A comparison of histochemical detection of GM1 ganglioside in cryostat sections using cholera toxin B-subunit after fixation with 4% formaldehyde and dry acetone gave tissue-dependent results. In the liver no pre-treatment showed detectable differences related to GM1 reaction products, while studies in the brain showed the superiority of acetone pre-extraction (followed by formaldehyde), which yielded sharper images compared with the diffuse, blurred staining pattern associated with formaldehyde. Therefore, the aim of our study was to define the optimal conditions for the GM1 detection using cholera toxin B-subunit.Ganglioside extractability with acetone, the ever neglected topic, was tested comparing anhydrous acetone with acetone containing admixture of water. TLC analysis of acetone extractable GM1 ganglioside from liver sections did not exceed 2% of the total GM1 ganglioside content using anhydrous acetone at −20°C, and 4% at room temperature. The loss increased to 30.5% using 9:1 acetone/water. Similarly, photometric analysis of lipid sialic acid, extracted from dried liver homogenates with anhydrous acetone, showed the loss of gangliosides into acetone 3.0±0.3% only. The loss from dried brain homogenate was 9.5±1.1%.Thus, anhydrous conditions (dry tissue samples and anhydrous acetone) are crucial factors for optimal in situ ganglioside detection using acetone pre-treatment. This ensures effective physical fixation, especially in tissues rich in polar lipids (precipitation, prevention of in situ diffusion), and removal of cholesterol, which can act as a hydrophobic blocking barrier.Key words: fixation, GM1 ganglioside, cholera toxin, anhydrous acetone, 4% formaldehyde.  相似文献

Synopsis The ovary of the domestic pigeon,Columba livia, has been assayed histochemically for the localization of 5-3-hydroxysteroid dehydrogenase (5-3-HSDH), 17-hydroxysteroid dehydrogenase (17-HSDH), 11-hydroxysteroid dehydrogenase (11-HSDH), glucose-6-phosphate dehydrogenase (G6P-DH) and NADH-diaphorase activities during different periods of the reproductive cycle. 5-3-HSDH, 17-HSDH, 11-HSDH, G6P-DH and NADH-diaphorase activity was found in the theca interna of growing, atretic and postovulatory follicles, the granulosa of ovulatory, atretic and postovulatory follicles, and interstitial gland cells during the pre-incubation and the laying periods. During the incubation and squab feeding periods only 5-3-HSDH, G6P-DH and NADH-diaphorase activities were observed in the above mentioned cells. The steroidogenic potential of atretic follicles depends upon the type of atresia a follicle undergoes.  相似文献

Lipid extraction using a monophasic chloroform/methanol/water mixture, coupled with functional group selective derivatization and direct infusion nano-ESI-high-resolution/accurate MS, is shown to facilitate the simultaneous analysis of both highly polar and nonpolar lipids from a single retina lipid extract, including low abundance highly polar ganglioside lipids, nonpolar sphingolipids, and abundant glycerophospholipids. Quantitative comparison showed that the monophasic lipid extraction method yielded similar lipid distributions to those obtained from established “gold standard” biphasic lipid extraction methods known to enrich for either highly polar gangliosides or nonpolar lipids, respectively, with only modest relative ion suppression effects. This improved lipid extraction and analysis strategy therefore enables detailed lipidome analyses of lipid species across a broad range of polarities and abundances, from minimal amounts of biological samples and without need for multiple lipid class-specific extractions or chromatographic separation prior to analysis.  相似文献

Summary Osmium tetroxide and OTAN histochemical reactions have been reappraised following recent publications in this journal by Ellbder and Lojda. Provided that the standard OTAN reaction is used on frozen sections of conventional thickness, unsaturated hydrophilic polar lipids are stained in an orange or red shade while unsaturated hydrophobic non-polar lipids are stained brown-black or black. We have confirmed the anomaly — pointed out by Elleder and Lojda — that the polar lipid reaction of the atherosclerotic plaque is essentially extinguised by acetone. Solubility and blockading methods provide circumstantial evidence that Elleder and Lojda's assumed OsO4-protein reactions are due to the lipid moiety of a lipoprotein complex.  相似文献

