The aim of the current study was to investigate the effects of ambient temperature on thyroid activity of the soft-shelled turtle (Lissemys punctata pucntata). Turtles exposed to low ambient temperature (10 degrees C for 15 days) showed a significant decrease in relative thyroid weight, follicular cell size (cell became squamous from cuboidal type) and epithelial height in both the peripheral and central follicles of the gland, with the appearance of homogeneous colloid materials in the follicular lumen. Thyroid peroxidase activity declined significantly. In contrast, high ambient temperatures (32/34 degrees C for 15 days) caused reverse changes to those observed after exposure with low ambient temperature. No significant difference was marked in thyroid activity between 32 and 34 degrees C temperature treatments. The findings provide evidence that low ambient temperature inhibits thyroid activity and high ambient temperature stimulates the gland activity in soft-shelled turtles. Ambient temperature acts presumably via the hypothalamo-hypophysial (TRF-TSH) axis which in turn alters thyroid function in turtles.  相似文献

The effects of some exogenous peripheral hormones (thyroxine, corticosterone, epinephrine, norepinephrine and insulin) on thyroid activity were investigated in juvenile female soft-shelled turtles, Lissemys punctata punctata. Each hormone was injected in three different doses (25 microg, 50 microg or 100 microg each per 100 g body weight, once daily at 9 AM) for 10 consecutive days. Thyroid activity was evaluated by gravimetry, histology (epithelial height) and thyroperoxidase assay. The findings revealed that thyroxine in low dose (25 microg) stimulated thyroid activity by increasing the relative thyroid weight, epithelial height and thyroperoxidase activity, but inhibited gland activity at a high dose (100 microg) by decreasing the values of all these parameters. The medium dose (50 microg) had no significant effect. All other hormones, in all doses, significantly decreased thyroid activity by decreasing the values of all the parameters. Thyroid responses to exogenous hormones are generally dose-dependent in turtles. The mechanisms of actions of the hormones administered are suggested.  相似文献

An exposure to ambient temperature of 25 degrees C had no perceptible effect on interrenal function but further increase of temperature to 35 degrees C caused nuclear hypertrophy with increase of nuclear diameter, RNA concentration, acid phosphatase and alkaline phosphatase activities, accompanied by quantitative depletions of cholesterol (free, esterified and total) and ascorbic acid levels in the interrenal gland of the soft-shelled turtle Lissemys p. punctata. Similar manifestations of stimulation, except in the nucleus, were marked after exposure to 38 degrees C, but the degree of response in respect of esterified and free cholesterol levels was higher at 38 degrees C than at 35 degrees C. Moreover, withdrawal of 38 degrees C temperature and subsequently maintaining at 25 degrees C for 15 days showed reverse manifestations to those of 35 degrees C/38 degrees C, leading to a tendency towards normalcy. It is suggested that high a ambient temperature of 35 degrees C significantly stimulates interrenal function of Lissemys turtles, but further increase of 38 degrees C does not cause further overall stimulation, and withdrawal of higher temperature (38 degrees C) shows a tendency towards normalcy. It is also suggested that (a) high ambient temperature causes thermal stress, (b) it is reversible and (c) it acts on interrenal activity presumably via CRF-ACTH-axis in turtles.  相似文献

Adrenocortical responses to diverse stressful situations (dehydration, formaldehyde treatment and salt loading) were studied in the adult female soft-shelled turtle, Lissenmys p. punctata. Dehydration, formaldehyde treatment (formalin, 1%: 0.1 ml/100 g body weight daily) or salt loading (NaCl, 1%: 0.1 ml/100 g body weight daily) treatments consecutively for 7 days caused hypertrophy of the adrenocortical cells with their nuclear diameter increased, and depletions of adrenal cholesterol and ascorbic acid concentrations followed by decreased acid phosphatase and alkaline phosphatase activities in turtles. Corticosterone levels were elevated in both the adrenal gland and serum of turtles after dehydration and formalin stress, but the hormone level remained unaltered after salt loading in turtles. The results suggest active involvement of adrenal cortex in stress for homeostasis in Lissemys turtles.  相似文献

The current study was undertaken to ascertain the seasonal influence of thyroid activity in female soft-shelled turtles, Lissemys p. punctata. Thyroid gland was studied month-wise throughout the year from relative gland weight, histology, epithelial height, glandular peroxidase activity, and RIA of T3 and T4 levels from blood serum and the thyroid gland. The values of all the parameters, except those of T3 and T4, were higher during March through May, decreased from June through August and began to rise thereafter (September through February). Whereas T3 and T4 levels were highest in May, lower during June to November and began to rise thereafter. The difference in the peaks between T3 and T4 levels and other parameters have been explained. The findings suggest that thyroid activity of Lissemys turtles varies seasonally and that seasonal factors like temperature play an important role in influencing thyroid activity in soft-shelled turtles.  相似文献

