玉米品种遗传多态性与根系内生细菌种群的相互关系   总被引:6,自引:0,他引:6

对辽宁省14个玉米主栽品种根系内主要细菌种群的分析结果表明,芽孢杆菌属(Bucillus ssp.)为玉米内生细菌的主要种群,其它种属包括肠杆菌属、沙雷氏杆菌属、假单孢菌属、黄单孢菌属和棍状杆菌属。内生细菌在不同玉米品种和不同生育期之间存在程度不同的差异。从90条随机引物中筛选出条带清晰、重复性好的13个引物,对14个玉米主栽品种进行遗传多态性的RAPD分析,共扩增出条带139条,其中多态性条带101条,多态性比率72.7%。利用SPSS统计分析软件对14个玉米品种在DNA水平和内生细菌种群水平上进行聚类分析,结果表明,品种亲缘关系在这两个水平上表现基本一致,即品种内生细菌的种类及其数量在很大程度上受品种的遗传背景控制。  相似文献

从3个供试玉米品种(川单13、川单418、川单416)苗期分离、纯化内生细菌68株,经形态及理化特征鉴定,分属5个不同的属,微球菌属和芽孢杆菌属为优势属.芽孢杆菌属细菌鉴定出5个种.供试品种中内生细菌种群数量各不相同,种群数量由多到少依次是川单13、川单418和川单416.通过抑菌谱和拮抗菌发酵液对种子萌芽率的影响,筛选出两株抑菌谱广,对玉米种子萌发无抑制作用的内生芽孢细菌BH和B98.盆栽实验显示,两株细菌均对玉米植株具有促生作用,对玉米纹枯病具有防治效果.  相似文献

武夷山地衣表生和内生芽孢杆菌种群的多样性   总被引:2,自引:0,他引:2  
摘要:【目的】分析武夷山地衣表生和内生芽孢杆菌种群的多样性。【方法】从武夷山自然保护区采集扁枝衣属(Evernia)、珊瑚枝属(Stereocaulon)、孔叶衣属(Menegazzia)等分属于7科9属的地衣样品9份,分离地衣表面附生(表生)和内生芽孢杆菌,并根据16S rRNA基因序列同源性对分离得到的芽孢杆菌进行种的鉴定。【结果】未从扁枝衣属、树花属(Ramalina)和茶渍属(Lecanora)的3个样品中分离出表生或内生芽孢杆菌,从另外6个样品中分离出芽孢杆菌34株,分别鉴定为芽孢杆菌属(Bacillus)、类芽孢杆菌属(Paenibacillus)、短芽孢杆菌属(Brevibacillus)、赖氨酸芽孢杆菌属(Lysinibacillus)和绿芽孢杆菌属(Viridiibacillus)的24个种。类芽孢杆菌属、芽孢杆菌属是这些地衣表生、内生芽孢杆菌的优势种群,分别占分离得到菌株的41.2%(14株)和35.3%(12株);短芽孢杆菌属、赖氨酸芽孢杆菌属和绿芽孢杆菌属为首次报道从地衣中分离获得。从不同的地衣样品分离的表生和内生芽孢杆菌的种类、数量存在差异。蜈蚣衣属(Physcia)表生的芽孢杆菌数量最多、可达3.85×106 cfu/g,而珊瑚枝属表面附生和内生芽孢杆菌的种类最多、共有12个种。分离到的芽孢杆菌大部分来自单独的一种地衣;台中类芽孢杆菌(Paenibacillus taichungensis)、土壤短芽孢杆菌(Brevibacillus agri)等4个种存在于2-3种地衣中;蕈状芽孢杆菌(Bacillus mycoides)分布最广,在蜈蚣衣属、珊瑚枝属等4种地衣中都有存在。【结论】武夷山地衣表生和内生芽孢杆菌存在种类和数量的多样性。  相似文献

