In this study, we evaluated the efficiency of different adsorbents for the removal of plant-derived impurities during the pre-purification of paclitaxel from plant cell cultures. Using the synthetic adsorbents sylopute and active clay and their major components SiO2 and MgO, we performed adsorbent treatment and analyzed the paclitaxel precipitates recovered from hexane precipitation. When SiO2 was used, the highest purity (~58.1%) and yield (~91.5%) of paclitaxel were obtained. We also determined differences in the effectiveness of the adsorbent treatment according to changes in the surface area, pore volume and pore diameter of SiO2. Adsorbent treatment was more effective when pore diameter was larger (silica I [2.19 nm] < silica II [4.92 nm] < silica III [9.07 nm]). The highest purity (~74.3%) and yield (~92.9%) of paclitaxel were obtained when silica III was used in the adsorbent treatment. Pore diameter had a greater effect on the removal of plant-derived impurities during the pre-purification of paclitaxel compared with surface area and pore volume. This result could be confirmed by HPLC analysis of the absorbent after treatment and TGA of the organic substances that were bonded to the adsorbent.  相似文献

Dopamine D1-like receptors (D1R and D5R) stimulate adenylyl cyclase (AC) activity, whereas the D2-like receptors (D2, D3 and D4) inhibit AC activity. D1R, but not the D5R, has been reported to regulate AC activity in lipid rafts (LRs). We tested the hypothesis that D1R and D5R differentially regulate AC activity in LRs using human embryonic kidney (HEK) 293 cells heterologously expressing human D1 or D5 receptor (HEK-hD1R or HEK-hD5R) and human renal proximal tubule (hRPT) cells that endogenously express D1R and D5R. Of the AC isoforms expressed in HEK and hRPT cells (AC3, AC5, AC6, AC7, and AC9), AC5/6 was distributed to a greater extent in LRs than non-LRs in HEK-hD1R (84.5 ± 2.3% of total), HEK-hD5R (68.9 ± 3.1% of total), and hRPT cells (66.6 ± 2.2% of total) (P < 0.05, n = 4/group). In HEK-hD1R cells, the D1-like receptor agonist fenoldopam (1μM/15 min) increased AC5/6 protein (+ 17.2 ± 3.9% of control) in LRs but decreased it in non-LRs (− 47.3 ± 5.3% of control) (P < 0.05, vs. control, n = 4/group). By contrast, in HEK-hD5R cells, fenoldopam increased AC5/6 protein in non-LRs (+ 67.1±5.3% of control, P < 0.006, vs. control, n = 4) but had no effect in LRs. In hRPT cells, fenoldopam increased AC5/6 in LRs but had little effect in non-LRs. Disruption of LRs with methyl-β-cyclodextrin decreased basal AC activity in HEK-D1R (− 94.5 ± 2.0% of control) and HEK-D5R cells (− 87.1 ± 4.6% of control) but increased it in hRPT cells (6.8 ± 0.5-fold). AC6 activity was stimulated to a greater extent by D1R than D5R, in agreement with the greater colocalization of AC5/6 with D1R than D5R in LRs. We conclude that LRs are essential not only for the proper membrane distribution and maintenance of AC5/6 activity but also for the regulation of D1R- and D5R-mediated AC signaling.  相似文献

Continuous removal of anthraquinones (AQ) by Amberlite polymeric adsorbents (XAD-4, XAD-7 and XAD-16) through in situ adsorption in Morinda elliptica cell suspension cultures is studied for product recovery and improvement of the overall titre. Ethanol was the best eluting solvent for effective recovery of AQ from all adsorbents. Pre-treatment of XAD-4 with sodium acetate not only enhanced intracellular AQ, but also AQ release and subsequent recovery from the adsorbent. The addition of sodium acetate pretreated XAD-4 on day 18 for 6-day contact period, achieved comparable cell growth to control (41 g/L), but with 1.3-fold higher intracellular AQ (124 mg/g DW) and two-fold increase in extracellular AQ (14.3 mg/L). High amount of adsorbent and longer contact period for the cultures entering stationary growth phase, stimulated AQ release and recovery but at the expense of cell growth. With 5–8.3 g XAD-4 adsorbent per litre M. elliptica culture in production (P) medium, between 60 and 90% AQ was recovered from extracellular AQ after 24–26 days of culture period.  相似文献