Summary Complex carbohydrates in the human cervix were studied histochemically using lectins, conjugated to horseradish peroxidase and correlated procedures. Stratified squamous epithelium of the exocervix and columnar epithelium of the endocervix in some, but not all specimens showed staining for terminal -N-acetyl-d-galactosamine, -d-galactose, -d-galactose and -l-fucose. the staining for -N-acetylgalactosamine and -galactose, the terminal sugars in blood group A and B antigens respectively, corresponded to a large extent with ABO blood type. One exception was the lack of staining for terminal -N-acetylgalactosamine in endocervical secretions in three of nine blood type A patients. A second exception was the staining for terminal -galactose in endocervical secretions in about half of blood type O and A specimens. The type and amount of glycoprotein formed by endocervical columnar cells differed according to location in superficial compared with deep portions of the glands and according to location at the junction with exocervix compared with the more internal regions. Staining of endothelial cells for blood group A and B antigens was confined to subjects of blood type A and B respectively, although three of nine type A specimens showed no lectin reactivity for group, A antigen. Endothelial cells evidenced affinity forUlex europeus I agglutinin demonstrative of fucose in all specimens. Mast cells disclosed lectin affinity consistent with the presence of terminal or internal mannose orN-acetylglucosamine residues. Two blood type O specimens were examined with conjugated lectins at the ultrastructural level. Secretory granules stained for content of terminal -galactose, -galactose and fucose. These results support and concur with biochemical studies of complex carbohydrates in human cervical tissues. They reveal, in addition, the location of the blood group antigens in the human exocervix and endocervix and the marked heterogeneity among endocervical columnar cells in glycoprotein production.  相似文献

Summary A considerable portion of polar lipids survives the routine dehydration procedure for paraffin embedding with ethanol, acetone and xylene and can be detected in dehydrated blocks of tissue. Sphingomyelin, cerebrosides, sulphatides and gangliosides can be demonstrated with appropriate histochemical methods and chromatographically even in ordinary paraffin sections especially when the amount of these lipids in tissues is sufficiently high, e.g. in lipidoses and in normal myelin. In blocks of tissue dehydrated with acetone and cleared with benzen a considerably higher amount of polar lipids is present. Factors governing the preservation of polar lipids in paraffin sections are discussed.  相似文献

Incubation of Chironomus salivary glands with -amanitine in concentrations from 1 to 10 /ml results in the suppression of puffing and chromosomal 3H-uridine incorporation after 30 to 60 min in 80% of the cells. Nucleolar 3H-uridine incorporation remains completely unaffected. Even 4 h after the injection of high doses of -amanitine into living larvae, nucleolar incorporation is still pronounced. The distribution of resistant cells within the salivary glands suggests that the uptake of -amanitine is subject to physiological restrictions.—A puff typically induced during in vitro incubation of salivary glands was found to be less sensitive to -amanitine than the Balbiani rings.  相似文献

Summary As a prerequisite for the histochemical study of sequence iron-hematoxylin stains the iron alum-acidified hematein procedure was developed which does not require differentiation.Histochemical blocking and extraction procedures demonstrated that carboxyl and hydroxyl groups are essential for the binding of cationic iron.The iron alum-Prussian blue reaction colored collagen, reticulum fibers and basement membranes more intensely than muscle fibers. Treatment of tissue sections mordanted in iron alum with the acidified hematein solvent resulted in practically complete removal of iron from all tissue structures. It must therefore be concluded that the selective staining of muscle fibers, terminal bars and related structures with sequence iron-hematein stains is not due to high affinity of iron for these tissue components.Observations by R. and M. Heidenhain on sequence hematoxylin-potassium dichromate and hematoxylin-alum stains and data from modern textile chemistry indicate that the staining patterns obtained with metal-hematein sequence stains are determined by the affinity of the hematein moiety for certain tissue structures.  相似文献

