COLI-AEROGENES BACTERIA IN FARM WATER SUPPLIES   总被引:2,自引:2,他引:0  
SUMMARY: A series of 825 cultures of coli-aerogenes bacteria isolated at 30° and a series of 735 cultures isolated at 37° from 645 samples of farm water supplies were classified according to the recommendations of the Coliform Sub-Committee of the Society for Applied Bacteriology (Report, 1949). Klebsiella constituted 50% of the cultures isolated at 30°, whereas Escherichia coli I was the dominant type, forming 57%, among the cultures isolated at 37°. It would thus appear that isolation at 30° is as selective for Klebsiella as isolation at 37° is for Escherichia . Coli-aerogenes organisms, mainly 37° negative strains of Citrobacter freundii I and K. cloacae , were found in waters of high sanitary quality derived from protected springs and wells; but the coli-aerogenes microflora of polluted water was dominated by E. coli I, which formed 43% of the isolates at 30° and 76% of those at 37°. The results for a series of fortnightly samples from 11 farm water supplies showed a marked seasonal variation in the incidence of different types isolated at 30°; E. coli I formed a higher proportion in summer than in winter, while 37° negative strains of Klebsiella and Citrobacter formed a higher proportion in winter than in summer.  相似文献

We report our first results of attempts to rear four species of eutardigrades inhabiting different substrates, feeding on different kinds of food and characterized by different sexual conditions and modes of reproduction. Attempts were carried out to follow individual terrestrial carnivorous (Macrobiotus richtersi, M. joannae) and limnic herbivorous (Diphascon cf. scoticum; Isohypsibius monoicus) species. Carnivorous leaf litter-dwelling species were reared in small dishes containing agar as substrate and bacteriophagous nematodes as food. Five generations were obtained with the triploid thelytokous strain of M. richtersi, whereas three generations were obtained with the hermaphrodite species M. joannae. Diphascon cf. scoticum and I. monoicus were reared in small dishes containing algae as food and substrate. Several generations were obtained for both species. Males were never found in D. cf. scoticum and I. monoicus was hermaphroditic. Specimens isolated from hatchings were maintained and reproduced in both species, demonstrating parthenogenesis in the first one and self-fertilization in the latter. Consideration of the problems and on the future applications of tardigrade rearing are discussed.  相似文献

A reappraisal of oxygen uptake by Sarotherodon mossambicus was undertaken using a continuous flow respirometer. Measurements were obtained over the temperature range 16°C–37°C for fish weighing between 10 g and 150 g. Oxygen uptake was converted to energy equivalents ( Q ox) using the value 13.68 J mg O2–1and the routine metabolic energy expenditure can be described by the equation E =0.0086 t 2 0783 M 0 652 where E is the energy requirement for routine metabolism expressed in J h-1, t the temperature in °C and M the mass in g.  相似文献

Hatchery reared Atlantic salmon ( Salmo salar L.) parr were tagged with the standard I.C.E.S./I.C.N.A.F. salmon tag at water temperatures of 15°C and 6°C. They were then subjected to swimming trials in a tunnel respirometer. Following this treatment the 15°C fish developed necrotic infarcts around the tag insertions. Provided they did not become infected these lesions healed, but if the fish were retested 9 days later, the wound enlarged, became extremely inflamed, and the fish died soon after. This effect was not seen in 6°C fish.  相似文献

A. MCLEOD AND M.C.M. PEROMBELON. 1992. A conjugated Staphylococcus aureus slide agglutination test was used to detect and identify the potato blackleg pathogen, Erwinia carotovora subsp. atroseptica. Agglutination was obtained with > 108 cfu/ml of the homologous strain with a polyclonal antiserum (171) against E.c. atroseptica serogroup I which is the predominant E.c. atroseptica serogroup on potatoes in Scotland. The titre of antiserum 171 against live cells of E.c. atroseptica groups I and XXII was 2000 whereas that of other serogroups was considerably less; only 1 and 4 out of 22 serogroups of E. carotovora subsp. carotovora reacted at 1:1500 and 1:1000 antiserum dilutions, respectively and one of the three less common other E.c. atroseptica serogroups reacted at 1:1000. When tested against 24 different bacterial species including E. chrysanthemi and saprophytic bacteria present in potato tuber rots, negative results were obtained with 1:1000 antiserum dilution. The titre against heat-treated (1 h, 70°C) cells of E.c. atroseptica serogroups I and XXII was1700–2000 whereas it was < 10 against other bacteria including E.c. carotovora. Detection of E.c. atroseptica serogroups I and XXII in diseased potato tissues was achieved directly by the slide agglutination test, but lower antiserum dilutions (1:700–1000) were needed. Still lower antiserum dilutions were needed with heat-treated test material for E.c. atroseptica identification.  相似文献

