Three doubled dihaploid potatoes were hybridised with six tetraploid cultivars and the offspring examined for commercially important characters. Doubled dihaploids used as parents did not reduce the within progeny phenotypic variation for most characters below that found in material produced by intercrossing heterozygous tetraploids. They could however prove useful for improving some characters in a breeding programme. The products of the hybridisations were used to examine the genetic control of discrete variables such as tuber skin pigmentation, tuber flesh colour, and skin russetting. Doubled dihaploids were found to be useful for such studies because they have only three out of the five possible tetraploid genotypes. Presence or absence of red tuber skin in the progenies of cv. Désirée was concluded to be due to the segregation of a dominant major gene which was simplex in the parental cultivar. The yellow flesh of Desiree was found to be recessive. On the hypothesis that russet skin was due to three complementary dominant genes, possible genotypes of the doubled dihaploids and cultivars could also be deduced.  相似文献

Numerically unreduced (2n) gametes from first division restitution (FDR) are considered to be superior to 2n-gametes from second division restitution (SDR) because they transfer a larger proportion of the total parental heterozygosity and epistasis intact to the tetraploid progeny. This supposed superiority was investigated by comparing 12 sets of reciprocal 4x-2x crosses. Each diploid parent used in a reciprocal set produced 2n-pollen by FDR and 2n-eggs by SDR. Six agronomic characters were investigated. FDR progenies (from 4x.2x) were found to have higher mean yields due to more and bigger tubers. With respect to underwater weight, the overall progeny mean of FDR progenies was significantly higher than that of SDR progenies (from 2x.4x). However, the absolute difference found between both overall progeny means was too small to be of practical significance. No differences between FDR and SDR progeny means were found for vine maturity and chip colour. In addition to the progeny mean, within-progeny variation is important in potato breeding. For vine maturity a higher within-progeny variation was detected in SDR progenies, whereas within-progeny variations for yield, underwater weight and chip colour were not different in FDR and SDR progenies. With regard to vine maturity, we conclude that SDR 2n-gametes are superior to FDR 2n-gametes because, with the same progeny means of FDR and SDR progenies, the within-progeny variation was higher in SDR progenies. Therefore the assumed superiority of FDR 2n-gametes was confirmed for yield but was not observed for vine maturity, underwater weight and chip colour.  相似文献

