Lepidocaryum tenue, Mauritia flexuosa and Mauritiella armata belong to the subtribe Mauritiinae, one early divergent lineage of the Arecaceae and one of the few of Calamoideae that occur in South America. These species occur in swampy environments and have fruits that are characteristically covered with scales. The objective of this study was to describe the formation of the layers of the pericarp within this subtribe and attempt to correlate fruit structure with the environment where species typically occur. Toward this goal, flowers in pre-anthesis and anthesis and fruits throughout development were analyzed using standard methods for light microscopy. The ontogeny of the layers of the pericarp of all three species was found to be similar. The scales were formed from non-vascularized emergences composed of exocarp and mesocarp. The median mesocarp accumulates lipids only in M. flexuosa and M. armata. The inner mesocarp together with the endocarp becomes papyraceous and tenuous in all species. This internal region of pericarp showed collapsed cells due to seed growth at the end of fruit development. Fruits of Mauritiinae are baccate, and the characters of the pericarp, especially the inner mesocarp and endocarp, help to maintain moisture. On the other hand, many species close to Mauritiinae show pericarp with sclerenchyma adjacent to the seed. This variation can contribute to understand the importance of this striking character in dispersal, germination and colonization in Arecaceae.  相似文献

比较了福建省 1 0个主栽龙眼品种果实的果皮形态和结构 ,结果表明 :不同品种在果皮厚度、外果皮表面颜色、龟状纹、放射线、瘤状突、刺毛、外果皮皮孔、周皮层厚度、栓质层厚度和连续性、中果皮薄壁组织细胞排列、石细胞大小、含量、排列和分布 ,维管束发达状况、排列和分布 ,内果皮表皮细胞排列和角蜡质层厚度等方面均存在着明显差异。风梨味、东壁、油潭本、乌龙岭、红核子、蕉眼龙眼果皮厚 ,外果皮表面瘤状突和剌毛多 ,外果皮周皮层、栓质层厚且连续性好 ,中果皮石细胞 (团 )含量多且排列紧密 ,分布在中果皮外侧且在中果皮中所占比例大 ,维管束发达且排列有序 ,内果皮角蜡质层厚 ;这些品种果实耐贮运、抗病性强。而水涨、赤壳、福眼、普明庵龙眼果皮薄 ,外果皮周皮层薄、栓质层不发达 ,中果皮石细胞 (团 )含量少、分布分散 ,维管束不发达 ,薄壁组织细胞胞间隙大 ,皮孔间隙大、皮孔通道与中果皮组织细胞间隙相通 ;这些品种的果实不耐贮运、抗病性弱。讨论了龙眼外果皮表面主色为褐色和内果皮比外果皮更容易褐变的解剖学原因及龙眼果皮形态结构与果实耐贮运的关系。  相似文献

Lignin is a major component of stone cells in pear fruit, which significantly affects fruit quality. Hydroxycinnamoyl CoA: shikimate hydroxycinnamoyl transferase (HCT), a recently discovered enzyme in plants, is an important gene that participates in the formation of lignin. Although HCT gene cloning and expression patterns have been studied in several species, including pear, there is still no extensive genome-wide bioinformatics analysis on the whole gene family, and the evolutionary history of HCT gene family is still unknown. A total of 82 HCT genes were identified in pear, most of which have one or two exons, and all with the conserved HXXXD motif and transferase domains. Based on the structural characteristics and phylogenetic analysis of these sequences, the HCT gene family genes could be classified into four main groups. Structural analysis also revealed that 25 % of HCT genes share a MYB binding site. Expansion of the HCT gene family mostly occurred before the divergence between Arabidopsis and Rosaceae, with whole-genome duplication or segmental duplication events playing the most important role in the expansion of the HCT gene family in pear. At the same time, purifying selection also played a critical role in the evolution of HCT genes. Five of the 82 HCT genes were verified by qRT-PCR to correspond to the pattern of stone cell formation during pear fruit development. The genome-wide identification, chromosome localization, gene structures, synteny, and expression analyses of pear HCT genes provide an overall insight into HCT gene family and their potential involvement in growth and development of stone cells.  相似文献

Factors Determining the Use and Cultivation of Moringa oleifera Lam. in the Republic of Benin. Despite its nutritious leaves and considerable economic importance, the agroforestry species Moringa oleifera Lam. is still considered a neglected and underutilized species. To contribute to the development of an effective valorization strategy for M. oleifera, this study identified factors driving its use and cultivation in Benin. To this end, an ethnobotanical survey through individual interviews (n?=?801) was performed in 46 localities across biogeographical zones in Benin. Conditional inference tree–based classification models allowed us to identify factors that mostly influence the use, cultivation, and cultivation system of M. oleifera. Awareness, knowledge of the plant biology, gender, cultivation system, and age are factors influencing the use of M. oleifera. Cultivation systems are driven by ethnicity, knowledge of the plant’s biology, and the main socioprofessional activity. Effective valorization of M. oleifera requires awareness rising on its usefulness while providing knowledge on the plant biology.  相似文献

