Summary In a study of the expression of folate-sensitive fragile sites in five normal individuals using RPMI medium containing methotrexate (MTX), we observed a high frequency of sister chromatid intercrossings (SCI) that is, the intersection of sister chromatids. The location of SCIs corresponded to fragile sites in 54.2% of the cases. Of the SCIs observed in each individual, 43%–53% were located at the same bands as their expressed fragile sites. Furthermore when RPMI+MTX medium was used instead of F-10 medium, the incidence of SCIs increased tenfold. We suggest that SCIs could indicate the existence of a pre-lesion.  相似文献

Summary The hypothesis that annual climatic variation can affect the nutritive value of host-plant tissue for a herbivore was tested. Larvae of a single generation of western spruce budworm, Choristoneura occidentalis Free., were fed foliages that had been collected on five consecutive springs from a stand of balsam fir Abies balsamea (L.) Mill. Budworm of both sexes were about 20 percent heavier at pupation when fed folages that had been produced in early rather than later spring seasons. This could result in an estimated 25 percent difference in the reproductive rate of females. It is concluded that annual variations in spring climate can affect the nutritive value of forage.  相似文献

The predatory bug,Orius majusculus(Reuter),is an important predator of thrips commercially produced for augmentative releases using the eggs of the Mediterranea n flour moth Ephestia kuehniella(Zeller).In this study,we assessed the potential for using frozen adults of fruit flies,Drosophila melanogaster(Meigen),either as nymphal rearing diet or as diet throughout the entire life-cycle.We compared life-hi story traits and reproduction of predators when fed D.melanogaster with high lipid body content(lipid-rich)and with high protein body content(protein-rich),using a diet of 100%E.kuehniella eggs as control.We also analyzed the biochemical composition of both prey and predator in order to assess the nutritional quality of each diet,which partially explained the adequacy of the different diets for O.majusculus.There were significant differences between predators fed the two types of D.melanogaster,with the protein-rich flies as diet providing the best results in terms of mortality and fecundity.Furthermore,we show that while feeding O.majusculus throughout their development with D.melanogaster increases mortality and reduces reproduction,protein-rich D.melanogaster can be used as nymphal diet with minimal reduction in reproductivity and minimal increase in mortality.  相似文献

Tachinids are usually considered as koinobionts as none of them kills or paralyzes the host when first entering it. These parasitoids, however, do not fit well into the koinobiont/idiobiont dichotomy because only some species show a high degree of physiological adaptation to the host, whereas the larvae of other species grow quickly following attack and kill the host rapidly, thus behaving more as idiobionts. The in vitro rearing technique provides further evidence for the poor adequacy of the koinobiont/idiobiont dichotomy for tachinids. In fact, while typical koinobionts are known to be difficult to culture on artificial media, some tachinids displaying non-synchronized development with the host have been successfully reared in vitro on insect material-free artificial media, thus behaving similarly to idiobionts. I therefore suggest not to use the koinobiont/idiobiont classification for tachinids and to class them instead only on the basis of the presence or absence of developmental synchrony with their hosts.  相似文献

Aldosterone-producing adenoma (aldosteronoma)--the most frequent form of primary hyperaldosteronism (PH)--is considered a specific form of diabetes mellitus (DM). In a previous study we demonstrated insulin resistance in patients with PH. We have therefore undertaken a study to evaluate the incidence of abnormalities of glucose metabolism in patients with PH (36 subjects) compared to control subjects with essential hypertension (EH) (21 patients). The following parameters were measured in all studied subjects: office blood pressure (by mercury sphygmomanometer in the sitting position), body mass index (BMI), plasma potassium, plasma glucose and insulin levels during oral glucose tolerance test (OGTT) (0, 60, 120 min), plasma renin activity and plasma aldosterone. Although patients with PH tended to have higher stimulated plasma glucose levels after 60 and 120 min compared to EH, these differences did not attain statistical significance. Patients with EH tended to have higher insulin levels at each measured interval, but due to a high variability these differences were again not significant. There were no significant differences between PH and EH in the proportion of diabetics (20% vs. 14%) or patients with impaired glucose tolerance (18% vs. 10%). In conclusion, we have found the absence of significant differences in the frequency of diabetes mellitus, impaired glucose tolerance and insulin resistance in patients with EH and PH. Our data thus do not support the idea of primary hyperaldosteronism as a specific type of diabetes mellitus. Furthermore, our results indicate that glucose metabolic characteristics in essential hypertension and primary hyperaldosteronism tend to be similar. The definitive conclusion with respect to the possible causal relationship between DM and PH, however, can be obtained only on larger groups of subjects, in particular after the evaluation of the effect of surgical/pharmacological treatment of primary hyperaldosteronism.  相似文献