A diallel set of crosses, including selfs and some reciprocal crosses, was made between 15 parents, chosen for their fertility, from those included in a tetraploid potato (Solanum tuberosum subsp. tuberosum) breeding programme at the Scottish Crop Research Institute. The progenies were grown in randomised complete block trials with two replicates at a high-grade seed site from 1994 to 1996 inclusive and at a ware site in 1995 and 1996. The parents were included in the ware trials. Tubers were assessed for visual preference in all trials and for fry colour at both sites in 1996. Emergence and maturity were recorded in the ware trials and the tubers were assessed for yield, dry matter, size, appearance (regularity of shape), scab, uniformity, sprouting in store and keeping quality. There were very few growth cracks and very few internal defects. No reciprocal differences were found. Inbreeding depression was marked for emergence, yield, tuber size and appearance, and visual preference. In contrast, the selfs had a lighter fry colour than the parents and F1s. Combining-ability analysis (selfs omitted) identified fry colour, emergence, maturity, yield, dry matter and sprouting resistance as traits for which the GCA (general combining ability) variance and narrow-sense heritability were high enough for good progress from full-sib family selection. Correlations between GCAs for pairs of traits were examined, including those from previously published seedling progeny tests. For fry colour, an unfavourable correlation with low yield (r = 0.596) was compensated by a favourable one with high dry matter content (r = 0.652), whereas unfavourable ones between foliage and tuber blight resistance and sprouting susceptibility (r = 0.578 and 0.596) were identified for monitoring. Clones with high GCAs were identified for use as parents in future breeding and the extent to which GCAs could be predicted from the parents, and the offspring means from the midparent means, was determined by regression and correlation analysis. The offspring-midparent regression was highest for fry colour, followed by dry matter, emergence and sprouting. Values were lower for scab due to environmental variation; for maturity, yield and tuber size due to SCA (specific combining ability); and for visual preference due to both factors.  Two diploid potato parents, W5295.7 and W5337.3, and their colchicine-doubled tetraploids were used as male to cross with three cultivars: ‘Raritan’, ‘Shepody’ and ‘Superior’. Both diploids produced 2n gametes via first division restitution (FDR) during meiosis. Field experiments were carried out on seedlings of the 12 hybrid progenies in 1991 and 1992. Six of the progenies represented the tetraploid-diploid (4x-2x) hybrids and the other six tetraploid-tetraploid (4x-4x) progenies. The genetic consequence of FDR gametes was examined by comparing the mean and genotypic variance of six tuber traits of 4x-2x and 4x-4x progenies. Tuber appearance, eye depth, specific gravity and tuber size showed significant differences between the means of 4x-2x and 4x-4x progenies from either both or one of the diploid parents. The genotypic variances of these traits in 4x-2x progenies were reduced in various degrees from those observed in 4x-4x progenies. Major genes responsible for the inheritance of these traits are probably located close to the centromere. The two diploid parents may also carry alleles of different size and direction of non-additive genic effects in their 2n gametes. The 4x-2x progenies also outperformed the 4x-4x progenies with respect to the means of total and marketable yield. There were thus heterotic gene loci for high yield located close to the centromere. No marked reduction of genotypic variance, however, was observed. The results suggest that genes for yield would be scattered between the centromere and the site for maximum recombination. Summary The objective of this research was to compare the efficiency of the 4x×2x breeding scheme with the traditional 4x×4x method with respect to potato improvement. The basis for such a comparison was the parental value of four 2x and four 4x male parents from the International Potato Center (CIP) as measured by multitrait selection and progeny testing. The 2x parents produced 2n pollen by parallel spindles at anaphase II, which is genetically equivalent to a first division restitution (FDR) mechanism. Both 2x and 4x parents were crossed with four common 4x female parents. Thus, 32 families were evaluated over 2 years at four Peruvian locations. A selection index which considered tuber yield, tuber number, average tuber weight and specific gravity was used for multitrait selection. Three FDR 2x parents had better selection index scores than the 4x parents over the four locations. Estimates of broad-sense heritability for total yield using different number of replications and locations were calculated by using the variance components. The 4x × 2x breeding scheme was found to be better than the traditional 4x × 4x method since fewer replications and locations are required to evaluate tuber yield in 4x × 2x progenies than in 4x × 4x progenies. The FDR 2x parents were also better material than the 4x parents for testing combining ability for tuber yield of the 4x progenitors. This could be the result of the mode of FDR 2n pollen formation. A diallel set of crosses, including selfs and some reciprocal crosses, was made between 15 parents chosen for their male fertility from those included in a tetraploid potato (Solanum tuberosum subsp. tuberosum) breeding programme at the Scottish Crop Research Institute. Seedling progeny tests were used to evaluate the progenies for non-race-specific resistance to late blight (Phytophthora infestans) in both foliage and tubers, quantitative resistance to the white potato cystnematode (PCN) (Globodera pallida) and the commercial worth of their tubers as judged by breeders' visual preference. No reciprocal differences were found. Comparisons of the selfs and crosses revealed inbreeding depression for breeders' preference, which varied among the parents from negligible to severe, whilst there were also statistically significant differences for foliage and tuber blight, but not for PCN. When the selfs were omitted from the combining ability analyses, large differences in general combining ability (GCA) were found for all four traits, and smaller differences in specific combining ability for tuber blight and breeders' preference. The only statistically significant correlation between GCAs for different traits was a favourable one of r = 0.56 between foliage and tuber resistance to late blight. It was concluded that prospects were good for simultaneously improving all four traits by multitrait genotypic recurrent selection.  相似文献

