The intrinsic viscosities, weight-average molecular weights (M?w), and radii of gyration [(R2g)12≈] of Streptococcus salivarius levan in various solvents were respectively obtained from viscosity and light-scattering measurements. The data showed that the levan in water is not aggregated by hydrogen bonds, and that the values of both the refractive index and (R2g)12 are lower in water than in aqueous solutions of urea. Urea may break intramolecular hydrogen-bonds, e.g., between branches, allowing the molecule to expand.  相似文献

A mathematical analysis is described which measures the effects of actinic light intensity and concentration of an artificial electron donor on the steady-state light-induced redox level of a reaction-center pigment (e.g. P-700) and on the overall light-induced electron flux (e.g. reduction of NADP+). The analysis led to a formulation (somewhat similar to the Michaelis-Menten equation for enzyme kinetics) in which a parameter, I12, is defined as the actinic light intensity that, at a given concentration of electron donor, renders the reaction-center pigment half oxidized and half reduced. To determine the role of a presumed reaction-center pigment, I12 is compared with another parameter, equivalent to I12, that is obtained independently of the reaction-center pigment by measuring the effect of actinic light intensity and concentration of electron donor on the overall electron flow.The theory was tested and validated in a model system with spinach Photosystem I chloroplast fragments by measurements of photooxidation of P-700 and light-induced reduction of NADP+ by reduced 2,6-dichlorophenolindophenol. A possible extension of this mathematical analysis to more general electron-transport systems is discussed.  相似文献

Isometric force levels, ranging between 0 and 100% of maximal force P0 at 2 to 3 °C, were elicited in frog sartorius muscle by means of rapidly cooling a Ringer solution containing 1·25 to 2·0 mm-caffeine. Equatorial X-ray diffraction patterns were obtained in the resting state and during contraction. The ratio of the intensities I11I10 increased with force almost linearly, with a slight upward curvature. The individual intensities for the contracting state were normalized relative to both the intensity of the undiffracted beam and the intensity of each reflection in the resting state. These normalized intensities were found to vary in a reciprocal way: I10 decreased while I11 increased throughout the range of forces studied.The gradual change in I11I10 with force level indicates that this ratio is a sensitive measure of the number of cross-bridges in the isometric state. A two-state model in which myosin projections are either in a resting or attached state and in which force is proportional to the fraction of projections in the attached state was applied to the experimental data of the individual reflections. I10 deviates from this model in a way that suggests that formation of the first few cross-bridges may decrease the regularity of the remaining unattached myosin projections.  相似文献

We previously described a model for the electrical transfer of excitation from one cell to the next which utilized the electric potential generated in the junctional cleft between the cells. Low-resistance connections between the cells were not used in the model, and it was assumed that the junctional membranes were excitable. This model was analyzed for the static case without capacitances and for the dynamic case in which capacitances were part of the circuit elements. For simplicity, the Na+ resistance (RNa), after a threshold potential was exceeded, was allowed to decrease exponentially (to 1% of its initial value) within 0·25–1·0 ms, and possible changes in the K+ resistance were ignored. In this paper, we have incorporated the Hodgkin-Huxley equations into the operation of the lumped membrane units for the electrical equivalent circuit of the cell membrane. The parameters varied are the membrane capacitances, resistances, maximum Na+ conductance (gNa), and the radial cleft resistance (Rjc). We demonstrated that our model worked very well, i.e. the successful transfer of action potentials was achieved, with the membrane units following Hodgkin-Huxley dynamics for changes in gNa and gK. The calculations indicate that transmission is facilitated when the junctional units have a higher gNa and a lower capacitance and when Rjc is elevated. Lowering the resistance of the junctional membrane units several fold, relative to the surface membrane units, also facilitated transmission; however, the absolute resistance of the junctional membrane was still well above the maximum value that would allow sufficient local-circuit current to flow to effect transmission. Thus, the electric field model provides an alternative means of cell-to-cell propagation between myocardial cells which is electrical in nature but does not require the presence of low-resistance connections between cells.  相似文献

