Semi-thin plastic sections reveal that the carotid baroreceptor region in the rock hyrax comprising the origin of the internal carotid artery has a preponderantly elastic structure and a thick tunica adventitia. In contrast, the common carotid artery has a musculoelastic structure, whereas the cranial segment of the internal carotid artery (immediately distal to the baroreceptor areas) shows the features of a muscular artery. Electron microscopy discloses the presence of sensory nerve endings within the parts of the tunica adventitia adjoining the preponderantly elastic zone of the internal carotid artery. These nerve endings are characterized by varicose regions containing a large quantity of mitochondria. Bundles of collagen fibers in the tunica adventitia form convolutions or whorls around the nerve terminals and often terminate on the surface of the elastic fibers or into the basement membranes of the neuronal profiles. The large content of elastic tissue in the tunica media of the baroreceptor region may render the vessel wall highly distensible to intraluminal pressure changes. This, in turn, would facilitate the transmission of the stimulus intensity to the sensory nerve terminals located in the tunica adventitia. It is suggested that the stretching of elastic fibers may form the main mechanical event leading to the distortion of the associated nerve terminals. However, a change in the geometrical configuration of the bundles of collagen under the influence of the elastic fibers may provide a better insight into the mechanisms of distortion of the baroreceptors related to and/or in contact with collagen fibers.  相似文献

We examined the fine structure of the baroreceptors and the adrenergic innervation of the guinea-pig carotid sinus. The tunica adventitia contained many nerve bundles whose perineuria enclosed unmyelinated nerve fibers, alone or together with myelinated nerve fibers. Baroreceptors, which lay close to elastic and collagen fibers in the adventitia and media, were surrounded by “terminal” cells with ultrastructural features characteristic of Schwann cells and contained inclusions of various types. Morphologic features of the baroreceptors included densely packed mitochondria, osmiophilic lamellated and homogeneous bodies, clear and granular vesicles, lamellar systems, glycogen granules, neurofilaments, neurotubuli, and vacuolated mitochondria. In animals that had been treated with 6-hydroxydopamine, occasional electrondense endings (or fibers) were observed in the adventitial layer. The baroreceptors in the guinea-pig carotid sinus appear to have most of the morphologic features reported for other species.  相似文献

The topography of the activity of succinate dehydrogenase, cytochrome oxidase, monoaminoxidase and esterase was studied in the arc of the aorta of the monkey, dog, cat, rabbit, rat. It was established that terminal portions of baroreceptors were accumulators of the activity of enzymes of the succinoxidase system connected with the mitochondria of a nervous component of sensory endings and the activity of cholinesterase localized in the zone of terminal branches of the baroreceptors fibres in the underlying specialized tissue. The activity of monoaminoxidase was found in the cytoplasm of Schwann cells of the baroreceptor fibres. No specific esterase was revealed in the baroceptors of the aorta arc of the animals in question. The enzymic organization of the cat's aorta baroreceptors is different from that of the other animals.  相似文献

Summary Two laminae composed of smooth muscles, elastic tissue and collagen have been described in relation with the gill skeleton in Perca fluviatilis. A transverse smoothmuscle lamina joins the base of the cartilage rods of the two opposite hemibranchs. A longitudinal smooth-muscle lamina runs parallel to the afferent branchial artery and joins the cartilage rods from one filament to the other. In both laminae, the formaldehyde-induced fluorescence technique (Falck-Hillarp) reveals a network of nerve fibers displaying a green fluorescence characteristic of catecholamines. At the ultrastructural level, the presence of nerve endings containing clear and granular vesicles, and the degeneration of these endings after 6-hydroxydopamine treatment confirm the aminergic nature and the sympathetic origin of this innervation. Surgical denervation brings evidence that the innervation of both laminae is supplied by the metatrematic branches of the branchial nerves. The role of these smooth-muscle laminae remains speculative.  相似文献

