峰值骨密度是由遗传和环境因素及其相互作用共同决定的复杂性状。维生素D受体基因、雌激素α受体基因、白介素6基因、副甲状腺素基因、Ⅰ型胶原α2基因、骨钙素基因、α2巯基糖蛋白基因是与骨代谢相关的7个重要的候选基因。本研究旨在检测这7个候选基因之间的相互作用对中国女性峰值骨密度的影响。样本为中国上海的361个无关、健康的绝经前女性,均为汉族人,年龄为20—44岁。采用Hologic QDR2000+双能X射线扫描仪对腰椎与髋部的骨密度进行了检测。采用聚合酶链式反应-限制性片段长度多态分析方法对每个个体的以下8个多态性标记位点进行基因分型:维生素D受体基因的Apa Ⅰ位点,雌激素α受体基因的PvuⅡ和XbaⅠ位点,白介素6基因的BsrB Ⅰ位点,副甲状腺素基因的BstB Ⅰ位点,Ⅰ型胶原α2基因的Msp Ⅰ位点,骨钙素基因的Hind Ⅲ位点,娃巯基糖蛋白基因的SacⅠ位点。采用二元方差分析对基因相互作用与骨密度的关系进行研究。结果表明,白介素基因和雌激素α受体基因(PvuⅡ)的相互作用对髋部(P=0.019)、转子间区(P=0.016)和股骨颈(P=0.019)的骨密度有显著作用。在这3个部位,GGPp基因型携带者比GGpp基因型携带者的骨密度值分别高出18.0%、19.5%和14.8%。另外观察到醒巯基糖蛋白基因与自介素6基因的相互作用对股骨颈骨密度有显著影响(P=0.046)。GGSS基因型携带者的股骨颈骨密度值比GGSs基因型携带者高出18.8%。该项群体水平的统计分析表明:对于中国女性峰值骨密度的遗传决定,白介素基因和雌激素α受体基因、α2巯基糖蛋白基因的相互作用显著。  相似文献

研究组前期的全基因组关联研究发现PHACTR3基因与骨折关联,为了检测该基因与骨密度的关联关系,采用精细定位关联研究来检测PHACTR3基因内及其附近的SNPs与骨密度的关系。首先在中国样本(1627个不相关的汉族样本)和美国样本(2286个不相关高加索样本)中对PHACTR3基因的140个SNPs进行基因分型,然后采用Plink软件检测PHACTR3基因与腰椎和髋部骨密度的关联关系。发现研究组以前报道的与骨折关联的SNPs rs1555364和rs6064822与腰椎和髋部骨密度关联(P=4.89×10^-2-1.26×10^-2)。另外还发现位于PHACTR3基因内含子中3个SNPs位点(rs6027138,rs1182531和rs1182532)与中国人群和白人腰椎骨密度均显著关联,将中国人与白人样本合并起来进行荟萃分析(Meta—analysis),得到合并P值为1.40×10^-3到4.00×10^-4,另外发现rs6064820与髋部BMD相关联,合并P值为6.70×10^-3。本研究进一步证实了PHACTR3基因在骨密度变异中的作用,对骨质疏松发病机制的认识提供了新的理论依据。  相似文献

骨大小是一种独立于骨密度(BMD)的骨质疏松性骨折的重要风险因子。由于其高遗传率,充分了解控制骨大小的遗传因素有很重要的临床意义。文章研究目的为检测中国人群中α2-HS糖蛋白基因(AHSG)多态性和腰椎及髋部骨大小变异之间的关联。我们总共征集了来自中国401个核心家庭(包括父母亲及至少一个女儿)的1260个研究样本,并且分型了AHSG基因第7个外显子的Sac Ⅰ位点多态性。该位点核苷酸的替换(C→G)引起第238号丝氨酸被苏氨酸取代,因此可能对基因功能有影响。在任何骨骼位点,没有发现显著的群体分层。发现-HSG基因SacⅠ位点多态性和转子间(P=0.019)以及全髋的(P=0.035)骨大小呈显著性相关。该多态性位点能分别解释转子间和全髋3.74%和3.16%的骨大小变异。连锁分析没有检测到显著性结果,可能的主要原因是样本中同胞对的数目较少,统计效力较低,以及SacⅠ位点多态相对于微卫星标记对连锁分析提供的信息量少。结果表明,月HSG基因多态性可能和中国人群中髋部骨大小变异有关。  相似文献