SUMMARY: The modification of the 44° test proposed by Mackenzie, Taylor & Gilbert (1948) is useful for the rapid identification of Escherichia coli I in water and foods. False positive tests caused by other coli-aerogenes bacteria, or by their association with other organisms, can be considered rare. Only a few E. coli I fail to produce indole or ferment lactose at 44°, and further confirmation for routine purposes is necessary only when the results at 44° are discordant, e.g. indole positive-lactose negative or indole negative-lactose positive.  相似文献

The male gamete, a cell widely used for evaluating phylogeny in different animal groups, remains relatively unknown in tardigrades. In this paper the spermatozoa of thirteen species of eutardigrades among four genera and three families is evaluated in order to determine whether sperm morphology can be used as a taxonomic character. Spermatozoa of Amphibolus volubilis and A. weglarskae are very similar and this resemblance is congruent with the remarkable similarity of sclerified parts of the species. In addition, the spermatozoa of eight species of the genus Macrobiotus were examined yielding two groups showing strong intragroup similarities. The first group includes M. pseudohufelandi, M. sandrae, M. macrocalix, M. terminals and M.joannae , and the second M. richtersi, M. areolatus and M. harmsworthi. Again, these groupings were congruent with those determined by analysis of sclerified structures. In contrast, a marked similarity was found between the spermatozoa ot Diphascon (Adropion) scoticum and Platicrista angustata , whereas Diphascon (Diphascon) humicus was appreciably different from both species. Resemblances found in this study between spermatozoa and sclerified body parts suggests that the spermatozoa are suitable characters for use in systematic studies.  相似文献

Two types of alkaline serine proteases were isolated from the culture filtrate of an alkalophilic actinomycete, Nocardiopsis dassonvillei OPC-210. The enzymes (protease I and protease II) were purified by acetone precipitation, DEAE-Sephadex A-50, CM-Sepharose CL-6B, Sephadex G-75 and phenyl-Toyopearl 650 M column chromatography. The purified enzymes showed a single band on sodium dodecyl sulphate polyacrylamide gel electrophoresis. The molecular weights of proteases I and II were 21000 and 36000, respectively. The pIs were 6.4 (protease I) and 3.8 (protease II). The optimum pH levels for the activity of two proteases were pH 10–12 (protease I) and pH 10.5 (protease II). The optimum temperature for the activity of protease I was 70°C and that for protease II was 60°C. Protease I was stable in the range of pH 4.0–8.0 up to 60°C and protease II was stable in the range of pH 6.0–12.0 up to 50°C.  相似文献

This is the first publication describing scale-bearing Chrysophyceae from India by means of electron microscopy. Twenty seven such taxa are described from samples for ponds, rivers and thermal springs. Twenty of these are Mallomonas spp.: M. akrokomos, M. bronchartiana, M. caudata, M. ceylanica, M. costata, M. crassisquanta, M. cyathellata var. cyathellata, M. cyathellata var. chilensis, M. cyathellata var. kenyana, M. guttata, M. heterospina, M. mangofera f. mangofera, M. mangofera f. foveata, M. mangoferea I. reticulata, M. matvienkoae I. matvienkoae, M. mat-vienkoae var. grandis, M. morrisonensis, M. peronoides, M. portae-ferreae , and M. tasmanica . Three are Synura taxa: S. curtispina, S. petersenii f. petersenii , and S. petersenii I. kufferathii . Two are Spiniferomonas species: S. coronacircumspina and S. enigmata. Paraphysomonas and Chrysosphaerella were each represented by one species: P. vestita and C. longispina . The water bodies from which these samples were taken were mostly eutrophic and nutrient rich. The majority of the taxa were obtained during the summer months when water temperatures were high. This contradicts the widely held belief that silica-scaled chrysophytes are mainly found in cold oligotrophic and mesotrophic waters. Mallomonas portae-ferreae and one unidentified species of Mallomonas were recorded also from thermal springs having water temperatures up to 50°C.  相似文献