Summary The carotid baroreceptor field of normotensive (NTR) and spontaneously hypertensive rats (SHR) examined in this study extends for about 0.5 mm along the length and about 1/3 to 1/2 of the circumference of the wall of the internal carotid artery opposite to the carotid body. The vascular wall of the baroreceptor field exhibits neither a marked dilation to form a carotid sinus nor histological differences in the intima and media compared to other parts of the carotid artery. Histologically the adventitia of the baroreceptor field is characterized by (1) an increased thickness and by less well developed elastic lamellae in comparison with other parts of the arterial wall, (2) a profuse blood and nerve supply, and (3) a richness of cellular elements. The presumptive baroreceptor terminals are localized in the inner 1/3 of the adventitia and display local enlargements that appear to show preferential association with the cell body or processes of the Schwann cell but not with other components of the adventitia. The enlargements are characterized by an accumulation of very densely packed mitochondria, and glycogen particles. No morphological alterations were noted in the baroreceptor terminals of SHR except for proliferated basal laminae that invest the terminals. Our work does not support the concept that resetting of the baroreceptors is due to degeneration of the terminals.Supported by a grant from the Edward G. Schlieder Educational Foundation. The skin of the pig-tail macaque is basically similar to that of the rhesus monkey and the stump-tail macaque. The epidermis is thin and contains occasional basal melanocytes. The dermis, rich in elastic fibers, is practically free of pigment-containing cells. The upper dermis is highly vascular in the perianal region and sex skin. Cholinesterase-reactive nerve endings are plentiful beneath the friction surfaces of the pes and manus, mucous membranes, and junction of the hairy gluteus and glabrous ischial callosity. Hederiform-like endings are present in the eyelid, pinna, and frontal scalp. Apocrine and eccrine sweat glands occur throughout the hairy skin in a 2–3: 1 ratio. Both types are invested by nerves reactive for acetyl- and butyrylcholinesterase.  相似文献

This paper reports observations on the innervation of gill filaments of the lamprey, Lampetra japonica. Nerve fibers run on each side of the afferent filament artery (AFA nerve) and in the connective tissue compartment along the efferent filament artery (EFA nerve). The AFA nerve supplies vasomotor fibers to the afferent filament artery and arteriovenous anastomoses and special visceral motor fibers to branchial muscle fibers (musculus compressor branchialis circularis). Nerve endings of the vasomotor fibers contain large, cored vesicles (60–180 nm in diameter) with a variable number of small, clear vesicles (30–70 μm in diameter), whereas those of the visceral motor fibers have many small, clear vesicles with few large, cored vesicles. The EFA nerve supplies vasomotor fibers to the efferent filament artery. Their endings, containing mixtures of predominantly large, cored vesicles and small, clear vesicles make close synaptic contacts with reticular cells. The latter in turn are connected with each other or with smooth muscle cells in the wall of the efferent filament artery by nexuses. No nerves are found in the axial plate between the afferent and efferent filament arteries nor in the secondary lamellae of individual gill filaments. No afferent nerve supply to the gill filament has been found.  相似文献

Merkel nerve endings of sinus hairs in the upper lip of the rat were observed after having cut the nervus infraorbitalis. The nerve terminal of the Merkel nerve ending has already been degenerated 24 hours after denervation and was phagocytised by neighbouring keratinocytes. The Merkel cells did not change in structure after having lost their nerve terminal even within 169 days after nerve crush. Their position in the stratum basale of the sinus hairs remained constant; the number, size and position of the osmiophilic granules in the cytoplasm of the Merkel cells did not change. These results may show, that Merkel cells are not modified keratinocytes. The possibilities of their origin are discussed.  相似文献

Summary The stratum radiatum of h 3 and h 4 in the hippocampus of the rahbit, where the mossy fiber endings are distributed, was investigated under the electron microscope. These regions contain a certain number of electron dense presynaptic endings. These are characterized by highly dense synaptic vesicles and mitochondrial matrices. The dense endings are not considered as degenerated. Electron dense silver particles, substituted for zinc, occurred on the synaptic vesicles of these dense terminals as well as the mossy fiber endings after the application of Timm's histochemical method modified for electron microscopy. It is concluded that the dark synaptic endings observed might represent mossy fiber terminals in a special functional phase, or might be the result of structural alteration in the course of tissue preparation. The zinc localized in the synaptic vesicles is thought to be associated with the neurotransmitter present in these endings.  相似文献

