Ursus americanus) in Forillon National Park (Québec, Canada) using a spatial capture–recapture estimator based on hair‐snag stations. We calculated average speed using telemetry data acquired from four different bear populations located outside our study area and estimated densities using the REM. The reference density, determined with a Bayesian spatial capture–recapture model, was 2.87 individuals/10km2 [95% CI: 2.41–3.45], which was slightly lower (although not significatively different) than the different densities estimated using REM (ranging from 4.06–5.38 bears/10km2 depending on the average speed value used). Average speed values obtained from different populations had minor impacts on REM estimates when the difference in average speed between populations was low. Bias in speed values for slow‐moving species had more influence on REM density estimates than for fast‐moving species. We pointed out that a potential overestimation of density occurs when average speed is underestimated, that is, using GPS telemetry locations with large fix‐rate intervals. Our study suggests that REM could be an affordable alternative to conventional spatial capture–recapture, but highlights the need for further research to control for potential bias associated with speed values determined using GPS telemetry data.  相似文献

Population studies often incorporate capture‐mark‐recapture (CMR) techniques to gather information on long‐term biological and demographic characteristics. A fundamental requirement for CMR studies is that an individual must be uniquely and permanently marked to ensure reliable reidentification throughout its lifespan. Photographic identification involving automated photographic identification software has become a popular and efficient noninvasive method for identifying individuals based on natural markings. However, few studies have (a) robustly assessed the performance of automated programs by using a double‐marking system or (b) determined their efficacy for long‐term studies by incorporating multi‐year data. Here, we evaluated the performance of the program Interactive Individual Identification System (I3S) by cross‐validating photographic identifications based on the head scale pattern of the prairie lizard (Sceloporus consobrinus) with individual microsatellite genotyping (N = 863). Further, we assessed the efficacy of the program to identify individuals over time by comparing error rates between within‐year and between‐year recaptures. Recaptured lizards were correctly identified by I3S in 94.1% of cases. We estimated a false rejection rate (FRR) of 5.9% and a false acceptance rate (FAR) of 0%. By using I3S, we correctly identified 97.8% of within‐year recaptures (FRR = 2.2%; FAR = 0%) and 91.1% of between‐year recaptures (FRR = 8.9%; FAR = 0%). Misidentifications were primarily due to poor photograph quality (N = 4). However, two misidentifications were caused by indistinct scale configuration due to scale damage (N = 1) and ontogenetic changes in head scalation between capture events (N = 1). We conclude that automated photographic identification based on head scale patterns is a reliable and accurate method for identifying individuals over time. Because many lizard or reptilian species possess variable head squamation, this method has potential for successful application in many species.  相似文献

Intramyocellular lipid (IMCL) utilization is impaired in older individuals, and IMCL accumulation is associated with insulin resistance. We hypothesized that increasing muscle total carnitine content in older men would increase fat oxidation and IMCL utilization during exercise, and improve insulin sensitivity. Fourteen healthy older men (69 ± 1 year, BMI 26.5 ± 0.8 kg/m2) performed 1 h of cycling at 50% VO2max and, on a separate occasion, underwent a 60 mU/m2/min euglycaemic hyperinsulinaemic clamp before and after 25 weeks of daily ingestion of a 220 ml insulinogenic beverage (44.4 g carbohydrate, 13.8 g protein) containing 4.5 g placebo (n = 7) or L‐carnitine L‐tartrate (n = 7). During supplementation, participants performed twice‐weekly cycling for 1 h at 50% VO2max. Placebo ingestion had no effect on muscle carnitine content or total fat oxidation during exercise at 50% VO2max. L‐carnitine supplementation resulted in a 20% increase in muscle total carnitine content (20.1 ± 1.2 to 23.9 ± 1.7 mmol/kg/dm; p < 0.01) and a 20% increase in total fat oxidation (181.1 ± 15.0 to 220.4 ± 19.6 J/kg lbm/min; p < 0.01), predominantly due to increased IMCL utilization. These changes were associated with increased expression of genes involved in fat metabolism (ACAT1, DGKD & PLIN2; p < 0.05). There was no change in resting insulin‐stimulated whole‐body or skeletal muscle glucose disposal after supplementation. This is the first study to demonstrate that a carnitine‐mediated increase in fat oxidation is achievable in older individuals. This warrants further investigation given reduced lipid turnover is associated with poor metabolic health in older adults.  相似文献

