Ostrinia nubilalis is (Lepidoptera: Crambidae) a severe pest ofcorn in the major corn growing areas of theUnited States. The efficacy of a Beauveria bassiana application, for season-long suppression of O. nubilalis was evaluated in 1996 and 1997 at locationsacross Iowa. Beauveria bassiana,Mycotech 726 (Mycotech Corporation, Butte, MT)formulated on corn grit granules (14–20 mesh)at 2.2 × 109 conidia/g and applied with ahand-held applicator at the rate of 0.4 g/plant(8.8 × 108 conidia/plant). Applicationswere made when plants were in the V7 or R1growth stage. The length of larval tunneling,percentage of plants not infested with O.nubilalis, percentage of plants with anendophyte, and yield from treated and controlplots were determined. Whorl-stage applicationof B. bassiana in 1996 resulted in asignificant reduction in centimeters oftunneling (46–55%) and the percentageplants not infested by O. nubilalis. In1997, B. bassiana caused significantreductions in larval tunneling at all locations(20–53%); however, a significant increase inthe percentage of plants not infested with O. nubilalis occurred at only one location. Treatment of plants with B. bassiana in1997 did not significantly increase thepercentage of plants with an endophyte;however, the trend, with the exception of onesite, was for a greater percentage ofendophytic plants in treated versus untreatedplots. A whorl-stage application of a granularformulation of B. bassiana was mostefficacious in reducing O. nubilalis larval damage.  相似文献

Intraplant epizootics of entomopathogens,Beauveria bassiana (Balsamo) Vuillemin, andNosema pyrausta (Paillot) were studied in a corn,Zea mays L., agroecosystem. Egg masses of the European corn borer,Ostrinia nubilalis (Hübner), infected withN. pyrausta were placed on midwhorl-stage corn plants. Conidia ofB. bassiana were applied in an aqueous suspension. Frass from the initial insects remaining within the plants was contaminated with sufficientN. pyrausta spores to infest 80% of the filial generation in each year of a 2-yr study. Viability ofN. pyrausta within the frass was monitored throughout the winter. Potential for impact of this inoculum on the filial generation is discussed. Conidia from cadavers of the European corn borer that were killed by the initial inoculum ofB. bassiana and/or conidia from the initial inoculum ofB. bassiana significantly reduced tunneling by the filial generation of the European corn borer. However, neither the concentration ofB. bassiana nor the age of the larvae exposed toB. bassiana, had any significant (P<.05) effect on tunneling by the 2nd-generation larvae. This paper reports the results of research only. Oviposition response of the polyphagous European corn borer,Ostrinia nubilalis (Hübner) (Lepidoptera: Pyralidae), to chemical constituents in host plants was investigated in the laboratory using two-choice bioassays. Foliar extracts of corn (Zea mays L.), pepper (Capsicum annuum L.) and potato (Solanum tuberosum L.) were prepared using the solvents pentane, acetone and methanol. In all three host plants, chemicals soluble in pentane stimulated oviposition. In potato, chemicals extractable in acetone also elicited a positive oviposition response. When presented with a choice between pentane extracts of corn and pepper, females preferred corn. No preferences were exhibited between pentane extracts of corn and potato or pepper and potato. Pentane extracts of corn husks, tassels, silk, and corn leaves from plants at early whorl and tassel (pre-pollen shed) stages of development also stimulated oviposition. Similar extracts from plants at 2-leaf and blister (when kernels resemble blisters) stages were not stimulatory. This indicates that plant phenology affects chemically mediated oviposition response in European corn borer. The potential use of plant chemicals for management ofO. nubilalis in the field is suggested.  相似文献