Summary With the use of a whole mount silver impregnation technique, sensory nerve endings were located in the connective tissue at the base of the modified pectoral fin ray in the gurnard,Aspitrigla cuculus, and within the perichondrium of the barbel in the goatfish,Mullus surmuletus. The location of these endings and their planar receptory fields in such highly mobile structures, suggests that the sensory endings are proprioceptive in nature and that they are associated in monitoring the positional state of the modified pectoral fin ray and barbel, respectively, during voluntary movement. This investigation addresses itself to the general problem of proprioception in teleost fishes and provides histological evidence for the presence of proprioceptive nerve endings.  相似文献

The effects of clonidine (1.5 mcg/kg of body weight), L-DOPA (4 mg/kg) and propranolol (0.1 mg/kg) on the reflex bradycardia caused by stimulation of the arterial baroreceptors were studied in 46 waking cats in encephale isole preparations. The drugs were administered into the vertebral artery. They increased the gain of the cardiovagal reflex from the arterial baroreceptors by 162%, 162.5%, 150% respectively with regard to the control values. A similar effect of clonidine and propranolol was manifested also in cats with spinal cord destroyed between the segments C1 to Th12. The effects of clonidine and L-DOPA on the cardiovagal component of the reflex were abolished by aceperon (4 mg/kg) injected into the vertebral artery.  相似文献

Summary Smooth feather muscles (mm. pennati) consist of bundles of smooth muscle cells which are attached to the feather follicles by short elastic tendons. In addition, some muscle bundles are interrupted by elastic tendons. The elastic tendon is composed of longitudinally arranged elastic fibers which branch and wavy bundles of collagen fibrils. Smooth muscle cells of the muscle bundles are attached to each other by desmosome-like junctions and by fusion of the basal laminae. The cytoplasm of the muscle cells is characterized by conspicuous thick filaments and abundant thin and intermediate filaments. These are attached to band-like dense patches (dense bands) at the plasma membrane which are particularly broad at the tapering end of the muscle cell. The contact surface between smooth muscle cells and their elastic tendon is considerably increased (i) by deep finger-like invaginations and indentations located at the tapering muscle end, and (ii) by branching of the coarse elastic fibers into slender processes, which are attached to the richly folded surface of the muscle cell endings by peripheral microfibrils. This intimate interlocking closely resembles the myotendinous junctions in skeletal muscle. In addition to fibroblasts and fibrocytes, the myotendinous junction of the young growing chicks contains numerous so-called myofibroblasts, which are suggested to represent smooth muscle cells differentiating into fibroblasts of the developing tendon.Dedicated to Professor Dr. Helmut Leonhardt on the occasion of his 60th birthdaySupported by a grant from the Deutsche Forschungsgemeinschaft (Dr. 91/1)  相似文献

Many receptors,including thermal receptors and mechanical receptors,are only activated by stimuli within a clearly defined range of intensities.Differences in the receptive ranges enable individual receptors and their sensory centers to precisely detect the intensity of the stimulus and changes in intensity.Baroreceptors are the sensory terminals of the baroreflex.It is well understood that an increasing number of baroreceptors are recruited to produce afferent action potentials as the blood pressure increases,indicating that individual baroreceptors have different pressure thresholds.The present study revealed that individual baroreceptors could stop their afferent signals when the blood pressure exceeds a certain level,indicating that individual baroreceptors are sensitive to a specific range of blood pressure.The receptive ranges of individual baroreceptors differ in terms of the total range,the lower threshold,and the upper threshold.Of 85 baroreceptors examined in this study,the upper thresholds for about half were within the physiological blood pressure range.These results indicate that supraphysiological blood pressure is unlikely to be encoded by the recruitment of more baroreceptors.Instead,supraphysiological blood pressure levels might be signaled by an increase in the frequency of action potentials or by other mechanisms.In conclusion,our results indicate that rabbit baroreceptors are activated by blood pressure levels within specific receptive ranges.These findings should encourage further studies to examine the role of population coding of blood pressure by baroreceptors in the baroreflex.  相似文献