对东湖磷细菌种群结构研究结果表明,至少有芽孢杆菌属,微球菌属,不动细菌属,气单胞菌属,产碱杆菌属,贝内克氏菌属,无色杆菌属,短杆菌属,肠杆菌属,黄杆菌尾,微杆菌 ,假单胞菌属,沙雷氏菌属,黄单胞菌属和发酵单胞菌属15个属的不同菌株以其对有机磷,无机磷化合物的分解能力推动着湖泊中磷的循环,其中以芽孢杆菌属和微球菌属居于明显的优势。磷细菌在湖泊中的分布随水质的污染程度和磷化合物含量的不同而表现有明显的判别:在数量上,湖泥高于湖水,污染较重的I站高于水质较好的II站,在调查期间,有机磷细菌数在多数情况下亦高于无机磷细菌,在种群结构上,1站湖水中种类最多,Ⅲ站次之,II站最少。  相似文献

从盐生植物海滨锦葵块根中分离内生细菌43株,经形态学特征和16S r DNA序列相结合的方法鉴定,分属10个种属,其中芽孢杆菌属是优势属,其次是假单胞菌属和农杆菌属,蜡样芽孢杆菌(Bacillus cereus)和地衣芽孢杆菌(Bacillus licheniformis)是优势种。对海滨锦葵内生细菌ACC脱氨酶活性的测定显示,其中5种菌明显具有ACC脱氨酶活性。用筛选到的5种细菌接种盐胁迫下小麦根系并测定其对于小麦耐盐性的影响。结果表明,蜡样芽孢杆菌、巨大芽孢杆菌(Bacillus megaterium)、短小芽孢杆菌(Bacillus pumilus)、地衣芽孢杆菌四种芽孢杆菌均能显著提高盐胁迫下小麦幼苗的干物质重和叶绿素含量,并能显著提高保护酶(SOD、POD、CAT)活性,对盐胁迫的毒害有一定的缓解作用。绿针假单胞菌(Pseudomonas chlororaphis)对小麦幼苗株高、根长、鲜重、干重、叶绿素含量和保护酶活性的提高也具有一定的作用。上述分析表明从海滨锦葵块根中分离出的5株具有ACC脱氨酶活性的内生细菌均能提高小麦幼苗的耐盐性。  相似文献

为了探明内生细菌在番茄中的分布和数量变化规律,有目的地筛选防治番茄青枯病的内生细菌,我们对广西可培养的番茄内生细菌的类群和数量动态进行了调查。从广西部分县市采集的303个番茄样本中分离到624株内生细菌菌株,初步确定有芽孢杆菌(Bacillus)、假单胞菌(Pseudomonas)、黄单胞菌(Xanthomonas)、棒杆菌(Corynebacterium)、土壤杆菌(Agrobacterium)、微杆菌(Microbacterium)、肠杆菌(Enterobacter)和欧文氏菌(Erwinia)8个属,其中以芽孢杆菌、假单胞菌和土壤杆菌为芽孢杆菌为优势类群。番茄内生细菌在植株器官中的分布以根部数量最多,其次是茎和叶。内生细菌的总量在番茄生育期的变化趋势是从苗期到花期数量上升,而从结果期到成熟期数量逐渐下降。多数内生细菌种群的数量变化动态符合细菌总量的变化趋势,只有微杆菌在番茄植株整个生育期中始终保持下降的趋势。春季种植的番茄植株的内生细菌类群数量比秋季种植的少。  相似文献

【背景】植物内生菌长期与宿主共生,对宿主生长发育产生影响。葛根作为重要的药食两用作物,葛根内生菌的研究具有重要实践意义。【目的】对广西葛根根部内生细菌进行分离、鉴定及促植物生长特性分析,旨在了解该药食同源植物内生细菌种群结构及其促生特性,为分析内生菌群体在药食同源植物产量和品质形成的作用及其内生细菌资源的开发利用提供参考。【方法】采用6种不同的培养基从广西葛根的根瘤、根系和根愈伤组织分离内生细菌,16S rRNA基因测序和系统发育分析内生细菌的分布特征和遗传多样性,采用生理生化方法测定分离菌株的固氮活性、溶磷特性、产生嗜铁素、分泌吲哚乙酸(indole-3-aceticacid,IAA)等促生特性。【结果】从葛根根瘤、根系和根部愈伤组织中共分离得到223个菌株,16S rRNA基因测序鉴定这些菌株隶属于2门4纲10科19属,其中芽孢杆菌属、假单胞菌属、土壤杆菌属、肠杆菌属为葛根优势菌群;内生细菌数量和群落组成存在明显的组织特异性,其数量表现为根瘤>根系>根愈伤组织,但其种群多样性表现为根愈伤组织>根系>根瘤。不同培养基分离出的细菌种群丰富度有差异。从供试菌株中筛...  相似文献

