泰和乌骨鸡的核型与带型研究   总被引:18,自引:2,他引:16

采用外周血淋巴细胞培养--空气干燥法,对泰和乌骨鸡染色体核型和带型进行了研究。结果表明:泰和乌骨鸡体细胞染色体数目2n=78,染色体基本臂数AF=90,1、9号染色体及Z、W性染色体为中央着丝粒(m)染色体,2、4、7号染色体为亚中央着丝粒(sm)染色体,3、6、8、10号染色体为端着丝粒(t)染色体。G带研究表明:前10对大型染色体可分为29个区,190条带。C带处理发现,所有母鸡分裂相中W性染色体都出现C带并整条深染。Ag-NORs研究发现:Ag-NORs常定位于1、2号常染色体短臂和Z性染色体短臂端部;Ag-NORs数目分布范围为1~6;平均每个细胞的Ag-NORs数在雌、雄鸡中分别为2.94和2.96。 Abstract:This study made the chromosome slides of Taihe Silkies by the peripheral blood lymphocyte culture-drying method,and analyzed Taihe Silkies karyotype and band pattern.The results are as follow:The diploid chromosome number of Taihe Silkies was 2n=78,the basic number of chromosome arms was AF=90 and the sex chromosome type was ZZ(♂)/ZW(♀).According to the measured relative length,arm ratio and centromeric index,the first 10 pairs of macro-chromosomes are described as follows:No.1 ,9 and Z,W chromosomes were metacentrics,No.2,4,7 were submetacentrics,and No.3,6,8,10 were telocentrics.Studies on Taihe Silkies′ G-band showed that the first 10 pairs of macro-chromosomes can be divided into 29 zones and 190 bands.Being treated by C-banding technique,a totally dark-stained and easily indentified W-chromosome always showed up in the female metaphase configurations.Ag-NORs were located in the short arms′ telomere of No.1,2 euchromosomes and Z sexchromosome,the Ag-NORs number varied from 1-6. -NORs  相似文献

以骨髓细胞为材料研究赤链蛇的染色体,结果表明该物种2n=46,由8对大型的和15对微小的染色体组成,AF=50.性染色体的大小介于3号和4号之间,为ZW型;8对大型染色体均显示着丝粒C带,1-6号还显示浅染端粒C带.W染色体为整条C带阳性;该物种一对NOR位于7号染色体近着丝粒区.不同地理分布群赤链蛇核型可能经历过Z与W染色体不等交换。 Abstract:The Karyotype,C-bands and Ag-NORs of Dinodon rufozonaturn (Cantor) reported is in this paper including the diploid number(2n=46)comprising 8 pairs of macro-and 15 pairs of microchromosomes,and AF=50 in the D.rufozonatum.The sex chromosome in size locates between the autochromosones No.3 and No.4,which belongs to ZW type.The C-banding technique revealed that the all macrochromosomes there are centromerc C band,the telomeric C band was only observed in Nos.1~6,while a whole W chromosome is constitutive heterochromatinization.Two NOR were observed on the pericentric of the No.7 chromosome.  相似文献