This study aimed to explore the function of IFN‐γ+IL‐17+Th17 cells on fibrosis in systemic scleroderma (SSc). Blood and skin samples were collected from 20 SSc cases and 10 healthy individuals. The percentage of IFN‐γ+IL‐17+Th17 cells was detected using flow cytometry. The in vitro induction of IFN‐γ+IL‐17+Th17 cells was performed adopting PHA and rIL‐12. Gene expression was detected via quantitative real‐time polymerase chain reaction (qRT‐PCR), whereas western blot analysis was adopted for protein analysis. The distribution of IFN‐γ+IL‐17+Th17 cells was significantly increased in SSc cases and positively correlated with SSc stages (P = .031), disease duration (P = .016), activity (P = .025) and skin scores (P < .001). In vitro, IFN‐γ+IL‐17+Th17 cells could promote the expressions of α‐SMA and COL1A1, revealing increased fibroblasts’ proliferation and enhanced collagen‐secreting capacity. In addition, IL‐21 expression was significantly increased in co‐culture medium of IFN‐γ+IL‐17+Th17 cells and fibroblasts (P < .001). IL‐21 neutralizer treatment resulted in the down‐regulation of α‐SMA and COL1A1. IL‐21 was confirmed as an effector of IFN‐γ+IL‐17+Th17 cells in fibrosis process. The distribution of IFN‐γ+IL‐17+Th17 cells was significantly increased in SSc cases and positively correlated with disease activity. IFN‐γ+IL‐17+Th17 cells could promote fibroblast proliferation and enhance collagen‐secreting ability via producing IL‐21, thus contributing to fibrosis in SSc.  相似文献

In genetically heterogeneous mice produced by the CByB6F1 x C3D2F1 cross, the “non‐feminizing” estrogen, 17‐α‐estradiol (17aE2), extended median male lifespan by 19% (p < 0.0001, log‐rank test) and 11% (p = 0.007) when fed at 14.4 ppm starting at 16 and 20 months, respectively. 90th percentile lifespans were extended 7% (p = 0.004, Wang–Allison test) and 5% (p = 0.17). Body weights were reduced about 20% after starting the 17aE2 diets. Four other interventions were tested in males and females: nicotinamide riboside, candesartan cilexetil, geranylgeranylacetone, and MIF098. Despite some data suggesting that nicotinamide riboside would be effective, neither it nor the other three increased lifespans significantly at the doses tested. The 17aE2 results confirm and extend our original reports, with very similar results when started at 16 months compared with mice started at 10 months of age in a prior study. The consistently large lifespan benefit in males, even when treatment is started late in life, may provide information on sex‐specific aspects of aging.  相似文献

  • 1 The severe and early destruction and fragmentation of woodland habitats due to human activities is thought to have been a leading factor in the extirpation from Britain of several large, forest‐dependent mammal species, such as the Eurasian lynx Lynx lynx. However, during the 20th century, Scotland in particular has experienced rapid, large‐scale reafforestation. In order to assess if this reafforestation has been sufficient to permit the potential restoration of extirpated forest mammal species with large spatial requirements, a Geographical Information System (GIS) analysis of potential habitat of one species, the Eurasian lynx, was performed for the Scottish mainland.
  • 2 A rule‐based analysis, incorporating data and expert opinion from Switzerland, an environmentally similar area where lynx now occur, was used to identify patches of suitable lynx habitat in Scotland. A connectivity analysis was used to investigate whether and how these patches are connected to form larger interconnected networks of potential lynx habitat that would allow lynx to sufficiently interact with one another to form a single interbreeding population.
  • 3 Scotland has over 20 000 km2 of suitable lynx habitat split into two main networks of interconnected patches: the Highlands (c. 15 000 km2) and the Southern Uplands (c. 5000 km2). A further 800 km2 of potential habitat, contiguous with the Southern Uplands lynx habitat network, lies across the border in England. Although connectivity between the Highlands and Southern Uplands networks is currently weak, the implementation of measures to mitigate the barrier effects of busy roads in central Scotland could facilitate the movement of lynx between the two areas.
  • 4 Based on the availability of prey resources, Scotland could support around 400 adult and subadult lynx in the Highlands and around 50 in the Southern Uplands. A Scottish population of this size would be the fourth largest lynx population in Europe considering current population estimates.