兰科植物内生细菌与菌根真菌的协作对宿主植物的生长、抗病、抗逆及植物修复环境能力等具有重要意义,揭示其内生细菌多样性及与生境之间的关系有助于阐明兰科植物的适应与进化机制。本研究基于16SrDNA序列分析探讨了不同生境下东南亚特有种五唇兰根部可培养内生细菌多样性及其空间异质性。结果表明:从不同生境下五唇兰根部共分离出内生细菌59株,其中从土生型五唇兰根部分离出内生细菌45株(76.27%),从石生型五唇兰根部分离出内生细菌14株(23.73%);基于内生细菌16SrDNA序列同源性分析及构建的系统发育树显示,五唇兰根部内生细菌分属于7属,即芽孢杆菌属(Bacillus)、伯克氏菌属(Burkholderia)、草酸菌属(Pandoraea)、土壤杆菌属(Agrobacterium)、类芽孢杆菌属(Paenibacillus)、泛菌属(Pantoea)、欧文氏菌属(Erwinia),其中优势属为芽孢杆菌属,次优势属为泛菌属和伯克氏菌属;多样性分析显示,土生型五唇兰根部内生细菌群落的Shannon多样性指数大于石生型五唇兰,不同生境下五唇兰根部内生细菌群落结构差异极显著(P0.01)。土生型五唇兰根部内生细菌群落优势属为芽孢杆菌属和泛菌属,石生型五唇兰根部内生细菌群落优势属为芽孢杆菌属和伯克氏菌属。  相似文献

采用化学法进行表面灭菌处理,运用平板涂布法及平板划线法从番茄茎内得到17株内生细菌.通过形态观察和生理生化指标鉴定,17株内生细菌分属于6个属,即葡萄球菌属(Staphylococcus)、短芽孢杆菌属(Brevi-bacillus)、芽孢杆菌属(Bacillus)、棍状杆菌属(Clavibacter)、欧文氏菌属(Erwinia)和乳杆菌属(Lactobacillus).采用滤纸片法从17株内生菌中筛选出1株对青枯菌(Pseudomonas solanacearum)有拮抗作用的菌株,编号为TS-06,属于芽孢杆菌属(Bacillus).其抑菌圈半径为2.5 mm.  相似文献

【目的】植物内生细菌在促进植物生长和重金属污染土壤修复中发挥着重要作用。本研究对镉污染水稻种子内生细菌进行分离和功能鉴定,从而确定其镉耐受性、植物促生性以及对镉胁迫下水稻种子萌发和幼苗生长的影响。【方法】采用乙醇-次氯酸钠联合灭菌法对水稻种子进行表面灭菌,采用标准平板培养法对内生细菌进行分离、纯化;对菌株16SrRNA基因进行扩增和测序,鉴定菌株的分类学地位;运用微量稀释法、电感耦合等离子体质谱法、比色法测定内生细菌的镉耐受性、镉去除率和植物促生性;采用浸种法进行菌剂侵染,观察施加菌剂对水稻种子萌发、幼苗生长和镉积累的影响。【结果】从两个水稻品种的种子中共分离得到133株内生细菌,分属于12个属的24个种。在低镉品种728B中分离得到83个菌株,主要包括假单胞菌属(34.94%)、芽孢杆菌属(28.92%)和类芽孢杆菌属(10.84%)菌株。在高镉品种博B中分离得到50个菌株,主要包括泛菌属(40%)、短小杆菌属(22%)和微杆菌属(12%)菌株。对24个代表性菌株的镉耐受性进行测定,发现镉离子对菌株的最小抑制浓度介于80–2 560μmol/L,其中芽孢杆菌HNR-4具有最高的镉耐受...  相似文献