对家猪不同品种及家系间的染色体组型、C-带、Ag-NORs多态性进行的研究表明:杜洛克猪、约克夏猪、长白猪体细胞染色体数2n=38,核型2n=10sm+12m+4st+12t, 而13/17易位纯合子猪(36, rob. 13/17)的体细胞染色体数2n=36,核型2n=10sm+12m+6st+8t; 13/17易位杂合子猪(37,rob. 13/17)的体细胞染色体数目为 2n=37,核型为2n=10sm+12m+5st+10t。5种家猪的C-带在13~18号染色体上存在大、中、小三种类型, 且呈多态性分布。杜洛克猪、约克夏猪、长白猪、13/17易位纯合子猪、13/17易位杂合子猪的Ag-NORs均数分别为2.05、2.06、2.00、1.99、1.98,说明Ag-NORs在品种、个体及细胞间具有多态性。 Abstract:The present experiment is carried out to make comparative stdy on polymorphism of chromosome Karyotypes, C-band ?and Ag-NORs of different breeds and strains in domestic Pigs. The results showed that the chromosome number of somatic cell in Duroc, Yorkshire and Landrace Pig Was 38 and the karyotype was 2n=10sm+12m+4st+12t. But in heterozygous Rob 13/17(37, rob. 13/17) Pig, the chromosome number of somatic cell was 37 andthe karyotype was 2n=10sm+12m+5st+10t, while in homozygousRob 13/17(36, rob. 13/17) Pig, the somatic cell chromosome number was 36 and the karyotype was 2n=10sm+12m+6st+8t. It was also showed that there were three C-band types in size on chromosome No. 13-18 showing polymorphism in three breeds and two strains of domestic Pigs. The average of Ag-NORs in Duroc, yorkshire,Landrace, homozygous Rob. 13/17 and heterozygous Rob. 13/17 Pig was 2.05, 2.06, 2.00, 1.99 and 1.98, respectively. It is suggested that Ag-NORs have polymorphism among strains, which can be used as a genetic index to analyse relationships among domestic animals with high reliability.  相似文献

本文对家燕和金腰燕的核型、C带和Ag-NORs带进行了比较研究。结果表明,两者的核型基本一致。核型公式:2n=80=2M+4ST+zMwSM+2M+2ST+2SM+8 T+2M+56mT/D。但两者在染色体的相对长度、W染色体大小、G带带型、A g-NORs的数目和分布均存在着差异。文中在家燕No.1、7、W染色体和金腰燕No.1、 6、7染色体的形态划分,金腰燕W染色体的确定及其染色体数目等方面,均与卞小庄、李庆伟的结果不同。 Abstract:A comparative study on karyotype and C-band and Ag-NORs of house swallow and red-rumped swallow has been made.The result shows that the karyotype of them are similar,and their karyotypical formulae is 2n=80=2M+4ST+zMwSM+2M+2ST+8T+2M+56mT/D.But they are different in the relative length of chromosomes,the size of W chromosome,C-band patterns and the number and distribution of Ag-NORs.The chromosomes,the size of W chromosome,C-band patterns and the number and distribution of Ag-NORs.THE result in this paper is different from that of Mr.Bian and Mr.Li in the types of No.1,7,W chromosomes of house swallow and No.1,6,7 chromosomes of red-rumped swallow and the determination of W chromosome and diploid number of red-rumped swallow.  相似文献

以骨髓细胞为材料研究了棕黑锦蛇赤峰亚种的染色体, 结果表明,该物种的2n=36,由8对大型的和10对微小的染色体组成,AF=50。No.4为性染色体(ZW型);所有大型染色体均显示端粒深染C带,但仅NO.2、3、5和Z染色体显示着丝粒浅染C带。W染色体为整条C带阳性;该物种一对NOR分布于微小染色体。锦蛇属核型可能经历过染色体间的着丝粒融合的罗伯逊易位。 Abstract:This paper reports the karyotype,C-bands and Ag-NORs of Elaphe schrenckii anomala(Boulenger).The diploid number,2n=36,comprising 8 pairs of macro- and 10 pairs of microchromosomes in the E.s.anomala.AF=50.The No.4 is sex chromosome,which belong to ZW type.The C-banding technique revealed telomeric constitutive heterochromatin in the whole macrochromosome.But the centromeric C band was only observed in No.2,3,5 and Z chromosome,while a whole W chromosome is constitutive heterochromatinization.Two NORs was observed in group of microchromosome.  相似文献