Extracorporeal immunoadsorption (ECI) therapy using Staphylococcal Protein A columns has proven effective for removing autoantibodies and circulating immune complexes from patients selectively, providing a promising treatment for autoimmune diseases. However, due to the drawbacks of Protein A in terms of cost and stability, the widespread use of Protein A based ECI is limited. In this study, we investigated the feasibility of 4-mercaptoethylpyridine (MEP, MW 139 Da), a simple and inexpensive synthetic compound, as an alternative to Protein A for autoantibody removal therapy. MEP-based adsorbents were prepared by coupling MEP to Sepharose CL-6B. We found that ligand density was an adjustable parameter for the synthesis of adsorbents aiming at different pathogenic factors, depending on the class of antibody. MEP-Sepharose with a ligand density of 98.8 μmol/ml could remove 80% of the anti-double-stranded DNA antibodies from human serum, whereas a ligand density of 64.5 μmol/ml was enough to remove 96% of the rheumatoid factor (RF) in the serum. Moreover, MEP-based adsorbents showed a lower degree of individual differences compared to Protein A-Sepharose. RF removal of 90% was achieved for all 12 serum samples from different individuals. Among the 14 serum samples derived from systemic lupus erythematosus patients, 11 samples had markedly reduced antinuclear antibody titers. In addition, non-specific adsorption of plasma components to MEP-Sepharose was limited, and the binding capacity of the absorbent for IgG was still about 20 mg/ml of gel after 10 cycles. The results indicated that MEP-based adsorbent could offer a new type of adsorber for the treatment of autoimmune diseases.  相似文献

In this study, crab shells were recycled as an adsorbent for the removal of phosphate. The effects of shell particle size, temperature, pH and phosphate concentration on phosphate removal were investigated. Shell particles less than 1000 μm in diameter removed more than 85% of 500 mg/L phosphate in 24 h while particles 3350 μm in diameter exhibited only 50% removal efficiency. Temperature showed negligible effect on phosphate removal in the range of 15–45 °C. Although removal efficiency was highest at pH 2.0, the efficiency remained 50–60% at pH of 4.0–10.0. The maximum removal capacity was calculated as 108.9 mg/g through Langmuir isotherm plotting, which was 17.0 and 4.7 times higher than those of coal fly ash and scallop shells, respectively. Although calcium carbonate played an active role in the removal of phosphate, both proteins composing 12.5% of crab shells and cellulose-like backbone of the crab shells also played an important role in phosphate removal.  相似文献

The removal of Remazol Blue and Reactive Black B by the immobilized thermophilic cyanobacterial strain Phormidium sp. was investigated under thermophilic conditions in a batch system, in order to determine the optimal conditions required for the highest dye removal. In the experiments, performed at pH 8.5, with different initial dye concentrations between 9.1 mg l−1 and 82.1 mg l−1 and at 45 °C, calcium alginate immobilized Phormidium sp. showed high dye decolorization, with maximum uptake yields ranging from 50% to 88% at all dye concentrations tested. When the effects of high dye concentrations on dye removal were investigated, the highest uptake yield in the beads was 50.3% for 82.1 mg l−1 Remazol Blue and 60.0% for 79.5 mg l−1 Reactive Black B. The highest color removal was detected at 45 °C and 50 °C incubation temperatures for all dye concentrations. As the temperature decreased, the removal yield of immobilized Phormidium sp. also decreased. At about 75 mg l−1 initial dye concentrations, the highest specific dye uptake measured was 41.29–41.17 mg g−1 for Remazol Blue and 47.69–43.82 mg g−1 for Reactive Black B at 45 °C and 50 °C incubation temperatures, respectively, after 8 days incubation.  相似文献