Evolution of RNA secondary structure is studied using simulation techniques and statistical analysis of fitness landscapes. The transition from RNA sequence to RNA secondary structure leads to fitness landscapes that have local variations in their ruggedness. Evolution exploits these variations. In stable environments it moves the quasispecies toward relatively flat peaks, where not only the master sequence but also its mutants have a high fitness. In a rapidly changing environment, the situation is reversed; evolution moves the quasispecies to a region where the correlation between secondary structures of neighboring RNA sequences is relatively low. In selection for simple secondary structures the movement toward flat peaks leads to pattern generation in the RNA sequences. Patterns are generated at the level of polynucleotide frequencies and the distribution of purines and pyrimidines. The patterns increase the modularity of the sequence. They thereby prevent the formation of alternative secondary structures after mutations. The movement of the quasispecies toward relatively rugged parts of the landscape results in pattern generation at the level of the RNA secondary structure. The base-pairing frequency of the sequences increases. The patterns that are generated in the RNA sequences and the RNA secondary structures are not directly selected for and can be regarded as a side effect of the evolutionary dynamics of the system. Summary The nature of tissue calcium, detectable with glyoxal-bis-(2-hydroxyanil), (GBHA), was investigated using gelatin films as model. The results indicate that in the films the procedure detects only the calcium fraction which was ionized in the original gelatin solution. The GBHA staining intensity (absorbance) appeared to be linear with the amount of ionized calcium in the range from 0 to 2 g/cm2. The method allows detection of amounts of ionized calcium as low as 0.15 g/cm2 or 0.0015 pg/2.For the measurement of calcium in pancreatic tissue of fed rats, the tissue was subjected to freeze-substitution at –80°C in acetone containing 1% oxalic acid. Adjacent sections were stained with either GBHA or aldehyde-fuchsin (AF). Exocrine tissue hardly stained with GBHA whereas islet tissue stained intensely. For GBHA as well as for AF a variation in staining intensity (visual evaluation) between islets was observed. Islet GBHA- and AF-staining intensities did not correlate. The AF-staining intensity but not the GBHA-staining intensity decreased with increasing islet diameter. Also in pancreatic islet tissue the GBHA method appears to be very sensitive and reproducible and small differences in islet GBHA-staining intensity can be detected. The results indicate that between islets differences in ionized calcium content exist. These differences do not correlate with the degree of B-cell granulation.  相似文献

The diversity of culturable bacteria associated with sea ice from four permanently cold fjords of Spitzbergen, Arctic Ocean, was investigated. A total of 116 psychrophilic and psychrotolerant strains were isolated under aerobic conditions at 4°C. The isolates were grouped using amplified rDNA restriction analysis fingerprinting and identified by partial sequencing of 16S rRNA gene. The bacterial isolates fell in five phylogenetic groups: subclasses and of Proteobacteria, the BacillusClostridium group, the order Actinomycetales, and the Cytophaga–Flexibacter–Bacteroides (CFB) phylum. Over 70% of the isolates were affiliated with the Proteobacteria subclass. Based on phylogenetic analysis (<98% sequence similarity), over 40% of Arctic isolates represent potentially novel species or genera. Most of the isolates were psychrotolerant and grew optimally between 20 and 25°C. Only a few strains were psychrophilic, with an optimal growth at 10–15°C. The majority of the bacterial strains were able to secrete a broad range of cold-active hydrolytic enzymes into the medium at a cultivation temperature of 4°C. The isolates that are able to degrade proteins (skim milk, casein), lipids (olive oil), and polysaccharides (starch, pectin) account for, respectively, 56, 31, and 21% of sea-ice and seawater strains. The temperature dependences for enzyme production during growth and enzymatic activity were determined for two selected enzymes, -amylase and -galactosidase. Interestingly, high levels of enzyme productions were measured at growth temperatures between 4 and 10°C, and almost no production was detected at higher temperatures (20–30°C). Catalytic activity was detected even below the freezing point of water (at –5°C), demonstrating the unique properties of these enzymes.  相似文献