采用Giemas染色、C-带、Ag-NORs、荧光染色和复制带显带的技术对黄颡鱼染色体进行了研究。结果表明, 黄颡鱼只有部分的染色体呈现阳性C-带, 可分为三类, 其中NORs区是染色最深、染色面积最大的区域, 为深染居间C-带。其Ag-NORs位于m5q末端。CMA3染色显示 NORs区呈现出明亮的荧光。中复制染色体上着丝粒区、端粒区和居间区浅染。发现核仁缢痕、深染居间C-带、Ag-NORs、CMA3明亮区和中复制带浅染NORs区位置基本一致, C-带阳性区和中复制带浅染区具有对应性。 Abstract Metaphase chromosomes of Pelteobagrus fulvidraco were analyzed by means of Giemsa staining, C-banding, Ag-NORs, fluorochrome staining and replication banding. Only parts of chromosomes showed C-band positive, the C-band heterochromatin was located in three regions. The NORs-bearing chromosomes had the largest and strongest C-bands. The long arms of chromosome pair 5 were the regions showing positive labeling with Ag-staining. Fluorochrome CMA3positively stained the NORs. In mid-period, negative replication bands were located in the centromere, the terminal and interstitial regions of the chromosomes. The distributions of secondary ?constri-ctions, positive C-bands, Ag-NORs, positive fluorescence bands and negative replication bands of mid-period were coincident.  相似文献

利用聚合酶链式反应(PCR)特异扩增家鸡W染色体高度重复序列EcoRI家族的447bp片段,作为鉴定家鸡W染色体存在的依据.4个表现型为雄性的嵌合体公鸡的精液中检测到了W染色体,通过人工授精获得嵌合体子一代.检测了其中188个雏鸡的血液,51.6%(97/188)含有W染色体,x2检验结果显示,雌雄后代符合1:1的比例,这表明在这188个成活的嵌合体子代中,基本上没有W染色体精子所产生的雏鸡,携带W染色体的精子没有将其W染色体传递到其子代雏鸡中.本研究利用PGR性别鉴定技术,从分子水平上对精液中携带W染色体的嵌合体公鸡(ZW→ZZ型)子代的性别分化作了初步的探讨。 Abstract:The W chromosome was detected in four cocks' sperms by a method based on the polymerase chain reaction which amplifies the EcoRI repeat unit of the chicken W chromosome.188 offsprings of these chimeric cocks were obtained by artificial insemination and 51.6% (97/188) was detected W chromosome in blood.The result of χ2 test suggests that the sex ratio is 1:1.It seems as if no evidence was found for skewed sex ratios of these offsprings.Therefore it seems as if the sperms with W chromosome could not contribute to these 188 offspring chicks.  相似文献

The aneuploid with isochromosome or telochromosome is ideal material for exploring the position of centromere in lingkage map.For obtaining these aneuploids in rice,the primary trisomics from triplo-1 to triplo-12 and the aneuploids derived from a triploid of indica rice variety Zhongxiao 3037 were carefully investigated.From the offsprings of triplo-10,a primary trisomic of chromosome 10 of the variety,an isotetrasomic “triplo-10-1” was obtained.Cytological investigation revealed that a pair of extra isochromosomes of triplo-10-1 were come from the short arm of chromosome 10.In the offsprings of the isotetrasomic,a secondary trisomic “triplo-10-2”,in which the extra-chromosome was an isochromosome derived from the short arm of chromosome 10,was identified.With the isotetrasomic,secondary trisomic,primary trisomic and diploid of variety Zhongxiao 3037,different molecular markers were used for exploring the position of the centromere of chromosome 10.Based on the DNA dosage effect,it was verified that the molecular markers G1125,G333 and L169 were Located on the short arm,G1084 and other 16 available molecular markers were on the long arm of chromosome 10.So the centromere of chromosome 10 was located somewhere between G1125 and G1084 according to the RFLP linkage map given by Kurata et al[1].The distance from G1125 to G1084 was about 3.2cM.  相似文献