ObjectivesIdiopathic pulmonary fibrosis (IPF) is marked by the excessive accumulation of extracellular matrix, which participates in a variety of chronic diseases or injuries and seriously threatens human health. Due to the side effects of clinical drugs, there is still a need to develop novel and less toxic drugs to treat pulmonary fibrosis.Materials and MethodsSKLB‐YTH‐60 was developed through computer‐aided drug design, de novo synthesis and high‐throughput screening. We employed the bleomycin (BLM)‐induced lung fibrosis animal models and used TGF‐β1 to induce the epithelial‐mesenchymal transition (EMT) of A549 cells in vitro. Meanwhile, the protein expression of collagen I and the α‐smooth muscle actin (α‐SMA), E‐cadherin, p‐FGFR1, p‐PLCγ, p‐Smad2/3 and p‐Erk1/2 was detected by western blot.ResultsYTH‐60 has obvious anti‐proliferative activity on fibroblasts and A549 cells. Moreover, YTH‐60 could impair the EMT of A549 cells and suppressed fibrosis by inhibiting FGFR and TGF‐β/Smad‐dependent pathways. Intraperitoneal administration of preventive YTH‐60 could significantly reduce the degree of fibrosis in mice and regulate the imbalance of the immune microenvironment. In addition, we observed that therapeutic YTH‐60 treatment attenuated fibrotic changes in mice during the period of fibrosis. Importantly, YTH‐60 has shown an acceptable oral bioavailability (F = 17.86%) and appropriate eliminated half‐life time (T 1/2 = 8.03 hours).ConclusionsTaken together, these preclinical evaluations suggested that YTH‐60 could be a promising drug candidate for treating IPF.  相似文献

Laboratory protocols using magnetic beads have gained importance in the purification of mRNA for vaccines. Here, the produced mRNA hybridizes specifically to oligo(dT)‐functionalized magnetic beads after cell lysis. The mRNA‐loaded magnetic beads can be selectively separated using a magnet. Subsequently, impurities are removed by washing steps and the mRNA is eluted. Magnetic separation is utilized in each step, using different buffers such as the lysis/binding buffer. To reduce the time required for purification of larger amounts of mRNA vaccine for clinical trials, high‐gradient magnetic separation (HGMS) is suitable. Thereby, magnetic beads are selectively retained in a flow‐through separation chamber. To meet the requirements of biopharmaceutical production, a disposable HGMS separation chamber with a certified material (United States Pharmacopeia Class VI) was developed which can be manufactured using 3D printing. Due to the special design, the filter matrix itself is not in contact with the product. The separation chamber was tested with suspensions of oligo(dT)‐functionalized Dynabeads MyOne loaded with synthetic mRNA. At a concentration of cB = 1.6–2.1 g·L–1 in lysis/binding buffer, these 1 μm magnetic particles are retained to more than 99.39% at volumetric flows of up to 150 mL·min–1 with the developed SU‐HGMS separation chamber. When using the separation chamber with volumetric flow rates below 50 mL·min–1, the retained particle mass is even more than 99.99%.  相似文献

Poly(ADP‐ribose) polymerase‐1 (PARP‐1) is a nuclear chromatin‐associated enzyme involved in the DNA damage response. SNP rs1136410 C>T, the most studied polymorphism in PARP‐1 gene, is highly implicated in the susceptibility of cancer. However, the roles of PARP‐1 rs1136410 C>T on cancer risk vary from different studies. We comprehensively screened all qualified publications from several databases, including PubMed, EMBASE, MEDLINE, CNKI and Wanfang. The searching was updated to April 2020. Our meta‐analysis included 60 articles with 65 studies, comprised of a total of 23 996 cases with cancer and 33 015 controls. Overall, pooled data showed that the PARP‐1 rs1136410 C>T polymorphism was significantly but a border‐line associated with an increased risk of overall cancer (CC vs. TT/TC: OR = 1.11, 95% CI = 1.00‐1.24; C vs T: OR = 1.07, 95% CI = 1.01‐1.14). Subgroup analysis indicated that rs1136410 C allele contributed to high risk among gastric, thyroid, and cervical cancer, but lower risk among brain cancer. Furthermore, increased cancer risk was detected in the subgroups of Asian, controls from population‐based design studies, and HWE ≤ 0.05 studies. Sensitivity analysis and Egger''s test showed that results of the meta‐analysis were fairly stable. The current study indicated that PARP1 rs1136410 C>T polymorphism may have an impact on certain types of cancer susceptibility.  相似文献