Various hemoglobin derivatives have been labeled at the Cys-β93 residue with a bulky and “strongly immobilized” nitroxide maleimide (I) and a smaller, more flexible and “weakly immobilized” nitroxide iodoacetamide (II) and crystallized. The angular dependence of the paramagnetic resonance of the spin-label was measured for the ab, ac1 and bc1 planes at 298 K and 77 K for spin-labeled crystals of Oxyhemoglobin, methemoglobin fluoride, and methemoglobin azide. In the case of the methemoglobin crystals, the angular variation of the heme resonance was also monitored at 77 K. From the hyperfine splitting data, the spin-label I was found to assume specific orientations at both temperatures with some motional narrowing at 298 K. Spin-label II is specifically oriented only at room temperature but is frozen at 77 K in random orientations. Oxyhemoglobin labeled with I (I-HbO22) has the most prominent spin-label orientation (zb, xa) and the less abundant spin-labels with (zb ± 15 °) (Ohnishi et al., 1966). The corresponding spin-label orientations for I-Hb+ F? are (z∥a, x∥c1) and (z∥c1, x∥a). Crystals of I-Hb+ N?3 have spin-labels oriented along angular directions similar, but not identical to those of I-Hb+F?. Therefore, there are probably significant peptide segmental displacements when HbO2 is oxidized to methemoglobins. At 25 °C II-Hb+ N?3 has spin-label orientations not too different from those in I-Hb+ N?3, whereas in HbO2 the two spin-labels show significant differences in their orientations.  相似文献

Quenching of singlet molecular oxygen (1ΔgO2) by α-tocopherol (I) involves the hydroxy function of the chromanol ring of I. In phosphatidylcholine (PC) uni- and multilamellar vesicles this structural element of I is localized at the interface polar headgroup/hydrophobic core. A dielectric constant of ? ~ 25 was determined for this special region of the PC bilayer. The ratio kQ/kR of rate constants of quenching processes (kQ) and irreversible reactions (kR) of I with 1ΔgO2 increases with decreasing polarity of the solvent. In ethanolic solutions where ? = 25.5, kQ/kR is about 40. Extrapolation of these results to phospholipid bilayers suggests that at the nearness of the ester carbonyl oxygen of the PC fatty acid moieties, α-tocopherol can deactivate approximately 40 1ΔgO2 molecules before being destroyed. It is concluded that in vivo, one may expect to find a higher kQ/kR ratio if the chromanol ring of I hides within the more hydrophobic interiors of the membrane surface peptides.  相似文献

The relative effectiveness of oxidizing (.OH, H2O2), ambivalent (O2?) and reducing free radicals (e? and CO2?) in causing damage to membranes and membrane-bound glyceraldehyde-3-phosphate dehydrogenase of resealed erythrocyte ghosts has been determined. The rates of damage to membranebound glyceraldehyde-3-phosphate dehydrogenase (R(enz)) were measured and the rates of damage to membranes (R(mb)) were assessed by measuring changes in permeability of the resealed ghosts to the relatively low molecular weight substrates of glyceraldehyde-3-phosphate dehydrogenase. Each radical was selectively isolated from the mixture produced during gamma-irradiation, using appropriate mixtures of scavengers such as catalase, superoxide dismutase and formate. .OH, O2? and H2 O2 were approximately equally effective in inactivating membrane-bound glyceraldehyde-3-phosphate dehydrogenase, while e? and CO2? were the least effective. R(enz) values of O2? and H2O2 were 10-times and of .OH 15-times that of e?. R(mb) values were quite similar for e? and H2O2 (about twice that of O2?), while that of .OH was 3-times that of O2?. Hence, with respect to R(mb): .OH >e? = H2O2 >O2? , and with respect to R(enz): .OH >O2? = H2O2 >e?. The difference between the effectiveness of the most damaging and the least damaging free radicals was more than 10-fold greater in damage to the enzyme than to the membranes. Comparison between H2O2 added as a chemical reagent and H2O2 formed by irradiation showed that membranes and membrane-bound glyceraldehyde-3-phosphate dehydrogenase were relatively inert to reagent H2O2 but markedly susceptible to the latter.  相似文献