最近的全基因组关联研究发现ZBTB40(zinc finger and BTB domain containing 40)基因是一个潜在的调节骨密度的新基因,为了可靠地验证该基因与骨密度的关联关系,采用精细定位关联研究来检测ZBTB40基因内及其附近的SNPs与骨密度的关系.首先在中国样本(1 627个不相关的汉族样本)和美国样本(2 286个不相关高加索样本)中对ZBTB40基因的50个SNPs进行基因分型,然后采用Plink软件检测ZBTB40基因与腰椎和髋部骨密度的关联关系.证实了以前报道的SNPs rs7524102与腰椎和髋部骨密度的关联关系.另外还发现5个SNPs(rs10917209、rs6426748、rs4433361、rs3856183和rs7550872)与腰椎和髋部骨密度相关联.其中最显著的SNP是位于ZBTB40基因上游区域的rs6426748,其与腰椎骨密度的关联P值为9.82×10-4,多重检验校正后仍然显著.连锁不平衡分析表明rs6426748与rs10917209,rs7524102和rs7550872呈现高度连锁关系,位于长度为27 kb的单体型块内,因此ZBTB40基因的功能致病位点可能是rs6426748或该单体型块内某一未分型的SNP.  相似文献

我们先前通过全基因组扫描发现lp36与体重指数显提示性连锁(LOD=2.09)。肿瘤坏死因子受体2(1NFR2)定位于lp36,是肥胖的一个极好的图位和功能侯选基因。本研究采用数量传递连锁不平衡检验在两个大的独立的白人样本中进行了TNFR2基因与肥胖表型的连锁与关联检验。第一组受试者由来自79个多代家系的1836个个体组成;第二组受试者由来自157个核心家庭的636个个体组成。所检测的肥胖表型包括体重指数、脂肪量和脂肪量百分数。在多代家系中我们发现TNFR2基因变异与BMI显著连锁(P=0.0056)。结果表明,TNFR2基因是影响白人BMI变异的一个数量性状位点。  相似文献

绝经后骨质疏松症(PMO)是一种多基因调控的遗传性疾病。雌激素受体β亚型基因是骨质疏松症的重要侯选基因。此文采用病例对照设计(78名股骨颈PMO病人和122名对照以及108名腰椎PMO病人和92名对照)研究中国人(汉族)雌激素受体β基因(ESR2)第5内含子CA重复序列多态性与PMO的相关性。以CA重复序列平均数22次为界将重复序列基因分为短基因(<22)和长基因(≥22),分别以S和L表示。股骨颈及腰椎(L2-4)部位,病例组中LL基因型和L等位基因者频率显著高于对照组(P<0.01),SL、LL及SL LL基因型者较SS基因型者患PMO风险显著增高(P<0.05);调整年龄、绝经时间、绝经年龄及体质指数后,Logistic回归分析显示ESR2(CA)n多态性仍然与股骨颈(OR4.923,95%CI1.986~12.203,P=0.001)及L2-4(OR2.267,95%CI1.121~4.598,P=0.023)PMO显著相关。结果显示:ESR2基因CA重复序列多态性与股骨颈和L2-4部位PMO独立关联,L等位基因显性影响PMO的发病风险。  相似文献

目的:探讨白细胞介素-10(IL-10)启动子-627C/A基因多态性和等位基因频率与过敏性哮喘血清IgE、IL-10浓度以及病情严重程度的相互关系。方法:从哮喘病人DNA文库中选择青岛地区过敏性哮喘病人518例和健康志愿者501例,采用PCR—RFLP方法对IL.10基因启动子.627位点多态性进行观察,比较两组基因型和等位基因的分布频率,同时测定血清中总IgE、IL-10浓度和肺功能检查(FEV1、FVC、FEV1/FVC)。结果:轻度和中一重度哮喘组AA、CA和CC基因型所占比例分别为38.1%、46.0%、15.9%和45.6%、46.2%和8.2%(P=0.0168,X2=8.232,df=2)。A等位基因与哮喘病轻的严重程度有明显相关性(P〈0.05)。AA基因型哮喘病人血清的IgE浓度显著升高(P〈0.01),但其血清IL-10浓度比CC基因型携带者明显降低(P〈0.01)。结论:IL-10基因启动子-627位点多态性与过敏性哮喘的发生有一定的相关性,等位基因A是哮喘患病的风险基因,而等位基因C则是哮喘病的保护基因。  相似文献