The phloem-unloading pathway of sucrose and mechanism of sugar-to-oil transition are still unknown in Camellia oleifera Fruit. Here, transmission electronic microscopy (TEM) and confocal laser-scanning microscopy (CLSM) were used to observe the cellular structure of vascular bundles and symplastic tracer, carboxyfluorescein (CF), transport in phloem zone. The results showed that sucrose was transported via symplast system in the early and late phases, whereas apoplast system exerted the function in middle stage. Moreover, enzymatic assays showed that acid invertase had a higher activity at the transition stage during the whole fruit development. The cell wall bound invertase (CWI) activity reached the highest at the middle stage of fruit development and the switch in phloem-unloading coincided with fruit developmental phase change and oil accumulation. Correlation analysis showed that the oil accumulation was significantly negatively correlated with content of soluble sugar at P < 0.05 level. However, the soluble acid invertase (SAI), CWI, and neutral invertase showed a significant positive correlation with oil accumulation at P < 0.01 level. In summary, our data provide new cytological insights into the transition of unloading transfer between symplasmic and apoplasmic patterns in C. oleifera fruit and suggest that invertases are positively involved in sugar–oil transition process.  相似文献

The drumstick tree (Moringa oleifera Lam.) is a perennial crop that has gained popularity in certain developing countries for its high-nutrition content and adaptability to arid and semi-arid environments. Here we report a high-quality draft genome sequence of M. oleifera. This assembly represents 91.78% of the estimated genome size and contains 19,465 protein-coding genes. Comparative genomic analysis between M. oleifera and related woody plant genomes helps clarify the general evolution of this species, while the identification of several species-specific gene families and positively selected genes in M. oleifera may help identify genes related to M. oleifera’s high protein content, fast-growth, heat and stress tolerance. This reference genome greatly extends the basic research on M. oleifera, and may further promote applying genomics to enhanced breeding and improvement of M. oleifera.  相似文献

Camellia oleifera is a subtropical evergreen plant. Cultivated C. oleifera is the most important woody oil crop in China. Wild C. oleifera is an essential genetic resource for breeding. The patterns of genetic differentiation among altitudes/latitudes in wild C. oleifera are still unknown. Camellia oleifera may be predominantly hexaploid. The characteristics of polyploidy may lead to considerable biases in estimates of genetic diversity and differentiation. Our study used C. oleifera as a case study for analysing genetic diversity, structure and differentiation in polyploid plants using simple sequence repeats (SSRs). Wild C. oleifera samples were collected at different altitudes on the Jinggang and Lu mountains of China. The ploidy levels were determined with flow cytometry analysis. Eight highly polymorphic SSRs were used to genotype the samples. Genetic diversity and structure were analysed. Various estimates of genetic differentiation were compared. The flow cytometry results indicated that wild C. oleifera samples were all hexaploid at various altitudes of the Jinggang and Lu mountains. High levels of genetic diversity were found on both the Jinggang and Lu mountains. Genetic structure analyses indicated clear genetic differentiation between the Jinggang and Lu mountains and lower genetic differentiation among altitudes within each mountain. Classical genetic differentiation estimates of Fst failed to discriminate genetic differentiation between and within mountains. The Rho statistic showed a moderate level of genetic differentiation between mountains and lower levels of genetic differentiation within each mountain. Our study demonstrates that Rho is the statistic of choice for estimating genetic differentiation in polyploids.  相似文献

Abstract

Leaf characteristics, soluble protein and total soluble sugar contents as well as Rubisco-related gene expression in three cultivars of C. oleifera were measured during a combined drought and heat stress period and after subsequent rainfall events. Leaf relative water content (RWC) was significantly correlated with leaf area (LA), leaf fresh weight (FW), SPAD chlorophyll meter readings, and the levels of Co-rbcL and Co-rbcS expression. Results suggest that leaf RWC, LA, leaf FW, SPAD readings together with Co-rbcL and Co-rbcS expression can be used for evaluating responses of C. oleifera cultivars to combined drought and heat stress and subsequent recovery after rainfall events. Rubisco activase might be used for evaluating plant recovery after rainfall. This study identified cultivars differing in tolerance to the combined stress and recovery. Information derived from this study should be valuable for improving survivability and productivity of C. oleifera cultivars.