NH2OH-treated, non-water-splitting chloroplasts can oxidize H2O2 to O2 through Photosystem II at substantial rates (100–250 μequiv · h?1 · mg?1 chlorophyll with 5 mM H2O2) using 2,5-dimethyl-p-benzoquinone as an electron acceptor in the presence of the plastoquinone antagonist dibromothymoquinone. This H2O2 → Photosystem II → dimethylquinone reaction supports phosphorylation with a Pe2 ratio of 0.25–0.35 and proton uptake with H+e values of 0.67 (pH 8)–0.85 (pH 6). These are close to the Pe2 value of 0.3–0.38 and the H+e values of 0.7–0.93 found in parallel experiments for the H2O → Photosystem II → dimethylquinone reaction in untreated chloroplasts. Semi-quantitative data are also presented which show that the donor → Photosystem II → dibromothymoquinone (→O2) reaction can support phosphorylation when the donor used is a proton-releasing reductant (benzidine, catechol) but not when it is a non-proton carrier (I?, ferrocyanide).  相似文献

The following peptides were synthesized by classical methods in solution: Ac-Gly-Gly- Val-Arg-Gly-Pro-Arg-Val-Val-Glu-Arg-NHCH3 (A), Ac-Ala-Glu-Gly-Gly-Gly-Val- Arg-Gly-Pro-Arg-Val-Val-Glu-Arg-NHCH3 (B), and Ac-Phe-Leu-Ala-Glu-Gly-Gly- Gly-Val-Arg-Gly-Pro-Arg-Val-Val-Glu-Arg-NHCH3 (C). The rates of hydrolysis of the Arg-Gly bond of these three peptides by thrombin were measured, and the values of kcatKm were found to be 0.05 × 10?7 (A), 0.02 × 10?7 (B), and 1.6 × 10?7 (C) [(NIH units/ liter)s]?1. The value ofkcatKm for peptide C is less than 1% of that for fibrinogen [although the value of kcat itself, for peptide C (but not for A or B), is comparable to that for fibrinogen]. These results indicate that phenylanine and leucine at positions P9 and P8, respectively, play a key role in the reaction of thrombin with fibrinogen. The data also show that factors outside of the 16 residues of peptide C are important in determining the rate of hydrolysis of fibrogen by thrombin.  相似文献

(1) Aqueous solutions of 1–10 μM ferricytochrome c treated with 100 μM–100 mM H2O2 at pH 8.0 emit chemiluminescence with quantum yield Ф ? 10?9 and absolute maximum intensity Imax ? 105 hv/s per cm3 (λ = 440), and exhibit exponential decay with a rate constant of 0.15 s?1. (2) The emission spectrum of the chemiluminescence covers the range 380–620 nm with the maximum at 460 ± 10 nm. (3) Neither cytochrome c nor haemin fluoresce in the spectral region of the chemiluminescence. In the reaction course with H2O2, a weak fluorescence in the region 400–620 nm with λmax = 465–510 nm (λexc 315–430 nm) gradually arises. This originates from tryptophan oxidation products of the formylkynurenine type or from imidazole derivatives, respectively. (4) Frozen solutions (77 K) of cytochrome c exhibit phosphorescence typical of tryptophan (λexc = 280 nm, λem = 450 nm). During the peroxidation, an additional phosphorescence gradually appears in the range 480–620 nm with λmax = 530 nm (λexc = 340 nm). This originates from oxidative degradation products of tryptophan. (5) There are no red bands in the chemiluminescence spectra of cytochrome c or haemin. This result suggests that singlet molecular oxygen O2(1Δg) is not involved in either peroxidation or chemiluminescence. (6) The haem Fe3+ group and H2O2 appear to be crucial for the chemiluminescence. It is suggested that the generation of electronically excited, light-emitting states is coupled to the production of conformational out-of-equilibrium states of peroxy-Fe-protoporphyrin IX compounds.  相似文献

The interaction of |CnH2n+1N+(CH3)3| · I? (n = 3, 6, 9, 12, 14, 16 or 18) with egg-yolk phosphatidylcholine-water dispersions has been studied by 31P-NMR spectroscopy. It is shown that the effective anisotropy of 31P chemical shift (?Δσeff) of the lamellar phospholipid liquid-crystalline phase Lα increases with increasing concentration and alkyl chain length of the drug. Addition of |C6H13N+(CH3)3| ·I ? or |C9H19N+(CH3)3I? to the phospholipid-water dispersion at a molar ratio ammonium salt:phospholipid > 0.8 induces in the dispersion a structure with an effective isotropic phospholipid motion. This structure is unstable and slowly transforms into the hexagonal phase. These effects have not been observed in phospholipid-water dispersions mixed with the ammonium derivatives with the longer alkyl chains n  12, 14, 16 or 18. It is proposed that these results might explain the effects of the investigated drugs on the nerve, muscle and bacterial cells.  相似文献