Daily afternoon injections of 25 micrograms melatonin for 12 weeks had no effect on testicular weights of male rats kept in long photoperiod (14L:10D); similarly, exposure of rats to short photoperiod (2L:22D) had no effect on gonadal weight. However, rats maintained in a long or short photoperiod and implanted every 2 weeks with a 15 mm Silastic pellet containing testosterone showed a significant reduction in testicular weight; this effect was more pronounced in rats exposed to a short photoperiod. Melatonin injections in testosterone-treated rats in a long photoperiod exacerbated the inhibitory effects of testosterone alone. Subcutaneous 2-weekly implants of a beeswax pellet containing 1 mg melatonin reversed the effects of the melatonin injections on relative testicular weights but not those due to short photoperiod exposure. Testosterone implants significantly reduced pituitary LH values in long and short photoperiod-exposed animals, more particularly in those exposed to short photoperiod. Melatonin injections alone or in combination with melatonin pellets did not further exaggerate the depression in pituitary LH due to testosterone alone in long photoperiod-exposed animals; similarly melatonin pellets did not reverse the depression in pituitary LH observed. No significant differences in plasma prolactin concentrations or in thyroxine concentrations or free thyroxine index were observed after any combination of treatments. We therefore suggest that the effects observed with short photoperiod may be due to melatonin.  相似文献

Testicular size, plasma testosterone levels, copulatory behavior, and daily locomotor activity are reduced in male hamsters after 10 weeks of exposure to short days. The role of testosterone in the short day-induced decline in locomotor activity was investigated, determining whether or not photoperiod could alter the effect of testosterone on activity. Castrated adult hamsters were allowed to acclimate to running wheels (wired to digital counters) and then were kept on either long (L:D 14:10) or short (L:D 6:18) days for 60 days. On Day 60, half of the animals on each light cycle were implanted with 12-mm-long testosterone-filled Silastic capsules; half received empty capsules. Digital counting of wheel-running activity continued for another 140 days. Blood samples taken on Day 200 confirmed L:D 14:10 and L:D 6:18 testosterone-treated hamsters had equivalent plasma testosterone levels. After an initial decline in activity, L:D 14:10 animals exhibited a progressive rise in mean running activity (from ~2000 to ~5000 wheel revolutions per day) through 100 days after the initiation of testosterone treatment. In contrast, activity levels in testosterone-treated L:D 6:18 animals remained uniform (~2000 wheel revolutions per day) during this time, indicating exposure to short days rendered the hamsters less sensitive to the stimulatory effect of testosterone on activity. Of further interest was a marked increase in activity after 160–200 short days in animals treated with either testosterone-filled or empty capsules. It appears the total amount of daily locomotor activity in the hamster is modulated by circulating testosterone levels in a manner which is dependent upon the environmental photoperiod.  相似文献

The objective of this work was to evaluate the role of short photoperiod in timing the onset and duration of reproductive activity in ewes. The perception of photoperiod was disrupted by pinealectomy following transfer from long (17L:7D) to short (8.5L:15.5D) photoperiod and the subsequent reproductive response was monitored. Ovariectomized ewes given Silastic implants containing estradiol-17 beta were exposed to long days until Day 0 (May 24) and then were allocated to the following groups (n = 5-6/group): Group 1) short-day control--moved to short days; Groups 2 to 5) pinealectomy after 0, 30, 60, or 90 short days, respectively; Group 6) long-day hold--kept on long days; Group 7) long days after 60 short days--moved to short days on Day 0 and returned to long days on Day 60. Six ewes kept outdoors served as additional controls. Reproductive neuroendocrine activity was assessed from plasma LH concentrations, high values being indicative of the breeding season and low values indicative of anestrus. Time of reproductive neuroendocrine activity onset (LH rise) did not differ among animals in the 7 groups kept indoors, but was advanced (p less than 0.05) relative to that of ewes outdoors. Adult rams were exposed to photoperiod treatments over 2 years to study the influence of light regimes on pituitary-testicular activity and semen quality. Initially, all rams (12 per group) were exposed to 3 months of long days (16L:8D). Group 1 was then exposed to a regime of continuous short days (8L:16D) and Groups 2, 3, and 4 were exposed to 4 months of short days alternated with 1, 2, or 4 months, respectively, of long days. Every 2 weeks, serum hormone levels and scrotal circumference were determined and semen quality was evaluated. Regular cycles in pituitary and testicular activities corresponding to the period of the lighting regime resulted in Groups 2, 3, and 4, but not in Group 1. In general, the change from long days to short days induced increases in follicle-stimulating hormone (FSH), luteinizing hormone (LH), and testosterone levels, scrotal size and sperm numbers and a decrease in prolactin. The reverse occurred after subsequent exposure to long days. After 4 months of long days, testicular regression was complete, but when long-day exposure was reduced, less regression occurred. With continuous exposure to short days, FSH and testosterone remained above basal levels, prolactin levels were depressed, scrotal size remained near the maximum, and elevated numbers of motile sperm were sustained.  相似文献

