药用昆虫蜣螂对灵芝多糖生物合成的影响   总被引:4,自引:0,他引:4  
采用液体深层发酵方式,研究了几种药用昆虫对灵芝多糖生物合成的影响。结果表明,药用昆虫蜣螂在添加量为5g/L时能显著促进灵芝胞内多糖(IPS)和胞外多糖(EPS)的形成(P<0.05)。胞内多糖和胞外多糖的产量分别由对照的(1.93±0.09)g/L和(520.3±20.2)mg/L提高到(2.41±0.12)g/L和(608.9±20.2)mg/L。灵芝胞内多糖和胞外多糖在DEAE纤维素柱上都可分离得到5种主要组分,其中IPS-1和EPS-1分别为2类多糖的主要组分。进一步用凝胶柱分离显示,IPS-1由3个单个的组分组成,EPS-1由2个单个的组分组成。添加蜣螂发酵后,灵芝胞内多糖和胞外多糖中没有出现新的组分,且各组分的相对含量也没有显著变化(P>0.05),提示添加蜣螂发酵后,灵芝胞内多糖和胞外多糖主要组分的合成途径并未改变。  相似文献

将11种中药水提物分别加入孤苓核生柄孔菌液体发酵中,以生物量、胞外多糖和三萜类物质为指标,从中选出最佳中药、最佳中药浓度以及最佳中药浓度组合。结果表明,对孤苓核生柄孔菌生长与发酵产物影响最大的中药是何首乌和菊花2种。菊花对三萜类物质的影响较大,何首乌则对孤苓核生柄孔菌生长和发酵胞外多糖影响较大。孤苓核生柄孔菌发酵生物量达到最大(23.97g/L)的条件为添加何首乌水提物浓度15g/L;产胞外多糖的最佳(4.15g/L)条件为添加何首乌水提物浓度5g/L;总三萜类物质产量达到最高值(397.99mg/L)的条件为组合中药水提物:何首乌和菊花水提物浓度均为25g/L。  相似文献

采用液体发酵蝉拟青霉,对蝉拟青霉的发酵条件进行优化,以提高蝉拟青霉胞外多糖产量及生物量。摇瓶发酵条件下,在单因素基础上设计正交实验确定各因素的最佳组合。优化后得最佳发酵培养基:蔗糖8%,牛肉膏0.75%,酵母膏0.125%,MgSO_4·7H_2O 0.3%,KH_2PO_4 0.2%,麸皮0.5%。该条件下胞外多糖产量为5.96 g/L,生物量为42 g/L,较优化前提高了1倍。采用发酵罐进行扩大培养,对分批发酵时的初糖浓度进行了优化,并分析了补料分批发酵对发酵过程的影响。发酵罐培养时最适初糖浓度为5%,此时生物量最高为38 g/L,多糖含量最高为5.5 g/L;采用补料分批发酵时,多糖产量最高为5.89 g/L,生物量最高为40 g/L,效果优于分批发酵。  相似文献

对10株灵芝菌株发酵菌丝体生物量、胞内多糖含量、胞外粗多糖得率及其多糖含量和单糖组成特征进行了系统分析。结果表明,随着培养时间的延长,10株菌株的生物量均有不同程度增加,但不同菌株胞内多糖含量和胞外粗多糖得率变化趋势有所不同。在培养至7d时,G0023和G0160菌株胞外粗多糖得率均较高,分别为3.02g/L和3.14g/L,其多糖含量分别达到了84.11%和91.03%,可作为发酵高产胞外多糖的优良菌株。分级醇沉胞外多糖分析结果表明,各菌株胞外液20%乙醇沉淀所得20E组分的得率和多糖含量均高于50%乙醇沉淀所得的50E组分,说明胞外液中主要以大分子量多糖为主。20E主要由葡萄糖组成,含有少量木糖和甘露糖;50E主要由葡萄糖和甘露糖组成,含有少量木糖和半乳糖。胞外液表观粘度随剪切速率变化曲线分析结果显示,各菌株胞外液均呈现剪切变稀非牛顿流体特性,其表观粘度大小与菌株对应20E组分的得率及多糖含量呈正相关。  相似文献

为筛选促进灰树花Grifola frondosa深层发酵的中药提取物,以期提高灰树花发酵产胞外多糖的产量,在灰树花液体深层发酵体系中添加12种中药(白花蛇舌草、金银花、淡竹叶、柚子皮、银杏叶、何首乌、薏苡仁、菊花、甘草、山药、枸杞、天麻)水提物和醇提物,以生物量和胞外多糖为指标,从中筛选出2种最适中药,并通过在发酵体系中单一添加和组合添加2种最适中药醇提物,确定2种中药的最适浓度和最适组合浓度。试验结果表明:对灰树花生物量与胞外多糖产量影响最大的中药是薏苡仁和天麻;添加薏苡仁醇提物质量浓度8 g/L时,灰树花生物量达到最大值,为14.43 g/L;发酵产胞外多糖的最佳条件为组合中药醇提物,即薏苡仁和天麻醇提物质量浓度均为6 g/L时,胞外多糖产量达3.61 g/L。试验结果为提高灰树花液体深层发酵的生物量和代谢产物合成量提供了新思路和新方法。  相似文献

以菌丝生物量及胞外多糖(exopolysaccharides,EPS)含量为指标对大球盖菇产胞外多糖液体培养基组成和发酵条件进行了优化。结果表明,最适碳源是麦芽糖,最适氮源是酵母膏,正交试验确定最佳培养基组成为马铃薯150 g/L,麦芽糖20 g/L,酵母膏1 g/L,KH2PO41 g/L,MgSO4.7H2O 2.5 g/L。最佳发酵条件为28℃,摇床转速160 r/min,起始pH值6.5,装液量100 mL/250 mL、接种量10%,发酵时间6 d。在此条件下,大球盖菇菌丝生物量及EPS含量分别比对照增加了31.8%和51.6%。  相似文献