摘要目的:探讨新疆地区维吾尔族、汉族脑啡肽酶(Neprilysin,NEP)基因单核苷酸多态性与阿尔茨海默病(Alzheimer’SdiseaseAD)的关系。方法:对新疆地区维吾尔族、汉族≥50岁8284名人群进行AD流行病学调查,参照ADRDA.NINCDS的标准,选取散发性阿尔茨海默病(sporadicAlzheimer’s disease,SAD)患者209例(AD组)与正常对照220例(对照组),应用聚合酶链反应一限制性片段长度多态性技术(PCR)检测NEP基因多态性,采用病例一对照的关联分析方法对NEP基因rs3736187位点进行基因型和等位基因频率分析。结果:(1)新疆维吾尔族、汉族两民族AD组与对照组间NEP基因基因型频率和等位基因频率分布差异均有统计学意义(P〈0.05)。携带T等位基因个体出现AD的危险性高于携带c等位基因的个体(0R=1.981,P〈0.05)。(2)新疆维吾尔族、汉族不同民族之间比较NEP基因基因型频率和等位基因频率分布差异均无统计学意义(P〉0.05),而在同一民族中AD组和对照组之间比较NEP基因等位基因频率分布差异均具有统计学意义(P〈0.05)。(3)两个年龄分段(〈65岁及≥65岁)之间NEP基因基因型频率和等位基因频率分布差异均无统计学意义(P〉0.05),而在同一年龄段内部AD组与对照组间等位基因频率分布差异具有统计学意义(P〈0.05)。(4)男性、女性之间NEP基因基因型频率和等位基因频率分布差异均无统计学意义,而在女性AD组与对照组间等位基因频率分布差异有统计学意义(P〈0.05)。结论:NEP基因rs3736187位点基因型频率和等位基因频率在新疆维吾尔族、汉族两民族间的分布相似;NEP基因的T等位基因是SAD的危险因素,在新疆维吾尔族、汉族两民族及女性SAD的发病中起重要作用。  相似文献

为了探讨RANK/RANKL/OPG和WNT信号通路中6个基因10个单核苷酸多态性位点与中老年男性髋部骨强度的关联,征集788名广州地区40岁以上男性为研究对象,采用Mass Array检测基因型,双能X线吸收仪测定股骨颈骨密度(bone mineral density,BMD)、横截面积(cross-sectional area,CSA)、平均皮质厚度(average cortical thickness,ACT)、剖面模数(section modulus,SM)和抗屈曲率(buckling ratio,BR),以多重线性回归分析各位点与骨表型的关联。结果发现:OPG基因rs3134069的C等位基因与BMD(β=-0.017,P=0.024)和ACT(β=-0.004,P=0.040)负相关;LRP5基因rs3736228的T等位基因与低BMD、CSA、ACT(β=-0.050~-0.003,P0.050)和高BR(β=0.242,P=0.043)相关,rs491347的G等位基因与低BMD相关(β=-0.012,P=0.046)。研究结果支持了OPG和LRP5基因与中国中老年男性人群髋部骨强度相关。  相似文献

利用PCR—SSCP技术对高繁殖力绵羊品种(小尾寒羊、湖羊、德国肉用美利奴羊)和低繁殖力绵羊品种(多赛特羊、萨福克羊)的雌激素受体(estrogen receptor,ESR)基因第一外显子部分序列进行单核苷酸多态性研究。结果表明:小尾寒羊、湖羊和德国肉用美利奴羊中存在3种基因型(AA、BB、AB),而在多赛特羊和萨福克羊中只存在两种基因型(AA、AB)。统计结果表明:湖羊、德国肉用美利奴羊、小尾寒羊、萨福克羊和多赛特羊A等位基因频率分别为0.672、0.786、0.846、0.857和0.867,B等位基因频率分别为0.328、0.214、0.154、0.143和0.133。测序结果表明:BB型和AA型相比在外显子1第363位发生1处碱基突变(C→G)。独立性检验表明:小尾寒羊和湖羊之间基因型分布差异极显著(P〈0.01),湖羊和多赛特羊之间基因型分布差异显著(P〈0.05),其他各个绵羊品种之间基因型分布差异均不显著。A8基因型和BB基因型小尾寒羊产羔数比AA基因型分别多0.51只(P〈0.05)和0.7只(P〈0.05)。研究结果表明:ESR基因可能是控制小尾寒羊多胎性能的一个主效基因或与之存在紧密的遗传连锁。  相似文献