Chronic exposure of female Syrian hamsters (Mesocricetus auratus) for 9 weeks to a short photoperiod (10L:14D) depressed the pituitary-thyroid axis as indicated by a drop in circulating titers of thyroid stimulating hormone (TSH), thyroxine (T4), triiodothyronine (T3) and the free thyroxine index (FT4I) compared to animals maintained under long photoperiodic conditions (14L:10D). 2. Short day treatment also reduced plasma cholesterol levels. 3. The effects of long 'summer' (22L:2D) and short 'winter' (2L:22D) photoperiod on post-weaning growth and food consumption of the collared lemming were examined. Growth was described by Gompertz equations. After 10 weeks, lemmings maintained under short photoperiods developed white winter pelage and heavy bifid claws; body mass and estimated asymptotic mass were significantly greater than for lemmings reared under long photoperiod. Sexual dimorphism in body size was observed within treatments, males growing larger than females. There were no significant differences in the overall growth rate constant k as a result of either sex or photoperiod treatment. However, instantaneous growth rates (dW/dt) were significantly higher in 'winter' lemmings. Mass at weaning was a significant determinant of adult mass at 90 days.
Cumulative food consumption at 90 days was not significantly different between photoperiod groups, even though 'winter' lemmings gained more than double the mass of their 'summer' counterparts over the post-weaning period. Increased gut length observed for winter morphs may act as the physiological mechanism promoting greater gross energy efficiency.
The present study suggests that the overwintering strategy of the collared lemming with respect to body mass changes and energy requirements differs considerably from that of other microtines.  相似文献

We investigated the effects of photoperiod on testicular activity in wild rabbits (Oryctolagus cuniculus) captured on Zembra Island (North Tunisia) and maintained in experimental photoperiodic conditions. Sexually inactive animals were subjected to alternate 3-mo periods of short days (8L:16D) and long days (16L:8D) for 1 yr. Testicular activity increased significantly and then decreased to levels equivalent to or lower than those measured during sexual quiescence after 1 mo of 8L:16D or 16L:8D, respectively. Eight groups of sexually active animals were also exposed to 8L:16D for 60 days. The light phase was divided into two photofractions (7.5 and 0.5 h). The short photofraction interrupted the dark phase 9.5-18.5 h after the beginning of the main photofraction. Testicular activity was inhibited if the short photofraction interrupted the dark phase 12.5 h or more after the beginning of the main photofraction. These results clearly confirm that photoperiod affects reproduction in this species: Short days stimulate reproduction, whereas long days inhibit it. The asymmetric pattern of skeleton photoperiods used demonstrated the existence of a circadian rhythm for photogonadosensitivity, with the photosensitive phase beginning 12.5 h after dawn. In this species, photoperiod length controls both the beginning and the end of the reproductive period. These results differ from those obtained with continental populations of wild rabbits, in which reproduction is inhibited by short day length. This difference may reflect genetic drift linked to the geographic isolation of this population, which is known to have been present on this small island for more than 2000 yr.  相似文献

Seasonal cycles in the size of the testes, blood plasma concentration of testosterone, FSH and prolactin, intensity of the sexual skin flush, timing of rutting behaviour and moulting of the body coat were recorded in Soay rams after s.c. implantation of melatonin contained in a Silastic envelope which increased the circulating blood levels of melatonin to 200-600 pg/ml for many months. Two groups of 8 adult rams were held under alternating periods of short days (8L:16D) and long days (16L:8D) to drive the seasonal cycles and the treatments with melatonin were initiated during the long or short days, and one group of 8 ram lambs was kept out of doors and given implants during the long days of summer (4 melatonin-implanted and 4 control (empty implants) rams per group). The treatments demonstrated that melatonin implants during exposure to long days resulted in a rapid 'switch on' of reproductive redevelopment similar to that produced by exposure to short days melatonin implants prevented the rams from showing the normal responses to changes in the prevailing photoperiod rendering them nonphotoperiodic; and long-term cyclic changes in testicular activity, prolactin secretion and other characteristics occurred in the melatonin-implanted rams; the pattern was similar to that previously observed in rams exposed to prolonged periods of short days. The overall results are consistent with the view that melatonin is the physiological hormone that relays the effects of changing photoperiod on reproduction and other seasonal features, and that continuous exogenous melatonin from an implant interferes with the normal 'signal' and produces an over-riding short-day response.  相似文献