The corn rootworm complex is the most damaging insect pest of corn (Zea mays L.). This study was conducted to determine the efficacy of whorl and pollen-shed stage applications of a granular formulation of Beauveria bassiana (Balsamo) Vuillemin for control of adult western corn rootworm (Diabrotica virgifera virgifera Le Conte). The effect of application time (whorl-stage, pollen-shed) and plant surface exposed (leaves and leaf collars; silks; leaves, leaf collars, and silks) on level of beetle fungal infection were investigated. In addition, the number of colony forming units of B. bassiana in the corn leaf collar area was quantified. In the three years (1998–2000) of the study, application of B. bassiana at whorl-stage did not significantly increase beetle fungal infection. Beauveria bassiana applied to plants at pollen-shed in 1998 resulted in a significant increase in beetle infection with 51% of beetles from treated plants infected and 6.0% from control plants. Similar applications at pollen-shed in 1999 and 2000 resulted in very low infection levels. Beauveria bassiana application at pollen-shed stage significantly increased the number of colony forming units per leaf collar during all years of the study. Beetle infection with B. bassiana did not differ consistently among plant surface to which beetles were exposed for either application. Increased fungal load in leaf collars was not correlated with increased levels of adult infection. Increased rates of B. bassiana and application when beetles are present on the plants are likely needed to significantly increase infection rates.  相似文献

利用生物种间互做关系抑制农业害虫的暴发是生物防治的重要手段。为探讨二种交配型内共生球孢白僵菌与玉米之间的互惠关系及其形成的共生体在亚洲玉米螟控制中的生态效应,以玉米为宿主植物,以球孢白僵菌孢子悬浮液进行灌根,在温室内构建了二种交配型(MAT1-1-1型,B5;MAT1-2-1型,B2)球孢白僵菌-玉米共生体,并研究了共生体对玉米的生长、对亚洲玉米螟的产卵选择和幼虫发育及其对球孢白僵菌生物学特性的影响。结果显示:通过叶片离体培养、ITS基因和交配型基因MAT检测,均能检测到白僵菌的内生定殖;MAT1-2-1型B2菌株定殖检出率高,MAT1-1-1型B5菌株在混合型接种中定殖有优势。回收后的球孢白僵菌菌落直径和毒力无显著性变化,但其产孢量都显著提高其中回收B5处理组来源菌株的产孢量提高最显著。接种过球孢白僵菌的玉米植株地上部生长速度、生物量和地下根系生物量均优于对照组,其中根系干重明显增加,而地上植株干重也相对增加。MAT1-1-1型菌株B5对共生体玉米植株地上高度促生长贡献明显;MAT1-2-1型菌株B2对共生体玉米植株地下干重增加贡献明显。总体上球孢白僵菌内生定殖对玉米地下根系生物量影响大于对地上植株生物量的影响。在产卵选择性试验中,各处理组亚洲玉米螟的产卵量显著少于对照组。共生体对亚洲玉米螟产卵具有明显的趋避作用,MAT1-2-1型菌株B2对产卵的趋避作用明显,而MAT1-2-1型菌株B5的趋避作用较弱。在人工接种幼虫的试验中,处理组回收的亚洲玉米螟幼虫存活率均显著低于对照组,其中,B5组回收幼虫的存活率最低,仅为38.33%;处理组的化蛹率与对照组差异不显著,但B5组的回收幼虫化蛹率显著低于B2组和对照组,仅为34.77%,这说明MAT1-1-1型B5菌株对玉米螟幼虫发育抑制最明显。上述结果表明,不同交配型球孢白僵菌内生定殖效率有差异,在经过内生定殖后在产孢量方面有显著性提高,两个交配型菌株在联合应用时具有协同增效作用;两个交配型菌株均能够通过内生定殖与玉米形成共生体并促进玉米植株的生长,这显示球孢白僵菌和玉米之间已经建立具有互惠关系的共生体。这种共生体通过趋避亚洲玉米螟产卵、抑制幼虫存活和降低化蛹率等方面的潜力虽然不一样,但都有助于对亚洲玉米螟的可持续生态防治,也证明了共生体的建成有效提高了玉米的生态适应性,为利用球孢白僵菌内共生性实施亚洲玉米螟防控提供了新思路。  相似文献