A number of recent studies have provided evidence that potato dihaploids (S. tuberosum) contain and express DNA from the male (dihaploid inducer) parent, S. phureja. The importance of this for breeding programmes that use dihaploid potatoes is to some extent dependent upon whether the S. phureja DNA influences dihaploid morphology. In the present study, 21 characters were used to compare the morphology of six dihaploids with those of their parents: S. tuberosum (cvs `Pentland Dell' and `Pentland Crown') and S. phureja (IVP48). Characteristics of S. phureja were found in all of the dihaploids examined. In principal component analyses, dihaploids formed intermediate groupings positioned between those of the parents, although much closer to S. tuberosum. It is concluded there is evidence that DNA originating from the dihaploid inducer can affect the morphology of potato dihaploids. Implications of the findings are discussed. Summary The level and mode of 2n megaspore formation was studied in full-sib diploid potato clones with either normal or desynaptic (ds-1ds-1) meiosis. Cytological analysis revealed that functional 2n megaspores produced by normal and desynaptic clones originate exclusively from second division restitution (SDR) and first division restitution (FDR), respectively. SDR 2n megaspores resulted from the omission of the second meiotic division following chromosome doubling after anaphase I, whereas FDR 2n megaspores resulted from a direct equational division of univalent chromosomes at anaphase I (pseudohomotypic division). Comparative data strongly indicated that the observed mechanisms of SDR and FDR 2n megaspore formation are extremes of a continuum that is being brought about by common genes for precocious chromosome division. Depending on the relative timing of cell cycle and chromosome division, this precocious chromosome division may impose postreductional (SDR) or prereductional (FDR) restitution of the sporophytic chromosome number under normal synaptic and desynaptic conditions, respectively. The observed frequencies of 2n megaspores closely correlated with seed set, following pollination by tetraploid varieties and by desynaptic diploid clones with exclusive FDR 2n pollen formation. Up to 54.0 and 21.5 seeds/ fruit were obtained from normal synaptic (SDR) and desynaptic (FDR) progeny, respectively. The high frequency of segregants with either SDR or FDR 2n megaspore formation (78.0 and 45.2%, respectively) supports the hypothesis that sexual polyploidization is the driving force behind the origin and evolution of polyploid Solanum species. The present identification of diploid potato clones with consistent FDR 2n megaspore formation extends the opportunities for direct transfer of enhanced diploid germ plasm to tetraploids, and particularly advocates the feasibility of 2x(ds-1; FDR)×2x(ds-1; FDR) breeding schemes in cultivar development and the production of relatively vigorous and uniform true potato seed (TPS) varieties. 为了解桉树重要性状在杂交种亲本、子代间的遗传变异规律,以尾叶桉(Eucalyptus urophylla)为母本、粗皮桉(E. pellita)和细叶桉(E. tereticornis)为父本、8个全同胞杂交子代为材料,对亲本和子代的叶片形态性状进行了分析。结果表明,亲缘关系近的尾叶桉和粗皮桉的叶片性状大多无显著差异;但他们与亲缘关系远的细叶桉间的差异显著;尾粗桉与父母本间的性状无显著差异或仅与母本差异显著,属偏父本型;尾细桉与双亲间的多数性状都差异显著,且多偏向于母本。在树种内的家系间或个体间,叶片数量性状差异显著,而质量性状仅叶面革质厚度、叶面朝向差异显著;杂交种家系部分性状有超亲现象,且尾细桉家系超亲频率显著高于尾粗桉。叶长、叶长宽比、形状因子、叶色、叶面革质是区别叶片形态特征的决定性状。  相似文献

 The 2n-pollen grains formed by first-division restitution without crossover (FDR-NCO) are unique breeding tools, since they can transmit almost 100% of non-additive genetic effects from the parent to the progeny. FDR-NCO gametes are considered superior to those formed by FDR with crossing over (FDR-CO), which can pass on to the progenies approximately 80% of the heterozygosity and a large fraction of the epistasis. However, 2n-pollen formation by FDR-NCO mechanism requires the incorporation (in homozygous condition) of at least two recessive alleles. In the present work, 40 tetraploid families derived from complete 4x-2x factorial crosses were evaluated under short-day conditions to verify whether or not the postulated genetic superiority of FDR-NCO over FDR-CO gametes holds true for eight quantitative traits in potato. Families were derived from crosses between four 4x commercial cultivars, and a random sample of ten diploid Solanum phureja-haploid S. tuberosum hybrids producing 2n-pollen by either FDR-CO or FDR-NCO. The results indicated no significant superiority of FDR-NCO over FDR-CO families for total tuber yield (TTY) and six other traits (haulm maturity – HM; plant vigor – PV; plant uniformity – PU; eye depth – ED; number of tubers per hill – NTH; and commercial over total yield index – CTI). Based upon cytological observations, the FDR-CO and FDR-NCO gametes are expected to be genetically equivalent for all loci between the centromeres and the chromosomal site of maximum recombination. In our experiment, differences between FDR-CO- and FDR-NCO-derived progenies were not observed for TTY. Therefore, our results can be interpreted as additional evidence for the hypothesis that genes with major effect on TTY expression might have a physical location between centromeres and proximal crossovers in the potato chromosomes. In addition, a similar trend was observed for HM, PV, and ED but apparently not for commercial yield – CY (i.e., tubers with more than 33 mm in diameter). Genetic parameters for apple (Malus x domestica) fruit external traits (fruit size, ground colour, proportion of over colour and attractiveness) and sensory traits (firmness, crispness, texture, juiciness, flavour, sugar, acidity and global taste) were estimated using 2,207 pedigreed genotypes from breeding programmes in six European countries. Data were scored for 3 years and four periods during storage. Analyses were performed with a restricted maximum likelihood method using VCE 5.1.2 software. Heritability estimates ranged from medium to high for instrumental traits. Genetic correlations between firmness and sugar were medium and low between firmness and acidity. Sensory traits showed low to high heritability, acidity and flavour being, respectively, the most and the less heritable. Global taste was strongly correlated with texture, juiciness, and flavour and relatively less correlated with crispness and acidity. Sensory sugar and acidity showed highly negative correlations whereas their instrumental measurements showed low and increasing positive correlations from harvest to 4 months post-harvest. Sugar exhibited a higher sensory/instrumental divergence. Conversely, instrumental and sensory firmness were highly correlated. Fruit external characteristics had medium heritability. Fruit attractiveness had highest and lowest correlations with fruit size and ground colour, respectively. Best linear unbiased predictors of breeding values were computed for all genotypes with the software PEST. The results were analysed with regard to the dynamic and the reliability of genetic parameters according to the scoring dates. Original issues of the study and the importance of the obtained results for efficient designs of further apple fruit quality breeding programmes were discussed.  相似文献