Calcium sensing receptor (CASR) is a central factor involved in calcium metabolism. Interleukin-6 (IL-6) is a pleiotropic cytokine that plays an important role in osteoclast differentiation. Thus, both CASR and IL-6 are important in bone and mineral metabolism and are prominent candidate genes for osteoporosis. The study aimed to test association and/or linkage between the CASR and IL-6 genes with bone mineral density (BMD) variation in a Chinese population. A cytosine-adenine (CA)n repeat polymorphism in the CASR gene and the IL-6 gene was genotyped, respectively, in 1 263 subjects from 402 Chinese nuclear families. Employing tests implemented in the program QTDT (quantitative transmission disequilibrium tests), a significant total association of the CASR (CA)12 allele (P = 0.006) and (CA)18 allele (P = 0.02) with BMD at the femoral neck was found. For the IL-6 gene, significant within-family associations were found between the (CA)14 allele and BMD at the total hip (P = 0.021), the femoral neck (P = 0.041), and the intertrochanteric region (P = 0.029). A significant linkage was also observed between IL-6 CA repeat polymorphism and BMD at the spine (P = 0.001). The results suggest that the CASR gene and the IL-6 gene may have effects on BMD variation in Chinese.  相似文献

Osteoporosis is a disease that is strongly genetically determined. Aromatase converts androgens to estradiol in postmenopausal women, therefore polymorphisms of the gene for this enzyme may be associated with bone mass and fracture. We investigated the association of the TTTA microsatellite polymorphism in intron 4 of the aromatase (CYP19) gene with bone mineral density (BMD) and fracture in 1,257 women aged 70 yr and greater. The data obtained were stratified based on the presence or absence of a [TTTA]n of 7 (A2), determined from a preliminary analysis of hip dual-energy X-ray absorptiometry BMD, which was present in 27% of the population. The presence of an A2 allele was associated with a higher free estradiol index (0.52 +/- 0.49, P = 0.049) compared with the absence of an A2 allele (0.47 +/- 0.45); higher BMD at all sites of the hip (3.4% total hip, 2.3% femoral neck, 3.6% intertrochanter, 4.1% trochanter) and the lumbar spine (12.7%); higher values for the calcaneal quantitative ultrasound parameters broadband ultrasound (1.3%), speed of sound (0.4%), and stiffness (3.7%); and higher peripheral quantitative computed tomography measures for total (3.4%), trabecular (3.3%), and cortical BMD (3.3%) and the derived stress strain index (SSI) parameters SSI polar (6.4%) and SSI x (6.8%) values. A lower deoxypryridinoline creatinine ratio was observed in subjects with an A2 allele (30.3 +/- 10.4 vs. 27.1 +/- 9.1, P = 0.03). The A2 allele was associated with a lower prevalence of vertebral fracture in subjects who were osteoporotic (odds ratio 0.27, confidence interval 0.09-0.79). Therefore, a common polymorphism of the aromatase gene, perhaps in linkage disequilibrium with a functionally significant CYP19 polymorphism, is associated with bone structure and bone turnover, either by local effects or by effects on circulating bioactive estrogen.  相似文献

探讨细胞色素P450 19 (CYP19) 基因Val80多态性及护骨素(OPG) 基因A163G多态性与绝经后女性骨密度 (BMD) 的关系。随机选择居住在重庆的绝经后女性200例, 采用多聚酶链反应-限制性片段长度多态性法检测Val80及A163G多态性, 采用Norland公司XR-46系列双能X线骨密度仪测量股骨近端及腰椎BMD。 200名绝经后女性中Val80基因型GG、GA及AA的频率分别为19.5%、44.5%及36.0%; A163G基因型GG、GC 及CC的频率分别为: 13.0%, 42.0%及45.0%; 基因型频率分布均符合Hardy-Weinberg平衡 (P>0.05)。协方差分析及多元逐步回归分析显示CYP19基因第3外显子Val80多态性与绝经后女性BMD无相关性 (P>0.05)。除大转子外, A163G位点AG/GG/AG+GG基因型者股骨颈、Ward’s三角及腰椎BMD均较AA基因型者低, A163G基因型与股骨颈、Ward’s三角及腰椎BMD有相关性 (P<0.05)。OPG基因启动子区A163G多态性分布存在明显的种族差异, 且与绝经后女性BMD有一定关联, AA型对BMD具有一定的保护作用, G等位基因是BMD降低的危险因素。  相似文献

