Three hundred and sixty-nine macroalgal and non-algal samples were collected from six coastal sites around the island of Hawai‘i on a biweekly basis over a 1-year period to ascertain (1) the presence of potentially toxigenic benthic dinoflagellates, and (2) substrate and environmental preferences of the dinoflagellates. Twenty-six genera/species of dinoflagellates were encountered including the (potentially) toxigenic species Amphidinium sp., Coolia monotis, Gambierdiscus sp., Ostreopsis ovata, Prorocentrum concavum, Prorocentrum hoffmannianum, Prorocentrum lima, and Prorocentrum mexicanum. Twenty of the species are being reported for the first time as present in Hawaiian coastal waters. There was some evidence of macroalgal host preference (e.g., C. monotis on Tolypiocladia glomerulata), although host morphology preferences was greater (e.g., Gambierdiscus sp., and P. lima on filamentous turfs; O. sp. 1 on sheet-like macroblades). While some dinoflagellate groups were significantly correlated with nutrient concentrations (e.g., total dinoflagellates with nitrate + nitrite and phosphate concentrations), others were not (e.g., Gambierdiscus sp., Prorocentrum emarginatum, P. lima, and Sinophysis microcephalus). The presence of several potentially toxigenic dinoflagellate species in Hawai‘i merits future study on possible impacts of these dinoflagellates on coastal food webs and human health.  相似文献

Species of the benthic dinoflagellate Gambierdiscus produce polyether neurotoxins that caused ciguatera fish/shellfish poisoning in human. The toxins enter marine food webs by foraging of herbivores on the biotic substrates like macroalgae that host the toxic dinoflagellates. Interaction of Gambierdiscus and their macroalgal substrate hosts is believed to shape the tendency of substrate preferences and habitat specialization. This was supported by studies that manifested epiphytic preferences and behaviors in Gambierdiscus species toward different macroalgal hosts. To further examine the supposition, a laboratory-based experimental study was conducted to examine the growth, epiphytic behaviors and host preferences of three Gambierdiscus species towards four macroalgal hosts over a culture period of 40 days. The dinoflagellates Gambierdiscus balechii, G. caribaeus, and a new ribotype, herein designated as Gambierdiscus type 7 were initially identified based on the thecal morphology and molecular characterization. Our results showed that Gambierdiscus species tested in this study exhibited higher growth rates in the presence of macroalgal hosts. Growth responses and attachment behaviors, however, differed among different species and strains of Gambierdiscus over different macroalgal substrate hosts. Cells of Gambierdiscus mostly attached to substrate hosts at the beginning of the experiments but detached at the later time. Localized Gambierdiscus-host interactions, as demonstrated in this study, could help to better inform efforts of sampling and monitoring of this benthic toxic dinoflagellate.  相似文献

Ciguatera fish poisoning is a common form of seafood poisoning caused by toxins (ciguatoxins) that accumulate in demersal (reef) food webs. The precursors of ciguatoxins are produced by dinoflagellates of the genus Gambierdiscus, and enter the food web via herbivory and detritivory. The Gambierdiscus genus was recently revised and new research on the physiology and ecology of the revised species is needed. While it has been demonstrated that Gambierdiscus spp. are predominately epiphytic, the variability in epiphytic behavior among the various Gambierdiscus species is not known. Five Gambierdiscus species isolated from the Greater Caribbean Region were the focus of this study (G. belizeanus, G. caribaeus, G. carolinianus, G. carpenteri, and G. yasumotoi). Cells of Gambierdiscus were grown in wells with algae fragments from eight different macroalgal host genera (Acanthophora, Caulerpa, Dasya, Derbesia, Dictyota, Laurencia, Polysiphonia, and Ulva) where the epiphytic behavior and growth of the different Gambierdiscus species were monitored over 29 days. The results of this experiment demonstrate that epiphytic behavior (growth and attachment) differs among the Gambierdiscus species toward the various macroalgal hosts. Results tended to be specific to Gambierdiscus – host pairings with few commonalities in the way a particular Gambierdiscus species interacted across hosts or how the various Gambierdiscus species responded to a particular host. The Gambierdiscus – host pairings that resulted in the highest growth and attachment combinations were examined in terms of known cellular toxicity and host palatability to determine which pairings could represent the most likely vectors for the transfer of ciguatoxins (or precursors) into the demersal food web. Two pairings, Gambierdiscus belizeanusPolysiphonia and G. belizeanusDictyota, best met these criteria, providing a hypothetical approach to better focus sampling and monitoring efforts on such potential vectors in the benthic environment.  相似文献