Control of plant growth by nitrogen and phosphorus in mesotrophic fens   总被引:7,自引:0,他引:7  
A fertilization experiment was carried out in 3 mesotrophic fens to investigate whether plant growth in these systems is controlled by the availability of N, P or K. The fens are located in an area with high N inputs from precipitation. They are annually mown in the summer to prevent succession to woodland. Above-ground plant biomass increased significantly upon N fertilization in the two mid-succession fens studied. In the late-succession fen that had been mown for at least 60 years, however, plant biomass increased significantly upon P fertilization. The mowing regime depletes the P pool in the soil, while it keeps N inputs and outputs in balance. A long-term shift occurs from limitation of plant production by N toward limitation by P. Hence, mowing is a suitable management tool to conserve the mesothrophic character of the fens.  相似文献

Summary The sensory innervation of 46 poles of long chain intrafusal muscle fibers was studied histochemically by staining for NADH-TR in periodic, 8 m thick transverse sections of cat muscle spindles. Each long chain fiber carried terminals of the primary sensory axon, and 23 of the fiber poles also displayed secondary sensory endings. With the NADH-TR reaction there was no apparent difference in the cross-sectional appearance of sensory endings on the long chain and on other nuclear chain fibers. However, the contact area between the secondary endings and the muscle fiber tended to be shorter on the long chain than on the neighboring chain fibers of shorter polar length. This was also the case for one long chain fiber in which the sensory innervation was examined in serial, 1 m thick sections stained with toluidine blue. Discharges of the secondary sensory axons in cat spindles may be affected more by contraction of the shorter nuclear chain fibers than by activation of the long chain fibers.  相似文献

The effect of different growth regulators on growth and the production of terpenoid indole alkaloids as well as some enzymes involved in the biosynthesis were studied in Catharanthus roseus seedlings. The seedlings were grown on MS solid medium containing different concentrations of each growth regulator for a period of one month. Extracted alkaloids were analyzed by HPLC for determination of terpenoid indole alkaloid quantities. Continuous availability of growth regulators induced different alkaloids with variable effects among the regulators. Gibberellic acid at concentration of either 5.8 M or 11.6 M resulted in elongation of shoots with lowering the number of leaves. Abscisic acid has a retardant effect on growth. Ethylene did not effect the growth pattern at concentration of 100 M but seedlings were not tolerant to higher concentrations. Methyljasmonate reduced the growth of the root system. Methyljasmonate was a general inducer for all alkaloids and increased the activity of strictosidine glucosidase. Ethylene applications promoted the pathways towards ajmalicine, serpentine, tabersonine and vindoline. Similar effect as for ethylene was observed for abscisic acid. Salicylic acid treatment increased the production of serpentine, tabersonine and higher concentration of salicylic acid induced vindoline accumulation. Peroxidase activity was also induced by salicylic acid. Gibberellic acid has little effect on alkaloid levels.  相似文献

An unambiguous, fully ab initio and automated technique denoted AIPAR (ab initio parametrization) implemented in the SJBR program has been proposed to yield intermolecular interaction potentials between polar molecules and water. The AIPAR procedure has been applied to several organic molecules covering a wide range of structure and functional groups, namely methanol, acetone (propanone), methanethiol (methyl mercaptan), imidazole (1,3-diazole), oxazole and furan. The AIPAR-derived sets of parameters compare well with the empirical OPLS ones, mainly when the all-atoms model is employed in the OPLS procedure. Monte Carlo simulations were performed for an aqueous solution of methanol and for an equimolar binary mixture methanol–water using the AIPAR and OPLS parameters. The thermodynamic and geometric results obtained with the parameters obtained with the AIPAR procedure compare favorably with the OPLS simulations, even for the binary mixture, demonstrating the precision, robustness and transferability of the parameters obtained with the AIPAR procedure.Figure Superimposed configurations of water (without the hydrogen atoms) around the methanol molecule obtained with the AGOA procedure.  相似文献