The granulation process using synthetic wastewater containing pentachlorophenol (PCP) in four 1.1 l laboratory scale upflow anaerobic sludge blanket (UASB) reactors was studied, and the anaerobic biotransformation of PCP during the granulation process investigated. After 110 days granular sludge was developed and up to 160 and 180 mg/l of PCP was added into the reactors R1 and R2, respectively, when they were inoculated with acclimated anaerobic sludge from an anaerobic digester of a citric acid plant. The inoculum was predominately composed of bacilli and filamentous bacteria. Granulation did not occur in reactors R3 and R4 which were inoculated with acclimated anaerobic sludge from aerobic sludge of the municipal sewage treatment plant which consisted mainly of cocci. Despite similar bacilli in the granule, the filamentous bacteria from reactor R1 were thicker than those of reactor R2. The granular sludge had a maximum diameter of 2.5 and 2.2 mm, and SMA of 1.44 and 1.32 gCOD/gTVS per day for reactors R1 and R2, respectively. Over 98% chemical oxygen demand (COD) removal rate and 99% of PCP removal rate were achieved when reactors R1 and R2 were operated at PCP and COD loading rates of 150 and 7.5 g/l per day, respectively. H2-producing acetogens were the dominant anaerobes in the granular sludge.  相似文献

A comparative study of the performance of two types of adsorbent (Streamline Quartz Base and Upfront Matrices), derivatized with the same affinity ligand (RPAP) to recover C595 diabody fragment (dbFv) from Escherichia coli lysates has been undertaken. Both streamline and Upfront Matrices are characterized by a particle size range of 100–300 μm. Streamline has a density of 1.20 g cm−3 and ligand concentration of 0.85 μmol ml−1. Upfront has a density of 1.35 g cm−3 and ligand concentration of 0.83 μmol ml−1. The release of C595 dbFv from E. coli cells was achieved by a chemical lysis method. The recovery performance of both adsorbents was evaluated in terms of operational productivity and elution yield of C595 dbFv in packed bed (clarified feedstock) and expanded bed (unclarified and clarified feedstock) chromatography systems. Streamline and Upfront adsorbents exhibited diabody operational productivities of 131 and 202 mg l−1, respectively, with an elution yield of 92 and 94%, respectively, in packed bed operation. Streamline and Upfront adsorbents exhibited diabody operational productivities of 54.5 and 123.7 mg l−1, respectively, with an elution yield of 89 and 92%, respectively, in expanded bed operation.  相似文献

The bioaccumulation of chromium(VI), nickel(II), copper(II), and reactive dye by the yeast Rhodotorula mucilaginosa has been investigated in media containing molasses as a carbon and energy source. Optimal pH values for the yeast cells to remove the pollutants were pH 4 for copper(II) and dye, pH 6 for chromium(VI) and dye, and pH 5 for nickel(II) and dye in media containing 50 mg l?1 heavy metal and 50 mg l?1 Remazol Blue. The maximum dye bioaccumulation was observed within 4–6 days and uptake yields varied from 93% to 97%. The highest copper(II) removal yields measured were 30.6% for 45.4 mg l?1 and 32.4% for 95.9 mg l?1 initial copper(II) concentrations. The nickel(II) removal yield was 45.5% for 22.3 mg l?1, 38.0% for 34.7 mg l?1, and 30.3% for 62.2 mg l?1. Higher chromium(VI) removal yields were obtained, such as 94.5% for 49.2 mg l?1 and 87.7% for 129.2 mg l?1 initial chromium(VI) concentration. The maximum dye and heavy metal bioaccumulation yield was investigated in media with a constant dye (approximately 50 mg l?1) and increasing heavy metal concentration. In the medium with 48.9–98.8 mg l?1 copper(II) and constant dye concentration, the maximum copper(II) bioaccumulation was 27.7% and 27.9% whereas the maximum dye bioaccumulation was 96.1% and 95.3%. The maximum chromium(VI) bioaccumulation in the medium with dye was 95.2% and 80.3% at 48.2 and 102.2 mg l?1 chromium(VI) concentrations. In these media dye bioaccumulation was 76.1% and 35.1%, respectively. The highest nickel(II) removal was 6.1%, 20.3% and 16.0% in the medium with 23.8 mg l?1 nickel(II) + 37.8 mg l?1 dye, 38.1 mg l?1 nickel(II) + 33.4 mg l?1 dye and 59.0 mg l?1 nickel(II) + 39.2 mg l?1 dye, respectively. The maximum dye bioaccumulation yield in the media with nickel(II) was 94.1%, 78.0% and 58.7%, respectively.  相似文献