The comparative study of adhesive, hemolytic, DNA-ase, lecithinase, antilysozymic, anticomplementary activities of mono- and associated cultures of 57 Enterobacter spp., 61 Citrobacter spp. and 55 Serratia spp. strains, isolated from patients with pyoinflammatory, intestinal and urological diseases is carried out. Different variations of cocultivated bacteria including Enterobacter and Citrobacter, Enterobacter and Serratia, Citrobacter and Serratia are used. It was shown, that cocultivated Enterobacter, Citrobacter and Serratia bacteria increased the persistent properties of mixt cultures.  相似文献

以定向分离培养和基于16S rDNA的PCR-DGGE (Denaturing gradient gel electrophoresis, DGGE)方法, 分析感黄龙病柑橘与健康柑橘植株不同部位的内生细菌多样性, 分离柑橘组织共获得19株可培养的兼性厌氧型内生细菌, 经形态、生理生化结合16S rDNA分子方法鉴定其隶属于12个属, 其中短小杆菌属Curtobacterium sp. (IF: 29.07%)、芽孢杆菌属Bacillus sp. (IF: 23.12%)和微杆菌属Microbacterium sp. (IF: 21.09%)为罹病植株的优势菌群, 芽孢杆菌属Bacillus sp. (IF: 21.03%)、动性球菌属Planococcus sp. (IF: 20.69%)和假单胞菌属Pseudomonas sp. (IF: 17.44%)为无症健株的优势菌群。对DGGE方法得到的50条16S rDNA目标条带进行序列比对, 共鉴定出9个属的细菌, 结果表明沙雷氏菌属Serrations sp. (IF: 28%)是优势菌属, 泛菌属Pantoea sp. (IF: 14%)是次优势菌属; 病果桔络中黄龙病菌含量最高(>1%), 而罹病植株其他部位的黄龙病菌丰度较低。PCR-DGGE 图谱也显示感病和健康柑橘组织的内生细菌存在差异。  相似文献

Spermosphere establishment by bacteria which were coated onto seeds was studied using soybean seeds treated with four bacterial strains at levels of log10 1 to 4 colony-forming units (cfu) per seed planted in a field soil mix, and incubated 48 h. Each strain at every inoculum level developed spermosphere population densities of log10 4 to 8 cfu/seed, demonstrating an average multiplication of log10 3 cfu/seed. An alternative method was developed to differentially rank bacteria for spermosphere colonizing capacity, based upon incorporation of bacteria into a soil and monitoring the resulting spermosphere population densities around noninoculated seeds after 4 days at 14 degrees C. Fifty-seven bacterial strains which were isolated from soybean roots or from water samples, including Pseudomonas putida, P. putida biovar B, P. fluorescens, Serratia liquefaciens, Enterobacter aerogenes, and Bacillus spp. were tested in the spermosphere colonization assay. Average spermosphere population densities for the 57 strains ranged from 0 to log10 7.0 cfu/seed. Strains of a given taxon demonstrated marked diversity with ranges from 0 to log10 6.0 cfu/seed for Bacillus spp. and from log10 1.4 to 7.0 cfu/seed for Pseudomonas putida. The relative ranking of representative strains was consistent in repeating experiments. The potential usefulness of the assay for efforts to develop competitive bacterial inoculants for crop seeds is discussed.  相似文献

Five different species of freshwater bacteria ( Pseudomonas sp., Vibrio sp., Klebsiella sp., Enterobacter sp., Serratia sp.) and a mixed natural population were used separately to inoculate cultures of axenic duckweed ( Lemna minor ). Inoculation with Vibrio sp. caused the final population density of Lemna plants to be significantly greater after 52 d than that of either axenic controls or Lemna inoculated with a mixed bacterial community. Inoculation with Pseudomonas sp. caused the final population density of Lemna to be significantly higher than with the mixed bacterial treatment. Inoculation of Lemna with Klebsiella sp., Enterobacter sp. or Serratia sp. resulted in higher plant populations compared with controls, but these differences were not statistically significant. The presence of a mixed community of bacteria did not significantly affect the final population density of Lemna compared with the controls. However, Lemna plants inoculated with a natural population of bacteria showed significantly higher levels of senescence compared with the other five treatments and the controls. None of the five single bacterial taxa used appeared to have any significant effect of the sensescence of duckweed.  相似文献