Chromosomal mapping of the human estrogen receptor beta (ERbeta) gene by fluorescence in situ hybridization (FISH) reveals that ERbeta is located at human chromosome 14, region q23-24.1, where the aberration of DNA copy number in the bone disorders is frequently involved. Then, we investigated the association between dinucleotide (cytosine-adenine; CA) repeat polymorphism located in the flanking region of ERbeta gene and bone mineral density (BMD) in 204 healthy postmenopausal Japanese women. The genotype was classified into "A" through "O" according to the number of the repeats, from 18 to 32. BMD was expressed in Z score (a deviation from the weight-adjusted average BMD of each age using the standard deviation as a unit). When we separate the subjects into two groups bearing at least one I allele (26 CA repeats) and those who did not, the former subjects had significantly higher Z score of L2-4 BMD (mean +/- standard deviation; 0.674 +/- 1.53 vs 0.128 +/- 1.38; P = 0.027). These data suggest that genetic variation at the ERbeta locus may be associated with some determinants for BMD and the possible involvements of this polymorphism in the cause of postmenopausal osteoporosis in Japanese women.  相似文献

Osteoporosis is a highly heritable common bone disease leading to fractures that severely impair the life quality of patients. Wrist fractures caused by osteoporosis are largely due to the scarcity of wrist bone mass. Here we report the results of a genome-wide association study (GWAS) of wrist bone mineral density (BMD). We examined ~500000 SNP markers in 1000 unrelated homogeneous Caucasian subjects and found a novel allelic association with wrist BMD at rs11023787 in the SOX6 (SRY (sex determining region Y)-box 6) gene (P=9.00×10(-5)). Subjects carrying the C allele of rs11023787 in SOX6 had significantly higher mean wrist BMD values than those with the T allele (0.485:0.462 g cm(-2) for C allele vs. T allele carriers). For validation, we performed SOX6 association for BMD in an independent Chinese sample and found that SNP rs11023787 was significantly associated with wrist BMD in the Chinese sample (P=6.41×10(-3)). Meta-analyses of the GWAS scan and the replication studies yielded P-values of 5.20×10(-6) for rs11023787. Results of this study, together with the functional relevance of SOX6 in cartilage formation, support the SOX6 gene as an important gene for BMD variation.  相似文献

Osteoporosis is a major public health problem for old people.Genetic factors are considered tobe major contributors to the pathogenesis of postmenopausal osteoporosis.The vitamin D receptor(VDR)gene is a prominent candidate gene for the regulation of postmenopausal bone mass;however,despite exten-sive studies,controversy remains regarding its association with postmenopausal body mineral density(BMD)variation.In this study,a total of 260 healthy postmenopausal Chinese women were genotyped at the VDRApaI locus using polymerase chain reaction(PCR)-restriction fragment length polymorphism (RFLP).Rawhip BMD was significantly associated with VDR ApaI polymorphism with and without adjusting for age(P=0.015 and 0.040,respectively).This genetic effect can explain 3.32% of hip BMD variation.However,the significant association vanished after correcting for both age and body mass index(BMI)(P=0.169).Inaddition,we observed a significant association between VDR ApaI polymorphism with unadjusted BMI(P=0.042)or BMI adjusted for age(P=0.049).The raw hip BMD was also found to be significantly corre-lated with BMI(r=0.517,P=0.0001),with BMI explaining 26.35% of the variation of hip BMD.All of thesefacts prompt us to conclude that the significant association between the VDR ApaI genotype and hip BMDmay be modified by BMI in postmenopausal Chinese women.Our findings may partially explain the earlierinconsistent association results concerning the VDR gene and BMD,and highlight the importance of incorpo-rating covariates such as BMI into osteoporosis association studies.  相似文献