Minces of the testes of bank voles, born and reared in a long (18L:6D) photoperiod until weaning (18-22 days of age) and subjected thereafter to a short (6L:18D, Group S) or a long (18L:6D, Group L) photoperiod for 6-9 weeks, were incubated with [4-14C]17 alpha-hydroxyprogesterone in the presence of cofactors (NADP/NADPH, 1.3 mmol/1) for 1 h at 37 degrees C. The radioactive metabolites were characterized and identified by thin-layer chromatography with derivative formation and chromatography to constant specific activity and isotope ratio. In Group L virtually all of the substrate was utilized and it was readily converted to androgens (48% of the radioactivity recovered) such as androstenedione and testosterone. The only pregnane metabolite identified was 17 alpha-hydroxy,20 alpha-dihydroxyprogesterone (43.3%). In Group S there was a decreased production of 17 alpha-hydroxy,20 alpha-dihydroprogesterone and androgens (25.4% and 10.4% respectively) and a substantial portion of the substrate was not metabolized (38.8%). The main androgen metabolites identified, androst-4-ene-3 beta,17 beta-diol and 5 alpha-androstane-3,17-dione are hormonally quite inert steroids. No androstenedione or testosterone was found. The results indicate that exposure to short photoperiod induces a decrease in the testicular C17-C20 lyase and 20 alpha-hydroxysteroid dehydrogenase.  相似文献

The purpose of this study was to evaluate whether the insertion of a continuous-release melatonin implant into ewes provides a short-day photoperiodic signal or acts as a functional pinealectomy (provides no specific photoperiodic signal but renders ewes incapable of responding to changes in photoperiod). Ewes primed with 60 long days (18L:6D) during the spring were moved to intermediate day length (13L:11D) for 66 days and then given one of five treatments: 1) short-day control, second drop in photoperiod to 8L:16D; 2) intermediate-photoperiod control, kept on 13L:11D; 3) pinealectomy and kept on 13L:11D; 4) melatonin implant and kept on 13L:11D; 5) melatonin implant and moved to 8L:16D. Mean number of estrous cycles per group and total duration of reproductive activity were determined. Ewes in all groups began to exhibit estrous cycles after the initial reduction in photoperiod. The number of estrous cycles and duration of reproductive activity differed among groups. The number of estrous cycles and duration of reproductive activity was extended in ewes receiving the second drop in photoperiod compared to that of the intermediate-photoperiod controls. Pinealectomized ewes had a number of estrous cycles and duration of reproductive activity similar to those of ewes maintained on the intermediate photoperiod. Melatonin implants increased the number of estrous cycles and prolonged reproductive activity in ewes maintained on the intermediate photoperiod; melatonin implants did not prevent the extension of reproductive activity in ewes receiving the second photoperiodic drop to the short daylength.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