Diatoms have been long collected from the Southern Ocean but almost no data exist for epiphytic communities, despite their high ecological significance as an important food source in Antarctic coastal food chains. Here, we present a first growth form analysis of diatoms associated with rhodophyte hosts from Terra Nova Bay, Ross Sea, Antarctica. We performed this study to gather baseline information on the species composition of epiphytic diatom communities, determine the influence of some environmental variables on the diatom distribution patterns, and assess the caveats that must be taken into account in terms of sampling design. Macroalgal material was collected during the Italian Antarctic expeditions between 1990 and 2004. Epiphytic diatoms were studied by means of scanning electron microscopy. In terms of growth forms, there were no significant differences between the diatom communities on the different macroalgal host species. Motile (mainly small-celled Navicula perminuta and other Navicula spp.) and adnate (Cocconeis spp.) diatoms dominated the community throughout the study period. Many of the macroalgal blades examined were also covered by epiphytic animals (calcareous bryozoans, hydroids) over most of their surface, with a significant effect on the associated diatom community structure. Our findings suggest that the bio-physicochemical characteristics of each sampling site affected the epiphytic diatom communities more than the substrate type provided by the macroalgal host or the sampling depth.  相似文献

We studied the influence of site conditions on epiphytic orchids under a subtropical climate in the Kathmandu Valley, Nepal. We analysed 96 systematically distributed grid points situated in Kathmandu Valley across a land-use intensity gradient (national park to urbanised city area). Geographical Information System (GIS) and remote sensing were used for classification of land-use types. We identified 23 species of epiphytic orchids, within 13 genera, from 42 different host tree species. Host preference is obvious for some orchid species (e.g., Dendrobium nobile), with certain tree species (e.g., Schima wallichii, Ficus religiosa) hosting more orchid species than others. The orchid Rhynchostylis retusa was the most common species found on many different host tree species across the land-use intensity gradient. Host species and host bark characteristics (e.g., rugosity, pH and exposure to wind) played a vital role for orchid distribution, with lower abundance in areas of higher impact. Under strong human impact (urban city area), F. religiosa was the dominant host tree, with large individual trees (mean diameter in breast height, dbh?=?1.3?m) providing the habitat for considerable populations of R. retusa individuals. In general, epiphytic orchids were found on larger host trees in urban areas than in areas of lower human impact. We found that some hosts are more likely to harbour orchid species, especially native host species. Older larger trees with rougher bark, low pH, exposed to wind and reduced human impact provided better habitats for orchids. We suggest these characteristics should be considered in urban planning to reduce human impact on the associated orchid epiphytic community.  相似文献

Fucus evanescens is a brown alga of arctic origin that has invaded European coasts. The epiphytic community of F. evanescens in southern Sweden was compared with that of the native Fucus vesiculosus, to examine to what extent an invading seaweed can modify local biodiversity. F. evanescens was much less fouled than F. vesiculosus, supporting both less biomass and fewer species of epiphytes. Multivariate analysis of the most common epiphyte taxa showed that the epiphytic community composition of F. evanescens was not entirely separated from that of F. vesiculosus, but host species contributed significantly to explain the variation in community composition. The biomass of free-living invertebrates was also lower on F. evanescens, although the pattern differed between taxonomic groups. While the biomass of amphipods was lower on F. evanescens, there was no significant difference in biomass of isopods or gastropods between the Fucus species. The good correlation between biomass of epiphytes and free-living animals suggests that the epiphytes play an important role in providing a suitable habitat for many species of free-living epifauna. The study shows that the invasion of F. evanescens affects the environmental conditions for many species associated with the Fucus community but that the direct effect on biodiversity is probably low.  相似文献