In cell therapy protocols, many tissues were proposed as a source of mesenchymal stem cells (MSC) isolation. So far, bone marrow (BM) has been presented as the main source of MSC despite the invasive isolation procedure related to this source. During the last years, the umbilical cord (UC) matrix was cited in different studies as a reliable source from which long term ex vivo proliferating fibroblasts were isolated but with contradictory data about their immunophenotype, gene expression profile, and differentiation potential. Hence, an interesting question emerged: Are cells isolated from cord matrix (UC-MSC) different from other MSCs? In this review, we will summarize different studies that isolated and characterized UC-MSC. Considering BM-MSC as gold standard, we will discuss if UC-MSC fulfill different criteria that define MSC, and what remain to be done in this issue.  相似文献

Mating frequency of the bulb mite, Rhizoglyphus robini Claparède (Astigmata: Acaridae) was recorded for 23 days on 6 concentrations of peanut extract. The largest numbers of couplings and of adults were obtained on the undiluted (100%) extract, and the fewest on the most diluted (10%) diet. Good association between mating frequency and adult numbers was also observed on several other foods.  相似文献

In this paper, we study an analytical model describing predatory behaviour. It is assumed that the parameter describing the predator's behaviour is its satiation. Using semigroup methods and compactness arguments we prove that a stable satiation distribution is reached if t. In this paper we analyse a stochastic model for invertebrate predation taking account of the predator's satiation. This model approximates Holling's hungry mantid model when handling time is negligible (see Part I). Formalin has long been the standard fixative for clinical routines worldwide. After the Formaldehyde Standard became law in the US in 1987, as a result of increasing concerns about the potential carcinogenicity of formaldehyde, attempts have been made to find safer alternatives. Alcoholic formalin is a useful fixative, because in addition to fixation, dehydration also is begun. For centuries, honey has been known to be an antibacterial agent with the potential to preserve compounds without harmful effects on its users. We compared the effects of honey fixation with other routine fixatives using conventional histochemical and immunohistochemical staining methods. Our results demonstrated that tissues fixed in either honey or alcoholic formalin and 10% neutral buffered formalin (NBF) have similar histomorphology. Honey fixation showed minor histomorphological differences among the various tissues; however, it did not influence affect correct diagnostic conclusions. Our results suggested that honey can be used as a safe alternative to formalin in histopathology.  相似文献

Tree and shrub species can be differentiated into two major groups based on their substantially different leaf anatomy: heterobaric and homobaric. In contrast to homobaric leaves, heterobaric leaves have bundle sheath extensions (BSEs) that create transparent regions on their lamina. Recent studies have shown that BSEs transfer visible light to internal mesophyll layers, thus affecting the photosynthetic performance of heterobaric leaves. Whether the two leaf types also differ in other functional and structural traits has not been addressed, nor have any structure-function relations. Here, we measured key anatomical and physiological parameters and tested their relationships in 30 species with different leaf types. Heterobaric leaves were thinner with lower leaf mass per area, had higher nitrogen concentration per mass, were (13)C-enriched, and achieved comparable photosynthetic capacity per area but had higher photosynthetic capacity per mass compared to homobaric leaves. Relations between leaf construction cost, nitrogen concentration, and photosynthesis followed the general pattern of the "leaf economic spectrum," but differed between homobaric and heterobaric leaves. We suggest that the mechanisms controlling these relations differ between the two leaf types, presumably due to their distinct anatomy.  相似文献

Mate guarding is a male strategy to monopolize matings and thus to ensure paternity. Since in callitrichids female reproductive status is advertised by scent marks, one may expect mate guarding by chemical means. We addressed this question during an episode of consortship observed in a polyandrous trio of wild saddleback tamarins (Saguinus fuscicollis). During consortship, the consort male was the only one to allomark the female. Scent marking frequency decreased for all individuals, although the consort male marked more than the other male during consortship, while there was no difference in the previous period. During consortship, almost 50% of female scents were overmarked by the consort, and more than 56% of the consort's scent marks were employed to overmark the female's scents. Therefore, the other male had limited access to female scent marks. Mate guarding may thus have a chemical component in tamarins, and olfactory communication may play an important role in mating competition.  相似文献