A quantitative structure-activity relationship has been formulated for 53 alkyl phosphonates [R2OPO(CH3)SR3] inhibiting chymotrypsin: log ki = 1.47MROR2 + 0.34MRSR3 + 1.25σ31 ? 1.06I ? 3.43 log (β·10MROR2 + 1) ? 5.26; log β = ?3.85. In this so-called bilinear model, ki is the bimolecular rate constant (m?1 s?1), β is a disposable parameter evaluated by a computerized iterative procedure, MR is the molar refractivity of a substituent, σ31 is Taft's polar parameter, and I is an indicator variable for substituents containing a sulfonium group. The correlation coefficient for this equation is 0.985. This quantitative structure-activity relationship is compared with those previously formulated for the action of chymotrypsin on acylamino acid ester substrates.  相似文献

Washed platelets were surface-labelled by lactoperoxidase catalyzed iodination and either the platelets or membranes were solubilized in detergent and applied to a wheat germ agglutinin-Sepharose column and a Lens culinaris lectin Sepharose column coupled sequentially. The glycoproteins eluted from the lectin columns were separated by two-dimensional gel electrophoresis. Alternatively, labelled whole platelets or membranes were solubilized and then directly separated by two-dimensional polyacrylamide gel electrophoresis. Spots corresponding to specific glycoproteins identified by apparent isoelectric point (pI), apparent molecular weight (Mr), staining and labelling characteristics were cut from the gels and analyzed by tryptic peptide mapping. The maps of the individual glycoproteins (GP) Ia, Ib, IIa, IIb, GP132–1354–4.5 IIIa, IIIb and IIIc were all different. Glycoproteins with the same Mr but different pI were distinct with the exception of regions of GP Ib. There were minor differences in the maps of glycoproteins separated in the reduced or non-reduced state. Tryptic peptide maps provide a valuable additional parameter for the identification and characterization of platelet glycoproteins.  相似文献

The dimerization of deuteroferrihaem in aqueous solution has been investigated using a parameter, named the dimerization index (Robs). This is defined as the ratio of extinction coefficients at wavelengths corresponding to Soret band maxima for the monomeric and dimeric species, respectively. For solutions containing mainly monomeric species, Robs > 2, whereas for solutions containing mainly dimeric species Robs < 1.A computer programme has been applied to determine values of the dimerization constant, K, defined as: K = [dimer][H+][monomer]2.Phosphate buffer anions and Tris · HCl buffer enhanced dimerization. Monovalent and divalent cations also increased dimerization, but in a specific manner. The magnitudes of their effects increased in the order K+ < Na+ < Li+ < Sr2+ < Mg2+ ? Ca2+. Values of K were determined for several concentrations of Na+ and Sr2+. These data are interpreted in terms of a stabilization of the ferrihaem dimer by the formation of ion triplets with the added cation ‘sandwiched’ between carboxyl residues of the adjacent ferrihaem monomeric units. General guidelines are recommended for the choice of conditions which minimize dimerization.  相似文献

The formation of chlorophyll triplet states during illumination of Photosystem I reaction center samples depends upon the redox state of P-700, X and ferredoxin Centers A and B. When the reaction centers are in the states P-700+A1XFdBFd?A and P-700 A1XFd?BFd?A prior to illumination, we observe electron paramagnetic resonance (EPR) spectra from a triplet species which has zero-field splitting parameters (|D| and |E|) larger than those of either the chlorophyll a or chlorophyll b monomer triplet, and a polarization which results from population of the triplet spin sublevels by an intersystem crossing mechanism. We interpret this triplet as arising from photoexcited chlorophyll antenna species associated with reaction centers in the states P-700+Fd?A and P-700+X?, respectively, which undergo de-excitation via intersystem crossing. When the reaction centers are in the states P-700A1XFd?BFd?A and P-700A1X?Fd?BFd?A prior to illumination, we observe a triplet EPR signal with a polarization which results from population of the triplet spin sublevels by radical pair recombination, and which has a |D| value similar to that of chlorophyll a monomer. We interpret this triplet (the radical pair-polarized triplet) as arising from 3P-700 which has been populated by the process P-700+A?13P-700A1. We observe both the radical pair-polarized triplet and the chlorophyll antenna triplet when the reaction centers are in the state P-700 A1XFd?BFd?A, presumably because the processes P-700+A?1X → P-700+A1X? and P-700+A?1X3P-700 A1X have similar rate constants when Centers A and B are reduced, i.e., the forward electron transfer time from A?1 to X is apparently much slower in the redox state P-700 A1XFd?BFd?A than it is in state P-700 A1XFdBFdA. The amplitude of the radical pair-polarized triplet EPR signal does not decrease in the presence of a 13.5-G-wide EPR signal centered at g 2.0 which was recorded in the dark prior to triplet measurements in samples previously frozen under intense illumination. This g 2.0 signal, which has been attributed to phototrapped A?1 (Heathcote, P., Timofeev, K.N. and Evans, M.C.W. (1979) FEBS Lett. 101, 105–109), corresponds to as many as 12 spins per P-700 and can be photogenerated during freezing without causing any apparent attenuation of the radical pair-polarized triplet amplitude. We conclude that species other than A?1 contribute to the g 2.0 signal.  相似文献