The performance of native and heat-treated anaerobic granular sludge in removing of malachite green (MG) from aqueous solution was investigated with different conditions, such as pH, ionic strength, initial concentration and temperature. The maximum biosorption was both observed at pH 5.0 on the native and heat-treated anaerobic granular sludge. The ionic strength had negative effect on MG removal. Kinetic studies showed that the biosorption process followed pseudo-second-order and qe for native and heat-treated anaerobic granular sludge is 61.73 and 59.17 mg/g at initial concentration 150 mg/L, respectively. Intraparticle diffusion model could well illuminate adsorption process and faster adsorption rate of native anaerobic granular sludge than heat-treated anaerobic granular sludge. The equilibrium data were analyzed using Langmuir and Freundlich model, and well fitted Langmuir model. The negative values of ΔG° and ΔH° suggested that the interaction of MG adsorbed by native and heat-treated anaerobic granular sludge was spontaneous and exothermic. Desorption studies revealed that MG could be well removed from anaerobic granular sludge by 1% (v/v) of HCl–alcohol solution.  相似文献

In the present study, separation of l-lysine from dilute aqueous solution by solid-phase extraction based on molecular imprinting technique using a polar porogen was investigated. l-Lysine imprinted polymer (LLIP) was prepared by free radical solution polymerization of methacrylic acid and ethylene glycol dimethacrylate as functional and cross-linking monomers, in the presence of l-lysine as an imprint molecule, mixture of water and methanol as solvent and AIBN as an initiator. Non-imprinted polymer (NIP) as control was also prepared by the same procedure in the absence of template molecules. LLIP particles were applied to determine the optimum operational condition for l-lysine separation from dilute aqueous solution. In adsorption step, optimum pH and retention time were 7.8 and 90 min, while corresponding values in extraction step were 12 and 50 min, respectively. l and d-Lysine recovery by LLIP at optimum condition were found to be 96 and 58% with corresponding distribution coefficients of 8000 and 460, respectively. The retention capacity of LLIP was 27.26 mg l-lys/g of polymer at optimum condition.  相似文献

The combined effects of initial sucrose and initial Remazol Turquoise Blue-G (RTBG) reactive dye concentrations on the specific growth rate and dye bioaccumulation efficiency of Candida utilis was investigated and optimized using response surface methodology (RSM) in this study. A 22 full factorial central composite design was successfully used for experimental design and analyses of the results. Two numerical correlations fitted to a second-order quadratic equation were obtained to estimate the responses of specific growth rate and dye uptake yield. The statistical analysis indicated that both the microbial growth and removal yield of dye enhanced with raising sucrose concentration up to 15 g l?1 and diminished with the increase in initial RTBG dye concentration up to approximately 500 mg l?1 due to inhibition caused by high concentrations of RTBG dye. The optimum combination predicted via RSM confirmed that C. utilis was capable of bioaccumulating RTBG with the maximum uptake yield of 82.0% in 15 g l?1 sucrose and 50 mg l?1 dye containing growth medium.  相似文献