We evaluated the egg parasitoid Trichogramma ostriniae (Hymenoptera: Trichogrammatidae) to control European corn borer [Lepidoptera: Crambidae: Ostrinia nubilalis (Hübner)] in field corn in 2001 and 2002. Inoculative releases of 75,000 T. ostriniae/ha occurred in New York and Virginia in 5–10 cornfields per state when corn was at mid-whorl. Incidence of egg mass parasitism, number of stalk tunnels, incidence of ear damage, and whole-plant yield were evaluated. Parasitism of European corn borer egg masses ranged from 0 to 75% in release plots and was greater in release plots than in control plots. Individual comparisons between paired release and control plots showed no reductions in either stalk or ear damage. However, when data were combined across both years and fields, stalk and ear damage were significantly reduced in New York. In Virginia, no significant differences were detected using data obtained from one year. There were no differences in yield between release and control plots. Low densities of European corn borer, drought conditions in 1 year, and a larger plant canopy in field corn are possible reasons why T. ostriniae releases provided less control than has been observed in previous trials in sweet corn. Additional research focused on improved timing and frequency and number of releases is warranted.  相似文献

The efficiency ofTrichogramma brassicae inundative releases in biological control of the European corn borerOstrinia nubilalis was analyzed in seven plots of 504 plants, each situated in a corn field naturally infested by the European corn borer. Different strategies ofTrichogramma releases were defined on the seven plots. Light and electron microscopy were used to describe the mode of penetration by the entomopathogenic fungus Beauveria bassiana (Balsamo) Vuillemin into corn, Zea mays L. After inoculation with a foliar spray of conidia, germinating hyphae grew randomly across the leaf surface. Often a germ tube formed from a conidium and elongated only a short distance before terminating its growth. Not all developing hyphae on the leaf surface penetrated the cuticle. However, when penetration did occur, the penetration site(s) was randomly located, indicating that B. bassiana does not require specific topographic signals at an appropriate entry site as do some phytopathogenic fungi. Long hyphal structures were observed to follow the leaf apoplast in any direction from the point of penetration. A few hyphae were observed within xylem elements. Because vascular bundles are interconnected throughout the corn plant, this may explain how B. bassiana travels within the plant and ultimately provides overall insecticidal protection. Virulency bioassays demonstrate that B. bassiana does not lose virulence toward the European corn borer, Ostrinia nubilalis (Hübner), once it colonizes corn. This endophytic relationship between an entomopathogenic fungus and a plant suggests possibilities for biological control, including the use of indigenous fungal inocula as insecticides.  相似文献

Three isolates of Beauveria bassiana were evaluated under laboratory conditions for pathogenicity against the European corn borer, Ostrinia nubilalis. Dose-mortality regressions and nonparametric statistical analyses revealed significant differences in pathogenicity between isolates and across O. nubilalis instars. First instars were found to be the most susceptible of the five larval stages. Little difference was noted for instars 2, 3, and 5 within any of the three B. bassiana isolates. Fourth instar larvae were found to be the most tolerant in all cases.  相似文献

We evaluated how manyTrichogramma nubilale should be released at a single location to controlOstrinia nubilalis in sweet corn. Six 8.6×16 m plots received 18.4 to 2 090 ΦΦT. nubilale/SAI when plants were in the mid to late whorl stage, where SAI, surface area index, is the plant surface area/m2. To evaluate the potential control by our releases, we exposed laboratory-rearedO. nubilalis egg masses to the released parasitoids at 4 times after the release. When an egg mass was parasitized byT. nubilale, 75.7% of the eggs in the egg mass were parasitized. We developed an equation to estimate the percent of egg masses that a single female was expected to parasitize in a day (efficiency of parasitism) and female disappearance (death and dispersal) rates, if both were constant during our experiment. The exponential disappearance rate was −0.52±0.03 day−1, which implied that 40% of the remaining ΦΦ disappeared per day. The efficiency of parasitism was 0.050% parasitism/Φ/SAI/day, which implied that at least 351,000 ΦΦ/ha would be needed to achieve 90% parasitism. Clearly, forT. nubilale to be a successful biological control agent, efficiency of parasitism must be increased and disappearance rates must be reduced.   相似文献

In order to assess the effects of high temperatures prevailing during the releases ofTrichogramma brassicae Bezdenko parasitoid used against the European corn borerOstrinia nubilalis Hübner, we exposed wasps in the white pupa stage (WP) to single or double shocks (at 35 or 44°C for 6 h) in the laboratory. The locomotor activity of adults in the presence ofO. nubilalis egg masses is greater than in pure air. Single or double shocks at 35°C have little effect on the females. On the contrary, wasps exposed to single or double shocks at 44°C had a lower motility than control wasps.  相似文献