Water use patterns of two species of strangler fig, Ficus pertusa and F. trigonata, growing in a Venezuelan palm savanna were contrasted in terms of growth phase (epiphyte and tree) and season (dry and wet). The study was motivated by the question of how C3 hemiepiphytes accommodate the marked change in rooting environment associated with a life history of epiphytic establishment followed by substantial root development in the soil. During the dry season, stomatal opening in epiphytic plants occurred only during the early morning, maximum stomatal conductances were 5 to 10-fold lower, and midday leaf water potentials were 0.5–0.8 MPa higher (less negative) than in conspecific trees. Watering epiphytes of F. pertusa during the dry season led to stomatal conductances comparable to those exhibited by conspecific trees, but midday leaf water potentials were unchanged. During the rainy season, epiphytes had lower stomatal conductances than conspecific trees, but leaf water potentials were similar between the two growth phases. There were no differences in 13C between the two growth phases for leaves produced in either season. Substrate water availability differed between growth phases; tree roots extended down to the permanent water table, while roots of epiphytic plants were restricted to material accumulated behind the persistent leaf bases of their host palm tree, Copernicia tectorum. Epiphytic substrate moisture contents were variable during both seasons, indicating both the availability of some moisture during the dry season and the possibility of intermittent depletion during the rainy season. Epiphytic strangler figs appear to rely on a combination of strong stomatal control, maintenance of high leaf water potentials, and perhaps some degree of stem water storage to cope with the fluctuating water regime of the epiphytic environment.  相似文献

Ciguatera fish poisoning (CFP) is a serious health problem in tropical regions and is caused by the bioaccumulation of lipophilic toxins produced by dinoflagellates in the genus Gambierdiscus. Gambierdiscus species are morphologically similar and are difficult to distinguish from one another even when using scanning electron microscopy. Improved identification and detection methods that are sensitive and rapid are needed to identify toxic species and investigate potential distribution and abundance patterns in relation to incidences of CFP. This study presents the first species‐specific, semi‐quantitative polymerase chain reaction (qPCR) assays that can be used to address these questions. These assays are specific for five Gambierdiscus species and one undescribed ribotype. The assays utilized a SYBR green format and targeted unique sequences found within the SSU, ITS, and the D1/D3 LSU ribosomal domains. Standard curves were constructed using known concentrations of cultured cells and 10‐fold serial dilutions of rDNA PCR amplicons containing the target sequence for each specific assay. Assay sensitivity and accuracy were tested using DNA extracts purified from known concentrations of multiple Gambierdiscus species. The qPCR assays were used to assess Gambierdiscus species diversity and abundance in samples collected from nearshore areas adjacent to Ft. Pierce and Jupiter, Florida USA. The results indicated that the practical limit of detection for each assay was 10 cells per sample. Most interestingly, the qPCR analysis revealed that as many as four species of Gambierdiscus were present in a single macrophyte sample.  相似文献

Abstract The distribution of epiphytic organisms is limited by the availability of, and dispersal to suitable hosts. We examined the distribution of a hemi‐epiphytic strangler fig, Ficus watkinsiana (Moraceae) in Cooloola National Park (Queensland, Australia), in order to determine whether this species exhibits a preference for certain host species and why. We assessed host bark roughness and flakiness, fruit type, and size to explain the observed distribution of F. watkinsiana. We surveyed over 1900 potential host trees of the 30 most common forest canopy species and found that host size measured by diameter at breast height accounted for most variation in fig prevalence (Binary Logistic Regression log‐likelihood = ?588.178, G = 314.494, d.f. = 1, P < 0.005). After controlling for host size, F. watkinsiana prevalence still differed significantly between host species (χ2 = 54.612, d.f. = 24, P < 0.005), a difference that was only partly explained by variation in the bark roughness of host trunks. These results suggest that variation in the rate at which tree species host strangler figs are primarily related to individual tree size – figs may simply be more likely to colonize and thrive upon host species that grow larger.  相似文献