本文研究了红鳍鲅、青稍红鲅、蒙古红鲅、翘嘴红鲅、CAN条、团头鲂和长春鳊等7种鲅亚科鱼类的银染带。结果表明,在中期分裂相中各自的A g-NORs数在4-8个之间。综合分析比较国内已发表的鲤科银染资料,得出多数中国鲤科鱼类的Ag-NORs数为4位于两对sm染色体短臂端部。鲅亚科鱼在进化过程中NORs数目有增加的趋势。 Abstract:The Ag-NORs of 7 species in Abramidinae,namely Culter erythropterus,Erythroculter dabryi,E.ilishaeformis,E.mongolicus,Hemiculter leucisclus,Megalobrama amblycephala and Parabramis pekinensis were studied.The results showed that 4~8 Ag-NORs were observed in their metaphases respectively.Based on the Ag-NORs data of Cyprinid fishes published in China,we suggested that the number of Ag-NORs in major Chinese Cyprinidae is 4,locating on the short arm tips of two pairs smchromosomes.The trend of Ag-NORs number in Abramidinae fishes is towards increasing in course of evolution.  相似文献

本文从小麦遗传图谱、物理图谱、比较基因组、基因组测序和EST 5个方面,介绍国内外小麦基因组的研究进展。我们利用W7984×Opata重组近交系的RFLP作图群体,对33个与小麦水分利用效率有关的性状进行了QTL遗传图谱比较分析,结果显明:在第一部分同源群染色体(1A,1B)上的着丝粒周围,分布有控制光合作用和根系特性的基因簇。在第二部分同源染色体上,有控制单株水分利用效率、根冠形态和生长发育的基因簇存在。在第六部分同源染色体上,6A和6B上都分别有由控制根系多个QTL组成的基因簇,6D染色体着丝粒周围有一个大的基因簇,由7个控制叶片和单株水分利用效率的QTL组成,说明第六部分同源染色体在小麦水分利用效率遗传方面起重要作用。 Abstract:Research development of genetic mapping,physics mapping,genome sequencing and expressed sequence tags in wheat have been reviewed in this paper.RFLP genetic linkage map of wheat recombinant inbred lines derived from W7984×Opata,was used to study QTL of 33 traits associated with water use efficiency.Compared with QTL map of 7 group homeologues chromosomes,the results were showed as follows:nearby the centromeric region of 1A and 1B chromosome,the gene cluster of controlling photosynthetic and root traits were located.The gene clusters of controlling water use efficiency per plant,root and plant height and growth rate were located on the 2 group chromosomes.The gene clusters of controlling root traits were located on the 6A an 6B chromosome,there was a big gene cluster mad up by 7 QTLs controlling water use efficiency of wheat leaf and per plant nearby the centromeric region of 6D chromosome.It showed that 6th homeologous chromosomes play an important role in controlling water use efficiency in wheat.  相似文献

Three species of marsupials from the Amazon region (Marmosa cinerea, Caluromys lanatus, and Didelphis marsupialis) and two from the region of S?o Paulo (Didelphis marsupialis and Didelphis albiventris) were studied. The G-banding pattern of the species with 2n = 14 (M. cinerea and C. lanatus) was very similar, as well as the pattern of G-bands in the species with 22 chromosomes (Didelphis). All of the autosomes of M. cinerea and D. albiventris have centromeric C-bands and the Y chromosome is totally C-band positive. The long arm of the M. cinerea X chromosome is completely C-band positive except for a negative band close to the centromeric region. In D. albiventris the long arm of the X chromosome is C-band positive except for a negative band close to the telomeric region. In M. cinerea the silver-stained nucleolar organizer regions (Ag-NORs) are found in the acrocentric chromosomes, being located in the telomeric region of one pair and in the centromeric region of the other pair. Caluromys lanatus has centromeric Ag-NORs in one acrocentric and in one submetacentric chromosome pairs. Didelphis marsupialis has three chromosome pairs with telomeric Ag-NORs. In D. albiventris the Ag-NORs are terminal and located in both arms of one pair and in the long arm of two pairs of chromosomes.  相似文献