Fourteen derivatives of l-alanine of the type CH3CH(NHCO-3-C5H4N)COOR3 have been synthesized and their hydrolysis by chymotrypsin was studied with the object of characterizing enzymic space (?3) to which R3 binds. The binding of R3 (log1Km) was shown via correlation analysis to correlate with molar refractivity (MR) of R3 rather than hydrophobicity (π). The results confirmed our earlier predictions. A correlation equation for the hydrolysis of 77 acyl-amino acid esters of the general formula R2CH(NHCOR1)COOR3 relating log(kcatkm) to molar refractivity of R1, R2, and R3 and to σ1 (Taft's polar parameter) of R3 was formulated. The general picture of ligand interactions with chymotrypsin as seen with correlation analysis is discussed.  相似文献

Turtle bladders bathed on both surfaces with identical HCO?3/CO2-rich, Cl?-free Na+ media and treated with ouabain and amiloride exhibit a transepithelial potential serosa electronegative to mucosa and a short-circuit current (Isc) which is a measure of the net luminal acidification rate. Addition of calcium ionophore A23187 (10 μM) to the mucosal side of the epithelium rapidly reverses the direction of the potential difference and Isc and decreases tissue resistance. The resulting positive Isc resembles that previously observed in response to isobutylmethylxanthine (IBMX) and cAMP analogs. Reversal of the Isc is enhanced in bladders from severely alkalotic turtles. In contrast, in severely acidotic turtles, ionophore A23187 decreases, but does not reverse, the Isc. The data suggest that, like IBMX and cAMP analogs, the Ca ionophore stimulates an electrogenic alkalinization mechanism, but, unlike the former agents inhibits the concurrent acidification process as well.  相似文献

The molecular weight of the adenovirus type 2 hexon was calculated from sedimentation equilibrium, light scattering and sedimentation and diffusion experiments. The extinction coefficient, E1 cm1%, was determined to be 14.3 at 279 nm, from quantitative nitrogen and carbon analyses combined with the N,C content calculated from the amino acid composition. Other parameters determined were: the partial specific volume, \?gn = 0.738 cm3 g?1; the refractive index increment, (?n?c)T,P = 0.193 cm3 g?1 at 435.8 nm; the sedimentation coefficient, s20,w0 = 13.0 S; and the diffusion constant, D20, w0 = 3.32 × 10?7 cm2 s?1. All molecular weights were between 355,000 and 363,000. Crystal density measurements were made on native and glutaraldehyde cross-linked crystals and the molecular weights calculated from these data were compared with the precise molecular weight determined by physico-chemical methods.Only one polypeptide of molecular weight 120,000 was found in reduced, or reduced and alkylated, hexon. Four or six organomercurial molecules were bound per 120,000 molecular weight of native hexon upon titration with 2-chloromercuri-4-nitrophenol and 2-chloromercuri-4,6-dinitrophenol, respectively. With 5,5′-dithiobis (2-nitrobenzoic acid) only one SH-group per 120,000 could be titrated in native hexon, but after denaturation in 1% sodium dodeeyl sulphate five more SH-groups reacted per 120,000 molecular weight. Thus there are three identical polypeptides of molecular weight 120,000 per hexon of total molecular weight 360,000.  相似文献