The dinoflagellate Gambierdiscus was exposed to ballast water from a trans-oceanic vessel, and maintained at a variety of temperatures in the dark to determine if viability would be maintained. Logarithmically growing Gambierdiscus inocula were admixed (1:6, vol:vol) with ballast water, maintained in the dark at 22.6 °C, 24.6 °C, 26.8 °C and 29.0 °C and assessed for numerical abundance over six days. Calculated growth rates from the biomass time series showed no indication that ballast water negatively impacted Gambierdiscus viability; accompanying microscopic inspections supported this conclusion. Filtration of large volumes of collected ballast water failed to show the presence of any Gambierdiscus cells contained therein. Recovery and microscopic examination of the experimental inocula after 10 weeks in the dark, failed to show cyst development at any temperature regime. This examination of ballast water showed no evidence of cytotoxicity to Gambierdiscus spp.  相似文献

Although increases in sedimentation have been proposed to interfere with benthic communities in many coastal areas worldwide, few experimental studies have investigated the effect of sedimentation on community composition and assessed species-specific responses. In a field experiment on a rocky shore on the Swedish east coast (northern Baltic Proper) we confirmed the hypotheses that ambient sedimentation influences macroalgal abundance and community composition, and that species-specific responses to sedimentation correlate with reproductive strategies. We followed the establishment and development of macroalgal vegetation on artificial substrates at 8-m and 15-m depth for 4.5 years while manipulating the depositional environment by regularly removing accumulated sediment. Sediment removal significantly favoured macroalgal development and vegetation cover. Responses of macroalgal species to the sediment treatments were clearly species-specific; for example, the ephemeral green algae (Cladophora glomerata and Enteromorpha spp.) were highly tolerant to sedimentation while belt-forming perennial brown algae (Fucus vesiculosus and Sphacelaria arctica) were not. Accordingly, multivariate analyses (redundancy analysis) showed that variance in species abundances were significantly correlated to sediment conditions. The effect of sediment removal was higher at 15-m than at 8-m depth and some species distributions seemed limited in depth by the present sediment load (e.g. F. vesiculosus). Vegetative propagation was common in the study area and many species mainly depended on dispersal by fragmentation. Generally, species with an extended reproductive period, either by long continuous spore release (C. glomerata and Enteromorpha spp.) or vegetative dispersal by fragmentation (e.g. Furcellaria lumbricalis and Polysiphonia fucoides), were most tolerant to sedimentation. This paper demonstrates long-term effects of sediment deposition on the development of a macroalgal community over several growing seasons. The results indicate that variation in sediment loads is an important constraint for species local distributions and abundances, and affects the composition of sublittoral rocky-shore macroalgal communities.  相似文献

In the context of global warming and climate change, ciguatera disease is put forward as an indicator of environmental disturbance. However, to validate this indicator, some unknown parameters such as the delay between environmental perturbation and outbreaks of ciguatera need to be investigated. The main goal of this study was to investigate the temporal link between the growth of Gambierdiscus spp., and one of its influencing factors and the declared cases of ciguatera disease in humans. Algal cell density and seawater temperature (SWT) were recorded monthly from February 1993 to December 2001 on the Atimaono barrier reef of Tahiti Island. Reports of ciguatera cases were obtained from three community health clinics near the study sites. The autoregressive integrated moving average model (ARIMA) shows: (1) SWT were positively associated with Gambierdiscus spp. growth at a lagtime of 13 and 17 months (p < 0.001); (2) Gambierdiscus spp. growth measured at a given time is related to a peak number of cases of ciguatera recorded 3 months after peak densities of this dinoflagellate (p < 0.001). These results allow the construction of a predictive model of the temporal link between ciguatera disease in humans and its etiologic agent: Gambierdiscus spp. This model constructed by using 1993–1999 data, then validated by 2000–2001 data, demonstrates an appreciable ability to predict changes in the incidence of ciguatera disease following algae blooms.  相似文献