Summary Two main populations of smooth muscle cells exist in the arterial media of adult mammals with respect to expression of two intermediate filament proteins: vimentin-positive/desmin-negative cells (V+/D-) and vimentin-positive/desmin-positive ones (V+/D+). However, it is still not understood how this phenotypic diversity is established. Since the proportion and the distribution patterns of the two muscle cell populations depend both on the type of blood vessel and the species examined, the aim of the present study was to determine and to compare their developmental origin in various artery segments from two different species. Using confocal scanning laser microscopy and sections stained by means of immunofluorescence, the distribution patterns of desmin and vimentin were compared in transverse sections of thoracic and abdominal aortas (elastic arteries) and of the femoral artery (muscular artery) of newborn and adult rats (n=12) and rabbits (n=12). The comparison of sections labelled with specific antibodies showed the existence of a subpopulation of smooth muscle cells in the aortas, but not in the femoral artery which expressed desmin in newborns but not in adults. These data suggest that the phenotype of smooth muscle cells in elastic arteries but not in muscular arteries is modulated during development.  相似文献

Summary Previous studies have demonstrated that substance P-(SP) and calcitonin gene-related peptide-like immunoreactivities (CGRP-LI) coexist in sensory nerve fibres in the guinea-pig carotid body and carotid sinus. In the present study the ultrastructure of these nerve fibres was investigated by means of single-and double-labelling immunocytochemistry. In both, carotid body and carotid sinus immunoreactive fibres were unmyelinated axons of small dianeter (0.12–0.56 m). At the subcellular level, SP-and CGRP-LI were colocalized in intra-axonal dense core vesicles, suggesting corelease and simultaneous action of these two compounds. SP/CGRP-LI nerve fibres within the carotid body were mainly found in the interparenchymal connective tissue, but also occurred in relationship to blood vesslesl and nests of glomus cells. Neither in the carotid body not in the carotid sinus, SP/CGRP-LI axons corresponded to the large terminals which are generally considered to represent the main chemoreceptor and baroreceptor endings, respectively. Thus, SP/CGRP-LI fibres either belong to the chemo-and baroreceptors of the C-fibre class or constitute a fibre population not directly involved in conduction of baro-and chemoreflexes.This study was supported by the DFG, grant He 919/6-2  相似文献

The embryonic development of the rat carotid body was studied with electron microscopy. In the 11 mm embryo a cell aggregation consisting of undifferentiated cells and unmyelinated nerve fibers appears on the anterior wall of the third branchial artery. Granule-containing cells appear in the 12 mm embryo and continue to increase in number as the cellular aggregation increases in size and becomes separated from the wall of the third branchial artery. Synapse formation and the appearance of fenestrated capillaries occur almost simultaneously at the 17 mm stage. There are two types of synapses, one with membrane densification and vesicles clustered inside the nerve endings, the other with dense material and vesicles inside the granule-containing cells. At the 20 mm stage the undifferentiated cells send enveloping cytoplasmic processes toward adjacent granule-containing cells and the carotid body anlage displays rudimentary lobules.  相似文献

We recently discovered 1) that intravenous injection of the antidiabetic drug metformin in the rat rapidly reduces arterial pressure elevations maintained by the alpha-adrenoceptor agonist phenylephrine (PE) and 2) that direct administration of metformin to isolated rat tail arterial tissue rings rapidly relaxes PE-induced contractions. To further characterize this potential direct vasodilator action, we examined effects of metformin on contractions induced not only by PE but also by norepinephrine (NE) and by nonadrenergic agonists (5-hydroxytryptamine, 5HT; arginine vasopressin, AVP). Also, because the rat tail artery contains abundant adrenergic nerve endings we conducted these tests not only in arterial rings with nerve endings intact but in rings in which they had been removed by pretreatment with 6-hydroxydopamine. In intact rings, metformin at levels from approximately 0.2 to 20 mmol/L rapidly relaxed half-maximal contractions induced by PE and NE similarly and to a markedly greater degree than contractions induced by 5-HT (p<0.05). Metformin did not relax AVP-induced contractions. In addition, removal of adrenergic nerve endings facilitated metformin's relaxant effects (p<0.05). Thus, the acute vasodilator action of metformin appears 1) to be selectively more powerful on arterial smooth muscle contractions induced adrenergically versus nonadrenergically and 2) to be buffered by a possible metformin-induced release of endogenous NE from adrenergic nerve endings. Such results were not seen during relaxation produced by either the calcium channel inhibitor nifedipine or the nitrovasodilator nitroprusside suggesting that metformin's effects are mediated by other mechanisms.  相似文献