Silica-based immobilized metal affinity chromatography adsorbents with various ligand densities were prepared for the purification and immobilization of poly(His)-tagged d-hydantoinase (DHTase). An adsorbent with a ligand density of 13.0 μmol Cu2+/g gel exhibiting the optimal selectivity and a capacity of 1.4 mg/g gel toward the poly(His)-tagged enzyme was identified. The adsorbent was used for the one-step purification of His-tagged enzymes from crude cell lysate with a purity above 90%. The silica-based affinity adsorbents are particularly well suited for industrial scale operations due to their robustness. A packed-bed bioreactor with the DHTase-loaded adsorbents was used for the continuous conversion of d,l-p-hydroxyphenylhydantoin (d,l-HPH) to N-carbamoyl-d-hydroxyphenylglycine, an intermediate for the production of d-hydroxylphenylglycine. Under optimal conditions, 60 °C and pH 8.0, a conversion of 60% was obtained at a residence time of 30 min. Upon extended operation, the catalytic activity of the biocatalysts declined significantly due to enzyme leakage and enzyme denaturation. The extent of enzyme leakage can be attenuated by crosslinking with glutaraldehyde. In this study, we successfully demonstrate that a packed-bed bioreactor containing silica-based IMAC adsorbents can be used for the direct purification and immobilization of poly(His)-tagged enzymes for biotransformation.  相似文献

This glasshouse study examined the effect of three damage types on plant growth and nutrient allocation of the invasive aquatic plant, alligator weed (Alternanthera philoxeroides). The damage included: repeated leaf removal, a single application of herbicide, and one-time shoot removal. Damage types were meant to simulate the effects of insect herbivory, chemical, and mowing/grazing, respectively. Response variables included plant biomass and both the concentration and abundance of nutrients. Complete shoot removal and herbicide treatments caused an initial decline in growth rate, followed by several weeks of increasing rates and finally a second decline during the fourth week. Plants from control and repeated leaf removal treatments showed a steady increase in growth rate from the treatment application to the final harvest, but control plants were accumulating biomass three times faster than repeated defoliation plants by the fifth week (9.7 and 3.5 g week−1, respectively). Not surprisingly, all treatments led to lower total cumulative biomass 5 weeks after treatment application (mean 30.8 g) when compared with controls (49.0 g). However, despite the repeated leaf removal and complete shoot removal treatments removing similar quantities of biomass (mean 8.0 and 7.5 g respectively), repeated removal of leaves produced less total biomass (26.2 g) and led to less cumulative above ground biomass (20.1 g) than the other treatments (mean total = 33.1 g, mean above ground = 25.7 g). Repeated leaf removal also produced less below ground biomass (6.1 g) than the shoot removal treatment (8.5 g) and had the greatest negative effect on nitrogen and potassium abundance in plant tissues after 5 weeks. In addition, it reduced the amount of phosphorous to a lower level than herbicide treated or control plants. These results indicate that repeated leaf removal was the treatment most effective in reducing biomass and depleting nutrients in A. philoxeroides plants.  相似文献

Integration of partial nitrification (nitritation) and anaerobic ammonium oxidation (anammox) in constructed wetlands creates a sustainable design for nitrogen removal. Three wetland treatment systems were operated with synthetic wastewater (60 mg NH3–N L?1) in a batch mode of fill – 1-week reaction – drain. Each treatment system had a surface flow wetland (unplanted, planted, and planted plus aerated, respectively) with a rooting substrate of sandy loam and limestone pellets, followed by an unplanted subsurface flow wetland. Meanwhile, three surface flow wetlands with a substrate of sandy loam and pavestone were operated in parallel to the former surface flow wetlands. Influent and effluent were monitored weekly for five cycles. Aeration reduced nitrogen removal due to hindered nitrate reduction. Vegetation maintained pH near neutral and moderate dissolved oxygen, significantly improved ammonia removal by anammox, and had higher TN removal due to coexistence of anammox and denitrification in anaerobic biofilm layers. Nitrite production was at a peak at the residence time of 4–5 d. Relative to pavestone, limestone increased the nitrite mass production peak by 97%. The subsurface flow wetlands removed nitrogen via nitritation and anammox, having an anammox activity of up to 2.4 g N m?3 d?1 over a startup operation of two months.  相似文献

