Soluble transforming growth factor-beta (TGF-beta)/bone morphogenetic protein (BMP)-binding proteins are widely distributed in mammalian tissues and control cytokine access to membrane signaling receptors. The serum and bone-resident glycoprotein alpha2-HS-glycoprotein/fetuin (ASHG) binds to TGF-beta/BMP cytokines and blocks TGF-beta1 binding to cell surface receptors. Therefore, we examined bone growth and remodeling phenotypes in ASHG-deficient mice. The skeletal structure of Ahsg(-/-) mice appeared normal at birth, but abnormalities were observed in adult Ahsg(-/-) mice. Maturation of growth plate chondrocytes was impaired, and femurs lengthened more slowly between 3 and 18 months of age in Ahsg(-/-) mice. However, bone formation was increased in Ahsg(-/-) mice as indicated by greater cortical thickness, accelerated trabecular bone remodeling, and increased osteoblast numbers on bone surfaces. The normal age-related increase in cortical thickness and bone mineral density was accelerated in Ahsg(-/-) mice and was associated with increased energy required to fracture. Bone formation in response to implanted BMP cytokine extended further from the implant in Ahsg(-/-) compared with Ahsg(+/+) mice, confirming the interaction between ASHG and TGF-beta/BMP cytokines in vivo. Our results demonstrate that ASHG blocks TGF-beta-dependent signaling in osteoblastic cells, and mice lacking ASHG display growth plate defects, increased bone formation with age, and enhanced cytokine-dependent osteogenesis.  相似文献

A report on the 64th annual American Society of Human Genetics meeting held in San Diego, USA, 18-22 October, 2014.Fueled by rapid technological innovations, the ability of geneticists to assay accurately not only genetic data but also gene expression levels, epigenetic markers, biomarker levels and microbiome composition has expanded the field of human genetics vastly. Alongside this expansion, the annual American Society of Human Genetics (ASHG) conference has grown to draw over 6,500 scientific participants with a broad range of interests. Here, we focus on some of the most notable advances made in the area of population genetics that were presented at the 64th annual ASHG meeting. This year, the sophistication of the analysis methods employed was striking, particularly for drawing inferences from subtle differences in human genetic variation.  相似文献

A subcommittee of The Social Issues Committee of The American Society of Human Genetics (ASHG), aware of the growing interest in the implications of inheritance and adoption, in 1987 began surveying the 50 states' and Washington DC's public adoption agencies regarding this issue, and it completed the survey in 1988. In 1987, two surveys obtained data on each state's legal requirements for obtaining genetic information and on what each public adoption agency collected as genetic history. These results were presented in a poster at the 1987 ASHG annual meeting in San Diego. In 1988, a questionnaire was sent to the same agencies to elicit opinions as to whether adoption agencies should systematically collect genetic information to share with the adoptive family and adoptee, whether legislation should be mandated to collect such information, and whether genetic education programs should be developed and implemented for adoption-agency staff. On the basis of responses to the 1988 questionnaire, it was concluded that there is an interest in developing a uniform set of genetic information which should be part of a child's adoption record and that there is a need for genetic education programs for adoption workers. Responses to the desirability of mandating legislation for this purpose were less consistent.  相似文献

In 1995, the American Society of Human Genetics (ASHG) and American College of Medical Genetics and Genomics (ACMG) jointly published a statement on genetic testing in children and adolescents. In the past 20 years, much has changed in the field of genetics, including the development of powerful new technologies, new data from genetic research on children and adolescents, and substantial clinical experience. This statement represents current opinion by the ASHG on the ethical, legal, and social issues concerning genetic testing in children. These recommendations are relevant to families, clinicians, and investigators. After a brief review of the 1995 statement and major changes in genetic technologies in recent years, this statement offers points to consider on a broad range of test technologies and their applications in clinical medicine and research. In 2020, research entities at the Institut Pasteur (IP) in Paris, as elsewhere around the world, were closed because of the coronavirus disease 2019 (COVID-19) pandemic. However, IP core facilities, laboratories, services, and departments working on severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and priority projects were authorized to continue working both on site and remotely. Given the importance of its role in SARS-CoV-2 genome-sequencing initiatives, the IP Biomics core facility was fully functional during the first (i.e., March–June 2020) and second (i.e., November–December 2020) national lockdowns. We describe here how Biomics successfully implemented an emergency management plan to deal with this health crisis. We highlight the internal deployment of the institutional business continuity plan (BCP) through a series of actions. We also address the impact of the COVID-19 crisis on Biomics staff and collaborators. The added value of quality management and the limitations of risk management systems are discussed. Finally, we suggest that the Biomics infrastructure and the BCP described here could be used for benchmarking purposes, for other next-generation sequencing core facilities wishing to implement/improve their processes, and for future major crisis management.  相似文献

National educational organizations have called upon scientists to become involved in K-12 education reform. From sporadic interaction with students to more sustained partnerships with teachers, the engagement of scientists takes many forms. In this case, scientists from the American Society of Human Genetics (ASHG), the Genetics Society of America (GSA), and the National Society of Genetic Counselors (NSGC) have partnered to organize an essay contest for high school students as part of the activities surrounding National DNA Day. We describe a systematic analysis of 500 of 2443 total essays submitted in response to this contest over 2 years. Our analysis reveals the nature of student misconceptions in genetics, the possible sources of these misconceptions, and potential ways to galvanize genetics education.  相似文献

Despite recent advances in modeling forest–rainfall relationships, the current understanding of changes in observed rainfall patterns resulting from historical deforestation remains limited. To address this knowledge gap, we analyzed how 40 years of deforestation has altered rainfall patterns in South America as well as how current Amazonian forest cover sustains rainfall. First, we develop a spatiotemporal neural network model to simulate rainfall as a function of vegetation and climate inputs in South America; second, we assess the rainfall effects of observed deforestation in South America during the periods 1982–2020 and 2000–2020; third, we assess the potential rainfall changes in the Amazon biome under two deforestation scenarios. We find that, on average, cumulative deforestation in South America from 1982 to 2020 has reduced rainfall over the period 2016–2020 by 18% over deforested areas, and by 9% over non-deforested areas across South America. We also find that more recent deforestation, that is, from 2000 to 2020, has reduced rainfall over the period 2016–2020 by 10% over deforested areas and by 5% over non-deforested areas. Deforestation between 1982 and 2020 has led to a doubling in the area experiencing a minimum dry season of 4 months in the Amazon biome. Similarly, in the Cerrado region, there has been a corresponding doubling in the area with a minimum dry season of 7 months. These changes are compared to a hypothetical scenario where no deforestation occurred. Complete conversion of all Amazon forest land outside protected areas would reduce average annual rainfall in the Amazon by 36% and complete deforestation of all forest cover including protected areas would reduce average annual rainfall in the Amazon by 68%. Our findings emphasize the urgent need for effective conservation measures to safeguard both forest ecosystems and sustainable agricultural practices.  相似文献

In 2007, the American Society of Human Genetics issued recommendations for what the new and largely self-regulating industry offering genetic tests directly to consumers should disclose to potential customers. Websites for every DTC company offering health-related genetic tests as identified by a public policy group were evaluated for compliance with those transparency recommendations. The results showed that only six of the 25 companies studied met even 70% of the standards and that overall, the industry complied with the disclosure standards just 44% of the time. Further, the study revealed that even when companies met the letter of the law, they often failed to disclose to consumers the shortcomings associated with the tests and thus promoted genetic determinism. By failing to meet the spirit of the ASHG transparency recommendations, the DTC genetic testing industry demonstrates disdain toward the ethical principle of informed consent.  相似文献