The ecotype population of goats (Capra hircus) was created by long‐term artificial selection and natural adaptation. Mile red‐bone goat is an indigenous breed with visible red bones, and its special bone structure has received extensive attention. This study aimed to identify genetic variants and candidate genes associated with specific bone phenotypes using next‐generation sequencing technology (NGS). The results revealed that 31,828,206 single nucleotide polymorphisms (SNPs) were obtained from 72 goats (20 Mile red‐bone goats and 52 common goats) by NGS. A total of 100 candidate genes were identified on the basis top 1% window interaction from nucleotide diversity (π), π ratio (π A/π B), and pairwise fixation index (F ST). Exactly 77 known signaling pathways were enriched. Specifically, three coding genes (NMNAT2, LOC102172983, and PNLIP) were annotated in the vitamin metabolism signaling pathways, and NCF2 was annotated to the osteoclast (OC) differentiation pathway. Furthermore, 5862 reliable copy number variations (CNVs) were obtained, and 14 and 24 genes were annotated with the top 1‰ CNV based on F ST (>0.490) and V ST (>0.527), respectively. Several pathways related to bone development and metabolism of exogenous substances in vivo, including calcium signaling pathway, OC differentiation, and glycerophospholipid metabolism, were annotated. Specifically, six genes from 19 candidate CNVs, which were obtained by interaction of the top 1‰ CNVs with F ST and V ST, were annotated to mucin‐type O‐glycan biosynthesis and metabolic pathways. Briefly, the results implied that pseudopurpurin and specific genetic variants work together to contribute to the red‐bone color and specific bone structure of Mile red‐bone goat. This study is helpful to understanding the genetic basis of the unique bone phenotype of Mile red‐bone goats.  相似文献

We examined vervet monkey (Chlorocebus pygerythrus) space use using GPS/UHF telemetry data from 10 vervet monkeys across six troops over 9 months within a 420 ha suburban eco‐estate. We documented a mean home range of 0.99 km2 (95% MCP) and 1.07 km2 (95% KDE) for females (n = 6), 1 km2 (95% MCP) and 1.50 km2 (95% KDE) for males (n = 4) and 0.87 km2 (95% MCP) and 1.12 km2 (95% KDE) for troops (n = 6), respectively, indicating that males and larger troops had larger home ranges. These relatively small home ranges included shared territorial boundaries and high home range overlap. Vervet monkey movements indicated higher morning activity levels, and habitat selection indicated significantly more use of golf course, urban residential and forest, thicket and woodland areas, and avoidance of wetland, grassland and shrub, and urban built‐up areas. Our results suggest that modified habitat use by vervet monkeys is a consequence of behavioural facilitation to access highly available food resources, thereby facilitating their persistence in green spaces in urban areas of South Africa. Conflict management is dependent on the conservation of sufficient natural habitats and food resources, to minimise their dependence on anthropogenic supplementary food resources and consequently reduce human–monkey conflict.  相似文献

Off the Ningaloo coast of North West Western Australia, Spangled Emperor Lethrinus nebulosus are among the most highly targeted recreational fish species. The Ningaloo Reef Marine Park comprises an area of 4,566 km2 of which 34% is protected from fishing by 18 no-take sanctuary zones ranging in size from 0.08–44.8 km2. To better understand Spangled Emperor movements and the adequacy of sanctuary zones within the Ningaloo Reef Marine Park for this species, 84 Spangled Emperor of a broad spectrum of maturity and sex were tagged using internal acoustic tags in a range of lagoon and reef slope habitats both inside and adjacent to the Mangrove Bay Sanctuary zone. Kernel Utilisation Distribution (KUD) was calculated for 39 resident individuals that were detected for more than 30 days. There was no relationship with fish size and movement or site fidelity. Average home range (95% KUD) for residents was 8.5±0.5 km2 compared to average sanctuary zone size of 30 km2. Calculated home range was stable over time resulting in resident animals tagged inside the sanctuary zone spending ∼80% of time within the sanctuary boundaries. The number of fish remaining within the array of receivers declined steadily over time and after one year more than 60% of tagged fish had moved outside the sanctuary zone and also beyond the 28 km2 array of receivers. Long term monitoring identified the importance of shifting home range and was essential for understanding overall residency within protected areas and also for identifying spawning related movements. This study indicates that despite exhibiting stable and small home ranges over periods of one to two years, more than half the population of spangled emperor move at scales greater than average sanctuary size within the Ningaloo Reef Marine Park.  相似文献

