以Actinobacillus succinogenes NJ113为出发菌株,经硫酸二乙酯(DES)诱变,在含8~20 g/L硫酸铵平板中筛选到一株耐铵型突变株YZ25,该菌株在含8 g/L硫酸铵培养基中厌氧发酵,琥珀酸产量达32.68 g/L,比出发菌提高了180.5%,对葡萄糖收率达65.4%,副产物乙酸、甲酸产量分别下降3.5%、28.7%,琥珀酸/乙酸比值由0.63提高到2.5。在7.5 L发酵罐中,用氨水调节pH分批实验,发酵34 h琥珀酸产量达27.13 g/L,较出发菌株提高了85.3%。 相似文献

以实验室筛选到的一株枯草芽孢杆菌(Bacillus subtilis)B-1为出发菌株,采用紫外诱变技术对出发菌株进行反复诱变,得到一株能够利用玉米原料生产γ-聚谷氨酸的优良高产菌株B-115,摇瓶发酵γ-聚谷氨酸的产量由原菌株的12.5g/L提高到19.5g/L。再以该菌株为研究对象利用响应面法进行碳源、氮源、谷氨酸钠、金属离子等发酵条件的优化实验,经48h摇瓶发酵,γ-聚谷氨酸产量达到40.98g/ L。 相似文献

以肌苷产生菌枯草杆菌7171-9-1为出发菌株,经物理、化学诱变剂连续处理,获得一株腺嘌呤、组氨酸、硫胺素三重营养缺陷型并对8-氮杂鸟嘌呤、6-巯基嘌呤有双重抗性的突变株JSIM-1019。在摇瓶和发酵罐试验中,该变异株的肌苷产量显著高于亲株。摇瓶试验产肌苷达20g/L,最高可达24.83g/L。工业生产试验最高达14.5g/L,稳定在12g/L。发酵周期平均为43.8小时。菌株遗传性状稳定。 相似文献

戊糖乳杆菌(Lactobacillus pentosus)是能利用木质纤维素水解液发酵产乳酸的潜力菌株,发酵条件优化与高产菌株的选育是提高乳酸产量的重要手段。通过单因素试验、Plackett-Burman设计与响应面试验,对戊糖乳杆菌ATCC 8041产乳酸的发酵培养基及发酵条件进行了优化。结果表明,该菌株发酵培养基的最佳组合为葡萄糖93.11 g/L、酵母浸粉5.19 g/L、碳酸钙29.43 g/L、蛋白胨10.00 g/L、Na2HPO4·12H2O 5.00 g/L、Mg SO4 0.20 g/L、Mn SO4 50 mg/L;最佳发酵条件为37℃、p H6.5、接种量6%、装液量80%。在此优化条件下,该菌株发酵产乳酸为54.12 g/L。进一步以戊糖乳杆菌ATCC 8041为出发菌株,通过原生质体进行紫外诱变,经多重筛选,最终获得一株遗传稳定性好的高产乳酸突变株,命名为戊糖乳杆菌Lactic UVC-02,由中国典型培养物保藏中心保存,注册号为CCTCC M 2013209。该突变株Lactic UVC-02经葡萄糖发酵,乳酸产量达64.17 g/L,比出发菌株ATCC 8041(54.12g/L)提高18.6%。 相似文献

对拉曼被孢霉突变株F5发酵生产γ—亚麻酸的最适碳源、氮源、发酵时间及温度、无机盐离子添加、最适碳源浓度及补加碳源时间等发酵条件进行了研究探讨。最适发酵培养基组成为 (g/L) :葡萄糖 1 0 0 ,酵母浸出粉 4 ,蛋白胨 1 ,K2 HPO4 1 ,CaCl2 1× 1 0 - 2 ,MgSO4 5× 1 0 - 2 ,FeSO4 1× 1 0 - 2 ,ZnSO4 7.5× 1 0 - 3,CuSO4 0 .5× 1 0 - 3,MnSO4 2× 1 0 - 3,pH 6.0。培养温度为 2 5℃ ,1 4 0r/min振荡培养 1 0天 ,培养 8天后 (即收获前 2天 )补加 5 %葡萄糖。发酵结果为 :DC 2 4 .5 9g/L ,TL 1 0 .84g/L ,TL/DC 4 4.0 9% ,GLA/TL 1 0 .67% ,GLA产量为 1 1 5 6.63mg/L。GLA产量较初始结果提高 1 5 6.1 5 %。该菌株已达到工业化生产菌株要求 相似文献

为了获得高产γ-亚麻酸(γ-linolenic acid,GLA)的菌株,利用苏丹黑染色法筛选获得1株GLA产生菌EIM-10,通过摇瓶培养,其生物量可达11.882g/L,菌丝体油脂含量可达18.86%。气相色谱-质谱(GC—MS)分析表明其γ-亚麻酸质量分数(占总脂肪酸)高达27.68%。为进一步鉴定该菌株,克隆测定了该菌18SrRNA基因序列,并对其进行系统进化树分析,结果表明该菌属于毛霉属,与Mucor racemosus、Mucor plumbeus、Mucor ramosissimus与Mucor circinelloides同属一个分支。 相似文献