According to current theoretical predictions, any deleterious mutations that reduce nonsexual fitness may have a negative influence on mating success. This means that sexual selection may remove deleterious mutations from the populations. Males of good genetic quality should be more successful in mating, compared to the males of lower genetic quality. As mating success is a condition dependent trait, large fractions of the genome may be a target of sexual selection and many behavioral traits are likely to be condition dependent. We manipulated the genetic quality of Drosophila subobscura males by inducing mutations with ionizing radiation and observed the effects of the obtained heterozygous mutations on male mating behavior: courtship occurrence, courtship latency, mating occurrence, latency to mating and duration of mating. We found possible effects of mutations. Females mated more frequently with male progeny of nonirradiated males and that these males courted females faster compared to the male progeny of irradiated males. Our findings indicate a possible important role of sexual selection in purging deleterious mutations.  相似文献

Forest fragments embedded within agricultural landscapes have the potential to provide a “forest reset effect” by mitigating agricultural effects on water quality, and acting as refugia and conservation reserves for aquatic species. We investigated the ability of forest fragments to reset agricultural effects using four catchments in the South Island, New Zealand. Two catchments were dominated by agricultural activities, but each had an isolated forest fragment in the lower valley, and two catchments had continuous riparian forest along the valley floor. Riffles sampled in continuous forest were generally deeper than those in agricultural and forest fragments, and not surprisingly streams in forest fragments and continuous forest received less light than those in agricultural land. All sites had circum-neutral pH, but both conductivity and temperature were significantly lower at continuous forest sites than agricultural and forest fragment sites. Taxonomic richness, Margalef’s index and numbers of Ephemeroptera, Plecoptera and Trichoptera (EPT) taxa were significantly higher in continuous forest than at forest fragment sites, but overall invertebrate densities did not differ between fragments and continuous forest. Several taxa were abundant at agricultural and forest fragment sites, but absent or at low densities in continuous forest. They included the blackfly Austrosimulium spp. and two caddisflies Pycnocentrodes sp. and Hydrobiosis parumbripennis. Conversely, the mayflies Austroclima sp. and Coloburiscus humeralis and the blepharicerid Neocurupira chiltoni were either restricted to continuous forest, or abundant in continuous forest but rare in agricultural and forest fragments. An ordination of communities separated those in agricultural and continuous forest sites, but communities at forest fragment sites were clustered among the agricultural sites. In this study forest fragments of 5–7 ha, located in the lower reaches of the catchment did not mitigate the negative upstream effects of agriculture on stream functioning. Fragment size (or riparian forest length), riparian forest width and vegetation type, and fragment location in the catchment may have critical roles in enabling forest fragments to reset the negative impacts of agriculture. Determining these characteristics of fragments has important consequences for stream remediation.  相似文献

Chronic calcium pyrophosphate crystal arthritis is a clinical consequence of the formation and deposition of these crystals in joints and can result in persistent arthritis. Curative treatment would require the removal of crystals from joints and tissues, but to date all agents tested have proven ineffective. Management of the inflammatory manifestations of chronic calcium pyrophosphate disease includes glucocorticoids, non-steroidal anti-inflammatory drugs, or colchicine, and responses are usually satisfactory. However, in some patients, the response to these agents is poor or they are contraindicated. Methotrexate had been reported as a promising option in small case series; however, in a recent issue of Arthritis Research & Therapy, a clinical trial failed to confirm the anticipated benefits. Here, we discuss some issues that might have influenced the results of the study, before deciding to abandon methotrexate as a therapeutic option for patients with chronic calcium pyrophosphate arthritis.  相似文献

Host-specific parasites of humans are used to track ancient migrations. Based on archaeoparasitology, it is clear that humans entered the New World at least twice in ancient times. The archaeoparasitology of some intestinal parasites in the New World points to migration routes other than the Bering Land Bridge. Helminths have been found in mummies and coprolites in North and South America. Hookworms (Necator and Ancylostoma), whipworms (Trichuris trichiura) and other helminths require specific conditions for life-cycle completion. They could not survive in the cold climate of the northern region of the Americas. Therefore, humans would have lost some intestinal parasites while crossing Beringia. Evidence is provided here from published data of pre-Columbian sites for the peopling of the Americas through trans-oceanic or costal migrations.  相似文献