Abstract Canada lynx (Lynx canadensis) were listed as a federally threatened species in 14 states at the southern extent of their geographic range in March 2000, with Maine being the only state in the northeastern United States known to support a resident population. Relatively little information is known about the ecology of lynx living at the southern edge of their range, including range requirements, movements, and spatial organization. Basic knowledge of lynx ecology is needed for federal recovery planning efforts. Between 1999 and 2004, we trapped and radiocollared 43 lynx (21 M, 22 F) in northern Maine in an intensively managed and predominantly early successional forested landscape. We estimated diurnal annual and seasonal home-range size for male and female lynx using the 85% fixed-kernel home-range estimator. Annual home ranges of adult male lynx (x̄ = 53.6 km2) were more than twice the size of adult female home ranges (x̄ = 25.7 km2). Home ranges of adult females during snow periods (x̄ = 38.3 km2) were nearly 3 times larger than their snow-free-period ranges (x̄ = 14.3 km2), whereas, snow-free ranges of adult males (x̄ = 58.8 km2) were slightly larger than their snow-period ranges (x̄ = 45.2 km2). We observed a limited amount of home-range overlap among lynx of the same sex (F: x̄ = 17.2%; M: x̄ = 11.8%). Lynx of opposite sex showed more extensive overlap (x̄ = 24.3%). Most home-range shifts of resident lynx were typically not extensive. Based on territory mapping, we estimated a minimum lynx density of 9.2–13.0 lynx/100 km2. We observed lynx spatial ecology and densities that were more similar to northern lynx populations when hares were abundant than to other southern lynx populations, suggesting that region-specific studies under varying habitat conditions and hare densities are needed to ensure realistic recovery goals and effective management of lynx at the southern extent of their range.  相似文献

AimUnderstanding spatial patterns of the distribution of adult native oyster, Ostrea edulis, and the invasive Magallana (Crassostrea) gigas is important for management of these populations. The aim of this study was to use ensemble SDM’s to (a) identify and predict conservation hotspots, (b) assess the current level of protection for O. edulis, and (c) quantify the amount of overlap between the two species where interactions with M. gigas are most likely.LocationSkagerrak, Sweden.MethodsWe used data collected by video at depths from 0.5 to 10 m in 436 sites. Models of occurrence and densities >1 m−2 were fitted and assessed using ensemble methods (“biomod2” package). Models of high‐density hotspots were used to predict, map, and quantify areal extent of the species in order to assess the degree of overlap with protected areas and the potential for interactions between the two species.ResultsBoth species were widely distributed in the region. Observations of high‐density habitats, mainly occurring at depths of ≈3 and 0.5 m for O. edulis and M. gigas, respectively, were found in 4% and 2% of the sites. Models provided useful predictions for both species (AUC = 0.85–0.99; sensitivity = 0.74–1.0; specificity = 0.72–0.97). High‐density areas occupy roughly 15 km2 each with substantial overlap between species. 50% of these are protected only by fisheries regulations, 44% are found in Natura 2000 reserves and 6% of the predicted O. edulis enjoys protection in a national park.Main conclusionsData collection by video in combination with SDM’s provides a realistic approach for large‐scale quantification of spatial patterns of marine population and habitats. O. edulis and M. gigas are common in the area, but a large proportion of the most valuable O. edulis habitats are not found in protected areas. The overlap between species suggests that efforts to manage the invasive M. gigas need to be integrated with management actions to conserve the native O. edulis.  相似文献