A newly isolated strain of Cunninghamella echinulata grown on glucose produced significant quantities of biomass and cellular lipids in media with high C/N ratio. The oil yield from glucose consumed increased after nitrogen exhaustion in the growth medium, but gamma-linolenic acid (GLA) content in cellular oil systematically decreased during the lipid accumulation process. When lipid accumulation was completed, GLA concentration in the cellular lipids progressively increased. The highest GLA production (720 mg/l) was achieved in medium with a C/N ratio equal to 163. C. echinulata was also able to grow on orange peel. The C/N ratio in the orange peel decreased from 50 to 26 during solid-state fermentation. Maximum oxygen uptake was observed during assimilation of reducing sugars, whereas a polygalacturonase activity was detected after reducing sugars had been exhausted. The maximum GLA production was 1.2-1.5 mg/g of fermented peel, calculated on a dry weight basis. After enrichment of the pulp with inorganic nitrogen and glucose, an increase in the production of oil and GLA was observed. 相似文献

In this work, a glutaryl-7-aminocephalosporanic acid acylase (GLA) coding gene was cloned from Pseudomonas diminuta NK703 which was screened from oilfield. The concerted effects of the expression system, inducing condition and culture medium on the expression of NK703 GLA in E. coli were firstly investigated. The best combination was the recombinant E. coli strain of pET-28a + GLA/BL21(DE3) with 2.0% (w/v) lactose inducing in YT medium at 25 °C. Then, by optimizing the components of culture medium, a synthetic medium with dextrin and a feeding medium with glycerol as the carbon sources were developed to further enhance the GLA yield and improve the GLA solubility. In the end, the NK703 GLA activity increased about 50-fold, reached 14,470 ± 465 U/L, and the GLA productivity and the proportion of soluble GLA to the total soluble protein attained 206.0 ± 9.033 U L−1 h−1 and 60.13%, respectively. Scaling up the GLA production in 3.7 L fermenter under the optimized conditions identified in shake flask, the GLA activity also reached 12,406 ± 521 U/L, which was the highest report at fermenter level. 相似文献

Twenty standard amino acids were examined as single nitrogen source on the growth, total fatty acids production, and yield of γ-linolenic acid (GLA) in Mucor circinelloides. Of the amino acids, tyrosine gave the highest biomass and lipid accumulation and thus resulted in a high GLA yield with respective values of 17.8 g/L, 23 % (w/w, dry cell weight, DCW), and 0.81 g/L, which were 36, 25, and 72 % higher than when the fungus was grown with ammonium tartrate. To find out the potential mechanism underlying the increased lipid accumulation of M. circinelloides when grown on tyrosine, the activity of lipogenic enzymes of the fungus during lipid accumulation phase was measured. The enzyme activities of glucose 6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase, and ATP-citrate lyase were up-regulated, while NADP-isocitrate dehydrogenase was down-regulated by tyrosine during the lipid accumulation phase of the fungus which suggested that these enzymes may be involved in the increased lipid biosynthesis by tyrosine in this fungus. 相似文献

Abstract γ-Linolenic acid (GLA) production using a high GLA producing marine green alga, Chlorella sp. NKG 042401, was studied. GLA was presented in the galactolipid fraction (37.9%/total fatty acids). The effects of growth conditions on GLA production were studied. Optimum salinity for GLA production was 5 g 1−1 , at which salinity the highest cell concentration was achieved, resulting in a 1.6-fold increase in GLA productivity. Total fatty acid, however, was not drastically affected by change of salinity. Nitrogen starvation decreased the ratio of unsaturated fatty acids, and consequently GLA ratio in total fatty acid decreased. The urea adduct method was used to concentrate GLA from crude extract. As a result, after 5 sequential concentration procedures, GLA was concentrated 5-fold with a yield of 49%. 相似文献

A mutant of Arthrospira platensis PCC 9108, strain M9108, obtained by mutagenesis with UV treatment, was able to mixotrophically grow in an SOT medium containing 40 g of glucose/l. The biomass and specific growth rate of strain M9108 (4.10 g/l and 0.70/d) were 1.9-fold and 1.4-fold higher, respectively, than those of the wild type (2.21 g/l and 0.58/d) under mixotrophic culture condition. In addition, when compared with the wild type, the content of gamma- linolenic acid (GLA) in the mutant was increased when glucose concentration was increased. Compared with the wild type, the GLA content of the mutant was 2-fold higher in autotrophic culture and about 3-fold higher in mixotrophic culture. Thus, the mutant appears to possess more efficient facility to assimilate and metabolize glucose and to produce more GLA than its wild-type strain. 相似文献

研究了不同培养条件对钝顶螺旋藻(Sp)NS-90020脂肪酸合成的影响。随着温度升高,其不饱和脂肪酸,γ一亚麻酸(GLA)相对含量降低,总脂肪酸含量升高,当温度为40℃时总脂肪酸和γ-亚麻酸绝对含量都达到最大值,分别为73.4mg/g干重和11.9mg/g干重;当培养基中NaCl浓度高于0.017mol/L时,其GLA相对含量降低,但低于0.0017mol/L时,对其脂肪酸组成无显著影响;氨水使其脂肪酸和GLA绝对含量升高,并在50mgN(NH3·H2O)时达到最大值,分别为67.96mg/g干重和13.63mg/g千重;暗处理92h使其总脂肪酸和GLA绝对含量升高;缺乏Fe2 或Mg2 或Mo2 时,其总脂肪酸和GLA绝对含量降低,而缺乏PO43-时,其总脂肪酸和GLA绝对含量略有升高。 相似文献