Nosema pyrausta (Paillot) (Microsporida: Nosematidae) is an obligatory intracellular parasite of the European corn borer, Ostrinia nubilalis (Hübner) (Lepidoptera: Crambidae). This pathogen is maintained in natural populations of O. nubilalis by both horizontal and vertical transmission. The impact of N. pyrausta on fecundity of adults and survival of larvae has been well documented in laboratory and field research. In an extensive study covering a 6 year period at one site, we described the effect of N. pyrausta within O. nubilalis populations in a continuous corn following corn ecosystem. We documented the presence of the pathogen through all life stages of O. nubilalis (egg, larva, pupa, adult), by collecting throughout the crop season and examining each insect stage in the laboratory for the frequency of infection with N. pyrausta. The percentage of infection with N. pyrausta and magnitude of the O. nubilalis population fluctuated throughout generation 1 and generation 2. Both horizontal and vertical transmission played a role in maintaining N. pyrausta in the population in both generations. There were strong correlations between percentage adults with N. pyrausta and percentage larvae with N. pyrausta, and between percentage eggs with N. pyrausta and percentage larvae with N. pyrausta. There was a weak correlation between percentage adults with N. pyrausta and percentage eggs with N. pyrausta. The percentage of insects infected with N. pyrausta was always lowest in the egg.  相似文献

The specialist parasitoidMacrocentrus grandii Goidanich (Hymenoptera: Braconidae) appears to parasitize its polyphagous host, European corn borer (Ostrinia nubilalis (Hübner)) (Lepidoptera: Pyralidae), in only certain habitats. To determine whether it differed in its olfactory response to host-habitat odours, volatiles from four plants were isolated using Tenax. Wind tunnel bioassays of the extracts revealed that, besides corn which was tested in an earlier study, olfactory stimuli for attraction ofM. grandii females were present in potato and snap bean but not in pepper or soybean. To further characterize the response to pepper and soybean, these extracts were bioassayed in combination with an attractive extract. The results indicated that pepper volatiles evoked a neutral response inM. grandii while response to soybean volatiles appeared to be neutral or slightly negative. The innate response to soybean volatiles was altered to one of attraction after oviposition experience on soybean. Seven days after oviposition, experienced females continued to respond positively to soybean volatiles. Components of soybean volatiles responsible for the change in flight behaviour resulting from oviposition experience were eluted by nonpolar and slightly polar solvents. These results support the idea that plant odour may be a factor determining the range of plants on whichM. grandii parasitizes its host. The study indicates the occurrence of associative learning of plant-related volatiles during oviposition inM. grandii, and suggests the involvement of diverse plant compounds in the learning process.  相似文献

Experiments were done to test if Beauveria bassiana can become an endophyte in sorghum and confer protection from stem borer. Four-week-old sorghum seedlings were treated with B. bassiana. The plants were examined for endophytic presence of B. bassiana, 30 and 60 days after treatment. Stem cultures from treated plants showed growth of B. bassiana. PCR amplification using fungal specific primers for a conserved region of β tubulin gene yielded identical 360 bp products from both B. bassiana and treated sorghum plants. In a subsequent experiment, B. bassiana treated and untreated (control) sorghum plants were artificially infested with stem borer (Chilo partellus) larvae 15 days post treatment and the extent of damage was compared. About 40% of the control plants developed dead heart while no plant in the B. bassiana treated plot did. In the surviving control plants, stem tunneling by shoot borer was significantly higher compared to B. bassiana treated sorghum plants.  相似文献

A conidial suspension of the entomopathogenic fungus Beauveria bassiana (Balsamo) Vuillemin, was injected into corn plants at anthesis to suppress populations of the European corn borer Ostrinia nubilalis (Hübner) (Lepidoptera: Pyralidae). The fungus colonized the plants and moved, primarily upward, within the pith, possibly along with plant photosyn‐thates. Beauveria bassiana injected into the corn plants significantly reduced the amount of European corn borer tunneling. Colonization of corn plants by B. bassiana was independent of European corn borer infestation. The impact of environmental conditions on the movement and colonization of the plants by B. bassiana is discussed.  相似文献