Human‐mediated habitat fragmentation in freshwater ecosystems can negatively impact genetic diversity, demography, and life history of native biota, while disrupting the behavior of species that are dependent on spatial connectivity to complete their life cycles. In the Alouette River system (British Columbia, Canada), dam construction in 1928 impacted passage of anadromous sockeye salmon (Oncorhynchus nerka), with the last records of migrants occurring in the 1930s. Since that time, O. nerka persisted as a resident population in Alouette Reservoir until experimental water releases beginning in 2005 created conditions for migration; two years later, returning migrants were observed for the first time in ~70 years, raising important basic and applied questions regarding life‐history variation and population structure in this system. Here, we investigated the genetic distinctiveness and population history of Alouette Reservoir O. nerka using genome‐wide SNP data (n = 7,709 loci) collected for resident and migrant individuals, as well as for neighboring anadromous sockeye salmon and resident kokanee populations within the Fraser River drainage (n = 312 individuals). Bayesian clustering and principal components analyses based on neutral loci revealed five distinct clusters, largely associated with geography, and clearly demonstrated that Alouette Reservoir resident and migrant individuals are genetically distinct from other O. nerka populations in the Fraser River drainage. At a finer level, there was no clear evidence for differentiation between Alouette Reservoir residents and migrants; although we detected eight high‐confidence outlier loci, they all mapped to sex chromosomes suggesting that differences were likely due to uneven sex ratios rather than life history. Taken together, these data suggest that contemporary Alouette Reservoir O. nerka represents a landlocked sockeye salmon population, constituting the first reported instance of deep‐water spawning behavior associated with this life‐history form. This finding punctuates the need for reassessment of conservation status and supports ongoing fisheries management activities in Alouette Reservoir.  相似文献

Recent studies have demonstrated a marked decrease in peripheral lymphocyte levels in patients with coronavirus disease 2019 (COVID‐19) caused by severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2). Few studies have focused on the changes of NK, T‐ and B‐cell subsets, inflammatory cytokines and virus‐specific antibodies in patients with moderate COVID‐19. A total of 11 RT‐PCR‐confirmed convalescent patients with COVID‐19 and 11 patients with non‐SARS‐CoV‐2 pneumonia (control patients) were enrolled in this study. NK, CD8+ T, CD4+ T, Tfh‐like and B‐cell subsets were analysed using flow cytometry. Cytokines and SARS‐CoV‐2‐specific antibodies were analysed using an electrochemiluminescence immunoassay. NK cell counts were significantly higher in patients with COVID‐19 than in control patients (P = 0.017). Effector memory CD8+ T‐cell counts significantly increased in patients with COVID‐19 during a convalescent period of 1 week (P = 0.041). TIM‐3+ Tfh‐like cell and CD226+ Tfh‐like cell counts significantly increased (P = 0.027) and decreased (P = 0.022), respectively, during the same period. Moreover, ICOS+ Tfh‐like cell counts tended to decrease (P = 0.074). No abnormal increase in cytokine levels was observed. The high expression of NK cells is important in innate immune response against SARS‐CoV‐2. The increase in effector memory CD8+ T‐cell counts, the up‐regulation of inhibitory molecules and the down‐regulation of active molecules on CD4+ T cells and Tfh‐like cells in patients with COVID‐19 would benefit the maintenance of balanced cellular and humoural immune responses, may prevent the development of severe cases and contribute to the recovery of patients with COVID‐19.  相似文献

Optimal foraging models predict that individual animals will optimize net energy gain by intensifying forage activity and/or reducing forage energy cost. Then, the free distribution model predicts an animal''s distribution in a patchy landscape will match the distribution of the resources. If not modified by other factors, such patterns may be expected to be particularly explicit in variable and extreme, forage‐limited, and patchy environments, notably alpine and Arctic environments during winter. The large ungulate wild mountain reindeer (Rangifer tarandus tarandus) surviving in such environments is used as a model during the forage‐limited winter season. The largest wild reindeer area in Western Europe (Hardangervidda, 8130 km2) is actively managed to sustain 10,000–12,000 wild reindeer. Since 2001, 104 different individuals have been GPS‐tracked at 3‐hr intervals. In winter, mountain reindeer may either choose to seek out and forage in patchy snow‐free habitats, typically on top of wind‐blown ridges, or use energy‐demanding digging through the snow to reach ground forage (cratering). We use late April satellite data from Landsat 5 and 8 (30 × 30 m), airborne laser scanning subsampling (processed to 1 × 1 m grid), and topographic information (1 m resolution) derived from digital aerial photographs (0.25 × 0.25 m resolution) to delineate snow‐free patches, constituting less than 694 km2. By overlaying recorded wild reindeer GPS positions winters 2001–2017 (188,942 positions), we document a strong positive selection for snow‐free patches, which were used about four times more frequently than expected from a “random walk” model. On a daily basis, the preference for snow‐free areas was slightly stronger in the evenings. In the sustainable management of wild mountain reindeer, the area of snow‐free patches is an important predictor of winter forage availability and important winter source areas.