以肾细胞作材料,采用秋水仙素-低渗-空气干燥法、Ag-NORs、C-带和G-带显带技术对准噶尔雅罗鱼(Leuciscus merzbacheri)染色体进行了研究。结果表明:(1)准噶尔雅罗鱼2n=50,核型组成为18m+14sm+6st+12t,NF=82,没有异型性染色体分化。(2)Ag-NORs的数目在不同的细胞中表现出多态性,数目为1~2个,出现1个Ag-NORs的频率最低(10%),出现2个的频率最高(70%);Ag-NORs主要出现在m1对和m4对同源染色体上;未发现有Ag-NORs联合的现象。(3)准噶尔雅罗鱼的染色体均呈现C-带阳性,可分为着丝粒C-带和端粒C-带。(4)同源染色体上G-带带纹基本一致,其带纹在每对染色体上的数目及分布具有明显特征性。  相似文献

利用骨髓细胞蒸气固定法制备染色体标本,研究了福建大头蛙(Limnonectesfujianensis)黄山居群的核型、C 带和Ag NORs。结果表明,福建大头蛙核型为2n =2 2 =2 0M 2SM ,NF =44,次缢痕位于No 1 0q ;各染色体均有着丝粒C 带,3p、9q出现插入型C 带;Ag NORs位于1 0q。  相似文献

本文以染色体分带技术,发现鼷鹿云南亚种的染色体数目为2n=32。全部为双胃染色体,NF=64。所有染色体着丝点区分布有C带,多数染色体的端部或两端也有C带。某些染色体还有插入C带。Y染色体C带阳性,有一Ag-NORs。文章对鼷鹿云南亚种染色体独特的C带分布以及和核型进化的关系进行了讨论。  相似文献

Observations were made of the C-banding patterns in several cells from 182 Japanese quail embryos to detect presence of stable variants. Each of the eight largest autosomes contains a C-band at the centromeric region. The short arm of autosome 8 is C-band positive, as is the entire W chromosome. The Z chromosome consistently contains an interstitial C-band in the long arm and a less prominent one in the short arm. Distinct variants of chromosome 4 and the Z chromosome were observed. In the Z chromosome a C-band at the terminal region of the short arm was markedly elongated in some embryos. Likewise, the short arm of chromosome 4 was much more prominent in one or both of the homologues in some embryos. Most of the microchromosomes contain a prominent C-band. The heteromorphisms are useful chromosome markers to detect the origins of heteroploidy in early embryos.  相似文献

本文以骨髓细胞常规空气干燥法和Sumner(1972)的方法(C带),Howell(1980)的方法(银带),对近年来昆明过冬的红嘴鸥的核型及其C带和银带进行了分析,结果显示出:红嘴鸥的2n应为68±。多数染色体的着丝粒区均显示出一个深浅不同的C带。Z染色体上有一个微小而模糊的C带,而W染色体大部分染色质被深染。其染色体经快速银染后表明,2对NORs均在微小染色体上。  相似文献

采用复制带、C带和硝酸银染色等分带技术研究了水貂的核型和带型。结果表明,2n=30,枝型为10(M)+16(SM)+2(A),XX(M)。C-带显示该水貂的一些染色体的结构异染色质比较丰富,从着丝粒区域延伸到两臂上,No.5染色体着丝粒结构异染色质有些弱化;X染色体的结构异染色质较常染色体的丰富。Ag-NORs有3个,分布在No.8染色体的次缢痕区域和一条No.2染色体长臂接近着丝粒的区域。  相似文献

本文对分布在我国的石貂北方亚种染色体进行了较详细的研究。结果表明2n=38,核型为14(M)+4(SM)+18(ST),XY(M,A)。C-带显示该亚种的一些染色体着丝粒区域结构异染色质弱化或消失。No,9染色体的短臂完全异染色质化;X染色体长臂丰出现插入杂色质带;Y为完全结构异染色质组成。  相似文献