An aqueous suspension of Nosema marucae spores was sprayed on foliage of sorghum plants that had been infested with neonate Chilo partellus larvae at 3, 5, 7 and 9 weeks after (plant) emergence (WAE). These periods corresponded to plants at the tillering, early booting, soft dough and late maturation phenological stages, respectively. The extent of borer infestation and plant damage was monitored until crop harvest, and compared with plants which had been similarly infested but not sprayed with the pathogen, and with plants in insect‐free control plots. When applied early, infestation with neonate borer larvae caused most damage to the plants and the greatest reduction in yield in the untreated plots. Early treatment with N. marucae spores at 3 and 5 WAE resulted in the greatest reduction in damage to plants and the highest improvement in yield of seeds. It is apparent, therefore, that for maximum crop protection the microsporidian needs to be applied when sorghum is in an early phenological stage.  相似文献

Parasitism ofOstrinia nubilalis egg masses byTrichogramma minutum was observed in maize-bare ground monocultures and polycultures of maize/bean/squash and maize/clover. Parasitism rates were 1.9 times higher in monocultures than in polycultures; seasonal phenology of parasitism, however, was similar in both. Parasitism first occurred during the late whorl/early tassel stage of maize; peak parasitism occurred during the mid to late tassel stage, and then dropped off rapidly. We speculate that direct predation of egg masses, and ammensal preemptive competition for egg masses byColeomegilla maculata may have contributed to the decline in parasitism rates in both monocultures and polycultures during the latter part of the season.   相似文献

We tested the combined effect of the fungus Beauveria bassiana and the microsporidium Nosema pyrausta on the European corn borer larvae, Ostrinia nubilalis, in the laboratory. The first instar of O. nubilalis larvae was the most sensitive to the B. bassiana infection followed by the fifth, second, third, and fourth instar (LC50s were 4.91, 6.67, 7.13, 9.15, and 6.51 × 105 conidia/ml for the first to fifth instars, respectively). Mortality of each instar increases positively with concentration of conidia. When B. bassiana and N. pyrausta were used in combination, mortality increased significantly in all instars. Relative to the B. bassiana treatment alone, the B. bassiana + N. pyrausta treatment decreased the LC50s by 42.16%, 37.63%, 21.60%, 27.11%, and 33.95% for the first to fifth instars, respectively. The combined effects of the two pathogens were mostly additive. However, at the two highest concentrations the pathogens interacted synergistically in the first and second instar. Individuals that survived the B. bassiana and B. bassiana + N. pyrausta treatments and developed into adults had significantly shorter lifespans and females oviposited fewer eggs than non-exposed insects. The effects on the longevity and the egg production were most pronounced at high concentration of B. bassiana conidia.  相似文献

The relative potential of the pathogenic fungi Beauveria bassiana and Zoophthora radicans for use as autodisseminated biological control agents of the diamondback moth (Plutella xylostella) was compared. The LC50 of B. bassiana conidia to third instar larvae was 499 conidia/mm2 of leaf surface and individual cadavers of mycosed fourth instar larvae yielded a mean of 67.5 × 106 (±7.5 × 106) conidia. All concentrations of B. bassiana tested in inoculation chambers (0.24, 2.4, and 6.2 μg/mm2) induced 100% mortality in adult male moths within 7 days. The times to death and sporulation were concentration and exposure duration dependent. A standard procedure for inoculating male moths resulted in >85% mortality from Z. radicans and >93% mortality from B. bassiana. Pairing of inoculated males with clean moths of both sexes yielded higher rates of passive transmission of B. bassiana than Z. radicans, but there was no evidence for sexual transmission of either pathogen. Similarly, B. bassiana was more effectively transmitted from inoculated male moths to larvae foraging on whole plants. Single sporulating cadavers producing B. bassiana or Z. radicans conidia placed on plants infested with larvae resulted in a similar rate of transmission for both pathogens. However, an increase of the density of sporulating cadavers from one to three/plant increased Z. radicans transmission (greater than fourfold) but had no effect on B. bassiana transmission. Simultaneous inoculations of larvae with conidia of both fungi reduced the mortality induced by each pathogen, the reduction being most acute for B. bassiana-induced mortality. Inoculation of adults with both fungi showed that, at concentrations required for effective passive transmission to larvae, B. bassiana severely inhibited Z. radicans mycosis in adults.  相似文献