Tje paper describes the ultrastructure of axons in the endings of various types from the corium in the glans penis in green monkey. In the Meissner's endings the axons are mostly completely enveloped in the plasma of Schwann cells. They contain numerous mitochondria which are partially vacuolated or are quite converted into vacuoles. Next, there are pseudomyelinated figures, light vesicles and further organelles. In the papillar simple glomerular endings with accumulation of Schwann cells there are axons irregular in shape, eccentrically placed in the plasma of Schwann cell, rounded smaller axons either completely or partially surrounded by the plasma of Schwann cell and finally axons with a concentric system of lamellae up to four in number, In the complicated glomerular endings the axons vary in appearance and are enveloped in one to five lamellae of Schwann cells, which is typical of those formations. About some of these systems there is a sign of a capsule formed by an elongated lamella probably of the perineurium. When the axons are not enveloped in the plasma of Schwann cell, they are covered by the basement membrane. In close neighbourhood of the epidermis so-called free endings forming groups were found. The plasma of Schwann cell covers them either partially or completely or it again forms around them a lamellar system amounting up to four layers. It is noticeable that these axons are very poor in organelles. A comparison of the simple sensory corpuscles in the nose skin in hedgehog, the funtional properties of Meissner's endings and the simple corpuscles results in the view that the complexes having a larger amount of lamellae correspond to an extent to the simple sensory corpuscles ant that the Meissner's endings and the complicated glomerules are probably a morphological and functional equivalent of simple sensory corpuscles in the non-primate mammals and that the gloverular endings may also be the first (developmental) stage of the simple sensory corpuscles.  相似文献

The hypothesis tested was that there are significant transient changes in the cardiovascular variables after rapid onset and release of mild lower body negative pressure (LBNP, -20 mmHg), even in experimental situations where there is no detectable change in steady-state values. Twelve subjects participated in the study. Heart rate, stroke volume (SV), cardiac output, mean arterial pressure (MAP), total peripheral resistance (TPR), acral and nonacral skin blood flow, and blood flow velocity in the brachial artery were continuously recorded during the pre-LBNP period (0-120 s), during LBNP (120-420 s), and during the post-LBNP period (420-600 s). The main finding was that MAP is transiently but strongly affected by rapid changes in LBNP as small as -20 mmHg. There was also a characteristic asymmetry in cardiovascular responses to the onset and release of LBNP, particularly in the responses in SV. The transient changes in MAP indicate that the neural responses that affect TPR are not fast enough to compensate for the rapid changes in LBNP. In this case, the arterial baroreceptors will be activated as well as the low-pressure baroreceptors that sense central venous pressure. This must be taken into consideration in future discussions of the results of LBNP protocols.  相似文献

Light- and electron-microscopic observations of the chemosensory areas of the arteries of the tortoise (Testudo hermanni) reveal that clusters of nonmuscular cells are found in the adventitial layer of restricted regions of the carotid artery, aortic arch, and pulmonary artery. In these clusters, three types of cells are complexly interwoven: the G-cell closely resembles the glomus cell, which has been found in the arterial chemoreceptor area of several animal species; the LG-cell has very large electron-dense granules; the third type is a G- and LG-cell supporting cell. Membrane specializations are often observed at apposing membranes between G-cells. Two kinds of nerve endings synapse with G-cells, one with numerous clear synaptic vesicles, the other without vesicles. Some G-cells are in membrane-to-membrane contact with smooth-muscle cells (g-s connection), and here a membrane thickening is visible. Nerve terminals with numerous synaptic vesicles synapse with the LG-cells. The G-cell in the carotid artery, the aorta, and the pulmonary artery is a chemoreceptor element ultrastructurally the same as the glomus cell in the arterial chemoreceptor area of various vertebrate species.  相似文献