In Exp. 1, the time course for the photoperiodic response in juvenile male musk shrews was examined by exposing animals to short (10L:14D) or long (14L:10D or 18L:6D) daylengths for 10, 20, 40 or 56 days. When compared with the response of animals maintained in long days, those exposed to short days showed an inhibition of reproductive maturation by 40 days of treatment. In Exp. 2, the combined effects of photoperiod and social cues were investigated in juvenile males that were either housed in short days (with or without a female) or in long days (with or without a female) for 40 days. The short photoperiod was generally inhibitory to sexual maturation, while the presence of an adult female was generally stimulatory, i.e. animals living with a female in long days had the greatest level of sexual maturity, while those living alone in short days had the lowest level of maturity. Animals that received opposing signals, i.e. short days in the presence of a female, had an intermediate response which was equivalent to the response seen in males living alone in long days. These results suggest that the presence of a female can partly reverse inhibitory effects of short days. In Exp. 3, the effects of photoperiod and social cues on the reproductive physiology of the adult male (same design as in Exp. 2) were investigated. The oviduct of the Indian fresh water soft-shelled turtle Lissemys punctata punctata was examined throughout the year under light and scanning electron microscopes to determine the location, histomorphological characteristics, and function of sperm storage structure, as well as their changes at different phases of the seasonal reproductive cycle. Sperm storage structures in the form of tubules were observed in the wall of isthmus throughout the year. These tubules developed either by folding or fusion of the oviductal mucosal folds and were lined by both ciliated and nonciliated epithelial cells. The height and secretory activities of the epithelia were markedly high during the breeding phase (August to September) but low in the nonbreeding phase (October to June). A few short tubules lined by cuboidal epithelium appear in the wall of infundibulum only during the breeding phase. Following mating (May), inseminated sperm were stored within the tubules of isthmus up to the pre-ovulatory stage (August). Thereafter, sperm associated with PAS-positive materials secreted from the epithelium (referred to as a carrier matrix) moved forward to the infundibulum and were stored within the storage tubules of the infundibulum for a short time. Subsequently, sperm evacuated the storage tubules and entered the oviductal lumen to fertilize the subsequently ovulated eggs during or prior to ovulation. The isthmus-tubules become shorter and narrower in the regressive phase (October to November) and remained so until the early preparatory phase (April). Sperm release might have been stimulated by estrogen secreted from the ovarian follicles of pre-ovulatory turtles. Stored sperm not utilized for fertilization remained viable not less than six months in the present turtle species.  相似文献

Siberian hamsters are photoperiodic rodents that typically exhibit several physiological changes when exposed to a short-day photoperiod. However, development of the winter phenotype in short days is largely conditional on prior photoperiod history: Hamsters that have been reared in an exceptionally long day length (18 L) do not usually exhibit the winter phenotype after transfer to short days, whereas animals reared under "moderately" long days (16 L) are more variable in responsiveness to subsequent short-day exposure, with 20% to 30% generally failing to exhibit winter-type responses. Hamsters reared exclusively in an "intermediate" day length (14 L) are almost uniformly responsive to short photoperiod. In the present study, the authors examine the influence of photoperiod history on short-day responsiveness in a breeding line of hamsters that has been subjected to artificial selection for resistance to the effects of short days. The results demonstrate that photoperiod history is an important determinant of short-day responsiveness in both random-bred (UNS) hamsters and animals artificially selected and bred for nonresponsiveness to short photoperiod (PNR). The PNR hamsters have a reduced requirement for long-day exposure to evoke a state of unresponsiveness to short days. The results are discussed in relation to possible significance for the origin of population and species differences in photoperiod responsiveness.  相似文献

The presence of a reproductive diapause in the life cycle of the neotropical cassidine beetle, Chelymorpha alternans , was investigated by exposing groups of beetles to conditions differing in photoperiod and humidity. Diapause was characterized by the absence of egg-laying in females up to 70 days after emergence and was induced in response to a short photoperiod of 12:12 h L: D but averted under a longer photoperiod of 13:11 h L:D. High (>90% RH) and low (55–75% RH) humidity conditions did not influence diapause induction. Males appeared to court and attempt to mate with females under all conditions between 16 and 30 days after emergence, but declined after this time in short photoperiods and it is not known if matings in these groups were successful. Adults induced to diapause by exposure to a 12:12 h L:D photoperiod and subsequently exposed to a 13:11 h L: D photoperiod 70 or 72 days after emergence did not show a rapid response by commencing egg-laying. However, all diapausing groups in long and short photoperiod and high and low humidity, in both experiments performed, had commenced egg-laying after 128 days, suggesting an endogenous rhythm for diapause termination. Long photoperiod and high humidity combined may hasten diapause termination since egg-laying began after only 95 days in this group in experiment two. This strategy of induction and termination is discussed with reference to the seasonality of the natural environment of C. alternans in Panama.  相似文献

Castrated hamsters which were transferred from long (14L:10D) to short (9L:15D) days and received testosterone-filled capsules for 1 week after transfer failed to show a significant suppression in the plasma levels of FSH and LH after capsule removal. In contrast, gonadotrophin concentrations were suppressed in hamsters in which the long-day castration response had been blocked with exogenous testosterone. After castration on long days and exposure to 10 weeks of short days pituitary gland weight and gonadotrophin content, as well as plasma FSH titres, were higher in control animals than in those that had received testosterone implants for 7 weeks of short days. The results suggest that failure of castrated hamsters to respond to the suppressive effects of short days reflects castration-induced changes in hypothalamo-pituitary physiology rather than a neuroendocrine mechanism by which photoperiod modulates gonadotrophin secretion.  相似文献