Summary Sterile AC hybrids between cultivated Oryza sativa (AA) and a distant wild species, O. officinalis (CC), were backcross to O. sativa. Most of the BC1 progenies were allotriploid (AAC), a few were hypotriploid. AAC progenies were again backcrossed to O. sativa. BC2 progenies consisting of disomic or aneuploid individuals were examined for the presence of O. officinalis traits. Eleven different traits from O. officinalis were identified in these progenies. Segregation data in the subsequent generations suggest that these traits are monogenic in nature. Two of these genes — for resistance to BPH and WBPH — are of value in rice improvement. The extremely low recovery of recombinant progenies is in agreement with the very low amount of pairing between A and C genomes. Because of this restricted recombination, the genotype of the recurrent parent was reconstituted after two backcrosses only. Thus, the BC2 progenies look remarkably similar to O. sativa. Most of them are stable and fertile and also interfertile with other O. sativa breeding lines. Some of the BPH-and WBPH-resistant progenies are comparable in yield to the best O. sativa parents and are being evaluated as varietal possibilities.  相似文献

The intertribal sexual hybrids between three Brassica napus (2n = 38) cultivars and Lesquerella fendleri (2n = 12) with the latter as pollen parent were obtained and characterized for their phenotypes and chromosomal and genomic constitutions. F1 plants and their progenies mainly resembled female B. napus parents, while certain characters of L. fendleri were expressed in some plants, such as longer flowering period, basal clustering stems and particularly the glutinous layer on seed coats related to drought tolerance. Twenty-seven F1 plants were cytologically classified into five types: type I (16 plants) had 2n = 38, type II (2) had 2n = 38–42, type III (3) had 2n = 31–38, type IV (5) had 2n = 25–31, and type V (1) had 2n = 19–22. Some hybrids and their progenies were mixoploids in nature with only 1–2 chromosomes or some chromosomal fragments of L. fendleri included in their cells. AFLP (Amplified fragments length polymorphism) analysis revealed that bands absent in B. napus, novel for two parents and specific for L. fendleri appeared in all F1 plants and their progenies. Some progenies had the modified fatty acid profiles with higher levels of linoleic, linolenic, eicosanoic and erucic acids than those of B. napus parents. The occurrence of these partial hybrids with phenotypes, genomic and fatty acid alterations resulted possibly from the chromosome elimination and doubling accompanied by the introgression of alien DNA segments and genomic reorganization. The progenies with some useful traits from L. fendleri should be new and valuable resource for rapeseed breeding.  相似文献

Summary Combining ability studies for grain yield and its primary component traits in diallel crosses involving seven diverse wheat cultivars of bread wheat (Triticum aestivum L.) over generations F1-F5 are reported. The general and specific combining ability variances were significant in all generations for all the traits except specific combining ability variance for number of spikes per plant in the F5. The ratio of general to specific combining ability variances was significant for all the traits except grain yield in all the generations. This indicated an equal role of additive and non-additive gene effects in the inheritance of grain yield, and the predominance of the former for its component traits. The presence of significant specific combining ability variances in even the advanced generations may be the result of an additive x additive type of epistasis or evolutionary divergence among progenies in the same parental array. The relative breeding values of the parental varieties, as indicated by their general combining ability effects, did not vary much over the generations. The cheap and reliable procedure observed for making the choice of parents, selecting hybrids and predicting advanced generation (F5) bulk hybrid performance was the determination of breeding values of the parents on the relative performance of their F2 progeny bulks.  相似文献