In this research, micro and nanoparticles of Spirulina platensis dead biomass were obtained, characterized and employed to removal FD&C red no. 40 and acid blue 9 synthetic dyes from aqueous solutions. The effects of particle size (micro and nano) and biosorbent dosage (from 50 to 750 mg) were studied. Pseudo-first order, pseudo-second order and Elovich models were used to evaluate the biosorption kinetics. The biosorption nature was verified using energy dispersive X-ray spectroscopy (EDS). The best results for both dyes were found using 250 mg of nanoparticles, in these conditions, the biosorption capacities were 295 mg g?1 and 1450 mg g?1, and the percentages of dye removal were 15.0 and 72.5% for the FD&C red no. 40 and acid blue 9, respectively. Pseudo-first order model was the more adequate to represent the biosorption of both dyes onto microparticles, and Elovich model was more appropriate to the biosorption onto nanoparticles. The EDS results suggested that the dyes biosorption onto microparticles occurred mainly by physical interactions, and for the nanoparticles, chemisorption was dominant.  相似文献

The adsorption of papain on Reactive Blue 4 dye–ligand affinity membrane was investigated in a batch system. The combined effects of operating parameters such as initial pH, temperature, and initial papain concentration on the adsorption were analyzed using response surface methodology. The optimum adsorption conditions were determined as initial pH 7.05, temperature 39 °C, and initial papain concentration 11.0 mg/ml. At optimum conditions, the adsorption capacity of dye–ligand affinity membrane for papain was found to be 27.85 mg/g after 120 min adsorption. The papain was purified 34.6-fold in a single step determined by fast protein liquid chromatography. More than 85% of the adsorbed papain was desorbed using 1.0 M NaCl at pH 9.0 as the elution agent. The purification process showed that the dye–ligand immobilized composite membrane gave good separation of papain from aqueous solution.  相似文献

The studies on adsorption of hexavalent chromium were conducted by varying various parameters such as contact time, pH, amount of adsorbent, concentration of adsorbate and temperature. The kinetics of adsorption of Cr(VI) ion followed pseudo second order. Langmuir adsorption isotherm was employed in order to evaluate the optimum adsorption capacity of the adsorbent. The adsorption capacity was found to be pH dependant. Sawdust was found to be very effective and reached equilibrium in 3 h (adsorbate concentration 30 mg l−1). The rate constant has been calculated at 303, 308, 313 and 318 K and the activation energy (Ea) was calculated using the Arrhenius equation. Thermodynamic parameters such as standard Gibbs energy (ΔG°) and heat of adsorption (ΔHr) were calculated. The ΔG° and ΔHr values for Cr(VI) adsorption on the sawdust showed the process to be exothermic in nature. The percentage of adsorption increased with decrease in pH and showed maximum removal of Cr(VI) in the pH range 4.5–6.5 for an initial concentration of 5 mg l−1.  相似文献

The adsorption characteristics of various filter media and treatment efficiency of small pilot-scale constructed wetlands (CWs) were investigated in order to design optimum CWs for treating greenhouse wastewater. Calcite was the best filter medium for the adsorption of ammonium nitrogen and phosphorus under various temperature and pH conditions. However, removal efficiency of calcite for total nitrogen (T-N) removal was low due primarily to high nitrate levels. Thus, several hybrid CWs (containing calcite as filter media) consisting of combinations of vertical flow (VF) and horizontal flow (HF) beds were evaluated for improving efficiency for T-N removal. Both 2- and 3-stage combinations of the VF and HF beds were tested. The optimum hybrid CWs was demonstrated to be a 3-stage combination of horizontal flow (HF)–vertical flow (VF)–horizontal flow (HF), which provided suitable conditions for both nitrification and denitrification, which improved removal of T-N in wastewater containing nitrate. In the HF–VF–HF 3-stage hybrid CWs, the reduction in chemical oxygen demand (COD), T-N, and total phosphorus (T-P) in the effluent were 95.1, 68.4 and 94.3%, respectively. The removal of COD, T-N and T-P in 3-stage HF–VF–HF CWs was rapid in order of VF (second stage)  HF (first stage)  HF (third stage), VF (second stage)  HF (third stage) > HF (first stage) and VF (second stage)  HF (first stage)  HF (third stage), respectively.  相似文献