Eutrophication of shallow coastal waters often leads to blooms of macroalgae. Grazing by crustaceans, such as amphipods and isopods, can reduce macroalgal biomass accumulation. At the same time, growth of the macroalgae can be stimulated by epiphyte removal. The role of grazing by isopods and amphipods on Ulva spp. biomass development was investigated in the Veerse Meer, a brackish lagoon situated in the southwest Netherlands. Exclusion of grazing in the field did not stimulate Ulva spp. growth. In fact, growth rates were higher in exclosures that allowed grazers to enter. Edibility tests identified the amphipod Gammarus locusta, and the isopods Idotea chelipes and Sphaeroma hookeri as potential grazers on Ulva spp. However, when epiphytic diatoms were present on the Ulva spp. thalli, Gammarus and Sphaeroma grazed on ephiphytes and not on Ulva tissue. Only Idotea continued to graze on Ulva spp. A laboratory growth experiment revealed a positive effect of Gammarus presence on Ulva spp. growth, probably caused by preferential removal of epiphytic diatoms from the Ulva spp. thalli. The growth stimulation by epiphyte removing grazers such as Gammarus may explain the higher growth rates in the presence of grazers observed in the field. When determining the potential role of invertebrate grazers in controlling macroalgal biomass accumulation, it is important to include an assessment of the epiphyte abundance on the macroalgae, as preferential removal of epiphytes may stimulate growth and thus have the opposite effect.  相似文献

Chondracanthus chamissoi is a red alga which is acquiring an increasing economic importance in Chile and becoming a target species for cultivation. Because epiphytism is a major problem in the mariculture of seaweeds, the recruitment of Ulva sp. and Enteromorpha sp. on gametophytic (cystocarpic) and tetrasporophytic thalli of C. chamissoi was tested in vitro on four populations from different locations in Chile. For Ulva sp., the density, cover and length of the recruits varied between the reproductive phases of the host and among the studied populations of C. chamissoi. In most experiments, a larger number of epiphytic thalli was observed on gametophytic plants of C. chamissoi; populations from Calderilla and Lechagua had a higher resistance to epiphytism. For Enteromorpha sp., its ability to grow epiphytically on both phases of the host showed a greater variability, but was inferior to that of Ulva sp. The differential resistance to epiphytism in both reproductive phases and in plant origin indicates the possibility of obtaining selected strains of C. chamissoi with lower susceptibility to epiphytism that could be utilized in planning maricultural programs.  相似文献

Previous studies on the interaction between bacteria and harmful algal bloom species have mostly considered the bacteria in the bulk solution. Here, we document the abundance and mode of attachment of bacteria growing on the cell surface of the domoic acid-producing diatom Pseudo-nitzschia multiseries (Hasle) Hasle in culture, compared with diatoms in field samples. The epiphytic bacteria were examined by scanning electron microscopy to visualize their morphology and mode of attachment. Two P. multiseries cultures were studied: clone CLN-1 and sub-clone CLN-1-NRC; the latter had been maintained in another laboratory for 2 years. Each of these P. multiseries cultures exhibited a clearly different assemblage of epibiotic bacteria, even though both originated from the same parent culture. The bacterial diversity was greater in clone CLN-1 (nine distinct morphotypes seen) than in sub-clone CLN-1-NRC (six morphotypes). The former clone also produced more domoic acid than the latter. There was a succession of bacterial morphotypes as well as an increase in the number of epiphytic bacteria per diatom cell during the progression from exponential to stationary phase. The most diverse and common morphotypes were rod-shaped cells (e.g. a Caulobacter-like bacterium attached by a discoid holdfast). Epibionts showed a preference for attachment at specific regions of the host diatom frustule, e.g. the raphe or cingulum, locations where organic matter may be extruding from the diatom cell. Most diatom cells carried only one to five bacteria, and up to ca. 60% of the intact diatom cells (although intact cells themselves were infrequent) were still free of epibiotic bacteria at the end of the 31-day batch culture experiment. Sequencing of the SSU rRNA gene showed that five of the eight bacterial strains isolated from the P. multiseries cultures were members of the Alphaproteobacteria, three of the Gammaproteobacteria and one of the Bacteroidetes. A morphologically diverse assemblage of epibiotic bacteria was also found on both centric and pennate planktonic diatoms in natural coastal waters. Of the eight morphotypes recorded, all but two were also found in the cultures. Relatively fewer wild diatom cells carried bacteria compared to cells in culture. We hypothesize that the diversity and abundance of epiphytic bacteria may explain some of the variability seen in the production of DA by different P. multiseries clones, and should be considered as another important and controllable variable that influences diatom cell physiology.  相似文献