The extent of allelic variation of the E and E polypeptide chains of the I-E antigens from the H-2> d ,H-2 k , H-2 p , H-2 r , and H-2 u haplotypes is described. E and E chains were individually labeled with arginine or lysine and compared by tryptic peptide analysis. The results indicate minimum variability among the E polypeptides encoded by the d, k, p, and r haplotypes. However, the E u chain differed significantly from the other allelic E gene products. On the other hand, the E alleles demonstrated substantial variability with the E d being notably less similar to the other alleles than they are to each other. Summary A total of 147 muscle spindles was studied histochemically in serial transverse sections of 42 cat tenuissimus muscle specimens. Nuclear bag1, nuclear bag2 and nuclear chain intrafusal muscle fibers were distinguished by the differential staining resulting from the reactions for myosin adenosine 5-triphosphatase and nicotinamide adenine dinucleotide tetrazolium reductase. The majority of intrafusal fibers were of the same histochemical type at both fiber poles. However, seven muscle spindles contained one nuclear bag fiber each that presented as a bag1 in one pole and as a bag2 in the other pole. These mixed nuclear bag fibers were found in spindles that also contained at least one bag1 and one bag2 fiber of equivalent histochemical presentation in both fiber poles. The mixed bag fibers displayed differences of apparent fiber diameter and relative polar length between the two fiber poles. The motor innervation pattern, as revealed by staining for cholinesterase, was also dissimilar between the two poles of mixed bag fibers. The study indicates that the spindle equatorial region may in some instances serve as a boundary between two morphologically and histochemically different poles of the same intrafusal fiber.  相似文献

The Escherichia coli -galactosidase gene is frequently used as a reporter gene in transgenic studies because its activity can be easily detected at the cellular level. Here we report a procedure for monitoring -galactosidase activity directly in tissue sections, which involves the use of a mixture of ethanol and poly-ethylene-glycol as a fixative (Kryofix) and a special paraffin characterized by a lower fusion point of 42 °C. After embedding and cutting, the sections are stained by the chromogenic substrate 5-bromo-4-chloro-3-indoyl--d galactopyranoside (X-Gal). This procedure allows both the retention of a high level of -galactosidase activity and the preservation of good tissue morphology. Furthermore, it can be combined with immunohistochemical methods to detect other cellular components without compromising reporter gene detection  相似文献

Summary Life history theory predicts that migratory fishes should delay reproduction, be larger at first reproduction, and have higher fecundities than nonmigrants. We tested this hypothesis by comparing life histories of anadromous (estuary) and resident freshwater (upstream) threespine sticklebacks (Gasterosteus aculeatus L.) from the Navarro River, California, USA. Using a split-brood, two-environment breeding design, families from cach population were divided and reared in both freshwater and seawater overwintering environments. In both treatments, the more migratory estuary sticklebacks were larger at first reproduction and had large initial clutch sizes; in the freshwater treatment, the estuary sticklebacks matured later than the upstream fish. Population means varied little across treatments, indicating that the average effects of the different overwintering conditions were slight. The responses of individual families to a given overwintering treatment were highly variable in both populations, as reflected in significant family x treatment effects for all traits. Phenotypic correlations among life history traits were significant and positive for most traits, and were similar in magnitude in both populations. Differences in the relative degree of specialization for migration may in part explain variation in life history between these populations.  相似文献

Summary The reaction of substances with osmium tetroxide in tissue sections is evident when primary or secondary blackening occurs. Many substances are not colored, however, despite the fact that they may react with osmium tetroxide. Osmium derivatives bound in such structures may be either in an insufficient amount or in a higher valency state or both. For the demonstration of such weakly colored bound osmium derivatives some chemical reagents including those used in analytical chemistry were tested. Not all reagents used were equally effective. Tschugajev's reagent with thiourea is unreliable for in situ detection in tissue sections because the resulting colored complex is soluble and escapes from the section into solution. Yellow ammonium sulphide is not sensitive enough. N-phenyl-p-phenylene diamine, N-dimethyl-p-phenylene diamine, benzidine, o-dianisidine, N-diethyl-p-phenylene diamine sulphate, p-aminophenol, p-phenylene diamine, N-methyl-p-aminophenol sulphate, 2,4-diaminophenol hydrochloride, -naphthylamine, -naphthylamine, diphenylcarbazone, 8-amino-1-naphthol-3,6-disulphonic acid (H acid), aniline, tannin, proved to be the most suitable detecting reagents. By these reagents large quantities of osmium pigments could be demonstrated in many structures even after the most efficient lipid extraction methods.  相似文献