本文报道了液体培养灵芝Gl8801菌林产生胞外多糖的最佳发酵条件和胞外多搪的化学组成。适宜的液体培养基(g/L):饴糖40,花生饼粉30,KH_2PO_41.5,(NH_4)_2SO_41.5,MgSO_4·7H_2O_(0.75),CaCO_32,最适起始pH5.5。300ml三角瓶装培养基60ml,发酵温度28℃。摇瓶(165rpm)培养7~8天,发酵液纯多糖含量可达880mg/L。灵芝胞外多糖分为水溶性多糖和水不溶性多糖两种。水溶性多糖的单糖组份为半乳糖、葡萄糖、阿拉伯糖和木糖,所含单糖重量比为6:4:4:1。不溶性多糖为葡聚糖。灵芝胞外多糖的糖环构型为吡喃糖,末端糖基构型,半乳糖为α型,葡萄糖为β型。  相似文献

研究了云芝、灵芝、柱状田头菇等3种食药用真菌胞外多糖及复合多糖对黑腹果蝇寿命的影响.分别将3种食药用真菌胞外多糖及复合多糖以1、5g/L的剂量加入果蝇培养基中,观察对黑腹果蝇寿命的影响.结果表明,3种食药用真菌的胞外多糖及复合多糖对果蝇寿命均有不同程度的延长,云芝5g/L、柱状田头菇5g/L、云芝2.5g/L+柱状田头菇2.5g/L、灵芝2.5g/L+柱状田头菇2.5g/L对果蝇平均寿命延长率分别达到了36.38%、46.60%、36.69%、47.08%.  相似文献

通过摇瓶发酵实验研究了培养温度、光照强度等培养条件对发菜细胞悬浮培养生物量和代谢产物发菜多糖累积的影响,通过均匀设计试验对培养条件进行了优化。结果表明:在培养温度24℃、培养基初始pH8.0、光照强度60μmol/(m2.s)、转速150r/min的条件下培养20d,发菜细胞生物量(细胞质量浓度)达到1.34g/L,胞外多糖产量达到208.32mg/L;与优化前相比,发菜细胞生物量和胞外多糖产量分别提高27.3%、111.17%。  相似文献

研究了几种昆虫的水提物和醇提物对灵芝深层发酵生产菌体和多糖的影响。结果表明,松毛虫水提物在剂量为30mg/L时,能促进灵芝菌体的生长,生物量从15.23±0.53g/L提高到16.26±0.67g/L。其它昆虫提取物对灵芝菌体的生长无显著影响,其中斑蝥提取物表现出明显的抑制作用。对灵芝多糖产量的影响试验表明,蜣螂水提物在剂量为50mg/L时,能显著提高灵芝胞内多糖的产量,产值从1.93±0.09g/L提高到2.34±0.13g/L;地鳖虫醇提物在剂量为55mg/L,以及松毛虫醇提物在剂量为35mg/L时,对胞内多糖的生产也具有促进作用。此外,蜣螂醇提物在剂量为20mg/L时,对胞外多糖的生产具有促进作用,产量从520.3±20.2g/L提高到589.5±24.1mg/L。结果提示药用昆虫中含有促进灵芝多糖生物合成的功能因子。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. Rasmussen’s encephalitis (RE) is a rare pediatric neurological disorder, and the exact etiology is not clear. Viral infection may be involved in the pathogenesis of RE, but conflicting results have reported. In this study, we evaluated the expression of both Epstein-Barr virus (EBV) and human herpes virus (HHV) 6 antigens in brain sections from 30 patients with RE and 16 control individuals by immunohistochemistry. In the RE group, EBV and HHV6 antigens were detected in 56.7% (17/30) and 50% (15/30) of individuals, respectively. In contrast, no detectable EBV and HHV6 antigen expression was found in brain tissues of the control group. The co-expression of EBV and HHV6 was detected in 20.0% (6/30) of individuals. In particular, a 4-year-old boy had a typical clinical course, including a medical history of viral encephalitis, intractable epilepsy, and hemispheric atrophy. The co-expression of EBV and HHV6 was detected in neurons and astrocytes in the brain tissue, accompanied by a high frequency of CD8+ T cells. Our results suggest that EBV and HHV6 infection and the activation of CD8+ T cells are involved in the pathogenesis of RE.  相似文献

正Dear Editor,Chikungunya virus (CHIKV), an arbovirus in the family of Togaviridae, genus Alphavirus, is transmitted by the A.aegyptii or A. albopictus mosquito, and causes disease in humans characterized by fever, rash, and arthralgia (Silva and Dermody 2017; Suhrbier 2019). It was first reported in 1953 in Tanzania, and caused only a few outbreaks and sporadic cases in Africa and Asia in last century. However, in the epidemic in 2004, CHIKV acquired mutations that conferred enhanced transmission by the A. albopictus mosquito(Schuffenecker et al. 2006). Since then, it has successively caused outbreaks in Africa, the Indian Ocean, South East Asia, the South America, and Europe (Zeller et al. 2016).  相似文献

In conclusion, the novel visual RT-LAMP assay is a simple, rapid, and sensitive approach for detection of SARS-CoV-2, and it is ready for application in primary care and community hospitals or health care centers, and even patients' own houses in response to the current SARS-CoV-2 epidemic because the assay does not require sophisticated equipment and skilled personnel. Furthermore, it is also ready to be used in fields for screening samples from wild animals and environments to facilitate the identification of potential intermediate hosts that mediate the cross-species transmission of SARS-CoV-2 from bats to humans.  相似文献