One of the applications of genomics is to identify genetic markers linked to loci responsible for variation in phenotypic traits, which could be used in breeding programs to select individuals with favorable alleles, particularly at the seedling stage. With this aim, in the framework of the European project FruitBreedomics, we selected five main peach fruit characters and a resistance trait, controlled by major genes with Mendelian inheritance: fruit flesh color Y, fruit skin pubescence G, fruit shape S, sub-acid fruit D, stone adhesion-flesh texture F-M, and resistance to green peach aphid Rm2. They were all previously mapped in Prunus. We then selected three F1 and three F2 progenies segregating for these characters and developed genetic maps of the linkage groups including the major genes, using the single nucleotide polymorphism (SNP) genome-wide scans obtained with the International Peach SNP Consortium (IPSC) 9K SNP array v1. We identified SNPs co-segregating with the characters in all cases. Their positions were in agreement with the known positions of the major genes. The number of SNPs linked to each of these, as well as the size of the physical regions encompassing them, varied depending on the maps. As a result, the number of useful SNPs for marker-assisted selection varied accordingly. As a whole, this study establishes a sound basis for further development of MAS on these characters. Additionally, we also discussed some limitations that were observed regarding the SNP array efficiency.  相似文献

The degradation of triglycerides in oil palm fruit due to an endogenous lipase in the pulp is the main reason for acidification of palm oil, which causes major economic losses and is currently mainly associated with the FLL1 gene. We designed this study to identify all the major genes controlling differences in acidity and lipase activity in the oil palm fruit mesocarp and determine a molecular markers kit to allow marker-assisted selection of commercial varieties with low acidity. Not only one gene (FLL1) but three closely linked genes including FLL1 were found and characterized in LM2T_EgCIR184O12c, a bacterial artificial chromosome sequence of 231 kb. Intra-gene PCR-based markers were designed for these genes. A QTL gene co-localization analysis for oil acidity (percentage of fatty acids released) was performed on two mapping populations. It evidenced a single major QTL at our lipase gene loci, explaining 84 to 92% of phenotypic variation, and validating the main genetic control of palm oil acidification by FLL1 and/or by the two new lipase genes. The three lipase genes had high homology to demonstrated triacylglycerol lipases. While FLL1 shows the highest expression levels, the two other genes may also contribute to oil acidity. Our molecular markers of lipase genes and the associated major QTL is an important step towards marker-assisted selection of commercial varieties with low acidity.  相似文献

Development and structure of the pericarp of Lannea discolor (Sonder) Engl.(Anacardiaceae). The exocarp develops from the outer epidermis and subepidermal, parenchymatous cell layers of the ovary wall. A parenchymatous zone with secretory cavities more or less delimits the exocarp internally. The inner part of the parenchymatous mesocarp is tanniniferous. The parenchymatous transition zone between mesocarp and sclercnchymatous endocarp or sderocarp, contains vascular tissue. The inner endocarp and operculum develop from the inner epidermis and subepidermal parenchyma of the ovary wall, while the outer endocarp develops from the parenchymatous zone with procambium strandS. Comparing the pericarp of L.discolor with those of Sclerocarya birrea subsp. caffra and Rhus lancea , the close affinity with Sclerocarya birrea subsp. caffra is evident.  相似文献

Rhynchospora is one of the most species-rich genera of Cyperaceae and one of the few with entomophilous species. Considering the few anatomical studies of the genus, especially of the reproductive structures, this study comparatively analysed the anatomy of flowers of two representative species of Rhynchospora, R. consanguinea and R. pubera, including the floral vasculature and the anatomy of the style base, which is persistent with the fruit. Both species have congested inflorescences with light-coloured bracts and bisexual flowers, and phenolic idioblasts in the anthers and gynoecium, characteristics that suggest insect pollination. In R. consanguinea, the bisexual and the most proximal male flower has perianths, a new character state reported for the genus. The floral vasculature pattern is similar in both species, but differs from that previously described for Rhynchospora. In both species, there are two vascular bundles in the rachilla, which split into three receptacular bundles, the latter forming a vascular plexus where the bundles of stamens, gynoecium and ovule are connected. No lateral carpellary trace was observed, and the presence of the abaxial receptacular bundle was interpreted as a vestige of the tricarpellate ancestral condition. In the fruit of both species, the thickened style base (stylopodium) has a parenchyma with idioblasts containing phenolic compounds and idioblasts with helical or reticulate cell wall thickenings. The stylopodium is a homologous structure in the species of Rhynchospora and to other genera of Cyperoideae and evolved several times in the subfamily.  相似文献