The thermophilic microbiota of solid-waste composting, with major emphasis on Bacillus spp., was examined with Trypticase soy broth (BBL Microbiology Systems) with 2% agar as the initial plating medium. Five 4.5-liter laboratory units at 49 to 69 degrees C were fed a mixture of dried table scraps and shredded newspaper. The composting plants treating refuse at Altoona, Pa., and refuse-sludge at Leicester, England, were also sampled. Of 652 randomly picked colonies, 87% were identified as Bacillus spp. Other isolates included two genera of unidentified nonsporeforming bacteria (one of gram-negative small rods and the other of gram-variable coccobacilli), the actinomycetes Streptomyces spp. and Thermoactinomyces sp., and the fungus Aspergillus fumigatus. Among the Bacillus isolates, the following, in order of decreasing frequency, were observed: B. circulans complex, B. stearothermophilus, B. coagulans types A and B, B. licheniformis, B. brevis, B. sphaericus, Bacillus spp. types i and ii, and B. subtilis. About 15% of the Bacillus isolates could be assigned to species only by allowing for greater variability in one or more characteristics than has been reported by other authors for their strains. In particular, growth at higher temperatures than previously reported was found for strains of several species. A small number of Bacillus isolates (less than 2%) could not be assigned to any recognized species.  相似文献

A wide range of potentially pathogenic species of Gram-negative bacteria were far more resistant to extreme acidity (pH 2.0–3.5) when cultured at pH 5.0 (habituated to acid) than after pH 7.0 culture. The differences were particularly great for Citrobacter spp., Enterobacter spp., Klebsiella spp. and for Vibrio parahaemolyticus ; substantial habituation was also observed for Proteus mirabilis and Aeromonas formicans but the effect was less marked for Serratia marcescens and Acinetobacter calcoaceticus . Growth at pH 5.0 was substantially poorer than at pH 7.0 for most of the above species and also for Salmonella typhimurium and Salm. enteritidis but phosphate markedly enhanced growth at pH 5.0 for many of these species without affecting growth at pH 7.0.  相似文献

【目的】研究降雨条件对浴场细菌总数和优势菌群组成的影响。【方法】2014年8月强降雨前后采集秦皇岛西浴场3个站位的海水样品,采用荧光显微镜计数法和平板计数法分别对细菌总数和可培养细菌总数进行计数;对群落结构组成进行分析,并对可培养细菌进行鉴定。【结果】雨前3个站位细菌总数和可培养细菌总数平均值分别为5.6×10~9 CFU/L和8.3×10~7 CFU/L,雨后分别为9.2×109 CFU/L和2.1×10~8 CFU/L。在可培养菌群中,变形菌门(Proteobacteria,雨前占80%,雨后占73%)是主要的微生物类群,其次为拟杆菌门(Bacteroides,雨前占12%,雨后占13%)、厚壁菌门(Firmicutes,雨前占7%,雨后占11%)等;肠杆菌属(Enterobacter spp.,21株)、海杆菌属(Marinobacter spp.,13株)、弓形菌属(Arcobacter spp.,13株)、假单胞菌属(Pseudomonas spp.,10株)、芽孢杆菌属(Bacillus spp.,10株)和弧菌属(Vibrio spp.,6株)为雨前可培养细菌优势属,而雨后可培养细菌优势属为肠杆菌属(22株)、海杆菌属(21株)、芽孢杆菌属(14株)、不动杆菌属(Acinetobacter spp.,11株)、假单胞菌属(9株)和弓形菌属(5株)等。【结论】降雨对细菌总数有显著的影响,同时降雨后浴场微生物群落结构发生了改变。  相似文献