It has been proposed that seaweed secondary metabolites, e.g. brown algal phlorotannins, may have an ecologically important function as a chemical defence against epiphytes, by acting against colonisation of epiphytic organisms. We tested whether the low epiphytic abundance on the invasive brown seaweed Fucus evanescens, compared to the congeneric F. vesiculosus, is due to a more effective chemical defence against epiphyte colonisation. A field survey of the distribution of the common fouling organism Balanus improvisus (Cirripedia) showed that the abundance was consistently lower on F. evanescens than on F. vesiculosus. However, contrary to expectations, results from experimental studies indicated that F. vesiculosus has a more effective anti-settlement defence than F. evanescens. In settlement experiments with intact fronds of the two Fucus species, both species deterred settlement by barnacle larvae, but settlement was lower on F. vesiculosus both in choice and no-choice experiments. Phlorotannins from F. vesiculosus also had a stronger negative effect on larval settlement and were active at a lower concentration than those from F. evanescens. The results show that Fucus phlorotannins have the potential to inhibit settlement of invertebrate larvae, but that settlement inhibition cannot explain the lower abundance of the barnacle Balanus improvisus on F. evanescens compared to F. vesiculosus. Assessment of barnacle survival in the laboratory and in the field showed that this pattern could instead be attributed to a higher mortality of newly settled barnacles. Observation suggests that the increased mortality was due to detachment of young barnacles from the seaweed surface. This shows that the antifouling mechanism of F. evanescens acts on post-settlement stages of B. improvisus.  相似文献

The epiphytic algae on surfaces of the macrophyte Ranunculus penicillatus (Dumort.) Bab. var. calcareus (R. W. Butcher) C. D. K. Cook and on Cladophora glomerata (L.) Kütz growing in the River Itchen at Otterbourne near Southampton were studied between February 1984 and June 1985. The river at this site has a mean flow rate of 0.33 m s–1, and is about 16 m wide and on average 20 cm deep, with a discharge ranging through the year between 0.34 and 2.46 m3 s–1. The pH of the river varies little around 8.2, with a mean alkalinity of 236 mg HCO inf3 sup1 l–1, because of its origin from chalk springs. Ranunculus grows throughout the year, with peaks of biomass in spring and autumn. It forms a very large surface for attachment of epiphytes, and covers on average 40% of the stream bed. Numbers of epiphyte cells removed from Ranunculus ranged through the year between 52 × 103 and 271 × 103 cells mm–2 stream floor, with maximum numbers in April, and a secondary peak in October. This pattern partly reflects fluctuations in the biomass of Ranunculus; the number of cells per unit area of plant surface showed a broader spring peak and lower fluctuations in other seasons. Diatoms formed 65 to 98% of these epiphyte cells, with chlorophytes reaching their peak (10%) in summer and cyanophytes (25%) in autumn. Estimates of biomass of these epiphytic forms, derived from measurements of chlorophyll c, indicate a range between 30 and 100 g dry weight m–2 of weed bed. Colonisation studies showed that the algae grow and reproduce throughout the year, with a mean generation time of about 5 days, suggesting an annual production of about 3 kg dry weight m–2 of weed bed, which makes epiphytic algae the principal primary producers in the stream. The numbers and biomass of epiphytic algae on Cladophora are considerably less.The species of epiphytic algae found on Ranunculus were generally similar to those growing on Cladophora, and to epilithic algae on pebbles of the stream bed, but different species were dominant on the different substrata. Algal cells in the water column were all derived from benthic habitats, although their relative abundance was very different.  相似文献

Species of the marine benthic dinoflagellate genus Gambierdiscus are the principal cause of Ciguatera fish poisoning. This genus has been recorded from tropical to temperate oceans, although Gambierdiscus species have rarely been found in Chinese waters. Our work revealed the morphological and genetic characteristics of three potentially toxic Gambierdiscus species observed in the temperate to subtropical waters of China. The fine thecal morphology was determined based on light microscopy and scanning electron microscopy analyses, and these species were also characterized by sequencing the D1–D3 and D8–D10 regions of the LSU rDNA. The morphological and genetic data indicated that these three Gambierdiscus species were G. pacificus, G. australes and G. caribaeus. This work provides the first report of these species in Chinese waters, which increases the known species distribution of this genus.  相似文献