Drosophila suzukii (Matsumura) has been recently detected causing damage to strawberries in Brazil. Infestation in strawberry culture has often been observed jointly with the presence of Zaprionus indianus Gupta. This study investigated the susceptibility of strawberries at three ripening stages to infestation of D. suzukii and Z. indianus and their interaction. In the laboratory, strawberries cv. Albion at different ripening stages (green, semi-ripe and ripe) were exposed to D. suzukii and Z. indianus for 24 h in choice and no-choice bioassays. Additionally, we evaluated the effects of mechanical damage incurred artificially or by D. suzukii oviposition on Z. indianus infestation. In no-choice bioassay, there were no significant differences in fruit susceptibility to D. suzukii infestation at different ripening stages. However, in choice bioassay, D. suzukii adults preferred to oviposit on R fruit. The presence of mechanical damage did not increase susceptibility of fruit to D. suzukii oviposition. For Z. indianus, there was greater susceptibility of R fruit in relation to SR and G fruit in both the choice and no-choice bioassays. There was a significant and positive interaction of mechanical damage and damage caused by D. suzukii to R fruit and infestation by Z. indianus, which was not observed in SR and G fruit. Although infestation of Z. indianus is related to attack damaged or decaying fruit, this work shows that this species has the ability to oviposit and develop in healthy strawberry fruit with and increased infestation level when the fruit has damage to its epidermis.  相似文献

β-glucosidase (BG) was believed to take part in abscisic acid (ABA) synthesis via hydrolysis of ABA glucose ester to release active ABA during plant growth and development. However, there is no genetic evidence available to indicate the role of genes during fruit ripening. Here, the expression patterns of three genes (VvBG1, VvBG2, and VvBG3) encoding β-glucosidase were analyzed during grape fruit development, and it was found that β-glucosidase activity increased in grape fruit in response to various stresses. Furthermore, to verify the function of β-glucosidase during fruit ripening, heterogeneous expression of the VvBG1 gene in strawberry fruit was validated, and the results showed that the VvBG1 over-expression increased β-glucosidase and promoted the fruit ripening process in strawberry. In addition, we found that ABA contents increased in the VvBG1 over-expression of strawberry fruit, which induced fruit anthocyanin, soluble solid accumulation, and fruit softening. Moreover, genes related to coloring (CHS, CHI, F3H, and UFGT), softening (PG1, PL1, and EXP1), and aroma (SAAT, and QR) were up-regulated. This work will elucidate the specific roles of VvBGs in the synthesis of ABA and provide some new insights into the ABA-controlled grape ripening mechanism.  相似文献

Background

The shells of various Haliotis species have served as models of invertebrate biomineralization and physical shell properties for more than 20 years. A focus of this research has been the nacreous inner layer of the shell with its conspicuous arrangement of aragonite platelets, resembling in cross-section a brick-and-mortar wall. In comparison, the outer, less stable, calcitic prismatic layer has received much less attention. One of the first molluscan shell proteins to be characterized at the molecular level was Lustrin A, a component of the nacreous organic matrix of Haliotis rufescens. This was soon followed by the C-type lectin perlucin and the growth factor-binding perlustrin, both isolated from H. laevigata nacre, and the crystal growth-modulating AP7 and AP24, isolated from H. rufescens nacre. Mass spectrometry-based proteomics was subsequently applied to to Haliotis biomineralization research with the analysis of the H. asinina shell matrix and yielded 14 different shell-associated proteins. That study was the most comprehensive for a Haliotis species to date.

Methods

The shell proteomes of nacre and prismatic layer of the marine gastropod Haliotis laevigata were analyzed combining mass spectrometry-based proteomics and next generation sequencing.

Results

We identified 297 proteins from the nacreous shell layer and 350 proteins from the prismatic shell layer from the green lip abalone H. laevigata. Considering the overlap between the two sets we identified a total of 448 proteins. Fifty-one nacre proteins and 43 prismatic layer proteins were defined as major proteins based on their abundance at more than 0.2% of the total. The remaining proteins occurred at low abundance and may not play any significant role in shell fabrication. The overlap of major proteins between the two shell layers was 17, amounting to a total of 77 major proteins.

Conclusions

The H. laevigata shell proteome shares moderate sequence similarity at the protein level with other gastropod, bivalve and more distantly related invertebrate biomineralising proteomes. Features conserved in H. laevigata and other molluscan shell proteomes include short repetitive sequences of low complexity predicted to lack intrinsic three-dimensional structure, and domains such as tyrosinase, chitin-binding, and carbonic anhydrase. This catalogue of H. laevigata shell proteins represents the most comprehensive for a haliotid and should support future efforts to elucidate the molecular mechanisms of shell assembly.