 Two sets of plants (Lb and Lc), regenerated from different single-genotype-derived embryogenic suspension cultures of Lolium perenne cv Citadel, were evaluated for agronomic traits in a modified polycross design in the field. Seed from the primary regenerated plants was harvested to evaluate morphological and phenological traits of corresponding progenies in a replicated field experiment. When compared to seed-grown plants of the same cultivar, primary regenerants of the Lb set showed a significant delay in ear emergence and a more-erect growth habit, while primary regenerants from the Lc set showed a significantly higher seed yield. However, progenies of regenerated plants did not differ from those of seed-grown plants. Embryogenic suspension cells of L. perenne have the potential for producing fertile, well-performing, material which can be integrated into breeding programs. Summary Genetic variation in disease resistance to vascular-streak dieback (Vsd) was studied in 54 cocoa hybrid progenies from Trinitario x Amazonian crosses. The disease was assessed by the number of Vsd infected branches per tree, total length of infected branches, average depth of disease penetration within branches and percentages of infected and dead plants. Highly significant differences due to general combining ability for both Trinitario female parents and Amazonian male parents were obtained for all characters. Percent of dead plants was the only character where specific combining ability was also important. Gene effects were predominantly additive for most characters, indicating that selection for progenies resistant to Vsd is effective in the present breeding population. There is strong evidence that resistance to Vsd is in the form of horizontal resistance. It is polygenic and largely inherited as additive genes. Therefore, breeding for resistance is an effective means of controlling Vsd in Papua New Guinea.  相似文献

Summary The applicability and reliability of RAPD markers were evaluated for an examination of the possible use of RAPD markers to confirm hybridity of somatic hybrids between dihaploids of potato (Solanum tuberosum L.). Most of the primers examined detected polymorphism among either tetraploids or dihaploids, and polymorphism was easily detected even among closely related clones. Most of the examples of polymorphism were confirmed as being the result of amplification from the nuclear genome by a comparison of patterns generated by PCR of clones that carried the same cytoplasm. All the bands of dihaploids were transmitted stably to the respective hybrids. In the absence of primers that generated complementary polymorphic bands for both parents, a mixture of two appropriate primers, each of which generated a band specific to one parent, permitted the simple confirmation of hybridity. Hybridity of all the fusion-derived regenerants of 32 fusion combinations was unequivocally confirmed, a result that suggests that RAPD analysis could be universally applicable to the confirmation of hybridity in the dihaploid breeding of potato.  相似文献

Twelve 4x families (obtained from a sub‐set of crosses between seven 4x‐potato cultivars and three 2x haploid Tuberosum‐Solanum chacoense hybrids) were evaluated at Hancock, Wisconsin (USA). The 4x‐parents were elite cultivars selected for adaptation in three continents (Europe, South America, and North America). The 2x male clones were able to produce 2n‐pollen grains by a mechanism akin to first‐division restitution with crossover (FDR‐CO). The estimation of the degree of heterosis for total tuber yield (TTY) was obtained by comparing the field performance of the progenies with their respective 2x and 4x parents. Haulm maturity (HM) and general tuber appearance (GTA) were also evaluated. For TTY, the 4x‐2x families (as a group) outyielded both the 4x and 2x parental groups by 10.6% and 42.5%, respectively. In addition, 5 out of 12 families outyielded their corresponding 4x‐parents. These best five families outyielded the group of 4x‐parents by 40.6%. A considerable variability was observed for HM but, in general, the families were later maturing than the 4x cultivars. The identification of 4x‐2x families with GTA within the range of the 4x commercial cultivars was another important observation. An overall lack of parent‐offspring correlation was detected indicating that performance of the parents per se cannot provide a reliable prediction about the performance of the families. Therefore, progeny testing would be an imperative step for selection of parental clones at both ploidy levels. Our study indicated that haploid Tuberosum‐S. chacoense hybrids are able to generate heterotic 4x‐2x families for TTY in combination with good GTA. These results reinforce the view that selection of superior clones for the Northern Hemisphere can be feasible using germplasm with ~25% genomic contribution of this wild South American species.  相似文献
